Le fardeau vasculaire (présence de maladies vasculaires et/ou cérébrovasculaires) est associé à une augmentation des troubles cognitifs chez les personnes âgées, ainsi qu’à un plus haut risque de démence vasculaire (DV) et de démence de type Alzheimer (DTA). Un nombre restreint de travaux a porté sur l’impact du fardeau vasculaire sur la cognition des personnes avec trouble cognitif léger (TCL). Pourtant, les personnes avec TCL représentent une population d’intérêt puisqu’elles sont à haut risque d’évoluer vers une démence. Cette thèse comprend trois articles de revue qui visent à exposer les connaissances entourant la santé vasculaire et la cognition des personnes âgées et trois articles empiriques (Chapitres 5, 6, 7). La première étude empirique traite de l’impact du fardeau vasculaire sur la cognition des personnes TCL et a comme objectif de spécifier quelles fonctions cognitives sont affectées par le fardeau vasculaire et dans quelle mesure le fardeau vasculaire influence l’hétérogénéité des TCL (Chapitre 5). Dans le cadre de la deuxième étude, nous avons examiné l’intégrité des processus stratégiques et non stratégiques de mémorisation des TCL avec et sans fardeau vasculaire, afin d’évaluer si ces processus diffèrent quantitativement et qualitativement entre ces deux groupes (Chapitre 6). Enfin, dans la troisième étude nous avons évalué les capacités d’association (binding) ainsi que la résistance à l’interférence des personnes TCL, les liens entre ces processus mnésiques et différents marqueurs cérébraux en plus des facteurs permettant de prédire l’évolution vers une démence (Chapitre 7). Les résultats présentés dans cette thèse appuient l’hypothèse selon laquelle le fardeau vasculaire influence le profil cognitif des TCL. Dans un premier temps, nous avons montré que le fardeau vasculaire est associé à une atteinte exécutive plus importante chez les TCL (Chapitre 5). De plus, nos résultats suggèrent que le fardeau vasculaire influence la classification clinique de ces derniers, puisque ceux dont le fardeau est élevé répondent davantage aux critères de TCL amnestique domaine multiple (trouble de mémoire plus au moins un autre déficit cognitif) tandis que ceux sans fardeau répondent davantage aux critères de TCL amnestique domaine unique (trouble isolé de la mémoire). Dans un deuxième temps, nous avons montré des différences dans la nature des processus mnésiques atteints chez les TCL avec et sans fardeau vasculaire (Chapitre 6). Alors que les premiers présentent une atteinte prédominante des processus stratégiques de mémorisation, les seconds présentent une atteinte des processus stratégiques et non stratégiques de mémorisation. Lorsque seuls les résultats des TCL ayant évolué vers une démence sont analysés, le patron d’atteinte est similaire puisque les TCL vasculaires sont quantitativement moins touchés que les TCL non-vasculaires au niveau des processus non-stratégiques de mémorisation. Enfin, tant les TCL qui progressent vers une démence que les TCL qui restent stables après un suivi de trois ans éprouvent tous des difficultés de mémoire associative et sont sensibles à l’interférence proactive (Chapitre 7). De plus, le fardeau vasculaire est associé à la sensibilité à l’interférence alors que le volume hippocampique est associé aux difficultés de mémoire associative. Enfin, nos résultats indiquent que les TCL qui éprouvent des difficultés de mémoire associative sont plus à risque d’évoluer vers une démence que les TCL ne présentant pas ces mêmes difficultés. De façon globale, les résultats de cette thèse révèlent que le fardeau vasculaire joue un rôle important dans l’hétérogénéité des TCL.

L’adénovirus a été étudié dans l’optique de développer de nouveaux traitements pour différentes maladies. Les vecteurs adénoviraux (AdV) sont des outils intéressants du fait qu’ils peuvent être produits en grandes quantités (1X1012 particules par millilitre) et de par leur capacité à infecter des cellules quiescentes ou en division rapide. Les AdVs ont subi bon nombre de modifications pour leur permettre de traiter des cellules tumorales ou pour transporter des séquences génétiques exogènes essentielles pour le traitement de maladies monogéniques. Toutefois, les faibles niveaux d’expression du récepteur primaire de l’adénovirus, le CAR (récepteur à l’adénovirus et au virus coxsackie), réduit grandement l’efficacité de transduction dans plusieurs tumeurs. De plus, certains tissus normaux comme les muscles n’expriment que très peu de CAR, rendant l’utilisation des AdVs moins significative. Pour pallier à cette limitation, plusieurs modifications ont été générées sur les capsides virales. L’objectif de ces modifications était d’augmenter l’affinité des AdVs pour des récepteurs cellulaires spécifiques surexprimés dans les tumeurs et qui seraient exempts dans les tissus sains avoisinant. On peut mentionner dans les approches étudiées: l’utilisation de ligands bispécifiques, l’incorporation de peptides dans différentes régions de la fibre ou la substitution par une fibre de sérotypes différents. Notre hypothèse était que les domaines d’interaction complémentaire (K-Coil et ECoil) permettraient aux ligands de s’associer aux particules virales et d’altérer le tropisme de l’AdV. Pour ce faire, nous avons inclus un domaine d’interaction synthétique, le K-Coil,dans différentes régions de la fibre virale en plus de générer des mutations spécifiques pour abolir le tropisme naturel. Pour permettre la liaison avec les récepteurs d’intérêt dont l’EGF-R, l’IGF-IR et le CEA6, nous avons fusionné le domaine d’interaction complémentaire, le E-Coil, soit dans les ligands des récepteurs ciblés dont l’EGF et l’IGF-I, soit sur un anticorps à un seul domaine reconnaissant la protéine membranaire CEA6, l’AFAI. Suite à la construction des différents ligands de même que des différentes fibres virales modifiées, nous avons determiné tout d’abord que les différents ligands de même que les virus modifiés pouvaient être produits et que les différentes composantes pouvaient interagir ensemble. Les productions virales ont été optimisées par l’utilisation d’un nouveau protocole utilisant l’iodixanol. Ensuite, nous avons démontré que l’association des ligands avec le virus arborant une fibre modifiée pouvait entraîner une augmentation de transduction de 2 à 21 fois dans différentes lignées cellulaires. À cause de la difficulté des adénovirus à infecter les fibres musculaires occasionnée par l’absence du CAR, nous avons cherché à savoir si le changement de tropisme pourrait accroître l’infectivité des AdVs. Nous avons démontré que l’association avec le ligand bispécifique IGF-E5 permettait d’accroître la transduction autant dans les myoblastes que dans les myotubes de souris. Nous avons finalement réussi à démontrer que notre système pouvait induire une augmentation de 1,6 fois de la transduction suite à l’infection des muscles de souriceaux MDX. Ces résultats nous amènent à la conclusion que le système est fonctionnel et qu’il pourrait être évalué dans des AdVs encodant pour différents gènes thérapeutiques.

Fine particles of lithium ferrite were synthesized by the sol-gel method. By subsequent heat treatment at different temperatures, lithium ferrites of different grain sizes were prepared. A structural characterization of all the samples was conducted by the x-ray diffraction technique. A grain size of around 12 nm was observed for Li0.5Fe2.5O4 obtained through the sol-gel method. Magnetic properties of lithium ferrite nanoparticles with grain size ranging from 12 to 32 nm were studied. Magnetization measurements showed that Li0.5Fe2.5O4 fine particles exhibit a deviation from the predicted magnetic behaviour. The as-prepared sample of lithium ferrite showed a maximum saturation magnetization of 75 emu g−1. Variation of coercivity is attributed to the transition from multi-domain to single domain nature. Dielectric permittivity and ac conductivity of all the samples were evaluated as a function of frequency, temperature and grain size. Variation of permittivity and ac conductivity with frequency reveals that the dispersion is due to the Maxwell–Wagner type interfacial polarization

The cupin superfamily is a group of functionally diverse proteins that are found in all three kingdoms of life, Archaea, Eubacteria, and Eukaryota. These proteins have a characteristic signature domain comprising two histidine- containing motifs separated by an intermotif region of variable length. This domain consists of six beta strands within a conserved beta barrel structure. Most cupins, such as microbial phosphomannose isomerases (PMIs), AraC- type transcriptional regulators, and cereal oxalate oxidases (OXOs), contain only a single domain, whereas others, such as seed storage proteins and oxalate decarboxylases (OXDCs), are bi-cupins with two pairs of motifs. Although some cupins have known functions and have been characterized at the biochemical level, the majority are known only from gene cloning or sequencing projects. In this study, phylogenetic analyses were conducted on the conserved domain to investigate the evolution and structure/function relationships of cupins, with an emphasis on single- domain plant germin-like proteins (GLPs). An unrooted phylogeny of cupins from a wide spectrum of evolutionary lineages identified three main clusters, microbial PMIs, OXDCs, and plant GLPs. The sister group to the plant GLPs in the global analysis was then used to root a phylogeny of all available plant GLPs. The resulting phylogeny contained three main clades, classifying the GLPs into distinct subfamilies. It is suggested that these subfamilies correlate with functional categories, one of which contains the bifunctional barley germin that has both OXO and superoxide dismutase (SOD) activity. It is proposed that GLPs function primarily as SODs, enzymes that protect plants from the effects of oxidative stress. Closer inspection of the DNA sequence encoding the intermotif region in plant GLPs showed global conservation of thymine in the second codon position, a character associated with hydrophobic residues. Since many of these proteins are multimeric and enzymatically inactive in their monomeric state, this conservation of hydrophobicity is thought to be associated with the need to maintain the various monomer- monomer interactions. The type of structure-based predictive analysis presented in this paper is an important approach for understanding gene function and evolution in an era when genomes from a wide range of organisms are being sequenced at a rapid rate.

This review summarizes the recent discovery of the cupin superfamily (from the Latin term "cupa," a small barrel) of functionally diverse proteins that initially were limited to several higher plant proteins such as seed storage proteins, germin (an oxalate oxidase), germin-like proteins, and auxin-binding protein. Knowledge of the three-dimensional structure of two vicilins, seed proteins with a characteristic beta-barrel core, led to the identification of a small number of conserved residues and thence to the discovery of several microbial proteins which share these key amino acids. In particular, there is a highly conserved pattern of two histidine-containing motifs with a varied intermotif spacing. This cupin signature is found as a central component of many microbial proteins including certain types of phosphomannose isomerase, polyketide synthase, epimerase, and dioxygenase. In addition, the signature has been identified within the N-terminal effector domain in a subgroup of bacterial AraC transcription factors. As well as these single-domain cupins, this survey has identified other classes of two-domain bicupins including bacterial gentisate 1, 2-dioxygenases and 1-hydroxy-2-naphthoate dioxygenases, fungal oxalate decarboxylases, and legume sucrose-binding proteins. Cupin evolution is discussed from the perspective of the structure-function relationships, using data from the genomes of several prokaryotes, especially Bacillus subtilis. Many of these functions involve aspects of sugar metabolism and cell wall synthesis and are concerned with responses to abiotic stress such as heat, desiccation, or starvation. Particular emphasis is also given to the oxalate-degrading enzymes from microbes, their biological significance, and their value in a range of medical and other applications.

The recently described cupin superfamily of proteins includes the germin and germinlike proteins, of which the cereal oxalate oxidase is the best characterized. This superfamily also includes seed storage proteins, in addition to several microbial enzymes and proteins with unknown function. All these proteins are characterized by the conservation of two central motifs, usually containing two or three histidine residues presumed to be involved with metal binding in the catalytic active site. The present study on the coding regions of Synechocystis PCC6803 identifies a previously unknown group of 12 related cupins, each containing the characteristic two-motif signature. This group comprises 11 single-domain proteins, ranging in length from 104 to 289 residues, and includes two phosphomannose isomerases and two epimerases involved in cell wall synthesis, a member of the pirin group of nuclear proteins, a possible transcriptional regulator, and a close relative-of a cytochrome c551 from Rhodococcus. Additionally, there is a duplicated, two-domain protein that has close similarity to an oxalate decarboxylase from the fungus Collybia velutipes and that is a putative progenitor of the storage proteins of land plants.

The cupin superfamily of proteins, named on the basis of a conserved β-barrel fold (‘cupa’ is the Latin term for a small barrel), was originally discovered using a conserved motif found within germin and germin-like proteins from higher plants. Previous analysis of cupins had identified some 18 different functional classes that range from single-domain bacterial enzymes such as isomerases and epimerases involved in the modification of cell wall carbohydrates, through to two-domain bicupins such as the desiccation-tolerant seed storage globulins, and multidomain transcription factors including one linked to the nodulation response in legumes. Recent advances in comparative genomics, and the resolution of many more 3-D structures have now revealed that the largest subset of the cupin superfamily is the 2-oxyglutarate-Fe2+ dependent dioxygenases. The substrates for this subclass of enzyme are many and varied and in total amount to probably 50–100 different biochemical reactions, including several involved in plant growth and development. Although the majority of enzymatic cupins contain iron as an active site metal, other members contain either copper, zinc, cobalt, nickel or manganese ions as a cofactor, with each cofactor allowing a different type of chemistry to occur within the conserved tertiary structure. This review discusses the range of structures and functions found in this most diverse of superfamilies.

The model amyloid peptide AAKLVFF was expressed as a His-tagged fusion protein with the immunoglobulin-binding domain B1 of streptococcal protein G (GB1), a small (56 residues), stable, single-domain protein. It is shown that expression of this model amyloid peptide is possible and is not hindered by aggregation. Formylation side reactions during the CNBr cleavage are investigated via synthesis of selectively formylated peptides.

Empowerment is a standard but ambiguous element of development rhetoric and so, through the socially complex and contested terrain of South Africa, this paper explores its potential to contribute to inclusive development. Investigating micro-level engagements with the national strategy of Broad-Based Black Economic Empowerment (B-BBEE) in the South African wine industry highlights the limitations, but also potential, of this single domain approach. However, latent paternalism, entrenched interests and a ‘dislocated blackness’ maintain a complex racial politics that shapes both power relations and the opportunities for transformation within the industry. Nonetheless, while B-BBEE may not, in reality, be broad-based its manifestations are contributing to challenging racist structures and normalising changing attitudes. This paper concludes that, to be transformative, empowerment needs to be re-embedded within South Africa as a multi-scalar, multi-dimensional dialogue and, despite the continuation of structural constraints, positions the local as a critical scale at which to initiate broader social change.

Objectives: Cognitive decline related to neurocysticercosis (NC) remains poorly characterized and underdiagnosed. In a cross-sectional study with a prospective phase, we evaluated cognitive decline in patients with strictly calcified form (C-NC), the epidemiologically largest subgroup of NC, and investigated whether there is a spectrum of cognitive abnormalities in the disease. Methods: Forty treatment-naive patients with C-NC aged 37.6 +/- 11.3 years and fulfilling criteria for definitive C-NC were submitted to a comprehensive cognitive and functional evaluation and were compared with 40 patients with active NC (A-NC) and 40 healthy controls (HC) matched for age and education. Patients with dementia were reassessed after 24 months. Results: Patients with C-NC presented 9.4 +/- 3.1 altered test scores out of the 30 from the cognitive battery when compared to HC. No patient with C-NC had dementia and 10 patients (25%) presented cognitive impairment-no dementia (CIND). The A-NC group had 5 patients (12.5%) with dementia and 11 patients (27.5%) with CIND. On follow-up, 3 out of 5 patients with A-NC with dementia previously still presented cystic lesions with scolex on MRI and still had dementia. One patient died and the remaining patient no longer fulfilled criteria for either dementia or CIND, presenting exclusively calcified lesions on neuroimaging. Conclusions: Independently of its phase, NC leads to a spectrum of cognitive abnormalities, ranging from impairment in a single domain, to CIND and, occasionally, to dementia. The objective of this work were apply and provide a preliminary evaluation of the Weather Research and Forecasting model coupled with Chemistry (WRF-Chem) performance, for Londrina region. We performed comparison with measurements obtained in meteorological stations. The model was configured to run with three domains with 27,9 and 3 km of grid resolution, using the ndown program and also was realized a simulation with the model configured to run with a single domain using a land use file based in a classified image for region of MODIS sensor. The emission files to supply the chemistry run were generated based in the work of Martins et al., 2012. RADM2 chemical mechanism and MADE/SORGAM modal aerosol models were used in the simulations. The results demonstrated that model was able to represent coherently the formation and dispersion of the pollution in Metropolitan Region of Londrina and also the importance of using the appropriate land use file for the region.

Background: Dementia is a multifaceted disorder that impairs cognitive functions, such as memory, language, and executive functions necessary to plan, organize, and prioritize tasks required for goal-directed behaviors. In most cases, individuals with dementia experience difficulties interacting with physical and social environments. The purpose of this study was to establish ecological validity and initial construct validity of a fire evacuation Virtual Reality Day-Out Task (VR-DOT) environment based on performance profiles as a screening tool for early dementia. Objective: The objectives were (1) to examine the relationships among the performances of 3 groups of participants in the VR-DOT and traditional neuropsychological tests employed to assess executive functions, and (2) to compare the performance of participants with mild Alzheimer’s-type dementia (AD) to those with amnestic single-domain mild cognitive impairment (MCI) and healthy controls in the VR-DOT and traditional neuropsychological tests used to assess executive functions. We hypothesized that the 2 cognitively impaired groups would have distinct performance profiles and show significantly impaired independent functioning in ADL compared to the healthy controls. Methods: The study population included 3 groups: 72 healthy control elderly participants, 65 amnestic MCI participants, and 68 mild AD participants. A natural user interface framework based on a fire evacuation VR-DOT environment was used for assessing physical and cognitive abilities of seniors over 3 years. VR-DOT focuses on the subtle errors and patterns in performing everyday activities and has the advantage of not depending on a subjective rating of an individual person. We further assessed functional capacity by both neuropsychological tests (including measures of attention, memory, working memory, executive functions, language, and depression). We also evaluated performance in finger tapping, grip strength, stride length, gait speed, and chair stands separately and while performing VR-DOTs in order to correlate performance in these measures with VR-DOTs because performance while navigating a virtual environment is a valid and reliable indicator of cognitive decline in elderly persons. Results: The mild AD group was more impaired than the amnestic MCI group, and both were more impaired than healthy controls. The novel VR-DOT functional index correlated strongly with standard cognitive and functional measurements, such as mini-mental state examination (MMSE; rho=0.26, P=.01) and Bristol Activities of Daily Living (ADL) scale scores (rho=0.32, P=.001). Conclusions: Functional impairment is a defining characteristic of predementia and is partly dependent on the degree of cognitive impairment. The novel virtual reality measures of functional ability seem more sensitive to functional impairment than qualitative measures in predementia, thus accurately differentiating from healthy controls. We conclude that VR-DOT is an effective tool for discriminating predementia and mild AD from controls by detecting differences in terms of errors, omissions, and perseverations while measuring ADL functional ability.

In the current model for bacterial cell division, the FtsZ protein forms a ring that marks the division plane, creating a cytoskeletal framework for the subsequent action of other essential division proteins such as FtsA and ZipA. The putative protein complex ultimately generates the division septum. The essential cell division protein FtsZ is a functional and structural homolog of eukaryotic tubulin, and like tubulin, FtsZ hydrolyzes GTP and self-assembles into protein filaments in a strictly GTP-dependent manner. FtsA shares sequence similarity with members of the ATPase superfamily that include actin, but its actual function remains unknown. To test the division model and elucidate functions of the division proteins, this dissertation primarily focuses on the analysis of FtsZ and FtsA in Escherichia coli. ^ By tagging with green fluorescent protein, we first demonstrated that FtsA also exhibits a ring-like structure at the potential division site. The localization of FtsA was dependent on functional FtsZ, suggesting that FtsA is recruited to the septum by the FtsZ ring. In support of this idea, we showed that FtsA and FtsZ directly interact. Using a novel E. coli in situ assay, we found that the FtsA-FtsZ interaction appears to be species-specific, although an interspecies interaction could occur between FtsA and FtsZ proteins from two closely related organisms. In addition, mutagenesis of FtsA revealed that no single domain is solely responsible for its septal localization or interaction with FtsZ. To explore the function of FtsA, we purified FtsA protein and demonstrated that it has ATPase activity. Furthermore, purified FtsA stimulates disassembly of FtsZ polymers in a sedimentation assay but does not affect GTP hydrolysis of FtsZ. This result suggests that in the cell, FtsA may function similarly in regulating dynamic instability of the FtsZ ring during the cell division process. ^ To elucidate the structure-function relationship of FtsZ, we carried out thorough genetic and functional analyses of the mutagenized FtsZ derivatives. Our results indicate that the conserved N-terminal domain of FtsZ is necessary and sufficient for FtsZ self-assembly and localization. Moreover, we discovered a critical role for an extreme C-terminal domain of FtsZ that consists of only 12 residues. Truncated FtsZ derivatives lacking this domain, though able to polymerize and localize, are defective in ring formation in vivo as well as interaction with FtsA and ZipA. Alanine scanning mutagenesis of this region pinpointed at least five residues necessary for the function of FtsZ. Studies of protein levels and protein-protein interactions suggested that these residues may be involved in regulating protein stability and/or FtsZ-FtsA interactions. Interestingly, two of the point mutants exhibited dominant-negative phenotypes. ^ In summary, results from this thesis work have provided additional support for the division machinery model and will contribute to a better understanding of the coordinate functions of FtsA and FtsZ in the cell division process. ^

The NRM intensity, AF demagnetization characteristics, hysteresis parameters, initial susceptibility, and thermomagnetic characteristics of 18 basalt specimens from Deep Sea Drilling Project Hole 504B were determined. In six samples, the grain size was large enough to allow microprobe analysis. We conclude (1) that the dominant magnetic mineral is titanomagnetite/titanomaghemite; (2) that, except for the upper few meters of the core where the grains are in the stable monodomain state, the grain size of the magnetic mineral lies in the pseudo-single-domain range (< 10 µm); (3) that maghemitization (i.e., low-temperature (< 350°C) oxidation) has taken place. We discuss possible geological histories.
