The relative contribution of migration of Rhizoctonia solani anastomosis group 3 (AG-3) on infested potato seed tubers originating from production areas in Canada, Maine, and Wisconsin (source population) to the genetic diversity and structure of populations of R. solani AG-3 in North Carolina (NC) soil (recipient population) was examined. The frequency of alleles detected by multilocus polymerase chain reaction-restriction fragment length polymorphisms, heterozygosity at individual loci, and gametic phase disequilibrium between all pairs of loci were determined for subpopulations of R. solani AG-3 from eight sources of potato seed tubers and from five soils in NC. Analysis of molecular variation revealed little variation between seed source and NC recipient soil populations or between subpopulations within each region. Analysis of population data with a Bayesian-based statistical method previously developed for detecting migration in human populations suggested that six multilocus genotypes from the NC soil population had a statistically significant probability of being migrants from the northern source population. The one-way (unidirectional) migration of genotypes of R. solani AG-3 into NC on infested potato seed tubers from Canada, Maine, and Wisconsin provides a plausible explanation for the lack of genetic subdivision (differentiation) between populations of the pathogen in NC soils or between the northern source and the NC recipient soil populations.

The yacon (Polymnia sonchifolia) is used largely for the high fructan content of its tubers; consequently, it is a good alternative for diabetics. One of the more important restricting factors of the commercial production of yacon is its susceptibility to nematode attack. This, as well as germplasm bank maintenance, justifies the importance of in vitro propagation of this species. In this way, our work aimed to verify the best asepsis method for yacon for the in vitro establishment from the rhizophore and the axillary buds of the aerial parts, and the effect of benzylaminopurine (BAP) addition to the culture medium. The number of contaminated cultures, the occurrence of phenolic oxidation and the occurrence of a vitreous aspect, showed differences with bud source, immersion time for asepsis, and BAP use. The results contribute to establishing a yacon micro propagation procedure.

The objective of this study was to determine the influence of five different water levels on the crop development of Calla. The crop parameters evaluated were leaf area and evapotranspiration. The study was conducted in glass greenhouse with 50% of sunlight reduction. The plants were grown in PVC pots with 150 mm diameter, which were filled with substrate. The plant tubers weighed from 10 g to 12 g. The pots were placed within containers, under water level constant automatically.. The table water levels used were 10, 17, 24, 31 and 38 cm. Nine evaluations during the growth cycle checked the growth development. The evaporation varied from 26.89 to 46.14 L.plant-1 for 38 and 10 cm water levels, respectively and leaf area per plant showed 1011.6 to 2016.3 for the same levels. The substrate water was more available in the treatment 24 cm, with more restrictions in the upper and lower treatments. There was positive correlation between leaf area and evapotranspiration at the final of the culture.

A synbiotic yoghurt based on a combination of soymilk and yacon water extract (from yacon root tubers) was developed as a novel food product fermented with a probiotic culture of Enterococcus faecium CRL 183 and Lactobacillus helveticus ssp jugurti 4l6. Response surface methodology (RSM) was used to optimize the independent variables soymilk protein concentration and percentage of yacon extract in the formulation through a Central Composite Rotatable Design (CCRD), consisting of a 22 factorial design with two levels (-1, +1), two central points (0) and four axial points (± a, 0) (0, ± α). The responses were assessed by consumer acceptance tests. The optimization indicated that a formulation with a soymilk protein concentration of 1.74g/L and 25.86% of yacon extract gave the best average values, 5.91 for the taste and 6.00 for the overall impression responses. The formulation with 40% of yacon extract and the same concentration of soymilk protein achieved similar acceptance values: taste (5.94) and overall impression (5.87), however, with the extra yacon, it probably had a greater content of prebiotic fructooligosaccharides. Consequently, both formulations may give useful functional foods, with sensory properties comparable with those of soy yoghurt (control formulation). In tropical conditions such as Brazil, depending on the local altitude, potatoes can be planted and harvested in every month of the year successively, which favors pest and disease incidence, especially aphids transmission viruses. Therefore, obtaining good yield depends on constant pest and disease control and quality potato seed acquisition. One of the main strategies to increase a healthier seed potato multiplication rate and production is the use of hydroponic systems, with or without substrates, in channels, pots or boxes. In 2005 and 2008, researches investigated several hydroponic systems with and without the use of substrates. In the hydroponic systems without substrate, the aeroponic system resulted in the highest multiplication rate, with 47 tubers plant-1 compared to NFT and DFT, 35 and 37 tubers plant-1, respectively. With the use of substrates, the pot system obtained better results, reaching 12 tubers plant-1, followed by the capillary system and boxes, with 8 and 7 tubers plant-1, respectively. Potato seed production was influenced by hydroponics systems. Among hydroponic systems with substrate, the pots were shown as the best option. However, higher yields were obtained in the hydroponics systems without substrate, with the aeroponic system that provided the best results.

The use of cultivars efficient in nutrient use is essential to increase yield and reduce production costs. The objective of this study was to evaluate absorption and nutrients use efficiency on five potato cultivars. The experiment was conduced in Itaí, São Paulo State, Brazil, in winter cropping season of 2008 on Oxisol. The experimental design was randomized blocks with split-plots and four replications. Plots comprised the five potato cultivars and subplots were established by sampling times, performed at planting and every seven days after emergence. At 97 DAP was calculated the nutrients use efficiency for dry matter production and at 122 DAP were calculated the nutrients use efficiency for fresh tuber and tuber dry matter yield. Mondial and Asterix cultivars showed greater dry matter production, higher fresh tubers yield and nutrients accumulation. Nutrients use efficiency for total dry matter and tubers dry matter, was similar among cultivars, but Ágata was more efficient in nutrients use efficiency for fresh tubers yield.

A simplified procedure for the preparation of immobilized beta-amylase using non-purified extract from fresh sweet potato tubers is established in this paper, using differently activated agarose supports. Beta-amylase glutaraldehyde derivative was the preparation with best features, presenting improved temperature and pH stability and activity. The possibility of reusing the amylase was also shown, when this immobilized enzyme was fully active for five cycles of use. However, immobilization decreased enzyme activity to around 15%. This seems to be mainly due to diffusion limitations of the starch inside the pores of the biocatalyst particles. A fifteen-fold increase in the Km was noticed, while the decrease of Vmax was only 30% (10.1 U mg-1 protein and 7.03 U mg-1 protein for free and immobilized preparations, respectively). © 2013 Elsevier Ltd.
