Sovint, la sistemàtica, basada principalment en caràcters morfològics, no es correspon amb els processos evolutius relacionats amb l'aparició dels grups d'organismes. En l'actualitat, la utilització de les dades moleculars es fa indispensable per a una revisió i millora de la classificació biològica de diversos organismes, com els peixos Acanthopterygii. A la sèrie Mugilomorpha la incongruència entre la taxonomia i la filogènia sorgeix de l'elevada semblança morfològica trobada per part dels seus membres. Pel que fa referència a la sèrie Atherinomorpha, la problemàtica principal resideix en determinar la seva proximitat evolutiva respecte a la sèrie anterior i en establir les relacions filogenètiques dins de la mateixa. Per tant, s'hi ha volgut estimar tant la divergència genètica dins de cada sèrie com inferir les relacions filogenètiques entre ambdues mitjançant la seqüenciació directa del DNA de les regions mitocondrials corresponents al tRNA-Phe, 12S rRNA, COI, cytb, tRNA-Thr, tRNA-Pro i regió control.

The open vegetation corridor of South America is a region dominated by savanna biomes. It contains forests (i.e. riverine forests) that may act as corridors for rainforest specialists between the open vegetation corridor and its neighbouring biomes (i.e. the Amazonian and Atlantic forests). A prediction for this scenario is that populations of rainforest specialists in the open vegetation corridor and in the forested biomes show no significant genetic divergence. We addressed this hypothesis by studying plumage and genetic variation of the Planalto woodcreeper Dendrocolaptes platyrostris Spix (1824) (Aves: Furnariidae), a forest specialist that occurs in both open habitat and in the Atlantic forest. The study questions were: (1) is there any evidence of genetic continuity between populations of the open habitat and the Atlantic forest and (2) is plumage variation congruent with patterns of neutral genetic structure or with ecological factors related to habitat type? We used cytochrome b and mitochondrial DNA control region sequences to show that D. platyrostris is monophyletic and presents substantial intraspecific differentiation. We found two areas of plumage stability: one associated with Cerrado and the other associated with southern Atlantic Forest. Multiple Mantel tests showed that most of the plumage variation followed the transition of habitats but not phylogeographical gaps, suggesting that selection may be related to the evolution of the plumage of the species. The results were not compatible with the idea that forest specialists in the open vegetation corridor and in the Atlantic forest are linked at the population level because birds from each region were not part of the same genetic unit. Divergence in the presence of gene flow across the ecotone between both regions might explain our results. Also, our findings indicate that the southern Atlantic forest may have been significantly affected by Pleistocene climatic alteration, although such events did not cause local extinction of most taxa, as occurred in other regions of the globe where forests were significantly affected by global glaciations. In this study, we addressed the phylogenetic and taxonomic relationships of Trypanosoma vivax and related trypanosomes nested in the subgenus Duttonella through combined morphological and phylogeographical analyses. We previously demonstrated that the clade T. vivax harbours a homogeneous clade comprising West African/South American isolates and the heterogeneous East African isolates. Herein we characterized a trypanosome isolated from a nyala antelope (Tragelaphus angasi) wild-caught in Mozambique (East Africa) and diagnosed as T. vivax-like based on biological, morphological and molecular data. Phylogenetic relationships, phylogeographical patterns and estimates of genetic divergence were based on SSU and ITS rDNA sequences of T. vivax from Brazil and Venezuela (South America), Nigeria (West Africa), and from T. vivax-like trypanosomes from Mozambique, Kenya and Tanzania (East Africa). Despite being well-supported within the T. vivax clade, the nyala trypanosome was highly divergent from all other T. vivax and T. vivax-like trypanosomes, even those from East Africa. Considering its host origin, morphological features, behaviour in experimentally infected goats, phylogenetic placement, and genetic divergence this isolate represents a new genotype of trypanosome closely phylogenetically related to T. vivax. This study corroborated the high complexity and the existence of distinct genotypes yet undescribed within the subgenus Duttonella.

A alface-d'água (Pistia stratiotes) é uma das principais entre as macrófitas aquáticas que causam problemas em corpos hídricos no Brasil e são consideradas como plantas daninhas. O presente trabalho foi realizado com os objetivos de conhecer melhor a variabilidade genética dessa macrófita e relacionar essa variabilidade com a resposta à aplicação do herbicida glyphosate. Para isso, foram coletados indivíduos em 12 corpos hídricos em diferentes cidades do território nacional (Americana, Cambaratiba, Curitiba, Itapura, Jaboticabal, Lagoa Santa, Piraí, Rio Grande, Rubinéia, Salto Grande, Santa Gertrudes e Três Lagoas). Os acessos foram caracterizados pelo uso de marcadores RAPD (DNA Polimórfico Amplificado ao Acaso), que permitiram, com o auxílio de iniciadores aleatórios, a caracterização dos locos polimórficos identificados por uma matriz de ausência e presença de bandas. Utilizando essa matriz, a análise de agrupamento permitiu nítida classificação dos acessos em três grupos com diferenças genéticas entre eles. Um ensaio de controle químico, com plantas mantidas em vasos plásticos (5 L) e pulverizadas com o herbicida glyphosate nas concentrações de 0,0, 0,6, 1,2, 1,8 e 2,4 kg ha-1, identificou, utilizando avaliações aos 7, 14 e 21 dias após aplicação, que as duas maiores doses promoveram melhor efeito herbicida. Foi verificado também que os acessos de Curitiba e Cambaratiba apresentaram menor suscetibilidade ao herbicida glyphosate. Não houve correspondência entre a estrutura de grupos dos acessos pela análise multivariada de agrupamento com a técnica RAPD e a suscetibilidade da alface-d'água ao glyphosate.

A divergência genética de 16 cultivares de arroz quanto à resistência ao percevejo-do-colmo-do-arroz, Tibraca limbativentris Stål, foi avaliada por meio de técnicas de análise multivariada. O experimento foi conduzido em casa de vegetação, em delineamento experimental de blocos ao acaso, com oito repetições. Na avaliação foram considerados oito caracteres de resistência ao ataque do inseto. A divergência genética foi avaliada por procedimentos multivariados: distância generalizada de Mahalanobis (D²), método de agrupamento de otimização de Tocher e técnica de variáveis canônicas. As cultivares mais dissimilares foram Bico Ganga e Marabá Branco, enquanto Agulha e Branco Tardão foram as mais similares. Foram formados cinco grupos pelo método de otimização de Tocher. As três primeiras variáveis canônicas explicaram 88,5% da variabilidade total disponível. Conclui-se que as técnicas de análise multivariada são eficientes para a análise da divergência genética entre as cultivares de arroz, com destaque para Marabá Branco e Bico Ganga, consideradas as mais promissoras a serem utilizadas em futuros cruzamentos para melhoramento visando resistência ao percevejo-do-colmo-do-arroz.

Marcadores moleculares fAFLP foram utilizados para estimar a diversidade genética entre 36 acessos de maracujá-amarelo (Passiflora edulis f. flavicarpa Deg.) coletados em 18 estados do Brasil. Os resultados obtidos permitiram concluir que os marcadores fAFLP se mostraram consistentes na avaliação da variabilidade genética, detectando e quantificando a ampla divergência genética entre os 36 acessos analisados, bem como a não-formação de estruturação geográfica. Tais resultados podem auxiliar na definição de estratégias mais eficientes a serem utilizadas em programas de melhoramento de maracujá-amarelo.

Avaliou-se a divergência genética entre quinze linhagens (Hav 13, Hav 14, Hav 21, Hav 22, Hav 25, Hav 38, Hav 40, Hav 41, Hav 49, Hav 53, Hav 56, Hav 64, Hav 65, Hav 67 e Hav 68) e cinco cultivares (Macarrão Favorito AG480, Macarrão Preferido AG482, Manteiga Maravilha AG481, Teresópolis AG484 e Macarrão Bragança) de feijão-vagem de crescimento indeterminado, utilizando-se vinte características agronômicas. O ensaio foi conduzido na AGENCIARURAL - EE de Anápolis, no período de 30/04 a 10/08/1998. Os dados foram submetidos às análises de variância e multivariada (distância D² de Mahalanobis e o método de agrupamento de Tocher). Houve diferenças significativas entre os genótipos para as características consideradas. Os genótipos Hav 13, Hav 49, Hav 56, Hav 64, Hav 68, Favorito AG480 e Teresópolis AG484 destacaram-se com relação ao conjunto de características favoráveis a produtores e consumidores. Houve maior freqüência de pares com maiores distâncias, quando um dos componentes era a cultivar Teresópolis AG484 ou Hav 49, e de pares com menores distâncias quando seus componentes tiveram como ancestral comum a linhagem Hab 229. Os genótipos distribuíram-se em quatro grupos, sendo um constituído exclusivamente pela linhagem Hav 49, outro englobando as cultivares Manteiga Maravilha AG481 e Teresópolis AG484. A linhagem Hav 41 e as cultivares Macarrão Favorito AG480 e Macarrão Preferido AG482 um terceiro grupo, e os demais genótipos um único grupo. As características que mais contribuíram para a divergência entre os genótipos foram o número de dias para o início de floração e o comprimento das vagens, com 58,11% do total, seguidas da porcentagem de palha na vagem seca, da largura das vagens, das alturas das plantas nas duas épocas avaliadas, do peso médio de vagem e do número de vagens por planta que, em conjunto, contribuíram com 85,73% do total.

The aim of this study to evaluate by means of quantitative traits, the genetic dissimilarity among sunflower cultivars in different locations, in addition to the agreement between the methods in order to extract lines for future crosses. There were eight sunflower hybrids grown in two locations in northwestern Rio Grande do Sul, mainly with soil type Oxisol. Multivariate methods were used to determine the genetic diversity, using the Mahalanobis distance. Although the different yield between locations and cultivars, methods of grouping agreed among them selves. To obtain segregating populations, regardless of location, the cultivate 'Olisun 5', demonstrated greater potential for hybridization, with major contributions through number of achenes per chapter and height of the insertion section.

The fauna of Brazilian reef fishes comprises approximately 320 species distributed along the coast of the mainland and islands ocean. Little is known about the levels of connectivity between their populations, but has been given the interest in the relations between the offshore and the islands of the Brazil, in a biogeographical perspective. The oceanic islands Brazilian hosting a considerable number of endemic species, which are locally abundant, and divide a substantial portion of its reef fish fauna with the Western Atlantic. Among the richest families of reef fish in species are Pomacentridae. This study analyzed through analysis of sequences of the mitochondrial DNA control region (D-loop), the standards-breeding population of C. Multilineata in different areas of the NE coast of Brazil, involving both oceanic islands (Fernando de Noronha Archipelago and of St. Peter and St. Paul) and continental shelf (RN and BA). To this aim, partial sequences were used in the region HVR1 of mtDNA (312pb). The population structure and parameters for the estimates of genetic variability, molecular variance (AMOVA), estimation of the index for fixing (FST) and number of migrants were determined. The phylogenetic relationships between the populations were estimated using neighbor-joining (NJ) method. A group of Bayesian analysis was used to verify population structure, according to haplotype frequency of each individual. The genetic variability of populations was extremely high. The populations sampled show moderate genetic structure, with a higher degree of genetic divergence being observed for the sample of the Archipelago of St. Peter and St. Paul. At smaller geographical scale, the sample of Rio Grande do Norte and the Archipelago of Fernando de Noronha do not have genetic differentiation. Three moderately differentiated population groups were identified: a population group (I), formed by the Rio Grande do Norte (I') and the archipelago of Fernando de Noronha (I''), and two other different groups formed by the island population of the archipelago of Saint Peter and St. Paul (II) and Bahia (III). The genetic patterns found suggest that the species has suffered a relatively recent radiation favoring the absence of shared haplotypes. C. multilineata seems to constitute a relatively homogenous population along the West Atlantic coast, with evidence of a moderate population genetic structure in relation to the Archipelago of St. Peter and St. Paul. These data supports the importance of the dispersal larvae by marine current and the interpopulation similarity this species.

The objective of this work was to study the effect of selective thinning on! the genetic divergence in progenies of Pinus caribaea var. bahamensis, aiming to identify the most productive and divergent progenies for the use of improvement program. The test of progenies containing 119 progenies and two commercial controls were planted in March 1990, using 11 x 11 square lattice design, sextuple, partially balanced, disposed in lineal plots with six trees in the spacing of 3,0 x 3,0m. 13 years after planting thinning was realized (selection for DBH), with 50% selection intensity based on Multi-effect index, leaving three trees per plot in all the experiment. The evaluations were done at four situations: A (before the thinning); B (thinned trees); C (remaining trees after thinning) and D (one year after thinning). The analyzed traits were: height, diameter at breast height (DBH), volume, form of stem and wood density. The genetic divergence among the progenies was studied with aid of the canonical variables and of clustering of Tocher method using the generalized distance matrix of Mahalanobis (D(2)) as estimate of the genetic similarity. The progenies were grouped in four groups in situation A, fourteen in the situation B, two in the situation C and three in the situation D. The selective thinning of the trees within of the progenies caused a change in the genetic divergence among the progenies, genetically homogenizing the progenies, as demonstrated by the generalized distances of Mahalanobis, clustering of Tocher' and canonical variables methods; The. thinning made possible a high uniformity in respect to the relative contribution, of the traits for the total genetic divergence. The techniques, of clustering were efficient to identify groups of divergent,progenies for the use hybridization and little divergent progenies for the use in backcross program.

The present research has proposed to estimate the genetic variation of growth traits and to estimate the expected gain by multi-effect index (MEI), in order to transform a Pinus caribaea var. caribaea progeny trial into a seedling seed orchard. The progeny trial was set up in 1989, in Selviria, MS, Brazil, using a 10 x 10 triple lattice design, with 99 progenies and a commercial control, with linear plots of ten plants, by the 3 x 3 m spacing between plants and rows. Total plant height, diameter at breast height (Dbh), wood volume, stem form, wood density at breast height, and survival were the evaluated quantitative traits. The trial was measured through 14, 15, and 16 years old. The 50% intensity of thinning at 14.3 years old was done. No significant was the genetic variation of different traits. Heritability estimates have presented low magnitude with low variation by the different ages. The application of MEI to DBH, at two years after thinning, resulted in higher gains than the selection of within and among progenies. The best selection strategy to obtain higher gains and to keep genetic diversity is to select until five plants per progenies.

Neoplecostomus paranensis Langeam, 1990, from the upper Rio Parana, is the only Neoplecostomus species described in this basin and is distinguished from its congeners by the lack or reduction of the adipose fin. Neoplecostomus specimens with a normal and always present adipose fin were caught in the Rio Corumba, upper Rio Parana basin. In the present study two samples of populations, one from a tributary of Rio Paranapanema (identified as a typical N. paranensis) and the other from the Rio Corumba were compared through allozyme electrophoresis. Six diagnostic loci were found, Acp-A, Adh-A, Est-A, Gpi-A, Ldh-A and Ldh-B. In addition, the locus Gpi-B showed significant differences between allelic frequencies for the two samples. Nei's genetic identity between the populations was 0.731. The expressive genetic divergence together with the presence of an adipose fin show that the sample from the Rio Corumba is distinct from N. paranensis and probably represents a new species.
