The presence of enteric viruses in biosolids can be underestimated due to the inefficient methods (mainly molecular methods) used to recover the viruses from these matrices. Therefore, the goal of this study was to evaluate the different methods used to recover adenoviruses (AdV), rotavirus species A (RVA), norovirus genogroup II (NoV GII) and the hepatitis A virus (HAV) from biosolid samples at a large urban wastewater treatment plant in Brazil after they had been treated by mesophilic anaerobic digestion. Quantitative polymerase chain reaction (PCR) was used for spiking experiments to compare the detection limits of feasible methods, such as beef extract elution and ultracentrifugation. Tests were performed to detect the inhibition levels and the bacteriophage PP7 was used as an internal control. The results showed that the inhibitors affected the efficiency of the PCR reaction and that beef extract elution is a suitable method for detecting enteric viruses, mainly AdV from biosolid samples. All of the viral groups were detected in the biosolid samples: AdV (90%), RVA, NoV GII (45%) and HAV (18%), indicating the viruses' resistance to the anaerobic treatment process. This is the first study in Brazil to detect the presence of RVA, AdV, NoV GII and HAV in anaerobically digested sludge, highlighting the importance of adequate waste management.

We determined the serum IgE levels and T-helper (Th)17-related cytokines during distinct hepatitis A virus (HAV)-induced clinical courses in children. A significantly higher concentration of macrophage inflammatory protein 3α, interleukin (IL)-17E and IL-17F in HAV-infected children with intermediate liver injury compared with those with minor liver damage was found. A reduction in the IgE levels in those patients who showed the highest levels of IL-17F in the group of intermediate liver injury was found. The data suggested that the Th17-related profile is associated with the severity of HAV infection and might play a role on the modulation achieved by HAV during allergies.

An increasing amount of research has been conducted on immunoglobulin Y (IgY) because the use of IgY offers several advantages with respect to diagnostic testing, including its easy accessibility, low cost and translatability to large-scale production, in addition to the fact that it can be ethically produced. In a previous work, immunoglobulin was produced and purified from egg yolks (IgY) reactive to hepatitis A virus (HAV) antigens. In the present work, this anti-HAV-specific IgY was used in an indirect immunofluorescence assay to detect viral antigens in liver biopsies that were obtained from experimentally infected cynomolgus monkeys. Fields that were positive for HAV antigen were detected in liver sections using confocal microscopy. In conclusion, egg yolks from immunised hens may be a reliable source for antibody production, which can be employed for immunological studies.

This study was conducted to analyse the course and the outcome of the liver disease in the co-infected animals in order to evaluate a possible synergic effect of human parvovirus B19 (B19V) and hepatitis A virus (HAV) co-infection. Nine adult cynomolgus monkeys were inoculated with serum obtained from a fatal case of B19V infection and/or a faecal suspension of acute HAV. The presence of specific antibodies to HAV and B19V, liver enzyme levels, viraemia, haematological changes, and necroinflammatory liver lesions were used for monitoring the infections. Seroconversion was confirmed in all infected groups. A similar pattern of B19V infection to human disease was observed, which was characterised by high and persistent viraemia in association with reticulocytopenia and mild to moderate anaemia during the period of investigation (59 days). Additionally, the intranuclear inclusion bodies were observed in pro-erythroblast cell from an infected cynomolgus and B19V Ag in hepatocytes. The erythroid hypoplasia and decrease in lymphocyte counts were more evident in the co-infected group. The present results demonstrated, for the first time, the susceptibility of cynomolgus to B19V infection, but it did not show a worsening of liver histopathology in the co-infected group.

Hepatitis A virus (HAV), the prototype of genus Hepatovirus, has several unique biological characteristics that distinguish it from other members of the Picornaviridae family. Among these, the need for an intact eIF4G factor for the initiation of translation results in an inability to shut down host protein synthesis by a mechanism similar to that of other picornaviruses. Consequently, HAV must inefficiently compete for the cellular translational machinery and this may explain its poor growth in cell culture. In this context of virus/cell competition, HAV has strategically adopted a naturally highly deoptimized codon usage with respect to that of its cellular host. With the aim to optimize its codon usage the virus was adapted to propagate in cells with impaired protein synthesis, in order to make tRNA pools more available for the virus. A significant loss of fitness was the immediate response to the adaptation process that was, however, later on recovered and more associated to a re-deoptimization rather than to an optimization of the codon usage specifically in the capsid coding region. These results exclude translation selection and instead suggest fine-tuning translation kinetics selection as the underlying mechanism of the codon usage bias in this specific genome region. Additionally, the results provide clear evidence of the Red Queen dynamics of evolution since the virus has very much evolved to re-adapt its codon usage to the environmental cellular changing conditions in order to recover the original fitness.

The increase in seafood production, especially in mariculture worldwide, has brought out the need of continued monitoring of shellfish production areas in order to ensure safety to human consumption. The purpose of this research was to evaluate pathogenic protozoa, viruses and bacteria contamination in oysters before and after UV depuration procedure, in brackish waters at all stages of cultivation and treatment steps and to enumerate microbiological indicators of fecal contamination from production site up to depuration site in an oyster cooperative located at the Southeastern estuarine area of Brazil. Oysters and brackish water were collected monthly from September 2009 to November 2010. Four sampling sites were selected for enteropathogens analysis: site 1- oyster growth, site 2- catchment water (before UV depuration procedure), site 3 - filtration stage of water treatment (only for protozoa analysis) and site 4- oyster's depuration tank. Three microbiological indicators ! were examined at sites 1, 2 and 4. The following pathogenic microorganisms were searched: Giardia cysts, Cryptosporidium oocysts, Human Adenovirus (HAdV), Hepatitis A virus (HAV), Human Norovirus (HnoV) (genogroups I and II), JC strain Polyomavirus (JCPyV) and Salmonella sp. Analysis consisted of molecular detection (qPCR) for viruses (oysters and water samples); immunomagnetic separation followed by direct immunofluorescence assay for Cryptosporidium oocysts and Giardia cysts and also molecular detection (PCR) for the latter (oysters and water samples); commercial kit (Reveal-Neogee (R)) for Salmonella analysis (oysters). Giardia was the most prevalent pathogen in all sites where it was detected: 36.3%, 18.1%, 36.3% and 27.2% of water from sites 1, 2, 3 and 4 respectively; 36.3% of oysters from site 1 and 54.5% of depurated oysters were harboring Giardia cysts. The huge majority of contaminated samples were classified as Giardia duodenalis. HAdv was detected in water and o! ysters from growth site and HnoV GI in two batches of oysters ! (site 1) in huge concentrations (2.11 x 10(13), 3.10 x 10(12) gc/g). In depuration tank site, Salmonella sp., HAV (4.84 x 10(3)) and HnoV GII (7.97 x 10(14)) were detected once in different batches of oysters. Cryptosporidium spp. oocysts were present in 9.0% of water samples from site four. These results reflect the contamination of oysters even when UV depuration procedures are employed in this shellfish treatment plant. Moreover, the molecular comprehension of the sources of contamination is necessary to develop an efficient management strategy allied to shellfish treatment improvement to prevent foodborne illnesses. Hoitotieteen pääkokoelma sijaitsee Terveystieteiden osastokirjastossa, jossa painettu yleiskokoelma koostuu 790 nimekkeestä monografioita (painettujen monografiasarjojen osat ovat mukana luvussa). Hoitotiede-kokoelmasta yli 40 % (329 nimekettä) käsittelee käytännön hoitotyötä joko yleisesti (21 % / 168) tai jollakin erityisalueella (20 % /161). Erityisalueista suurimmat ovat geriatrinen hoitotyö (6 % / 42) ja psykiatrinen hoitotyö (4 % 35). Lähes kolmasosa kokoelmasta on yleisteoksia (27 % / 241) käsittäen mm. koulutusta (5 % /41), tutkimusta (11 % /86) ja hoitotyötä ammattina (6 % /51) tarkastelevaa kirjallisuutta. Hoitotyön etiikkaa, filosofiaa ja psykologiaa on kokoelmassa vajaa viidennes (17 % / 137). Hallintoa, esimiestehtäviä ja hoitotyön opetusta käsittelevää kirjallisuutta oli 5 % (43 nimekettä). By reconstructing the nutrient balance of a Catalan v illage circa 1861-65 we examine the sustainability of organic agricultural sy stems in the northwest Mediterranean bioregion prior to the green rev olution and the question of whether the nutrients extracted f rom the soil were replenished. With a population density of 59 inhabitants per square km, similar to other northern European rural areas at that time, and a lower liv estock density per cropland unit, this v illage experienced a manure shortage. The gap was f illed by other labour-intensiv e way s of transf erring nutrients f rom uncultiv ated areas into the cropland. Key elements in this agricultural sy stem were v iney ards because they hav e f ew nutrient requirements, and woodland and scrublands as sources of relev ant amounts of nutrients collected in sev eral ways.

There is evidence that virtual reality (VR) pain distraction is effective at improving pain-related outcomes. However, more research is needed to investigate VR environments with other pain-related goals. The main aim of this study was to compare the differential effects of two VR environments on a set of pain-related and cognitive variables during a cold pressor experiment. One of these environments aimed to distract attention away from pain (VRD), whereas the other was designed to enhance pain control (VRC). Participants were 77 psychology students, who were randomly assigned to one of the following three conditions during the cold pressor experiment: (a) VRD, (b) VRC, or (c) Non-VR (control condition). Data were collected regarding both pain-related variables (intensity, tolerance, threshold, time perception, and pain sensitivity range) and cognitive variables (self-efficacy and catastrophizing). Results showed that in comparison with the control condition, the VRC intervention significantly increased pain tolerance, the pain sensitivity range, and the degree of time underestimation. It also increased self-efficacy in tolerating pain and led to a reduction in reported helplessness. The VRD intervention significantly increased the pain threshold and pain tolerance in comparison with the control condition, but it did not affect any of the cognitive variables. Overall, the intervention designed to enhance control seems to have a greater effect on the cognitive variables assessed. Although these results need to be replicated in further studies, the findings suggest that the VRC intervention has considerable potential in terms of increasing self-efficacy and modifying the negative thoughts that commonly accompany pain problems.

Within caring science, investigations and explorations have been carried out on the ontology of caring, and many aspects of the field have been the subject of scientific research. The main subject for this study is grounded on the human need for aesthetics. The purpose is to find how the aesthetic dimension is taken into consideration and how the aesthetic surroundings are evaluated and attended to, in the general hospitals in Norway. The theoretical perspective is founded basicly on the study of litterature from caring science and philosophy. The aim is to develop a disposition for a framework on the aesthetic surroundings in the hospitals, and to develop phenomenological and ontological knowledge and understanding of the aesthetic dimension. The study aspires to attain a deeper understanding of the aesthetic acknowledgment and of the aesthetic needs. The focus is how the aesthetic dimension can promote health and wellbeing, both for patients and for the caring staff, in the general hospitals and why the aesthetic dimension should be obligatory in `evident care¿. The study concentrates on 11 selected categories in the hospital environment, where aesthetics is of importance. The research is implemented on 5 part studies: 1. part is a study of caring science and philosophical theories about aesthetics, as a framework for the investigation. 2. part is a survey of the physical environment, in Norwegian somatic hospitals, with focus on aesthetics. This by analyzing the strategy plans for the hospitals. 3. and 4. part is questionnaires to patients and nurses to get their opinion and evaluation of the aesthetic environment in the hospitals they are connected to, and their opinion on how this influences the health and wellness for both patients and caring staff. 5. part is qualitative interviews with 16 experts, to get their opinion and evaluation of the aesthetic environment in hospitals they are or have been connected to. How would the experts like the aesthetic surroundings to be, and also their opinion on what influence they think aesthetics has on health and wellness. The main literature of caring science is rooted in K. Erikssons caring theory as well as philosophic literature; mainly I. Kant, Platon and Y. Hirn's theories on aesthetics. Various scientificresearchers of aesthetics have also been referred to. The methodological approach is a triangulation with a hermeneutic exploration, where H.G. Gadamer and Ricoeur provides the inspirational foundation. The findings and conclusions result in the development of new hypothesis for the caring science foundation and suggestions, a disposition for a framework related to future planning of the aesthetic environments in general hospitals. It might be said that a common thread arises/appears in the invariance's (invariables) that are discerned from the analysis and interpretation of the interviews and also important angles shows in the variances that crystallized. Based on the conclusions the study confirms that there is a clearconnection between health, wellness and aesthetics in the environment and that it is an ethical obligationfor those in the caring professions to be aware of and attend to the aesthetic dimension.

Avhandlingen söker en djupare förståelse för vad som är det vårdande för människor som befinner sig i djupt lidande som gränsar till det outhärdliga lidandet. Människans övergripande livsrum är sammansatt av flera mindre rum dit tillträde ges i olika omfattning. Vissa av människans handlingar innebär att de rum som ger möjlighet till näring och växt är stängda. Endast de yttersta livsrummen återstår att vistas i. Vårdaren bör vara beredd att ge sig in i dessa tillsammans med patienten. Fokus är på vad som är det vårdande i en samtalsform som här benämns hermeneutiska vårdande samtal. Patienterna som samtalen förs med befinner sig inom rättspsykiatrisk vårdkontext. Där vårdas människor, som överlämnats till vård efter att de begått brottsliga handlingar. Skälet till att de vårdas och inte befinner sig i fängelse är att de befunnits lida av en allvarlig psykisk störning (som den juridiska termen lyder). Grunden för samtalen är den caritativa vårdvetenskapen där ethos anger tonen och riktningen. Substansen i samtalen hämtas från de filosofiska fundamenten varat och transcendensen, det Oändliga samt gränssituation. En annan del av samtalsplattformen utgörs av några centrala begrepp såsom lidande, livsförståelsemörker och försoning. Skönlitterära texter, poesi och musik har även utgjort en viktig förutsättning för samtalen att växa sig mot allt större djup. Samtalen ses som en gemensam bildningsresa för forskaren och forskningspersonen som handlar om att lämna det bekanta för det främmande och sedan återvända som en annan. Rytmen och rörelsen i samtalen är väsentliga för uppnåendet av den horisontsammansmältning som kan leda fram till en gemensam förståelse av saken, det vill säga vad som är det vårdade. Resultatet framträder genom en mönsterskapande fas, ett sökandet efter det universella samt en allomfattande tolkning. De mönster som visar sig är att bära och hantera skuld, lindrat lidande genom utveckling i livsförståelse, möten i vårdade gemenskap samt försoningens vandring och slutligen hopp. Dessa mönster ger glimtar av en ny förståelsehorisont. Förståelsen av vad som är det vårdande i de hermeneutiska vårdade samtalen utifrån dessa mönster, mynnar ut i följande: Den vårdade gemenskapen innebär att vårdaren är tillsammans med patienten i gränssituationen, den gemensamma förståelsen av saken betyder att patienten så småningom kan ge sig själv rätten att inneha det mänskliga ämbetet, lidandet öppnar för det universella som ibland kan ta formen av en gudstro, enda vägen till försoning är att för alltid bära skulden, reningen ger kraft samt slutligen att vara varandras hopp. Den nya förståelsen har kontextspecifika drag, men öppnar för ett vidare inseende i vad som är det vårdande för den som befinner sig i ett stort lidande. Den funna kunskapen betraktas utifrån sin universella karaktär som överförbar till andra människor och till andra vårdade kontexter än den som är aktuell i studien.

Undervattensmiljön i skärgårdarna och de kustnära områdena i norra Östersjön är mångformiga och produktiva. Arternas levnadsmil-jöer styrs bl.a. av bottentyp, exponeringsgrad och ljustillgång. Kunskap och information om undervattensarterna är av stor vikt för en hållbar planering av kust- och havsmiljön. Denna rapport beskriver indelningen av de viktigaste undervattenshabitaten i norra Östersjöns kustvatten samt hur ett naturvärde på dessa miljöer kan uppskattas på basen av de olika ekologiska funktioner som habitatet uppfyller. De mest inflytelserika mänskliga hoten, både små- och storskaliga, som allvarligt kan hota habitatens fortbestånd behandlas också. Habitaten som beskrivs är både av typen hårdbotten och mjukbotten. Hårdbottenssamhällena bildar distinkta bälten från ytan mot bott-nen: trådalgszonen, zonen med stora fleråriga brunalger (Fucus vesiculosus, F. radicans), rödalgszonen och blåmusselzonen (Mytilus edulis/trossulus). Mjukbottensamhällena är uppbyggda kring ålgräs (Zostera marina), andra vattenlevande kärlväxter eller kransalger. En stor del av både de grunda och djupa mjukbottnarna saknar vegetation och samhällena struktureras då av ryggradslösa djur i och på sedi-mentytan. Habitaten har klassificerats och värderats enligt en tvåstegsmodell. Först har habitaten namngivits efter den dominerande arten eller artgruppen som bestämts enligt täckningsgrad. Därefter har varje habitat getts ett värde enligt sin ekologiska betydelse. Naturvärdet är indelat i skalan 1-5. Projektet NANNUT – The Nature and Nurture in the Northern Baltic Sea – har gjort inventeringar av undervattensmiljön på olika håll av kusterna och skärgårdarna i Finland 2010-2011. Målsättningen har varit att utveckla enkla och kostnadseffektiva metoder både för kartering och värdering av undervattenshabitat. Det huvudsakliga undersökningsområdet var förlagt till Raseborgsstad i södra Finland.

Vad händer i tidvattenzonen? Var går gränsen mellan land och hav, vad händer i tidvattenzonen och vem ansvarar för detta? I västra Indiska oceanen (VIO) kan avståndet mellan den lägsta nivån för lågvattnet och den högsta nivån för högvattnet vara flera kilometer och nivåskillnaderna upp till 6 meter och detta skapar ett stort och föränderligt område. Syftet med min avhandling är att öka förståelsen för tidvattenzonen i tropiska och subtropiska västra Indiska oceanen. Sammanfattningsvis visar mina studier att det finns ett mycket stort värde i den komplexa tidvattenzonen, men också att det här området hotas från både land och hav, genom t.ex. överexploatering, erosion och föroreningar. Uttnyttjandet av tidvattenzonen är stort och min avhandling har visat att aktiviteter såsom fiske i form av plocking av musslor och andra ryggradslösa djur och hamnaktiviteter påverkar den biologiska mångfalden negativt, vilket leder till försämrad levnadsstandard för resursutnyttjande människor i regionen. För att förbättra situationen krävs det mer forskning, miljöövervakning och bättre förvaltning av tidvattenzonen. Experter i regionen har rangordnat förslag på förvaltningsstrategier som skulle kunna testas för att förbättra miljön och skapa ett mer hållbart nyttjande. Avhandlingen visar även att det är möjligt att använda fjärranalysteknik såsom satellitbildsanalys för att kvantifiera mängden sjögräsvegetation (i form av biomassa), vilket kan ha stor betydelse för att förbättra storskalig miljöövervakning av kustnära naturtyper (habitat). I avhandlingsarbetet har jag använt mig av ett multidisciplinärt tillvägagångssätt och använt metoder såsom ekologisk och biologisk provtagning, intervjuer, observationer, diskussionsgrupper, frågeformulär och fjärranalys. Resultaten presenterade i denna avhandling ger en ökad kunskap om tidvattenzonen i utvecklingsländerna inom VIO-regionen som kan användas för att initiera och fortsätta att utveckla hållbara förvaltningsstrategier av biologiska resurser.
