A novel variant genotype C of hepatitis B virus identified in isolates from Australian Aborigines: complete genome sequence and phylogenetic relatedness

There have been no reports of DNA sequences of hepatitis B virus (HBV) strains from Australian Aborigines, although the hepatitis B surface antigen (HBsAg) was discovered among them. To investigate the characteristics of DNA sequences of HBV strains from Australian Aborigines, the complete nucleotide sequences of HBV strains were determined and subjected to molecular evolutionary analysis. Serum samples positive for HBsAg were collected from five Australian Aborigines. Phylogenetic analysis of the five complete nucleotide sequences compared with DNA sequences of 54 global HBV isolates from international databases revealed that three of the five were classified into genotype D and were most closely related in terms of evolutionary distance to a strain isolated from a healthy blood donor in Papua New Guinea. Two of the five were classified into a novel variant genotype C, which has not been reported previously, and were closely related to a strain isolated from Polynesians, particularly in the X and Core genes. These two strains of variant genotype C differed from known genotype C strains by 5.9-7.4% over the complete nucleotide sequence and 4.0-5.6 % in the small-S gene, and had residues Arg(122), Thr(127) and Lys(160) characteristic of serotype ayw3, which have not been reported previously in genotype C. In conclusion, this is the first report of the characteristics of complete nucleotide sequences of HBV from Australian Aborigines. These results contribute to the investigation of the worldwide spread of HBV, the relationship between serotype and genotype and the ancient common origin of Australian Aborigines.

A soluble form of CD83 is released from activated dendritic cells and B lymphocytes, and is detectable in normal human sera

CD83 is an inducible glycoprotein expressed predominantly by dendritic cells (DC) and B lymphocytes. Expression of membrane CD83 (mCD83) is widely used as a marker of differentiated/ activated DC but its function and ligand(s) are presently unknown. We report the existence of a soluble form of CD83 (sCD83). Using both a sCD83-specific ELISA and Western blotting, we could demonstrate the release of sCD83 by mCD83(+) B cell and Hodgkin's disease-derived cell lines, but not mCD83(-) cells. Inhibition of de novo protein synthesis did not affect the release of sCD83 during short-term (2 h) culture of cell lines although mCD83 expression was significantly reduced, suggesting sCD83 is generated by the release of mCD83. Isolated tonsillar B lymphocytes and monocyte-derived DC, which are mCD83(low), released only low levels of sCD83 during culture. However, the differentiation/activation of these populations both up-regulated mCD83 and increased sCD83 release significantly. Analysis of sera from normal donors demonstrated the presence of low levels (121 +/- 3.6 pg/ml) of circulating sCD83. Further studies utilizing purified sCD83 and the analysis of sCD83 levels in disease may provide clues to the function and ligand(s) of CD83.

Experimental investigation of liquid flow shift due to gas cross flow in non-wetted packed beds

An experimental study has been carried out for the gas-liquid two-phase flow in a packed bed simulating conditions of the gas and liquid flows in the lower part of blast furnace. The localised liquid flow phenomenon in presence of gas cross flow, which usually occurs around the cohesive zone and raceway in blast furnace, was investigated in detail. Such liquid flow is characterised in terms of liquid shift distance or liquid shift angle that can effectively be measured by the experiments involved in the current study. It is found that liquid shift angle does not significantly increase or decrease with different packing depth. This finding supports the hypothesis of the force balance model where a vectorial relationship among acting forces, i.e. gas drag force, gravitational force and solid-liquid friction force, and liquid shift angle does exist. Liquid shift angle is inversely proportional to particle size and liquid density, and proportional to square of gas superficial velocity, but is almost independent on liquid flowrate and liquid viscosity. The gas-liquid drag coefficient, an important aspect for quantifying the interaction between gas and liquid flows, was conceptually modified based on the discrete feature of liquid flow through a packed bed and evaluated by the combined theoretical and experimental investigation. Experimental measurements suggest that the gas-liquid drag coefficient is approximately a constant (C-DG(')=5.4+/-1.0) and is independent on liquid properties, gas velocity and packing structure. The result shows a good agreement with previous experimental data and prediction of the existing liquid flow model.

Effect of storage time and conditions on the seed coat colour of Australian adzuki beans

Two varieties of adzuki beans (Vigna angularis), Bloodwood and Erimo, were stored at temperatures of 10, 20 or 30degreesC, and relative humidities (RH) 40 or 65%, and samples were analysed at 0, 1.5, 3 and 6 months. Storage at 30degreesC for > 1.5 months caused a significant decrease in the a(star) and b(star) colour values and darkening of the seed coat. Beans stored at 65% RH had lower L-star but higher a(star) and b(star) colour values than those stored at 40% RH. Bloodwood and Erimo samples showed similar trends in colour during storage. The best storage conditions for the preservation of the adzuki colour were 10degreesC and 65% RH. The Australian beans had lower L-star, a(star) and b(star) colour values than Japanese Erimo-shouzu beans and storage increased the difference.

Mão-de-obra do Brasil para o Japão: a aspectos econômicos e impactos nas empresas do fenômeno dekassegui

A crise econômica no Brasil e principalmente o desalento quanto às perspectivas de melhoras têm levado muitos brasileiros a procurar trabalhos desqualificados no exterior. Um movimento de transferência de trabalhadares brasileiros é aquele para o Japão, que vem ocorrendo desde 1985. Estes trabalhadores, japoneses e descendentes, seriam mais de 100.000. Neste artigo, procura-se verificar as causas e as conseqüências mais imediatas e perceptíveis deste processo, com ênfase nos impactos no mercado de trabalho e nas empresas.

Área de influência: um estudo no varejo de supermercados

O artigo procura dar uma contribuição ao conhecimento varejista no Brasil, integrando aspectos teóricos e empíricos sobre a área de influência, conceito de fundamental importância no marketing varejista. A metodologia engloba uma revisão do conhecimento teórico existente e desenvolve uma investigação sobre o fenômeno da área de influência com base em pesquisa empírica com clientes de supermercados brasileiros. A descoberta de certas regularidades na área de influência ocorreu quando se passou a analisar o fenômeno por meio de curvas acumuladas de clientes. Verificou-se que o tamanho da loja e a densidade populacional parecem ser fatores determinantes da extensão da área de influência.

Capacidades dinâmicas e rituais de interação entre alta e média gerência: proposta de um framework

O presente artigo foi desenvolvido considerando situações de incerteza ambiental, em que a ordem do dia ou questão estratégica (strategic issue) é a inovação dos recursos, ou seja, transformação, renovação ou desenvolvimento de uma nova capacidade funcional (ordinária). Com o objetivo de compreender como as capacidades dinâmicas desempenham esse papel na inovação dos ativos organizacionais em tais contextos ambientais, propõe-se aqui uma perspectiva alternativa de análise a partir de uma vertente microssociológica, a qual recorre aos elementos constitutivos de rituais de interação segundo Collins (2004). Essa perspectiva oferece possibilidades de investigação da influência das conversações estratégicas, que ocorrem em meio a práticas ritualísticas de interação entre alta e média gerência, sobre o potencial de inovação das capacidades organizacionais. Para compreensão da articulação teórica realizada no trabalho, apresenta-se um framework de análise que agrega a incerteza no nível macro com variáveis comportamentais e organizacionais no nível micro, enquanto elementos mais salientes dos rituais de interação. Visando estudos futuros e melhor compreensão do framework do trabalho, são apresentadas proposições acerca das relações do modelo após o detalhamento da estrutura analítica.
