Introduction The aim of this study was to conduct an epidemiological study comparing the genetic similarity of yeasts isolated from blood cultures. Methods Random amplification of polymorphic DNA (RAPD) techniques were used for the Candida samples obtained from patients at the Hospital Universitário da Universidade Federal do Mato Grosso do Sul (HU/UFMS) in Campo Grande, state of Mato Grosso do Sul, Brazil, from 1998-2000. Results The most frequently isolated species was Candida albicans (45.8%). DNA amplification from genomic yeast isolates indicated a genetic similarity of over 90%. Conclusions The RAPD profiles obtained were able to differentiate between the isolated Candida species, thereby suggesting that the method might be useful in epidemiological studies.

Lithiasic cholecystitis is classically associated with the presence of enterobacteria, such as Escherichia coli, Enterococcus, Klebsiella, and Enterobacter, in the gallbladder. Cholecystitis associated with fungal infections is a rare event related to underlying conditions such as diabetes mellitus, steroid use, and broad-spectrum antibiotic use for prolonged periods, as well as pancreatitis and surgery of the digestive tract. Here, we present the first reported case of a gallbladder infection caused by Candida famata.

Introduction The aim of this study was to investigate the effects of Rosmarinus officinalis essential oil on germ tube formation by Candida albicans isolated from denture wearers. Methods Ten C. albicans isolates recovered from denture wearers were tested using 10% fetal bovine serum with or without 4% R. officinalis essential oil. Results The essential oil from R. officinalis completely inhibited germ tube formation in the investigated C. albicans isolates. Conclusions The results demonstrate that the essential oil of R. officinalis modulates C. albicans pathogenicity through its primary virulence factor (i.e., germ tube formation was suppressed).

AbstractINTRODUCTION:Candida parapsilosis is a common yeast species found in cases of onychomycosis and candidemia associated with infected intravascular devices. In this study, we differentiated Candida parapsilosis sensu stricto, Candida orthopsilosis , and Candida metapsilosis from a culture collection containing blood and subungual scraping samples. Furthermore, we assessed the in vitro antifungal susceptibility of these species to fluconazole, itraconazole, voriconazole, posaconazole, amphotericin B, and caspofungin.METHODS:Differentiation of C. parapsilosis complex species was performed by amplification of the secondary alcohol dehydrogenase (SADH) gene and digestion by the restriction enzyme Ban I. All isolates were evaluated for the determination of minimal inhibitory concentrations using Etest, a method for antifungal susceptibility testing.RESULTS:Among the 87 isolates, 78 (89.7%) were identified as C. parapsilosis sensu stricto , five (5.7%) were identified as C. orthopsilosis , and four (4.6%) were identified as C. metapsilosis . Analysis of antifungal susceptibility showed that C. parapsilosis sensu strictoisolates were less susceptible to amphotericin B and itraconazole. One C. parapsilosis sensu stricto isolate was resistant to amphotericin B and itraconazole. Moreover, 10.2% of C. parapsilosis sensu stricto isolates were resistant to caspofungin. Two C. parapsilosis sensu strictoisolates and one C. metapsilosis isolate were susceptible to fluconazole in a dose-dependent manner.CONCLUSIONS:We reported the first molecular identification of C. parapsilosiscomplex species in State of Goiás, Brazil. Additionally, we showed that although the three species exhibited differences in antifungal susceptibility profiles, the primary susceptibility of this species was to caspofungin.

As leveduras da espécie Candida albicans são comensais do ser humano, podendo em certos casos, como a toma prolongada de antibióticos de largo espectro, a diminuição da imunidade, ou a quimioterapia, causar infecções severas. Estas infecções classificam-se em superficiais ou sistémicas, consoante a zona anatómica ou os órgãos afectados. As infecções hospitalares por fungos, nomeadamente as causadas por C. albicans, têm vindo a aumentar nos últimos anos, assim como a mortalidade e a morbilidade associadas, e ainda os custos relacionados com os cuidados de saúde. Torna-se por isso importante a implementação de terapêutica de uma forma rápida, eficaz e correcta. Assim, é imperativo que os laboratórios de microbiologia clínica possuam métodos de diagnóstico rápidos, simples e económicos em relação a uma correcta identificação ao nível de espécie e em relação ao estudo da sensibilidade aos antifúngicos. Contudo, esta identificação ao nível de espécie não permite averiguar a origem das infecções, facto importante para a compreensão da evolução das infecções nosocomiais. Para tal, é necessário realizar um estudo ao nível de estirpe através de métodos moleculares que permitam fazer a distinção acurada entre isolados de C. albicans. Os métodos moleculares têm vindo a desenvolver um papel importante no diagnóstico de infecções: são rápidos, sensíveis, precisos e possuem a vantagem de se poder trabalhar pequenas quantidades de amostra. Têm no entanto a desvantagem de ser métodos caros, por vezes demasiado elaborados para serem instituídos num laboratório de microbiologia clínica com grande volume de amostras diárias. O principal objectivo deste trabalho consistiu na diferenciação de estirpes de C. albicans através de uma metodologia molecular inovadora, nunca antes efectuada no ocidente, simples e rápida, por meio de simples amplificações por PCR. Para tal, foram seleccionados 60 isolados clínicos de leveduras obtidas a partir de amostras clínicas de três localidades: Covilhã (Centro Hospitalar Cova de Beira, E.P.E) Lisboa (Instituto Português de Oncologia e Hospital de Santa Maria, E.P.E) e Viseu (Hospital de São Teotónio). O trabalho baseou-se na genotipagem de isolados clínicos de C. albicans por amplificação por PCR com primers dirigidos às sequências 25S rDNA e às regiões ALT das sequências RPS. Foi possível a sua diferenciação e distinção em estirpes, fazendo deste método um bom recurso para estudos epidemiológicos. Realizou-se ainda um estudo de sensibilidade in vitro das estirpes de C. albicans ao antifúngicos Fluconazol e Voriconazol pelo método de difusão em disco, segundo os procedimentos padronizados e publicados pelo CLSI. Através deste estudo foi verificada elevada susceptibilidade aos antifúngicos estudados. Com este trabalho conclui-se que a diferenciação ao nível de estirpe é muito importante para compreender padrões de surtos de infecções e ainda para averiguar a origem, endógena ou exógena, destas infecções nosocomiais. Como tal, este estudo epidemiológico torna-se essencial para definir um controlo mais rigoroso das infecções.

Estudou-se o comportamento de leveduras do gênero Candida a antifungicos, pela determinação da Concentração Inibitória Mínima (CIM) e da Concentração Fungicida Mínima (CFM) "in vitro"de 30 cepas de Candida frente aos antifungicos: miconazol, cetoconazol e anfotericina B. Empregou-se o método de diluição em meio líquido e os antifungicos foram diluídos visando proporcionar concentrações a partir de 0,06 a 128 μg/mL. O inóculo foi padronizado ajustando-se a suspensão para conter 1X 106 ufc/mL. A concentração para a qual houve maior convergência de cepas foi de 1 μg/mL (26,5%) para anfotericina B. Para miconazol foi de 16 μg/mL (26,0%) e para cetoconazol 32 μg/mL (23,0%). Os valores de CFM foram de 2 μg/mL (23,5%) para anfotericina B, de 16 e 64 μg/mL (26,5%) para miconazol e 32 e 64 μg/mL (30,0%) para cetoconazol. Este imidazólico mostrou os valores de CIM e CFM mais elevados atingindo até 128 μg/mL em algumas espécies. As espécies de Candida mostraram-se mais sensíveis à Anfotericina B, quando comparadas em relação aos antifungicos testados. Sobre o desempenho das espécies de Candida, melhor padrão de comportamento foi verificado para C. albicans com níveis mais baixos de sensibilidade.

Candida bracarensis is an uncommon Candida species found during an epidemiological study of candidiasis performed in Braga, Portugal. Initially, it was identified as C. glabrata, but recently detailed analyses pointed out their differences. So, little information is still available about C. bracarensis virulence factors and antifungal susceptibilities. Therefore, the main goal of this work is to evaluate the ability of C. bracarensis to form biofilms, to produce hydrolytic enzymes (proteases, phospholipases and hemolysins), as well as its susceptibility to amphotericin B and fluconazole. It was shown, for the first time, that all C. bracarensis strains were able to form biofilms and display proteinase and hemolytic activities. Moreover, although planktonic cells presented antifungal susceptibility, amphotericin B and fluconazole were unable to inhibit biofilm formation and eradicate pre-formed biofilms. Due to the propensity of C. bracarensis to display antifungal resistance and virulence attributes, the control of these emerging pathogens is recommended.

Candida parapsilosis is nowadays an emerging opportunistic pathogen and its increasing incidence is part related to the capacity to produce biofilm. In addition, one of the most important C. parapsilosis pathogenic risk factors includes the organisms\textquoteright selective growth capabilities in hyper alimentation solutions. Thus, in this study, we investigated the role of glucose in C. parapsilosis biofilm modulation, by studying biofilm formation, matrix composition and structure. Moreover, the expression of biofilm-related genes (BCR1, FKS1 and OLE1) were analyzed in the presence of different glucose percentages. The results demonstrated the importance of glucose in the modulation of C. parapsilosis biofilm. The concentration of glucose had direct implications on the C. parapsilosis transition of yeast cells to pseudohyphae. Additionally, it was demonstrated that biofilm related genes BCR1, FKS1 and OLE1 are involved in biofilm modulation by glucose. The mechanism by which glucose enhances biofilm formation is not fully understood, however with this study we were able to demonstrate that C. parapsilosis respond to stress conditions caused by elevated levels of glucose by up-regulating genes related to biofilm formation (BCR1, FKS1 and OLE1).

The severity and frequency of opportunistic fungal infections still growing, concomitantly to the increasing rates of antimicrobial drugs resistance. Natural matrices have been used over years due to its multitude of health benefits, including antifungal potential. Thus, the present work aims to evaluate the anti-Candida potential of the phenolic extract and individual phenolic compounds of Glycyrrhiza glabra L. (licorice), by disc diffusion assay, followed by determination of the minimal inhibitory concentration (MIC) and minimal fungicidal concentration (MFC) for both planktonic cells and biofilms. Licorice extract evidenced inhibitory potential against the nineteen tested Candida strains, but no pronounced effect was observed by testing the most abundant individual phenolic compounds. Candida tropicalis strains were the most sensible, followed by Candida glabrata, Candida parapsilosis and, then, Candida albicans. Lower MIC and MFC values were achieved to C. glabrata and C. tropicalis, which confirms its susceptibility to licorice extract; however, for C. tropicalis strains a higher variability was observed. Anti-biofilm potential was also achieved, being most evident in some C. glabrata and C. tropicalis strains. In general, a twice concentration of the MIC was necessary for planktonic cells to obtain a similar potential to that one observed for biofilms. Thus, an upcoming approach for new antifungal agents, more effective and safer than the current ones, is stablished; notwithstanding, further studies are necessary in order to understand its mechanism of action, as also to assess kinetic parameters.

Objectives: This research work intends to clarify the role of artificial saliva, in particularly the role of mucin, a salivary protein, on the surface properties and adhesion ability of Candida spp. oral clinical isolates to abiotic surfaces. Methods: Four oral clinical isolates of Candida spp. were used: two Candida albicans strains (AC; AM) and two Candida parapsilosis strains (AD; AM2). The strains were isolated from patients using oral prosthesis. The microorganisms were cultured in the absence or presence of mucin and artificial saliva, and their adhesion to an abiotic surface (coated with mucin and artificial saliva) was evaluated. Results: The presence of mucin per se onto the abiotic surface decreased the adhesion of all strains, although the combination of mucin with artificial saliva had reduced this effect. No direct correlation between adhesion and the surface free energies of adhesion of the microorganisms was found. Significance: Candida spp. were human commensal microorganisms that became pathogenic when the host immune defenses were compromised. Medical devices were colonized by Candida spp. particularly, oral prostheses, which might lead to the degradation of the prostheses and systemic infections. The salivary secretions that constantly cover the oral cavity influenced Candida spp. adhesion process. Therefore, it was important to understand the interactions between Candida spp., salivary proteins and the characteristic of oral prosthesis when developing materials for oral prostheses.
