First published November 10, 2010; doi:10.1152/ajpregu.00169.2010.-Endocrine pancreas from pregnant rats undergoes several adaptations that comprise increase in beta-cell number, mass and insulin secretion, and reduction of apoptosis. Lactogens are the main hormones that account for these changes. Maternal pancreas, however, returns to a nonpregnant state just after the delivery. The precise mechanism by which this reversal occurs is not settled but, in spite of high lactogen levels, a transient increase in apoptosis was already reported as early as the 3rd day of lactation (L3). Our results revealed that maternal islets displayed a transient increase in DNA fragmentation at L3, in parallel with decreased RAC-alpha serine/threonine-protein kinase (AKT) phosphorylation (pAKT), a known prosurvival kinase. Wortmannin completely abolished the prosurvival action of prolactin (PRL) in cultured islets. Decreased pAKT in L3-islets correlated with increased Tribble 3 (TRB3) expression, a pseudokinase inhibitor of AKT. PERK and eIF2 alpha phosphorylation transiently increased in islets from rats at the first day after delivery, followed by an increase in immunoglobulin heavy chain-binding protein (BiP), activating transcription factor 4 (ATF4), and C/EBP homologous protein (CHOP) in islets from L3 rats. Chromatin immunoprecipitation (ChIP) and Re-ChIP experiments further confirmed increased binding of the heterodimer ATF4/CHOP to the TRB3 promoter in L3 islets. Treatment with PBA, a chemical chaperone that inhibits UPR, restored pAKT levels and inhibited the increase in apoptosis found in L3. Moreover, PBA reduced CHOP and TRB3 levels in beta-cell from L3 rats. Altogether, our study collects compelling evidence that UPR underlies the physiological and transient increase in beta-cell apoptosis after delivery. The UPR is likely to counteract prosurvival actions of PRL by reducing pAKT through ATF4/CHOP-induced TRB3 expression.

Aim: To investigate the effects of swimming training on the renin-angiotensin system (RAS) during the development of hypertensive disease. Main methods: Male spontaneously hypertensive rats (SHR) were randomized into: sedentary young (SY), trained young (TV), sedentary adult (SA), and trained adult (TA) groups. Swimming was performed 5 times/wk/8wks. Key findings: Trained young and adult rats showed both decreased systolic and mean blood pressure, and bradycardia after the training protocol. The left ventricular hypertrophy (LVH) was observed only in the TA group (12.7%), but there was no increase on the collagen volume fraction. Regarding the components of the RAS, TV showed lower activity and gene expression of angiotensinogen (AGT) compared to SY. The TA group showed lower activity of circulatory RAS components, such as decreased serum ACE activity and plasma renin activity compared to SA. However, depending on the age, although there were marked differences in the modulation of the RAS by training, both trained groups showed a reduction in circulating angiotensin II levels which may explain the lower blood pressure in both groups after swimming training. Significance: Swimming training regulates the RAS differently in adult and young SHR rats. Decreased local cardiac RAS may have prevented the LVH exercise-induced in the TV group. Both groups decreased serum angiotensin II content, which may, at least in part, contribute to the lowering blood pressure effect of exercise training. (C) 2011 Elsevier Inc. All rights reserved.

The Fiji government’s 2008 'budget for hope' rests on some fundamental assumptions: political stability, the recovery of key sectors of the economy from the effects of the 2006 coup, as well as the recovery of the tourism industry, an industry which in the past has been resilient to coups. With 34 per cent of the population already living below the poverty line, low economic growth could push significantly more people into poverty. If the increases in global food and fuel prices persist, creating even more difficulties for the poor, the prospect of social instability could heighten. In the absence of the expected recovery of these key sectors and the loss of skilled workers, achieving economic and social recovery will require political consensus to resolve the political impasse that has gripped the country. The Interim Government also needs to revisit its fiscal austerity package.

The swimming backward for biomimetic carangiform robot fish is analyzed and implemented in this paper. The swimming law of the carangiform robot fish is modified according to the European Eel swimming mode based on the multiple-link structure to implement the backward motion. The motion mode difference between the eel and carangiform fish is discussed, and a qualitative kinematic analysis of the carangiform swimming in water is given to analyze the propulsion produced by the undulation of the multi-links tail. The experiments conducted demonstrate the good performance of the proposed method, and the results are given.

Spermatogenesis in the blue swimming crab, Portunus pelagicus, is described by light and electron microscopy. The testis is composed of anterior (AT) and posterior (PT) lobes, that are partitioned into lobules by connective tissue trabecula, and further divided into zones (germinal, transformation and evacuation), each with various stages of cellular differentiation. The vas deferens is classified into three distinct regions: anterior (AVD), median (MVD), and posterior (PVD), on the presence of spermatophores and two secretions, termed substance I and II. Based on the degree and patterns of heterochromatin, spermatogenesis is classified into 13 stages: two spermatogonia (SgA and SgB), six primary spermatocytes (leptotene, zygotene, pachytene, diplotene, diakinesis, and metaphase), a secondary spermatocyte (SSc), three spermatids (St 1–3), and a mature spermatozoon. Spermatid stages are differentiated by chromatin decondensation and the formation of an acrosomal complex, which is unique to brachyurans. Mature spermatozoa are aflagellated, and have a nuclear projection and a spherical acrosome. AUT-PAGE and Western blots show that, during chromatin decondensation, there is a reduction of most histones, with only small amounts of H2B and H3 remaining in mature spermatozoa.

Significant world events often cause the behavioral convergence of the expression of shared sentiment. This paper examines the use of the blogosphere as a framework to study user psychological behaviors, using their sentiment responses as a form of ‘sensor’ to infer real-world events of importance automatically. We formulate a novel temporal sentiment index function using quantitative measure of the valence value of bearing words in blog posts in which the set of affective bearing words is inspired from psychological research in emotion structure. The annual local minimum and maximum of the proposed sentiment signal function are utilized to extract significant events of the year and corresponding blog posts are further analyzed using topic modeling tools to understand their content. The paper then examines the correlation of topics discovered in relation to world news events reported by the mainstream news service provider, Cable News Network, and by using the Google search engine. Next, aiming at understanding sentiment at a finer granularity over time, we propose a stochastic burst detection model, extended from the work of Kleinberg, to work incrementally with stream data. The proposed model is then used to extract sentimental bursts occurring within a specific mood label (for example, a burst of observing ‘shocked’). The blog posts at those time indices are analyzed to extract topics, and these are compared to real-world news events. Our comprehensive set of experiments conducted on a large-scale set of 12 million posts from Livejournal shows that the proposed sentiment index function coincides well with significant world events while bursts in sentiment allow us to locate finer-grain external world events.

Although long-distance transport of marine organisms is constrained by numerous oceanic and biological factors, some species have evolved life-histories reliant on such movements. We examine the factors that promote long-distance transport in a transoceanic migrant, young loggerhead sea turtles (Caretta caretta), from the southeastern U.S. Empirical data from near-surface buoys and simulations in two ocean circulation models indicated that passive drifters are often retained for long periods shoreward of oceanic fronts that delineate coastal and offshore waters. Further simulations revealed that offshore swimming aided newly hatched turtles in moving past fronts and increased turtles’ probability of survival, reaching distant foraging grounds, and encountering favorable temperatures. Swimming was most beneficial in regions that were more favorable under scenarios assuming passive drift. These results have broad implications for understanding the movement processes of many marine species, highlighting likely retention of more planktonic species and potential for dispersal in more nektonic species.
