This research introduces a novel dressing for burn wounds, containing silver nanoparticles in hydrogels for infected burn care. The 2-acrylamido-2-methylpropane sulfonic acid sodium salt hydrogels containing silver nanoparticles have been prepared via ultraviolet radiation. The formation of silver nanoparticles was monitored by surface plasmon bands and transmission electron microscopy. The concentration of silver nitrate loaded in the solutions slightly affected the physical properties and mechanical properties of the neat hydrogel. An indirect cytotoxicity study found that none of the hydrogels were toxic to tested cell lines. The measurement of cumulative release of silver indicated that 70%–82% of silver was released within 72 hr. The antibacterial activities of the hydrogels against common burn pathogens were studied and the results showed that 5 mM silver hydrogel had the greatest inhibitory activity. The results support its use as a potential burn wound dressing.

Serious Childhood Sexual Assault (CSA) has the potential for a wide range of extreme difficulties, not only in childhood but for many years to come. However, the lasting negative impacts are not inevitable and with the right help, those who have experienced CSA can go on to enjoy whole and fulfilling lives. The question posed in this research is “What comprises “the right help”? The present study qualitatively investigated the narratives of men and women with regard to what was deemed important components of counselling which facilitated their healing from childhood sexual assault. Participants consisted of 21 women and 11 men who had sought both individual and group counselling to work through issues stemming from traumatic sexual assault in childhood. Findings presented here describe three superordinate themes which were essential in the healing process, incorporating abuse-specific, client-specific and practitioner-specific components. A final superordinate theme of negative experiences in counselling is also presented, describing unhelpful or negative counselling experiences. Similarities between men and women as to the important components of counselling was remarkable, however important gender differences were also observed.

This study was based on the premise that variation is the foundation of statistics and statistical investigations. The study followed the development of fourth-grade students' understanding of variation through participation in a sequence of two lessons based on measurement. In the first lesson all students measured the arm span of one student, revealing pathways students follow in developing understanding of variation and linear measurement (related to research question 1). In the second lesson each student's arm span was measured once, introducing a different aspect of variation for students to observe and contrast. From this second lesson, students' development of the ability to compare their representations for the two scenarios and explain differences in terms of variation was explored (research question 2). Students' documentation, in both workbook and software formats, enabled us to monitor their engagement and identify their increasing appreciation of the need to observe, represent, and contrast the variation in the data. Following the lessons, a written student assessment was used for judging retention of understanding of variation developed through the lessons and the degree of transfer of understanding to a different scenario (research question 3).

By modifying the electrodeposition technique, we have stabilized the silver nanowires (AgNWs) in high-energy hexagonal closed packed (hcp)structure. The conductivity noise measurements show that the noise magnitude in hcp silver nanowires is several orders of magnitude smaller than that of face centered cubic (fcc) silver nanowires, which is obtained by standard over potential lectrodeposition (OPD)technique. The reduction of noise can be attributed to the restricted dislocation dynamics in hcp AgNWs due to the presence of less number of slip systems. Temperature dependent noise measurements show that the noise magnitude in hcp AgNWs is weakly temperature dependent while in fcc AgNWs it is strong function of temperature.

Electrical conductivities and dielectric properties of glassy Ag4P2O7 have been investigated as a function of temperature and frequency. The variation of the properties is consistent with the structure of this glass which consists of a variety of polymeric anion species. Upon crystallization Ag4P2O7 appears to retain some of the anionic species in the solid solution as evident from the phase transition behaviour at higher temperatures.

A commercial acrylic fiber with 92% (w/w) acrylonitrile content was partially hydrolyzed converting a fraction of the nitrile (-CN) groups to carboxylic acid (-COOH) groups, to coat the fiber with polyethylenimine (PEI) resin, which was then crosslinked with glutaraldehyde and further quaternized with ethyl chloroacetate to produce a novel strong-base anionic exchanger in the form of fiber. Designated as PAN(QPEI.XG)(Cl-), the fibrous sorbent was compared with a commercial bead-form resin Amberlite IRA-458(Cl-) in respect of sorption capacity, selectivity, and kinetics for removal of silver thiosulfate complexes from aqueous solutions. Though the saturation level of [Ag(S2O3)(2)](3-) on PAN(QPEI.XG)(Cl-) is considerably less than that on IRA-458(Cl-), the gel-coated fibrous sorbent exhibits, as compared to the bead-form sorbent, a significantly higher sorption selectivity for the silver thiosulfate complex in the presence of excess of other anions Such as S2O32-, SO42-, and Cl-, and a remarkably faster rate of both sorption and stripping. The initial uptake of the sorbate by the fibrous sorbent is nearly instantaneous, reaching up to similar to 80% of the saturation capacity within 10 s, as compared to only similar to 12% on the bead-form sorbent. The high initial rate of uptake fits a shell-core kinetic model for sorption on fiber of cylindrical geometry. With 4M HCl, the stripping of the sorbed silver complex from the fibrous sorbent is clean and nearly instantaneous, while, in contrast, a much slower rate of stripping on the bead-form sorbent leads to its fouling due to a slow decomposition of the silver thiosulfate complex in the acidic medium.

The removal of noncoding sequences, or introns, from the eukaryotic messenger RNA precursors is catalyzed by a ribonucleoprotein complex known as the spliceosome. In most eukaryotes, two distinct classes of introns exist, each removed by a specific type of spliceosome. The major, U2-type introns account for over 99 % of all introns, and are almost ubiquitous. The minor, U12-type introns are found in most but not all eukaryotes, and reside in conserved locations in a specific set of genes. Due to their slow excision rates, the U12-type introns are expected to be involved in the regulation of the genes containing them by inhibiting the maturation of the messenger RNAs. However, little information is currently available on how the activity of the U12-dependent spliceosome itself is regulated. The levels of many known splicing factors are regulated through unproductive alternative splicing events, which lead to inclusion of premature STOP codons, targeting the transcripts for destruction by the nonsense-mediated decay pathway. These alternative splice sites are typically found in highly conserved sequence elements, which also contain binding sites for factors regulating the activation of the splice sites. Often, the activation is achieved by binding of products of the gene in question, resulting in negative feedback loops. In this study, I show that U11-48K, a protein factor specific to the minor spliceosome, specifically recognizes the U12-type 5' splice site sequence, and is essential for proper function of the minor spliceosome. Furthermore, the expression of U11-48K is regulated through a feedback mechanism, which functions through conserved sequence elements that activate alternative splicing and nonsense-mediated decay. This mechanism is conserved from plants to animals, highlighting both the importance and early origin of this mechanism in regulating splicing factors. I also show that the feedback regulation of U11-48K is counteracted by a component of the major spliceosome, the U1 small nuclear ribonucleoprotein particle, as well as members of the hnRNP F/H protein family. These results thus suggest that the feedback mechanism is finely tuned by multiple factors to achieve precise control of the activity of the U12-dependent spliceosome.

We describe the fabrication of silver nanotriangle array using angle resolved nanosphere lithography and utilizing the same for enhancing fluorescence. The well established nanosphere lithography is modified by changing the angle of deposition between the nanosphere mask and the beam of silver being deposited resulting in nanotriangles of varying surface area and density. The 470 nm plasmon resonance wavelength of the substrate was determined using minimum reflectivity method which closely matches with excitation wavelength of the fluorophore. Ten times enhancement in fluorescence emission intensity is obtained from fluorescein isothiocyanate coated on top of silver nanotriangle array separated by a spacer layer of poly vinyl alcohol as compared to glass. The enhanced fluorescence emission is attributed to the increase in local field enhancement.

The effect of silver nanoparticles (nAg) in PS/PVME polystyrene/poly(vinyl methyl ether)] blends was studied with respect to the evolution of morphology, demixing temperature, and segmental dynamics. In the early stage of demixing, PVME developed an interconnected network that coarsened in the late stage. The nAg induced miscibility in the blends as supported by shear rheological measurements. The physicochemical processes that drive phase separation in blends also led to migration of nAg to the PVME phase as supported by AFM. The segmental dynamics was greatly influenced by the presence of nAg due to the specific interaction of nAg with PVME. Slower dynamics and an increase in intermolecular cooperativity in the presence of nAg further supported the role of nAg in delaying the phase separation processes and augmenting the demixing temperature in the blends. Different theoretical models were assessed to gain insight into the dynamic heterogeneity in PS/PVME blends at different length scales.

A silver ion (Ag+)-triggered thixotropic metallo(organo)gel of p-pyridyl-appended oligo(p-phenylenevinylene) derivatives (OPVs) is reported for the first time. Solubilization of single-walled carbon nanohorns (SWCNHs) in solutions of the pure OPVs as well as in the metallogels mediated by pi-pi interactions has also been achieved. In situ fabrication of silver nanoparticles (AgNPs) in the SWCNH-doped dihybrid gel leads to the formation of a trihybrid metallogel. The mechanical strength of the metallogels could be increased step- wise in the order: freshly prepared gel indicate the formation of three distinct morphologies, that is, nanoscale metal organic particles (NMOPs), flowerlike aggregates of SWCNHs and AgNPs, and also their integration with each other. Detailed studies suggest lamellar organizations of the linear metal ligand complexes in the NMOPs, which upon association create a three-dimensional network that eventually immobilizes the solvent molecules.

Computational general relativity is a field of study which has reached maturity only within the last decade. This thesis details several studies that elucidate phenomena related to the coalescence of compact object binaries. Chapters 2 and 3 recounts work towards developing new analytical tools for visualizing and reasoning about dynamics in strongly curved spacetimes. In both studies, the results employ analogies with the classical theory of electricity and magnitism, first (Ch. 2) in the post-Newtonian approximation to general relativity and then (Ch. 3) in full general relativity though in the absence of matter sources. In Chapter 4, we examine the topological structure of absolute event horizons during binary black hole merger simulations conducted with the SpEC code. Chapter 6 reports on the progress of the SpEC code in simulating the coalescence of neutron star-neutron star binaries, while Chapter 7 tests the effects of various numerical gauge conditions on the robustness of black hole formation from stellar collapse in SpEC. In Chapter 5, we examine the nature of pseudospectral expansions of non-smooth functions motivated by the need to simulate the stellar surface in Chapters 6 and 7. In Chapter 8, we study how thermal effects in the nuclear equation of state effect the equilibria and stability of hypermassive neutron stars. Chapter 9 presents supplements to the work in Chapter 8, including an examination of the stability question raised in Chapter 8 in greater mathematical detail.

As práticas discursivas que constituem e sustentam as concepções de sexo e de sexualidade nas sociedades contemporâneas são fortemente arraigadas no discurso normalizado e normalizador segundo o qual há apenas dois sexos que se evidenciam em corpos masculinos ou femininos, naturalmente distintos biológica e fisionomicamente e reconhecíveis em modos de ser diversos - mas perfeitamente identificáveis com o sexo biológico (nascer com pênis/nascer com vagina). Esse sistema dicotômico afirmou por muitos anos uma concepção inequívoca de o que é e como ser homem ou mulher; restringiu os corpos a uma performance estereotipada de masculinidade e de feminilidade; conformou os discursos sobre gênero e sexo e naturalizou a heterossexualidade. Entretanto, o sistema binário no qual se funda a heterossexualidade encontra fissuras ante as identidades sexuais e de gênero que ora se evidenciam e desestabilizam a (hetero)sexualidade normativa. Sustentadas pela teoria queer e por autores como Judith Butler, Michel Foucault e Beatriz Preciado, minhas discussões nesta tese focalizam as repercussões e marcas dos discursos escolares na produção de corpos, gêneros e sexualidades de lésbicas, gays, bissexuais e transgêneros LGBT. Interessa indagar, a partir das narrativas desses sujeitos, as formas como as sexualidades têm sido trabalhadas como conteúdo escolar e discutir os possíveis efeitos das práticas escolares na constituição das sexualidades LGBT. Como constituir-se como sujeito fora da norma em um espaço-tempo altamente regulado como o currículo escolar? Busco evidenciar que, mesmo às margens dos discursos socialmente legitimados que circulam na escola, cuja pretensão é manter a hegemonia da história universal onde apenas alguns sujeitos são inscritos e reconhecidos como inteligíveis, os sujeitos LGBT produzem suas histórias e criam modos de vida. Minhas contribuições ao debate do tema se referem à possibilidade de desconstrução dos discursos predominantes no espaço-tempo da escola que enfatizam as concepções naturalizadas de sexo, de gênero e de sexualidade. Neste sentido acentuo a necessidade de novas/outras abordagens que incluam todas as sexualidades no espaço de inteligibilidade habitado pela norma heterossexual e apontar indícios de como os discursos predominantes contribuem para a manutenção da heteronormatividade e do heterossexismo. As narrativas dos sujeitos LGBT apontam a necessidade de repensar as práticas escolares, visando desconstruir concepções naturalizadas em torno da vivência da sexualidade e avançar das políticas de tolerância para uma política da diferença na qual a concepção do que é ser humano esteja sempre em aberto.

Esse estudo, inscrito no registro das teorias pós-fundacionais, se propõe a investigar os discursos do perfil docente nas políticas de formação de professores. Para tal, realiza uma análise de um conjunto de textos da produção acadêmica, que constam dos Anais da 36 Reunião Anual da Associação Nacional de Pós-Graduação e Pesquisa em Educação - ANPEd, buscando interpretar os mecanismos que operam nessa construção discursiva. Nesse sentido, orienta-se pelas contribuições teórico-metodológicas da Teoria do Discurso proposta por Ernesto Laclau e Chantal Mouffe, buscando problematizar perspectivas essencialistas de professor. Defende que o discurso do protagonismo docente, sedimentado nos textos investigados, não é a representação plena de um fundamento de docência dado a priori, mas se constitui na luta política por hegemonizar um modelo de professor e de formação significados como via para o alcance da qualidade da educação. Dessa forma, os significantes do protagonismo docente, que assumem centralidade no discurso, se constituem em resposta a um antagonismo o gerencialismo docente identificado nas políticas neoliberais , construindo sentidos de professor na contingência. O estudo aponta para a potencialização de dois discursos nessa construção discursiva: a relação entre perfil docente e qualidade da educação; e a ideia de controle da identidade, do trabalho, da profissão e da formação de professores. Problematiza essa possibilidade de controle, pela compreensão do social como linguagem, colocando em questão a ideia de um fundamento inteligível, de uma racionalidade que consiga saturar todas as relações sociais, que possibilite acessar o sentido originário de professor, de trabalho docente, de profissão, de formação. Assumem centralidade no discurso produzido a partir dessa investigação: a significação da dimensão epistemológica da cultura como constitutiva do professor, do seu trabalho, da sua profissão, da sua formação; a diferença, significada como proliferação e adiamento de sentidos, como tensão entre regulação e escape no processo de significação. A partir desse estudo, defende-se que os discursos do perfil docente, bem como os projetos de formação de professores, se constituem na decisão razoável, em resposta à diferença, entendida como própria de um social que não é expressão de um fundamento, que não pode ser apreendido por uma racionalidade, mas que vai sendo constituído provisoriamente e contingencialmente. Ressalta-se, ainda, a produtividade da diferença, do conflito, do dissenso, como possibilidade de emergência de alternativas na significação do perfil docente.
