In this work, the biodegradation mechanism of phenol and sub products (such as catechol and hydroquinone) in Chromobacterium violaceum was investigated by cloning and molecular characterization of a phenol monooxygenase gene in Escherichia coli. This gene (Cvmp) is very similar (74 and 59% of similarity and identity, respectively) to the ortholog from Ralstonia eutropha, bacteria capable of utilizing phenol as the sole carbon source. The phenol biodegradation ability of E. coli recombinant strains was tested by cell-growth in a minimal medium containing phenol as the sole source of carbon and release of intermediary metabolites (catechol and hydroquinone). Interestingly, during the growth of these strains on phenol, catechol, and hydroquinone accumulated transiently in the medium. These metabolites were further analyzed by HPLC. These results indicated that phenol can be initially orto or para hydroxylated to produce cathecol or hydroquinone, respectively, followed by meta-cleavage of aromatic rings. To verify this information, the metabolites obtained from HPLC were submitted to LC/MS to confirm their chemical structure, thereby indicating that the recombinant strains utilize two different routes simultaneously, leading to different ring-fission substrates for the metabolism of phenol. An important feature of a database management systems (DBMS) is its client/server architecture, where managing shared memory among the clients and the server is always an tough issue. However, similarity queries are specially sensitive to this kind of architecture, since the answer sizes vary widely. Usually, the answers of similarity query are fully processed to be sent in full to the user, who often is interested in just parts of the answer, e.g. just few elements closer or farther to the query reference. Compelling the DBMS to retrieve the full answer, further ignoring its majority is at least a waste of server processing power. Paging the answer is a technique that splits the answer onto several pages, following client requests. Despite the success of paging on traditional queries, little work has been done to support it in similarity queries. In this work, we present a technique that not only provides paging in similarity range or k-nearest neighbor queries, but also supports them in two variations: the forward similarity query and the backward similarity query. They return elements either increasingly farther of increasingly closer to the query reference. The reported experiments show that, depending on the proportion of the interesting part over the full answer, both techniques allow answering queries much faster than it is obtained in the non-paged way. An outbreak of infections affecting 311 patients who had undergone different invasive procedures occurred in 2004 and 2005 in the city of Belem, in the northern region of Brazil. Sixty-seven isolates were studied; 58 were from patients who had undergone laparoscopic surgeries, 1 was from a patient with a postinjection abscess, and 8 were from patients who had undergone mesotherapy. All isolates were rapidly growing nonpigmented mycobacteria and presented a pattern by PCR-restriction enzyme analysis of the hsp65 gene with BstEII of bands of 235 and 210 bp and with HaeIII of bands of 200, 70, 60, and 50 bp, which is common to Mycobacterium abscessus type 2, Mycobacterium bolletii, and Mycobacterium massiliense. hsp65 and. rpoB gene sequencing of a subset of 20 isolates was used to discriminate between these three species. hsp65 and rpoB sequences chosen at random from 11 of the 58 isolates from surgical patients and the postinjection abscess isolate presented the highest degrees of similarity with the corresponding sequences of M. massiliense. In the same way, the eight mesotherapy isolates were identified as M. bolletii. Molecular typing by pulsed-field gel electrophoresis (PFGE) grouped all 58 surgical isolates, while the mesotherapy isolates presented three different PFGE patterns and the postinjection abscess isolate showed a unique PFGE pattern. In conclusion, molecular techniques for identification and typing were essential for the discrimination of two concomitant outbreaks and one case, the postinjection abscess, not related to either outbreak all of which were originally attributed to a single strain of M. abscessus.

Recent movements within world Anglicanism towards a more democratic representation of the church are in contrast to Torres Strait Islanders' assertion of their own male-led conservative and hierarchical body. These characteristics have marked Torres Strait Island Anglicanism for many years. On the surface, the various strands leading to a conflict over a choice of leader in 1997 focused upon discordant relationships and faulty decision-making procedures, especially the surrender of the diocese of Carpentaria to the adjacent diocese of North Queensland and a subsequent choice of a bishop where Torres Strait clergy claimed that the terms of the surrender had been dishonoured. Yet below the surface, the cleavage between Island and European leadership was also a sign of the ideological shift which was occurring in the Anglican Church of Australia. Supported by European elements within that church opposed to the ordination of women, Islander clergy charged that the mainland body was deserting the faith and order of the 'church of the fathers'. With the Islanders newly empowered, as they perceived it, by the Mabo judgement of the High Court of Australia in 1992, their perception appears to have been that, in spirit, the mainland church denied what the High Court's decision recognised: the ultimate control by Islanders over their own affairs.

This article considers the decision of the Family Court of Australia in Re Kevin (Validity of Marriage of a Transsexual) [2001] FamCA 1074, which was upheld by the Full Court of the Family Court of Australia in February 2003. Re Kevin was the first case in Australia to deal directly with the question of whether a transsexual person could marry under Australian law. In the past, Australia had adhered to the judgement of Ormrod J in Corbett v Corbett [1971] P. 83, which set the benchmark for what is ‘male’ and what is ‘female’ under the common law. Prior to Re Kevin the question of what is a man and what is a woman for the purposes of marriage in Australia mirrored the strict biological test established in Corbett. In other words, the Australian courts relied upon biological factors, as espoused by Ormrod J, when determining a person's true sex. In Re Kevin, Chisholm J examined in detail what it is to be a man or woman, but unlike Ormrod J considered ‘brain sex’ to have a significant impact on a person's view of their own innate sexual identity. The Full Court of the Family Court agreed with the powerful and well-reasoned judgement of Chisholm J at first instance.

1. With the aim of determining whether patterns of variation in macroinvertebrate assemblage composition across the hierarchy of spatial units in two lowland rivers changed during a supra-seasonal drought (1997–2000), patterns during a reduced flow season (1999–2000) were compared with those during two preceding higher flow seasons (1997–98 and 1998–99) using samples from the Glenelg and Wimmera Rivers, two lowland regulated rivers in western Victoria, Australia.

2. We hypothesised that (i) differences between reaches would increase during the reduced flow season owing to decreased hydrological connectivity, (ii) differences between the habitats would decrease because the cessation of flow in run habitats should cause them to become more similar to pool habitats and (iii) differences between microhabitats would also decrease because of reduced scour of inorganic substrata and large woody debris.

3. During each season, macroinvertebrates were sampled from three microhabitats (sand/silt substratum, large woody debris and macrophytes) that were hierarchically nested within a run or pool habitat and within one of three reaches within each river. A range of physico-chemical variables was also sampled.

4. Analysis of similarity showed that assemblage composition in both rivers during the higher flow seasons differed more among microhabitats than other spatial units. However, during the reduced flow season, assemblage composition in the Wimmera River differed most among reaches. This change in pattern was associated with the combined effects of decreased flow and longitudinal increases in salinity. In contrast, the fauna of the Glenelg River appeared to be resistant to the effects of the reduced flow season, owing to limited decline in water quality despite lower river discharge.

5. As salinisation and poor water quality in the Wimmera River result from human activities in the catchment, these results support the idea that human impacts on rivers can change macroinvertebrate scaling patterns and exacerbate the effects of drought beyond the tolerance of many riverine macroinvertebrates.

Relationships between businesses, businesses and end customers, as well as between customers are an important area of practical and scientific interest. In the present era, largely due to digital technologies such as the database, public and private networks, and data collection and information distribution via TCP/IP (Transmission Control Protocol/Internet Protocol) interface tools such as the World Wide Web (Web), the interest in relationships and related aspects such as trust as it relates to Web interactivity continues. An important antecedent empirical study established that arts patrons (customers) of a New York theatre company could be segmented according to their relational orientation, and that this orientation mediated between component attitudes and future purchase intentions. The study reported in this paper employs Web-based data collection and postal data collection methods in an investigation of the mediation effects of these data collection methods used with the same population of a premier football club in Australia. While a future aim is to more closely compare the outcomes established in the arts study with those from a similarly constructed study in the sporting arena, the focus of this initial paper is the differences in response exhibited by online respondents relative to postal survey respondents. The paper reports findings which do not support those of the antecedent arts and entertainment study concerning the weakness of overall satisfaction on the purchase intentions of high relational orientation customers. The paper also reports findings which give confidence to users of online surveys that despite differences in demographic profiles of these respondents and postal survey respondents, there is a degree of similarity in the responses of the two groups on the measures used in this study. The paper also suggests the need for further research into these data collection effects as they relate to relationship marketing.

A widely recognised theme of construction economics suggests that the cost of construction per square metre increases as building height rises. However, after many years, research conducted regarding the height and cost issue have established a classic relationship between those two, well known as a U-shaped curve. This paper describes the study of height-cost relationship of high-rise residential buildings in Shanghai and Hong Kong. Initial findings indicated that the curved relationships of height-cost of residential buildings in Shanghai and Hong Kong exhibit different profiles. The differences suggest that, Hong Kong contractors have more expertise in multi-storey and high-rise construction than contractors in Shanghai. The dissimilarities also imply that different sets of criteria should be applied in the judgement of height affects cost in different locations. Many factors could be contributors, such as the history and experience in constructing residential high-rise buildings, location, linkage and relationships to the neighbourhood provinces, design and construction regulations, and government policy on residential construction.

We have determined the structure of the reduced form of the DsbA oxidoreductase from Vibrio cholerae. The reduced structure shows a high level of similarity to the crystal structure of the oxidized form and is typical of this class of enzyme containing a thioredoxin domain with an inserted α-helical domain. Proteolytic and thermal stability measurements show that the reduced form of DsbA is considerably more stable than the oxidized form. NMR relaxation data have been collected and analyzed using a model-free approach to probe the dynamics of the reduced and oxidized states of DsbA. Akaike's information criteria have been applied both in the selection of the model-free models and the diffusion tensors that describe the global motions of each redox form. Analysis of the dynamics reveals that the oxidized protein shows increased disorder on the pico- to nanosecond and micro- to millisecond timescale. Many significant changes in dynamics are located either close to the active site or at the insertion points between the domains. In addition, analysis of the diffusion data shows there is a clear difference in the degree of interdomain movement between oxidized and reduced DsbA with the oxidized form being the more rigid. Principal components analysis has been employed to indicate possible concerted movements in the DsbA structure, which suggests that the modeled interdomain motions affect the catalytic cleft of the enzyme. Taken together, these data provide compelling evidence of a role for dynamics in the catalytic cycle of DsbA.

Recently enacted legislation in New Zealand, the Parole (Extended Supervision) Amendment Act 2004, allows for the imposition of up to 10 years of supervision in the community for child-victim sex offenders following their release from prison. The Act requires reports to be written specifically assessing the risk of sexual re-offending against children. This study examined the application of actuarial measures used by the New Zealand Department of Corrections in these assessments, including a computer-scored instrument based on static factors (the Automated Sexual Recidivism Scale; ASRS) and a clinically-based judgement of dynamic risk factors (the SONAR). It was expected that a conservative approach would be taken in making recommendations for or against extended periods of supervision, such that a high score on either measure would predict a recommendation for extended supervision. It was found, however, that a more individualized approach was often taken, whereby a baseline assessment of risk as predicted by the ASRS was adjusted by clinicians based on SONAR ratings. Implications for the practice of risk assessment in sexual re-offending are discussed.

Riparian zones are a characteristic component of many landscapes throughout the world and increasingly are valued as key areas for biodiversity conservation. Their importance for bird communities has been well recognised in semi-arid environments and in modified landscapes where there is a marked contrast between riparian and adjacent non-riparian vegetation. The value of riparian zones in largely intact landscapes with continuous vegetation cover is less well understood. This research examined the importance of riparian habitats for avifauna conservation by investigating the ecological interactions contributing to the pattern of bird assemblages in riparian and adjacent non-riparian habitats. Specifically, the focus is on the bird assemblages of riparian zones and those of adjacent non-riparian vegetation types and the influence that associated differences in resource availabilities, habitat structure and conditions have on observed patterns. This study was conducted in the foothill forests of the Victorian Highlands, south-east Australia. Mixed-species eucalypt (genus Eucalyptus) forests dominate the vegetation of this region. Site selection was based on the occurrence of suitable riparian habitat interspersed within extensive, relatively undisturbed (i.e. no recent timber harvesting or fire events) forest mosaics. A series of 30 paired riparian and non-riparian sites were established among six stream systems in three forest areas (Bunyip State Park, Kinglake National Park and Marysville State Forest). Riparian sites were positioned alongside the stream and the non-riparian partner site was positioned on a facing slope at a distance of approximately 750 m. Bird surveys were carried out during 29 visits to each site between July 2001 and December 2002. Riparian sites were floristically distinct from non-riparian sites and had a more complex vegetation structure, including a mid-storey tree layer mostly absent from non-riparian sites, extensive fine litter and coarse woody debris, and dense ground-layer vegetation (e.g. sedges and ground ferns). The characteristic features of non-riparian habitats included a relatively dense canopy cover, a ground layer dominated by grasses and fine litter, and a high density of canopy-forming trees in the smaller size-classes. Riparian zones supported a significantly greater species richness, abundance and diversity of birds when compared to non-riparian habitats. The composition of bird assemblages differed significantly between riparian and non-riparian habitats, with riparian assemblages displaying a higher level of similarity among sites. The strongest contributors to observed dissimilarities between habitat types included species that occurred exclusively in either habitat type or species with large contrasts in abundance between habitat types. Much of the avifauna (36%) of the study area is composed of species that are common and widespread in south-east Australia (i.e. forest generalists). Riparian habitats were characterised by a suite of species more typical of wetter forest types in south-east Australia and many of these species had a restricted distribution in the forest mosaic. Some species (7%) occurred exclusively in riparian habitats (i.e. riparian selective species) while others (43%) were strongly linked to these habitats (i.e. riparian associated species). A smaller proportion of species occurred exclusively (2%) in non-riparian habitats (i.e. non-riparian selective species) or were strongly linked to these habitats (10%; i.e. non-riparian associated species). To examine the seasonal dynamics of assemblages, the variation through time in species richness, abundance and composition was compared between riparian and non-riparian sites. Riparian assemblages supported greater richness and abundance, and displayed less variation in these parameters, than non-riparian assemblages at all times. The species composition of riparian assemblages was distinct from non-riparian assemblages throughout the annual cycle. An influx of seasonal migrants elevated species richness and abundance in the forest landscape during spring and summer. The large-scale movement pattern (e.g. coastal migrant, inland migrant) adopted by migrating species was associated with their preference for riparian or non-riparian habitats in the landscape. Species which migrate north-south along the east coast of mainland Australia (i.e. coastal migrants) used riparian zones disproportionately; eight of eleven species were riparian associated species. Species which migrate north-south through inland Australia (i.e. inland migrants) were mostly associated with non-riparian habitats. The significant differences in the dynamics of community structure between riparian and non-riparian assemblages shows that there is a disproportionate use of riparian zones across the landscape and that they provide higher quality habitat for birds throughout the annual cycle. To examine the ecological mechanisms by which riparian assemblages are richer and support more individual birds, the number of ecological groups (foraging, nest-type and body mass groups) represented, and the species richness of these groups, was compared between riparian and non-riparian assemblages. The structurally complex vegetation and distinctive habitat features (e.g. aquatic environments, damp sheltered litter) provided in the riparian zone, resulted in the consistent addition of ecological groups to riparian assemblages (e.g. sheltered ground – invertebrates foraging group) compared with non-riparian assemblages. Greater species richness was accommodated in most foraging, nest-type and body mass groups in riparian than non-riparian assemblages. Riparian zones facilitated greater richness within ecological groups by providing conditions (i.e. more types of resources and greater abundance of resources) that promoted ecological segregation between ecologically similar species. For a set of commonly observed species, significant differences in their use of structural features, substrates and heights were registered between riparian and non-riparian habitats. The availability and dynamics of resources in riparian and non-riparian habitats were examined to determine if there is differential availability of particular resources, or in their temporal availability, throughout the annual cycle. Riparian zones supported more abundant and temporally reliable eucalypt flowering (i.e. nectar) than non-riparian habitats throughout the annual cycle. Riparian zones also supported an extensive loose bark resource (an important microhabitat for invertebrates) including more peeling bark and hanging bark throughout the year than at non-riparian sites. The productivity of eucalypts differed between habitat types, being higher in riparian zones at most times for all eucalypts combined, and for some species (e.g. Narrow-leaved Peppermint Eucalyptus radiata). Non-riparian habitats provided an abundant nectar resource (i.e. shrub flowering) at particular periods in the annual cycle. Birds showed clear relationships with the availability of specific food (i.e. nectar) and foraging resources (i.e. loose bark). The demonstration of a greater abundance of resources and higher primary productivity in riparian zones is consistent with the hypothesis that these linear strips that occupy only a small proportion of the landscape have a disproportionately high value for birds. Riparian zones in continuous eucalypt forest provide high quality habitats that contribute to the diversity of habitats and resources available to birds in the forest mosaic, with positive benefits for the landscape-level species pool. Despite riparian and non-riparian habitat supporting distinct assemblages of birds, strong linkages are maintained along the riparian-upslope gradient. Clearly, the maintenance of diverse and sustainable assemblages of birds in forest landscapes depends on complementary management of both riparian and non-riparian vegetation.

This thesis analyses the ways in which moral judgements of so-called privileged Jews are constructed in Holocaust representations. ‘Privileged’ Jews include those prisoners in the camps and ghettos who held positions which gave them access to material and other benefits. Subject to extreme levels of coercion, these victims were compelled to act in ways that have often been judged as both self-serving and harmful to fellow inmates. Such controversial figures constitute an intrinsically important, frequently misunderstood and hastily judged facet of the Holocaust. Scholars have neglected the problem of judgement in relation to ‘privileged’ Jews; nonetheless, Holocaust texts frequently portray these liminal figures.

Of crucial importance to the thesis is Primo Levi’s paradigmatic essay entitled ‘The Grey Zone,’ which directly engages with the complex and sensitive issue of ‘privileged’ Jews. Levi argues that due to the extreme ethical dilemmas that ‘privileged’ Jews confronted, any judgement of these victims needs to be suspended. However, if, as Levi suggests, judgement is at times impossible, the thesis challenges Levi’s assumption by contending that representations of ‘privileged’ Jews inevitably take a moral position. In this way, the thesis conceptualises judgement as a ‘limit’ of representation. Indeed, it is shown that Levi himself cannot abstain from judging those for whom he argues judgement should be suspended.

The thesis takes Levi’s concept of the ‘grey zone’ as a point of departure in order to examine the problems of judgement and representation in relation to ‘privileged’ Jews. Analysis focuses on Raul Hilberg’s influential historical work and examples of documentary and fiction films. The thesis examines how Hilberg and several filmmakers employ conventions as a means of conveying judgement. It is argued that self-reflexive representations of ‘privileged’ Jews in film, particularly fictional dramatisation, have the potential to provide a nuanced representation of ‘privileged’ Jews, which engages with Levi’s ideas by questioning the possibility of judgement.

The blogosphere has grown to be a mainstream forum of social interaction as well as a commercially attractive source of information and influence. Tools are needed to better understand how communities that adhere to individual blogs are constituted in order to facilitate new personal, socially-focused browsing paradigms, and understand how blog content is consumed, which is of interest to blog authors, big media, and search. We present a novel approach to blog subcommunity characterization by modeling individual blog readers using mixtures of an extension to the LDA family that jointly models phrases and time, Ngram Topic over Time (NTOT), and cluster with a number of similarity measures using Affinity Propagation. We experiment with two datasets: a small set of blogs whose authors provide feedback, and a set of popular, highly commented blogs, which provide indicators of algorithm scalability and interpretability without prior knowledge of a given blog. The results offer useful insight to the blog authors about their commenting community, and are observed to offer an integrated perspective on the topics of discussion and members engaged in those discussions for unfamiliar blogs. Our approach also holds promise as a component of solutions to related problems, such as online entity resolution and role discovery.

While the largest common subgraph (LCSG) between a query and a database of models can provide an elegant and intuitive measure of similarity for many applications, it is computationally expensive to compute. Recently developed algorithms for subgraph isomorphism detection take advantage of prior knowledge of a database of models to improve the speed of on-line matching. This paper presents a new algorithm based on similar principles to solve the largest common subgraph problem. The new algorithm significantly reduces the computational complexity of detection of the LCSG between a known database of models, and a query given on-line.
