Background The majority of introns in gene transcripts are found within the coding sequences (CDSs). A small but significant fraction of introns are also found to reside within the untranslated regions (5′UTRs and 3′UTRs) of expressed sequences. Alignment of the whole genome and expressed sequence tags (ESTs) of the model plant Arabidopsis thaliana has identified introns residing in both coding and non-coding regions of the genome. Results A bioinformatic analysis revealed some interesting observations: (1) the density of introns in 5′UTRs is similar to that in CDSs but much higher than that in 3′UTRs; (2) the 5′UTR introns are preferentially located close to the initiating ATG codon; (3) introns in the 5′UTRs are, on average, longer than introns in the CDSs and 3′UTRs; and (4) 5′UTR introns have a different nucleotide composition to that of CDs and 3′UTR introns. Furthermore, we show that the 5′UTR intron of the A. thaliana EFIα-A3 gene affects the gene expression and the size of the 5′UTR intron influences the level of gene expression. Conclusion Introns within the 5′UTR show specific features that distinguish them from introns that reside within the coding sequence and the 3′UTR. In the EFIα-A3 gene, the presence of a long intron in the 5′UTR is sufficient to enhance gene expression in plants in a size dependent manner.

This volume stems from the 1st International Conference on Epithelial-Mesenchymal Transitions (EMT), which was convened by the editors on October 5–8, 2003 in the beautiful setting of Port Douglas, Queensland, Australia. EMT, the name given to the transformation of cells arranged in a coherent layer – epithelial cells – to more individualistic and potential motile cells – mesenchymal cells – was recognized decades ago by Prof. Elisabeth (Betty) Hay (Harvard Medical School, Boston, Mass., USA) as a primary mechanism in embryogenesis for remodelling tissues. More recently EMT has been seen as crucial to the spread and invasion of carcinoma, and more recently still, EMT-like changes have been detected in various pathologies marked by fi brosis. Despite the basic and clinical importance of EMT, this extremely rapidly growing fi eld had never previously had a conference devoted to it, and indeed the disciplines of developmental biology, cancer and pathology rarely interact although they have much to share. The chapters assembled for this volume encompass these three major themes of the meeting, development, pathology and cancer, and further highlight the commonality in terms of mechanisms and outcomes among them...

Aims: Caveolin-1 (cav1) is reported to have both cell survival and pro-apoptotic characteristics. This may be explained by its localisation or phosphorylation in injured cells. This study investigated the role of cav1 in kidney cells of different nephron origin and developmental state after oxidative stress. Methods: Renal MCDK distal tubular, HK2 proximal tubular epithelial cells and HEK293T renal embryonic cells were treated with 1mM hydrogen peroxide. Apoptosis, loss of cell adhesion, and cell survival were compared with expression of cav1 in its non-phosphorylated and phosphorylated (p-cav1) forms. Cav1 was transfected into the HEK293T cells, or caveolae were disrupted with filipin or nystatin in HK2 cells, to investigate functions of cav1 and p-cav1. Results: Oxidative stress induced more apoptosis in HK2s than MDCKs (p<0.05). HK2s had lower endogenous cav1 and p-cav1 than MDCKs (p<0.05). Both cell lines had increased p-cav1, but not cav1, with oxidative stress. This increase was greatest in MDCKs (p<0.01). Cav1 was located mainly in the plasma membrane of untreated cells and translocated to the cytoplasm with oxidative stress in both cell lines, more so in MDCKs. Disruption of caveolae caused cytoplasmic translocation of cav1 in HK2s, but did not alter high levels of oxidative stress-induced apoptosis. When HEK293Ts lacking endogenous cav1 were transfected with cav1, oxidant-induced apoptosis and loss of cell adhesion was decreased (p<0.01), and p-cav1 was induced by treatment. Conclusion: Cav1 expression and localisation in kidney cells is not anti-apoptotic, but increased expression of p-cav1 may promote cell survival after oxidative stress. © 2008 Royal College of Pathologists of Australasia.

The banana industry worldwide is under threat from a fungal disease known as Fusarium wilt, a disease for which there is no chemical control. Conventional breeding approaches to generate resistant banana varieties are lengthy and very difficult. As such, genetic engineering for disease resistance is considered the most viable control option. In this PhD thesis, genetically modified banana plants were generated using several different stress tolerance genes. When challenged with Fusarium wilt in glasshouse trials, some lines showed increased resistance to the disease. The promising elite lines generated in this study will now require testing in field trials.

Many countries over the last decade, have used performance-based contracting (PBC) to manage and maintain roads. The implementation of PBC provides additional benefits for the government/public such as cost savings and improved conditions of contracted road assets. In Australia, PBC is already being implemented on all categories of roads: national, state, urban and rural. Australian PBC arrangement is designed to turn over control and responsibility for roadway system maintenance, rehabilitation, and capital improvement projects to private contractors. Contractors’ responsibilities include determination of treatment types, the design, programming and the undertaking of works needed to maintain road networks at predetermined performance levels. Indonesia initiated two PBC pilot projects in 2011, the Pantura Section Demak-Trengguli (7.68 kilometers) in Central Java Province and Section Ciasem-Pamanukan (18.5 kilometers) in West Java Province. Both sections are categorized as national roads. The contract duration for both of these projects is four years. To facilitate a possible way forward, it is proposed to conduct a study to understand Australia's experiences of advancing from pilot projects to nation-wide programs using PBC. The study focuses on the scope of contracts, bidding processes, risk allocation, and key drivers, using relevant PBC case studies from Australia. Recommendations for future PBC deployment nation-wide should be based on more research associated with risk allocation. This will include investigation of standard conditions of contract. Implications of the contract clauses for the risk management strategy to be adopted by contractors. Based on the nature of risks, some are best managed by the project owner. It is very important that all parties involved to be open to the new rules of contract and to convince themselves about the potential increased benefits of the use of PBC. The most recent states of challenging issues were explored and described.

Background The most common pathway to development of diabetes foot ulcers is repetitive daily activity stress on the plantar surface of the neuropathic foot. Studies suggest an association between different diabetic foot complications and physical activity. However, to the best of the authors knowledge the steps/day and sleep patterns of people with diabetic foot ulcers has yet to be investigated. This observational study aims to investigate the physical activity and sleep patterns of three groups of adults with type 2 diabetes and different foot complications Methods Participants with type 2 diabetes were recruited into three groups: 1. those with no reported foot complications (DNIL), 2. those with diagnosis of neuropathy (DPN) and 3. those with a neuropathic ulcer (DFU). Exclusion criteria included peripheral arterial disease and mobility aid use. Participants wore a SenseWear Pro 3 Armband continuously for 7 days and completed an Epworth Sleepiness Scale. The Armband is a validated automated measure of activity (walking steps, average Metabolic Equivalent Task (MET), physical activity (>3 METs) duration), energy expenditure(kJ) (total and physical activity (>3 METs)) and sleep (duration). Data on age, sex, BMI, diabetes duration and HbA1c were also collected. Results Sixty-Six (14 DNIL, 22 DPN and 30 DFU's participants were recruited; 71% males, mean age 61(±12) years, diabetes duration 13(±9) years, HbA1c 8.3(±2.8), BMI 32.6(±5.9), average METs 1.2(0.2). Significant differences were reported in mean(SD) steps/day (5,859(±2,381) in DNIL; 5,007(±3,349) in DPN and 3,271(±2,417) in DFU's and daily energy expenditure (10,868(±1,307)kJ in DNIL; 11,060(±1,916)kJ in DPN and 13,006(± 3,559) in DFU's(p <0.05). No significant differences were reported for average METs, physical activity duration or energy expenditure, sleep time or Epworth score (p>0.1). Conclusions Preliminary findings suggest people with diabetes are sedentary. Results indicate that patients with a diabetic foot ulcer work significantly less than those with neuropathy or nil complications and use significantly more energy to do so. Sleep Parameters showed no differences. Recruitment is still on going.

Tutkielma käsittelee naisleskien ja orpojen elämää luo- ja kisii-yhteisöissä Länsi-Kenian maaseudulla. Tarkoituksena on selvittää, kuinka yhteiskunnassa tapahtuvat taloudelliset ja sosiaaliset muutokset ovat vaikuttaneet orpojen ja leskien asemaan patriarkaalisissa yhteisöissä. Tutkielma pyrkii osoittamaan paikallistason muutosten kulttuurisidonnaisuuden erilaisia muutosteemoja tarkastelemalla. Erityisesti on tarkasteltu aids-epidemian ja maapulan vaikutuksia leskien ja orpojen lähisuhteisiin, maankäyttöoikeuksiin ja luojen leviraattisuhteeseen. Tutkielman aineisto koostuu kahden kuukauden kenttätyöjakson aikana tehdyistä haastatteluista ja osallistuvasta havainnoinnista. Kenttätyö on tehty Suomen Luterilaisen Evankeliumiyhdistyksen ja Kenian Evangelic Lutheran Churchin orpotyöprojektin yhteydessä syksyllä 2007. Aids-epidemian myötä huollettavien määrä maaseutuyhteisöissä on kasvanut. Samanaikaisesti on havaittu, että sukulaisten muodostama turvaverkko on heikentynyt. Turvaverkon heikentyminen liittyy resurssipulaan ja arvojen muutokseen. Lesket ja orvot ovat vaarassa jäädä ilman tarvitsemaansa tukea ja apua. Kisiiden ja luojen sukulaisuusjärjestelmässä naisilla ei ole siskon tai tyttären rooliin liittyviä oikeuksia. Tutkielmasta käy ilmi, että leskien ja orpojen häätäminen kotitiloilta on yleistä, vaikka perinteisesti heidän asemansa maankäyttäjänä on ollut turvattu. Luo-leski saatetaan häätää myös hänen kieltäytyessään leviraattisuhteesta. Häätöjen keskeinen motiivi on saada lesken käytössä ollut maapala miesvainajan sukulaisten haltuun. Häädetyt lesket rinnastuvat Thomas Håkanssonin tutkimiin avoliitosta häädettyihin kisii-naisiin, joita hän nimittää suvuttomiksi naisiksi. Pekka Seppälän tutkimusta luhyoiden sukupolvenvaihdoksista käytetään maankäyttöoikeuksiin liittyvän analyysin pohjana. Luojen leviraattisuhteessa on tapahtunut merkittäviä muutoksia aids-epidemian aikana. Länsi-Kenian aids-tilanne on muuta maata huonompi. Alueen asukkailla on valistuskampanjoiden ansiosta hyvät tiedot sairaudesta, mutta he toimivat usein tämän tiedon vastaisesti. Tutkielman mukaan keskeisin syy näennäiselle välinpitämättömyydelle on leimautumisen pelko. Aidsiin liittyvä stigma on voimakasta ja sen vuoksi tauti pyritään salaamaan mahdollisimman pitkään. Sukulaismiehet kieltäytyvät usein leskenperijän roolista hiv-tartunnan pelossa. Tämä on luonut kysyntää ammattiperijöille, jotka suorittavat seksuaalisia rituaaleja leskille korvausta vastaan. Tutkielmassa esitellään rituaalisen seksin merkitystä luoille ja todetaan sen olevan tärkein syy leviraattikäytännön jatkumiselle. Luo-leskien perinteistä asemaa tarkastellaan Betty Potashin tutkimusten valossa.

The aim of this paper is to analyze how active R&D policies affect the growth rate of an economy with endogenous growth and non-renewable resources. We know from Scholz and Ziemens (1999) and Groth (2006) that in infinitely lived agents (ILA) economies, any active R&D policy increases the growth rate of the economy. To see if this result also appears in economies with finite lifetime agents, we developed an endogenous growth overlapping generations (OLG) economy à la Diamond which uses non-renewable resources as essential inputs in final good’s production. We show analytically that any R&D policy that reduces the use of natural resources implies a raise in the growth rate of the economy. Numerically we show that in economies with low intertemporal elasticity of substitution (IES), active R&D policies lead the economy to increase the depletion of non-renewable resources. Nevertheless, we find that active R&D policies always imply increases in the endogenous growth rate, in both scenarios. Furthermore, when the IES coefficient is lower (greater) than one, active R&D policies affect the growth rate of the economy in the ILA more (less) than in OLG economies.

A presente dissertação de mestrado pretende abordar as vicissitudes do discurso do capitalista sobre o sujeito. Para tanto, toma como ponto de partida a afirmação de Lacan, em suas palestras em Saint-Anne sobre O saber do psicanalista, de que o que caracteriza o discurso do capitalista é a foraclusão da castração. A partir dessa colocação representada no matema do discurso do capitalista pelo desaparecimento da disjunção entre produto e verdade, bem como pela ausência de um vetor entre agente e Outro, o que demonstra que esse discurso não promove laço social , percorre-se algumas questões que reverberam sobre o sujeito, dentre as quais se destaca: se o sujeito não existe fora do laço social, que sorte de efeitos a ele resulta por freqüentar um discurso que se apresenta como dominante na contemporaneidade em que a castração está foracluída e, portanto, também o liame social? Tendo em vista que a Verwerfung é o que opera no discurso do capitalista, lança-se a hipótese de que a esquizofrenização no nível do discurso resulte na propagação do que Helene Deutsch convencionou chamar de como se. Esse fenômeno surgiria como uma suplência imaginária a que recorreria o sujeito como estabilização face aos efeitos decorrentes da foraclusão da castração no discurso do capitalista. Trata-se de um discurso psicotizante que, ao contrário dos quatro discursos apresentados por Lacan no seminário O avesso da psicanálise, não promove laço social. Se discurso é o que faz laço social e Freud, em 1930, anunciara que a principal fonte de sofrimento de que padece o homem é a relação com os demais, o mal-estar na civilização é o mal-estar dos laços sociais. Logo, os quatro discursos participam do mal-estar na civilização. Ao contrário destes, o discurso do capitalista, na tentativa de eliminar o mal-estar, foraclui o laço social. Por promover o gozo ao invés da renúncia à pulsão, o discurso do capitalista acaba por instigá-la. E, como toda pulsão é pulsão de morte, não é outra coisa senão o empuxo mortífero ao gozo que o discurso do capitalismo produz. A potência que o discurso do capitalista adquiriu na contemporaneidade é analisada a partir de sua íntima relação com o discurso da ciência. A partir de algumas colocações de Hannah Arendt acerca da banalidade do mal presente em um sistema totalitário como o nazista, cogita-se a hipótese de que a aliança entre os discursos do capitalista e da ciência resulte no surgimento de uma nova forma de totalitarismo: o totalitarismo de consumo.
