O sismo é uma das catástrofes naturais mais prejudiciais e fatais para o ser humano. Nos últimos anos, este fenómeno natural foi responsável pela morte de milhares de pessoas e pelo colapso de incontáveis edifícios e obras de arte. É prática corrente dos projetos de estruturas porticadas de betão armado desprezar a contribuição dos painéis de alvenaria de enchimento no comportamento e resposta sísmica destas estruturas, assumindo que se está do lado da segurança. Contudo, apesar deste procedimento ser conservativo para o caso das ações verticais, tal não se pode afirmar no caso de ações horizontais, das quais faz parte a ação sísmica. Assim, o presente trabalho tem como objetivo principal analisar numericamente a influência dos painéis de alvenaria de enchimento e das paredes resistentes em edifícios de betão armado, sujeitos à ação sísmica. Sismos recentes têm mostrado inúmeras fragilidades dos edifícios de betão armado. As principais causas são as pormenorizações de armaduras inadequadas, deficiente confinamento do betão, reduzida capacidade resistente das seções, e principalmente as irregularidades de rigidez e massa em altura e em planta. A incorreta construção inerente às paredes de alvenaria é responsável por estas mesmas irregularidades que têm provocado inúmeros danos nas construções, dos quais se destacam os mecanismos de soft-storey. Neste trabalho foram selecionados três edifícios casos de estudo com o intuito de dar resposta ao objetivo deste trabalho. Estes edifícios, alvo de estudos anteriores do LNEC, foram transformados em nove modelos diferentes cada, de forma a ser avaliada a influência das paredes de alvenaria e também do sistema estrutural. Foi usada a ferramenta de cálculo SeismoStruct, tendo sido efetuadas modelações numéricas de forma a realizar várias análises não lineares estáticas e dinâmicas. No final são apresentados e analisados os resultados decorrentes das análises.

Através do processamento de dados sísmicos convertem-se registos de campo em secções sísmicas com significado geológico, que revelam informações e ajudam a delinear as camadas geológicas do subsolo e identificar estruturas soterradas. Portanto, a interpretação dos dados sísmicos só é boa se o processamento também o for. Este trabalho é resultado de um estágio curricular na empresa de prospecção geofísica GeoSurveys, que consistiu principalmente em processar 18 linhas de dados de sísmica de reflexão multicanal de alta resolução adquiridas na ilha de Pulau Tekong em Singapura, que têm como finalidade investigação do solo da baia desta mesma ilha. Estes dados foram cedidos à GeoSurveys para fins académicos, caso em que se inclui esta dissertação. Para atingir os objectivos propostos que consistiam em avaliar o impacto das condições de operação na qualidade do sinal sísmico e interpretação das linhas, fez-se o processamento das linhas utilizando um fluxo processamento padrão utilizado na empresa, com recurso ao software Radex Pro. Este fluxo de processamento tem como mais-valia o método de correcções estáticas, o UHRS trim statics, além das técnicas habituais utilizadas para melhorar a resolução das secções sísmicas como é o caso da desconvolução, a atenuação de ruído através do stacking, correcções de NMO, e migração, entre outras técnicas. A interpretação das linhas sísmicas processadas foi feita no software Kingdom Suite (IHS), através da distinção da configuração interna dos reflectores em cada secção sísmica, estabelecendo deste modo as principais unidades sismo-estratigráficas e identificando as zonas de interface que delimitam os horizontes principais. Foi feito ainda um estudo geológico sumário da área de pesquisa e da evolução geodinâmica da região.

Los usuarios reales y potenciales del sistemas de salud en Colombia, encuentran en el camino de acceso a los servicios múltiples barreras, debido a las diferentes carencias que éste sistema presenta. Para apoyar las necesidades del usuario en éste tema de acceso, se constituyó en el año 2006 la Fundación RASA, quien ofrece de manera gratuita, a la comunidad en general, mecanismos de exigibilidad, acceso y protección de sus derechos en temas de salud. Este trabajo de grado, pretende dar un valor agregado diseñando una herramienta modular que facilite la implementación del banco de proyectos, con el fin de mejorar sus procesos y sea referencia para otras entidades dedicadas a velar por el bienestar de los individuos, en virtud de su objeto social orientado a la protección de derechos en temas de salud, además se realiza con el fin de Optar por el Título de Especialista en Alta Gerencia de la Universidad de Medellín. Con el fin de entender las necesidades de La Fundación RASA, será primordial conocer y tener un manejo adecuado de la teoría de la organización; teniendo clara su vertiente descriptiva que sugiere lo que se debe hacer para mejorar varios aspectos de la empresa, en éste caso la necesidad de diseñar un Banco de Proyectos. El trabajo se ha estructurado de la siguiente manera: Referente Teórico, Contextualización De La Fundación Rasa, Diseño De Una Herramienta Modular (Introducción, Objetivos, Recursos, Metodología y procedimientos, Evaluación y Monitoreo, Responsable de proceso, Presupuesto Estructural), Conclusiones y Recomendaciones.

En el presente documento se diseña e implementa un controlador de carga de baterías a pulsos, el cual está basado en el circuito integrado BQ24650 de National Instruments, para mejorar el sistema de alimentación de las estaciones sísmicas de la Red Sísmica del Austro. Además, se ha desarrollado un sistema de monitoreo de los niveles de tensión del panel solar y la batería. Esta información será transmitida a través de un radio enlace analógico existente en la RSA, usando el ancho de banda no ocupado del sistema analógico de transmisión de información sísmica, sin alterar, perturbar o añadir ruido al sistema. Posteriormente esta información será visualizada y almacenada en una base de datos en las oficinas de la RSA en la ciudad de Cuenca.

El sismo de magnitud 5.3 grados en la escala de Ritcher se convierte en el más importante después del terremoto de abril y julio de 1983, dado que es el primero que se registra en la recién instalada Red Sismográfica Nacional, operada  por el OVSICORI-UNA.   Este documento realiza una interpretación preliminar sobre las características epicentrales e ipocentrales del sismo.  Así como el estudio de la intensidad del evento principal. Se presenta el mapa de intensidades según la escala de Mercalli, combinando la información del mapa con una gira efectuada a la zona de mayores intensidades.  En la gira a la zona sur del país se utilizó un cuestionario de intensidades suministrados por los reporteros sísmicos de OVSICORI-UNA, como parte del proyecto de investigación “Sistematización de reportes de intensidad sísmica (UNA-CONICIT)”.  Durante el trabajo de campo se utilizó una encuesta para determinar la intensidad máxima alcanzada por el sismo en los lugares visitados.

Urban centres base their resilience on the ability to evolve and adapt as needed throughout their life. Although constantly developing, changing and subsuming nature for its needs, the current age of environmental awareness requires that cities progress in a more conscious and considered way. While they have become the dominant form of human habitation, there now exists a need to integrate 'green' solutions into urban centres to address social, physical and environmental wellbeing. The means of implementing the vast array of possible solutions without negative impacts is not clear; cities are complex systems, layering meaning, history and cultural memory ‐ they are a manifestation of shared cultural values, and as such, they do not allow a tabula rasa approach of 'blanket' solutions. All around us, cities are continuing to develop and change, and although their form is varied ‐ sprawling cities with density and sustainability problems; or collapsing cities with 'dead' centres and dilapidated districts – a common issue is the resilience of the local identity. The strength or resilience of cities lies in the elements which have become fixed points in the urban structure, giving character and identity to a shared urban experience. These elements need to be identified and either maintained or revitalised. Similarly, the identification of urban elements which can most viably be modified without compromising character and identity of place, will assist in making concrete contributions to increasing both the sustainability and experience of cities, making them more resilient. Through an examination of case studies, this paper suggests a framework to inform urban renewal assessing the widespread elements which generate an urban identity, beyond the traditional approach of heritage conservation for cultural or tourist purposes. The rapid contemporary alteration of urban structures requires an innovative methodology which satisfies on one side the need of new sustainable performances and, on the other, the resilience of the local character.

Exogenous prostacyclin is effective in reducing pulmonary vascular resistance in some forms of human pulmonary hypertension (PH). To explore whether endogenous prostaglandins played a similar role in pulmonary hypertension, we examined the effect of deleting cyclooxygenase (COX)-gene isoforms in a chronic hypoxia model of PH. Pulmonary hypertension, examined by direct measurement of right ventricular end systolic pressure (RVESP), right ventricular hypertrophy (n = 8), and hematocrit (n = 3), was induced by 3 weeks of hypobarichypoxia in wild-type and COX-knockout (KO) mice. RVESP was increased in wild-type hypoxic mice compared with normoxic controls (24.4 ± 1.4 versus 13.8 ± 1.9 mm Hg; n = 8; p < 0.05). COX-2 KO mice showed a greater increase in RVESP following hypoxia (36.8 ± 2.7 mm Hg; p < 0.05). Urinary thromboxane (TX)B2 excretion increased following hypoxia (44.6 ± 11.1 versus 14.7 ± 1.8 ng/ml; n = 6; p < 0.05), an effect that was exacerbated by COX-2 gene disruption (54.5 ± 10.8 ng/ml; n = 6). In contrast, the increase in 6-keto-prostacyclin1α excretion following hypoxia was reduced by COX-2 gene disruption (29 ± 3 versus 52 ± 4.6 ng/ml; p < 0.01). Tail cut bleed times were lower following hypoxia, and there was evidence of intravascular thrombosis in lung vessels that was exacerbated by disruption of COX-2 and reduced by deletion of COX-1. The TXA2/endoperoxide receptor antagonist ifetroban (50 mg/kg/day) offset the effect of deleting the COX-2 gene, attenuating the hypoxia-induced rise in RVESP and intravascular thrombosis. COX-2 gene deletion exacerbates pulmonary hypertension, enhances sensitivity to TXA2, and induces intravascular thrombosis in response to hypoxia. The concept of dispositional resistance to change has been introduced in a series of exploratory and confirmatory analyses through which the validity of the Resistance to Change (RTC) Scale has been established (S. Oreg, 2003). However, the vast majority of participants with whom the scale was validated were from the United States. The purpose of the present work was to examine the meaningfulness of the construct and the validity of the scale across nations. Measurement equivalence analyses of data from 17 countries, representing 13 languages and 4 continents, confirmed the cross-national validity of the scale. Equivalent patterns of relationships between personal values and RTC across samples extend the nomological net of the construct and provide further evidence that dispositional resistance to change holds equivalent meanings across nations.

The particles of Potato virus A (PVA; genus Potyvirus) are helically constructed filaments that contain multiple copies of a single type of coat-protein (CP) subunit and a single copy of genome-linked protein (VPg), attached to one end of the virion. Examination of negatively-stained virions by electron microscopy revealed flexuous, rod-shaped particles with no obvious terminal structures. It is known that particles of several filamentous plant viruses incorporate additional minor protein components, forming stable complexes that mediate particle disassembly, movement or transmission by insect vectors. The first objective of this work was to study the interaction of PVA movement-associated proteins with virus particles and how these interactions contribute to the morphology and function of the virus particles. Purified particles of PVA were examined by atomic force microscopy (AFM) and immuno-gold electron microscopy. A protrusion was found at one end of some of the potyvirus particles, associated with the 5' end of the viral RNA. The tip contained two virus-encoded proteins, the genome-linked protein (VPg) and the helper-component proteinase (HC-Pro). Both are required for cell-to-cell movement of the virus. Biochemical and electron microscopy studies of purified PVA samples also revealed the presence of another protein required for cell-to-cell movement the cylindrical inclusion protein (CI), which is also an RNA helicase/ATPase. Centrifugation through a 5-40% sucrose gradient separated virus particles with no detectable CI to a fraction that remained in the gradient, from the CI-associated particles that went to the pellet. Both types of particles were infectious. AFM and translation experiments demonstrated that when the viral CI was not present in the sample, PVA virions had a beads-on-a-string phenotype, and RNA within the virus particles was more accessible to translation. The second objective of this work was to study phosphorylation of PVA movement-associated and structural proteins (CP and VPg) in vitro and, if possible, in vivo. PVA virion structural protein CP is necessary for virus cell-to-cell movement. The tobacco protein kinase CK2 was identified as a kinase phosphorylating PVA CP. A major site of CK2 phosphorylation in PVA CP was identified as a single threonine within a CK2 consensus sequence. Amino acid substitutions affecting the CK2 consensus sequence in CP resulted in viruses that were defective in cell-to-cell and long-distance movement. The CK2 regulation of virion assembly and cell-to-cell movement by phosphorylation of CP was possibly due to the inhibition of CP binding to viral RNA. Four putative phosphorylation sites were identified from an in vitro phosphorylated recombinant VPg. All four were mutated and the spread of mutant viruses in two different host plants was studied. Two putative phosphorylation site mutants (Thr45 and Thr49) had phenotypes identical to that of a wild type (WT) virus infection in both Nicotiana benthamiana and N. tabacum plants. The other two mutant viruses (Thr132/Ser133 and Thr168) showed different phenotypes with increased or decreased accumulation rates, respectively, in inoculated and the first two systemically infected leaves of N. benthamiana. The same mutants were occasionally restricted to single cells in N. tabacum plants, suggesting the importance of these amino acids in the PVA infection cycle in N. tabacum.

The physical properties of surface soil horizons, essentially pore size, shape, continuity and affinity for water, regulate water entry into the soil. These properties are prone to changes caused by natural forces and human activity. The hydraulic properties of the surface soil greatly impact the generation of surface runoff and accompanied erosion, the major concern of agricultural water protection. The general target of this thesis was to improve our understanding of the structural and hydraulic properties of boreal clay soils. Physical properties of a clayey surface soil (0 - 10 cm, clay content 51%), with a micaceous/illitic mineralogy subjected to three different management practices of perennial vegetation, were studied. The study sites were vegetated buffer zones located side by side in SW Finland: 1) natural vegetation with no management, 2) harvested once a year, and 3) grazed by cattle. The soil structure, hydraulic properties, shrinkage properties and soil water repellency were determined at all sites. Two distinct flow domains were evident. The surface soil was characterized by subangular blocky, angular blocky and platy aggregates. Hence, large, partially accommodated, irregular elongated pores dominated the macropore domain at all sites. The intra-aggregate pore system was mostly comprised of pores smaller than 30 μm, which are responsible for water storage. Macropores at the grazed site, compacted by hoof pressure, were horizontally oriented and pore connectivity was poorest, which decreased water and air flux compared with other sites. Drying of the soil greatly altered its structure. The decrease in soil volume between wet and dry soil was 7 - 10%, most of which occurred in the moisture range of field conditions. Structural changes, including irreversible collapse of interaggregate pores, began at matric potentials around -6 kPa indicating, instability of soil structure against increasing hydraulic stress. Water saturation and several freezethaw cycles between autumn and spring likely weakened the soil structure. Soil water repellency was observed at all sites at the time of sampling and when soil was dryer than about 40 vol.%. (matric potential < -6 kPa). Therefore, water repellency contributes to water flow over a wide moisture range. Water repellency was also observed in soils with low organic carbon content (< 2%), which suggests that this phenomenon is common in agricultural soils of Finland due to their relatively high organic carbon content. Aggregate-related pedofeatures of dense infillings described as clay intrusions were found at all sites. The formation of these intrusions was attributed to clay dispersion and/or translocation during spring thaw and drying of the suspension in situ. These processes generate very new aggregates whose physical properties are most probably different from those of the bulk soil aggregates. Formation of the clay infillings suggested that prolonged wetness in autumn and spring impairs soil structure due to clay dispersion, while on the other hand it contributes to the pedogenesis of the soil. The results emphasize the dynamic nature of the physical properties of clay soils, essentially driven by their moisture state. In a dry soil, fast preferential flow is favoured by abundant macropores including shrinkage cracks and is further enhanced by water repellency. Increase in soil moisture reduces water repellency, and swelling of accommodated pores lowers the saturated hydraulic conductivity. Moisture- and temperature-related processes significantly alter soil structure over a time span of 1 yr. Thus, the pore characteristics as well as the hydraulic properties of soil are time-dependent.

H2O2, in addition to producing highly reactive molecules through hydroxyl radicals or peroxidase action, can exert a number of direct effects on cells, organelles and enzymes. The stimulations include glucose transport, glucose incorporation into glycogen, HMP shunt pathway, lipid synthesis, release of calcium from mitochondria and of arachidonate from phospholipids, poly ADP ribosylation, and insulin receptor tyrosine kinase and pyruvate dehydrogenase activities. The inactivations include glycolysis, lipolysis, reacylation of lysophospholipids, ATP synthesis, superoxide dismutase and protein kinase C. Damages to DNA and proteoglycan and general cytotoxicity possibly through oxygen radicals were also observed. A whole new range of effects will be opened by the finding that H2O2 can act as a signal transducer in oxidative stress by oxidizing a dithiol protein to disulphide form which then activates transcription of the stress inducible genes. Many of these direct effects seem to be obtained by dithiol-disulphide modification of proteins and their active sites, as part of adaptive responses in oxidative stress.

A Bacia de Almada, localizada no estado da Bahia, compartilha características similares com as outras bacias da margem leste do Brasil, quando é analisada segundo aspectos como os processos sedimentares e o regime de esforço dominante durante a sua formação. Observa-se uma diferença marcante em relação as outras bacias quando é analisada sob a ótica da composição da crosta transicional, uma vez que não se registra atividade vulcânica durante a fase rifte. A aquisição de um extenso levantamento sísmico 3D, com cabos de 6 km de comprimento e 9.2 segundos de tempo de registro (tempo sísmico duplo), resultaram em imagens sísmicas de boa qualidade das estruturas profundas do rifte. Adicionalmente, estudos de modelagem gravimétrica foram integrados com a análise sísmica para corroborar o modelo geológico. A Bacia de Almada é parte dos sistemas de rifte continentais, desenvolvidos durante o Berriasiano até o Aptiano, que antecederam a quebra do continente do Gondwana, evoluindo posteriormente para uma margem passiva divergente. O processo do rifteamento desenvolveu cinco sub-bacias de orientação NNE-SSO, desde posições terrestres até marinhas profundas, produzindo um arcabouço estrutural complexo. Os perfis da sísmica profunda mostram o afinamento progressivo da crosta continental até espessuras da ordem de 5 km, abaixo da sub-bacia mais oriental, com fatores de estiramento crustal próximo a 7 antes do desenvolvimento de crosta oceânica propriamente dita. As imagens sísmicas de boa qualidade permitem também o reconhecimento de sistemas de falhas lístricas que se iniciam na crosta superior, evoluem atravessando a crosta e conectando as sub-bacias para finalizar em um descolamento horizontal na crosta inferior estratificada. Adicionalmente, a bacia apresenta um perfil assimétrico, compatível com mecanismos de cisalhamento simples. As margens vulcânicas (VM) e não vulcânicas (NVM), são os extremos da análise composicional das margens divergentes continentais. Na Bacia de Almada não se reconhecem os elementos arquiteturais típicos das VM, tais como são as grandes províncias ígneas, caracterizadas por cunhas de refletores que mergulham em direção ao mar e por intenso vulcanismo pré- e sin-rifte nas bacias. Embora a margem divergente do Atlântico Sul seja interpretada tradicionalmente como vulcânica, o segmento do rifte ao sul do Estado da Bahia apresenta características não-vulcânicas, devido à ausência destes elementos arquiteturais e aos resultados obtidos nas perfurações geológicas que eventualmente alcançam a seqüência rifte e embasamento. Regionalmente a margem divergente sul-americana é majoritariamente vulcânica, embora a abundância e a influência do magmatísmo contemporâneo ao rifte seja muito variável. Ao longo da margem continental, desde a Bacia Austral no sul da Argentina, até a Bacia de Pernambuco no nordeste do Brasil, podem ser reconhecidos segmentos de caráter vulcânico forte, médio e não vulcânico. Nos exemplos clássicos de margens não vulcânicas, como a margem da Ibéria, a crosta transicional é altamente afinada podendo apresentar evidências de exumação de manto. Na Bacia de Almada, a crosta transicional apresenta importante estiramento embora não haja evidências concretas de exumação de manto. Os mecanismos responsáveis pela geração e intrusão dos grandes volumes de magma registrados nas margens divergentes são ainda sujeitos a intenso debate. Ao longo da margem divergente sul-americana há evidências da presença dos mecanismos genéticos de estiramento litosférico e impacto de plumas. Alternativamente estes dois mecanismos parecem ter tido um papel importante na evolução tectônica da margem sudeste e sul, diferenciando-as da margem continental onde foi implantada a Bacia de Almada.
