O presente trabalho, visou o estudo de mutantes resistentes à antibióticos na bactéria fitopatogênica Xanthomonas campestris (Pammel) Dowson. Foram obtidos mutantes resistentes à cinco antibióticos: penicilina, estreptomicina, aureomicina, cloranfenicol e éritromicina. O incremento de resistência em relação à linhagem original foi 4 vêzes para a penicilina, 8 para o cloranfenicol, 32 para a aureomicina e pelo menos 16 e 128 vêzes para a éritromicina e estreptomicina respectivamente. Em nenhum dos casos, verificou-se resistência cruzada ou sensibilidade colateral.

Foi comparado o crescimento de uma linhagem de Xanthomonas campestris (Pammel) Dowson, com o crescimento de 3 linhagens dessa bactéria resistente à estreptomicina, penicilina e aureomicina. O crescimento da linhagem original, não diferiu do crescimento dos mutantes resistentes à estreptomicina e penicilina. O crescimento do mutante resistente à aureomicina foi mais lento do que o da linhagem original. O mutante resistente à estreptomicina apresentou o mesmo índice de crescimento, tanto em meio sem antibiótico, como em meio suplementado com a droga, o mesmo acontecendo com o mutante resistente à aureomicina. O mutante resistente à penicilina teve crescimento reduzido em meio suplementado com a droga em questão. Misturas de células sensíveis e resistentes à estreptomicina mantiveram-se constantes quanto à porcentagem de células sensíveis e resistentes inicialmente inoculadas, o mesmo acontecendo com a mistura de bactérias sensíveis e resistentes à penicilina. Houve uma variação nessas porcentagens, para o caso da aureomicina.

Foi estudada a resistência da bactéria fitopatogênica Xanthomonas campestris (Pammel) Dowson com relação a dois antibióticos: Tilosina e Vancomicina. Para a vancomicina, colônias resistentes apareceram até na mais alta concentração usada (256 mcg/ml.). Para a tilosina, em concentrações maiores que 128 mcg/ml não foram encontradas bactérias, resistentes. Tais resultados indicam que a X. campestris seguiu o modelo de "um só passo" para adquirir resistência a vancomicina e, o modelo de "múltiplos passos" com relação a resistência a tilosina.

No presente trabalho foi observado o comportamento das variedades Stoneville-20, Nu-16, Gregg, Mebane, RM2 e IAC-12, quando üioculadas com um isolamento de Xanthomanas malvacearum (E. F. Smiith) Dowson, em condições de casa de vegetação e das variedades Stoneville-20, Nu-16, RM4 e RM2, quando em condições de campo. Pelos dados dados, obtidos, as variedades Stoneville-20 e Nu-16 mostraram-se resistentes ao isolamento testado, enquanto que as variedades RM4, Gregg, Mebane e IAC-12 máostraram-se suscetíveis. A variedade RM2 testada continha plantas que apresentavam reações fisiológicas, de resistência e suscetibilidade. Dentre as, progênies obtidas de plantas da variedade RM2 selecionadas em campo como resistentes só aproximadamente 20% mostraram alto grau de resistência quando testadas em casa de vegetação.

No presente trabalho foi determinada a variabilidade de X. malvacearum, em condições de casa de vegetação, baseando-se na reação dos hospedeiros diferenciais para raças fisiológicas do patógeno. A técnica de inoculação empregada foi a de riscos feitos na página inferior das folhas e para avaliação dos sintomas foi adotada uma escala que variou de 1 a 5. Foi detectada a ocorrência das raças fisiológicas 3, 8 e 10. A linhagem do algodoeiro IAC RM3-4133 71/523 foi resistente à raça fisiológica 3, enquanto que as linhagens (Acala x Nu.16) 71/213 e IAC 12-2 71/170 foram resistentes às raças 3, 8 e 10.

En las colecciones del IRTA-Mas Bové se han caracterizado más de 150 clones de nogal procedentes de prospección, material extranjero y de selecciones de distintos programas de mejora. El material se ha caracterizado agronómicamente siguiendo los descriptores del IPGRI (1994). Actualmente existen fichas descriptivas muy amplias de 43 accesiones. Toda esta información sobre el comportamiento del material vegetal en las condiciones climáticas de Tarragona, tiene interés a nivel científico y es muy válida para posibles plantaciones con nuevo material. El estudio de los caracteres de sensibilidad a Bacteriosis y afección de la caída prematura de flores (PFA), en dos decendencias de nogal de cruzamientos controlados, ha permitido obvervar que estos caracteres tienen una herencia compleja. Se ha establecido la virulencia de distintos aislados de Xanthomonas arboricola pv. juglandis, en nogal. Los aislados más virulentos se han utilizado para establecer un modelo de predicción, en condiciones de laboratorio, de esta bacteria en nogal. La temperatura a la que la severidad de la enfermedad es máxima es 26ºC. A esta temperatura un aumento en la duración del período de humedad relativa elevada de 0-24 horas, produce un aumento de la afección. En los próximos años, este modelo de predicción se validará en campo. Si los resultados son positivos, permitirán racionalizar los tratamientos para la Bacteriosis en las plantaciones de nogal. Los primeros resultados de un control de la iluminación en el interior de las copas de los árboles, realizado mediante medidas de iluminación interceptadas con un ceptómetro Delta T (SunScan), han puesto de manifiesto que la gran influencia de la luz en el crecimiento anual de las ramas. Así, en la formación y poda de los árbles hay que tener presente estos resultados, en especial en las formaciones en ejes libres, y favorecer la máxima entrada de luz en el interior de la copa de los árboles.

We analyze how individual happiness is affected over time by nine major life events using a panel of British individuals. Our aim is to test for the existence of adaptation and anticipation effects. Adaptation effects are found for all the life events considered with the possible exception of unemployment. Anticipation effects precede events that are easily predicted such as marriage, separation and the birth of a child.

Two mechanisms have attracted considerable attention from researchers studying the effects of income on happiness: adaptation and social comparison. In this paper we study both mechanisms using a panel of British households. Besides dealing with the UK case in detail, the paper contributes to the literature by considering the two mecha- nisms together and testing for them both separately and jointly. Our results strongly support the existence of adaptation effects but find only weak evidence in favour of social comparison.

This study assesses the industrial relations application of the „loyalty-exit-voice‟ proposition. The loyalty concept is linked to reciprocal employer-employee arrangements and examined as a job attribute in a vignette questionnaire distributed to low and medium-skilled employees. The responses provided by employees in three European countries indicate that reciprocal loyalty arrangements, which involve the exchange of higher effort for job security, are one of the most desirable job attributes. This attribute exerts a higher impact on the job evaluations provided by unionised workers, compared to their non-union counterparts. This pattern is robust to a number of methodological considerations. It appears to be an outcome of adaptation to union mediated cooperation. Overall the evidence suggests that the loyalty-job evaluation profiles of unionised workers are receptive to repeated interaction and negative shocks, such as unemployment experience. This is not the case for the non-union workers. Finally, unionised workers appear to „voice‟ a lower job satisfaction, but exhibit low „exit‟ intentions, compared to the non-unionised labour.
Local adaptation and matching habitat choice in female barn owls with respect to melanic coloration.

Local adaptation is a major mechanism underlying the maintenance of phenotypic variation in spatially heterogeneous environments. In the barn owl (Tyto alba), dark and pale reddish-pheomelanic individuals are adapted to conditions prevailing in northern and southern Europe, respectively. Using a long-term dataset from Central Europe, we report results consistent with the hypothesis that the different pheomelanic phenotypes are adapted to specific local conditions in females, but not in males. Compared to whitish females, reddish females bred in sites surrounded by more arable fields and less forests. Colour-dependent habitat choice was apparently beneficial. First, whitish females produced more fledglings when breeding in wooded areas, whereas reddish females when breeding in sites with more arable fields. Second, cross-fostering experiments showed that female nestlings grew wings more rapidly when both their foster and biological mothers were of similar colour. The latter result suggests that mothers should particularly produce daughters in environments that best match their own coloration. Accordingly, whiter females produced fewer daughters in territories with more arable fields. In conclusion, females displaying alternative melanic phenotypes bred in habitats providing them with the highest fitness benefits. Although small in magnitude, matching habitat selection and local adaptation may help maintain variation in pheomelanin coloration in the barn owl.

Background and aims: Family-centred care is an expected standard in PICU and parent reported outcomes are rarely measured. The Dutch validated EMPATHIC questionnaire provides accurate measures of parental perceptions of family-centred care in PICU. A French version would provide an important resource for quality control and benchmarking with other PICUs. The study aimed to translate and to assess the French cultural adaptation of the EMPATHIC questionnaire. Methods: In September 2012, following approval from the developer, translation and cultural adaptation were performed using a structured method (Wild et al. 2005). This included forward-backward translation and reconciliation by an official translator, harmonization assessed by the research team, and cognitive debriefing with the target users' population. In this last step, a convenience sample of parents with PICU experience assessed the comprehensibility and cultural relevance of the 65-item French EMPATHIC questionnaire. The PICUs in Lausanne, Switzerland and Lille, France participated. Results: Seventeen parents, including 13 French native and 4 French as second language speakers, tested the cognitive equivalence and cultural relevance of the French EMPATHIC questionnaire. The mean agreement for comprehensibility of all 65 items reached 90.2%. Three items fell below the cut-off 80% agreement and were revised for inclusion in the final French version. Conclusions: The translation and the cultural adaptation permitted to highlight a few cultural differences that did not interfere with the main construct of the EMPATHIC questionnaire. Reliability and validity testing with a new sample of parents is needed to strengthen the psychometric properties of the French EMPATHIC questionnaire.

Many new gene copies emerged by gene duplication in hominoids, but little is known with respect to their functional evolution. Glutamate dehydrogenase (GLUD) is an enzyme central to the glutamate and energy metabolism of the cell. In addition to the single, GLUD-encoding gene present in all mammals (GLUD1), humans and apes acquired a second GLUD gene (GLUD2) through retroduplication of GLUD1, which codes for an enzyme with unique, potentially brain-adapted properties. Here we show that whereas the GLUD1 parental protein localizes to mitochondria and the cytoplasm, GLUD2 is specifically targeted to mitochondria. Using evolutionary analysis and resurrected ancestral protein variants, we demonstrate that the enhanced mitochondrial targeting specificity of GLUD2 is due to a single positively selected glutamic acid-to-lysine substitution, which was fixed in the N-terminal mitochondrial targeting sequence (MTS) of GLUD2 soon after the duplication event in the hominoid ancestor approximately 18-25 million years ago. This MTS substitution arose in parallel with two crucial adaptive amino acid changes in the enzyme and likely contributed to the functional adaptation of GLUD2 to the glutamate metabolism of the hominoid brain and other tissues. We suggest that rapid, selectively driven subcellular adaptation, as exemplified by GLUD2, represents a common route underlying the emergence of new gene functions.
