Aquesta tesi doctoral està basada en el desenvolupament de nous agents antimicrobians derivats del pèptid híbrid cecropina A-melitina WKLFKKILKVL-NH2 (Pep3) que siguin sostenibles i útils per al control de malalties de plantes. Es van dissenyar i sintetitzar més de 133 anàlegs de Pep3 mitjançant química combinatòria. Es van obtenir anàlegs de Pep3 amb una elevada activitat contra fitopatògens i que presentaven baixa toxicitat. Els millors anàlegs van presentar eficàcies comparables amb pesticides de referència en la prevenció d'infeccions causades per fitopatògens. Es va estudiar el mecanisme d'acció de KKLFKKILKYL-NH2 (BP100) investigant la seva interacció amb models de membrana mitjançant tècniques espectroscòpiques. Es va observar la capacitat de BP100 a induir la permeabilització, la neutralització, i l'agregació de vesícules lipídiques aniòniques a una determinada concentració llindar. Es va deduir una equació que relaciona la CMI d'un pèptid antimicrobià amb la constant de partició i la concentració llindar en la membrana.

Forestry and other activities are increasing in the boreal mixedwood of Alberta, with a concomitant decrease in older forest. The Barred Owl (Strix varia) is an old-growth indicator species in some jurisdictions in North America. Hence, we radio-tagged Barred Owls in boreal mixedwood in Alberta to determine whether harvesting influenced habitat selection. We used three spatial scales: nest sites, i.e., nest tree and adjacent area of 11.7 m radius around nests, nesting territory of 1000 m radius around nests, and home range locations within 2000 m radius of the home range center. Barred Owls nested primarily in balsam poplar (Populus balsamifera) snags > 34 cm dbh and nest trees were surrounded by large, > 34 cm dbh, balsam poplar trees and snags. Nesting territories contained a variety of habitats including young < 80-yr-old, deciduous-dominated stands, old deciduous and coniferous-dominated stands, treed bogs, and recent clear-cuts. However, when compared to available habitat in the study area, they were more likely to contain old conifer-dominated stands and recent cutblocks. We assumed this is because all of the recent harvest occurred in old stands, habitat preferred by the owls. When compared with random sites, locations used for foraging and roosting at the home range scale were more likely to be in young deciduous-dominated stands, old conifer-dominated stands and cutblocks > 30 yr old, and less likely to occur in old deciduous-dominated stands and recent cutblocks. Hence, although recent clearcuts occurred in territories, birds avoided these microhabitats during foraging. To meet the breeding requirements of Barred Owls in managed forests, 10–20 ha patches of old deciduous and mixedwood forest containing large Populus snags or trees should be maintained. In our study area, nest trees had a minimum dbh of 34 cm. Although cut areas were incorporated into home ranges, the amount logged was low, i.e., 7%, in our area. Hence more research is required to determine harvest levels tolerated by owls over the long term.

The Short-eared Owl (Asio flammeus) is an open-country species breeding in the northern United States and Canada, and has likely experienced a long-term, range-wide, and substantial decline. However, the cause and magnitude of the decline is not well understood. We set forth to address the first two of six previously proposed conservation priorities to be addressed for this species: (1) better define habitat use and (2) improve population monitoring. We recruited 131 volunteers to survey over 6.2 million ha within the state of Idaho for Short-eared Owls during the 2015 breeding season. We surveyed 75 transects, 71 of which were surveyed twice, and detected Short-eared Owls on 27 transects. We performed multiscale occupancy modeling to identify habitat associations, and performed multiscale abundance modeling to generate a state-wide population estimate. Our results suggest that within the state of Idaho, Short-eared Owls are more often found in areas with marshland or riparian habitat or areas with greater amounts of sagebrush habitat at the 1750 ha transect scale. At the 50 ha point scale, Short-eared Owls tend to associate positively with fallow and bare dirt agricultural land and negatively with grassland. Cropland was not chosen at the broader transect scale suggesting that Short-eared Owls may prefer more heterogeneous landscapes. On the surface our results may seem contradictory to the presumed land use by a “grassland” species; however, the grasslands of the Intermountain West, consisting largely of invasive cheatgrass (Bromus tectorum), lack the complex structure shown to be preferred by these owls. We suggest the local adaptation to agriculture represents the next best habitat to their historical native habitat preferences. Regardless, we have confirmed regional differences that should be considered in conservation planning for this species. Last, our results demonstrate the feasibility, efficiency, and effectiveness of utilizing public participation in scientific research to achieve a robust sampling methodology across the broad geography of the Intermountain West.

In this work a new method for clustering and building a topographic representation of a bacteria taxonomy is presented. The method is based on the analysis of stable parts of the genome, the so-called “housekeeping genes”. The proposed method generates topographic maps of the bacteria taxonomy, where relations among different type strains can be visually inspected and verified. Two well known DNA alignement algorithms are applied to the genomic sequences. Topographic maps are optimized to represent the similarity among the sequences according to their evolutionary distances. The experimental analysis is carried out on 147 type strains of the Gammaprotebacteria class by means of the 16S rRNA housekeeping gene. Complete sequences of the gene have been retrieved from the NCBI public database. In the experimental tests the maps show clusters of homologous type strains and present some singular cases potentially due to incorrect classification or erroneous annotations in the database.

Apoptosis induced by the death-inducing ligand FasL (CD95L) is a major mechanism of cell death. Trophoblast cells express the Fas receptor yet survive in an environment that is rich in the ligand. We report that basal nitric oxide (NO) production is responsible for the resistance of trophoblasts to FasL-induced apoptosis. In this study we demonstrate that basal NO production resulted in the inhibition of receptor clustering following ligand binding. In addition NO also protected cells through the selective nitrosylation, and inhibition, of protein kinase Cepsilon (PKCepsilon) but not PKCalpha. In the absence of NO production PKCepsilon interacted with, and phosphorylated, the anti-apoptotic protein cFLIP. The interaction is predominantly with the short form of cFLIP and its phosphorylation reduces its recruitment to the death-inducing signaling complex (DISC) that is formed following binding of a death-inducing ligand to its receptor. Inhibition of cFLIP recruitment to the DISC leads to increased activation of caspase 8 and subsequently to apoptosis. Inhibition of PKCepsilon using siRNA significantly reversed the sensitivity to apoptosis induced by inhibition of NO synthesis suggesting that NO-mediated inhibition of PKCepsilon plays an important role in the regulation of Fas-induced apoptosis.

The effects on the intestinal microbiota of a short period of marginal over-eating, characteristic of holiday or festival periods, were investigated in a pilot study. Fourteen healthy male subjects consumed a diet rich in animal protein and fat for seven days. During this period, the subjects significantly increased their dietary energy, protein, carbohydrate and fat intakes by 56, 59, 53 and 58%, respectively (all P < 0.05). The mean weight gain of 0.27 kg was less than the expected 1 kg, but this was consistent with a degree of under-reporting on the baseline diet. Fluorescence in situ hybridisation analysis confirmed the relative stability of each individual’s faecal microbiota but showed considerable variations between them. The diet was associated with a significant increase in numbers of total faecal bacteria and the bacteroides group, as detected by the universal bacterial probe (DAPI) and Bacteroides probe (Bac 303), respectively. Overall, there was a decrease in numbers of the Lactobacillus/Enterococcus group (Lab 158 probe; 2.8 ± 3.0% to 1.8 ± 1.8%) and the Bifidobacterium group (Bif 164 probe; 3.0 ± 3.7% to 1.7 ± 1.2%), although there was considerable inter-individual variation. Analysis of the relative proportions of each bacterial group as a percentage of the subject’s total bacteria showed a trend for a change in the intestinal microbiota that might be considered potentially unhealthy.

The poor performance of the Stock Market in the US up to the middle of 2003 has meant that REITs are increasingly been seen as an attractive addition to the mixed-asset portfolio. However, there is little evidence to indicate the consistency of the role REITs should play a role in the mixed-asset portfolio over different investment horizons. The results highlight that REITs do play a significant role over both different time horizons and holding periods. The findings show that REITs attractiveness as a diversification asset increase as the holding period increases. In addition, their diversification qualities span the entire efficient frontier, providing return enhancement properties at the lower end, switching to risk reduction qualities at the top end of the frontier.

The recent increase in short messaging system (SMS) text messaging, often using abbreviated, non-conventional ‘textisms’ (e.g. ‘2nite’), in school-aged children has raised fears of negative consequences of such technology for literacy. The current research used a paradigm developed by Dixon and Kaminska, who showed that exposure to phonetically plausible misspellings (e.g. ‘recieve’) negatively affected subsequent spelling performance, though this was true only with adults, not children. The current research extends this work to directly investigate the effects of exposure to textisms, misspellings and correctly spelledwords on adults’ spelling. Spelling of a set of key words was assessed both before and after an exposure phase where participants read the same key words, presented either as textisms (e.g. ‘2nite’), correctly spelled (e.g. ‘tonight’) or misspelled (e.g. 'tonite’)words. Analysis showed that scores decreased from pre- to post-test following exposure to misspellings, whereas performance improved following exposure to correctly spelled words and, interestingly, to textisms. Data suggest that exposure to textisms, unlike misspellings, had a positive effect on adults’ spelling. These findings are interpreted in light of other recent research suggesting a positive relationship between texting and some literacy measures in school-aged children.

This paper examines the changes in the length of commercial property leases over the last decade and presents an analysis of the consequent investment and occupational pricing implications for commercial property investmentsIt is argued that the pricing implications of a short lease to an investor are contingent upon the expected costs of the letting termination to the investor, the probability that the letting will be terminated and the volatility of rental values.The paper examines the key factors influencing these variables and presents a framework for incorporating their effects into pricing models.Approaches to their valuation derived from option pricing are critically assessed. It is argued that such models also tend to neglect the price effects of specific risk factors such as tenant circumstances and the terms of break clause. Specific risk factors have a significant bearing on the probability of letting termination and on the level of the resultant financial losses. The merits of a simulation methododology are examined for rental and capital valuations of short leases and properties with break clauses.It is concluded that in addition to the rigour of its internal logic, the success of any methodology is predicated upon the accuracy of the inputs.The lack of reliable data on patterns in, and incidence of, lease termination and the lack of reliable time series of historic property performance limit the efficacy of financial models.
