Polyol sugars, displaying a plurality Of hydroxyl groups, were shown to modulate tetra hydroxyborate (borate) cross-linking in lidocaine hydrochloride containing poly(vinyl alcohol) scini-solid hydrogels. Without polyol, demixing of borate cross-linked PVA hydrogels into two distinct phases was noticeable upon lidocaine hydrochloride addition, preventing further use as a topical System. D-Mannitol incorporation was found to be particularly suitable in cicumventing network constriction induced by ionic and pH effects upon adding the hydrochloride salt of lidocaine. A test formulation (4% w/v lidocaine HCl, 2% W/V D-mannitol, 10% w/v PVA and 2.5%, w/v THB) was shown to constitute an effective delivery system, which was characterised by an initial burst release and a drug release mechanism dependent on temperature, changing from a diffusion-controlled system to one with the properties of a reservoir system. The novel flow properties and innocuous adhesion of PVA-tetrahydroxyborate hydrogels Support their application for drug delivery to exposed epithelial surfaces, Such as lacerated wounds. Furthermore, addition of a polyol, such as mannitol, allows incorporation of soluble salt forms of active therapeutic agents by modulation of cross-linking density. (C) 2008 Elsevier B.V. The reduction of guanine was studied by microelectrode voltammetry in the room temperature ionic liquids (RTILs) N-hexyltriethylammonium his (trifluoromethanesulfonyl) imide [N-6.2.2.2][N(Tf)(2)], 1-butyl-3-methylimidazolium hexafluorosphosphate [C(4)mim] [PF6], N-butyl-N-methyl-pyrrolidinium bis(trifluoromethanesulfonyl)imide [C(4)mpyrr][N(Tf)(2)], 1-butyl-3-methylimidazolium bis(trifluoromethanesulfonyl)imide [C-4mim][N(TF)(2)], N-butyl-N-methyl-pyrrolidinium dicyanamide [C(4)mpyrr][N(NC)(2)] and tris(P-hexyl)-tetradecylphosphonium trifluorotris(pentafluoroethyl)phosphate [P-14,P-6,(6,6)][FAP] on a platinum microelectrode. In [N-6,N-2,N-2,N-2][NTf2] and [P-14,P-6,P-6.6][FAP], but not in the other ionic liquids studied, guanine reduction involves a one-electron, diffusion-controlled process at very negative potential to produce an unstable radical anion. which is thought to undergo a dimerization reaction, probably after proton abstraction from the cation of the ionic liquid. The rate of this subsequent reaction depends on the nature of the ionic liquid, and it is faster in the ionic liquid [P-14,P-6,P-6.6[FAP], in which the formation of the resulting dimer can be voltammetrically monitored at less negative potentials than required for the reduction of the parent molecule. Adenine showed similar behaviour to guanine but the pyrimidines thymine and cytosine did not; thymine was not reduced at potentials less negative than required for solvent (RTIL) decomposition while only a poorly defined wave was seen for cytosine. The possibility for proton abstraction from the cation in [N-6,N-2,N-2,N-2],[NTF2] and [P-14,P-6,P-6.6][FAP] is noted and this is thought to aid the electrochemical dimerization process. The resulting rapid reaction is thought to shift the reduction potentials for guanine and adenine to lower values than observed in RTILs where the scope for proton abstraction is not present. Such shifts are characteristic of so-called EC processes where reversible electron transfer is followed by a chemical reaction. (C) 2009 Elsevier B.V. All rights reserved.

The quenching of the electronically-excited, lumophoric state of [Ru(bpy)(3)(2+)(Ph4B-)(2)] by oxygen is studied in a wide variety of neat plasticizers. The Stern-Volmer constant, K-SV, is found to be inversely dependent upon the viscosity of the quenching medium, although the natural lifetime of the electronically excited state of [RU(bPY)(3)(2+)(Ph4B-)(2)] is largely independent of medium. The least viscous of the plasticizers tested, triethyl phosphate, did not, however, produce highly sensitive optical oxygen sensors when used to plasticize [RU(bPY)(3)(2+)(Ph4B-)(2)]-containing cellulose acetate butyrate (CAB) and poly(methyl methacrylate) (PMMA) films, Instead, the compatibility of the polymer-plasticizer combination, as measured by the difference in the values of the solubility parameter of the two, appears to be a major factor in determining the overall oxygen sensitivity of the thin plastic films. For highly compatible polymer-plasticizer combinations, the plasticizer with the lowest viscosity produces films of the highest oxygen sensitivity. This situation arises because in the film the quenching process is partly diffusion-controlled and, as a result, the quenching rate constant is inversely proportional to the effective viscosity of the reaction medium.

Ultrasound promotes the reduction of hexacyanoferrate(III) by thiosulfate ions mediated by RuO2 . xH(2)O under diffusion-controlled conditions. There is a strong correlation between the measured first-order rate constant and the absorbance of the dispersion, which, in turn, is closely related to the specific surface area of the catalyst. The enhancement in rate with ultrasonic irradiation appears to be largely associated with the dispersive action of the ultrasound on the aggregated particles of RuO2 . xH(2)O. The rate of reaction increases with increasing %duty cycle and ultrasonic intensity. The measured overall activation energies for the reaction with and without ultrasound, i.e. 18 +/- 1 and 20 +/- 1 kJ mol(-1), respectively, are very similar to those expected for a diffusion-controlled reaction. The homogeneous reaction is not promoted by ultrasound.

The kinetics of the recovery of the photoinduced transient bleaching of colloidal CdS in the presence of different electron acceptors are examined. In the presence of the zwitterionic viologen, N,N'-dipropyl-2,2'-bipyridinium disulphonate, excitation of colloidal CdS at different flash intensities generates a series of decay profiles which are superimposed when normalized. The shape of the decay curves are as predicted by a first-order activation-controlled model for a log-normal distribution of particles sizes. In contrast, the variation in flash intensity in the presence of a second viologen, N,N'-dipropyl-4,4'-bipyridinium sulphonate, generates normalized decay traces which broaden with increasing flash intensity. This behaviour is predicted by a zero-order diffusion-controlled model for a log-normal distribution of particle radii. The photoreduction of a number of other oxidants sensitized by colloidal CdS is examined and the shape of the decay kinetics interpreted via either the first- or zero-order kinetics models. The rate constants and activation energies derived using these models are consistent with the values expected for an activation- or diffusion-controlled reaction.

The results of a kinetic study of the oxidation of water to oxygen by Ce(IV) ions in different acid media, mediated by anhydrous ruthenium(IV) oxide are described. In an acid medium which is predominantly HClO4 the kinetics are diffusion controlled and first order with respect to both [Ce(IV)] and [RuO2] and exhibit an activation energy of 19 kJ mol-1. In 0.5 mol dm-3 H2SO4 the kinetics are much slower and complex, the rate decreasing with increasing [Ce(III)]. The kinetics of catalysis observed in all the different acid media studied are readily interpreted using an electrochemical model in which the catalyst particles are considered as acting as microelectrodes which mediate electron transfer between a Nernstian reduction reaction (Ce(IV) --> Ce(III)) and an irreversible oxidation reaction (H2O --> 2H+ + 1/2O2). This electrochemical model is used to analyse the complex kinetics observed in 0.5 mol dm-3 H2SO4 and extract mechanistic information concerning the nature of the rate determining step.

A number of different, characterised, supported and unsupported oxides of Ru(IV) and Ir(IV) have been tested for activity as a chlorine catalyst in the oxidation of brine by Ce(IV) ions. All the different materials tested gave yields of chlorine of > 90% and first-order kinetics for the reduction of the Ce(IV) ions. The samples prepared by the Adams method were the most active of the materials tested and are typified by high surface areas and appreciable activities per unit area. The kinetics of the catalysed reduction of Ce(IV) ions by brine were studied in detail using an Ru(IV) oxide prepared by the Adams method and supported on TiO2 and the results were rationalised in terms of an electrochemical model in which the rate-determining step is the diffusion-controlled reduction of Ce(IV) ions. In support of this model the measured activation energies for the oxidation of brine by Ce(IV) ions, catalysed by either a supported or unsupported Adams catalyst, were both close (18-21 kJ mol-1) to that expected for a diffusion-controlled reaction (ca. 15 kJ mol-1).

Thermoresponsive polymeric platforms are used to optimise drug delivery in pharmaceutical systems and bioactive medical devices. However, the practical application of these systems is compromised by their poor mechanical properties. This study describes the design of thermoresponsive semi-interpenetrating polymer networks (s-IPNs) based on cross-linked p(NIPAA) or p(NIPAA-co-HEMA) hydrogels containing poly(e-caprolactone) designed to address this issue. Using DSC, the lower critical solution temperature of the co-polymer and p(NIPAA) matrices were circa 34 °C and 32 °C, respectively. PCL was physically dispersed within the hydrogel matrices as confirmed using confocal scanning laser microscopy and DSC and resulted in marked changes in the mechanical properties (ultimate tensile strength, Young's modulus) without adversely compromising the elongation properties. P(NIPAA) networks containing dispersed PCL exhibited thermoresponsive swelling properties following immersion in buffer (pH 7), with the equilibrium-swelling ratio being greater at 20 °C than 37 °C and greatest for p(NIPAA)/PCL systems at 20 °C. The incorporation of PCL significantly lowered the equilibrium swelling ratio of the various networks but this was not deemed practically significant for s-IPNs based on p(NIPAA). Thermoresponsive release of metronidazole was observed from s-IPN composed of p(NIPAA)/PCL at 37 °C but not from p(NIPAA-co-HEMA)/PCL at this temperature. In all other platforms, drug release at 20 °C was significantly similar to that at 37 °C and was diffusion controlled. This study has uniquely described a strategy by which thermoresponsive drug release may be performed from polymeric platforms with highly elastic properties. It is proposed that these materials may be used clinically as bioactive endotracheal tubes, designed to offer enhanced resistance to ventilator associated pneumonia, a clinical condition associated with the use of endotracheal tubes where stimulus responsive drug release from biomaterials of significant mechanical properties would be advantageous. © 2012 Elsevier B.V. The role of sodium surface species in the modification of a platinum (Pt) catalyst film supported on 8 mol% yttria-stabilised-zirconia (YSZ) was investigated under a flow of 20 kPa oxygen at 400 °C. Cyclic and linear sweep voltammetry were used to investigate the kinetics of the oxygen charge transfer reaction. The Pt/YSZ systems of both ‘clean’ and variable-coverage sodium-modified catalyst surfaces were also characterised using SEM, XPS and work function measurements using the Kelvin probe technique.

Samples with sodium coverage from 0.5 to 100% were used. It was found that sodium addition modifies the binding energy of oxygen onto the catalyst surface. Cyclic voltammetry experiments showed that higher overpotentials were required for oxygen reduction with increasing sodium coverage. In addition, sodium was found to modify oxygen storage and/or adsorption and diffusion increasing current densities at higher cathodic overpotential. Ex situ XPS measurements showed the presence of sodium hydroxide, carbonate and/or oxide species on the catalyst surface, while the Kelvin probe technique showed a decrease of approximately 250 meV in the work function of samples with more than 50% sodium coverage (compared to a nominally ‘clean’ sample).

Cervical cancer is the third most prevalent cancer in women and disproportionately affects those in low resource settings due to limited programs for screening and prevention. In the developed world treatment for the disease in the non-metastasised state usually takes the form of surgical intervention and/or radiotherapy. In the developing world such techniques are less widely available. This paper describes the development of an intravaginal ring for the localised delivery of a chemotherapeutic drug to the cervix that has the potential to reduce the need for surgical intervention and will also provide a novel anti-cancer therapy for women in low resource settings. Disulfiram has demonstrated antineoplastic action against prostate, breast and lung cancer. Both PEVA and silicone elastomer were investigated for suitability as materials in the manufacture of DSF eluting intravaginal rings. DSF inhibited the curing process of the silicone elastomer, therefore PEVA was chosen as the material to manufacture the DSF-loaded vaginal rings. The vaginal rings had an excellent content uniformity while the DSF remained stable throughout the manufacturing process. Furthermore, the rings provided diffusion controlled release of DSF at levels well in excess of the IC50 value for the HeLa cervical cancer cell line.

This study describes the preclinical development of a matrix-type silicone elastomer vaginal ring device designed to provide controlled release of UC781, a non-nucleoside re- verse transcriptase inhibitor. Testing of both human- and macaque-sized rings in a sink condition in vitro release model demonstrated continuous UC781 release in quantities consid- ered sufficient to maintain vaginal fluid concentrations at levels 82–860-fold higher than the in vitro IC50 (2.0 to 10.4 nM) and therefore potentially protect against mucosal trans- mission of HIV. The 100-mg UC781 rings were well tolerated in pig-tailed macaques, did not induce local inflammation as determined by cytokine analysis and maintained median con- centrations in vaginal fluids of UC781 in the range of 0.27 to 5.18 mM during the course of the 28-day study. Analysis of residual UC781 content in rings after completion of both the in vitro release and macaque pharmacokinetic studies revealed that 57 and 5 mg of UC781 was released, respectively. The pharmacokinetic analysis of a 100-mg UC781 vaginal ring in pig-tailed macaques showed poor in vivo–in vitro correlation, attributed to the very poor solubility of UC781 in vaginal fluid and resulting in a dissolution-controlled drug release mecha- nism rather than the expected diffusion-controlled mechanism.

The redox catalyst ruthenium dioxide, prepared via the Adams technique, i.e.Ru(Adams), is used as a water oxidation catalyst using the oxidants (i) Ce(IV) in 0.5M H2SO4 and (ii) periodate in 0.5 M H2SO4, water and 0.1 M KOH. Like Ce(IV),periodate is a very strong oxidant that is able to oxidise water to oxygen and can bereadily monitored spectrophotometrically at 280 nm, compared with 430 nm for Ce(IV).More importantly, unlike Ce(IV), which is unstable towards hydrolysis above pH 1,periodate is stable in acid, water and strong alkali. A spectrophotometric study of thekinetics of periodate reduction, and concomitant oxidation of water to O2, reveals thatin the presence of a suitable redox catalyst, Ru(Adams) in this work, periodate is ableto effect the stoichiometric oxidation of water, with a turnover number > 64. In justwater, the kinetics of the latter reaction appear diffusion-controlled, due to the largethermodynamic driving force, a measure of which is the difference in redox potential,i.e. ∆E = 423 mV. As this difference is decreased, ∆E = 396 mV in acid and 290 mVin strong alkali (0.1 M KOH), the kinetics become increasingly activation-controlledand slower. These findings are discussed briefly with regard to the possible use of (i)periodate as an alternative oxidant in the rapid screening of new potential wateroxidation catalyst material powders that are stable only under near neutral and/oralkaline conditions, and (ii) Ru(Adams) as a benchmark catalyst.

O objectivo deste trabalho é a produção de novos eléctrodos modificados com polioxotungstatos (POMs) do tipo Keggin, incluindo POMs lacunares e substituídos por metais de transição. A preparação e caracterização dos polioxotungstatos encontram-se descritas no Capítulo 2. No Capítulo 3 descreve-se a produção de eléctrodos de carbono vítreo funcionalizados com sais híbridos de tetra-n-butilamónio de vários silicotungstatos pelo método de evaporação da gota. As propriedades electroquímicas dos polioxotungstatos imobilizados foram comparadas com as das espécies solúveis correspondentes. A morfologia dos depósitos foi avaliada por microscopia óptica e por microscopia electrónica de varrimento. No capítulo 4 descreve-se a preparação de novos eléctrodos compósitos de carbono e poli(hexilmetacrilato) com fosfotungstatos. Os estudos electroquímicos revelaram que as principais características dos POMs são mantidas e que os processos de redução são controlados por difusão, dependendo da difusão dos protões da solução. O Capítulo 5 descreve a construção de filmes em multicamadas ultrafinos contendo POMs e polietilenimina, preparados pelo método de auto-montagem camada-sobre-camada em eléctrodos de carbono vítreo. Os filmes em multicamada foram caracterizados por voltametria cíclica e por microscopia electrónica de varrimento e foi usada a espectroscopia de absorção de UV-Vis em placas de quartzo para monitorar o crescimento de filme. Os resultados voltamétricos revelaram que os processos de redução dos POM são confinados à superfície. Alguns destes eléctrodos modificados revelaram propriedades electrocatalíticas relativamente à redução dos aniões nitrito, bromato e/ou iodato. A espectroscopia de impedância electroquímica também foi usada na caracterização destes filmes e os resultados revelaram que a resistência à transferência de carga aumenta com o aumento do número de bicamadas para ambas as espécies redox, indicando que a espessura do filme tem um efeito importante sobre a cinética de reacções de transferência de carga. No capítulo 6 descreve-se a síntese de filmes híbridos orgânicos/inorgânicos compostos por poli(3,4-etilenodioxitiofeno) (PEDOT) e por silicotungstatos do tipo Keggin através da polimerização electroquímica, em condições aquosas, na superfície de electrodos de carbono vítreo. A voltametria cíclica revelou que as características principais dos POMs são mantidas nos filmes. Verificou-se que estes filmes são muito estáveis, possivelmente devido a fortes interacções electrostáticas entre os POMs aniónicos e o polímero positivamente carregado. A espectroscopia de impedância electroquímica foi também utilizada e os resultados mostraram que a resistência de transferência de carga aumenta com o aumento do pH e para valores de potenciais mais elevados. O capítulo 7 apresenta as conclusões finais e possíveis trabalhos futuros.

We investigate the dynamic nature of metal speciation in colloidal dispersions using a recently proposed theory [J.P. Pinheiro, M. Minor, H.P. Van Leeuwen, Langmuir, 21 (2005) 8635] for complexing ligands that are situated on the surface of the particles. The new approach effectively modifies the finite rates of association/dissociation of the colloidal metal complexes, thus invoking consideration of the two basic dynamic criteria: the association/dissociation kinetics of the volume complexation reaction (the ‘‘dynamic’’ criterion), and the interfacial flux of free metal to a macroscopic surface due to dissociation of complex species (the ‘‘lability’’ criterion). We demonstrate that the conventional approach for homogeneous systems that assume a smeared-out ligand distribution, overestimates both the dynamics and the lability of metal complexes when applied to colloidal ligands. It is also shown that the increase of lability with increasing particle radius, as expected for a homogeneous solution, is moderated for spherical microelectrodes and practically eliminated for planar electrodes.

The modelling of the experimental data of the extraction of the volatile oil from six aromatic plants (coriander, fennel, savoury, winter savoury, cotton lavender and thyme) was performed using five mathematical models, based on differential mass balances. In all cases the extraction was internal diffusion controlled and the internal mass transfer coefficienty (k(s)) have been found to change with pressure, temperature and particle size. For fennel, savoury and cotton lavender, the external mass transfer and the equilibrium phase also influenced the second extraction period, since k(s) changed with the tested flow rates. In general, the axial dispersion coefficient could be neglected for the conditions studied, since Peclet numbers were high. On the other hand, the solute-matrix interaction had to be considered in order to ensure a satisfactory description of the experimental data.
