Problem High plasma levels of tumor necrosis factor-alpha (TNF-alpha) in pregnant women have been associated with the pathogenesis of pre-eclampsia (PE). This study evaluated TNF-alpha plasma levels and monocyte production in gestational hypertension (GH) and PE during gestation and at puerperium.Method of study This study included 128 women, of whom 20 were non-pregnant (NP) normotensive (NT), and 108 were pregnant: 36 NT, 27 with GH, and 45 with PE. Peripheral blood plasma was used for TNF-alpha and uric acid determination. TNF-alpha was determined in plasma and lipopolysaccharide (LPS)-stimulated and non-stimulated monocyte supernatants by L929 bioassay.Results Tumor necrosis factor-alpha and uric acid plasma levels were higher in PE than in GH pregnancies. In both hypertensive groups, these parameters positively correlated and were significantly more elevated than in NT and NP women. TNF-alpha plasma levels and monocyte production were higher in hypertensive than in NT women during gestation, and significantly decreased at puerperium. Although decreased, TNF-alpha release in LPS-stimulated PE monocytes, was still significantly higher than in the other pregnant groups.Conclusion In vivo monocyte activation in GH and PE pregnant women was characterized by in vitro TNF-alpha production. The fact that higher circulating concentrations of TNF-alpha and uric acid were observed in PE than in GH suggests an association with disease severity.

Development within the cleidoic egg of birds and reptiles presents the embryo with the problem of accumulation of wastes from nitrogen metabolism. Ammonia derived from protein catabolism is converted into the less toxic product urea or relatively insoluble uric acid. The pattern of nitrogen excretion of the green iguana, Iguana iguana, was determined during embryonic development using samples from allantoic fluid and from the whole homogenized egg, and in hatchlings and adults using samples of blood plasma. Urea was the major excretory product over the course of embryonic development. It was found in higher concentrations in the allantoic sac, suggesting that there is a mechanism present on the allantoic membrane enabling the concentration of urea. The newly hatched iguana still produced urea while adults produced uric acid. The time course of this shift in the type of nitrogen waste was not determined but the change is likely to be related to the water relations associated with the terrestrial habit of the adult. The green iguana produces parchment-shelled eggs that double in mass during incubation due to water absorption; the eggs also accumulate 0.02. mM of urea, representing 82% of the total measured nitrogenous residues that accumulate inside the allantois. The increase in egg mass and urea concentration became significant after 55. days of incubation then were unchanged until hatching. © 2012 Elsevier Inc.

High-grade prostate cancers express high levels of matrix metalloproteinases (MMPs), major enzymes involved in tumor invasion and metastasis. However, the tumor cell lines commonly employed for prostate cancer research express only small amounts of MMPs when cultivated as monolayer cultures, in common culture media. The present study was conducted to ascertain whether culture conditions that include fibronectin can alter MMP2 and MMP9 expression by the human prostatic epithelial cell lines RWPE-1, LNCaP and PC-3. These cells were individually seeded at 2×104cells/cm2, cultivated until they reached 80% confluence, and then exposed for 4h to fibronectin, after which the conditioned medium was analyzed by gelatin zymography. Untreated cells were given common medium. Only RWPE-1 cells express detectable amounts of MMP9 when cultivated in common medium, whereas the addition of fibronectin induced high expression levels of pro and active forms of MMP2 in all tested cell lines. Our findings demonstrate that normal and tumor prostate cell lines express MMP2 activity when in contact with extracellular matrix components or blood plasma proteins such as fibronectin. Future studies of transcriptomes and proteomes in prostate cancer research using these cell lines should not neglect these important conclusions. © 2012 Elsevier Inc..

Background: To investigate mechanisms of fetal-maternal cell interactions in the bovine placenta, we developed a model of transgenic enhanced Green Fluorescent Protein (t-eGFP) expressing bovine embryos produced by nuclear transfer (NT) to assess the distribution of fetal-derived products in the bovine placenta. In addition, we searched for male specific DNA in the blood of females carrying in vitro produced male embryos. Our hypothesis is that the bovine placenta is more permeable to fetal-derived products than described elsewhere. Methodology/Principal Findings: Samples of placentomes, chorion, endometrium, maternal peripheral blood leukocytes and blood plasma were collected during early gestation and processed for nested-PCR for eGFP and testis-specific Y-encoded protein (TSPY), western blotting and immunohistochemistry for eGFP detection, as well as transmission electron microscopy to verify the level of interaction between maternal and fetal cells. TSPY and eGFP DNA were present in the blood of cows carrying male pregnancies at day 60 of pregnancy. Protein and mRNA of eGFP were observed in the trophoblast and uterine tissues. In the placentomes, the protein expression was weak in the syncytial regions, but intense in neighboring cells on both sides of the fetal-maternal interface. Ultrastructurally, our samples from t-eGFP expressing NT pregnancies showed to be normal, such as the presence of interdigitating structures between fetal and maternal cells. In addition, channels-like structures were present in the trophoblast cells. Conclusions/Significance: Data suggested that there is a delivery of fetal contents to the maternal system on both systemic and local levels that involved nuclear acids and proteins. It not clear the mechanisms involved in the transfer of fetal-derived molecules to the maternal system. This delivery may occur through nonclassical protein secretion; throughout transtrophoblastic-like channels and/or by apoptotic processes previously described. In conclusion, the bovine synepitheliochorial placenta displays an intimate fetal-maternal interaction, similar to other placental types for instance human and mouse. © 2013 Pereira et al.

A dengue é causada pelo Vírus da dengue (VDEN - Flaviviridae, Flavivirus) e é considerada a arbovirose mais difundida no mundo. Atualmente, não há um animal que sirva como modelo para estudos sobre a patogênese do VDEN. Para investigar a susceptibilidade da espécie Callithrix penicillata ao VDEN foram inoculados amostras do VDEN-3 e do VDEN-2 de isoladas de casos humanos fatais. Para tal, vinte e dois animais foram infectados com VDEN-3 (3,23 × 103 PFU/mL) e, sessenta dias após a infecção (d.p.i.), 11 deles foram secundariamente infectados com VDEN-2 (4,47 × 104 PFU/mL). Sangue, plasma e soro, foram coletados diariamente durante os primeiros sete dias e em 15, 20, 45 e 60 d.p.i.. Foram investigados a produção de anticorpos IgM e inibidores da hemaglutinação, assim como foram análisados parâmetros hematologicos e bioquímicos e o perfil sérico de citocinas (IL-6, TNF-, IL-2, IFN-α, IL-4 e IL-5). A infecção primária (VDEN-3) revelou anticorpos IgM (15- 20 d.p.i.), anticorpos IH (15-60 d.p.i.), aumento dos níveis de TNF-α e IFN-γ e diminuição da IL-5. Na infecção secundária (VDEN-2), foi detectado anticorpos IgM (15-20 d.p.i.), anticorpos IH (15-60 d.p.i.) e diminuição dos níveis de IL-6, TNF-α, de IFN-γ e IL-5. Além disso, foi observado em ambas infecções leucopenia, neutropenia, linfocitopenia, monocitopenia, trombocitopenia, e aumentou da AST. O aumento dos níveis de INF-γ e TNF- α indicaram a ativação da resposta inflamatória, com a diferenciação para a resposta celular e supressão da proliferação de citocinas características da resposta humoral. A presença de anticorpos neutralizantes pode ter ocasionado a supressão da resposta inflamatória na infecção secundária o que pode ter levado a ausência de sinais de Febre Hemorrágica do Dengue/Sindrome do Choque do Dengue (FHD/SCD) durante a infecção secundária (VDEN- 2). Os resultados indicam que o primatas não humanos da espécie Callithrix penicillata demonstraram susceptibilidade à cepas de VDEN de origem humana, sugerindo que esses animais são um bom modelo para estudos da resposta imunológica da Febre do Dengue (FD), assim como para a avaliação de uma vacina tetravalente.
