The social wasp P. paulista is relatively common in southeast Brazil causing many medically important stinging incidents. The seriousness of these incidents is dependent on the amount of venom inoculated by the wasps into the victims, and the characteristic envenomation symptoms are strongly dependent on the types of peptides present in the venom. In order to identify some of these naturally occurring peptides available in very low amounts, an analytical protocol was developed that uses a combination of reversed-phase and normal-phase high-performance liquid chromatography (HPLC) of wasp venom for peptide purification, with matrix-assisted laser desorption/ionization time-of-flight post-source decay mass spectrometry (MALDI-Tof-PSD-MS) and low-energy collision-induced dissociation (CID) in a quadrupole time-of-flight tandem mass spectrometry (QTof-MS/MS) instrument for peptide sequencing at the sub-picomole level. The distinction between Leu and Ile was achieved both by observing d-type fragment ions obtained under CID conditions and by comparison of retention times of the natural peptides and their synthetic counterparts (with different combinations of I and/or L at N- and C-terminal positions). To distinguish the isobaric residues K and Q, acetylation of peptides was followed by Q-Tof-MS analysis. The primary sequences obtained were INWLKLGKMVIDAL-NH2 (MW 1611.98Da) and IDWLKLGKMVMDVL-NH2 (MW 1658.98Da). Micro-scale bioassay protocols characterized both peptides as presenting potent hemolytic action, mast cell degranulation, and chemotaxis of poly-morphonucleated leukocyte (PMNL) cells. The primary sequences and the bioassay results suggest that these toxins constitute members of a new sub-class of mastoparan toxins, directly involved in the occurrence of inflammatory processes after wasp stinging. The reproductive biology of the Brazilian sharpnose shark, Rhizoprionodon lalandii, off southeastern Brazil was investigated using data from gillnet landings. The size-at-maturity for males and females was estimated to be 59 and 62 cm total length (LT), respectively. Ovarian fecundity ranged from 3 to 7 follicles (mean = 4.S4), and uterine fecundity from 1 to S embryos (mean = 3.3). There was a slight positive relationship between female LT and the number of ovarian follicles, but uterine fecundity was not related to female LT. Embryonic growth is fast following fertilization during summer and autumn. Gestation requires 11 - 12 months, and peak parturition is between August and September. A comparison of size-at-maturity between animals from northeastern and southeastern Brazil suggests the existence of at least two stocks of R. lalandii along the Brazilian coast.

Quatro úteros provenientes de quatro fêmeas prenhas e dois indivíduos recém-nascidos de anequim, Isurus oxyrinchus, foram coletados na região Sudeste do Brasil durante os meses de setembro a novembro de 1993 e 1994. Todos os embriões estavam bem desenvolvidos, próximos ao estágio de nascimento, apresentando a dentição e órgãos internos bastante desenvolvidos. O comprimento total desses embriões variou entre 64,5 e 72,0 cm, e o maior número de embriões observado no interior de uma única fêmea foi 20. As observações aqui realizadas confirmam a oofagia como forma de nutrição dos embriões dessa espécie e sua periodicidade. A presença de dentes no estômago dos embriões sugere que a substituição dos dentes se inicia na fase uterina.

The biochemical and functional characterization of wasp venom toxins is an important prerequisite for the development of new tools both for the therapy of the toxic reactions due to envenomation caused by multiple stinging accidents and also for the diagnosis and therapy of allergic reactions caused by this type of venom. PLA(1) was purified from the venom of the neotropical social wasp Polybia paulista by using molecular exclusion and cation exchange chromatographies; its amino acid sequence was determined by using automated Edman degradation and compared to the sequences of other vespid venom PLA(1)'s. The enzyme exists as a 33,961.40 da protein, which was identified as a lipase of the GX class, liprotein lipase superfamily, pancreatic lipases (ab20.3) homologous family and RP2 sub-group of phospholipase. P. paulista PLA(1) is 53-82% identical to the phospholipases from wasp species from Northern Hemisphere. The use restrained-based modeling permitted to describe the 3-D structure of the enzyme, revealing that its molecule presents 23% alpha-helix, 28% beta-sheet and 49% coil. The protein structure has the alpha/beta fold common to many lipases; the core consists of a tightly packed beta-sheet constituted of six-stranded parallel and one anti-parallel beta-strand, surrounded by four alpha-helices. P. paulista PLA(1) exhibits direct hemolytic action against washed red blood cells with activity similar to the Cobra cardiotoxin from Naja naja atra. We investigated sex ratio, reproductive period, fecundity, and sexual dimorphism of Pamphorichthys hollandi in southeastern Brazil. Reproductive status was identified according to a female's gonads/embryos development scale. of the 613 collected specimens, 373 (60 %) were females and 240 (40 %) males giving a sex ratio of I male: 1.6 female. Sexual dimorphism was evident in respect to body size, body coloration, and anal and pelvic-fin morphologies. The highest proportion of reproductive females was registered in January and February, which represent wet and hot periods. The simultaneous presence of more than one litter at different stages of development within a single female characterized this species as superfetaceous.

Dissection of 286 specimens of the Bothrops neuwiedi pubescens, combined with data on captive individuals, provided information on the reproductive biology of this viperid snake from southern Brazil. Females attained larger body sizes than males, and reproduction was seasonal with mating taking place in autumn when males were more frequently encountered. Vitellogenesis occurred from summer to spring (January-September), sperm storage during autumn and winter (May-September), ovulation and fertilization in early spring (September), embryonic development during middle spring and summer (October-March), and parturition in the summer (January-March). Embryonic development was estimated to last from three to five months, a shorter time than was previously reported. The number of offspring of Bothrops neuwiedi pubescens born in one litter can vary from four to 25 ((x) over bar = 11). Fecundity is correlated with maternal body size. Neonates measure 17-25 cm SVL. Inferred growth rate of juveniles was low (10 mm/month in first year), with males attaining sexual maturity at about 16 months, but maturity is delayed in females for at least two additional years.
