Abstract Background Serological tests to detect antibodies specific to Plasmodium vivax could be a valuable tool for epidemiological studies, for screening blood donors in areas where the malaria is not endemic and for diagnosis of infected individuals. Because P. vivax cannot be easily obtained in vitro, ELISA assays using total or semi-purified antigens are rarely used. Based on this limitation, we tested whether recombinant proteins representing the 19 kDa C-terminal region of the merozoite surface protein-1 of P. vivax (MSP119) could be useful for serological detection of malaria infection. Methods Three purified recombinant proteins produced in Escherichia coli (GST-MSP119, His6-MSP119 and His6-MSP119-PADRE) and one in Pichia pastoris (yMSP119-PADRE) were compared for their ability to bind to IgG antibodies of individuals with patent P. vivax infection. The method was tested with 200 serum samples collected from individuals living in the north of Brazil in areas endemic for malaria, 53 serum samples from individuals exposed to Plasmodium falciparum infection and 177 serum samples from individuals never exposed to malaria. Results Overall, the sensitivity of the ELISA assessed with sera from naturally infected individuals was 95%. The proportion of serum samples that reacted with recombinant proteins GST-MSP119, His6-MSP119, His6-MSP119-PADRE and yMSP119-PADRE was 90%, 93.5%, 93.5% and 93.5%, respectively. The specificity values of the ELISA determined with sera from healthy individuals and from individuals with other infectious diseases were 98.3% (GST-MSP119), 97.7% (His6-MSP119 and His6-MSP119-PADRE) or 100% (yMSP119-PADRE). Conclusions Our study demonstrated that for the Brazilian population, an ELISA using a recombinant protein of the MSP119 can be used as the basis for the development of a valuable serological assay for the detection of P. vivax malaria.

Immunizing pregnant women with a malaria vaccine is one approach to protecting the mother and her offspring from malaria infection. However, specific maternal Abs generated in response to vaccination and transferred to the fetus may interfere with the infant's ability to respond to the same vaccine. Using a murine model of malaria, we examined the effect of maternal 19-kDa C-terminal region of merozoite surface protein-1 (MSP1(19)) and Plasmodium yoelii Abs on the pups' ability to respond to immunization with MSP1(19). Maternal MSPI,g-specific Abs but not A yoelii-specific Abs inhibited Ab production following MSP1(19) immunization in 2-wk-old pups. This inhibition was correlated with the amount of maternal MSP1(19) Ab present in the pup at the time of immunization and was due to fewer specific B cells. Passively acquired Ab most likely inhibited the development of an Ab response by blocking access to critical B cell epitopes. If a neonate's ability to respond to MSP1(19) vaccination depends on the level of maternal Abs present at the time of vaccination, it may be necessary to delay immunization until Abs specific for the vaccinating Ag have decreased.

Resumen: No momento em que a Eneida foi composta, entre os anos 29 e 19 a.C, a memória da divinizaão de Csar se fazia presente, lançando também a expectativa sobre a consecratio do próprio Imperador. No poema épico, o prenúncio da apoteose de Augusto surge na mesma profecia em que Júpiter assegura a Vênus a acolhida de seu filho, Enéias, entre os imortais (VIRGÍLIO, Eneida, I.259- 289). Este tema reaparece nas Metamorfoses de Ovídio cujas linhas finais do livro XV retratam a transformaão de Júlio Csar em astro – sidus - e mencionam o dia em que o Princeps deverá receber, nos cus, as preces de seus governados (OVÍDIO, Metamorfoses, XV.745-879). Tendo em vista a instituição do culto imperial, problematizaremos nas obras mencionadas as relaões entre mito, memória e poder no Principado.

In a recent study, we demonstrated the immunogenic properties of a new malaria vaccine polypeptide based on a 19 kDa C-terminal fragment of the merozoite surface protein-1 (MSP1(19)) from Plasmodium vivax and an innate immunity agonist, the Salmonella enterica serovar Typhimurium flagellin (FliC). Herein, we tested whether the same strategy, based on the MSP1(19) component of the deadly malaria parasite Plasmodium falciparum, could also generate a fusion polypeptide with enhanced immunogenicity. The His(6)FliC-MSP1(19) fusion protein was expressed from a recombinant Escherichia coil and showed preserved in vitro TLR5-binding activity. In contrast to animals injected with His(6)MSP1(19), mice subcutaneously immunised with the recombinant His6FliC-MSP1(19) developed strong MSP1(19)-specific systemic antibody responses with a prevailing IgG1 subclass. Incorporation of other adjuvants, such as CpG ODN 1826, complete and incomplete Freund`s adjuvants or Quil-A, improved the IgG responses after the second, but not the third, immunising dose. It also resulted in a more balanced IgG subclass response, as evaluated by the IgG1/IgG2c ratio, and higher cell-mediated immune response, as determined by the detection of antigen-specific interferon-gamma secretion by immune spleen cells. MSP(19)-specific antibodies recognised not only the recombinant protein, but also the native protein expressed on the surface of P. falciparum parasites. Finally, sera from rabbits immunised with the fusion protein alone inhibited the in vitro growth of three different P. falciparum strains. In summary, these results extend our previous observations and further demonstrate that fusion of the innate immunity agonist FliC to Plasmodium antigens is a promising alternative to improve their immunogenicity. A menudo se ha debatido sobre la problemática del destinatario en las Geórgicas de Virgilio. En esta oportunidad, analizamos la figura discursiva del enunciatario didáctico, construido a los largo de la obra, en el marco de la teoría de la enunciación inaugurada a partir de los trabajos de E. Benveniste y nutrida por diversos aportes a lo largo del tiempo. La caracterización identitaria resultante permite configurarlo como un miembro masculino de la clase dominante, culta política y económicamente poderosa de la sociedad romana.

En el libro I de Geórgicas, el poeta despliega ampliamente sus conocimientos sobre el arte de la agricultura, la roturación de los campos, la preparación para la siembra, la elección de las simientes, etc. Siguiendo a Patricia Johnston (1980), se puede observar que la concepción virgiliana sobre la agricultura se transforma a lo largo de sus obras y el mito de las edades, de raigambre hesiódica, adquiere diferentes matices en un discurso que acompaa los cambios sociales. En la presente comunicación, nos proponemos analizar un pasaje que anuncia el retorno de la paz tempus veniet (vv.493497), su vínculo con la edad de oro y la agricultura, la representación de la virtus y la figura del poeta, también agente de la paz a partir de sus enseñanzas.

Este texto presenta problemas de traducción en algunos versos que merecen ser atentamente examinados para no oscurecer el significado del poema. Es necesario dilucidar la cosmovisión y la concepción del tiempo que el poeta ha querido transmitir para poder traducir con adecuación y la mayor precisión posible el texto y no quebrar su unidad y coherencia. Los textos problemáticos a examinar: v. 5 'ab integro'; v. 8 'puero, quo'; v. 11 'decus hoc aevi'; v. 3436: 'alter', 'altera', 'altera'; v. 60 'risu'; v. 62 'risere parentes / parentem /parenti'.

En la presente comunicación analizaremos de qué modo los mitos presentes en la geórgica I (Ceres; la Edad de Hierro; Deucalión; el perjurio de Laomedonte) confluyen a articular la noción de labor como la condición para la agricultura. El labor implica la dedicación al trabajo diario del campo (remover la tierra, vallar los campos, marcar el ganado, fabricar y mantener las herramientas, etc), conocer la topografía del cielo y venerar a los dioses para que favorezcan la producción. Tales indicaciones son constitutivas de la parte didáctica del texto y constituyen asimismo parte del labor del poeta, que con esta obra se inserta en el proyecto político económico y agrario de Augusto. Sin embargo, el poema trasciende esta problemática formando parte del proyecto poético de Virgilio.
