Schistosomiasis affects more than 200 million people worldwide; another 600 million are at risk of infection. The schistosomulum stage is believed to be the target of protective immunity in the attenuated cercaria vaccine model. In an attempt to identify genes up-regulated in the schistosomulum stage in relation to cercaria, we explored the Schistosoma mansoni transcriptome by looking at the relative frequency of reads in EST libraries from both stages. The 400 genes potentially up-regulated in schistosomula were analyzed as to their Gene Ontology categorization, and we have focused on those encoding-predicted proteins with no similarity to proteins of other organisms, assuming they could be parasite-specific proteins important for survival in the host. Up-regulation in schistosomulum relative to cercaria was validated with real-time reverse transcription polymerase chain reaction (RT-PCR) for five out of nine selected genes (56%). We tested their protective potential in mice through immunization with DNA vaccines followed by a parasite challenge. Worm burden reductions of 16-17% were observed for one of them, indicating its protective potential. Our results demonstrate the value and caveats of using stage-associated frequency of ESTs as an indication of differential expression coupled to DNA vaccine screening in the identification of novel proteins to be further investigated as potential vaccine candidates.

Flavonóides são compostos fenólicos de ampla ocorrência na natureza exercendo diversas funções fisiológicas nos vegetais. Na família Leguminosae estes metabólitos secundários exercem o papel de antimicrobianos (fitoalexina e/ou fitoanticipinas) e como moléculas sinalizadoras, na simbiose com bactérias do solo no processo de fixação biológica do Nitrogênio. O gênero Lupinus, pertencente a esta família, é encontrado em todas as regiões fisiográficas do estado do Rio Grande do Sul. Entretanto, não existem relatos de estudos químicos ou microbiológicos. Neste trabalho, foi analisado o perfil químico de folhas, flores, raízes, nódulos e legumes da espécie Lupinus lanatus. Foram isolados oito compostos fenólicos, dois deles não foram identificados devido ao baixo rendimento, um derivado do ácido cafeico isolado a partir das raízes teve sua estrutura parcialmente identificada. Quatro flavonóides tiveram suas estruturas elucidadas por métodos espectroscópicos e espectrométricos; três flavonas-C-glicosiladas: citisosídeo a partir das flores e folhas, ramnosil-O-vitexina e ramnosil-O-citisosídeo, a partir das folhas; e a isoflavona angustona A, a partir dos nódulos. Foi analisada a atividade antimicrobiana das flavonas pelo método da bioautografia frente a Bradyrhizobium sp. e Agrobacterium sp., microrganismos simbionte e patógeno, respectivamente, sendo que nenhum metabólito obteve resultado positivo, entretanto quando analisados pelo mesmo método frente a Staphylococcus aureus, Klebsiella pneumoniae e Escherichia coli o resultado foi positivo para ramnosil-O-vitexina. Foi avaliada a atividade antioxidante pelo método da DPPH para as flavonas, onde citisosídeo obteve resultado positivo.

This study aimed to evaluate the effects of salinity in the growth and nitrogen fixation in leucaena. Plants were cultivated in Leonard pots containing clean and sterilized sand. The treatments were distributed in a completely randomized design, in a 3x3 factorial, with five replicates. The first factor corresponded to no inoculation and inoculation with Bradyrhizobium sp SEMIA 6070 and SEMIA 6153 strains. The second factor corresponded to NaCl concentrations: 0, 25 and 50 mol m(-3). Were analyzed the height and dry matter accumulation, the number of nodules and nitrogen in plants. The inoculation of plants with the strain SEMIA 6070 provided higher growth and greater nitrogen accumulation in plants thar were not subject to salinity. Nodulation was not affected by salinity. Inoculation with SEMIA 6153 strain provided greater tolerance to salinity.

O objetivo deste trabalho foi confrontar as sequências parciais do gene 16S rRNA de estirpes padrão de rizóbios com as de estirpes recomendadas para a produção de inoculantes no Brasil, com vistas à verificação da confiabilidade do sequenciamento parcial desse gene para a identificação rápida de estirpes. Foram realizados sequenciamentos através de reação em cadeia da polimerase (PCR) com iniciadores relativos à região codificadora do gene 16S rRNA entre as bactérias estudadas. Os resultados foram analisados pela consulta de similaridade de nucleotídeos aos do Basic Local Alignment Search Tool (Blastn) e por meio da interpretação de árvores filogenéticas geradas usando ferramentas de bioinformática. A classificação taxonômica das estirpes Semia recomendadas para inoculação de leguminosas com base em propriedades morfológicas e especificidade hospedeira não foi confirmada em todas as estirpes. A maioria das estirpes estudadas, consultadas no Blastn, é consistente com a classificação proposta pela construção de árvores filogenéticas das sequências destas estirpes, com base na similaridade pelo sequenciamento parcial do gene considerado.

The use of sewage sludge is a highly promising practice for the development of sustainable agricultural systems. The objective of this study was to evaluate doses of sewage sludge composted with and without Rhizobium inoculation in leaf N content, nodule number, nodule dry weight and plant during flowering. The experiment was conducted in the greenhouse of the Department of Soil Science and Natural Resources College of Agricultural Sciences of Botucatu, using as substrate used in vessels of 30 liters a Red Yelow Latosol sandy texture with experimental design adopted was randomized blocks constituted for 10 treatments and five doses of composted sewage sludge (0, 10, 20, 30, 40 t ha(-1)) with or without inoculation Bradyrhizobium japonic with three replications. There was an increase in the number and dry weight of nodules and shoot dry mass of soybeans due to the increase of the dose of sludge up to a dose of 20 t ha(-1) and after this dose there was a decrease of these parameters. At a dose of 10 t ha(-1) sludge compost inoculated seeds showed higher for foliar concentrations of N and number of nodules compared with uninoculated seeds. At a dose of 30 t ha(-1) inoculated seeds were higher compared to uninoculated in all parameters.

The relationship between soil fungi and actinomycetes, with rhizobia that produce nodulation in Neonotonia wightii (perennial soybean) was studied in culture medium in order to obtain material to be used in mixed inoculations of this legume. A field experiment was designed to provide soil samples and isolate microorganisms belonging to these groups that are capable of interfering in the development of two selected rhizobium strains. Results show that the Bradyrhizobium strains used in the experiment, which are efficient in nodulation of Neonotonia wightii, are highly sensitive to substances produced in the culture medium by actinomycetes and fungi.

The rule creation to clone selection in different projects is a hard task to perform by using traditional implementations to control all the processes of the system. The use of an algebraic language is an alternative approach to manage all of system flow in a flexible way. In order to increase the power of versatility and consistency in defining the rules for optimal clone selection, this paper presents the software OCI 2 in which uses process algebra in the flow behavior of the system. OCI 2, controlled by an algebraic approach was applied in the rules elaboration for clone selection containing unique genes in the partial genome of the bacterium Bradyrhizobium elkanii Semia 587 and in the whole genome of the bacterium Xanthomonas axonopodis pv. citri. The tree leguminous gliricídia (Gliricidia sepium), acácia (Acacia mangium), leucena (Leucaena leucocephala) and sombreiro (Clitoria fairchildiana) are indicated for agroforestry systems, reclamation of degraded lands, reforestation and other purposes in the wet tropic. Despite the importance of legumes the preamazon region it is lacking in information about the symbiotic capacity and diversity of indigenous rhizobia of these legumes. The aim of this work was to evaluate the phenotypic and genetic diversity of rhizobia species nodulating gliricidia sombreiro, leucena and acacia in the Maranhão pre-Amazon region and authenticate isolates of these species in siratro (Macroptilium atropurpureum). For this they were installed two experiments. Sampling was carried out on a alley cropping system, was sampled 20 plants of each species by collecting 10 nodules per plant. It was made isolation, cultural characterization, partial 16S rRNA gene sequencing and analysis of the symbiotic ability of bacterial strains with siratro. The authentication experiment was done in two steps for each legume (gliricidia, acácia, sombreiro and leucena), in the greenhouse and in a completely randomized design with three replications with sterile Hoagland nutrient solution as substrate. Gliricídia, Sombreiro, leucena and acacia are colonized by distinct groups of rhizobia. Gliricidia nodulate preferably with Rhizobium, sombreiro and acacia nodulate preferably with Bradyrhizobium and leucena has Mesorhizobium main symbiote. Endophytic strains of ten genera were found colonizing the gliricidia, sombrero, leucena and acacia nodules and a strain of Arthrobacter sp. had a positive nodulation with siratro. This is the first report on isolation of Methylobacterium sp. in gliricidia nodules and endophytic ability of Terriglobus sp. strains. Indigenous strains of pre-Amazon region of Bradyrhizobium, Mesorhizobium and Rhizobium genus nodulate with siratro, but are ineficiente or had low efficiency to promote their growth.

A presente invenção se refere a um processo biológico de produção de um bipolímero de polissacarídeo com propriedades similares a goma xantana por microorganismos do gênero Rhizobium e Bradyrhizobium, propondo a conformação de uma goma do tipo xantana que seja obtida por meio de microrganismos não patogênicos, ou seja, o microrganismo que é utilizado para a produção da goma trata-se de uma bactéria solo fixadora do nitrogênio atmosférico, portanto, benéfica, não causando danos a plantas de interesse agrícola ou ao meio ambiente, bem como não apresenta a pigmentação amarelada característica das gomas atualmente produzidas, fato este que reduz os custos de recuperação do polímero.
