Toxoplasma gondii is a protozoan parasite that induces behavioral changes in rodents. The aim of this study was to evaluate the effect of infection by T. gondii during the chronic phase in working memory and impulsivity in rodents as well as the effect of antipsychotics to reverse any behavioral changes resulting from infection. Female Wistar rats (n = 40) were infected with 25 cysts of the strain ME-49 T. gondii after 4 months the animals were subjected to behavioral tests: tolerance to delay gratification, in which the animal must choose between two rewards, a smaller and more immediate, but delayed and the test of spontaneous alternation, in which the animal must use spatial cues to remember previously visited arms. Antipsychotic drugs were intraperitoneally administered during the testing of the behavioral experiments, the antipsychotic is haloperidol (1.5 mg / kg) administered 60 min before the start of the session and the antipsychotic clozapine (2.5 mg / kg) 30 min before. Animals infected with the parasite did not show operating deficits of memory, and motor impairment did not develop, however motor impairment was observed only in animals treated with haloperidol. It was found that administration of clozapine and haloperidol increased the percentage of alternation in infected and control groups in task switching espontânea.Não no distinction between control animals and infected the test of tolerance to delay gratification in relation to the percentage of choices greatest reward, during the pre-training and training, in which there is a delay of 15 s to access the great reward, however it was observed that infected animals prefer the greatest reward, when there is a delay of 30 s when compared to control group. The administration of clozapine possible that infected animals chose the greatest reward in the delay of 30 seconds during the test. These data suggest that infected mice do not exhibit deficits in working memory and that clozapine has therapeutic efficacy in improving cognitive performance of mice infected

The involvement of neurotoxicants in the etiology of emotional pathologies is becoming an issue in neurotoxicology. Lead (Pb) exposure during childhood has been associated with increased impulsivity, aggressivity, and delinquency. Considering the paucity of experimental studies investigating the involvement of developmental Pb exposure in emotional disorders, our objective was to investigate whether Pb exposure during pregnancy and/or lactation could be related to depressive symptoms in adult male and female rats. Wistar dams received 10 mg of Pb, as Pb acetate, or 13.4 mg of Na acetate, by gavage, daily, during pregnancy and lactation. By cross-fostering at the time of birth, pups were either exposed to Ph or Na acetate during pregnancy only, lactation only, or during both pregnancy and lactation. At 70 days of age, animals were submitted to the open-field test followed by the forced swimming test. Ph levels were measured in the blood of dams (weaning) and pups (after behavioral evaluation). The results demonstrated that exposure to Ph during both pregnancy and lactation induced, in males, an increased emotionality state detected in the open-field test, and in females, depressive-like behavior detected in the forced swimming test. These alterations were observed at residual blood Pb levels (i.e., around 5 mug/dL).

Neuropsychological disorders are frequently associated with obstructive ventilatory disorders (OVD). Aim: To analyze the incidence of neuropsychological disorders in Brazilian children with OVD, using a screening questionnaire and to compare the answers given before and after surgery. Patients and Methods: We studied 30 children with clinical diagnosis of OVD. The children were divided into 3 groups: group I, children aged 4 to 7; group II, from 8 to 10; and group III, children over 11. The applied questionnaires were answered by the parents or tutors, and comprised 30 questions, 10 for each disorder: attention deficit, hyperactivity and impulsivity. The children were diagnosed with one of the disorders when presented 3 or more positive answers. The follow up interview occurred 6 months after adenotonsillectomy. Results: There was a predominance of male gender (60.6%) over female gender (39.4%). Group II presented the highest number of significant changes, with reductions raging from 87.5% to 33.3% of patients with attention deficit, 75% to 50% of the hyperactive patients, and 50% to 33% of the impulsive patients. Conclusion: There was neuropsychological improvement after the surgery, which occurred mainly in the children from group II. More interaction among health professionals is necessary when diagnosing and following up similar cases.

Although traditionally obsessive-compulsive disorder (OCD) and impulse control disorders (ICD) have represented opposing ends of a continuum, recent research has demonstrated a frequent co-occurrence of impulsive and compulsive behaviours, which may contribute to a worse clinical picture of some psychiatric disorders. We hypothesize that individuals with 'impulsive' OCD as characterized by poor insight, low resistance, and reduced control towards their compulsions will have a deteriorative course, greater severity of hoarding and/or symmetry/ordering symptoms, and comorbid ICD and/or substance use disorders (SUD). The sample consisted of 869 individuals with a minimum score of 16 on the Yale-Brown Obsessive Compulsive Scale (Y-BOCS). Of these, 65 had poor insight, low resistance, and reduced control towards compulsions ('poor IRC') and 444 had preserved insight, greater resistance and better control over compulsions ('good IRC'). These two groups were compared on a number of clinical and demographic variables. Individuals with poor IRC were significantly more likely to have a deteriorative course (p < 0.001), longer duration of obsessions (p = 0.017), greater severity of symmetry/ordering (p < 0.001), contamination/cleaning (p < 0.001) and hoarding (p = 0.002) symptoms, and comorbid intermittent explosive disorder (p = 0.026), trichotillomania (p = 0.014) and compulsive buying (p = 0.040). Regression analysis revealed that duration of obsessions (p = 0.037) and hoarding severity (p = 0.005) were significant predictors of poor IRC. In the absence of specific measures for impulsivity in OCD, the study highlights the utility of simple measures such as insight, resistance and control over compulsions as a phenotypic marker of a subgroup of OCD with impulsive features demonstrating poor clinical outcome. © 2012 Elsevier Ltd.

Transtorno do Déficit de Atenção e Hiperatividade (TDAH) é um fenômeno estudado em diversos países, composto de sintomas de desatenção, imperatividade e impulsividade, ocorre na infância podendo persistir até a idade adulta. Este transtorno causa prejuízos nas áreas acadêmicas, sociais e ocupacionais, reduz a auto-estima, pode evoluir para delinqüência, uso de drogas e álcool, gera estresse nas famílias, tem alto custo financeiro e impacto social. O diagnóstico é feito com base em critérios clínicos adotados pelo DSM IV e o tratamento indicado combina farmacoterapia e psicoterapia. Considerando-se que TDAH é um transtorno de desenvolvimento de autocontrole e que estudos sugerem que autocontrole pode ser adquirido em condições de treino, torna-se relevante a realização de pesquisas aplicadas utilizando princípios da análise do comportamento, para minimizar prejuízos e contribuir para melhoria de qualidade de vida de portadores de TDAH. Este estudo objetivou verificar a eficácia do uso de esquemas de reforçamento diferencial de outro comportamento (DRO) e de atraso de reforço na instalação e/ ou aumento de comportamentos de autocontrole em um menino de 9 anos diagnosticado com TDAH, que faz uso de medicamento. O participante foi exposto ao procedimento de treino de autocontrole, realizou tarefas, durante as quais, caso se comportasse como combinado, recebia reforço. Ao final de cada sessão, trocava os reforços por brinquedos e poderia escolher reservá-los para obter reforços de maior valor. O procedimento foi dividido em sete etapas: (1) entrevista com a neuropediatra; (2) análise do prontuário e convocação; (3) entrevista inicial com responsáveis; (4) visita à escola e entrevista com professores; (5) sessões de observação direta (linha de base, habituação às regras, instalação, manutenção, fading e avaliação da estabilidade); (6) followp-up; e (7) encerramento. Realizaram-se 18 sessões de observação direta, gravadas em vídeo e transcritas para a elaboração de um sistema de categorias de comportamentos para análise. Os resultados foram analisados por meio da comparação entre relatos de entrevistas, resultados de instrumentos padronizados e categorias de comportamentos observados. Identificou-se que os comportamentos de autocontrole do participante se ampliaram e generalizaram para outros contextos, durante as sessões e em domicílio, conforme os relatos dos pais e da professora da escola atual, e registro de observação direta da terapeuta-pesquisadora. Verificou-se que a utilização de esquema de reforçamento DRO com disponibilidade limitada pode favorecer o aumento de comportamentos de autocontrole e generalizações em crianças com TDAH.

Esse estudo foi realizado com o objetivo de contribuir com a avaliação dos efeitos de uma intervenção comportamental direcionada a pais/responsáveis de crianças com Transtorno do Déficit de Atenção e Hiperatividade-TDAH, investigando os efeitos desse modelo de treino parental em duas condições, setting terapêutico (Condição 1) e ambiente domiciliar (Condição 2), sobre a ocorrência de comportamentos de hiperatividade versus autocontrole. Os participantes foram pais de quatro crianças, na faixa etária entre cinco e nove anos. Utilizaram-se como instrumentos: Termo de Consentimento Livre e Esclarecido-TCLE, Inventário de Estilos Parentais-IEP, Lista de Verificação Comportamental para Crianças e Adolescentes-CBCL/TRF, Escala do TDAH versão para professor, Roteiro de Entrevista Inicial, Roteiro de Entrevista de Avaliação, Roteiro de Entrevista Final, Critério de Classificação Econômica Brasil-CCEB. O procedimento de pesquisa consistiu em: (a) contato com neuropediatra; (b) triagem e convite aos participantes; (c) distribuição de dois participantes para cada condição de intervenção; (d) avaliação inicial, incluindo entrevista com os responsáveis, aplicações do TCLE, IEP e CBCL; (e) visita à escola e aplicações do TCLE, da Escala do TDAH e do TRF, versões para professor; (f) realização de cinco sessões de intervenção, gravadas em áudio e vídeo, duas de linha de base, uma de habituação às regras e duas de manutenção das regras e instalação de comportamento de auto-observação, que envolveram situações de interação em jogos de regras, com participação da terapeuta-pesquisadora, da mãe e da criança; (g) realização de entrevista de avaliação da primeira fase; (h) reversão de contextos para os participantes e (i) avaliação final, realizada por meio de entrevista com os responsáveis e re-aplicação dos instrumentais padronizados com pais e professores, utilizados anteriormente, mais o CCEB. Os dados obtidos por meio dos instrumentos padronizados receberam o tratamento indicado nos manuais. Dois sistemas de categorias de análise do comportamento foram utilizados, um para descrever os comportamentos das mães e outro para comportamentos observados nas crianças. Os principais resultados sugerem que as crianças em ambiente de consultório tiveram maior ocorrência de emissão de comportamentos de autocontrole do que as em ambiente de domicílio, as quais, por sua vez, tiveram prevalência de comportamentos de hiperatividade/impulsividade. Do mesmo modo, as mães em ambiente de consultório obtiveram maiores escores em práticas educativas positivas e menos em negativas, comparadas às mães do grupo de domicílio. Houve aumento de práticas educativas positivas para a maioria das mães. Discute-se o contexto de consultório enquanto um ambiente eficaz de intervenção, embora se reconheça que as dificuldades de controle de comportamentos inadequados são maiores para os pais em ambiente domiciliar, por isso, intervenções em ambiente natural devem ser consideradas no processo terapêutico. Por outro lado, o treino parental demonstrou ser efetivo na aquisição, fortalecimento e manutenção de práticas educativas positivas em todas as mães, o que pode influenciar beneficamente os comportamentos das crianças com TDAH.

Aims To describe, in the context of DSM-V, how a focus on addiction and compulsion is emerging in the consideration of pathological gambling (PG). Methods A systematic literature review of evidence for the proposed re-classification of PG as an addiction. Results Findings include: (i) phenomenological models of addiction highlighting a motivational shift from impulsivity to compulsivity associated with a protracted withdrawal syndrome and blurring of the ego-syntonic/ego-dystonic dichotomy; (ii) common neurotransmitter (dopamine, serotonin) contributions to PG and substance use disorders (SUDs); (iii) neuroimaging support for shared neurocircuitries between behavioural and substance addictions and differences between obsessivecompulsive disorder (OCD), impulse control disorders (ICDs) and SUDs; (iv) genetic findings more closely related to endophenotypic constructs such as compulsivity and impulsivity than to psychiatric disorders; (v) psychological measures such as harm avoidance identifying a closer association between SUDs and PG than with OCD; (vi) community and pharmacotherapeutic trials data supporting a closer association between SUDs and PG than with OCD. Adapted behavioural therapies, such as exposure therapy, appear applicable to OCD, PG or SUDs, suggesting some commonalities across disorders. Conclusions PG shares more similarities with SUDs than with OCD. Similar to the investigation of impulsivity, studies of compulsivity hold promising insights concerning the course, differential diagnosis and treatment of PG, SUDs, and OCD.

Introduction: Compulsive buying (CB) is currently classified as an impulse control disorder (ICD) not otherwise classified. Compulsive buying prevalence is estimated at around 5% of the general population. There is controversy about whether CB should be classified as an ICD, a subsyndromal bipolar disorder (BD), or an obsessive-compulsive disorder (OCD) akin to a hoarding syndrome. To further investigate the appropriate classification of CB, we compared patients with CB, BD, and OCD for impulsivity, affective instability, hoarding, and other OCD symptoms. Method: Eighty outpatients (24 CB, 21 BD, and 35 OCD) who were neither manic nor hypomanic were asked to fill out self-report questionnaires. Results: Compulsive buying patients scored significantly higher on all impulsivity measures and on acquisition but not on the hoarding subdimensions of clutter and "difficulty discarding." Patients with BD scored higher on the mania dimension from the Structured Clinical Interview for Mood Spectrum scale. Patients with OCD scored higher on obsessive-compulsive symptoms and, particularly, higher on the contamination/washing and checking dimensions from the Padua Inventory; however, they did not score higher on any hoarding dimension. A discriminant model built with these variables correctly classified patients with CB (79%), BD (71%), and OCD (77%). Conclusion: Patients with CB came out as impulsive acquirers, resembling ICD- rather than BD- or OCD-related disorders. Manic symptoms were distinctive of patients with BD. Hoarding symptoms other than acquisition were not particularly associated with any diagnostic group. (C) 2012 Elsevier Inc. All rights reserved.

OBJETIVO: avaliar a memória operacional fonológica e relacionar com a impulsividade de pacientes em tratamento no Centro de Atenção Integrada à Saúde Mental. MÉTODO: 29 usuários: 21 do gênero masculino e 8 do feminino, usuários de substâncias psicoativas, com 37,9±10,5 anos de idade e 10,59±3,53 anos de escolaridade; e 30 voluntários: 19 do gênero masculino e 11 do feminino, com 32,4±11,9 anos de idade e 11,07±3,29 anos de escolaridade, sem histórico psiquiátrico ou de dependência química foram convocados à avaliação de: 1) memória operacional para palavras e pseudo-palavras; 2) impulsividade em seus fatores de segunda ordem (impulsividade atencional, motora e de não planejamento). RESULTADOS: o desempenho dos usuários de substâncias psicoativas na avaliação da memória em comparação ao grupo controle foi pior tanto no span auditivo de palavras e pseudo-palavras como também no número total de recordação de palavras e pseudo-palavras. Na avaliação da impulsividade, os usuários apresentaram escores elevados em contraposição aos sujeitos controle em todos os subtipos de impulsividade, inclusive no total. Na análise de correlação dos dados não foram encontradas relações entre os escores de impulsividade e memória. CONCLUSÃO: : este padrão de respostas indica comprometimento da memória operacional fonológica provavelmente independente do alto nível de impulsividade apresentado pelos usuários de drogas. Estas análises contribuem para propor estratégias de tratamento direcionadas às alterações detectadas.

People are daily faced with intertemporal choice, i.e., choices differing in the timing of their consequences, frequently preferring smaller-sooner rewards over larger-delayed ones, reflecting temporal discounting of the value of future outcomes. This dissertation addresses two main goals. New evidence about the neural bases of intertemporal choice is provided. Following the disruption of either the medial orbitofrontal cortex or the insula, the willingness to wait for larger-delayed outcomes is affected in odd directions, suggesting the causal involvement of these areas in regulating the value computation of rewards available with different timings. These findings were also supported by a reported imaging study. Moreover, this dissertation provides new evidence about how temporal discounting can be modulated at a behavioral level through different manipulations, e.g., allowing individuals to think about the distant time, pairing rewards with aversive events, or changing their perceived spatial position. A relationship between intertemporal choice, moral judgements and aging is also discussed. All these findings link together to support a unitary neural model of temporal discounting according to which signals coming from several cortical (i.e., medial orbitofrontal cortex, insula) and subcortical regions (i.e., amygdala, ventral striatum) are integrated to represent the subjective value of both earlier and later rewards, under the top-down regulation of dorsolateral prefrontal cortex. The present findings also support the idea that the process of outcome evaluation is strictly related to the ability to pre-experience and envision future events through self-projection, the anticipation of visceral feelings associated with receiving rewards, and the psychological distance from rewards. Furthermore, taking into account the emotions and the state of arousal at the time of decision seems necessary to understand impulsivity associated with preferring smaller-sooner goods in place of larger-later goods.

Pochi studi hanno indagato il profilo dei sintomi non-motori nella malattia di Parkinson associata al gene glucocerebrosidasi (GBA). Questo studio è mirato alla caratterizzazione dei sintomi non-motori, con particolare attenzione alla valutazione delle funzioni neurovegetativa, cognitiva e comportamentale, nel parkinsonismo associato a mutazione del gene GBA con la finalità di verificare se tali sintomi non-motori siano parte dello spettro clinico di questi pazienti. E’ stato condotto su una coorte di pazienti affetti da malattia di Parkinson che erano stati tutti sottoposti ad una analisi genetica per la ricerca di mutazioni in uno dei geni finora associati alla malattia di Parkinson. All’interno di questa coorte omogenea sono stati identificati due gruppi diversi in relazione al genotipo (pazienti portatori della mutazione GBA e pazienti non portatori di nessuna mutazione) e le caratteristiche non-motorie sono state confrontate nei due gruppi. Sono state pertanto indagati il sistema nervoso autonomo, mediante studio dei riflessi cardiovascolari e analisi dei sintomi disautonomici, e le funzioni cognitivo-comportamentali in pazienti affetti da malattia di Parkinson associata a mutazione del gene GBA. I risultati sono stati messi a confronto con il gruppo di controllo. Lo studio ha mostrato che i pazienti affetti da malattia di Parkinson associata a mutazione del gene GBA presentavano maggiore frequenza di disfunzioni ortosimpatiche, depressione, ansia, apatia, impulsività, oltre che di disturbi del controllo degli impulsi rispetto ai pazienti non portatori. In conclusione, i pazienti GBA positivi possono esprimere una sintomatologia non-motoria multidominio con sintomi autonomici, cognitivi e comportamentali in primo piano. Pertanto l’impostazione terapeutica in questi pazienti dovrebbe includere una accurata valutazione dei sintomi non-motori e un loro monitoraggio nel follow up clinico, allo scopo di ottimizzare i risultati e ridurre i rischi di complicazioni.

Children with attention-deficit/hyperactivity disorder (ADHD) have a higher rate of obesity than children without ADHD. Obesity risk alleles may overlap with those relevant for ADHD. We examined whether risk alleles for an increased body mass index (BMI) are associated with ADHD and related quantitative traits (inattention and hyperactivity/impulsivity). We screened 32 obesity risk alleles of single nucleotide polymorphisms (SNPs) in a genome-wide association study (GWAS) for ADHD based on 495 patients and 1,300 population-based controls and performed in silico analyses of the SNPs in an ADHD meta-analysis comprising 2,064 trios, 896 independent cases, and 2,455 controls. In the German sample rs206936 in the NUDT3 gene (nudix; nucleoside diphosphate linked moiety X-type motif 3) was associated with ADHD risk (OR: 1.39; P = 3.4 × 10(-4) ; Pcorr  = 0.01). In the meta-analysis data we found rs6497416 in the intronic region of the GPRC5B gene (G protein-coupled receptor, family C, group 5, member B; P = 7.2 × 10(-4) ; Pcorr  = 0.02) as a risk allele for ADHD. GPRC5B belongs to the metabotropic glutamate receptor family, which has been implicated in the etiology of ADHD. In the German sample rs206936 (NUDT3) and rs10938397 in the glucosamine-6-phosphate deaminase 2 gene (GNPDA2) were associated with inattention, whereas markers in the mitogen-activated protein kinase 5 gene (MAP2K5) and in the cell adhesion molecule 2 gene (CADM2) were associated with hyperactivity. In the meta-analysis data, MAP2K5 was associated with inattention, GPRC5B with hyperactivity/impulsivity and inattention and CADM2 with hyperactivity/impulsivity. Our results justify further research on the elucidation of the common genetic background of ADHD and obesity.

The German version of the Conners Adult ADHD Rating Scales (CAARS) has proven to show very high model fit in confirmative factor analyses with the established factors inattention/memory problems, hyperactivity/restlessness, impulsivity/emotional lability, and problems with self-concept in both large healthy control and ADHD patient samples. This study now presents data on the psychometric properties of the German CAARS-self-report (CAARS-S) and observer-report (CAARS-O) questionnaires.
