O Cul² prepara-se sob a forma de hidrosol cuja dosagem pode ser rigorosa. De suas propriedades quimicas já estudadas por TRAUBE é mais notavel a sensibilidade desse corpo ao oxijeno nacente. Basta em verdade 0,00001 de H²O² para que o Cul² se decomponha desprendendo-se I. Inversamente ele decompõe a agua oxijenada em solução neutra a 1 % (Perhydrol MERCK). O oxijeno do ar atmosferico só o decompõe muito lentamente. As soluções se conservam bem por 6 mezes em vidro escuro. Mesmo em 24 horas, uma corrente de ar não o altera. A ação hemolitica desse hidrosol depende principalmente da H²O distilada que entra em sua composição. Em uma solução isotonisada pelo NaCl, a ação hemolitica diminue proporcionalmente ao aumento de concentração de NaCl, ainda mesmo que se aumente a concentração em Cul². A concentração em Cul² só tem influencia manifesta sobre o fenomeno da precipitação da hemoglobina, sobre a qual ele aje como redutor. Tem ação fixadora sobre os leucocitos, e aglutinante sobre as hematias. Não destroe o fermento da fibrina nem a catalase do sangue. Precipita os albuminoides do soro, pelo menos in vitro. In vivo, esses fenomenos não são aparentes. Foi possivel a aplicação do Cul² por via venosa, sem que isso tivesse determinado acidentes nos animais experimentados (cão, cobaia, coelho, rato). Por via peritoneal não era suportado pelo coelho, nem pela cobaia, morrendo os animais dentro das primeiras 24 horas. Por via subcutanea não era tolerado facilmente por cobaias e ratos. Formava-se sempre escara no ponto da inoculação. O cão tolerava bem as inoculaçõis subcutaneas. Mostrou-se esse hidrosol ineficaz na esporotricose e tuberculose experimentais. The ability to detect early molecular responses to various chemicals is central to the understanding of biological impact of pollutants in a context of varying environmental cues. To monitor stress responses in a model plant, we used transgenic moss Physcomitrella patens expressing the beta-glucuronidase reporter (GUS) under the control of the stress-inducible promoter hsp17.3B. Following exposure to pollutants from the dye and paper industry, GUS activity was measured by monitoring a fluorescent product. Chlorophenols, heavy metals and sulphonated anthraquinones were found to specifically activate the hsp17.3B promoter (within hours) in correlation with long-term toxicity effects (within days). At mildly elevated physiological temperatures, the chemical activation of this promoter was strongly amplified, which considerably increased the sensitivity of the bioassay. Together with the activation of hsp17.3B promoter, chlorophenols induced endogenous chaperones that transiently protected a recombinant thermolabile luciferase (LUC) from severe heat denaturation. This sensitive bioassay provides an early warning molecular sensor to industrial pollutants under varying environments, in anticipation to long-term toxic effects in plants. Because of the strong cross-talk between abiotic and chemical stresses that we find, this P. patens line is more likely to serve as a direct toxicity bioassay for pollutants combined with environmental cues, than as an indicator of absolute toxicity thresholds for various pollutants. It is also a powerful tool to study the role of heat shock proteins (HSPs) in plants exposed to combined chemical and environmental stresses.

Ontogenetic changes in digestive capabilities were analyzed in larvae and first juveniles of the spider crab Maja brachydactyla. Activities of five proteinases (total proteases, trypsin, chymotrypsin, pepsin-like and aminopeptidase), three carbohydrases (amylase, maltase and chitinase), an esterase and an alkaline phosphatase were studied to evaluate digestive enzyme profiles of the species. Both quantitative (spectrophotometry and fluorometry) and qualitative (SDS-PAGE) approaches were used. All assayed enzymes were active from hatching (zoea I-ZI) throughout larval development and in first juveniles. Significant variations during ontogeny were found only in total activities likely as a consequence of digestive system development. Specific activity varied little over ontogeny, being significant only for chitinase. Total proteases, trypsin and pepsin-like activities showed a similar pattern of increase as larval ontogeny advanced, decreasing significantly in juveniles. Chymotrypsin continued to increase, showing maximum activity after metamorphosis. Proteinase zymograms confirmed strong proteolytic activity in first zoeas, with increasing bands over the course of ontogeny, decreasing after metamorphosis. A group of bands with high molecular mass was specific to larval stages. Amylase and maltase showed a parallel pattern of continuous increase of total activity as development advanced. Gel-SDS-PAGE showed unchanged patterns of amylase activity in first zoeas of different ages and the most complex set of bands during larval ontogeny in second zoea. Esterase total activity increased significantly as ZI's aged likely reflecting introduction of a lipid-enriched diet. The importance of lipid accumulation at the beginning of ontogeny was also confirmed by the protease/esterase and amylase/esterase activity ratios, which decreased from hatch to late ZI and might be explained as an adaptation, ensuring the next molt. The results suggest that larvae of M. brachydactyla are capable of digesting a variety of dietary substrates as soon as they hatch.

Sustained atrial fibrillation (AF) is maintained by sites displaying high dominant frequency (DF). In patients (pts) with long-standing persistent AF (LS-pAF), their spatial distribution and the presence of a left-to-right atrial DF gradient remain poorly known. We hypothesized that the pre-ablation bi-atrial frequency characteristics of LS-pAF pts terminated within the left atrium (LT) are different from that of non terminated (NT) ones. Methods: 23 consecutive pts (59±7y, LS-pAF duration 19±12m) underwent stepwise catheter ablation (step-CA) consisting in pulmonary veins isolation, left atrial (LA) defragmentation, and right atrial (RA) ablations for non terminated AF. A quadripolar catheter (CAT) was placed into the RA appendage (RAA), a decapolar CAT into the coronary sinus (CS) and a duodecapolar CAT into the LA divided into 8 segments. For each segment, 20-sec of bipolar recording was acquired. The DF was defined as the largest peak in the power spectrum (3-15 Hz). The inter-atrial DF gradient was defined as the DF difference between LA and RA appendages. Results: LS-pAF was terminated in 83% (19/23) of the pts: 17 LT, 2 during RA ablation and 4 NT. The figure shows that before ablation bi-atrial DF values of LT pts are significantly lower than that of NT pts for each LA segment as well as for the RAA (p < 0.05). No significant LA-to-RA DF gradient was observed both for LT (0.3±0.5 Hz, p=ns) and NT (0.5±0.03 Hz, p=ns) pts. No significant difference in DF values was observed between LA segments. Conclusions: The lower DF of LT pts is suggestive of a higher organization within both atria compared to NT pts. Our findings suggest that low bi-atrial DF values, but not inter-atrial DF gradient, might be of interest for selecting LS-pAF candidates for sinus rhythm restoration by step-CA.

Helicobacter pylori is the most common gastric bacteria of human beings. Animal-borne helicobacter have been associated with gastritis, ulceration, and gastric mucosa-associated lymphoid-tissue lymphoma in people. We attempted to identify the species of Helicobacter spp. that infect human beings in north Paraná, Brazil. Samples of gastric mucosa from 38 dyspeptic patients were analyzed by optic microscopy on silver stained slides, polimerase chain reaction (PCR), and enzymatic cleavage. Genus and species-specific primers to H. pylori, H. heilmannii, H. felis, and consensual primers to H. bizzozeronii or H. salomonis were used. The PCR products were submitted to enzymatic cleavage by VspI (Helicobacter spp. product) and HinfI (species products) enzymes. Thirty-two out of 38 patients evaluated had 3.2 to 5 µm long bacteria that resembled H. pylori in Warthin-Starry stained slides and were positive to the genus Helicobacter by PCR. In 30 of these patients the bacteria were identified as H. pylori. Two samples positive by silver stain were negative to all species tested by PCR. None of the 38 samples was positive to animal-origin helicobacter species. These results show that PCR and enzymatic restriction are practical methods to identify the species of helicobacters present in gastric mucosa of human beings. People in north Paraná appear to be infected mostly with H. pylori.

Primary cultures of cardiomyocytes represent a useful model for analyzing cardiac cell biology as well as pathogenesis of several cardiovascular disorders. Our aim was to standardize protocols for determining the damage of cardiac cells cultured in vitro by measuring the creatine kinase and its cardiac isotype and lactate dehydrogenase activities in the supernatants of mice cardiomyocytes submitted to different protocols of cell lysis. Our data showed that due to its higher specificity, the cardiac isotype creatine kinase was the most sensitive as compared to the others studied enzymatic markers, and can be used to monitor and evaluate cardiac damage in in vitro assays.

El presente documento tiene por objetivo identificar la evolución que ha tenido la inteligencia de negocios en la ultima década, para determinar en que punto se encuentra y para donde va, además de identificar las alternativas de desarrollo que tiene, apalancándose con las Tecnologías de la Información, y más específicamente con las herramientas empaquetadas de BI de tipo Open Source.Así mismo, la presente investigación identifica las prácticas más convenientes, para sistematizar la elección y adaptación de la inteligencia de negocios en las organizaciones, analizando estas en uncaso exitoso real de implantación de la inteligencia de negocios sobre una herramienta BI tipo Open Source.

Actualmente muchas empresas ya sean grandes o pequeñas, se plantean implantar en su organización un sistema de información que les ayude en la toma de decisiones. Una de las tendencias actuales es implantar un sistema de BI (Business Intelligence). Muchas organizaciones implantan este tipo de sistemas sin haber realizado un estudio previo de las necesidades y problemas de la empresa, de los beneficios que se pretenden obtener con la implantación del sistema, de la aceptación y formación de los usuarios, de las herramientas y proveedores de BI,... y así un largo etcétera de situaciones y condiciones que se deberían tener en cuenta para la implantación de esta clase de sistemas en cualquier tipo de organización, ya sea grande o pequeña.En el presente artículo se realiza una síntesis de la amalgama de información existente en los sistemas de BI, se realiza una definición aclaratoria, así como también se expone su estado actual, beneficios, factores de riesgo, pasos para conseguir una implantación exitosa y los componentes y herramientas open source de los sistemas de BI. Primero se exponen las principales herramientas open source existentes en el mercado y se profundiza más especialmente en la Suite open source de BI Pentaho.
