CAP1/Prss8 is a membrane-bound serine protease involved in the regulation of several different effectors, such as the epithelial sodium channel ENaC, the protease-activated receptor PAR2, the tight junction proteins, and the profilaggrin polypeptide. Recently, the V170D and the G54-P57 deletion mutations within the CAP1/Prss8 gene, identified in mouse frizzy (fr) and rat hairless (fr(CR)) animals, respectively, have been proposed to be responsible for their skin phenotypes. In the present study, we analyzed those mutations, revealing a change in the protein structure, a modification of the glycosylation state, and an overall reduction in the activation of ENaC of the two mutant proteins. In vivo analyses demonstrated that both fr and fr(CR) mutant animals present analogous reduction of embryonic viability, similar histologic aberrations at the level of the skin, and a significant decrease in the activity of ENaC in the distal colon compared with their control littermates. Hairless rats additionally had dehydration defects in skin and intestine and significant reduction in the body weight. In conclusion, we provided molecular and functional evidence that CAP1/Prss8 mutations are accountable for the defects in fr and fr(CR) animals, and we furthermore demonstrate a decreased function of the CAP1/Prss8 mutant proteins. Therefore, fr and fr(CR) animals are suitable models to investigate the consequences of CAP1/Prss8 action on its target proteins in the whole organism.

Studies in cystic fibrosis patients and mice overexpressing the epithelial Na(+) channel beta-subunit (betaENaC-Tg) suggest that raised airway Na(+) transport and airway surface liquid (ASL) depletion are central to the pathogenesis of cystic fibrosis lung disease. However, patients or mice with Liddle gain-of-function betaENaC mutations exhibit hypertension but no lung disease. To investigate this apparent paradox, we compared the airway phenotype (nasal versus tracheal) of Liddle with CFTR-null, betaENaC-Tg, and double mutant mice. In mouse nasal epithelium, the region that functionally mimics human airways, high levels of CFTR expression inhibited Liddle epithelial Nat channel (ENaC) hyperfunction. Conversely, in mouse trachea, low levels of CFTR failed to suppress Liddle ENaC hyperfunction. Indeed, Na(+) transport measured in Ussing chambers ("flooded" conditions) was raised in both Liddle and betaENaC-Tg mice. Because enhanced Na(+) transport did not correlate with lung disease in these mutant mice, measurements in tracheal cultures under physiologic "thin film" conditions and in vivo were performed. Regulation of ASL volume and ENaC-mediated Na(+) absorption were intact in Liddle but defective in betaENaC-Tg mice. We conclude that the capacity to regulate Na(+) transport and ASL volume, not absolute Na(+) transport rates in Ussing chambers, is the key physiologic function protecting airways from dehydration-induced lung disease.

Regulation of sodium balance is a critical factor in the maintenance of euvolemia, and dysregulation of renal sodium excretion results in disorders of altered intravascular volume, such as hypertension. The amiloride-sensitive epithelial sodium channel (ENaC) is thought to be the only mechanism for sodium transport in the cortical collecting duct (CCD) of the kidney. However, it has been found that much of the sodium absorption in the CCD is actually amiloride insensitive and sensitive to thiazide diuretics, which also block the Na-Cl cotransporter (NCC) located in the distal convoluted tubule. In this study, we have demonstrated the presence of electroneutral, amiloride-resistant, thiazide-sensitive, transepithelial NaCl absorption in mouse CCDs, which persists even with genetic disruption of ENaC. Furthermore, hydrochlorothiazide (HCTZ) increased excretion of Na+ and Cl- in mice devoid of the thiazide target NCC, suggesting that an additional mechanism might account for this effect. Studies on isolated CCDs suggested that the parallel action of the Na+-driven Cl-/HCO3- exchanger (NDCBE/SLC4A8) and the Na+-independent Cl-/HCO3- exchanger (pendrin/SLC26A4) accounted for the electroneutral thiazide-sensitive sodium transport. Furthermore, genetic ablation of SLC4A8 abolished thiazide-sensitive NaCl transport in the CCD. These studies establish what we believe to be a novel role for NDCBE in mediating substantial Na+ reabsorption in the CCD and suggest a role for this transporter in the regulation of fluid homeostasis in mice.

Lithium-induced nephrogenic diabetes insipidus (NDI) is accompanied by polyuria, downregulation of aquaporin 2 (AQP2), and cellular remodeling of the collecting duct (CD). The amiloride-sensitive epithelial sodium channel (ENaC) is a likely candidate for lithium entry. Here, we subjected transgenic mice lacking αENaC specifically in the CD (knockout [KO] mice) and littermate controls to chronic lithium treatment. In contrast to control mice, KO mice did not markedly increase their water intake. Furthermore, KO mice did not demonstrate the polyuria and reduction in urine osmolality induced by lithium treatment in the control mice. Lithium treatment reduced AQP2 protein levels in the cortex/outer medulla and inner medulla (IM) of control mice but only partially reduced AQP2 levels in the IM of KO mice. Furthermore, lithium induced expression of H(+)-ATPase in the IM of control mice but not KO mice. In conclusion, the absence of functional ENaC in the CD protects mice from lithium-induced NDI. These data support the hypothesis that ENaC-mediated lithium entry into the CD principal cells contributes to the pathogenesis of lithium-induced NDI.

Cirrhosis is a frequent and severe disease, complicated by renal sodium retention leading to ascites and oedema. A better understanding of the complex mechanisms responsible for renal sodium handling could improve clinical management of sodium retention. Our aim was to determine the importance of the amiloride-sensitive epithelial sodium channel (ENaC) in collecting ducts in compensate and decompensate cirrhosis. Bile duct ligation was performed in control mice (CTL) and collecting duct-specific αENaC knockout (KO) mice, and ascites development, aldosterone plasma concentration, urinary sodium/potassium ratio and sodium transporter expression were compared. Disruption of ENaC in collecting ducts (CDs) did not alter ascites development, urinary sodium/potassium ratio, plasma aldosterone concentrations or Na,K-ATPase abundance in CCDs. Total αENaC abundance in whole kidney increased in cirrhotic mice of both genotypes and cleaved forms of α and γ ENaC increased only in ascitic mice of both genotypes. The sodium chloride cotransporter (NCC) abundance was lower in non-ascitic KO, compared to non-ascitic CTL, and increased when ascites appeared. In ascitic mice, the lack of αENaC in CDs induced an upregulation of total ENaC and NCC and correlated with the cleavage of ENaC subunits. This revealed compensatory mechanisms which could also take place when treating the patients with diuretics. These compensatory mechanisms should be considered for future development of therapeutic strategies.

Les syndromes myotoniques congénitaux atypiques dus à des mutations du canal sodé voltage-dépendant Nav1.4 se distinguent des myotonies congénitales classiques (canal chlore ClC-1) par la présence de traits atypiques incluant des myotonies douloureuses aggravées au froid et à l’ingestion de potassium. La caractérisation clinique et moléculaire de plusieurs familles atteintes de ces conditions rares dans la région du Saguenay-Lac-St-Jean nous a permis de découvrir une nouvelle mutation SCN4A à effet fondateur causant un phénotype de myotonies douloureuses aggravées au froid, parfois accompagné de phénomènes dystrophiques ou paralytiques. L’ampleur de notre cohorte nous permet de commenter sur l’hétérogénité phénotypique observée, sur les traits caractéristiques des syndromes associés au gène SCN4A, sur les implications physiologiques probables d’une telle mutation ainsi que sur les facteurs modulant le phénotype observé. Enfin, notre étude nous permet de souligner l’importance du dépistage familial systématique afin de prévenir les complications anesthésiques potentielles associées à ces conditions.

La restriction de croissance intrautérine (RCIU) est associée à l’apparition de maladies à l’âge adulte et le phénotype de la condition pathologique peut être différent selon le sexe. Notre laboratoire a développé un modèle de RCIU chez le rat en administrant une diète faible en sodium lors du dernier tiers de la gestation entraînant une réduction de l’expansion volémique maternelle et de la perfusion utéroplacentaire. L'activité rénine et la concentration d'aldostérone plasmatique sont augmentées chez la mère et les foetus RCIU. Antérieurement, notre laboratoire a démontré une augmentation de l’expression génique et protéique rénale de la Na+-K+-ATPase-α1 uniquement chez les foetus femelles RCIU. Ainsi, nous émettons l’hypothèse que la diminution du volume circulant chez la rate gestante entraîne une augmentation et une expression différentielle, selon le sexe, des éléments de la cascade de signalisation du récepteur des minéralocorticoïdes (MR) dans les reins de foetus RCIU. L’expression des gènes est réalisée par qRT-PCR et celle des protéines par immunobuvardage de type Western. Bien que les résultats démontrent que la transcription génique de SGK1, α-ENaC et GILZ soit augmentée dans les reins de foetus RCIU, l’expression protéique de SGK1, pSGK1(Thr 256) et α-ENaC est similaire à celle des témoins. La protéine GILZ est indétectable. Pour CNKSR3, aucune différence de l’ARNm ou de la protéine n’a été observée entre les deux groupes. Par contre, même si l’expression génique du MR n’est pas différente, l’expression protéique est diminuée chez les RCIU. Aucun effet du sexe n’a été observé. En conclusion, l’augmentation d'aldostérone plasmatique chez les foetus ayant subi une RCIU stimule la transcription des gènes associés à la voie de réabsorption sodique, mais la quantité protéique demeure inchangée. Ceci suggère qu’il peut avoir des mécanismes de régulation post-transcriptionnelle ou une dégradation accélérée des protéines. Malgré la pertinence du sexe dans le développement de maladies, le sexe n’influence pas l’expression des composantes de la voie de rétention sodique chez le foetus. Il serait important de suivre cette voie en fonction de l’âge et de corréler les expressions génique et protéique avec l’apparition de maladies.

Le transport actif de sodium par les cellules épithéliales alvéolaires est le principal mécanisme impliqué dans la régulation du niveau de liquide dans le poumon distal. Le canal épithélial sodique (ENaC) exprimé par les cellules épithéliales alvéolaires est essentiel à la résorption du liquide des poumons à la naissance ainsi que la résolution de l'œdème pulmonaire chez l'adulte. L'activité et l'expression du canal ENaC sont modulées par de nombreux stress pathophysiologiques. L'inflammation pulmonaire constitue un facteur important dans l'inhibition de l'expression du canal ENaC et pourrait favoriser la formation d'œdème pulmonaire. Nous avons précédemment démontré que différentes cytokines pro-inflammatoires, ainsi que les lipopolysaccharides (LPS) de Pseudomonas aeruginosa, inhibent l'expression de l'ARNm αENaC par des mécanismes de régulation transcriptionnelle et post-transcriptionnelle. Ces résultats suggèrent que les mécanismes qui modulent la stabilité des ARNm αENaC pourraient jouer un rôle important dans la régulation du niveau d’expression du transcrit en condition inflammatoire. Le principal objectif de mes travaux était de caractériser les mécanismes de modulation de l’ARNm αENaC dans les cellules épithéliales alvéolaires lors de différents stress pathophysiologiques et déterminer si cette modulation pouvait s’expliquer en partie par une régulation de la stabilité du transcrit. Mes travaux montrent que les LPS et la cycloheximide inhibent l’expression de l’ARNm αENaC de façon similaire via l’activation des voies de signalisation des MAPK ERK1/2 et p38. Cependant, les mécanismes de modulation de l’expression de l'ARNm αENaC sont différents puisque les LPS répriment la transcription du gène, alors que la cycloheximide diminuerait la stabilité du transcrit via des mécanismes post-transcriptionnels impliquant la région 3' non traduite (3'UTR) de l'ARNm αENaC. Pour mieux étudier le rôle du 3'UTR dans ce processus, nous avons développé un modèle Tet-Off nous permettant de mesurer la demi-vie de l’ARNm αENaC indépendamment de l’utilisation d’un inhibiteur de la transcription comme l'actinomycine D (Act. D). Nous avons montré que la demi-vie de l’ARNm αENaC était de 100min, un temps beaucoup plus court que celui rapporté dans la littérature. Nous avons démontré que l’Act. D a un effet stabilisateur important sur l’ARNm αENaC et qu’il ne peut être utilisé pour évaluer la stabilité du transcrit. À l’aide de différents mutants de délétion, nous avons entrepris de déterminer la nature des régions du 3’UTR impliquées dans la modulation de la stabilité du transcrit. Nous avons trouvé que le 3’UTR joue un rôle à la fois de stabilisation (région 3’UTR proximale) et de déstabilisation (région 3’UTR distale) du transcrit. Notre système nous a finalement permis de confirmer que la diminution de l’ARNm αENaC observée en présence de TNF-α s’expliquait en partie par une diminution importante de la stabilité du transcrit induite par cette cytokine. Enfin, nous avons identifié la nature des protéines pouvant se lier au 3’UTR de l’ARNm αENaC et déterminé lesquelles pouvaient moduler la stabilité du transcrit. Des trois protéines candidates trouvées, nous avons confirmé que la surexpression de DHX36 et TIAL1 diminue le niveau de transcrit par un mécanisme impliquant la stabilité du messager. Les travaux présentés ici montrent la complexité des voies de signalisation induites par différents stress sur les cellules épithéliales alvéolaires et montrent comment la stabilité de l’ARNm αENaC et en particulier, les séquences du 3’UTR jouent un rôle important dans la modulation du niveau de transcrit. Le modèle Tet-Off que nous avons développé permet d’estimer le temps de demi-vie réel de l’ARNm αENaC et montre que le 3’UTR du messager joue un rôle complexe dans la stabilisation du messager en condition de base ainsi qu’en condition pro-inflammatoire. Enfin, nous avons identifié deux protéines liant l’ARNm qui pourraient jouer un rôle important dans la modulation de la stabilité du transcrit.

La Fibrosis Quística es la enfermedad autosómica recesiva mas frecuente en caucásicos. En Colombia no se conoce la incidencia de la enfermedad, pero investigaciones del grupo de la Universidad del Rosario indican que podría ser relativamente alta. Objetivo: Determinar la incidencia de afectados por Fibrosis Quística en una muestra de recién nacidos de la ciudad de Bogotá. Metodología: Se analizan 8.297 muestras de sangre de cordón umbilical y se comparan tres protocolos de tamizaje neonatal: TIR/TIR, TIR/DNA y TIR/DNA/TIR. Resultados: El presente trabajo muestra una incidencia de 1 en 8.297 afectados en la muestra analizada. Conclusiones: Dada la relativamente alta incidencia demostrada en Bogotá, se justifica la implementación de Tamizaje Neonatal para Fibrosis Quística en Colombia.

Previous studies have shown that "Mudanpi", a Chinese herbal medicine, has a significant cardioprotective effect against myocardial ischaemia. Based on these findings we hypothesised that paeonol, the main component of Mudanpi, might have an effect on the cellular electrophysiology of cardiac ventricular myocytes. The effects of paeonol on the action potential and ion channels of cardiac ventricular myocytes were studied using the standard whole-cell configuration of the patch-clamp technique. Ventricular myocytes were isolated from the hearts of adult guinea-pig by enzymic dispersion. The myocytes were continuously perfused with various experimental solutions at room temperature and paeonol applied in the perfusate. Action potentials and membrane currents were recorded using both current and voltage clamp modes of the patch-clamp technique. Paeonol, at concentrations 160 mu M and 640 mu M, decreased the action potential upstroke phase, an action associated with the blockade of the voltage-gated, fast sodium channel. The effects of paeonol on both action potential and Na+ current were concentration dependent. Paeonol had a high affinity for inactivated sodium channels. Paeonol also shortened the action potential duration, in a manner not associated with the blockade of the calcium current, or the enhancement of potassium currents. These findings suggest that paeonol, and therefore Mudanpi, may possess antiarrhythmic activity, which may confer its cardioprotective effects. (c) 2006 Elsevier B.V All rights reserved.

Human functional imaging provides a correlative picture of brain activity during pain. A particular set of central nervous system structures (eg, the anterior cingulate cortex, thalamus, and insula) consistently respond to transient nociceptive stimuli causing pain. Activation of this so-called pain matrix or pain signature has been related to perceived pain intensity, both within and between individuals,1,2 and is now considered a candidate biomarker for pain in medicolegal settings and a tool for drug discovery. The pain-specific interpretation of such functional magnetic resonance imaging (fMRI) responses, although logically flawed,3,4 remains pervasive. For example, a 2015 review states that “the most likely interpretation of activity in the pain matrix seems to be pain.”4 Demonstrating the nonspecificity of the pain matrix requires ruling out the presence of pain when highly salient sensory stimuli are presented. In this study, we administered noxious mechanical stimuli to individuals with congenital insensitivity to pain and sampled their brain activity with fMRI. Loss-of-function SCN9A mutations in these individuals abolishes sensory neuron sodium channel Nav1.7 activity, resulting in pain insensitivity through an impaired peripheral drive that leaves tactile percepts fully intact.5 This allows complete experimental disambiguation of sensory responses and painful sensations

In contrast to the many studies on the venoms of scorpions, spiders, snakes and cone snails, tip to now there has been no report of the proteomic analysis of sea anemones venoms. In this work we report for the first time the peptide mass fingerprint and some novel peptides in the neurotoxic fraction (Fr III) of the sea anemone Bunodosoma cangicum venom. Fr III is neurotoxic to crabs and was purified by rp-HPLC in a C-18 column, yielding 41 fractions. By checking their molecular masses by ESI-Q-Tof and MALDI-Tof MS we found 81 components ranging from near 250 amu to approximately 6000 amu. Some of the peptidic molecules were partially sequenced through the automated Edman technique. Three of them are peptides with near 4500 amu belonging to the class of the BcIV, BDS-I, BDS-II, APETx1, APETx2 and Am-II toxins. Another three peptides represent a novel group of toxins (similar to 3200 amu). A further three molecules (similar to similar to 4900 amu) belong to the group of type 1 sodium channel neurotoxins. When assayed over the crab leg nerve compound action potentials, one of the BcIV- and APETx-like peptides exhibits an action similar to the type 1 sodium channel toxins in this preparation, suggesting the same target in this assay. On the other hand one of the novel peptides, with 3176 amu, displayed an action similar to potassium channel blockage in this experiment. In summary, the proteomic analysis and mass fingerprint of fractions from sea anemone venoms through MS are valuable tools, allowing us to rapidly predict the occurrence of different groups of toxins and facilitating the search and characterization of novel molecules without the need of full characterization of individual components by broader assays and bioassay-guided purifications. Sodium channel toxins from sea anemones are employed as tools for dissecting the biophysical properties of inactivation in voltage-gated sodium channels. Cangitoxin (CGTX) is a peptide containing 48 amino acid residues and was formerly purified from Bunodosoma cangicum. Nevertheless, previous works reporting, the isolation procedures for such peptide from B. cangicum secretions are controversial and may lead to incorrect information. In this paper, we report a simple and rapid procedure, consisting of two chromatographic steps, in order to obtain a CGTX analog directly from sea anemone venom. We also report a substitution of N16D in this peptide sample and the co-elution of an inseparable minor isoform presenting the R14H substitution. Peptides are named as CGTX-II and CGTX-III, and their effects over Nav1.1 channels in patch clamp experiments are demonstrated. (c) 2008 Elsevier Ltd. All rights reserved.

To investigate the kdr (knockdown resistance) resistance-associated gene mutation and determine its frequency in pyrethroid-resistant horn fly (Haematobia irritans) populations, a total of 1,804 horn flies of 37 different populations from all Brazilian regions (North, Northeast, Central-West, Southeast, and South) were molecular screened through polymerase chain reaction (PCR). The kdr gene was not detected in 87.08% of the flies. However, the gene was amplified in 12.92% of the flies, of which 11.70% were resistant heterozygous and 1.22% were resistant homozygous. Deviation from Hardy-Weinberg equilibrium (HWE) was found only in 1 ranch with an excess of heterozygous. When populations were grouped by region, three metapopulations showed significant deviations of HWE (Central-West population, South population and Southeast population). This indicates that populations are isolated one from another and kdr occurrence seems to be an independent effect probably reflecting the insecticide strategy used by each ranch. Although resistance to pyrethroids is disseminated throughout Brazil, only 48% of resistant populations had kdr flies, and the frequency of kdr individuals in each of these resistant populations was quite low. But this study shows that, with the apparent exception of the Northeast region, the kdr mechanism associated with pyrethroid resistance occurs all over Brazil.

We have examined the effect of the uncharged species of lidocaine (LDC) and etidocaine (EDC) on the acyl chain moiety of egg phosphatidylcholine liposomes. Changes in membrane organization caused by both anesthetics were detected through the use of EPR spin labels (5, 7 and 12 doxyl stearic acid methyl ester) or fluorescence probes (4, 6, 10, 16 pyrene-fatty acids). The disturbance caused by the LA was greater when the probes were inserted in more external positions of the acyl chain and decreased towards the hydrophobic core of the membrane. The results indicate a preferential insertion of LDC at the polar interface of the bilayer and in the first half of the acyl chain, for EDC. Additionally, 2 H NMR spectra of multilamellar liposomes composed by acyl chain-perdeutero DMPC and EPC (1:4 mol%) allowed the determination of the segmental order (S-mol) and dynamics (T-1) of the acyl chain region. In accordance to the fluorescence and EPR results, changes in molecular orientation and dynamics are more prominent if the LA preferential location is more superficial, as for LDC while EDC seems to organize the acyl chain region between carbons 2-8, which is indicative of its positioning. We propose that the preferential location of LDC and EDC inside the bilayers creates a ""transient site"", which is related to the anesthetic potency since it could modulate the access of these molecules to their binding site(s) in the voltage-gated sodium channel.
