本工作用 ICP-AES 法测定稀土-6063变形铝合金中 La、Ce、Pr、Nd、Sm、Gd、Fe、Mg、Ti、Zn,Cu 和 Mn,本方法不用化学分离,可一次同时测定这些元素,并做了 ICP光源参数、基体量变化、酸度(HCl)变化、稀土总量变化、共存元素 Fe 和 Mg 量的变化对被测元素的影响。本方法速度快、准确度高(除 Cu 稍差)。回收率91.0~110.0%,相对标准偏差±1.4~8.0%。

中华绒螯蟹是我国重要的水产经济动物,近年来养殖规模不断扩大,产量持续增加。但是,伴随着养殖规模的扩大,养殖环境也日益恶化并导致了大量疾病的发生,严重制约了中华绒螯蟹养殖业的健康发展。因此,疾病预防和控制对中华绒螯蟹养殖业的可持续发展具有举足轻重的作用。与其他无脊椎动物一样,中华绒螯蟹的免疫系统没有免疫球蛋白和淋巴细胞,而是依靠由细胞免疫和体液免疫构成的固有免疫系统来对病原进行识别和清除。中华绒螯蟹的固有免疫机制的研究有助于推动中华绒螯蟹病害防治工作的开展。 本研究采用大规模EST测序方法,结合末端快速扩增(rapid amplification of cDNA ends,RACE)技术从中华绒螯蟹血淋巴中克隆到了过氧化物还原酶(peroxiredoxin,EsPrx6)和硫氧还蛋白(thioredoxin,EsTrx1)基因的cDNA 全长序列;采用实时荧光定量PCR 技术检测了这两个基因在健康个体中表达的组织分布情况以及鳗弧菌刺激后血淋巴细胞中的时序表达规律;同时,将这两个基因的编码区克隆到pET 系列载体,并在大肠杆菌中实现了重组表达,并进行了体外活性检测。 过氧化物还原酶是一个抗氧化蛋白超家族,在保护机体免受活性氧(reactive oxygen species,ROS)的伤害中发挥着重要作用。中华绒螯蟹Prx6(EsPrx6) 基因的cDNA 全长为1076 bp,5` UTR(untranslated region,UTR) 为69 bp,3` UTR 为347 bp,开放阅读框(open reading frame,ORF)为660 bp,编码219 个氨基酸的蛋白。mRNA 3`-端具有多聚腺苷酸加尾信号(polyadenylation signal)AATAAA 和polyA 尾巴。EsPrx6 的预测分子量为 24 kDa,理论等电点为6.21,具有一个保守的Prx 结构域、一个AhpC 结构域和过氧化物酶催化活性中心PVCTTE,表明EsPrx6 属于1-Cys 型Prx。在所检测的组织中均有EsPrx6 的表达,其中以肝胰腺表达量最高,为血淋巴细胞中表达量的17.4 倍。鳗弧菌刺激后,血淋巴细胞中EsPrx6 的表达下降,到12 h 时,实验组显著低于对照组(P<0.05);随时间推移,表达水平逐渐回升,但在整个实验期间,都没有恢复到起始水平。将EsPrx6 进行体外重组并在大肠杆菌E. coli BL21(DE3)中实现表达,重组EsPrx6 具有预期的抗氧化活性和过氧化物酶活性,其中抗氧化活力为14.69 U/mg 蛋白,高于相同条件下GSH 的抗氧化力(P<0.05),过氧化物酶活力为23.46 U/mg 蛋白。结果表明,EsPrx6 作为一种重要的抗氧化剂,在中华绒螯蟹抵御ROS 可能引起的氧化损伤方面具有重要作用。 硫氧还蛋白是广泛存在于生物体内的一种具有硫醇依赖性的具有还原活性的蛋白。中华绒螯蟹Trx1(EsTrx1)基因的cDNA 全长为641 bp,5` UTR 为17 bp,3` UTR 为306 bp,开放阅读框为318 bp,编码105 个氨基酸。EsTrx1 的预测分子量为12.2 kDa,理论等电点为4.8。EsTrx1 不含信号肽,其氨基酸序列与其他动物的Trx1s 具有高度相似性,如与地中海黄蝎的Trx1 相似度达到73%;而与其他物种Trx2 的同源性很低,相似度仅为14.3-22.8%,表明EsTrx1 属于Trx1 亚族。实时荧光定量PCR 检测发现,EsTrx1 在鳃、性腺、肝胰腺、肌肉、心脏和血淋巴细胞中都有表达。血淋巴细胞中EsTrx1 mRNA 的表达量在菌刺激后上升,刺激后6 h,实验组表达量显著高于对照组和空白组(P<0.05),然后逐渐恢复到刺激前水平。为进一步探讨其生物学功能,将EsTrx1 进行体外重组并在大肠杆菌E. coli BL21(DE3)得到表达,重组EsTrx1 具有预期的氧化还原调节活性,抗氧化活力为3.06 U/mg,且抗氧化活力高于GSH(P<0.05)。rEsTrx1 的二硫键还原活力为5.03,低于凡纳滨对虾的二硫键还原活力(10.44),接近于大肠杆菌(4.93),小牛胸腺(6.50)和小牛肝脏(5.09),而高于鲍鱼Trx2(1.83)活力。结果表明,EsTrx1 在生理条件下能够作为一种重要的抗氧化剂,参与对细菌感染的免疫应答反应。

Three kinds of steels were studied using electrically connected hanging specimen in the corrosion simulation device and offshore long scale hanging specimen. The experimental results obtained by the two methods show that the device can better reflect the offshore corrosion environment. A Ni-Cu-P steel specimen was studied through analysis of the specimen's corrosion products and corrosion types. The surface of the samples before and after the removal of the rust layer produced by these two methods were observed and compared after some experiments. The microstructure of the corrosion products under different marine environments were analyzed and compared through IR. It indicated good correlation between the electrically connected hanging specimen method and the long scale hanging specimen method.

A corrosion simulation device was studied using offshore long scale hanging specimens. An Ni-Cu-P steel specimen was studied by analysing its corrosion products and corrosion types. The appearance of the samples and the surface of the metallic substrate after the removal of the rust layer produced by these two methods were observed and compared after 470 days of exposure. The phase structure of the corrosion products under different marine environments were analysed and compared. It further indicated good correlation between the electrically connected hanging specimen method and the long scale hanging specimen method.

Effect of alloy elements on corrosion of low alloy steel was studied under simulated offshore conditions. The results showed that the elements Cu, P, Mo, W, V had evident effect on corrosion resistance in the atmosphere zone; Cu, P, V, Mo in the splash zone and Cr, Al, Mo in the submerged zone.

本文报道了高原鼢鼠和高原鼠兔整体骨骼及头骨、脊柱骨和下肢骨中Cu、Zn、Fe、M n、Co、N i、Mo、Cr、T i、F、Se 和Ge12种必需微量元素的含量, 并进行了显著性差异和相关性分析。结果表明: 只有Cu 的含量在两种动物骨骼中没有显著性差异(P > 0.05) , 其余11种元素均有显著性或极显著性差异(P < 0.05或P < 0.01) , 其中Zn、Fe、M n、Co、N i、Mo、F 和Se 的含量, 高原鼢鼠> 高原鼠兔, 而Cr、T i 和Ge 的含量, 高原鼠兔> 高原鼢鼠。微量元素在头骨、脊柱骨和下肢骨中分布很不均衡, 以头骨的微量元素最为丰富。

The effects of hypoxia on the levels of essential macroelements and trace elements (K, Na, Ca, Mg, Cu, Zn, Fe, and Mn) in the heart muscles of Wistar rats and plateau pikas (Ochotona curzoniae) were studied by atomic absorption spectrometry. Unlike the rat, the plateau pika is tolerant to hypoxia. The levels of K, Na, and the trace element Mn were not significantly changed in rat or pika hearts after exposure to hypoxia for 1, 10, or 25 d at simulated altitudes of 5000 and 7000 m. Other minerals (Ca, Mg, Cu, Zn, and Fe) were significantly affected by hypoxia and the levels followed different time-courses under different hypoxic regimes in these two animals. There were marked differences between the rat and pika in myocardial accumulation of essential elements such as Ca, which was increased to high levels in the rat but not affected in the pika. The results suggest that hypoxia affects animal physiological mechanisms by regulating the levels of essential elements.

随着荧光光谱技术的进展 ,三维荧光光谱被广泛应用于研究各种天然水体中溶解有机质 (包括腐殖质 )的各种环境行为 .本文利用三维荧光光谱和荧光猝灭滴定技术研究河流溶解有机质与Hg(Ⅱ )的相互作用 .结果表明 ,溶解有机质中含有的3种类型荧光基团都能够不同程度地被Hg(Ⅱ )猝灭 ,并采用非线性回归分析拟合出Hg(Ⅱ )与 3种荧光基团之间的条件稳定常数 .pH值对Hg(Ⅱ ) DOM体系具有显著影响 .Cl-对Hg(Ⅱ ) DOM体系具有强烈的竞争作用 .Ca(Ⅱ )能够 使Hg(Ⅱ ) DOM体系荧光增强 ,而Mg(Ⅱ )对之影响很小 .此外 ,Cu(Ⅱ )与Hg(Ⅱ ) 都是DOM的荧光猝灭剂 ,但是其猝灭机理存在差异

岩石风化成土作用是生态系统矿质元素的重要来源,也是全球矿质元素生物地球化学循环的重要环节。因此,岩石风化成土过程中矿质元素的行为与环境质量研究有助于我们深入了解矿质元素的生物地球化学循环及其生态环境效应,同时对农业生产和环境保护也具有重要意义。选择乌江流域沉积岩-土壤-植被-河流系统作为研究对象,采集了多个小流域盆地岩石、土壤、植物及溪水样品。通过测试样品的矿物、元素组成以及元素存在形态等,分析了喀斯特流域的矿物学与地球化学特征,研究了喀斯特流域的成土物源、风化成土方式及风化成土作用的控制因素,探讨了风化成土过程中矿质元素的行为与环境质量,为进一步进行喀斯特地区土壤-植被系统矿质元素的生物地球化学循环研究奠定基础,为喀斯特地区生态环境保护和修复以及稀土等矿质元素的农业利用提供科学依据,同时为核废料的地质处置提供参考。通过研究,取得了如下一些成果和认识: (1) 提出了弱淀积元素的概念,改进了侵蚀前后风化剖面中元素的演化模型。 (2) 硅酸盐岩地区土壤来源于下伏基岩的风化成土作用,其风化成土过程中存在脱硅富铝作用。 (3) 连续碳酸盐岩地区土壤最终来源于碳酸盐岩的风化成土作用,其风化成土作用存在四种方式和两个阶段。脱钙(镁)富铝硅和脱硅富铝两个阶段的研究结果,支持了王世杰等(1999)提出的碳酸盐岩两阶段风化成土模式。 (4) 岩性和地形是研究区风化成土作用的主要控制因素,植被和其它因素只是对其风化成土产物的理化性质进行了一些改造。 (5) 由于侵蚀、脱钙和元素本身的淀积作用,相对于研究区黄壤、中国土壤(CS)和世界土壤(WS),研究区石灰土富集Mn、V、P、REE 和U 等矿质元素。风化成土过程中矿质元素的行为主要受岩性和地貌部位控制,还受元素性质、土壤pH 值、氧化还原条件、铁锰(氢)氧化物和有机质含量、粘土矿物组成以及植被条件等的影响。 (6) 石灰岩等岩石风化成土作用可以通过植物营养供给和元素相互作用影响土壤质量和植物生长。 (7) 石灰岩等岩石风化成土作用控制着溪水和乌江河流的pH 值以及Mg、Ca、P、Mn 和U 等矿质元素的分布特征。 (8) NH4Ac-HAc单一提取法可用于评价石灰土中Mg、Ca、K、Mn和P等多数矿质元素的植物可利用性。与何子平等(2001)的研究结果不同,石灰土中多数矿质营养元素对植物的供给能力高于其它土壤。

由于缺乏相应的沉积地层,贵州新生代地质、环境演化恢复一直是学术界的难点问题。夷平面研究的兴起,为陆地地壳抬升、地貌演变等反演提供了理论基础,而风化壳正是夷平面研究的重要替代对象,尤其是对各类沉积地层不发育的地区。因而,贵州高原各级台地上发育有较为广泛的红色风化壳自然地、就成了主要研究载体。然而一与其它岩类不同,碳酸盐岩风化成土作用过程研究并没有得到同等重视,其研究范围和程度均很不充分。显然,要想利用碳酸盐岩风化壳进行夷平面等地质、地貌意义研究,就必需详细了解贵州或中国南方碳酸盐岩风化成土的基本过程。也只有如此,才能明确各种对比指标的使用条件和范围,才能明确各种测年结果的具体意义,才能真正地通过贵州碳酸盐岩风化壳获取全省地质、环境演化信息。本论文通过对贵州省岩溶台地碳酸盐岩石灰土、红色风化壳剖面形成地球化学过程的剖析,开展石灰土与红色风化壳之间的关联和差异性综合对比、红色风化壳之间发育程度的比较,以及风化壳剖面尝试性的侧年等方面研究,取得了以下儿点认识:1、石灰土形成地球化学过程及对比通过矿物组成、粒度分布特征、地球化学方法判别,确定了本文所选石灰土剖面物质来源于下伏碳酸盐岩中的酸不溶物,是碳酸盐岩风化后酸不溶物原地残余堆积的结果。石灰土形成的地球化学过程都遵循碳酸盐岩风化成土之两阶段模式:母岩-酸不溶物,大量可溶性物质Ca、Mg、Mn、P、Na淋失,而Si、K·Fe、Al、Ti则相对富集。该过程的矿物组成变化表现为,伴随碳酸盐矿物(方解石、白云石等)的溶解、迁移,硅酸盐相矿物(长石、2:1型粘土矿物等)残余、累积。酸不溶物,土层或土层的演化,K、Si(长石、伊利石)不断淋失,而Al(伊利石、高岭石、三水铝石)、Fe(针铁矿、赤铁矿等)逐渐富集,体现出脱硅、富铝(铁)过程。我们强调碳酸盐岩风化成土分为两个阶段,但也认识到风化作用是综合的、复杂的:在碳酸盐矿物大量淋失过程中,同时也进行着酸不溶物的风化。石灰土分为原生和次生两种成因类型。原生石灰土的风化程度总体较低,剖面中还残留一定量的CaCO3,剖面从下到上矿物组成、地球化学各种指标具有逐渐演化的过渡特征。原生石灰上土层演化过程显示出正风化序列,从下到上风化程度逐渐增大、可溶性物质逐渐减少、稳定和相对稳定组分(Al、Fe.、REE)含量不断增大。另外,主量元素的质量迁移系数在剖面中的变化与风化程度呈明显的相关性。而次生石灰土不但具有很高的风化程度,而且剖面无过渡特征,显示可能是由遭受强烈风化作用的物质直接转变形成的。剖面中也含有一定量后期加入的碳酸盐,但与高CIA值特征不相匹配;SiO2、Al2O3、Fe2O3组分具有不同的变化特征,与)成化程度无明显关联;REE在剖面岩一上界面出现富集层。2、碳酸盐岩红色风化壳形成地球化学过程及其与石灰土的对比红色风化壳形成的地球化学过程也遵循碳酸盐岩风化成土之两阶段模式:母岩一酸不溶物,大量可溶性物质Ca、Mg、Mn、P、Na淋失,而Si、K、Fe、Al、Ti则相对富集。该过程的矿物组成变化表现为,伴随碳酸盐矿物(方解石、白云石等)的溶解、迁移,硅酸盆相矿物(长石、2:1型粘土矿物等)残余、累积。酸不济物一土层或上层的演化,K、Si(长石、伊利石)不断淋失,而Al(伊利石、高岭石、三水铝石)、Fe(针铁矿、赤铁矿等)逐渐富集,体现出脱硅、富铝(铁)过程。石灰土与红色风化壳之间既存在共性,又有一定的差异性。红色风化壳与原生石灰土一样都具有过渡特征,但后者仅在剖面下部出现一定的过渡层位;原生石灰士的城化程度远低于红色风化壳,仅与后者底部土层相当;原生石灰土不具有红色风化壳之岩-土REE(超常)富集层。总体上,原生石灰土可以作为红色风化壳的前身。次生石灰土与红色风化壳在矿物组成、风化程度等各种地球化学指标等都十分接近(略低),显示与红色风化壳之间存在某种关联:由红色风化壳物质直接转变,或由酸不溶物已强烈风化的(白云岩)岩粉、碎裂岩风化形成。3、碳酸盐岩风化成土母岩差异性碳酸盐岩风化成土存在母岩差异性,石灰土母岩差异性较为明显,而红色风化壳的母岩差异性较弱,体现出红色风化壳的均一化特征。石灰土母岩差异性体现为:母岩为灰岩的石灰土剖面主量元素含量变化特征较为一致,而白云岩则具有不同的变化;灰岩石灰土Fe3+、Al具有不同的富集、亏损特征,且具有富集层的补偿亏损层,而白云岩石灰上两组分都呈现无补偿层位的相同富集态势;灰岩石灰土剖面出现负Eu异常和具以Gd为中心的倒“V”型MREE稀土富集特征(母岩标准化),而白云岩石灰土无明显Eu异常和具L既E或HREE稀士.富集特征;灰岩和白云岩石灰土的微量元素富集、亏损特征也有所不同,前者从上到下逐渐富集,而后者无明显变化趋势。红色风化壳形成的基本过程、主量元素地球化学行为无明显的母岩相关性,没有如石灰土剖面灰岩和白云岩之间存在的差异性,显示出红土化作用的均一化过程;微量元素中受重矿物影响的元素与母岩类型有一定的关联,但大多数元素的亏损、富集等与具体剖面的微环境关系密切。4、碳酸盐岩红色风化壳发育程度的对比及意义多利指标对比显示所选红色风化壳的风化程度有一定的强弱差别:平坝剖面>湖潮剖面>天龙剖面、大兴剖面>花溪剖面、新蒲剖面,但这种差异性不足以否定各剖面隶属于贵州山盆期广泛夷平面的基本推测。各剖面之间的微弱差异是红色风化壳形成时纬度分带和垂向分带的体现。低纬度的平坝、湖潮、天龙剖面经历的风化作用强于高纬度的新蒲、大兴剖面;高海拔的新蒲剖面风化程度低于海拔低的大兴剖面。5、红色风化壳次生石英裂变径迹测年红色风化壳次生石英裂变径迹方法测年尝试,得到一定的结果和认识:晶形相对较好的石英,是次生的,与母岩中碎屑及成岩阶段没有关联;各剖面上部与中、下部次生石英的诱发裂变径迹的密度有着明显的区别,可能暗示它们的形成环境不同;名一剖面石英的形成或退火年龄分布较为分散,1.2-25.2Ma,同一剖面的石英年龄也不均一,变化较大;从下到上,年龄值呈现出逐渐减小的规律性,与风化作用的正常序列相反。如果能确切其成因,次生石英裂变径迹年代学研究将是确定风化壳形成时代及揭示主要风化作用信,却钩有利武器。

BACKGROUND: When the nature and direction of research results affect their chances of publication, a distortion of the evidence base - termed publication bias - results. Despite considerable recent efforts to implement measures to reduce the non-publication of trials, publication bias is still a major problem in medical research. The objective of our study was to identify barriers to and facilitators of interventions to prevent or reduce publication bias. METHODS: We systematically reviewed the scholarly literature and extracted data from articles. Further, we performed semi-structured interviews with stakeholders. We performed an inductive thematic analysis to identify barriers to and facilitators of interventions to counter publication bias. RESULTS: The systematic review identified 39 articles. Thirty-four of 89 invited interview partners agreed to be interviewed. We clustered interventions into four categories: prospective trial registration, incentives for reporting in peer-reviewed journals or research reports, public availability of individual patient-level data, and peer-review/editorial processes. Barriers we identified included economic and personal interests, lack of financial resources for a global comprehensive trial registry, and different legal systems. Facilitators identified included: raising awareness of the effects of publication bias, providing incentives to make data publically available, and implementing laws to enforce prospective registration and reporting of clinical trial results. CONCLUSIONS: Publication bias is a complex problem that reflects the complex system in which it occurs. The cooperation amongst stakeholders to increase public awareness of the problem, better tailoring of incentives to publish, and ultimately legislative regulations have the greatest potential for reducing publication bias.

Coastal zooplankton have been investigated since 1984 at a Long Term Ecological Research station MC (LTER-MC) in the inner Gulf of Naples (Tyrrhenian Sea, Western Mediterranean). The sampling site, located between the littoral and the open sea systems, has very active hydrography that affects plankton communities. The present work was aimed at establishing whether, in such a dynamic and variable environment, species associations and homogeneous periods could be identified as characteristic and stable features of the mesozooplankton over the period 1984–2006. Hierarchical clustering was applied to assess species associations based on a matrix of similarities between species (R-mode), and homogeneous periods based on a matrix of similarities between observations (Q-mode). The Indicator Value index [IndVal, Dufrene and Legendre (1997) Species assemblages and indicator species: the need for a flexible asymmetrical approach. Ecol. Monogr., 67, 345–366] was calculated to identify species characterizing each period. Five taxonomic groups with well-defined composition and abundance were identified as robust associations that likely reflect different modes of community functioning. The temporal course of these associations was largely shaped by strong seasonal forcing comprising both physical and biological (e.g. trophic) signals. These associations persisted over the long term, thus indicating some stable characters in the Naples zooplankton time-series, providing evidence of resilience in communities in highly variable coastal conditions.

Decreased survival in patients with cystic fibrosis has been related to FEV1, BMI, and infection with Burkholderia cepacia complex (BCC). We have assessed the relationship of blood, sputum, and urine inflammatory markers to lung function, BMI, colonization with B cenocepacia (Bc), and patient survival. Thirty-nine stable cystic fibrosis (CF) patients (10 with Bc) were enrolled in a study to determine the effect of alpha-1-antitrypsin on airways inflammation. Pre-treatment measurements were used in this study. Demographics, sputum microbiology, heart rate, oxygen saturation, lung function were recorded. Blood samples were obtained for white blood count (WBC), C-Reactive Protein (CRP), and plasma neutrophil elastase/AAT complexes (pNEC). Neutrophil elastase (NE), neutrophil elastase/AAT complexes (sNEC), interleukin-8 (IL-8), TNF-receptor 1 (sTNFr), and myeloperoxidase (MPO) were measured in sputum and urinary desmosine concentration determined. Patients with Bc had significantly higher levels of pNEC, 332?±?91.4 ng/ml (mean?±?SEM) versus 106?±?18.2 ng/ml (P?=?0.0005) and sNEC, 369?±?76.6 ng/ml versus 197?±?36.0 ng/ml compared to those who were not. Five deaths were reported at the end of 1 year, (four with Bc) (P?=?0.011). Patients who subsequently died had significantly lower lung function FEV1, 1.2?±?0.2 L versus 2.0?±?0.1 L (P?=?0.03) and FVC, 2?±?0.3 L versus 3.1?±?0.2 L (P?=?0.01), compared to those that survived. There was significantly higher NE activity, 3.6?±?1.6 U/ml versus 1.5?±?0.6 U/ml (P?=?0.03), pNEC, 274?±?99 ng/ml versus 142?±?30 ng/ml (P?=?0.05), MPO, 163?±?62 mcg/ml versus 54?±?6.9 mcg/ml (P?=?0.03), and urinary desmosines 108?±?19.9 pM/mg creatinine versus 51.1?±?3.3 pM/mg creatinine (P?=?0.001), in those patients who subsequently died compared to those that survived. These data suggest there is increased neutrophil degranulation in patients infected with Bc and these patients have a poor outcome.
