We sequenced the small subunit (SSU) rRNA and glycosomal glyceraldehyde-3-phosphate dehydrogenase (gGAPDH) genes of two trypanosomes isolated from the Brazilian snakes Pseudoboa nigra and Crotalus durissus terrificus. Trypanosomes were cultured and their morphometrical and ultrastructural features were characterized by light microscopy and scanning and transmission electron microscopy. Phylogenetic trees inferred using independent or combined SSU rRNA and gGAPDH data sets always clustered the snake trypanosomes together in a clade closest to lizard trypanosomes, forming a strongly supported monophyletic assemblage (i.e. lizard-snake clade). The positioning in the phylogenetic trees and the barcoding based on the variable V7-V8 region of the SSU rRNA, which showed high sequence divergences, allowed us to classify the isolates from distinct snake species as separate species. The isolate from P. nigra is described as a new species, Trypanosoma serpentis n. sp., whereas the isolate from C. d. terrificus is redescribed here as Trypanosoma cascavelli.

Fluorochrome-labelled cells of two field isolates and Mycoplasma synoviae (Ms) were inoculated onto monolayer cultures of fluorochrome-labelled HEp-2 cells and monitored by confocal laser scanning microscopy (CLSM). Ms was detected initially adhered to and subsequently inside the host cells. Between 24 and 48 h of infection, Ms was detected in the perinuclear region, and after 72 h of infection was confirmed by gentamicin invasion assay. High and low passage Ms strains showed no differences in adherence or invasion. The morphology and the actin filaments of the infected HEp-2 cells were preserved throughout the study period. The observed invasion by Ms is consistent with the biology of Mollicutes, and could explain the difficulties in recovering field isolates of the mycoplasma and in controlling the infection in birds even after long-term antibiotic treatment. The thermal behavior of PANI nanofibers doped with beta-naphthalenesulfonic acid (beta-NSA) was investigated and their morphological and structural changes after heating were monitored by SEM, XRD and Raman techniques, respectively. By using electron-scanning microscopy it is possible to verify that the nanofiber morphology is stable and no polymer degradation is observed in thermogravimetric (TG) data up to 200 degrees C. Nevertheless, the heating promotes the formation of cross-linking structures (phenazine and/or oxazine-like rings), that is clearly demonstrated by the presence of bands at ca. 578, 1398, and 1644 cm(-1) in resonance Raman spectra of heated PANI-NSA samples. The most important consequence of the formation of cross-linking structures in PANI-NSA samples is that these samples retain their nanofiber morphology upon HCl doping in contrast to PANI-NSA nanofibers without heating. In the present study, porous nickel foams with three different porosities (i.e. 50 %, 60 % and 70 %) were fabricated using the space-holding sintering method. Ammonium bicarbonate particles with sizes ranging from 1- 2 mm were chosen as the space-holding material. The anisotropic behaviours of the nickel foam samples were investigated by compressive testing loading at different directions, i.e., in both directions of the major and minor axis of ellipsoidal cells. Electron scanning microscopy (SEM) and Image-Pro Plus was used to characterise the morphological characteristics of the porous nickel foam samples. Results indicated that the porous nickel foam samples exhibited obvious anisotropic mechanical properties. The foam sample shows significantly higher nominal stress for loading in the direction of the major axis of the pores than loading in the direction of the minor axis of pores. The nominal stress increases with the decreasing of the porosity.

Titanium and some of its alloys are well accepted as load-bearing implant materials due to their excellent mechanical properties, superior corrosion resistance, and outstanding biocompatibility. However, solid implant materials may suffer from the problems of adverse tissue reaction, biomechanical mismatch and lack of new bone tissue ingrowth ability. In the present study, porous titanium-molybdenum (Ti-Mo) alloy was fabricated by the space-holding sintering method. The pore size, pore shape and porosity can be controlled through choosing appropriate space-holding particle materials. Electron scanning microscopy (SEM) was used for the characterisation of the porous Ti-Mo alloy. The mechanical properties of the porous Ti-Mo alloy samples were investigated by compressive tests. Results indicated that the porous Ti-Mo alloy provides promising potential for new implant materials with new bone tissue ingrowth ability and mechanical properties mimicking those of natural bone.

In the present study, porous nickel foam samples with pore sizes of 20 μm and 150 μm and porosities of 60 % and 70 % were fabricated by the space-holding sintering method via powder metallurgy. Electron scanning microscopy (SEM) and Image-Pro Plus were used to characterise the morphological features of the porous nickel foam samples. The anisotropic mechanical properties of porous nickel foams were investigated by compressive testing loading in different directions, i.e. the major pore axis and minor pore axis. Results indicated that the nominal stress of the nickel foam samples increases with the decreasing of the porosity. Moreover, the foam sample exhibited significantly higher nominal stress for loading in the direction of the major pore axis than loading in direction of the minor pore axis. It is also noticeable that the nominal stress of the nickel foams increases with the decreasing of the pore size. It seems that the deformation behaviour of the foams with a pore size in the micron-order differs from those with a macro-porous structure.

Membrane is usually subject to fouling by various organic foulants, such as yeast, protein and sodium alginate during filtration. Backwashing is a common practice to reduce membrane fouling. It is essential to evaluate the effects of backwashing on fouling in order to optimize operational parameters. In this experiment, poly(vinylidene fluoride) (PVDF) membranes were used to filter organic foulants from suspensions in a dead-end stirred cell. Three types of organic foulants including yeast, protein and sodium alginate which were stained with fluorescent dyes before filtration were used with different combinations in the experiments. After filtration, the PVDF membrane was backwashed.

Consequently, a stack of images, instrumental data and sample data were captured from the fouling layers on the PVDF membrane surface using confocal laser scanning microscope (CLSM) and its associated image acquisition software LAS AF. Then, the quality of the images was enhanced for better visualization and a set of quantitative fouling data were derived by using the software code developed by the project team at Deakin University.

This collection contains raw image data of poly(vinylidene fluoride) (PVDF) membrane’s fouling layer when three types of organic foulants present, which are captured by confocal laser scanning microscopy (CLSM) and its software, and the instrumental and sample metadata, the processed image data and the geometrical structure properties of the fouling layer. By comparing with the same membrane without backwashing, the efficiency of backwashing was computed.

This data collection would be useful to evaluate the backwashing efficiency of PVDF membrane in order to optimize frequency and operational conditions of backwashing by membrane materials researchers and water researchers.

The physicochemical and rheological properties of yoghurt made from unstandardised unhomogenised buffalo milk were investigated during fermentation and 28 days of storage and compared to the properties of yoghurt made from homogenised fortified bovine milk. A number of differences observed in the gel network can be linked to differences in milk composition. The microstructure of buffalo yoghurt, as assessed by confocal laser scanning microscopy (CLSM) and cryo scanning electron microscopy (cryo-SEM), was interrupted by large fat globules and featured more serum pores. These fat globules have a lower surface area and bind less protein than the homogenised fat globules in bovine milk. These microstructural differences likely lead to the higher syneresis observed for buffalo yoghurt with an increase from 17.4 % (w/w) to 19.7 % (w/w) in the weight of whey generated at days 1 and 28 of the storage. The higher concentration of total calcium in buffalo milk resulted in the release of more ionic calcium during fermentation. Gelation was also slower but the strength of the two gels was similar due to similar protein and total solids concentrations. Buffalo yoghurt was more viscous, less able to recover from deformation and less Newtonian than bovine yoghurt with a thixotropy of 3,035 Pa.s-1 measured for buffalo yoghurt at the end of the storage, at least four times higher than the thixotropy of bovine yoghurt. While the titratable acidity, lactose consumption and changes in organic acid concentrations were similar, differences were recorded in the viability of probiotic bacteria with a lower viability of Lactobacillus acidophilus of 5.17 log (CFU/g) recorded for buffalo yoghurt at day 28 of the storage. Our results show that factors other than the total solids content and protein concentration of milk affect the structural properties of yoghurt. They also illustrate the physicochemical reasons why buffalo and bovine yoghurt are reported to have different sensory properties and provide insight into how compositional changes can be used to alter the microstructure and properties of dairy products. © 2013 Springer Science+Business Media New York.

De-alloying of S-phase in AA2024-T3 in the presence chlorides, is well-known. However, it is unclear how rare earth mercaptoacetate inhibitors affect this process when immersed in a 0.1. M NaCl solution. This paper analyses data obtained using EPMA on AA2024-T3 surfaces before and after a 16. min immersion period. Cerium and praseodymium mercaptoacetate inhibited the de-alloying process of S-phase particles. Although no significant change in composition was observed for cathodic intermetallics, each appeared to participate in local corrosion reactions as evidenced by the development of surface oxides. Clustering between S-phase and one of the Cu-containing intermetallic domains was also evident.

AA2024-Tx is one of the most common high-strength aluminium alloys used in the aerospace industry. This article reviews current understanding of the microstructure of sheet AA2024-T3 and chronicles the emergence of new compositions for constituent particles as well as reviews older literature to understand the source of the original compositions. The review goes on to summarise older and more recent studies on corrosion of AA2024-T3, drawing attention to areas of corrosion initiation and propagation. It pays particular attention to modern approaches to corrosion characterisation as obtained through microelectrochemical techniques and physicochemical characterisation, which provide statistical assessment of factors that contribute to corrosion of AA2024. These approaches are also relevant to other alloys.

The cobalt-chromium alloy is extensively used in the Odontology for the confection of metallic scaffolding in partial removable denture. During the last few years, it has been reported an increasing number of premature imperfections, with a few months of prosthesis use. The manufacture of these components is made in prosthetic laboratories and normally involves recasting, using parts of casting alloy and parts of virgin alloy. Therefore, the objective of the present study was to analyze the mechanical properties of a commercial cobalt-chromium alloy of odontological use after successive recasting, searching information to guide the dental prosthesis laboratories in the correct manipulation of the cobalt-chromium alloy in the process of casting and the possible limits of recasting in the mechanical properties of this material. Seven sample groups were confectioned, each one containing five test bodies, divided in the following way: G1: casting only with virgin alloy; G2: casting with 50% of the alloy of the G1 + 50% of virgin alloy; G3: casting with 50% of the alloy of the G2 + 50% of virgin alloy; G4: casting with 50% of the alloy of the G3 + 50% of virgin alloy; G5: 50% of alloy of the G4 + 50% of virgin alloy; G6: 50% of alloy of the G5 + 50% of virgin alloy and finally the G7, only with recasting alloy. The modifications in the mechanical behavior of the alloy were evaluated. Moreover, it was carried the micro structural characterization of the material by optic and electronic scanning microscopy, and X ray diffraction.and fluorescence looking into the correlatation of the mechanical alterations with structural modifications of the material caused by successive recasting process. Generally the results showed alterations in the fracture energy of the alloy after successive recasting, resulting mainly of the increasing presence of pores and large voids, characteristic of the casting material. Thus, the interpretation of the results showed that the material did not reveal significant differences with respect to the tensile strength or elastic limit, as a function of successive recasting. The elastic modulus increased from the third recasting cycle on, indicating that the material can be recast only twice. The fracture energy of the material decreased, as the number of recasting cycles increased. With respect to the microhardness, the statistical analyses showedno significant differences. Electronic scanning microscopy revealed the presence of imperfections and defects, resulting of the recasting process. X ray diffraction and fluorescence did not show alterations in the composition of the alloy or the formation of crystalline phases between the analyzed groups. The optical micrographs showed an increasing number of voids and porosity as the material was recast. Therefore, the general conclusion of this study is that the successive recasting of of Co-Cr alloys affects the mechanical properties of the material, consequently leading to the failure of the prosthetic work. Based on the results, the best recommendadition is that the use of the material should be limited to two recasting cycles

O objetivo do presente trabalho foi o de estudar a resistência à corrosão em placas de aço inoxidável 316L, com diferentes tipos de acabamento e tratamento superficial, e a possível interferência dessa reação corrosiva na consolidação óssea. Utilizaram-se placas semi-acabadas, polidas, tratadas com jatos de microesferas de vidro e passivadas, as quais foram aplicadas na epífise distal do rádio de cães. Foram utilizados 12 animais, divididos em dois grupos, nos quais, após osteotomia bilateral do rádio e ulna, foram realizadas osteossínteses do rádio, totalizando 24 procedimentos. Avaliou-se a evolução clínica e radiográfica das regiões que receberam os implantes aos 30, 60, 90, 180, 240 e 360 dias. Os animais do grupo 1 (GI) foram sacrificados aos 180 dias e os do GII aos 360 dias para estudo histológico e de microscopia eletrônica de varredura do local da osteotomia sob a região dos implantes metálicos e para estudo da resistência à corrosão no organismo, pelos implantes metálicos, por meio de análises química e metalográfica (microscopia óptica e eletrônica de varredura e espectroscopia de espalhamento de energia por raios X). Os animais recuperaram a função dos membros operados 24 horas após a cirurgia. Radiograficamente, verificou-se a consolidação óssea em todos os animais. Macro e microscopicamente não foram observados sinais de corrosão nos implantes metálicos, exceto em uma placa passivada, aplicada no rádio esquerdo de um animal, na qual a corrosão foi detectada pela microscopia óptica e eletrônica de varredura. Este estudo permite concluir que as placas de aço inoxidável 316L, independente do acabamento superficial a que foram submetidas, não sofreram corrosão ou reações adversas e foram efetivas no tratamento das fraturas experimentais do rádio e ulna de cães.

O presente trabalho teve como objetivo verificar a forma de penetração do fungo Metarhizium anisopliae [METSCH. (SOROKIN, 1883)] em carrapatos da espécie Rhipicephalus sanguineus (LATREILLE, 1806), assim como as lesões infringidas nos tecidos internos do ácaro. A forma de aderência e penetração do fungo foi estudada através da microscopia eletrônica de varredura e a ação do fungo nos tecidos internos avaliada em secções histológicas convencionais. Para observação destes eventos, realizaram-se infecções experimentais em 11 grupos de fêmeas ingurgitadas do carrapato R. sanguineus contendo 12 fêmeas ingurgitadas cada. Para tal, as fêmeas ingurgitadas foram banhadas durante 3 minutos, sob agitação manual, em suspensão com concentração 108 conídios/mL. No caso dos grupos controle o banho foi realizado apenas no veículo da suspensão. Os carrapatos foram processados para histopatologia e microscopia eletrônica em diversos tempos após a infecção, a saber: 1 e 18h, e um, dois, três, quatro, cinco, seis, sete, nove e onze dias. Observou-se que a maior parte dos conídios germinou em até 18h após a inoculação e que o fungo penetrou no ácaro através do tegumento 48h após a infecção. Após a penetração, o fungo invadiu o corpo do hospedeiro promovendo uma colonização difusa, sem preferência aparente por tecidos específicos. Dentre as lesões nos tecidos internos do ácaro, ressalta-se o rompimento da parede intestinal e vazamento do conteúdo para a hemocele. A morte do hospedeiro ocorreu entre 96 e 120h pós-infecção, e a esporulação do patógeno sobre o cadáver do ácaro iniciou-se em torno de 120 a 144h pós-infecção. Espera-se, com este trabalho, contribuir para o desenvolvimento e viabilização de técnicas de controle biológico dos carrapatos por fungos como alternativa ao uso de acaricidas.
