Zusammenfassung (deutsch) Seit den 1980iger Jahren wächst die Bedeutung der sog. Bildschaffenden Methoden für die Bestimmung der Qualität ökologischer Produkte. Zu diesen Methoden gehört die Biokristallisation, Steigbild und Rundfilter-Chromatographie. Die Ergebnisse dieser Methoden sind Bilder, die anhand definierter Kriterien ausgewertet werden. Bei der Biokristallisation sind es mehr oder weniger geordnete Kristallisationen auf einer Glasplatte, bei dem Steigbild zweidimensionale Strukturen auf Chromatographiepapier. In der Vergangenheit wurden die Bilder von Spezialisten ausgewertet, die nach einer längeren Schulung produktspezifische Kriterien entwickelt hatten. Im Gegensatz zur Dünnschicht-Chromatographie, wo der einzelne Stoff von der Matrix separiert wird, ist das Ziel beim Steigbild, Strukturen der möglichst ganzen Probe zu erzeugen. Die Methode wurde von Kolisko in den 1929iger Jahren entwickelt, wobei eine Kombination aus Chromatographieprozess und Metallkomplexreaktionen genutzt wurde. Die Firma WALA entwickelte die Methode für die Kontrolle ihrer Produkte und setze Silbernitrat und Eisensulfat ein. Bisher wurde die Methode qualitativ beschreibend ausgewertet, wobei einzelne Bildelemente und deren Interaktion beschrieben wurden. Deshalb musste für die vorliegende Arbeit Auswertungsmethoden entwickelt werden, mit denen auch eine statistische Bearbeitung der Ergebnisse möglich ist (nominale Unterscheidung von proben anhand der Bilder). Die Methode wurde bisher in einer Reihe von Studien eingesetzt (u.a. die Unterscheidung von Produktionsweisen). Obwohl die Bilder nur qualitativ ausgewertet wurden, konnten geschulte Prüfpersonen Proben aus verschiedenen Anbausystemen anhand der Bilder trennen. Die Ergebnisse wurden aber nicht so dokumentiert, dass sie den Erfordernissen internationaler Standardnormen für Laboratorien genügten. Deshalb mussten für diese Arbeit zunächst die Prozeduren dokumentiert und eine systematische Untersuchung zu den Einflussgrößen durchgeführt werden. Dazu wurde die visuelle Bildauswertung entwickelt und standardisiert. Die visuelle Bildauswertung basiert auf morphologischen Kriterien der Bilder von den untersuchten Weizen- und Möhrenproben. Ein Panel aus geschulten Personen entwickelte dann die Kriterien und legte sie anhand von Referenzbildern fest. Die Bilder der vorliegenden Arbeit wurden mit der einfach beschreibenden Prüfung ausgewertet, wie sie aus der sensorischen Prüfung von Lebensmitteln übernommen werden konnte. Mit geschulten und ungeschulten Prüfpersonen wurden Weizenproben und verschiedene Möhrensäfte mit der sog. Dreiecksprüfung ausgewertet (von ISO 4120). Alle Laborprozeduren wurden dokumentiert. Mit der Anwendung dieser Prozeduren wurden Vergleichsversuche mit Laboren in Dänemark und Holland (BRAD, LBI) durchgeführt. Die Ergebnisse waren sowohl für Weizen- als auch für Möhrenproben vergleichbar, wobei alle drei Labore zwischen jeweils zwei Proben unterscheiden konnten. Die systematische Untersuchung zu den Einflussgrößen zeigte, dass das Unterscheidungsvermögen der Methode vor allem von den klimatischen Bedingungen während der Steigphasen beeinflusst wird. Auch die Präkonditionierung der Papiere hat einen großen Einfluss, während die Wasserqualität (ultra-filtriert, de-ionisiert, destilliert) eine untergeordnete Bedeutung hat. Für Weizen- und Möhrenproben wurde sowohl die Wiederholbarkeit als auch die Reproduzierbarkeit getestet. Die Unterschiede in den Bildern der verschiedenen Proben waren dabei immer größer als die Variation durch Proben- und Bildwiederholung und das Labor. Die so charakterisierte Methode wurde auf kodierte Proben von definierten Feldversuchen und auf Marktproben (Paarvergleich von Anbausystemen ökologisch und konventionell) angewandt, wobei als Ergebnis mehr als 90% der Proben mit der einfach beschreibenden Prüfung anhand der Bilder unterschieden werden konnten. Die Auswertung mit der Dreiecksprüfung zeigte, dass sowohl Sorten und Verarbeitungsschritte (Saft) als auch Anbauweisen signifikant getrennt wurden. Darüber hinaus wurde die Methode auch erfolgreich auf Apfelproben angewandt. Weitere Untersuchungen müssen zeigen, ob sich das Potential der Methode, verschiedene Fragen wie die Authentizitätsprüfung von Lebensmitteln verifizieren lassen.

As tetraciclinas são compostos antibacterianos utilizados em bovinos de leite para tratamento de doenças infecciosas, como a mastite, mas também como aditivos em ração animal. O uso das tetraciclinas pode conduzir à presença de resíduos destes fármacos no leite, principalmente se não forem utilizados de acordo com as indicações, nem respeitado o período mínimo de eliminação dos antibióticos pelo leite. A presença de resíduos de antibióticos no leite interfere no processo industrial dos seus derivados, podendo inviabilizar a produção destes e, consequentemente, causar igualmente prejuízos económicos, como por exemplo, pela inibição de fermentos lácticos que são culturas de microorganismos utilizados na produção de iogurtes, queijos e outros produtos lácteos. Os resíduos de antibióticos no leite de consumo podem representar riscos à saúde humana, podendo causar reacções alérgicas em indivíduos sensíveis ou ter um efeito adverso na flora intestinal humana, prejudicando a sua acção protectora local, além de propiciar a selecção de populações bacterianas resistentes.(Denobile & Nascimento, 2004)

Plant root mucilages contain powerful surfactants that will alter the interaction of soil solids with water and ions, and the rates of microbial processes. The lipid composition of maize, lupin and wheat root mucilages was analysed by thin layer chromatography and gas chromatography-mass spectrometry. A commercially available phosphatidylcholine (lecithin), chemically similar to the phospholipid surfactants identified in the mucilages, was then used to evaluate its effects on selected soil properties. The lipids found in the mucilages were principally phosphatidylcholines, composed mainly of saturated fatty acids, in contrast to the lipids extracted from root tissues. In soil at low tension, lecithin reduced the water content at any particular tension by as much as 10 and 50% in soil and acid-washed sand, respectively. Lecithin decreased the amount of phosphate adsorption in soil and increased the phosphate concentration in solution by 10%. The surfactant also reduced net rates of ammonium consumption and nitrate production in soil. These experiments provide the first evidence we are aware of that plant-released surfactants will significantly modify the biophysical environment of the rhizosphere.

Parabens are used as preservatives in many thousands of cosmetic, food and pharmaceutical products to which the human population is exposed. Although recent reports of the oestrogenic properties of parabens have challenged current concepts of their toxicity in these consumer products, the question remains as to whether any of the parabens can accumulate intact in the body from the long-term, low-dose levels to which humans are exposed. Initial studies reported here show that parabens can be extracted from human breast tissue and detected by thin-layer chromatography. More detailed studies enabled identification and measurement of mean concentrations of individual parabens in samples of 20 human breast tumours by high-pressure liquid chromatography followed by tandem mass spectrometry. The mean concentration of parabens in these 20 human breast tumours was found to be 20.6 +/- 4.2 ng g(-1) tissue. Comparison of individual parabens showed that methylparaben was present at the highest level (with a mean value of 12.8 +/- 2.2 ng g(-1) tissue) and represents 62% of the total paraben recovered in the extractions. These studies demonstrate that parabens can be found intact in the human breast and this should open the way technically for more detailed information to be obtained on body burdens of parabens and in particular whether body burdens are different in cancer from those in normal tissues. The identification of lipophilic flavones and flavonols using a combination of high performance liquid chromatography, thin layer chromatography and UV spectral analysis is discussed. Data are provided for the flavones, apigenin, luteolin and tricetin and twelve of their methyl ethers, 8-hydroxyluteolin, 6-hydroxyluteolin and scutellarein and fourteen of their methyl ethers, and some 6,8-dihydroxyapigenin and 6,8-dihydroxyluteolin derivatives. Data for some forty two flavonols with extra 6- and/or 8-hydroxylation, mostly 6-hydroxykaempferol and quercetagetin derivatives, are also presented. The remaining compounds analysed include fourteen 5-deoxyflavones, four 5-methoxyflavones and five 5-deoxyflavonols plus further 5-hydroxylated flavones and flavonols without B-ring oxidation or with 2-, 5- or 6-hydroxylation. The isoprenoid metabolic pathway in protozoa of the Leishmania genus exhibits distinctive characteristics. These parasites, as well as other members of the Trypanosomatidae family, synthesize ergosterol, instead of cholesterol, as the main membrane sterol lipid. Leishmania has been shown to utilize leucine, instead of acetate as the main precursor for sterol biosynthesis. While mammalian dolichols are molecules containing 15-23 isoprene units, Leishmania amazonensis promastigotes synthesize dolichol of 11 and 12 units. In this paper, we show that the intracellular stages of L. amazonensis, amastigotes, synthesize mainly polyprenols of 9 isoprene units, instead of dolichol. (c) 2007 Elsevier Inc. Urban particulate matter (UPM) contributes to lung cancer incidence. Here, we have studied the mutagenic activity and DNA adduct-forming ability of fractionated UPM extractable organic matter (EOM). UPM was collected with a high-volume sampler in June 2004 at two sites, one at street level adjacent to a roadway and the other inside a park within the urban area of the city of Sao Paulo, Brazil. UPM was extracted using dichloromethane, and the resulting EOM was separated by HPLC to obtain PAH, nitro-PAH, and oxy-PAH fractions which were tested for mutagenicity with the Salmonella strains TA98 and YG1041 with and without S9 metabolic activation. The PAH fraction from both sites showed negligible mutagenic activity in both strains. The highest mutagenic activity was found for the nitro-PAH fraction using YG1041 without metabolic activation; however, results were comparable for both sites. The nitro-PAH and oxy-PAH fractions were incubated with calf thymus DNA under reductive conditions appropriate for the activation of nitro aromatic compounds, then DNA adduct patterns and levels were determined with thin-layer chromatography (TLC) (32)p-postlabeling method using two enrichment procedures-nuclease PI digestion and butanol extraction. Reductively activated fractions from both sites produced diagonal radioactive zones (DRZ) of putative aromatic DNA adducts on thin layer plates with both enrichment procedures. No such DRZ were observed in control experiments using fractions from unexposed filters or from incubations without activating system. Total adduct levels produced by the nitro-PAH fractions were similar for both sites ranging from 30 to 45 adducts per 10(8) normal nucleotides. In contrast, the DNA binding of reductively activated oxy-PAH fractions was three times higher and the adduct pattern consisted of multiple discrete spots along the diagonal line on the thin layer plates. However, DNA adduct levels were not significantly different between the sampling sites. Both samples presented the same levels of mutagenic activity. The response in the Salmonella assay was typical of nitroaromatics. Although, more mutagenic activity was related to the nitro-PAH fraction in the Salmonella assay, the oxy-PAH fractions showed the highest DNA adduct levels. More studies are needed to elucidate the nature of the genotoxicants occurring in Sao Paulo atmospheric samples. (C) 2008 Elsevier B.V. All rights reserved.

The present work had as objective the isolation of the five compounds by thin-layer Chromatography (TLC) from the essential oil of the Aloysia gratissima. For this, a number of systems of eluents were used for its separation, indicating that through the system acetone/hexane in proportions (v/v) 1:30 it was possible to isolate guaiol, elemol, pinocanphone (trans-3-pinanone), cis-pinocarvyl, and acorenone. The isolation of the compound acorenone from the other compounds was possible with the mixture of solvents hexane/dichloromethane in proportions (v/v) (1:1,3).

The oily sludge is a complex mix of hydrocarbons, organic impurities, inorganic and water. One of the major problems currently found in petroleum industry is management (packaging, storage, transport and fate) of waste. The nanomaterials (catalysts) mesoporous and microporous are considered promising for refining and adsorbents process for environment protection. The aim of this work was to study the oily sludge from primary processing (raw and treated) and vacuum residue, with application of thermal analyses technique (pyrolysis), thermal and catalytic pyrolysis with nanomaterials, aiming at production petroleum derived. The sludge and vacuum residue were analyzed using a soxhlet extraction system, elemental analysis, thin layer chromatography, thermogravimetry and pyrolysis coupled in gas chromatography/mass spectrometry (Py GC MS). The catalysts AlMCM-41, AlSBA-15.1 e AlSBA-15.2 were synthesized with molar ratio silicon aluminum of 50 (Si/Al = 50), using tetraethylorthosilicante as source of silicon and pseudobuhemita (AlOOH) as source of aluminum. The analyzes of the catalysts indicate that materials showed hexagonal structure and surface area (783,6 m2/g for AlMCM-41, 600 m2/g for AlSBA-15.1, 377 m2/g for AlSBA-15.2). The extracted oily sludge showed a range 65 to 95% for organic components (oil), 5 to 35% for inorganic components (salts and oxides) and compositions different of derivatives. The AlSBA-15 catalysts showed better performance in analyzes for production petroleum derived, 20% increase in production of kerosene and light gas oil. The energy potential of sludge was high and it can be used as fuel in other cargo processed in refinery

The main aim of this study was to compare the procedure for dehydration of Gracilaria birdiae prepared handmade and laboratory, collected in the northern coast of Rio Grande do Norte. The sample was collected in the Rio do Fogo beach in march 2009. The sample collected followed by two processing, the first the material prepared in laboratory was air-dried at 50°C for 24 hours in air-flow oven. The second the handmade sample was air-dried on the sun during three days. The extract was prepared in three different solvents: ethanol, hydroethanol and water, resulting in ethanol, hidroethanol and aqueous extracts from handmade and laboratory sample. In according with results only the ethanol extract was fractionated yielding the fractions hexane, dichloromethane and ethyl acetate fractions. The different process to obtain Gracilaria birdiae resulted in the samples with different shades. The soluble solids content was higher in the laboratory sample. The chemical composition the both samples were characterized by presenting a considerable amounts of carbohydrates, with amior percentage protein and ash, respectively, in the handmade and laboratory sample. In two samples showed a low content of lipids and the lipid profile showed a higher proportion of monounsaturated fatty acids, with the absence polyunsaturated handmade sample. The phytochemical screening by chemical reactions showed the presence of flavonoids, tannins, alkaloids and saponins the laboratory sample, presenting a greater diversity of bioactive compounds. Through of the analysis by thin layer chromatography was possible to identify the phytosterols β-sitosterol and stigmasterol the both samples, also suggest the presence of β-carotene and chlorophyll α the laboratory sample. The levels of total phenolics and flavonoids were more significant in the ethanol extract of the laboratory sample. The in vitro lethality showed that extracts of the laboratory sample and handmade from 125 to 500 μg/ mL, respectively, were highly lethal. In the evaluation of antioxidant capacity by the system β-carotene/ácido linoleic method and by DPPH radical scavernging assay, the ethanol extract from the laboratory process showed significantly greater activity than the other extracts, being and the first and second methods, respectively, lower and equivalent to the synthetic antioxidant BHT. The handmade ethanol extract has not demonstrated skill in deactivating free radicals, but showed activity in inhibiting lipid peroxidation, although the values were significantly lower than the laboratory sample. We conclude that the dehydration process in the laboratory is the most efficient technique to maintenance of the chemical composition present in the seaweed, providing beneficial properties such as antioxidant capacity. We emphasize that this property can be explored with the objective of adding commercial value to the final product, which will promote the expansion of production of this seaweed in the community of Rio do Fogo

This work was developed with the objetive to evaluate the insecticidal effect of the Neem (Azadirachta indica A. Juss) seeds against grass bug nymphs Collaria scenica Stal. For that, an extract was prepared from green fruits of Neem by rotaevaporacion. The extract was diluted in three ppm concentrations corresponding to treatments. Using thin-layer chromatography we identified the presence of Azaridactina. DCA essay was carried out with 4 treatments and 5 repetitions that included the three extract concentrations and a control. In each repetition were used 15 nymphs, randomly placed in plastic boxes with food and the corresponding concentration. Every day an extract application was made and recorded mortality percentage, number of exuviae and number of individuals who came to adulthood in each treatment. The results were analyzed using Kruskal-Wallis and Games-Howell for each variable. The three concentrations of Neem seed extract had a negative effect on the development of the bugs. The more concentrated treatment (250 ppm) was the most effective, showing a 97% mortality, fewer exuvias and and fewer adults in the end of the experiment.

In this work, experimental data for the system Lippia alba + CO2 is presented. The major constituents of the L. alba volatile oil are limonene and carvone. Thus, literature data for the systems limonene + CO2 and carvone + CO2, and the Peng-Robinson equation of state (PR-EOS) were used to select the operating temperature and pressure, which maximize the global yield in L. alba extract. Global yields were determined at 80, 100, and 120 bar and 40, 45, and 50 degrees C. L. alba extracts were also obtained by conventional processes (hydrodistillation, low-pressure ethanol extraction and Soxhlet ethanol). The chemical compositions of the extracts were determined by gas and thin layer chromatography (TLC). The secretor structures of L. alba were observed by scanning electron microscopy (SEM) before and after supercritical extraction. The largest yield (similar to 7%, mass of extract/mass of dry solid) of the CO2-extract was obtained at 318 K and 100 bar. The chemical compositions of the CO2-extracts were different from those of the extracts obtained by Soxhlet and low-pressure solvent extraction (LPSE) because of the co-extraction of heavy substances by ethanol. The operating conditions that maximized the carvone and limomene yields were 80 bar and 323 K (80 mass%) and 120 bar and 323 K (17 mass%), respectively. (c) 2004 Elsevier B.V All rights reserved.

Introduction: Rhodococcus equi is an opportunistic pathogen, causing rhodococcosis, a condition that can be confused with tuberculosis. Often, without identifying M. tuberculosis, physicians initiate empiric treatment for tuberculosis. R. equi and M. tuberculosis have different susceptibility to drugs. Identification of R. equi is based on a variety of phenotypic, chromatographic, and genotypic characteristics.Objective: This study aimed to characterize bacterial isolates from sputum samples suggestive of R. equi.Methods: The phenotypic identification included biochemical assays; thin-layer chromatography (TLC) and polymerase chain reaction (PCR) were used for genotypic identification.Results: Among 78 Gram-positive and partially acid-fast bacilli isolated from the sputum of tuberculosis-suspected patients, 51 were phenotypically and genotypically characterized as R. equi based on literature data. Mycolic acid analysis showed that all suspected R. equi had compounds with a retention factor (R-f) between 0.4-0.5. Genotypic characterization indicated the presence of the choE gene 959 bp fragments in 51 isolates CAMP test positive. Twenty-two CAMP test negative isolates were negative for the choE gene. Five isolates presumptively identified as R. equi, CAMP test positive, were choE gene negative, and probably belonged to other bacterial species.Conclusions: The phenotypic and molecular techniques used constitute a good methodological tool to identify R. equi. The yeast Kluyveromyces marxianus var. bulgaricus produced large amounts of extracellular inulinase activity when grown on inulin, sucrose, fructose and glucose as carbon source, This protein has been purified to homogeneity by using successive DEAE-Trisacryl Plus and Superose 6 HR 10/30 columns. The purified enzyme showed a relative molecular weight of 57 kDa by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and 77 kDa by gel filtration in Superose 6 HR 10/30, Analysis by SDS-PAGE showed a unique polypeptide band with Coomassie Blue stain and nondenaturing PAGE of the purified enzyme obtained from media with different carbon sources showed the band, too, when stained for glucose oxidase activity, the optimal hydrolysis temperature for sucrose, raffinose and inulin was 55 degrees C and the optimal pH for sucrose was 4.75, the apparent K-m values for sucrose, raffinose and inulin are 4.58, 7.41 and 86.9 mg/ml, respectively, Thin layer chromatography showed that inulinase from K. marxianus var. bulgaricus was capable of hydrolyzing different substrates (sucrose, raffinose and inulin), releasing monosaccharides and oligosaccharides, the results obtained suggest the hypothesis that enzyme production was constitutive.
