The upper 1200 m of pre-Pliocene sediment recovered by Cape Roberts Project (CRP) drilling off the Victoria Land coast of Antarctica between 1997-1999 has been subdivided into 54 unconformity-bound stratigraphic sequences, spanning the period c. 32 to 17 Ma. The sequences are recognised on the basis of the cyclical vertical stacking of their constituent lithofacies, which are enclosed by erosion surfaces produced during the grounding of the advancing ice margin onto the sea floor. Each sequence represents deposition in a range of offshore shelf to coastal glacimarine sedimentary environments during oscillations in the ice margin across the Western Ross Sea shelf, and coeval fluctuations in water depth. This paper applies spectral analysis techniques to depth- and time-series of sediment grain size (500 samples) for intervals of the core with adequate chronological data. Time series analysis of 0.5-1.0m-spaced grainsize data spanning sequences 9-11 (CRP-2/2A) and sequences 1-7 (CRP-3) suggests that the length of individual sequences correspond to Milankovitch frequencies, probably 41 k.y., but possibly as low as 100 k.y. Higher frequency periodic components at 23 k.y. (orbital precession) and 15-10 k.y. (sub-orbital) are recognised at the intrasequence-scale, and may represent climatic cycles akin to the ice rafting episodes described in the North Atlantic Ocean during the Quaternary. The cyclicity recorded by glacimarine sequences in CRP core provides direct evidence from the periphery of Antarctica for orbital oscillations in the size of the Oligocene-Early Miocene East Antarctic Ice Sheet.

Four temperature data-loggers were placed in each of five green sea turtle nests on Heron Island in the 1998-99 nesting season. Temperatures in all nests increased as incubation progressed due to general sand heating and increased metabolic heat production of the developing embryos. Even at the top of nests no daily diurnal fluctuation in temperature was evident. The temperature of eggs in the middle of the nest increased above those in the nest periphery during the last third of incubation. However, this metabolic nest heating would have little effect on hatchling sex ratio because it occurred after the sex-determining period. Small differences in temperature between regions of a nest persisted throughout incubation and may be important in ensuring the production of at least some individuals of the opposite sex in nests that have temperatures close to either the all-male or all-female determining temperatures. Location and degree of shading of nests had little effect on mean nest temperature, but deeper nests were generally cooler and therefore were predicted to produce a higher proportion of males than were shallower nests. Nest temperature profile data indicated that the 1998-99 nesting season on Heron Island would have produced a strongly female-biased sex ratio amongst hatchlings.

The intracellular assembly site for flaviviruses in currently not known but is presumed to be located within the lumen of the rough endoplasmic reticulum (RER), Building on previous studies involving immunofluorescence (IF) and cryoimmunoelectron microscopy of Kunjin virus (KUN)-infected cells, we sought to identify the steps involved in the assembly and maturation of KUN. Thus, using antibodies directed against envelope protein E in IF analysis, we found the accumulation of E within regions coincident with the RER and endosomal compartments. Immunogold labeling of cryosections of infected cells indicated that E and minor envelope protein prM were localized to reticulum membranes continuous with KUN-induced convoluted membranes (CM) or paracrystalline arrays (PC) and that sometimes the RER contained immunogold-labeled virus particles. Both proteins were also observed to be labeled in membranes at the periphery of the induced CIM or PC structures, but the latter were very seldom labeled internally. Utilizing drugs that inhibit protein and/or membrane traffic throughout the cell, we found that the secretion of KUN particles late in infection was significantly affected in the presence of brefeldin A and that the infectivity of secreted particles was severely affected in the presence of monensin and N-nonyl-deoxynojirimycin. Nocodazole did not appear to affect maturation, suggesting that microtubules play no role in assembly or maturation processes. Subsequently, we showed that the exit of intact virions from the RER involves the transport of individual virions within individual vesicles en route to the Golgi apparatus. The results suggest that the assembly of virions occurs within the lumen of the RER and that subsequent maturation occurs via the secretory pathway.

Selection in the thymus restricted by MHC and self-peptide shapes the diverse reactivities of the T-cell population which subsequently seeds into the peripheral tissues, in anticipation of the universe of pathogen antigens to which the organism may be exposed. A necessary corollary is the potential for T-cell self-reactivity (autoimmunity) in the periphery. Transgenic mouse models in which transgene expression in the thymus is prevented or excluded, have been particularly useful for determining the immunological outcome when T-cells encounter transgene-encoded 'self' antigen in peripheral tissues. Data suggest that non-mutually exclusive mechanisms of T-cells 'ignoring' self-antigen, T-cell deletion, T-cell anergy and T-cell immunoregulation have evolved to prevent self-reactivity while maintaining T-cell diversity. The peripheral T-cell repertoire, far from being static following maturation through the thymus, is in a dynamic stated determined by these peripheral selective and immunoregulatory influences. This article reviews the evidence with particular reference to CD8+ive T-cells.

Smooth muscle cells (SMC) exhibit a functional plasticity, modulating from the mature phenotype in which the primary function is contraction, to a less differentiated state with increased capacities for motility, protein synthesis, and proliferation. The present study determined, using Western analysis, double-label immunofluorescence and confocal microscopy, whether changes in phenotypic expression of rabbit aortic SMC in culture could be correlated with alterations in expression and distribution of structural proteins. Contractile state SMC (days 1 and 3 of primary culture) showed distinct sorting of proteins into subcellular domains, consistent with the theory that the SMC structural machinery is compartmentalised within the cell. Proteins specialised for contraction (alpha -SM actin, SM-MHC, and calponin) were highly expressed in these cells and concentrated in the upper central region of the cell. Vimentin was confined to the body of the cell, providing support for the contractile apparatus but not co-localising with it. In line with its role in cell attachment and motility, beta -NM actin was localised to the cell periphery and basal cortex. The dense body protein alpha -actinin was concentrated at the cell periphery, possibly stabilising both contractile and motile apparatus. Vinculin-containing focal adhesions were well developed, indicating the cells' strong adhesion to substrate. In synthetic state SMC (passages 2-3 of culture), there was decreased expression of contractile and adhesion (vinculin) proteins with a concomitant increase in cytoskeletal proteins (beta -non-muscle [NM] actin and vimentin). These quantitative changes in structural proteins were associated with dramatic chan-es in their distribution. The distinct compartmentalisation of structural proteins observed in contractile state SMC was no longer obvious, with proteins more evenly distributed throughout die cytoplasm to accommodate altered cell function. Thus, SMC phenotypic modulation involves not only quantitative changes in contractile and cytoskeletal proteins, but also reorganisation of these proteins. Since the cytoskeleton acts as a spatial regulator of intracellular signalling, reorganisation of the cytoskeleton may lead to realignment of signalling molecules, which, in turn, may mediate the changes in function associated with SMC phenotypic modulation. In the periphery, physiological dopamine increases renal blood flow, decreases renal resistance and acts on the kidney tubule to enhance natriuresis and diuresis. The loss of dopamine function may be involoved in the deterioration in kidney function associated with ageing and may have a role in the pathogenesis of hypertension and diabetes. Intravenous dopamine is used as a positive inotrope in the treatment of acute heart failure and cardiogenic shock and as a diuretic in renal failure. The clinical uses of dopamine are limited, as it must be given intravenously, and also has widespread effects. The levels of peripheral dopamine can be increased by the administration of L-dopa to increase synthesis, prodrugs to release dopamine (docarpamine, glu-dopa) or by inhibiting the breakdown of dopamine (nitecapone). Preliminary clinical trials suggest that docarpamine may be useful in patients with low cardiac output syndrome after cardiac surgery and in refractory cirrhotic ascites. Ibopamine is an agonist at dopamine D1 and D2 receptors, which may retard the progression of chronic renal failure. Gludopa is selective for the kidney thus avoiding widespread side effects. The early clinical studies with ibopamine as a diuretic in heart failure were favourable but the subsequent large mortality study showed that ibopamine increased mortality. Fenoldopam is a selective dopamine D1 receptor agonist. Intravenous fenoldopam may be useful in the treatment of hypertension associated with coronary artery bypass surgery or in hypertensive emergencies. Although our understanding of physiological and pathological roles of peripheral dopamine has been increasing rapidly in recent times, we still need more information to allow the design of clinically useful drugs that modify these roles. One priority is an orally-active selective dopamine D1 receptor agonist.

Early pregnancy factor (EPF) is a secreted protein, present in serum during early pregnancy and essential for maintaining viability of the embryo. It is a homologue of chaperonin 10 (Cpn10) but, unlike Cpn10, it has an extracellular role. EPF has immunosuppressive and growth regulatory properties. Previously we have reported the preparation of recombinant EPF (rEPF) and shown that treatment with rEPF will suppress clinical signs of MBP-EAE in Lewis rats and PLP-EAE in SJL/J mice. In the present study, these findings have been extended to investigate possible mechanisms involved in the action of EPF. Following treatment of mice with rEPF from the day of inoculation, there were fewer infiltrating CD3+ and CD4+ cells in the parenchyma of the spinal cord during the onset of disease and after the initial episode, compared with mice treated with vehicle. Expression of the integrins LFA-1, VLA-4 and Mac-1 and of members of the immunoglobulin superfamily of adhesion molecules ICAM-1 and VCAM-1 was suppressed in the central nervous system (CNS) following rEPF treatment. The expression of PECAM-1 was not affected. To determine if rEPF suppressed T cell activation in the periphery, the delayed-type hypersensitivity (DTH) reaction of normal BALB/c mice to trinitrochlorobenzene (TNCB) following treatment with rEPF was studied. The results showed that treatment with rEPF suppressed the DTH reaction, demonstrating the ability of EPF to downregulate the cell-mediated immune response. These results indicate that suppression of immunological mechanisms by rEPF plays a major role in the reduction of clinical signs of disease in experimental autoimmune encephalomyelitis (EAE). CD40 has emerged as a key signaling pathway for the function of B cells, monocytes, and dendritic cells (DC) in the immune system, and plays a major role in inflammatory pathways of nonhemopoletic cells. CD40 is expressed by monocytes and DC and is up-regulated when DC migrate from the periphery to draining lymph nodes (DLN) in response to microbial challenge. CD154 signaling by MHC-restricted, activated CD4* T cells induces differentiation of DC, as defined by an increased surface expression of MHC, costimulatory, and adhesion molecules. Thus, CD40 functions in the adaptive immune response as a trigger for the expression of costimulatory molecules for efficient T-cell activation. CD40 ligation of DC also has the capacity to induce high levels of the cytokine IL-12, which polarizes CD4(+) T cells toward a T helper 1 (Th1) type, enhances proliferation of CD8(+) T cells, and activates NK cells. CD40 may also play an important role in the decision between tolerance and immunity and the generation of regulatory CD4(+) T cells that are thought to maintain peripheral self-tolerance in vivo.

The work of Michel Foucault sees modern penal technology its ann expression of power that operates through and is motivated by a dry instrumental reason. This article draws upon Durkheim and Bakhtin to advance a radically alternative approach. It is suggested that such technology is invested with sacred and profane symbolism and is understood via emotionally charged, dramatically compelling narrative frames. Tensions between official and unauthorized discourses can be understood through a center/periphery model of culture. In an extended case study of the guillotine, it is shown dial the apparatus was initially legitimated as an expression of a sacred revolutionary code. Such a discourse was subsequently destabilized by popular medical debates that raised the specter of pain after decapitation. While inconclusive, these new motifs mobilized Gothic and grotesque themes that confronted the rationalist aesthetics of the guillotine. A situation of Bakhtinian hetoroglossia eventuated. Uncertainty, the uncanny and fable entered a discursive field of increasing complexity.

A tese afirma a vida por meio das biopotências que se manifestam em movimentações invisíveis aos olhos acostumados a permitir o ver, o julgar e o falar. Foca o tempo presente, na comunicação em redes, traz em si a potência de reatar a multidão, com a capacidade de sondar possibilidades mostrando o que antes parecia opaco e impossível. Perambular é ligar nas redes quentes de um bairro com/no/do território ao privilegiar o movimento, o processo, sempre caminhando pelas vias e conexões abertas, aposta estética-ética-política nos paradoxos sem superação e não hierarquizados. Toma como método de pesquisa-intervenção elementos de uma cartografia de movimentos e devires, traçando um perambular rizomático em que são problematizados a constituição do problema de pesquisa, que considera a construção do conhecimento diversificada, descentralizada e horizontalizada. Problematiza as práticas discursivas de si em suas relações com a biopolítica, a governabilidade e a biopolítica das populações. O que está(rá) rolando nesse bairro, no que foi chamado de criança, adolescente, escola, compor para que as coisas apareçam, junto com outros que vivem a loucura em duplas e trios. A tese é a possibilidade da existência de uma educação menor na periferia para as populações marginalizadas, muçulmanizadas. Educação menor, da sala de aula, do bairro, do cotidiano de professores, familiares e alunos. Educação que permite revolucionários, na medida em que alguma revolução ainda faz sentido na educação nesses dias. A educação menor constitui-se, assim, num empreendimento de militância, de professores militantes. Plano das afecções em que não há unidades, apenas intensidade. A tese fala do processo, de como reproduzir, ou não, os modos de subjetividade dominante, não se trata de medidas - "menor" ou "pequeno". Nesse sentido, é preciso considerar os efeitos de produção de subjetividade e a incorporação dos fatos à própria vida. A tese analisa movimentos instituintes buscando reconhecê-los em sua natureza contestatória e transgressora e ter conhecimento de como se organiza na escola e muito além dela. O trabalho buscou analisar questões de desigualdade regional no Espírito Santo através da linha de pesquisa denominada Nova Geografia Econômica (NGE). Uma forma de realizar essa análise é através do estudo da relação entre diferenciais de salário e mercado potencial. Mais precisamente, o trabalho procurou verificar o impacto de fatores geográficos de segunda natureza – mercado potencial – nos salário médios municipais. Inicialmente, por meio de uma Análise Exploratória de Dados Espaciais, verificou-se que os salários são maiores próximos às regiões com alto mercado potencial (litoral/RMGV). Por meio da utilização de técnicas de estatística e econometria espacial foi possível observar para os anos de 2000 e 2010 a existência de uma estrutura espacial de salários no Espírito Santo. O coeficiente de erro autorregressivo foi positivo e estatisticamente significativo, indicando o modelo SEM (spatial error model) como o mais apropriado para modelar os efeitos espaciais. Os resultados indicam ainda que não só fatores educacionais afetam os salários, fatores geográficos de segunda natureza possuem um efeito até maior quando comparados aos primeiros. Conclui-se, como demonstra o modelo central da NGE que, forças exclusivamente de mercado nem sempre levam ao equilíbrio equalizador dos rendimentos, pelo contrário, levam à conformação de uma estrutura do tipo centro-periferia com diferença persistente de rendimentos entre as regiões. Adicionalmente, verifica-se que os municípios que apresentam maior salário, maior mercado potencial e melhores indicadores sociais são àqueles localizados no litoral do estado, mais precisamente os municípios próximos à RMGV. Sendo assim, o trabalho reforça a necessidade de que se pense estratégias que fomentem a criação de novas centralidades no Espírito Santo, a fim de atuar na redução das desigualdades regionais. O trabalho se insere num grupo de vários outros estudos que analisaram questões de desigualdade e concentração produtiva no Espírito Santo. A contribuição está na utilização do referencial teórico da NGE, que ainda não havia sido empregada para o estado, e na utilização de técnicas de estatística espacial e econometria espacial.

A Política Nacional de Atenção Integral à Saúde do Homem propõe formas diferenciadas de atuação da equipe de saúde no atendimento da população masculina, uma vez que este público demanda estratégias diferenciadas de serviço. Ao se deparar com a realidade vivenciada pelos homens, surgem várias questões acerca do estereótipo social construído acerca das características masculinas e suas vivências. O objetivo da pesquisa foi compreender alguns aspectos relevantes para as práticas de saúde de homens usuários de Unidade de Saúde da Família, como qualidade de vida, consumo de álcool, representações sociais da bebida alcoólica e características de masculinidade. Foi utilizada uma amostra de 300 homens, frequentadores de Unidade de Saúde da Família, e aplicado um questionário contendo os dados sociodemográficos, o World Health Organization Quality of Life (Whoqol-bref), o Bem Sex-Role Inventory (BSRI), um exercício de evocação sobre bebida alcoólica, o Alcohol Use Disorders Identification Test (Audit) e um bloco para verificar os problemas ocasionados pelo consumo de álcool e a procura por tratamento. Os dados dos instrumentos quantitativos foram analisados com testes estatísticos de comparação de médias e de correlação. Os dados das evocações foram analisados com o software EVOC (Ensemble de Programmes Permettant l’Analyse des Évocations). Na primeira análise, constatou-se adesão mais alta a características femininas, alta percepção de qualidade de vida e padrões de consumo de álcool semelhantes às médias nacionais. Homens que declararam praticar sua religião apresentaram média significativamente menor de consumo de álcool. Apresentaram correlação inversamente proporcional ao consumo de álcool as características femininas de gênero, os domínios físico, social, psicológico e percepção global de qualidade de vida. Na análise das evocações, constatou-se que os elementos com tendência à centralidade são, em sua maioria, de cunho negativo. Os dados da população geral apresentaram o termo gosto como um aspecto positivo e central da bebida alcoólica. O grupo de abstinentes não apresentou avaliação positiva do termo e o grupo de bebedores apresentou o termo diversão na primeira periferia, referindo-se aos aspectos positivos e de socialização da bebida alcoólica. Os resultados indicaram uma qualidade de vida satisfatória, a religião e as características femininas destacaram-se como um fator de proteção ao uso de bebida alcoólica. Apresentaram, ainda, percepção dos problemas associados ao próprio consumo. Apesar de a maioria dos termos relacionados à bebida alcoólica ser negativo, este consumo ainda se dá em um nível considerável. Por isso, se faz necessária a construção de vínculo entre o profissional e o usuário do serviço de saúde a fim de dar oportunidade para que as reais práticas sobre a bebida alcoólica sejam evidenciadas. Esses dados podem ajudar profissionais de Saúde da Família a refletirem sobre as representações sociais que constroem acerca dos homens de classe popular usuários do serviço

The study intends to understand how countries from the periphery deal with new institutional challenges resulting from globalization, using the case of the Brazilian diplomatic service.

In the last fifty years, Brazil began a rapid process of structural transformation, following the first stage of industrial development in the 1930s. Currently the country integrates the small group of countries which evolved from an initial peripheral and subordinate insertion dating back to the nineteenth century, part of the most dynamic segment of the semiperiphery. But this category, intermediate between the "maturity" and "backwardness", according Modernization theorists, or between the "center" and "periphery", as theorists of the Dependence defend, has undergone a process of overcoming considerable positive progress in the direction of the group of states that dominate the current world system. In this way, during the years 2003-2010, foreign policy, along with the formulation of a new regionalism as a strategy of global integration and a new ideal model of State, has been a key factor
