In dieser Arbeit wurde zunächst ein humanisiertes Mausmodell entwickelt für die Analyse von humanen DCs in vivo. Darüber hinaus wurden erste Versuche mit Nanopartikelbeladenen DCs durchgeführt, mit der Intention, durch diese Kombination humane DCs zu untersuchen. Es wurden immunsupprimierte NOD/LtSz-scid IL2R (NSG) Mäuse verwendet und mit humanen CD34+ PBSCs transplantiert. Es wurden insgesamt 14 Modelle getestet, mit einer durchschnittlichen Humanisierungsrate von 76 %. In allen Modellen konnten ab Woche sechs nach Transplantation humane CD45+ Zellen sowie humane Bund NK-Zellen und CD14+ Monozyten gefunden werden. Darüber hinaus waren myeloide DC-Vorläuferzellen, konventionelle HLA DR CD11c DCs (cDCs) und plasmazytoide DCs (pDCs) vorhanden. Humane T-Zellen konnten nicht vor Woche 18 nach Transplantation beobachtet werden. Neben der Rekonstitution humaner DCs in peripheren Organen, wurde ebenfalls nach gewebsständigen DCs, insbesondere den Langerhans Zellen (LCs) der Epidermis geschaut. Waren humane LC vorhanden, konnten diese ab Woche zwölf nach Transplantation in der murinen Epidermis detektiert werden. Diese waren konstant bis in Woche 30 nach Transplantation nachweisbar. In Hinblick auf die Etablierung der DCs in diesem humanisierten Mausmodells wurden verschiedene Einflussgrößen getestet. IL-7 führte zu keiner veränderten Hämatopoese, wohingegen Flt3L zu einer Zunahme von CD14+ Monozyten und cDCs führte. Darüber hinaus konnte eine drastische Abnahmernhumaner B-Zellen beobachtet werden. Es zeigte sich, dass der Zeitpunkt der Flt3LrnApplikation einen entscheidenen Faktor für den Effekt von Flt3L auf die Rekonstitution humaner Zellen darstellt. Für die in dieser Arbeit durchgeführten funktionellen in vivo Studien, wurden humanisierten Mäusen alloreaktive CD8+ T-Zellen appliziert. Somit sollte die Funktionalität der rekonstituierten humanen APCs getestet werden. Es wurde deutlich, dass Monozyten und DCs ihre Funktionalität erst ab Woche 14 nach Transplantation zu entwickeln schienen,rnwohingegen B-Zellen bereits zu früheren Zeitpunkten als Zielzellen für die alloreaktiven T-Zellen dienten. Dies wurde durch den Rückgang der jeweiligen Zellen nach Applikation der T-Zellen sichtbar. Zu erwähnen ist, dass das Anwachsen einer humanen Hämatopoese stark spenderabhängig ist und somit keine allgemeingültigen Aussagen hinsichtlich der in vivo Funktion getroffen werden können. Um im Gewebe verbliebende APCs zu manipulieren gibt es verschiedene Möglichkeiten. Im Rahmen dieser Arbeit wurden auf Polystyren-basierende Nanopartikel getestet. Die verwendeten Partikel hatten eine Größe von 80 bis 160 nm und waren unfunktionalisiert oder mit Amino- bzw. Carboxy-Gruppen versehen. Zusätzlich wurden die Partikel mit BODIPY (Durchflusszytometrie und kLSM-Messungen), einem Infrarotnahem Farbstoff IR 780 (BFI-Messungen) und Platin (in vivo Messungen) beladen. Der Carboxy-funktionalisierte Partikel zeigte den geringsten Einfluss auf die Vitalität von humanen DCs, wohingegen der Amino-funktionalisierte Partikel bei steigender Konzentration toxisch wirkte. Bei unfunktionalisierten Partikeln stieg die Toxizität bei zunehmender Konzentration. Hinsichtlich der Expression diverser DC spezifischer Oberflächenmoleküle nach Beladung mit Nanopartikeln zeigte sich, dass allein der unfunktionalisierte, mit Lutensol AT50 hergestellte Partikel zu einer leichten Hochregulation von MHC-Klasse-II Molekülen führte. Die Expression von CD86 wurde im Gegenzug nur durch die Beladung mit den Amino-, bzw. Carboxy funktionalisierten Partikeln und dem unfunktionalisierten, mit SDS hergestellten Partikel leicht gesteigert. Trotz der teilweise leicht veränderten Expression von Oberflächenmarkern, konnte mit Hilfe von IFN-g ELISpots keine Beeinflussungrnder Funktion als APCs von Nanopartikel-beladenen DCs beobachtet werden. In den in vivo Untersuchungen zeigten alle vier Partikel eine konstante Zirkulation imrnOrganismus und konnten bis 96 h nach Applikation nachgewiesen werden. Alle Partikel konnten primär in der Leber detektiert werden, wobei der unfunktionalisierte, mit Lutensol AT50 hergestelle Partikel das weiteste Verbreitungsmuster zeigte. Erste Versuche im humanisierten Mausmodell zeigten keine Beeinflussung der Verteilung und Kinetik von Nanopartikeln durch die humane Hämatopoese. Mit dem in dieser Arbeit etablierten humanisierten Mausmodell ist es möglich, die Entwicklung, Differenzierung, Aktivierung und Funktionalität humaner DCs in vivo zu untersuchen. Darüber hinaus kann das gezielte Adressieren von DCs in vivo analysiert werden, was sowohl die Möglichkeit der Manipulation von DCs zur Vermeidung einer akuten GvHD bietet als auch Verwendung in anderen DC-vermittelten Therapien (z.B.Vakzinationsstudien) findet.

Cell differentiation are associated with activation of cell lineage-specific genes. The $LpS{\it 1}\beta$ gene of Lytechinus pictus is activated at the late cleavage stage. $LpS{\it 1}\beta$ transcripts accumulate exclusively in aboral ectoderm lineages. Previous studies demonstrated two G-string DNA-elements, proximal and distal G-strings, which bind to an ectoderm-enriched nuclear factor. In order to define the cis-elements which control positive expression of the $LpS{\it 1}\beta$ gene, the regulatory region from $-$108 to +17 bp of the $LpS{\it 1}\beta$ gene promoter was characterized. The ectoderm G-string factor binds to a G/C-rich region larger than the G-string itself and the binding of the G-string factor requires sequences immediately downstream from the G-string. These downstream sequences are essential for full promoter activity. In addition, only 108 bp of $LpS{\it 1}\beta\ 5\sp\prime$ flanking DNA drives $LpS{\it 1}\beta$ gene expression in aboral ectoderm/mesenchyme cells. Therefore, for positive control of $LpS{\it 1}\beta$ gene expression, two regions of 5$\sp\prime$ flanking DNA are required: region I from base pairs $-$762 to $-$511, and region II, which includes the G/C-rich element, from base pairs $-$108 to $-$61. A mesenchyme cell repressor element is located within region I.^ DNA-binding proteins play key roles in determination of cell differentiation. The zinc finger domain is a DNA-binding domain present in many transcription factors. Based on homologies in zinc fingers, a zinc finger-encoding gene, SpKrox-1, was cloned from S. purpuratus. The putative SpKrox-1 protein has all structural characteristics of a transcription factor: four zinc fingers for DNA binding; acidic domain for transactivation; basic domain for nuclear targeting; and leucine zipper for dimerization. SpKrox-1 RNA transcripts showed a transient expression pattern which correlates largely with early embryonic development. The spatial expression of SpKrox-1 mRNA was distributed throughout the gastrula and larva ectodermal wall. However, SpKrox-1 was not expressed in pigment cells. The SpKrox-1 gene is thus a marker of a subset of SMCs or ectoderm cells. The structural features, and the transient temporal and restricted spatial expression patterns suggest that SpKrox-1 plays a role in a specific developmental event. ^

Retinoids such as all-trans-retinoic acid (ATRA) are promising agents for cancer chemoprevention and therapy. ATRA can cause growth inhibition, induction of differentiation and apoptosis of a variety of cancer cells. These effects are thought to be mediated by nuclear retinoids receptors which are involved in ligand-dependent transcriptional activation of downstream target genes. Using differential display, we identified several retinoic acid responsive genes in the head and neck squamous carcinoma cells and lung cancer cells, including tissue type transglutaminase, cytochrome P450-related retinoic acid hydroxylase, and a novel gene, designated RAIG1. RAIG1 has two transcripts of 2.4 and 6.8 kbp, respectively, that are generated by alternative selection of polyadenylation sites. Both transcripts have the same open reading frame that encodes a protein comprised of 357 amino acid residues. The deduced RAIG1 protein sequence contains seven transmembrane domains, a signature structure of G protein-coupled receptors. RAIG1 mRNA is expressed at high level in fetal and adult lung tissues. Induction of RAIG1 expression by ATRA is rapid and dose-dependent. A fusion protein of RAIG1 and the green fluorescent protein was localized in the cell surface membrane and perinuclear vesicles in transiently transfected cells. The locus for RAIG1 gene was mapped to a region between D12S358 and D12S847 on chromosome 12p12.3-p13. Our study of the novel retinoic acid induced gene RAIG1 provide evidence for a possible interaction between retinoid and G protein signaling pathways.^ We further examined RAIG1 expression pattern in a panel of 84 cancer cell lines of different origin. The expression level varies greatly from very high to non-detectable. We selected a panel of different cancer cells to study the effects of retinoids and other differentiation agents. We observed: (1) In most cases, retinoids (including all-trans retinoic acid, 4HPR, CD437) could induce the expression of RAIG-1 in cells from cancers of the breast, colon, head and neck, lung, ovarian and prostate. (2) Compare to retinoids, butyrate is often a more potent inducer of RAIG-1 expression in many cancer cells. (3) Butyrate, Phenylacetate butyrate, (R)P-Butyrate and (S)P-Butyrate have different impact on RAIG1 expression which varies among different cell lines. Our results indicate that retinoids could restore RAIG1 expression that is down-regulated in many cancer cells.^ A mouse homologous gene, mRAIG1, was cloned by 5$\sp\prime$ RACE reaction. mRAIG1 cDNA has 2105 bp and shares 63% identity with RAIG1 cDNA. mRAIG1 encodes a polypeptide of 356 amino acid which is 76% identity with RAIG1 protein. mRAIG1 protein also has seven transmembrane domains which are structurally identical to those of RAIG1 protein. Only one 2.2 kbp mRAIG1 transcript could be detected. The mRAIG1 mRNA is also highly expressed in lung tissue. The expression of mRAIG1 gene could be induced by ATRA in several mouse embryonal carcinoma cells. The induction of mRAIG1 expression is associated with retinoic acid-induced neuroectoderm differentiation of P19 cells. Similarity in cDNA and protein sequence, secondary structure, tissue distribution and inducible expression by retinoic acid strongly suggest that the mouse gene is the homologue of the human RAIG1 gene. ^

Abacavir hypersensitivity is a severe hypersensitivity reaction which occurs exclusively in carriers of the HLA-B*57∶01 allele. In vitro culture of PBMC with abacavir results in the outgrowth of abacavir-reacting CD8+ T cells, which release IFNγ and are cytotoxic. How this immune response is induced and what is recognized by these T cells is still a matter of debate. We analyzed the conditions required to develop an abacavir-dependent T cell response in vitro. The abacavir reactivity was independent of co-stimulatory signals, as neither DC maturation nor release of inflammatory cytokines were observed upon abacavir exposure. Abacavir induced T cells arose in the absence of professional APC and stemmed from naïve and memory compartments. These features are reminiscent of allo-reactivity. Screening for allo-reactivity revealed that about 5% of generated T cell clones (n = 136) from three donors were allo-reactive exclusively to the related HLA-B*58∶01. The addition of peptides which can bind to the HLA-B*57∶01-abacavir complex and to HLA-B*58∶01 during the induction phase increased the proportion of HLA-B*58∶01 allo-reactive T cell clones from 5% to 42%. In conclusion, abacavir can alter the HLA-B*57∶01-peptide complex in a way that mimics an allo-allele ('altered self-allele') and create the potential for robust T cell responses.

Isochorismate is an important metabolite formed at the end of the shikimate pathway, which is involved in the synthesis of both primary and secondary metabolites. It is synthesized from chorismate in a reaction catalyzed by the enzyme isochorismate synthase (ICS; EC 5.4.99.6). We have purified ICS to homogeneity from elicited Catharanthus roseus cell cultures. Two isoforms with an apparent molecular mass of 64 kD were purified and characterized. The Km values for chorismate were 558 and 319 μm for isoforms I and II, respectively. The isoforms were not inhibited by aromatic amino acids and required Mg2+ for enzyme activity. Polymerase chain reaction on a cDNA library from elicited C. roseus cells with a degenerated primer based on the sequence of an internal peptide from isoform II resulted in an amplification product that was used to screen the cDNA library. This led to the first isolation, to our knowledge, of a plant ICS cDNA. The cDNA encodes a protein of 64 kD with an N-terminal chloroplast-targeting signal. The deduced amino acid sequence shares homology with bacterial ICS and also with anthranilate synthases from plants. Southern analysis indicates the existence of only one ICS gene in C. roseus.

Bacterial lipopolysaccharide (LPS) is a potent stimulator of B-cell activation, proliferation, and differentiation. We examined the genetic response of B-lineage cells to LPS via trapping of expressed genes with a gene-trap retrovirus. This analysis showed that expression of only a small fraction of genes is altered during LPS stimulation of B-lineage cells. Isolation of the cellular portion of the trapped LPS-response genes via 5' RACE (rapid amplification of cDNA ends) cloning identified novel genes for all the cloned loci. These novel LPS-response genes were also found to have differentiation stage-restricted expression within the B-lymphoid lineage. That LPS-response genes in B cells also have differentiation stage-restricted expression suggests that these genes may be involved in the control of B-cell function and differentiation, since the known members of this class of genes have frequently been found to play a role in the function and differentiation of B-lineage cells. The isolation of novel members of this class of genes, including a gene that contains a putative SH2 domain, will further increase our understanding of the molecular events involved in the control of B-cell differentiation and function.

We have developed a paracrine signaling assay capable of mimicking inductive events in the early vertebrate embryo. RNA encoding one or more secreted proteins is microinjected into a Xenopus laevis oocyte. After a brief incubation to allow translation, a piece of embryonic tissue competent to respond to the signaling protein is grafted onto the oocyte. The secreted protein's effect on the grafted explant is then scored by assaying expression of tissue-specific markers. Explants of ectodermal tissue from blastula or gastrula stage embryos were grafted onto oocytes that had been injected with RNA encoding activin or noggin. We found that the paracrine assay faithfully reconstitutes mesoderm induction by activin and neural induction by noggin. Blastula-stage explants grafted onto activin-expressing oocytes expressed the mesodermal marker genes brachyury, goosecoid, and muscle actin. Gastrula-stage explants grafted onto noggin-expressing oocytes expressed neural cell adhesion molecule (NCAM) and formed cement gland. By injecting pools of RNA synthesized from a cDNA expression library into the oocyte, we also used the assay to screen for secreted neural-inducing proteins. We assayed 20,000 independent transformants of a library constructed from LiCl-dorsalized Xenopus laevis embryos, and we identified two cDNAs that induced neural tissue in ectodermal explants from gastrula-stage embryos. Both cDNAs encode noggin. These results suggest that the paracrine assay will be useful for the cloning of novel signaling proteins as well as for the analysis of known factors.

Sulfate (SO42-) is required for bone/cartilage formation and cellular metabolism. sat-1 is a SO42- anion transporter expressed on basolateral membranes of renal proximal tubules, and is suggested to play an important role in maintaining SO42- homeostasis. As a first step towards studying its tissue-specific expression, hormonal regulation, and in preparation for the generation of knockout mice, we have cloned and characterized the mouse sat-1 cDNA (msat-1), gene (sat1; Slc26a1) and promoter region. msat-1 encodes a 704 amino acid protein (75.4 kDa) with 12 putative transmembrane domains that induce SO42- (also oxalate and chloride) transport in Xenopus oocytes. msat-1 mRNA was expressed in kidney, liver, cecum, calvaria, brain, heart, and skeletal muscle. Two distinct transcripts were expressed in kidney and liver due to alternative utilization of the first intron, corresponding to an internal portion of the 5'-untranslated region. The Sa1 gene (similar to6 kb) consists of 4 exons. Its promoter is similar to52% G+C rich and contains a number of well-characterized cis-acting elements, including sequences resembling hormone responsive elements T3REs and VDREs. We demonstrate that Sat1 promoter driven basal transcription in OK cells was stimulated by tri-iodothyronine. Site-directed mutagenesis identified an imperfect T3RE at -454-bp in the Sat1 promoter to be responsible for this activity. This study represents the first characterization of the structure and regulation of the Sat1 gene encoding a SO42-/chloride/oxalate anion transporter.

水母雪莲(Saussurea medusa Maxim)为名贵珍稀中药材,其主要药用成分为类黄酮,尤其是3-脱氧类黄酮。目前关于雪莲的研究主要集中在采用细胞培养生产类黄酮等方面,但对于雪莲类黄酮生物合成的分子机制了解甚少,极大限制了这一珍贵资源的利用。本研究采用水母雪莲红色系愈伤组织及悬浮细胞为材料,构建cDNA文库,从中克隆水母雪莲类黄酮次生代谢中的相关基因并对这些基因进行了深入的生物信息学分析、转基因研究初步确定其功能,以期了解雪莲类黄酮次生代谢的分子机制,为提高类黄酮的合成奠定基础。主要结果如下: 1. 成功地构建了水母雪莲红色系愈伤组织与悬浮细胞cDNA文库,原始文库滴度达到4×106pfu/ml,扩增文库滴度接近1011 pfu/ml,重组率达98%。PCR检测插入片段,均在0.5kb到3kb之间,1kb以上占62%。从文库中检测到了chs、dfr及Myb转录因子SmP,文库覆盖度达到要求且为PCR筛选文库提供了可能。 2. 采用部分简并引物,通过RT-PCR克隆了水母雪莲查尔酮异构酶基因Smchi特异探针,并根据这一探针序列设计特异引物,采用TD-PCR法筛选cDNA文库,获得Smchi cDNA序列,全长831bp,编码一个232氨基酸残基的蛋白。根据cDNA序列克隆了Smchi DNA序列,结果表明Smchi基因无内含子。Smchi cDNA序列与翠菊chi基因高度同源,ORF区域同源性高达84%,但推测氨基酸序列则只有79.3%。Smchi mRNA具有复杂的二级结构。SmCHI具有典型的Chalcone结构域,其二级结构与苜蓿CHI蛋白十分相似,7个α-螺旋与8个延伸链由随机结构联系起来。但其活性中心的第三个关键氨基酸残基N115为M115所取代,这一取代可能导致该蛋白无生物活性,也可能使它具有一般CHI不同的功能。构建Smchi正义、反义真核表达载体,通过农杆菌介导导入烟草,获得转正义、反义Smchi基因的烟草。转基因烟草花色未改变,但叶片总黄酮发生了显著的变化,50%转正义基因烟草总黄酮含量显著提高,最高比对照提高6倍,70%转反义基因烟草总黄酮含量显著下降,最多达85.1%,初步证明Smchi具有功能,并能有效调控烟草类黄酮次生代谢。因此,SmCHI可能是不同于已知CHI的一类新的CHI蛋白,它催化的反应可能与花色素合成无关,其反应机制也可能有所不同。 3. 伴随Smchi的克隆获得了一个黄烷酮3-羟化酶类似基因Smf3h的cDNA,全长1334bp,编码一个343aa的蛋白。根据这一cDNA序列克隆了Smf3h DNA序列,全长1630bp,结果表明该基因由4个外显子和3个内含子组成。Smf3h mRNA具有十分复杂的二级结构。 推测蛋白氨基酸同源性分析表明,SmF3H属于2OG-FeII_Oxy家族,与同一家族的的颠茄H6H的同源性为45%,与拟南芥F3H的同源性为40%,但对SmF3H、典型F3H及典型H6H推测蛋白二级结构、活性中心关键氨基酸残基的位置与相对距离、软件进行功能预测分析,发现SmF3H与F3H更相似。构建Smf3h的正义与反义真核表达载体,通过农杆菌介导导入烟草,但只获得一批转正义基因的烟草,反义基因导致烟草不能再生而未获得转反义基因烟草。转基因烟草花色未改变,叶片总黄酮也与对照相似,初步确认Smf3h与烟草类黄酮生物合成无关,而是一个既不属于f3h也不属于h6h的功能未确定的新基因。 4. 采用与克隆Smchi基因相似的方法,从cDNA文库中克隆了SmP基因cDNA,全长969bp,编码一个256 aa的蛋白质。根据cDNA序列克隆了SmP基因的DNA序列,结果表明,SmP基因无内含子。SmP基因cDNA 一级结构及mRNA二级结构预测分析表明,该基因A+T含量很高(63%),所形成二级结构以A-T配对为主,其稳定性可能较差。SmP推测蛋白序列具有R2R3-Myb转录因子的典型特征,在N-端具有两个Myb DNA-binding Domain,其二级结构与鸡Myb转录因子1A5J十分相似,与其他基因如水稻OsMYB、番茄ThMYB的同源区域主要集中在这一结构域,分别为71.3%和70.8%;C-端富含丝氨酸,与烟草NtMYB、葡萄VlMYB等类黄酮调控因子相似,都呈寡聚体分布,并具有相同的保守磷酸化位点S170与S206。构建SmP基因真核表达载体,通过农杆菌介导导入烟草,获得大量转基因烟草。转基因烟草花色未发生改变,但51%的转基因烟草叶片总黄酮含量都显著提高(0.5-6倍),表明SmP具有促进烟草类黄酮生物合成的功能,但所调控的支路与花色素合成无关。初步试验结果表明,转SmP基因烟草对蚜虫具有很高的抗性,可有效地抑制蚜虫在烟草上的生长,抑制率最高可达92%-100%。这一抗性与烟草中类黄酮的积累可能具有直接的联系,但还需要进一步的试验证明。 5. 与美国俄亥俄州立大学Erich Grotewold 博士实验室合作,完成了微型EST库50个克隆的测序并进行了分析,从中获得了水母雪莲花色素合酶基因SmANS及醛脱氢酶基因SmALDH的特异探针。根据SmANS特异探针设计引物,采用PCR从这50个克隆中筛选获得了SmANS的cDNA序列,全长1229bp,编码一个356aa的蛋白质。SmANS在cDNA水平上与同属的翠菊ANS基因高度同源,但同源区域集中在ORF区域,达到80%,mRNA 预测二级结构十分复杂;推测氨基酸序列与翠菊ANS同源性达到82.9%。SmANS属于2OG-FeII_Oxy家族,在2OG-FeII_Oxy结构域高度保守,与翠菊、甜橙ANS保守结构域同源性达到94%。预测蛋白二级结构以α-螺旋-β-折叠为主,由7个主螺旋和11个主β-折叠及随机结构连接而成,并具有2OG-FeII_Oxy家族活性中心的三个保守的组氨酸残基(His84、His235、His291)和一个天冬氨酸残基(Asp237)。 6. 根据微型EST库中获得的SmALDH特异探针设计引物,采用PCR从这50个克隆中筛选获得了SmALDH基因cDNA 序列,全长1664bp,编码一个491aa的蛋白质。SmALDH基因cDNA具有独特的碱基组成,3/-UTR富含A+T,占该区域碱基总量的80%,5/-UTR的A+T和G+C各占50%,比ORF区域(52%)还低,因此其mRNA二级结构中5/-UTR可以单独形成自身二级结构并且十分稳定,这可能影响基因的表达。这一现象在水稻、玉米等植物中也存在。SmALDH在cDNA水平上在ORF区域与拟南芥、藏红花、水稻等具有较高同源性,分别为64.03%、63.89%、63.72%,但在推测蛋白氨基酸序列水平上同源性反而较低,分别为54.9%、54.3%、54.0%。SmALDH缺少线粒体定位信号,为胞质醛脱氢酶,具有一个Aldedh 保守结构域,还具有与1OF7-H相似的以α-螺旋-β-折叠为主的二级结构,由10个主螺旋和15个主β-折叠及随机结构连接而成。由于ALDH在植物细胞乙醇发酵中具有解除醛类物质毒害的功能,因此SmALDH基因的克隆为改造细胞自身以适应发酵培养条件,解决水母雪莲细胞大规模培养中需氧问题提供了可能。

TNF receptor associated factor 1 (TRAF1) plays an important role in regulating the TNF signaling and protecting cells from apoptosis. In the present study, a TRAF1 gene has been cloned from grass carp (Ctenopharyngodon idella) by reverse transcription (RT)-PCR and rapid amplification of cDNA ends (RACE). The full-length cDNA is 2235 bp, including a 250 bp 5' UTR (untranslated region), a 1659 bp open reading frame, and a 326 bp 3'UTR. The polyadenylation signal (AATAAA, AATAA) and one mRNA instability motif (AUUUA) were found followed by a poly (A) tail in the 3'UTR. No signal peptide or transmembrane region has been found in the putative amino acids of grass carp TRAF1 (gcTRAF1). The putative amino acids of gcTRAF1 share 72% identity with the homologue in zebrafish. It is characterized by a zinc finger at the N-terminus and a TRAF domain (contains one TRAF-C and one TRAF-N) at the C-terminus. The identity of the TRAF domain among all the TRAF1 homologues in vertebrates varies from 52% to 58%, while the identities of TRAF-C were almost the same as 70%. The recombinant gcTRAF1 has been constructed successfully and expressed in Escherichia coli by using pET-32a expression vector. The polyclonal antibody for rabbit has been successfully obtained. The expression of gcTRAF1 in different organs was examined by real-time quantitative PCR and Western blotting, respectively. It was widely distributed in heart, head kidney, thymus, brain, gill, liver, spleen, and trunk kidney. This is the first report of TRAF1 homologue molecule found in fish. (c) 2007 Elsevier B.V. Tumor necrosis factor receptor-associated factor 2 (TRAF2) is a crucial component of almost the entire tumor necrosis factor receptor superfamily signaling pathway. In the present study, a TRAF2 gene has been cloned from grass carp (Ctenopharyngodon idella) by reverse transcription-polymerase chain reaction and rapid amplification of cDNA ends. The full-length cDNA is 3162 bp, including a 60 bp 5' untranslated region (UTR), a 1611 bp open reading frame, and a 1491 bp 3' UTR. The polyadenylation signal (AATAAA) and the mRNA instability motifs (ATTTTA, ATTTA) were followed by a poly(A) tail in the 3' UTR. No signal peptide or transmembrane region has been found in the putative amino acids of grass carp TRAF2 (gcTRAF2). Phylogenetic tree analysis clearly showed that gcTRAF2 is nearest to the TRAF2 gene of goldfish. The identity of gcTRAF2 with its homologs in other vertebrates ranges from 56% to 97%. It is characterized by one RING-type signature at the N-terminus, one zinc finger in the middle part, and one conserved TRAF domain consisting of a C-proximal (TRAF-C) subdomain and a N-proximal (TRAF-N) subdomain. The identity of TRAF-C among all TRAF2 homologs in vertebrates varies from 78% to 97%, whereas the identity of TRAF-N ranges from 56% to 100%. The recombinant gcTRAF2 has been expressed in Escherichia coli using pET-32a expression vector. The rabbit anti-gcTRAF2 polyclonal antibody was obtained. The expression of gcTRAF2 in different organs was examined by real-time quantitative polymerase chain reaction and Western blot analysis. It was widely distributed in heart, head kidney, thymus, brain, gill, liver, spleen, and trunk kidney. This is the first report of a TRAF2 homolog molecule in fish.

Using degenerate primers based on conserved regions of the UDP-glucose dehydrogenase (UDPGDH) gene, an initial 476-bp DNA fragment was amplified from the water-bloom forming cyanobacterium, Microcystis aeruginosa FACHB 905. TAIL-PCR and ligation-mediated PCR were used to amplify the flanking regions to isolate an about 2.5-kb genomic DNA fragment. Sequence analysis revealed an ORF encoding a putative 462 amino acid protein, designated Mud for Microcystis UDPGDH. The Mud amino acid sequence is closely related to UDPGDH sequences from cyanobacterium Synechocystis PCC6803 (73% identity, 81% similarity), and bacterium Bacillus subtilis (51% identity and 67% similarity). The cloned mud gene was expressed in Escherichia coli using the pGEX-4T-1 fusion expression vector system to generate a GST-Mud fusion protein that exhibited UDPGDH activity. The cytosolic fraction of M aeruginosa FACHB 905 was subjected to Western analysis with an anti-Mud antibody, which revealed a single band of approximately 49 kD, consistent with the deduced molecular mass of the enzyme. The Mud protein could thus be characterized as a UDP-glucose dehydrogenase, which was a key enzyme for polysaccharide synthesis and has, for the first time, been studied in algae.

Three β-hydroxysteroid dehydrogenase/Δ5-Δ4-isomerase (3β-HSD) catalyze the oxidative conversion of Δ5-3β-hydroxysteroids to the Δ4-3-keto configuration and is therefore essential for the biosynthesis of all classes of hormonal steroids, namely progesterone, glucocorticoids, mineralocorticoids, androgens, and estrogens. Using human 3β-HSD cDNA as probe, a human 3β-HSD gene was isolated from a λ-EMBL3 library of leucocyte genomic DNA. A fragment of 3β-HSD genomic DNA was also obtained by amplification of genomic DNA using the polymerase chain reaction. The 3β-HSD gene contains a 5′-untranslated exon of 53 base pairs (bp) and three successive translated exons of 232, 165, and 1218 bp, respectively, separated by introns of 129, 3883, and 2162 bp. The transcription start site is situated 267 nucleotides upstream from the ATG initiating codon. DNA sequence analysis of the 5′-flanking region reveals the existence of a putative TATA box (ATAAA) situated 28 nucleotides upstream from the transcription start site while a putative CAAT binding sequence is located 57 nucleotides upstream from the TATA box. Expression of a cDNA insert containing the coding region of 3β-HSD in nonsteroidogenic cells shows that the gene encodes a single 42-kDa protein containing both 3β-hydroxysteroid dehydrogenase and Δ5-Δ4-isomerase activities. Moreover, all natural steroid substrates tested are transformed with comparable efficiency by the enzyme. In addition to its importance for studies of the regulation of expression of 3β-HSD in gonadal as well as peripheral tissues, knowledge of the structure of the human 3β-HSD gene should permit investigation of the molecular defects responsible for 3β-HSD deficiency, the second most common cause of adrenal hyperplasia in children.

Rhabdomyosarcomas (RMS) are highly aggressive tumors that are thought to arise as a consequence of the regulatory disruption of the growth and differentiation of skeletal muscle progenitor cells. Normal myogenesis is characterized by the expression of the myogenic regulatory factor gene family but, despite their expression in RMS, these tumor cells fail to complete the latter stages of myogenesis. The RMS cell line RD-A was treated with 12-O-tetradecanoylphorbol-13-acetate to induce differentiation and cultured for 10 days. RNA was extracted on days 1, 3, 6, 8 and 10. A human skeletal muscle cDNA microarray was developed and used to analyze the global gene expression of RMS tumors over the time-course of differentiation. As a comparison, the genes identified were subsequently examined during the differentiated primary human skeletal muscle cultures. Prothymosin alpha (PTMA), and translocase of inner mitochondrial membrane 10 (Tim10), two genes not previously implicated in RMS, showed reduced expression during differentiation. Marked differences in the expression of PTMA and Tim10 were observed during the differentiation of human primary skeletal muscle cells. These results identify several new genes with potential roles in the myogenic arrest present in rhabdomyosarcoma. PTMA expression in RMS biopsy samples might prove to be an effective diagnostic marker for this disease.

Die vorliegende Dissertation beinhaltet Untersuchungen zur Expression und Funktion der respiratorischen Proteine Neuroglobin (Nbg) und Cytoglobin (Cygb) in Vertebraten. rnrnUm die Expression der Globine während der Entwicklung des Säugerhirns zu untersuchen, wurden die Hirne von Maus-Embryonen ab dem Fötalstadium MF10 bis zum Tag eins nach der Geburt (T1) mit Adulttieren verglichen. Quantifiziert wurde sowohl die mRNA- als auch die Protein-Expression. Beide Globine zeigten im Verlauf der Entwicklung einen stetigen Anstieg der mRNA-Expression, wobei Ngb zu Beginn in zehnfach höherer Konzentration vorlag und im zeitlichen Verlauf einen 130-fachen Anstieg zeigte. Cygb zeigte lediglich einen 16-fachen Anstieg bis zum Adultstadium. Auf Proteinebene konnte die Expressionszunahme beider Globine im Laufe der Entwicklung bestätigt werden. Weder in den hypoxieresistenten Frühembryonalstadien noch während der mit Sauerstoff-Stress verbundenen Geburt zeigte sich ein Expressionsmaximum. Dies spricht gegen eine Globin-Funktion in der Oxidanz-Abwehr. Eher ist zumindest Ngb mit der Reifung der Neurone und dem damit einhergehenden, gesteigerten oxidativen Stoffwechsel assoziiert.rnrnDes Weiteren sollte die zelluläre und intrazelluläre Lokalisation beider Globine anhand einer primären Zellkultur aus dem Hippocampus pränataler Ratten und in immortalen Zelllinien untersucht werden. Neuroglobin wurde dabei nur in Neuronen, nicht jedoch in Gliazellen nachgewiesen. Das Färbemuster war in allen Ngb-exprimierenden Zellen zytoplasmatisch. Cytoglobin wurde in der Primärkultur in den Neuronen jedoch ebenso in den mit anti-GFAP markierten Gliazellen beobachtet. In beiden Zellpopulationen war auch der Kern durch das CyGB-Antiserum markiert. rnEine genauere Untersuchung der intrazellulären Lokalisation sollte durch die Transfektion von Globin-pEGFP-Fusionsproteinen erfolgen. Nach Transfektion der Fusionskonstrukte wurde die GFP-Färbung bei beiden Globinen sowohl im Zytoplasma als auch im Kern beobachtet. Eine rein nukleäre Lokalisation, die insbesondere für Cygb von anderen Autoren postuliert wurde, konnte somit ausgeschlossen werden. rnrnIn primären Zellkulturen aus Cerebellum und Kortex, die mit Hilfe von Paraquat oxidativem Stress ausgesetzt wurden, wurde der Verlauf der Globin-mRNA-Expression mit dem unregulierten 18s rRNA-Referenzgen und mit den Antioxidanz-Enzymen Cu-Zn-SOD und Gpx verglichen. Neuroglobin zeigte einen Expressionsverlauf ähnlich dem der beiden Antioxidanz-Enzyme, jedoch liegt seine mRNA im Hirngewebe in hundertfach niedrigerer Menge als Cu-Zn-SOD und Gpx vor. Cytoglobin zeigte keine Veränderung der Expression. Eine Funktion der Globine im Sinne einer ROS-Abwehr kann aus den Befunden nicht abgeleitet werden. rnrnUntersuchungen von Tumor und Normalgewebe mittels eines cDNA-Cancer-Arrays zeigten, dass NGB in Tumoren verschiedenen Ursprungs nicht exprimiert wird, CyGB dagegen keine Änderung seiner Expression in Tumor versus Normalgewebe erfährt. Eine Induktion der beiden Globine z.B. durch Hypoxie in soliden Tumoren kann daher ausgeschlossen werden.rn
