One-third of botanical remedies from southern Italy are used to treat skin and soft tissue infections (SST's). Methicillin-resistant Staphylococcus aureus (MRSA), a common cause of SSTIs, is responsible for increased morbidity and mortality from infections. Therapeutic options are limited by antibiotic resistance. Many plants possess potent antimicrobial compounds for these disorders. Validation of traditional medical practices is important for the people who rely on medicinal plants. Moreover, identification of novel antibiotics and anti-pathogenic agents for MRSA is important to global healthcare.^ I took an ethnopharmacological approach to understand how Italian medicinal plants used for the treatment of SSTIs affect MRSA growth and virulence. My hypothesis was that plants used in folk remedies for SSTI would exhibit lower cytotoxicity and greater inhibition of bacterial growth, biofilm formation and toxin production in MRSA than plants used for remedies unrelated to the skin or for plants with no ethnomedical application. The field portion of my research was conducted in the Vulture-Alto Bradano area of southern Italy. I collected 104 plant species and created 168 crude extracts. In the lab, I screened samples for activity against MRSA in a battery of bioassays. Growth inhibition was analyzed using broth microtiter assays for determination of the minimum inhibitory concentration. Interference with quorum-sensing (QS) processes, which mediate pathogenicity, was quantified through RP-HPLC of δ-toxin production. Interference with biofilm formation and adherence was assessed using staining methods. The mammalian cytotoxicity of natural products was analyzed using MTT cell proliferation assay techniques.^ Although bacteriostatic activity was limited, extracts from six plants used in Italian folk medicine (Arundo donax, Ballota nigra, Juglans regia, Leopoldia comosa, Marrubium vulgare, and Rubus ulmifolius ) significantly inhibited biofilm formation and adherence. Moreover, plants used to treat SSTI demonstrated significantly greater anti-biofilm activity when compared to plants with no ethnomedical application. QSI activity was evident in 90% of the extracts tested and extracts from four plants ( Ballota nigra, Castanea saliva, Rosmarinus officinalis, and Sambucus ebulus) exhibited a significant dose-dependent response. Some of the plant remedies for SSTI identified in this study can be validated due to anti-MRSA activity.^

The exponential growth of studies on the biological response to ocean acidification over the last few decades has generated a large amount of data. To facilitate data comparison, a data compilation hosted at the data publisher PANGAEA was initiated in 2008 and is updated on a regular basis (doi:10.1594/PANGAEA.149999). By January 2015, a total of 581 data sets (over 4 000 000 data points) from 539 papers had been archived. Here we present the developments of this data compilation five years since its first description by Nisumaa et al. (2010). Most of study sites from which data archived are still in the Northern Hemisphere and the number of archived data from studies from the Southern Hemisphere and polar oceans are still relatively low. Data from 60 studies that investigated the response of a mix of organisms or natural communities were all added after 2010, indicating a welcomed shift from the study of individual organisms to communities and ecosystems. The initial imbalance of considerably more data archived on calcification and primary production than on other processes has improved. There is also a clear tendency towards more data archived from multifactorial studies after 2010. For easier and more effective access to ocean acidification data, the ocean acidification community is strongly encouraged to contribute to the data archiving effort, and help develop standard vocabularies describing the variables and define best practices for archiving ocean acidification data.

Algumas plantas são uma fonte natural de compostos bioativos, tais como polifenóis, vitaminas, carotenóides e ácidos gordos insaturados. Esta diversidade de biomoléculas permite a sua utilização em diversas áreas, especialmente como aditivos alimentares e ingredientes naturais para promoção da saúde. Estes fitoquímicos têm sido utilizados na industria farmacêutica, bem como na formulação de suplementos dietéticos, alimentos funcionais e nutracêuticos. No entanto, a utilização de matérias-primas de boa qualidade microbiológica é um dos requisitos essenciais na indútria, uma vez que os microrganismos podem contaminar o produto final, levando à sua deterioração. Assim, a irradiação é creditada para que a sua aplicação seja permitida em ingredientes secos, sendo cada vez mais reconhecida mundialmente, devido à eficiência na redução das perdas causadas por processos fisiológicos naturais (brotamento, maturação e envelhecimento), para eliminar ou reduzir microorganismos, parasitas e pragas, sem que ocorra qualquer alteração (química ou organoléptica) no alimento, tornando-o mais seguro para o consumidor [1-3]. O objetivo deste estudo foi avaliar os efeitos da aplicação de diferentes doses de radiação gama e feixe de eletrões na composição química e bioatividade de várias plantas (Ginkgo biloba L., Melissa officinalis L., Melittis melissophyllum L., Mentha piperita L., Aloysia citrodora Palàu, Arenaria montana L. e Thymus vulgaris L.).

Chestnut flowers, lemon balm plants and their decoctions were incorporated into "Serra da Estrela" cheese, to assess their potential to preserve its nutritional properties and provide new foodstuffs. The analyses were carried out after the normal ripening period of 1month and after 6months of storage. The most abundant nutrients were proteins and fats. The most abundant minerals were Ca and Na, while C16:0 and C18:1 were the main fatty acids. Saturated fatty acids were the most abundant, followed by the monounsaturated. Moisture seemed to be lower in the samples with the plants incorporated. The dried plants, when incorporated, seemed to be more efficient as preservers then the decoctions, although these better preserved the proteins. These plants can be regarded as promising natural preservers in foodstuffs cheese, given the preservation of key parameters and the slight impact on the nutritional value.

Liver diseases represent a major health problem around the world. in Mexico these are the 5th leading cause of death in the economically active population. in Mexico, it is estimated that about 60% of the population uses some medicine from plants to treat their illnesses. The purpose of this work was to search for medicinal plants in Mexico that have been evaluated for their hepatoprotective effect in different models. in this review we found only 13 plants evaluated for hepatoprotective activity: Amole tuber, Cochlospermum vitifolium, Heterotheca inuloides, Hibiscus sabdariffa, Leucophyllum frutescens, Prostechea michuacana, Psidium Guajava, Rosmarinus officinalis, Verbena Carolin, Centaurea americana, Juglans mollis, Krameria ramossisima and Turnera diffusa. This study describes the studies conducted in Mexico for each of them and the international literature reports of pharmacological and phytochemical studies.

Se ha localizado en el margen de la carretera comarcal CV 795 en su kilómetro 17 próximo a Ulls de Canals y dentro del término de Banyeres de Mariola, una buena población asilvestrada de Rosa gallica L. var. officinalis (hort. ex Andrews) Ser., que corresponde a la segunda población localizada en Alicante, puesto que se conocía una población pero situada próxima al litoral en la comarca de la Marina Alta. La llamada rosa de Provins o provincialis (de Provenza) es una de las rosas cultivadas en Europa desde más antiguo, probablemente la primera. Aparece dibujada en los frescos de Pompeya, fue también la rosa de las boticas (“Officinalis”) y se corresponde con la famosa rosa roja o Rosa de Lancanster, símbolo de este bando en la Guerra de civil inglesa llamada de “las dos rosas”. Su cultivo en España fue muy importante y era frecuentemente citada en la literatura botánica antigua, fue llamada “rosa de Castilla” pero con el tiempo y el abandono del campo, su presencia fue disminuyendo y de forma asilvestrada solo aparece en unas pocas provincias españolas, aunque se sigue manteniendo su cultivo a pequeña escala en colecciones particulares y rosaledas. Precisamente, en la Rosaleda “Pere Dot” del Jardín Botánico de Torretes (Ibi) mantenemos en cultivo dos de las variedades botánicas reconocidas y muchos de los cultivares e híbridos de este rosal, que hemos adquirido de colecciones extranjeras. El hallazgo de este material asilvestrado probablemente procedente de antiguos cultivos en la zona, tiene gran valor etnobotánico y puede tener valor genético. Aunque en su morfología no se han encontrado diferencias relevantes, se ha procedido a su multiplicación vegetativa y será próximamente incluido en la rosaleda del Jardín Botánico, junto con pliegos de herbario y semillas.

Calendula officinalis is grown widely as an ornamental plant across Europe. It belongs to the large. Asteraceae family. In this study, the aim was to explore the possibilities to use Calendula officinalis as a new model organism for flower development and secondary mechanism studies in Asteraceae. Tissue culture of Calendula officinalis was established using nine different cultivars. Murashige & Skoog (MS) medium with four different combinations of plant growth regulators were tested. Of all these combinations, the medium containing 1mg/l BAP, 0.1 mg/l IAA, and 1mg/l Zeatin achieved highest frequency of adventitious shoot regeneration from hypocotyl and cotyledon explants. Virus-induced gene silencing is a recent developed genetic tool for charactering the gene functions in plants, and extends the range of host plants that are not accessible for Agrobacterium transformation. Here, tobacco rattle virus (TRV)-based VIGS technique was tested in calendula (cv. Single Orange). We used TRV carrying Gerbera hybrid phytoene desaturase (PDS) gene fragment to induce PDS silencing in calendula. Vacuum infiltration and syringe infiltration methods both resulted in photo-bleaching phenotypes in leaves, bracts and petals. Loss-of-function phenotypes occurred on calendula 13 days post-infiltration. In conclusion, the data indicates that calendula explants can be regenerated through tissue culture which is a prerequisite for development of stable transformation methods. However, further optimization is still needed to improve the frequency. In addition, VIGS was applied to silence PDS marker gene expression indicating that this method has potential for gene functional studies in future.

Resumen: La producción de espárragos en Argentina está caracterizada por una elevada estacionalidad concentrada en el periodo octubre-diciembre, por lo que el empleo de invernaderos brinda la posibilidad de ampliar el calendario de oferta, anticipando la entrada en producción. Con el objetivo de evaluar el rendimiento de primicia de diferentes híbridos de espárrago verde, se realizó un ensayo en invernadero con ocho híbridos, en UCA Buenos Aires, iniciado el 15/11/2006, mediante plantines de 100 días a 1m*0,3m. Se evaluaron los siguientes genotipos: Italo, Zeno, Eros, Ercole, H-668, Marte, y Giove, de origen italiano, y UC-157 de origen americano, como testigo por ser el tradicionalmente cultivado en Argentina. Se evaluaron 22 cosechas, con una frecuencia de día por medio, del 17/08/2011-25/10/2011. Se estudiaron diferencias en kg totales y comerciales (PFT y PFC), Nº turiones totales y comerciales/ha (NTT y NTC) y distribución de calibres (DC): Jumbo (J), Extra-Large (XL), Large (L), Medium (M), Small (S) y Asparagina (A). Se efectuó un análisis multifactor ANOVA LSD test (P>0.05). En promedio se obtuvieron: PFT: 17053; PFC: 7904 kg.ha-1; NTT: 670566 y NTC: 520938 turiones.ha-1. Se destacaron: en PFT: Italo: 29458a, Zeno: 23056b, Giove: 23034b y H-668: 18568bc; en PFC: Italo: 14850a, Giove: 9856b, Zeno: 9130bc y H-668: 8228bcd; en NTT: Italo: 842512a, H-668: 754512a, Giove: 715000b y Eros: 707498b; en NTC: Italo: 844998a, H-668: 667502b y Eros: 542498bc y en DC: en J: Italoa, Gioveb, Zenobc y UC-157bcd; en XL: Italoa, Zenoab y Gioveb; en L: Giovea, Italoa, Zenoab, H-668abc y UC-157abc; en M: Italoa, H-668ab y Erosab; en S: H-668a, Erosa e Italoab y en A: H-668a y Marteab. Por lo expuesto resulta alentadora la productividad de Italo, Giove, Zeno y H-668 para producción de espárrago verde de primicia en invernadero.

稻属(Oryza L.)属于禾本科稻族(Oryzeae Dumortier)。本属包括AA,BB,BBCC,CC,CCDD,EE,FF,GG,HHJJ和HHKK十个基因组,二十余种。其中Oryza officinalis复合体包括BB,BBCC,CC,CCDD和EE五个基因组,九个种。众多的学者对该复合体进行了广泛深入的研究,为后续研究奠定了坚实的基础。然而,迄今为止,多倍体物种形成问题许多没有解决,基因组间的关系尚未完全阐明,甚至有些多倍体物种的基因组组成仍未确认。 本文评述了Oryza officinalis复合体中基因组研究的历史和现状,用基因组原位杂位(GISH)的方法,对该复合体四倍体物种的基因组组成作了验证;对B,C,D和E四个基因组间的关系进行了研究。同时对基因组原位杂交的方法,原位杂交鉴定多倍体基因组的组成以及研究基因组间关系的方法作了一些探讨。其主要研究结果如下: 一. 原杂交方法的研究:1)染色体制片:比较研究了不同的制片方法,发现压片法适用于大染色体的材料;将酶解/空气干燥法(Fukui et al., 1992)加以改进后,特别适宜于小染色体植物材料的制片。2)根尖储存时间和条件对原位杂交的影响:发现在-20 ℃的酒精(70%)中储存8个月以内的根尖材料,可用于原位杂交;而在-20 ℃的固定液中储藏18个月的根尖,DNA降解严重,不能用于GISH。3)探针标记:比较了随机引物法、缺口平移法和两步标记法(先用随机引物标记后,再用缺口平移法进行标记的方法)的优缺点。结果显示两步标记法是最佳标记方法。 二. GISH鉴定异源多倍体的方法:用两个异源四倍体Oryza minuta和Scilla sinensis (2n = 34)做染色体制片,进行原位杂交实验,结果表明:1)用二倍体亲本基因组之一做探针而不用封阻DNA, 可以鉴别Oryza minuta而不能鉴别Scilla sinensis中的基因组。2)用一个二倍体亲本做探针而用另一个做封阻,能够区分Scilla sinensis的两个基因组;但过量的封阻DNA将可以造成一些实验假象。3)同时用两个亲本的DNA做探针,不仅能够有效分辨不同的基因组,还能够根据交叉杂交程度推测基因组间的分化程度,是鉴别异源多倍体最有效的方法。 三. GISH鉴定稻属四倍体的基因组组成:1)Oryza minuta, O. punctata和O. malampuzhaensis的基因组的组成都为BBCC。2)Oryza minuta中B基因组和二倍体O. punctata中的B基因组之间存在着明显的基因组内分化。3)O. alta是一个异源多倍体,其基因组组成为CCDD。但C和D之间的分化不彻底,可以认为它不是一个严格意义上的异源多倍体。 四. GISH研究B,C,D和E基因组间的关系:1)B基因组和C基因组之间的关系最远;E和C之间的分化同E和B之间的分化程度接近,但E和C之间的分化比E和D之间的分化要小一些;C和D之间的分化不彻底,关系最近。

Different extraction processes were employed to extract the polyphenolic compounds from pitanga (Eugenia uniflora L) leaves: a one-step process using water, ethanol or supercritical CO(2) as solvents, and a two-step process using supercritical CO(2) followed by either water or ethanol. The total polyphenolic compounds, total flavonoids and antioxidant activity were determined in all the extracts obtained. The process performance was evaluated with respect to three variables: global extraction yield, concentration and yield of both polyphenols and flavonoids in the extracts. For the one-step extraction, the results showed that the extraction yield increased with solvent polarity. For the two-step process, the results suggested that water was more efficient in extracting the phenolic compounds from E. uniflora when the matrix was previously extracted with scCO(2). With respect to the antioxidant activity, the ethanolic extracts obtained from both processes, using either the DPPH radical scavenging method or the beta-carotene bleaching method, presented high antioxidant activities. This study evaluated the inhibitory activity of copaiba oil (Copaifera officinalis against the cariogenic microorganism, Streptococcus mutans. For such purpose, a minimum inhibition concentration test of copaiba oil against S. mutans was performed, using the serial dilution in broth technique, with a negative control, a positive control (0.12% chlorhexidine) and a 10% copaíba oil solution as a test. A minimum bactericidal concentration test with tubes presenting microbial inhibition was also conduced. In the minimum inhibitory concentration test, copaiba oil showed inhibition of bacterial growth at all concentrations tested up to 0.78 µL/mL of the 10% copaiba oil solution in the broth. In addition, the negative control had no inhibition, and the 0.12% chlorhexidine solution was effective up to 6.25 µL/mL in the broth. Copaiba oil showed a bacteriostatic activity against S. mutans at low concentrations, and could be a an option of phytotherapic agent to be used against cariogenic bacteria in the prevention of caries disease.
