Despite large changes in salt intake, the mammalian kidney is able to maintain the extracellular sodium concentration and osmolarity within very narrow margins, thereby controlling blood volume and blood pressure. In the aldosterone-sensitive distal nephron (ASDN), aldosterone tightly controls the activities of epithelial sodium channel (ENaC) and Na,K-ATPase, the two limiting factors in establishing transepithelial sodium transport. It has been proposed that the ENaC/degenerin gene family is restricted to Metazoans, whereas the α- and β-subunits of Na,K-ATPase have homologous genes in prokaryotes. This raises the question of the emergence of osmolarity control. By exploring recent genomic data of diverse organisms, we found that: 1) ENaC/degenerin exists in all of the Metazoans screened, including nonbilaterians and, by extension, was already present in ancestors of Metazoa; 2) ENaC/degenerin is also present in Naegleria gruberi, an eukaryotic microbe, consistent with either a vertical inheritance from the last common ancestor of Eukaryotes or a lateral transfer between Naegleria and Metazoan ancestors; and 3) The Na,K-ATPase β-subunit is restricted to Holozoa, the taxon that includes animals and their closest single-cell relatives. Since the β-subunit of Na,K-ATPase plays a key role in targeting the α-subunit to the plasma membrane and has an additional function in the formation of cell junctions, we propose that the emergence of Na,K-ATPase, together with ENaC/degenerin, is linked to the development of multicellularity in the Metazoan kingdom. The establishment of multicellularity and the associated extracellular compartment ("internal milieu") precedes the emergence of other key elements of the aldosterone signaling pathway.

Diplomityössä tutkittiin otsonin käytön soveltuvuutta ja etsittiin optimaalista soveltamistapaa kartonki- ja paperikoneen kiertoveden mikrobiologisen tilan hallintaan. Kirjallisuusosassa selvitettiin otsonin tuotannon nykytekniikkaa sekä otettiin selvää saatavilla olevista kapasiteetiltaan riittävistä otsonigeneraattoreista. Kirjallisuuden perusteella selvitettiin myös otsonin reaktioita paperi- ja kartonkikoneen kiertovesissä olevien orgaanisten ja epäorgaanisten aineiden kanssa. Lisäksi käsiteltiin otsonin käytön eri sovelluksia vedenpuhdistuksessa sekä perehdyttiin aikaisemmin tehtyihin tutkimuksiin otsonin käytöstä mikrobitorjuntaan paperi- ja kartonkikoneiden kiertovesistä. Kokeellisessa osassa määritettiin sopiva otsoniannos valkaisematonta sellua käyttävän koneen kiekkosuotimen kirkkaalle ja samealle suodokselle. Vesien otsonointi suoritettiin otsonipilotlaitteistolla. Työssä tutkittiin otsonoinnin vaikutusta kiertoveden mikrobiologisiin ja kemiallisiin ominaisuuksiin. Lisäksi tarkasteltiin kiertoveden otsonoinnin vaikutuksia koepaperikoneella valkaisemattomasta sel-lusta ja hylkymassasta valmistettujen lopputuotteiden laatuun. Työssä arvioitiin myös otsonin käytön mahdollisuutta osittain korvaamaan jo käytössä olevia hapettavia biosidejä. Samealle suodokselle sopivaksi otsoniannokseksi osoittautui 120 mg O3/l (0,3 mg O3/mg CODCr) ja kirkkaalle suodokselle riittävä annos olisi 60 mg O3/l (0,2 mg/mg CODCr). Näillä annoksilla kiertoveden mikrobireduktiot olivat 98–100 %. Kiertoveden hapetuskyky kasvoi 9-14 % ja pH laski yhdellä yksiköllä otsonoinnin vaikutuksesta. Kiertoveden kemiallisiin ominaisuuksiin otsonoinnilla oli edullinen vaikutus, sillä veden sameus, kiintoaine- ja CODCr-pitoisuudet laskivat. Koepaperikoneella havaittiin perälaatikon ilmapitoisuuden vähentyneen jopa 49 % käytettäessä ot-sonoitua kiertovettä. Mikrobiologiset analyysit suoritettiin koepaperikoneen massalle, viiravedelle ja lopputuotteelle. Tutkimusten perusteella mikrobientorjunnan kannalta parhaimmaksi otsonin annos-telupaikaksi osoittautui samea suodos. Massan, viiraveden ja lopputuotteen mikrobimäärät alenivat selvästi. Kemialliset analyysit osoittivat, että kirkasta suodosta käytettäessä oli koepaperikoneen vii-raveden kemiallinen tila parantunut LK-aineen muuttuessa hydrofiilisemmäksi ja sen sitoutumisky-vyn parantuessa. Samealla suodoksella ei muutosta havaittu. Valkaisemattomasta sellusta ja päällystetystä hylystä koepaperikoneella valmistettujen lopputuotteiden lujuus- ja aistinvaraisissa ominaisuuksissa ei tapahtunut merkittävää muutosta. Tulosten perusteella otsonia voisi käyttää korvaamaan hinnaltaan kalliimpia hapettavia biosidejä. Otsonin etuna on, ettei sille resistenttejä mikrobikantoja ole. Kirjallisuuden perusteella otsonin heikkoutena on sen tehon paikallisuus eli otsonin lisänä on käytettävä jotain muuta sekundääristä biosidia ylläpitämään matala mikrobitaso otsonoinnin jälkeen.

Kirjallisuusosassa käsitellään paperi- ja kartonkikoneiden eri vesijärjestelmiä, vedenkäytön vähentämistä ja sen vaikutusta prosessiin. Mikrobiologiaa, mikrobien aiheuttamia prosessiongelmia, mikrobien torjuntaa ja simulointia on myös käsitelty kirjallisuusosassa. Kokeellisessa osassa laadittiin Kartonkikone 1:n massa- ja vesijärjestelmän mikrobiologisen puhtauden simulointimalli Balas-prosessisimulointiohjelmalla. Kartongin mikrobiologinen puhtaus määritettiin kokonaispesäkelukuna. Kokeellinen osa sisälsi pilot-mittakaavan suodatinkoeajoja, joissa määritettiin tarvittavat erotusparametrit simulointimallille, selvitettiin Dynasand-hiekkasuodattimen soveltuvuus suihkuvesien puhdistukseen, Dynadisc-kiekkosuodattimen käyttökelpoisuus kiertoveden puhdistukseen kiertovesisuodattimena ja hiekkasuodattimien esisuodattimena sekä UV-reaktorin soveltuvuus suodatettujen vesien desifiointiin. Simulointimallilla tarkasteltiin eri prosessivaihtoehtojen vaikutus kartongin mikrobiologiseen puhtauteen. Suihkuvesien puhdistaminen hiekkasuodattimilla alentaa kokonaispesäkelukua vain vähän. Koneen ajettavuus paranee suihkuputkien roskaryöppyjen ja viiraosan limoittumisen vähenemisen takia. Kiertovesijärjestelmän pelkkä jakaminen hylkyprosessi- ja sakeudensäätövedeksi aiheuttaa kokonaispesäkeluvun kasvun prosessista poistettavien vesien puhdistumisen takia. Kokonaispesäkeluku minimoidaan poistamalla ylimääräinen vesi hylkysaostajan suodoksena. Suodatettua hylkysaostajan suodoksen ja viiraveden seosta käytetään sekä sakeudensäätö- että hylkyprosessivedeksi. Hylkymassalla on dominoiva vaikutus kokonaispesäkelukuun. Suurin vähennys kokonaispesäkeluvussa saadaan estämällä mikrobikasvu hylkymassassa. Tämä voidaan tehdä kuumentamalla hylkymassa 80 °C:n höyryllä. Suodatuskoeajojen perusteella Dynasand-hiekkasuodattimella voidaan puhdistaa pisaralämmönvaihtimelta saatavaa lämmintä suihkuvettä. Esisuodatettua kiertovettä voidaan puhdistaa Dynasand-hiekkasuodattimella viiran johtotelojen suihkuvedeksi. Dynadisc-kiekkosuodatinta voidaan käyttää hiekkasuodattimen esisuodattimena ja kiertovesisuodattimena. UV-säteily poistaa tehokkaasti mikrobeja suodatetuista vesistä.

Les bactéries du genre Pseudomonas ont la capacité étonnante de s'adapter à différents habitats et d'y survivre, ce qui leur a permis de conquérir un large éventail de niches écologiques et d'interagir avec différents organismes hôte. Les espèces du groupe Pseudomonas fluorescens peuvent être facilement isolées de la rhizosphère et sont communément connues comme des Pseudomonas bénéfiques pour les plantes. Elles sont capables d'induire la résistance systémique des plantes, d'induire leur croissance et de contrer des phytopathogènes du sol. Un sous-groupe de ces Pseudomonas a de plus développé la capacité d'infecter et de tuer certaines espèces d'insectes. Approfondir les connaissances sur l'interaction de ces bactéries avec les insectes pourraient conduire au développement de nouveaux biopesticides pour la protection des cultures. Le but de cette thèse est donc de mieux comprendre la base moléculaire, l'évolution et la régulation de la pathogénicité des Pseudomonas plante-bénéfiques envers les insectes. Plus spécifiquement, ce travail a été orienté sur l'étude de la production de la toxine insecticide appelée Fit et sur l'indentification d'autres facteurs de virulence participant à la toxicité de la bactérie envers les insectes. Dans la première partie de ce travail, la régulation de la production de la toxine Fit a été évaluée par microscopie à épifluorescence en utilisant des souches rapportrices de Pseudomonas protegens CHA0 qui expriment la toxine insecticide fusionnée à une protéine fluorescente rouge, au site natif du gène de la toxine. Celle-ci a été détectée uniquement dans l'hémolymphe des insectes et pas sur les racines des plantes, ni dans les milieux de laboratoire standards, indiquant une production dépendante de l'hôte. L'activation de la production de la toxine est contrôlée par trois protéines régulatrices dont l'histidine kinase FitF, essentielle pour un contrôle précis de l'expression et possédant un domaine "senseur" similaire à celui de la kinase DctB qui régule l'absorption de carbone chez les Protéobactéries. Il est donc probable que, durant l'évolution de FitF, un réarrangement de ce domaine "senseur" largement répandu ait contribué à une production hôte-spécifique de la toxine. Les résultats de cette étude suggèrent aussi que l'expression de la toxine Fit est plutôt réprimée en présence de composés dérivés des plantes qu'induite par la perception d'un signal d'insecte spécifique. Dans la deuxième partie de ce travail, des souches mutantes ciblant des facteurs de virulence importants identifiés dans des pathogènes connus ont été générées, dans le but d'identifier ceux avec une virulence envers les insectes atténuée. Les résultats ont suggéré que l'antigène O du lipopolysaccharide (LPS) et le système régulateur à deux composantes PhoP/PhoQ contribuent significativement à la virulence de P. protegens CHA0. La base génétique de la biosynthèse de l'antigène O dans les Pseudomonas plante-bénéfiques et avec une activité insecticide a été élucidée et a révélé des différences considérables entre les lignées suite à des pertes de gènes ou des acquisitions de gènes par transfert horizontal durant l'évolution de certaines souches. Les chaînes latérales du LPS ont été montrées comme vitales pour une infection des insectes réussie par la souche CHA0, après ingestion ou injection. Les Pseudomonas plante-bénéfiques, avec une activité insecticide sont naturellement résistants à la polymyxine B, un peptide antimicrobien modèle. La protection contre ce composé antimicrobien particulier dépend de la présence de l'antigène O et de la modification du lipide A, une partie du LPS, avec du 4-aminoarabinose. Comme les peptides antimicrobiens cationiques jouent un rôle important dans le système immunitaire des insectes, l'antigène O pourrait être important chez les Pseudomonas insecticides pour surmonter les mécanismes de défense de l'hôte. Le système PhoP/PhoQ, connu pour contrôler les modifications du lipide A chez plusieurs bactéries pathogènes, a été identifié chez Pseudomonas chlororaphis PCL1391 et P. protegens CHA0. Pour l'instant, il n'y a pas d'évidence que des modifications du lipide A contribuent à la pathogénicité de cette bactérie envers les insectes. Cependant, le senseur-kinase PhoQ est requis pour une virulence optimale de la souche CHA0, ce qui suggère qu'il régule aussi l'expression des facteurs de virulence de cette bactérie. Les découvertes de cette thèse démontrent que certains Pseudomonas associés aux plantes sont de véritables pathogènes d'insectes et donnent quelques indices sur l'évolution de ces microbes pour survivre dans l'insecte-hôte et éventuellement le tuer. Les résultats suggèrent également qu'une recherche plus approfondie est nécessaire pour comprendre comment ces bactéries sont capables de contourner ou surmonter la réponse immunitaire de l'hôte et de briser les barrières physiques pour envahir l'insecte lors d'une infection orale. Pour cela, les futures études ne devraient pas uniquement se concentrer sur le côté bactérien de l'interaction hôte-microbe, mais aussi étudier l'infection du point de vue de l'hôte. Les connaissances gagnées sur la pathogénicité envers les insectes des Pseudomonas plante-bénéfiques donnent un espoir pour une future application en agriculture, pour protéger les plantes, non seulement contre les maladies, mais aussi contre les insectes ravageurs. -- Pseudomonas bacteria have the astonishing ability to survive within and adapt to different habitats, which has allowed them to conquer a wide range of ecological niches and to interact with different host organisms. Species of the Pseudomonas fluorescens group can readily be isolated from plant roots and are commonly known as plant-beneficial pseudomonads. They are capable of promoting plant growth, inducing systemic resistance in the plant host and antagonizing soil-borne phytopathogens. A defined subgroup of these pseudomonads evolved in addition the ability to infect and kill certain insect species. Profound knowledge about the interaction of these particular bacteria with insects could lead to the development of novel biopesticides for crop protection. This thesis thus aimed at a better understanding of the molecular basis, evolution and regulation of insect pathogenicity in plant-beneficial pseudomonads. More specifically, it was outlined to investigate the production of an insecticidal toxin termed Fit and to identify additional factors contributing to the entomopathogenicity of the bacteria. In the first part of this work, the regulation of Fit toxin production was probed by epifluorescence microscopy using reporter strains of Pseudomonas protegens CHAO that express a fusion between the insecticidal toxin and a red fluorescent protein in place of the native toxin gene. The bacterium was found to express its insecticidal toxin only in insect hemolymph but not on plant roots or in common laboratory media. The host-dependent activation of Fit toxin production is controlled by three local regulatory proteins. The histidine kinase of this regulatory system, FitF, is essential for the tight control of toxin expression and shares a sensing domain with DctB, a sensor kinase regulating carbon uptake in Proteobacteria. It is therefore likely that shuffling of a ubiquitous sensor domain during the evolution of FitF contributed to host- specific production of the Fit toxin. Findings of this study additionally suggest that host-specific expression of the Fit toxin is mainly achieved by repression in the presence of plant-derived compounds rather than by induction upon perceiving an insect-specific signal molecule. In the second part of this thesis, mutant strains were generated that lack factors previously shown to be important for virulence in prominent pathogens. A screening for attenuation in insect virulence suggested that lipopolysaccharide (LPS) O-antigen and the PhoP-PhoQ two-component regulatory system significantly contribute to virulence of P. protegens CHAO. The genetic basis of O-antigen biosynthesis in plant-beneficial pseudomonads displaying insect pathogenicity was elucidated and revealed extensive differences between lineages due to reduction and horizontal acquisition of gene clusters during the evolution of several strains. Specific 0 side chains of LPS were found to be vital for strain CHAO to successfully infect insects by ingestion or upon injection. Insecticidal pseudomonads with plant-beneficial properties were observed to be naturally resistant to polymyxin B, a model antimicrobial peptide. Protection against this particular antimicrobial compound was dependent on the presence of O-antigen and modification of the lipid A portion of LPS with 4-aminoarabinose. Since cationic antimicrobial peptides play a major role in the immune system of insects, O-antigenic polysaccharides could be important for insecticidal pseudomonads to overcome host defense mechanisms. The PhoP-PhoQ system, which is well-known to control lipid A modifications in several pathogenic bacteria, was identified in Pseudomonas chlororaphis PCL1391 and P. protegens CHAO. No evidence was found so far that lipid A modifications contribute to insect pathogenicity in this bacterium. However, the sensor kinase PhoQ was required for full virulence of strain CHAO suggesting that it additionally regulates the expression of virulence factors in this bacterium. The findings of this thesis demonstrate that certain plant-associated pseudomonads are true insect pathogens and give some insights into how these microbes evolved to survive within and eventually kill the insect host. Results however also point out that more in-depth research is needed to know how exactly these fascinating bacteria manage to bypass or overcome host immune responses and to breach physical barriers to invade insects upon oral infection. To achieve this, future studies should not only focus on the bacterial side of the microbe-host interactions but also investigate the infection from a host-oriented view. The knowledge gained about the entomopathogenicity of plant-beneficial pseudomonads gives hope for their future application in agriculture to protect plants not only against plant diseases but also against insect pests.

Intestinal helminths are potent regulators of their host's immune system and can ameliorate inflammatory diseases such as allergic asthma. In the present study we have assessed whether this anti-inflammatory activity was purely intrinsic to helminths, or whether it also involved crosstalk with the local microbiota. We report that chronic infection with the murine helminth Heligmosomoides polygyrus bakeri (Hpb) altered the intestinal habitat, allowing increased short chain fatty acid (SCFA) production. Transfer of the Hpb-modified microbiota alone was sufficient to mediate protection against allergic asthma. The helminth-induced anti-inflammatory cytokine secretion and regulatory T cell suppressor activity that mediated the protection required the G protein-coupled receptor (GPR)-41. A similar alteration in the metabolic potential of intestinal bacterial communities was observed with diverse parasitic and host species, suggesting that this represents an evolutionary conserved mechanism of host-microbe-helminth interactions.

Gram-negative bacteria represent a major group of pathogens that infect all eukaryotes from plants to mammals. Gram-negative microbe-associated molecular patterns include lipopolysaccharides and peptidoglycans, major immunostimulatory determinants across phyla. Recent advances have furthered our understanding of Gram-negative detection beyond the well-defined pattern recognition receptors such as TLR4. A B-type lectin receptor for LPS and Lysine-motif containing receptors for peptidoglycans were recently added to the plant arsenal. Caspases join the ranks of mammalian cytosolic immune detectors by binding LPS, and make TLR4 redundant for septic shock. Fascinating bacterial evasion mechanisms lure the host into tolerance or promote inter-bacterial competition. Our review aims to cover recent advances on bacterial messages and host decoding systems across phyla, and highlight evolutionarily recurrent strategies.

The aim of this study was to examine suitability of BimTwin cleaning concept in card board machine to control microbiological activity and describe microbiological balance of the machine. In a review of literature is examined microbe and spore caused problems in paper industry. Biggest problems are deposits, which decrease runnability and cause quality errors. In this chapter is also introduced most common oxidizing biocides used in paper industry and described ATP assay as a microbiological monitoring method. In an experimental part are included BimTwin mill trial results, chemical condition monitoring methods and microbiological balance in a card board machine. In a second part are examined possibilities to effect hygiene of card board by chemical treatment of the surface size and coating. Result showed that BimTwin concept is suitable for card board machine as a cleaning concept, when chemical dosing is fitted right. For proper dosing and secure tolerable hygiene level, chemical and microbiological monitoring is significant. Determining of the microbiological balance would have need more sampling. According to acquired results, broke turned out to be the biggest microbe source. Sizing and coating experiments showed that it is possible to improve hygiene of card board by chemically treated surface size and coating color.

Erilaiset sterilointitarpeet ovat olennainen osa biotekniikkaa. Steriloinnit ovat huomattava kuluerä energia-, materiaali- ja laitekäyttöjen takia ja puutteellinen sterilointi voi aiheuttaa huomattavia ylimääräisiä kustannuksia. Eniten teollisesti käytettäviä sterilointimenetelmiä ovat steriilisuodatus, lämpösterilointi, kemiallinen sterilointi ja säteilysterilointi. Näistä lämpösterilointi on selvästi käytetyin menetelmä. Kirjallisuusosassa käsitellään lämpösteriloinnin kinetiikka ja sterilointityön laskentaan käytetyt menetelmät. Lämpösteriloinnin kinetiikka on mikrobista ja sen olomuodosta riippuvainen ja noudattaa Arrheniuksen lakia. Kirjallisuusosassa käsitellään myös lämpösterilointiin käytettyjä laitteistoja. Soveltavassa osassa käsitellään panos- ja jatkuvatoimia lämpösterilointeja. Työssä käydään läpi glukoosisiirapin sterilointi vastaavan kapasiteetin panos- ja jatkuvatoimisella sterilointilaitteistolla ja vertaillaan tarvittavia energia- ja hyödykemääriä. Soveltavan osan tietojen perusteella tehdään vertailu eri lämpösterilointimenetelmien kustannuksista sekä muista hyvistä ja huonoista puolista.

Työssä selvitettiin uudenaikaisten polymeerikalvojen soveltuvuutta maidon sekä heran mikrosuodatukseen. Kirjallisessa osassa perehdyttiin maidon ja heran koostumuksiin, mikrobikantoihin sekä erilaisiin maidon sekä heran rasvanpoisto- ja puhdistusmenetelmiin. Tämän lisäksi käsiteltiin mikrosuodatusmenetelmiä sekä niiden käyttöä heran ja maidon käsittelyssä meijeriteollisuudessa. Kokeellisessa osassa verrattiin kahdella eri suodatuslaitteistolla keraami- ja polymeerikalvojen suodatusominaisuuksia separoidun raakamaidon sekä heran poikkivirtausmikrosuodatuksessa. Tavoitteena oli erottaa suodatettavista liuoksista mikrobit sekä jäännösrasva ja tuottaa puhdasta, proteiinipitoista maitoa / heraa. Koeajoissa otetuista näytteistä analysoitiin ainesosakoostumus sekä mikrobipitoisuudet. Tuloksista määritettiin retentioarvot ja näiden perusteella vertailtiin polymeeri- ja keraamikalvojen erotustehokkuutta. Lisäksi kalvojen toimivuutta arvioitiin vuoarvojen avulla. Raakamaidon suodatuksissa polymeerikalvot pidättivät täydellisesti tai lähes täydellisesti jäännösrasvan ja mikrobit, mutta myös kaseiiniproteiinin. Tästä syystä yksikään polymeerikalvoista ei sovellu puhtaan maidon suodatukseen. Keraamikalvot soveltuvat tähän selkeästi paremmin ja yksikään toinen testatuista kalvoista ei päässyt samalle tasolle meijeriteollisuudessa jo käytössä olevan Membralox 1,4 μm -keraamikalvon kanssa. Heran suodatuksissa polymeerikalvot pidättivät täydellisesti tai lähes täydellisesti rasvan ja mikrobit, mutta keraamikalvo päästi huomattavasti enemmän haluttuja proteiineja permeaattiin. Membralox 0,8 μm -keraamikalvo pidätti täydellisesti mikrobit, mutta päästi jonkin verran jäännösrasvaa permeaattiin. Näin ollen kalvovalinta on kompromissi permeaatin puhtauden ja proteiinin saannon välillä. Keraamikalvojen vuohäviöt olivat merkittävästi pienemmät kuin polymeerikalvoilla molempien syöttöliuosten kanssa. Lisäksi keraamikalvojen puhdistus oli huomattavasti nopeampaa ja helpompaa. Tästä syystä keraamiset kalvot soveltuvat polymeerisiä paremmin maidon ja heran mikrosuodatukseen.

CD4+ T helper (Th) cells have an important role in the defence against diverse pathogens. Th cells can differentiate into several functionally distinct subtypes including Th1 and Th2 cells. Th1 cells are important for eradicating intracellular pathogens, whereas Th2 cells pro¬tect our body against extracellular parasites. However if uncontrolled, Th cells can mediate immunopathology such as asthma or allergies, but inappropriate Th response can also lead to autoimmune diseases such as multiple sclerosis or type 1 diabetes. Deeper knowledge of the regulation of the lymphocyte response both in vitro and in vivo is important for un¬derstanding the pathogenesis of immune-mediated diseases and microbe-host interactions. In the work presented in this thesis, the first goal was to elucidate the role of novel factors, PIM kinases and c-FLIP in the regulation of human Th cell differentiation. The oncogenic serine-threonine kinases of the PIM family were shown to be preferentially expressed in Th1 cells and in addition, by using RNA interference, they were also shown to be positive regulators of Th1 differentiation. The PIM depletion experiments suggest that PIM kinases promote the expression of the hallmark cytokine of Th1 cells, IFNγ, and influence the IL12/STAT4 pathway during the early Th1 cell differentiation. In addition to cytokine and T cell receptor (TCR) induced pathways, caspase activity has been shown to regulate Th cell proliferation. In the work presented in this thesis, the two isoforms of the caspase regulator protein, c-FLIP, were shown to be differentially ex¬pressed in Th1 and Th2 cells. Both of the isoforms were up-regulated in response to TCR activation, but the expression of the short isoform was selectively induced by IL4, the Th2 inducing cytokine. Furthermore, the c-FLIP isoforms had distinct and opposite roles during the early differentiation of Th1 and Th2 cells. The knockdown of the long isoform of c-FLIP led to the induction of Th1 marker genes, such as IFNγ and TBET, whereas the depletion of c-FLIP short down-regulated Th2 marker genes IL-4 and GATA3. The third goal was to elucidate the gene expression profiles characterizing the T- and B-lymphocyte responses in vivo during experimental infection by intracellular bacte¬rium Chlamydia pneumoniae. Previously, it has been shown that CD8+ and CD4+ T cells are important for the protection against Chlamydia pneumoniae. In this study, the analysis revealed up-regulation of interferon induced genes during recurrent infection underlining the importance of IFNγ secreted by Th1 and CD8+ T cells in the protection against this pathogen. Taken together, in this study novel regulators of Th cell differ¬entiation were discovered and in addition the gene expression profiles of lymphocytes induced by Chlamydia pneumoniae infection were characterized.

Infections with Salmonella serotypes are a major cause of food-borne diseases worldwide. Animal models other than the mouse have been employed for the study of nontyphoidal Salmonella infections because the murine model is not suitable for the study of Salmonella-induced diarrhea. The microbe has developed mechanisms to exploit the host cell machinery to its own purpose. Bacterial proteins delivered directly into the host cell cytosol cause cytoskeletal changes and interfere with host cell signaling pathways, which ultimately enhance disease manifestation. Recently, marked advances have been made in our understanding of the molecular interactions between Salmonella serotypes and their hosts. Here, we discuss the molecular basis of the pathogenesis of Salmonella-induced enteritis.

Caries is a plaque-associated multifactorial chronic disease. Oral hygiene habits, sugar, and oral micobiota interactions are important for caries to occur. Xylitol has been shown to reduce caries mainly due to its effects on mutans streptococci (MS). The purpose of this study was to evaluate the relationship of daily oral health habits and bacterial level on the caries occurrence and to study the effect of xylitol on the composition of oral microflora. A total of 192, 10-12 years old, male school children had been screened for salivary MS. Healthy subjects with high MS counts participated in two parallel double-blinded, randomised, controlled trials. In the first 5-week trial, subjects were assigned into xylitol (n=35) and sorbitol gum (n=38) groups. At baseline, children were examined using International Caries Detection and Assessment System (ICDAS) criteria and interviewed for oral health habits. In the second 4-week trial, subjects were assigned into xylitol (n=25) and saccharine mouthrinse (n=25) groups. In the end of both interventions, saliva samples were collected. The samples were analysed for changes in MS counts and changes in the composition of the oral microbiota assessed by the Human Oral Microbe Identification Microarray (HOMIM). Relationships between daily habits, bacterial levels and caries were evaluated. Daily use of sweets and soft drinks were the habits significantly associated with caries severity measured by ICDAS Caries Index (CI), while toothbrushing was the only habit associated with the low caries severity. Abiotrophia defectiva and Actinomyces meyeri/ A. odontolyticus were significantly higher in caries-affected children while Shuttleworthia satelles was significantly higher in caries-free children. Xylitol showed significant reduction in salivary levels of MS in both trials. No significant effects on other members of the microbiota were found when evaluated by HOMIM. In conclusion, other members of oral microbiota than MS may be associated with caries occurrence or absence. The use of xylitol had significant effect on MS with no effects on the other members of the salivary microbiota.

This study evaluated the microbiological quality of hamburgers and the microbe community on the hands of vendors in Cuiabá, Mato Grosso, Brazil, in relation to vendors´ awareness as to what constitute acceptable food-handling practices as part of a broad-spectrum research programme on street foods in Brazil . Sale of the hamburger known as the 'baguncinha' is common and widespread in urban Cuiabá, Mato Grosso, Brazil. Food inspectors encounter various difficulties in carrying out inspections. One hundred and five hamburgers samples were evaluated using conventional methods including tests for facultative aerobic and/or anaerobic mesophytic bacteria, coliform counts at 45 °C, the coagulase test for Staphylococcus, Gram-staining for the presence of Bacillus cereus, Clostridium sulphite reductase and Salmonella spp. The hamburgers were categorized as unsuitable for human consumption in 31.4% of samples, with those testing positive for coliforms and Staphylococcus at unacceptably high levels by Brazilian standards. High levels of microbiological contamination were detected on the hands of the food handlers and mesophytic bacterial counts reached 1.8 × 10(4) CFU/hand. Interviews were carried out by means of questionnaires to evaluate levels of awareness as to acceptable food handling practices and it was found that 80,1% of vendors had never participated in any kind of training.

Lysinuric protein intolerance (LPI) is a recessively inherited disorder characterised by reduced plasma and increased urinary levels of cationic amino acids (CAAs), protein malnutrition, growth failure and hyperlipidemia. Some patients develop severe immunological, renal and pulmonary complications. All Finnish patients share the same LPIFin mutation in the SLC7A7 gene that encodes CAA transporter y+LAT1. The aim of this study was to examine molecular factors contributing to the various symptoms, systemic metabolic and lipid profiles, and innate immune responses in LPI. The transcriptomes, metabolomes and lipidomes were analysed in whole-blood cells and plasma using RNA microarrays and gas or liquid chromatography-mass spectrometry techniques, respectively. Toll-like receptor (TLR) signalling in monocyte-derived macrophages exposed to pathogens was scrutinised using qRT-PCR and the Luminex technology. Altered levels of transcripts participating in amino acid transport, immune responses, apoptosis and pathways of hepatic and renal metabolism were identified in the LPI whole-blood cells. The patients had increased non-essential amino acid, triacylglycerol and fatty acid levels, and decreased plasma levels of phosphatidylcholines and practically all essential amino acids. In addition, elevated plasma levels of eight metabolites, long-chain triacylglycerols, two chemoattractant chemokines and nitric oxide correlated with the reduced glomerular function in the patients with kidney disease. Accordingly, it can be hypothesised that the patients have increased autophagy, inflammation, oxidative stress and apoptosis, leading to hepatic steatosis, uremic toxicity and altered intestinal microbe metabolism. Furthermore, the LPI macrophages showed disruption in the TLR2/1, TLR4 and TLR9 pathways, suggesting innate immune dysfunctions with an excessive response to bacterial infections but a deficient viral DNA response.

The Cochin estuary (CE), which is one of the largest wetland ecosystems, extends from Thanneermukkam bund in the south to Azhikode in the north. It functions as an effluent repository for more than 240 industries, the characteristics of which includes fertilizer, pesticide, radioactive mineral processing, chemical and allied industries, petroleum refining and heavy metal processing industries (Thyagarajan, 2004). Studies in the CE have been mostly on the spatial and temporal variations in the physical, chemical and biological characteristics of the estuary (Balachandran et al., 2006; Madhu et al., 2007; Menon et al., 2000; Qasim 2003;Qasim and Gopinathan 1969) . Although several monitoring programs have been initiated in the CE to understand the level of heavy metal pollution, these were restricted to trace metals distribution (Balachandran et al., 2005) or the influence of anthropogenic inputs on the benthos and phytoplankton (Madhu et al., 2007;Jayaraj, 2006). Recently, few studies were carried out on microbial ecology in the CE(Thottathil et al 2008a and b;Parvathi et al., 2009and 2011; Thomas et al., 2006;Chandran and Hatha, 2003). However, studies on metal - microbe interaction are hitherto not undertaken in this estuary. Hence, a study was undertaken at 3 sites with different level of heavy metal concentration tounderstand the abundance, diversity and mechanisms of resistance in metal resistant bacteria and its impact on the nutrient regeneration. The present work has also focused on the response of heavy metal resistant bacteria towards antibacterial agent’s antibiotics and silver nanoparticles
