Transposons, mobile genetic elements that are ubiquitous in all living organisms have been used as tools in molecular biology for decades. They have the ability to move into discrete DNA locations with no apparent homology to the target site. The utility of transposons as molecular tools is based on their ability to integrate into various DNA sequences efficiently, producing extensive mutant clone libraries that can be used in various molecular biology applications. Bacteriophage Mu is one of the most useful transposons due to its well-characterized and simple in vitro transposition reaction. This study establishes the properties of the Mu in vitro transposition system as a versatile multipurpose tool in molecular biology. In addition, this study describes Mu-based applications for engineering proteins by random insertional transposon mutagenesis in order to study structure-function relationships in proteins. We initially characterized the properties of the minimal Mu in vitro transposition system. We showed that the Mu transposition system works efficiently and accurately and produces insertions into a wide spectrum of target sites in different DNA molecules. Then, we developed a pentapeptide insertion mutagenesis strategy for inserting random five amino acid cassettes into proteins. These protein variants can be used especially for screening important sites for protein-protein interactions. Also, the system may produce temperature-sensitive variants of the protein of interest. Furthermore, we developed an efficient screening system for high-resolution mapping of protein-protein interfaces with the pentapeptide insertion mutagenesis. This was accomplished by combining the mutagenesis with subsequent yeast two-hybrid screening and PCR-based genetic footprinting. This combination allows the analysis of the whole mutant library en masse, without the need for producing or isolating separate mutant clones, and the protein-protein interfaces can be determined at amino acid accuracy. The system was validated by analysing the interacting region of JFC1 with Rab8A, and we show that the interaction is mediated via the JFC1 Slp homology domain. In addition, we developed a procedure for the production of nested sets of N- and C-terminal deletion variants of proteins with the Mu system. These variants are useful in many functional studies of proteins, especially in mapping regions involved in protein-protein interactions. This methodology was validated by analysing the region in yeast Mso1 involved in an interaction with Sec1. The results of this study show that the Mu in vitro transposition system is versatile for various applicational purposes and can efficiently be adapted to random protein engineering applications for functional studies of proteins.

This paper proposes a Single Network Adaptive Critic (SNAC) based Power System Stabilizer (PSS) for enhancing the small-signal stability of power systems over a wide range of operating conditions. SNAC uses only a single critic neural network instead of the action-critic dual network architecture of typical adaptive critic designs. SNAC eliminates the iterative training loops between the action and critic networks and greatly simplifies the training procedure. The performance of the proposed PSS has been tested on a Single Machine Infinite Bus test system for various system and loading conditions. The proposed stabilizer, which is relatively easier to synthesize, consistently outperformed stabilizers based on conventional lead-lag and linear quadratic regulator designs.

Electric power systems are exposed to various contingencies. Network contingencies often contribute to over-loading of network branches, unsatisfactory voltages and also leading to problems of stability/voltage collapse. To maintain security of the systems, it is desirable to estimate the effect of contingencies and plan suitable measures to improve system security/stability. This paper presents an approach for selection of unified power flow controller (UPFC) suitable locations considering normal and network contingencies after evaluating the degree of severity of the contingencies. The ranking is evaluated using composite criteria based fuzzy logic for eliminating masking effect. The fuzzy approach, in addition to real power loadings and bus voltage violations, voltage stability indices at the load buses also used as the post-contingent quantities to evaluate the network contingency ranking. The selection of UPFC suitable locations uses the criteria on the basis of improved system security/stability. The proposed approach for selection of UPFC suitable locations has been tested under simulated conditions on a few power systems and the results for a 24-node real-life equivalent EHV power network and 39-node New England (modified) test system are presented for illustration purposes.

Aim: The present study was conducted to overcome the disadvantages associated with the poor water solubility and low bioavailability of curcumin by synthesizing nanotized curcumin and demonstrating its efficacy in treating malaria. Materials and methods: Nanotized curcumin was prepared by a modified emulsion-diffusion-evaporation method and was characterized by means of transmission electron microscopy, atomic force microscopy, dynamic light scattering, Zetasizer, Fourier transform infrared spectroscopy, and differential thermal analysis. The novelty of the prepared nanoformulation lies in the fact that it was devoid of any polymeric matrices used in conventional carriers. The antimalarial efficacy of the prepared nanotized curcumin was then checked both in vitro and in vivo. Results: The nanopreparation was found to be non-toxic and had a particle size distribution of 20-50 nm along with improved aqueous dispersibility and an entrapment efficiency of 45%. Nanotized curcumin (half maximal inhibitory concentration IC50]: 0.5 mu M) was also found to be ten-fold more effective for growth inhibition of Plasmodium falciparum in vitro as compared to its native counterpart (IC50: 5 mu M). Oral bioavailability of nanotized curcumin was found to be superior to that of its native counterpart. Moreover, when Plasmodium berghei-infected mice were orally treated with nanotized curcumin, it prolonged their survival by more than 2 months with complete clearance of parasites in comparison to the untreated animals, which survived for 8 days only. Conclusion: Nanotized curcumin holds a considerable promise in therapeutics as demonstrated here for treating malaria as a test system.

This paper proposes a novel decision making framework for optimal transmission switching satisfying the AC feasibility, stability and circuit breaker (CB) reliability requirements needed for practical implementation. The proposed framework can be employed as a corrective tool in day to day operation planning scenarios in response to potential contingencies. The switching options are determined using an efficient heuristic algorithm based on DC optimal power flow, and are presented in a multi-branch tree structure. Then, the AC feasibility and stability checks are conducted and the CB condition monitoring data are employed to perform a CB reliability and line availability assessment. Ultimately, the operator will be offered multiple AC feasible and stable switching options with associated benefits. The operator can use this information, other operating conditions not explicitly considered in the optimization, and his/her own experience to implement the best and most reliable switching action(s). Cleome rosea é uma espécie nativa, de porte herbáceo, ocorrente em restingas brasileiras. Estudos recentes têm revelado o potencial medicinal da espécie para importantes propriedades farmacológicas, como por exemplo, as atividades anti-inflamatória, antigenotóxica, antiviral e antibacteriana. Porém, nos últimos anos, C. rosea não tem sido encontrada em várias regiões de seu ambiente natural, devido, principalmente, às ações antrópicas. Dessa forma, torna-se relevante o desenvolvimento de métodos de conservação que permitam o estudo e exploração das propriedades medicinais da espécie. O cultivo in vitro de raízes representa uma forma eficiente para produção de biomassa, devido ao rápido crescimento, produção estável de metabólitos, além de representar uma potencial fonte de explantes para a propagação em massa de diferentes espécies. O presente trabalho teve como objetivo a produção in vitro de culturas de raízes de C. rosea, associada à criopreservação, como forma de manutenção em longo prazo das culturas, monitorada através da análise de estabilidade genética. As culturas estabelecidas a partir de explantes radiculares de plantas propagadas in vitro de C. rosea demonstraram excelente capacidade de multiplicação de raízes em meio de cultura suplementado com o fitorregulador ANA, com manutenção dessa capacidade ao longo de sucessivas subculturas. Associado a esses resultados, o estabelecimento de protocolos de criopreservação pelo método de vitrificação resultou em elevados valores de frequência de recuperação do material após congelamento em nitrogênio líquido com as soluções de vitrificação PVS2 e PVS3. Os estudos de monitoramento da estabilidade genética, pela técnica de marcadores moleculares RAPD, revelaram a presença de polimorfismos significativos em uma das três culturas iniciadas a partir de raízes de C. rosea criopreservadas. Esses resultados demonstram as possibilidades de produção de raízes de C. rosea e conservação em longo prazo através da criopreservação, iniciando estudos inéditos para a espécie.

The genomes of many positive stranded RNA viruses and of all retroviruses are translated as large polyproteins which are proteolytically processed by cellular and viral proteases. Viral proteases are structurally related to two families of cellular proteases, the pepsin-like and trypsin-like proteases. This thesis describes the proteolytic processing of several nonstructural proteins of dengue 2 virus, a representative member of the Flaviviridae, and describes methods for transcribing full-length genomic RNA of dengue 2 virus. Chapter 1 describes the in vitro processing of the nonstructural proteins NS2A, NS2B and NS3. Chapter 2 describes a system that allows identification of residues within the protease that are directly or indirectly involved with substrate recognition. Chapter 3 describes methods to produce genome length dengue 2 RNA from cDNA templates.
The nonstructural protein NS3 is structurally related to viral trypsinlike proteases from the alpha-, picorna-, poty-, and pestiviruses. The hypothesis that the flavivirus nonstructural protein NS3 is a viral proteinase that generates the termini of several nonstructural proteins was tested using an efficient in vitro expression system and antisera specific for the nonstructural proteins NS2B and NS3. A series of cDNA constructs was transcribed using T7 RNA polymerase and the RNA translated in reticulocyte lysates. Proteolytic processing occurred in vitro to generate NS2B and NS3. The amino termini of NS2B and NS3 produced in vitro were found to be the same as the termini of NS2B and NS3 isolated from infected cells. Deletion analysis of cDNA constructs localized the protease domain necessary and sufficient for correct cleavage to the first 184 amino acids of NS3. Kinetic analysis of processing events in vitro and experiments to examine the sensitivity of processing to dilution suggested that an intramolecular cleavage between NS2A and NS2B preceded an intramolecular cleavage between NS2B and NS3. The data from these expression experiments confirm that NS3 is the viral proteinase responsible for cleavage events generating the amino termini of NS2B and NS3 and presumably for cleavages generating the termini of NS4A and NS5 as well.
Biochemical and genetic experiments using viral proteinases have defined the sequence requirements for cleavage site recognition, but have not identified residues within proteinases that interact with substrates. A biochemical assay was developed that could identify residues which were important for substrate recognition. Chimeric proteases between yellow fever and dengue 2 were constructed that allowed mapping of regions involved in substrate recognition, and site directed mutagenesis was used to modulate processing efficiency.
Expression in vitro revealed that the dengue protease domain efficiently processes the yellow fever polyprotein between NS2A and NS2B and between NS2B and NS3, but that the reciprocal construct is inactive. The dengue protease processes yellow fever cleavage sites more efficiently than dengue cleavage sites, suggesting that suboptimal cleavage efficiency may be used to increase levels of processing intermediates in vivo. By mutagenizing the putative substrate binding pocket it was possible to change the substrate specificity of the yellow fever protease; changing a minimum of three amino acids in the yellow fever protease enabled it to recognize dengue cleavage sites. This system allows identification of residues which are directly or indirectly involved with enzyme-substrate interaction, does not require a crystal structure, and can define the substrate preferences of individual members of a viral proteinase family.
Full-length cDNA clones, from which infectious RNA can be transcribed, have been developed for a number of positive strand RNA viruses, including the flavivirus type virus, yellow fever. The technology necessary to transcribe genomic RNA of dengue 2 virus was developed in order to better understand the molecular biology of the dengue subgroup. A 5' structural region clone was engineered to transcribe authentic dengue RNA that contains an additional 1 or 2 residues at the 5' end. A 3' nonstructural region clone was engineered to allow production of run off transcripts, and to allow directional ligation with the 5' structural region clone. In vitro ligation and transcription produces full-length genomic RNA which is noninfectious when transfected into mammalian tissue culture cells. Alternative methods for constructing cDNA clones and recovering live dengue virus are discussed.

In the process of interferometric testing, the measurement result is influenced by the system structure, which reduces the measurement accuracy. To obtain an accurate test result, it is necessary to analyze the test system, and build the relationship between the measurement error and the system parameters. In this paper, the influences of the system elements which include the collimated lens and the standard surface on the interferometric testing are analyzed, the expressions of phase distribution and wavefront error on the detector are obtained, the method to remove some element errors is introduced, and the optimization structure relationships are given. A radiação ultravioleta (UV) induz diversos efeitos nocivos nos organismos e a quantidade desta radiação que atinge a biosfera é afetada pela concentração de ozônio, latitude, altitude, clima e reflexão especular. As respostas de briófitas em relação aos efeitos da radiação UV e a presença de compostos que absorvem esta radiação têm sido estudadas. Sanionia uncinata, Holomitriopsis laevifolia e Leucobryum laevifolium são espécies de musgos encontrados em locais expostos a alta incidência de radiação UV e com habitats distintos. Considerando que as respostas de musgos contra os efeitos da radiação UV e seus mecanismos de proteção ainda são pouco caracterizados, o objetivo deste estudo foi investigar o potencial fotoprotetor e possíveis riscos toxicológicos associados aos extratos dos musgos S. uncinata, proveniente da Antártica e H. laevifolia e L. laevifolium, proveniente do Amazonas. Seus extratos metanólico (EM), aquoso (EA), hidroalcoólico (EH) e etanólico (EE) foram estudados com a caracterização química por absorção ao UV e visível e pela cromatografia líquida de alta eficiência; quantificação do índice total de compostos fenólicos; determinação da capacidade captadora do radical 2,2-difenil-1-picril-hidrazila a fim de avaliar as atividades antioxidantes; avaliação do potencial de fotoproteção cutânea pela determinação do fator de proteção solar; avaliações do potencial mutagênico e citototóxico, através do ensaio de Salmonella/microssoma, utilizando as cepas TA97, TA98, TA100, TA102 e TA104; do potencial fotomutagênico através do ensaio de fotomutagenicidade, usando as cepas TA102 e TA104; e investigação dos efeitos genotóxicos e fotogenotóxicos, pelo ensaio de micronúcleo e fotomicronúcleo, respectivamente, usando diferentes linhagens celulares estabelecidas. Foram encontradas atividades fotoprotetoras e antioxidantes e observou-se que os extratos se apresentaram singulares devido a sua composição química. Os resultados fotoprotetores, além dos mutagênicos/fotomutagênicos, genotóxicos/fotogenotóxicos e suas respectivas avaliações citotóxicas também permitiram selecionar extratos e suas concentrações, como promissores candidatos em fotoproteção Assim, os EA e EH de H. laevifolia e L. laevifolium apresentam, no geral, os resultados mais significativos, tornando-se potenciais para avaliações refinadas em fotoproteção e na separação de componentes que possam levar a futuras aplicações como antioxidantes e protetores solares ou como adjuvantes.

Cleome dendroides é uma espécie endêmica da Mata Atlântica dos estados do Rio de Janeiro e Espírito Santo, bioma alterado por intensa atividade antrópica, o que constitui uma ameaça à preservação de suas populações. Não existem estudos dos pontos de vista fisiológico, biotecnológico, fitoquímico ou farmacológico sobre a espécie. Considerando-se o perfil fitoquímico e o potencial medicinal do gênero, torna-se relevante definirem-se protocolos para a produção de plantas e metabólitos de C. dendroides utilizando diferentes sistemas de cultivo in vitro. No presente trabalho, foram realizados estudos sobre a germinação in vivo da espécie, avaliando-se a influência do substrato, temperatura e luz. Não se observou qualquer tipo de dormência, sendo as temperaturas de 20, 25 e 20-30C, em areia ou vermiculita, apropriadas para a germinação in vivo. Definiu-se também uma metodologia eficiente de germinação sob condições in vitro, e as plântulas obtidas foram utilizadas como fonte de explantes para os estudos de propagação in vitro. A resposta morfogênica foi avaliada considerando-se a origem e tipo do explante, tipos e concentrações de reguladores de crescimento e número de subculturas. A metodologia empregada mostrou-se eficiente para a produção de brotos e manutenção de estoques de plantas in vitro que serviram como fonte de explantes. A melhor condição para a propagação in vitro foi definida em meio solidificado contendo BA, independentemente do tipo de explante e da origem. Os brotos obtidos foram alongados, enraizados e aclimatizados. Também foi estabelecida a cultura de raízes e a regeneração de brotos a partir destas culturas. Avaliou-se o efeito da origem do explante, assim como dos tipos e concentrações de fitorreguladores sobre a proliferação de raízes e a regeneração de brotos. O fitorregulador AIB propiciou maior multiplicação das raízes, enquanto BA mostrou-se eficiente na regeneração de brotos a partir das raízes recém-formadas. Foi estabelecido ainda um protocolo de cultura de calos e de suspensões celulares. Avaliou-se o efeito da origem e do tipo de explante, dos tipos e das concentrações de fitorreguladores sobre a calogênese. A combinação de PIC com KIN foi a mais eficiente para a indução de calos em explantes de plântulas obtidas a partir de germinação in vitro, produzindo calos que foram mantidos por pelo menos dois anos. As suspensões celulares também foram estabelecidas em meio contendo PIC + KIN, mantendo uma produção de biomassa de cerca de cinco vezes o peso fresco inicial por três sucessivas subculturas. Análises histoquímicas e fitoquímicas revelaram a presença de alcaloides nos calos e nas suspensões celulares. Foram realizadas análises fitoquímicas de plantas de campo, plantas aclimatizadas, plantas mantidas em estoque in vitro e culturas de raízes, as quais indicaram a presença de derivados de glicosinolatos. Os resultados demonstraram a viabilidade de produção de material vegetal de C. dendroides por meio de métodos biotecnológicos e a produção in vitro de metabólitos de importância medicinal

盐角草(Salicornia europaea L.)属藜科盐角草属(Salicornia),是迄今已报道过的最为耐盐的真盐生植物之一。跟与其同属的海蓬子(Salicornia bigelovvi Torr.)相比, 它的分布更为广泛。它的种子含油丰富,因此具有发展为油料作物的潜力。此外,它也可以作为蔬菜和饲料。盐角草的组织培养工作为未来的转化研究和胁迫相关基因功能分析提供了有力的工具。本工作中,通过器官发生途径,建立了盐角草的体外再生体系。以完整的成熟种子为起始培养材料,在添TDZ 0.1 mg/L与NAA 1 mg/L的MS培养基上,暗培养三周后在下胚轴处形成愈伤组织,形成愈伤的平均频率为99%。愈伤组织在含TDZ 0.1 mg/L与NAA 1 mg/L的培养基上培养3-4周后分化出芽,分化频率约26.7%。采用2,4-D短时处理法结合添加NaCl,经过6-8周的培养获得了丛生芽,提高了再生频率。分化芽3周后转入含IBA 0.5 mg/L、KN 0.1 mg/L与0.05%活性炭的1/2 MS培养基,3周后生根形成完整植株。同时,本研究也进行了从直接不定芽途径建立盐角草再生体系的试验,但未获成功。 此外,本工作借鉴拟南芥的floral-dip转化法,对建立盐角草floral-dip转化系统进行了尝试。

对Egonol龙胆三糖苷及以Egonol衍生物对雌二醇生成活性及其相关机制进行了研究。发现Egonol龙胆三糖苷促雌二醇最高生成率在MCF-7、HepG2、ROS1728中分别为157% 、182.4%、226.8%(以空白组200μg/ml睾酮转换成E2值作为100%生成率)。活性的强弱可能与芳香化酶的组织特异性表达情况一致,说明Egonol龙胆三糖苷促雌二醇活性可能与芳香化酶有关。芳香化酶的组织特异性表达与特异性启动子有关系,Egonol龙胆三糖苷在各组织中皆有促雌二醇活性,说明该化合物不是通过调节该酶的基因表达而起作用。 在探究Egonol龙胆三糖苷及其衍生物是否介导cAMP-PKA途径从而影响芳香化酶的表达中,发现该系列化合物在HEK-293T细胞中对cAMP的影响非常弱小。在人HepG2细胞中显示了极强的提高cAMP的作用。而化合物对cAMP的作用与其促雌二醇活性强弱不呈正相关关系,对c AMP-PKA途径的激活可能与胞内雌激素有关。 Egonol龙胆三糖苷及其衍生物对HepG2细胞增殖影响显示,该系列化合物同雌二醇一样有相似的较弱促HepG2细胞增殖作用。而且存在一定剂量依赖性。在瞬时转染有ERE(雌激素作用元件)的HepG2中,Egonol龙胆三糖苷及其衍生物也显示了类似于雌二醇与ERE结合的作用,进一步提示Egonol龙胆三糖苷及其衍生物在HepG2细胞中具备雌激素样作用。 为研究Egonol龙胆三糖苷及其衍生物是否可能直接提高芳香化酶的活性,我们计划将芳香化酶从芳香化酶阳性细胞中克隆后表达到芳香化酶阴性的细胞中。在MCF-7细胞中以Oligo dT为引物合成的cDNA模板,和在ROS1728细胞中以Oligo dT及大鼠引物F链为引物合成的cDNA模板能成功扩增出与芳香化酶全长编码序列大小一致的片段。 Egonol衍生物在HepG2、ROS1728细胞中促雌二醇活性的实验表明,Egonol苯环上引入其它基团可以提高Egonol的活性。 从雌激素经典的基因组效应和非基因组效应两方面对雌激素信号转导研究进展进行了简单的综述。 The promoting effects of egonol gentiotrioside and egonol derivatives on the synthesis of estrogen E2 were studied. In vitro test, egonol gentiotrioside promoted the synthesis of estrogen E2 in MCF-7, HepG2,ROS1728 cell lines with mean yields of estrogen E2 57%,82.4% and 126.8%, higher than those of blank control at a concentration of 100 mg/ml. The difference of estrogen E2 synthesis promoting effects among the cell lines suggested tissue specificity. It is in accordance with tissue specific character of aromatase expression. The evidence implied that effect of egonol gentiotrioside on promoting the synthesis of estrogen E2 was related to the aromatase. Different expression levels of aromatase in different tissues are attributed to their specific promoters, but egonol gentiotrioside can promote the synthesis of estrogen E2, in many tissues,so the fact is controversary to the estimation that this compound regulates the aromatase on gene level. In order to investigate whether egonol gentiotrioside and its synthetic derivatives regulates aromatase activity through the cAMP-PKA signal pathway,we transfected the p CRE-Luc luciferase reporter gene into the HEK-293T cells and HepG2 cells. These compounds had weak activity in promoting the cAMP activity in HEK-293T cells but strong in HepG2 cells.The compounds’effect of promoting the cAMP may be related to their estrogenic activity in cells. The modified HepG2 cell proliferation assay was used to evaluate the estrogenic activity of egonol gentiotrioside and its derivatives. The weak estrogenic activity of egonol gentiotrioside and its derivatives at various concentrations expressed as proliferative effect relative to that of blank control was examined. We transfected the pERE-Luc luciferase reporter gene into the HepG2 cells. These compounds possessed significant activity on estrogen response element compared with the one treated with 10 n M estrogen E2. This evidence indicated that the estrogenic activity of egonol gentiotrioside and its derivatives. In order to investigate whether the egonol gentiotrioside and its derivatives can upregulate the activity of aromatase directly, The full-length of P450 aromatase cDNA encoding aromatase were amplified by using primer Oligo dT in MCF-7,and specific primer in ROS1728,respectively. The structure-activity relationship of Egonol in promoting the synthesis of E2 in HepG2 and ROS1728 cells indicated that introduction of some group on the basic sketon of egonol could improve the effect. The progress in research of signal pathway of estrogen in recent years was summarized.

鞑靼荞麦是我国特有的农业产品,具有抗寒耐旱特性和较高的营养保健功能。荞麦的开花习性及遗传特点导致其人工杂交授粉难以成功,这成为荞麦杂交育种难以获得突破的重要原因。因此利用转基因技术导入有益基因有可能成为荞麦遗传改良的新途径,而再生及转化体系的建立是开展转基因研究的基础。 本文研究了苗龄、外植体、几种激素配比对鞑靼荞麦(Fagopyrum tataricum Gaertn.)离体培养的影响,初步建立了鞑靼荞麦离体再生体系。结果表明,鞑靼荞麦离体再生的最佳取材时间为苗龄6-8d;诱导愈伤组织的最适培养基为MS+2.0 mg/L 2,4-D+1.5 mg/L 6-BA,子叶诱愈率达75%左右,下胚轴的可高达86.62%;愈伤组织分化的最适培养基为MS 0.1mg/L IAA+2.0mg/L 6-BA+1.0 mg/L KT+0.5mg/L TDZ,下胚轴的分化率可达9.52%。下胚轴的诱愈率与分化率均高于子叶,更适于离体再生培养。培养基中加入AgNO3后,能有效降低褐化率。生根最适培养基为含有0.5mg/L NAA的1/2MS培养基,生根率在50%左右。TDZ在诱导鞑靼荞麦的愈伤组织分化出芽的过程中起到明显的促进作用,可提高分化率约20%。 在上述研究基础上,本文还对鞑靼荞麦的遗传转化体系进行了探索性研究。分别利用根癌农杆菌(Agrobacterium tumefaciens)介导法和微粒轰击法(基因枪法)对黑水苦荞下胚轴进行遗传转化。 在农杆菌介导的方法中,携带有质粒pCAMBIA2301的农杆菌菌株EHA105用于转化。载体质粒pCAMBIA2301包含有gus和npt-II 基因, 并受35s启动子驱动。研究结果表明,在侵染方式选择上,浸泡方式比吸打方式更有效,根癌农杆菌侵染的较适浓度为OD600=0.5,共培养3天,恢复培养7天,能检测到gus基因的表达。 基因枪法使用质粒pBI121,同样包含有gus和npt-II基因, 并受CaMV35s 启动子驱动。轰击距离为9cm较合适,甘露醇前处理在本研究中未表现出明显优势。 两种转化方法比较,基因枪法比农杆菌介导法更快速有效。 本研究为进一步的遗传操作研究打下基础。 Tartary buckwheat (Fagopyrum tataricum Gaertn.), the traditional and unique agricultural product of China, is a kind of crop with strong drought and cold tolerance, abundant nutrition and high medical value. Artificial hybridization is hard in buckwheat because of its flowering habits and genetic characteristics, which leads to no breakthrough in tartary buckwheat breeding. However, biotechnological approaches, especially genetic transformation for the direct introduction of good genes into tartary buckwheat for quality improvement, hold great promise. In this study, we established tartary buckwheat regeneration system in vitro. It is the foundation for genetic manipulation of this crop. The effects of seedling age, hypocotyl and cotyledon as explants, and proportions of several growth regulators were tested in tissue culture of tartary buckwheat for establishing its in vitro regeneration system. The results showed that the best seedling age for callus induction was 6 to 8 days. On the MS medium containing 2.0mg/L 2, 4-D and 1.5mg/L 6-BA, the induction rate of callus from hypocotyls was up to 86.62%, while from cotyledons was about 75%. The suitable shooting medium was the MS medium+0.1mg/L IAA+2.0mg/L 6-BA+1.0 mg/L KT+0.5mg/L TDZ, and the shooting rate from hypocotyls was 9.52%. The callus induction and shooting rates were higher from hypocotyls than from cotyledons. Browning reduced when the medium mixed with AgNO3. Half strength MS supplemented with 0.5mg/L NAA was the best for rooting, the rate was around 50% after 30 days culture. TDZ can accelerate the shoot differentiation distinctively, and it could improve the shooting rate nearly 20%. On the base of above, the explorative research of the genetic transformation in tartary buckwheat was done. In the study, hypocotyls from Heishui tartary buckwheat were transformed by Agrobacterium-mediated method and microprojectile bombardment method (gene-gun), comparatively. In Agrobacterium-mediated method, a disarmed Agrobacterium tumefaciens strain EHA105 harboring plasmid pCAMBIA2301 was used. The vector pCAMBIA2301 contains gus and npt-II genes, driven by CaMV35s promoter. The results showed that the appropriate concentration of Agrobacterium tumefaciens for infecting was OD600=0.5, and co-culture time was 3d. Seven days later after coculture, GUS expression could be tested. In particle bombardment transformation, plasmid pBI121 was used. pBI121 also contains gus and npt-II genes, driven by 35s promoter. Hypocotyls pretreated with mannitol, no effect was observed, and the suitable distance of bombardment is 9cm. Comparing with Agrobacterium-mediated method, gene-gun method is more convenient and effective. All above results could be a basic work for further study in tartary buckwheat transformation.

This paper presents a new method for online determination of the Thèvenin equivalent parameters of a power system at a given node using the local PMU measurements at that node. The method takes into account the measurement errors and the changes in the system side. An analysis of the effects of changes in system side is carried out on a simple two-bus system to gain an insight of the effect of system side changes on the estimated Thévenin equivalent parameters. The proposed method uses voltage and current magnitudes as well as active and reactive powers; thus avoiding the effect of phase angle drift of the PMU and the need to synchronize measurements at different instances to the same reference. Applying the method to the IEEE 30-bus test system has shown its ability to correctly determine the Thévenin equivalent even in the presence of measurement errors and/or system side changes.

The small signal stability of interconnected power systems is one of the important aspects that need to be investigated since the oscillations caused by this kind of instability have caused many incidents. With the increasing penetration of wind power in the power system, particularly doubly fed induction generator (DFIG), the impact on the power system small signal stability performance should be fully investigated. Because the DFIG wind turbine integration is through a fast action converter and associated control, it does not inherently participate in the electromechanical small signal oscillation. However, it influences the small signal stability by impacting active power flow paths in the network and replacing synchronous generators that have power system stabilizer (PSS). In this paper, the IEEE 39 bus test system has been used in the analysis. Furthermore, four study cases and several operation scenarios have been conducted and analysed. The selective eigenvalue Arnoldi/lanczos's method is used to obtain the system eigenvalue in the range of frequency from 0.2 Hz to 2 Hz which is related to electromechanical oscillations. Results show that the integration of DFIG wind turbines in a system during several study cases and operation scenarios give different influence on small signal stability performance.
