Due to the effect of catabolite repression, sugar mixtures cannot be metabolized in a rapid and efficient way implicating in lower productivity in bioprocesses using lignocellulosic hydrolysates. In gram-negative bacteria, this mechanism is mediated by the phosphotransferase system (PTS), which concomitantly internalizes and phosphorylates sugars. In this study, we isolated a UV mutant of Burkholderia sacchari, called LFM828, which transports hexoses and pentoses by a non-PTS uptake system. This mutant presented released glucose catabolite repression over the pentoses. In mixtures of glucose, xylose, and arabinose, specific growth rates and the specific sugar consumption rates were, respectively, 10 and 23% higher in LFM828, resulting in a reduced time to exhaust all sugars in the medium. However, in polyhydroxybutyrate (PHB) biosynthesis experiments it was necessary the supplementation of yeast extract to maintain higher values of growth rate and sugar consumption rate. The deficient growth in mineral medium was partially recovered by replacing the ammonium nitrogen source by glutamate. It was demonstrated that the ammonium metabolism is not defective in LFM828, differently from ammonium, glutamate can also be used as carbon and energy allowing an improvement on the carbohydrates utilization for PHB production in LFM828. In contrast, higher rates of ammonia consumption and CO(2) production in LFM828 indicate altered fluxes through the central metabolism in LFM828 and the parental. In conclusion, PTS plays an important role in cell physiology and the elimination of its components has a significant impact on catabolite repression, carbon flux distribution, and PHB biosynthesis in B. sacchari.

Neutral trehalase from Neurospora crassa was expressed in Escherichia coli as a polypeptide of similar to 84 kDa in agreement with the theoretical size calculated from the corresponding cDNA. The recombinant neutral trehalase, purified by affinity chromatography exhibited a specific activity of 80-150 mU/mg protein. Optima of pH and temperature were 7.0 and 30 degrees C, respectively. The enzyme was absolutely specific for trehalose, and was quite sensitive to incubation at 40 degrees C. The recombinant enzyme was totally dependent on calcium, and was inhibited by ATP, copper, silver, aluminium and cobalt. K(M) was 42 mM, and V(max) was 30.6 nmol of glucose/min. The recombinant protein was phosphorylated by cAMP-dependent protein kinase, but not significantly activated. Immunoblotting with polyclonal antiserum prepared against the recombinant protein showed that neutral trehalase protein levels increased during exponential phase of N. crassa growth and dropped at the stationary phase. This is the first report of a neutral trehalase produced in E. coli with similar biochemical properties described for fungi native neutral trehalases, including calcium-dependence. The aim of this study was to evaluate the nitrogen topdressing influence in common bean irrigated (winter-spring), in the first year of no tillage implementation. The experimental design was split-plot with three replications in randomized blocks. The plots were formed by three types of ground cover, corn-grain, corn-grain intercropped with Brachiaria ruziziensis and only B. ruziziensis. The subplots were formed by five doses of nitrogen topdressing (0, 40, 80, 120 and 160 kg ha(-1)), using urea as nitrogen source. It was determined the dry mass of residue present as ground cover, the full flowering time, the leaf nitrogen content, the 100 grains mass, the grain yield, the processing income, the chemical soil properties and economic analysis of common bean grain yield. It was found the common bean grain productivity in succession to corn-grain was positively influenced by nitrogen fertilization, showing it is economically viable only when given 160 kg ha(-1) of nitrogen and the intercropping corn-grain and B. ruziziensis use is the best option when the common bean is sown in succession.

Based on the genetic analysis of the phytopathogen Xylella fastidiosa genome, five media with defined composition were developed and the growth abilities of this fastidious prokaryote were evaluated in liquid media and on solid plates. All media had a common salt composition and included the same amounts of glucose and vitamins but differed in their amino acid content. XDM1 medium contained amino acids threonine, serine, glycine, alanine, aspartic acid and glutamic acid, for which complete degradation pathways occur in X fastidiosa; XDM2 included serine and methionine, amino acids for which biosynthetic enzymes are absent, plus asparagine and glutamine, which are abundant in the xylem sap; XDM3 had the same composition as XDM2 but with asparagine replaced by aspartic acid due to the presence of complete degradation pathway for aspartic acid; XDM4 was a minimal medium with glutamine as a sole nitrogen source; XDM5 had the same composition as XDM4, plus methionine. The liquid and solidified XDM2 and XDM3 media were the most effective for the growth of X. fastidiosa. This work opens the opportunity for the in silico design of bacterial defined media once their genome is sequenced. (C) 2002 Federation of European Microbiological Societies. Quarenta bovinos machos, com 30 meses de idade e 371 kg, foram distribuídos em um delineamento em blocos casualizado com esquema fatorial 3 x 3, para avaliar o desempenho e rendimento de carcaça quando alimentados com diferentes fontes protéicas (Amiferm, uréia e farelo de soja) e volumosos (pastagem de capim-braquiária, cana-de-açúcar e silagem de milho). As dietas foram balanceadas para conterem níveis semelhantes de em e PB. A interação volumoso x fonte de N não foi significativa para nenhum dos parâmetros estudados. O ganho de peso vivo diário (GPV/dia) dos animais alimentados com cana-de-açúcar (0,83 kg) não diferiu dos mantidos em pastagem (0,82 kg), mas ambos foram menores que dos animais alimentados com silagem de milho (1,09 kg). As diferentes fontes de N proporcionaram GPV/dia semelhantes, com valores de 0,94; 0,83 e 0,97 kg, para os animais que receberam uréia, Amiferm e farelo de soja, respectivamente. As dietas contendo farelo de soja proporcionaram maior ganho de carcaça diário (0,57 kg) em relação ao uso de Amiferm, não diferindo da uréia (0,55 kg). Não houve diferença entre volumosos e fontes nitrogenadas para rendimento de carcaça, rendimento de carcaça do corpo vazio, rendimento de carcaça do ganho de peso e espessura de gordura, com média de 51,03%, 54,49%, 60,10% e 8,5 mm, respectivamente. O uso de Amiferm proporcionou ganhos de peso e rendimento de carcaça semelhantes às demais fontes protéicas.

O objetivo do experimento foi avaliar os efeitos da utilização do farelo de soja ou da glutenose sobre a degradabilidade ruminal in situ dos constituintes de dietas compostas por feno, milho e as fontes nitrogenadas fornecidas para bovinos adultos com 650 kg de peso. A glutenose foi considerada fonte nitrogenada de baixa degradabilidade e o farelo de soja, de média degradabilidade ruminal. As degradabilidades potenciais obtidas para o nitrogênio foram 39,7 e 90,3% e as degradabilidades efetivas 35,7 e 62,9%, respectivamente, para glutenose e farelo de soja. Utilizou-se a fibra mordente para determinar a taxa de passagem das dietas e obtiveram-se 6,6%/h para dietas com farelo de soja e 2,4%/h para dietas com glutenose. Não houve diferença na degradabilidade potencial da matéria seca do feno, nas frações A, B, C e na taxa de degradação, mas a degradabilidade efetiva da matéria seca foi inferior para as dietas com farelo de soja (29,3 vs 40,6%). Os demais ingredientes, à exceção do milho, apresentaram acentuadas diferenças entre as dietas nas variáveis de degradabilidade, na taxa de degradação e na degradabilidade efetiva do nitrogênio, com inferioridade na dieta com glutenose. O milho diferiu apenas quanto à degradabilidade efetiva do nitrogênio, superior na dieta com glutenose (68,7 vs 56,3%). O amido do milho apresentou menor degradabilidade efetiva na dieta com farelo de soja (52,5 vs 68,0%), sendo a degradabilidade efetiva da fibra do feno também inferior para essas dietas (27,3 vs 43,2%). Concluiu-se que fontes nitrogenadas com distintas degradabilidades podem afetar tanto a degradabilidade da fibra do volumoso quanto à do amido da própria fonte nitrogenada e do milho das dietas.

Os objetivos deste trabalho foram avaliar a produção de alface em ambiente protegido, utilizando diferentes compostos orgânicos como fonte de nitrogênio, e seu efeito residual em dois ciclos sucessivos. O experimento foi conduzido entre os meses de fevereiro e maio de 2005, em casa-de-vegetação com ventilação e temperatura controlados, utilizando vasos de 3,5 L e solo classificado como Latossolo Vermelho Escuro. Foram utilizados compostos orgânicos produzidos a partir da mistura de resíduos do processamento de quatro plantas medicinais e esterco bovino. Os compostos C1, C2, C3 e C4 nas doses 30, 60, 90 e 120 t ha-1 constituíram os tratamentos dispostos em delineamento inteiramente casualizado, com quatro repetições e uma testemunha (adicional). Foram determinados: matéria fresca da parte aérea (MFPA); massa de matéria seca da parte aérea (MSPA). Os compostos aplicados supriram satisfatoriamente as necessidades de nitrogênio da cultura, no primeiro ciclo, dispensando o uso de fertilizante mineral. A composição dos materiais aplicados influenciou significativamente a produção de alface no primeiro ciclo, promovendo efeito residual no segundo ciclo, porém em menores proporções.

The common bean is a leguminous of great importance in the Brazilian economy and nitrogen is the taken up nutrient in larger amount. Nitrogen fertilization management is of extreme importance to offer larger economical viability, besides increasing the efficiency of plant in the use of the available resources. The objective this study was to evaluate the effect of sources and doses of sidedressing nitrogen in the development and yield of winter common bean in no tillage system, as well as evaluate its economical viability. A randomized blocks design was used, in a factorial scheme 3x3 with 9 treatments constituted by three sources of nitrogen (ammonium sulphate, urea and ammonium sulphate 1/2 of N + urea 1/2 of N, applied at V(4-3) stadium) and different doses of sidedressing nitrogen (0, 40, 80 kg ha(-1)) in four replications. The study was conducted in Selviria county, MS State in 2004 in no season crop period, in a dystrophic Haplustox soil. The conclusion: independent of nitrogen source, nitrogen fertilization increasing provides increment in yield of winter common bean up to dose 80 kg/ha, and this provides, on average, an increase of 20% in yield compared with control (without sidedressing nitrogen). The urea is the nitrogen source of larger economical efficiency.

Realizou-se a análise comparativa de custos de produção de feijão, após um ano em sistemas de plantio direto e convencional, no período de inverno, com irrigação por pivô central. Os experimentos foram desenvolvidos na Fazenda Dois Irmãos, em Pereira Barreto (SP), em delineamento experimental de blocos ao acaso, com 11 tratamentos e quatro repetições, para cada sistema de plantio. Os tratamentos constaram de uma testemunha sem N, duas fontes de N (uréia e nitrato de amônio) e cinco dosagens (20, 40, 60, 80 e 100 kg.ha-1 de N), aplicados em cobertura aos 27 dias após a emergência das plantas. Utilizou-se densidade de 15 plantas por metro de linha e 0,50 m entre sulcos, com semeadura realizada em 20/5/2001. Na área do sistema de plantio direto, as plantas daninhas foram dessecadas após o cultivo do milho, e na de plantio convencional, realizaram uma aração e duas gradagens, sendo uma pesada e uma niveladora, no preparo do solo. Durante o desenvolvimento da cultura foram aplicados herbicidas em pós-emergência para o controle do mato. As duas fontes de nitrogênio mostraram efeito positivo na produção de grãos. A análise econômica revelou melhor resultado para uréia, na dose de 60 kg.ha-1 de N, em preparo convencional do solo, com índice de lucratividade de 40,6%. Para o plantio direto, o nitrato de amônio, na base de 40 kg.ha-1 de N, mostrou maior acréscimo na receita e lucratividade de 25,6%.

This paper presents the results from study investigating the capacity of Pseudomonas aeruginosa LBI growing on several carbon (Hydrophilic substrate) and nitrogen sources. The carbon source (2016) studied were: glycerol, manitol, fructose, glucose, lactose and the nitrogen sources (2; 3; 4 and 5016) were: NaNO3, NH4NO3, (NH4)(2)SO4 and (NH2)(2)CO. At the 96 h of fermentation, the medium with glycerol as a carbon source reached 7.9 g/L of rhamnolipids and 1. 2 g/L cellular protein. The surface tension reduction was 38.46 % for glycerol as a carbon source. The NaNO3 at 3% was the best concentration for rhamnolipids production (7.35 g/L) and cellular protein (1.12 g/L). The influences of metal ions [FeSO4.7H(2)O (0.001g/L, 0.005 g/L and 0.1 g/L) and MgSO4.7H(2)O (0.001 g/L, 0.005 g/L and 0.1 g/L)] on ramnolipids production were studied. Fe2+ had a negative influence on the studied concentrations while Mg2+ had a positive influence when its concentration was increased.

Penicillium citrinum grown in orange juice processing wastes medium under continuous agitation was studied in order to establish optimal conditions (incubation period, incubation temperature, initial pH and nitrogen addition) for biomass and ribonuclease production, as well as, biological depuration of the wastes. Nitrogen addition to wastes medium increased the biomass and ribonuclease production and provides COD reduction. The soy meal shows to be the best nitrogen source. The conditions for a more favorable enzyme and biomass production and COD reduction were initial pH 6.0 and temperature of 27°C. The maximum value obtained for these parameters on optimal conditions of cultivation was 11 U/mL of enzyme, 4 mg/mL of biomass (dry matter) and 55% of COD reduction, in 96 hours of incubation.
