The objective of this work was to typify, through physicochemical parameters, honey from Campos do Jordão’s microrregion, and verify how samples are grouped in accordance with the climatic production seasonality (summer and winter). It were assessed 30 samples of honey from beekeepers located in the cities of Monteiro Lobato, Campos do Jordão, Santo Antonio do Pinhal e São Bento do Sapucaí-SP, regarding both periods of honey production (November to February; July to September, during 2007 and 2008; n = 30). Samples were submitted to physicochemical analysis of total acidity, pH, humidity, water activity, density, aminoacids, ashes, color and electrical conductivity, identifying physicochemical standards of honey samples from both periods of production. Next, we carried out a cluster analysis of data using k-means algorithm, which grouped the samples into two classes (summer and winter). Thus, there was a supervised training of an Artificial Neural Network (ANN) using backpropagation algorithm. According to the analysis, the knowledge gained through the ANN classified the samples with 80% accuracy. It was observed that the ANNs have proved an effective tool to group samples of honey of the region of Campos do Jordao according to their physicochemical characteristics, depending on the different production periods.

The type A gasoline samples were analyzed by gas chromatography with flame ionization detector (GC-FID) which allowed quantifying and classifying of the various compounds into different classes of hydrocarbons. Several physicochemical parameters were evaluated according to the official methods in order to compare the results obtained against the limits established by the Agência Nacional de Petróleo, Gás Natural e Biocombustíveis (ANP, 2011). Additionally, principal component analysis (PCA) was applied to discriminate the samples studied, which revealed the separation of four groups according to their chemical composition determined in samples collected from the eight fuel distributors in the State of Pará.

Although the biopolymer poly-(3-hydroxybutyrate), P[3HB], presents physicochemical properties that make it an alternative material to conventional plastics, its biotechnological production is quite expensive. As carbon substrates contribute greatly to P[3HB] production cost, the utilization of a cheaper carbon substrate and less demanding micro-organisms should decrease its cost. In the present study a 23 factorial experimental design was applied, aiming to evaluate the effects of using hydrolysed corn starch (HCS) and soybean oil (SBO) as carbon substrates, and cheese whey (CW) supplementation in the mineral medium (MM) on the responses, cell dried weigh (DCW), percentage P[3HB] and mass P[3HB] by recombinant Escherichia coli strains JM101 and DH10B, containing the P[3HB] synthase genes from Cupriavidus necator (ex-Ralstonia eutropha). The analysis of effects indicated that the substrates and the supplement and their interactions had positive effect on CDW. Statistically generated equations showed that, at the highest concentrations of HCS, SO and CW, theoretically it should be possible to produce about 2 g L(1) DCW, accumulating 50% P[3HB], in both strains. To complement this study, the strain that presented the best results was cultivated in MM added to HCS, SBO and CW ( in best composition observed) and complex medium (CM) to compare the obtained P[3HB] in terms of physicochemical parameters. The obtained results showed that the P[3HB] production in MM (1.29 g L(-1)) was approximately 20% lower than in CM (1.63 g L(-1)); however, this difference can be compensated by the lower cost of the MM achieved by the use of cheap renewable carbon sources. Moreover, using differential scanning calorimetry and thermogravimetry analyses, it was observed that the polymer produced in MM was the one which presented physicochemical properties (Tg and Tf) that were more similar to those found in the literature for P[3HB].

During the manufacture of explosives, large amounts of water are used to remove unwanted by-products generated. This water in turn, ends up in wastewater treatment plants or water bodies. The aim of this study was to evaluate the toxic potential of effluent generated by 2.4.6-Trinitrotoluene (TNT) production, yellow water, red water and mixture of yellow and red water, produced from a plant located in the Paraiba Valley, Sao Paolo state, Brazil. Daphnia similis, Danio rerio, Escherichia coli, Pseudomonas putida and Pseudokircheneriella subcaptata were used as test organisms. Physicochemical parameters such as color, pH, conductivity, total dissolved solids, dissolved oxygen, chemical oxygen demand (COD) and biochemical oxygen demand (BOD) were evaluated. Effluent from 2.4.6-TNT production was extremely toxic to all test organisms. The physicochemical parameters evaluated showed high levels of conductivity (from 41.533 to 42.344 mu S /cm) and chemical oxygen demand (COD of 8471 to 27.364 mg/L) for the effluents analyzed.

Die exzitatorische Neurotransmission erfolgt über ionotrope Glutamat-Rezeptoren von denen dem NMDA-(N-Methyl-D-aspartat)-Rezeptor durch seine hohe Leitfähigkeit für Ca2+-Ionen eine besondere Rolle zugesprochen wird. Bei seiner Überaktivierung kommt es zu exzitotoxischen Prozessen, die direkt mit neurodegenerativen Erkrankungen einhergehen und nach einem Schlaganfall, bei akuten Epilepsien, Morbus Parkinson, Alzheimer Demenz aber auch im Bereich der neuropathischen Schmerzentstehung eine wichtige Rolle spielen.rnDurch das Eingreifen in die glutamatvermittelten pathologischen Prozesse verspricht man sich daher die Möglichkeit einer Neuroprotektion bei der Therapie verschiedener neurodegenerativer Erkrankungen, die primär auf völlig unterschiedliche Ursachen zurückzuführen sind.rnAusgehend von in früheren Arbeiten synthetisierten Hydantoin-substituierten Dichlor-indol-2-carbonsäure-Derivaten, die hochaffine Eigenschaften zur Glycin-Bindungsstelle des NMDA-Rezeptors aufweisen, sollten neue Derivate entwickelt und untersucht werden, die hinsichtlich ihrer Affinität zur Glycin-Bindungsstelle des NMDA-Rezeptors, ihrer Pharmakokinetik sowie physikochemischen Parameter in präparativ-organischen, radiopharmazeutischen und zell- bzw. tierexperimentellen Studien in vitro sowie in vivo charakterisiert werden sollten. Von besonderem Interesse war dabei die Evaluierung der synthetisierten Verbindungen in einem Verdrängungsassay mit dem Radioliganden [3H]MDL105,519 mit dem der Einfluss der strukturellen Modifikationen auf die Affinität zur Glycin-Bindungsstelle des Rezeptors untersucht wurde, sowie die Selektivität und die Potenz der Liganden abgeschätzt wurde.rnIm Rahmen der Struktur-Wirkungs-Untersuchungen mit Hilfe der Bindungsexperimente konnten bestimmte Strukturmerkmale als essentiell herausgestellt bzw. bekräftigt werden. Die Testverbindungen zeigten dabei IC50-Werte im Bereich von 0,0028 bis 51,8 μM. Die entsprechenden Ester dagegen IC50-Werte von 23,04 bis >3000 μM. Als vielversprechende Strukturen mit Affinitäten im niedrigen nanomolaren Bereich stellten sich Derivate mit einer 4,6-Dichlor-oder Difluor-Substitution am Indolgrundgerüst (2,8 bis 4,6 nM) heraus. Auch die Substitution des Phenylhydantoin-Teils durch das bioisostere Thienylhydantoin führte zu einer gleichbleibenden ausgeprägten Affinität (3,1 nM). rnZur Abschätzung der Bioverfügbarkeit, insbesondere der Fähigkeit zur Überwindung der Blut-Hirn-Schranke, wurden die Lipophilien bei einer Auswahl der Testverbindungen durch Bestimmung ihrer log P-Werte ermittelt. Neben dem Verfahren der potentiometrischen Titration wurde eine HPLC-Methode an einer RP-Phase verwendet.rnUm das Zytotoxizitätsprofil der synthetisierten Strukturen frühzeitig abschätzen zu können, wurde ein schnell durchführbares, zellbasiertes in vitro-Testsystem, der kommerziell erhältliche „Cell Proliferation Kit II (XTT-Test)“, eingesetzt. rnIm Rahmen von Positronen-Emissions-Tomographie-Experimenten an Ratten wurde eine Aussage bezüglich der Aufnahme und Verteilung eines radioaktiv markierten, hochaffinen Liganden an der Glycinbindungsstelle des NMDA-Rezeptors im Gehirn getroffen. Dabei wurden sowohl ein Carbonsäure-Derivat sowie der korrespondierende Ethylester dieser Testung unterworfen.rn

A disposable microarray was developed for detection of up to 90 antibiotic resistance genes in gram-positive bacteria by hybridization. Each antibiotic resistance gene is represented by two specific oligonucleotides chosen from consensus sequences of gene families, except for nine genes for which only one specific oligonucleotide could be developed. A total of 137 oligonucleotides (26 to 33 nucleotides in length with similar physicochemical parameters) were spotted onto the microarray. The microarrays (ArrayTubes) were hybridized with 36 strains carrying specific antibiotic resistance genes that allowed testing of the sensitivity and specificity of 125 oligonucleotides. Among these were well-characterized multidrug-resistant strains of Enterococcus faecalis, Enterococcus faecium, and Lactococcus lactis and an avirulent strain of Bacillus anthracis harboring the broad-host-range resistance plasmid pRE25. Analysis of two multidrug-resistant field strains allowed the detection of 12 different antibiotic resistance genes in a Staphylococcus haemolyticus strain isolated from mastitis milk and 6 resistance genes in a Clostridium perfringens strain isolated from a calf. In both cases, the microarray genotyping corresponded to the phenotype of the strains. The ArrayTube platform presents the advantage of rapidly screening bacteria for the presence of antibiotic resistance genes known in gram-positive bacteria. This technology has a large potential for applications in basic research, food safety, and surveillance programs for antimicrobial resistance.

The injurious effect of nonsteroidal anti-inflammatory drugs (NSAIDs) in the small intestine was not appreciated until the widespread use of capsule endoscopy. Animal studies found that NSAID-induced small intestinal injury depends on the ability of these drugs to be secreted into the bile. Because the individual toxicity of amphiphilic bile acids and NSAIDs directly correlates with their interactions with phospholipid membranes, we propose that the presence of both NSAIDs and bile acids alters their individual physicochemical properties and enhances the disruptive effect on cell membranes and overall cytotoxicity. We utilized in vitro gastric AGS and intestinal IEC-6 cells and found that combinations of bile acid, deoxycholic acid (DC), taurodeoxycholic acid, glycodeoxycholic acid, and the NSAID indomethacin (Indo) significantly increased cell plasma membrane permeability and became more cytotoxic than these agents alone. We confirmed this finding by measuring liposome permeability and intramembrane packing in synthetic model membranes exposed to DC, Indo, or combinations of both agents. By measuring physicochemical parameters, such as fluorescence resonance energy transfer and membrane surface charge, we found that Indo associated with phosphatidylcholine and promoted the molecular aggregation of DC and potential formation of larger and isolated bile acid complexes within either biomembranes or bile acid-lipid mixed micelles, which leads to membrane disruption. In this study, we demonstrated increased cytotoxicity of combinations of bile acid and NSAID and provided a molecular mechanism for the observed toxicity. This mechanism potentially contributes to the NSAID-induced injury in the small bowel.

Karst aquifers are known for their wide distribution of water transfer velocities. From this observation, a multiple geochemical tracer approach seems to be particularly well suited to provide a significant assessment of groundwater flows, but the choice of adapted tracers is essential. In this study, several common tracers in karst aquifers such as physicochemical parameters, major ions, stable isotopes, and d13C to more specific tracers such as dating tracers – 14C, 3H, 3H–3He, CFC-12, SF6 and 85Kr, and 39Ar – were used, in a fractured karstic carbonated aquifer located in Burgundy (France). The information carried by each tracer and the best sampling strategy are compared on the basis of geochemical monitoring done during several recharge events and over longer time periods (months to years). This study’s results demonstrate that at the seasonal and recharge event time scale, the variability of concentrations is low for most tracers due to the broad spectrum of groundwater mixings. The tracers used traditionally for the study of karst aquifers, i.e., physicochemical parameters and major ions, efficiently describe hydrological processes such as the direct and differed recharge, but require being monitored at short time steps during recharge events to be maximized. From stable isotopes, tritium, and Cl� contents, the proportion of the fast direct recharge by the largest porosity was estimated using a binary mixing model. The use of tracers such as CFC-12, SF6, and 85Kr in karst aquifers provides additional information, notably an estimation of apparent age, but they require good preliminary knowledge of the karst system to interpret the results suitably. The CFC-12 and SF6 methods efficiently determine the apparent age of baseflow, but it is preferable to sample the groundwater during the recharge event. Furthermore, these methods are based on different assumptions such as regional enrichment in atmospheric SF6, excess air, and flow models among others. 85Kr and 39Ar concentrations can potentially provide a more direct estimation of groundwater residence time. Conversely, the 3H–3He method is inefficient in the karst aquifer for dating due to 3He degassing.

In vitro incubation of acetylcholinesterase from brain tissue of several species with organophosphate compounds indicated that the concentrations required to inhibit 50% of acetylcholinesterase activity (IC(,50)) differed from species to species for the same compound (Murphy, et al., 1968; Andersen, et al., 1972, 1977 and 1978).^ The hypothesis that non-specific binding proteins (Lauwerys and Murphy, 1969a,b) exerts a protective effect on acetylcholinesterase, and thus cause the differences observed in IC(,50) studies was tested by a ('3)H-DFP binding experiment. It was found that differences in the amount of non-specific binding protein cannot explain the observed differences observed in IC(,50) studies.^ An alternative hypothesis, that acetylcholinesterase from different species have different affinities for binding and/or different rates of phosphorylation by organophosphate insecticides was tested by determining the apparent affinity constant (k(,a)) and apparent rate of phosphorylation (k(,p)). Kinetic studies indicated that acetylcholinesterases from different species have different sensitivities to inhibition by organophosphate insecticides, and the differences are due to different affinities for binding and/or different rates of phosphorylation by the same organophosphate compound.^ Studies of the spontaneous reactivation of acetylcholinesterase after inhibition by organophosphate insecticides also indicated that acetylcholinesterases from different species have different rates and extents of spontaneous reactivation. This further substantiates the hypothesis that acetylcholinesterases from different species have different kinetic characteristics with respect to organophosphate insecticides inhibition.^ Eleven paraoxon analogs were synthesized for a quantitative structure-activity relationship study. It was found that the electron-withdrawing power ((sigma)) and hydrophobicity ((PARAGR)) of the substituent are important in determining the anti-cholinesterase activity of paraoxon analogs. Thus, predictions of species differences in acetylcholinesterase sensitivities to paraoxon analogs can be made if the physicochemical parameters ((sigma) and (PARAGR)) of the substituents are known.^ In another approach, i.e. enzyme modeling, the sensitivity of rat brain acetylcholinesterase to organophosphate insecticides was used as the independent variable to predict the sensitivities of acetylcholinesterases from other species to the same compound. Regression equations were derived for each species based on nineteen organophosphate insecticides studied. It was found, that in addition to paraoxon analogs, this method is also applicable to other organophosphate compounds with wide variations in structure. Thus, the sensitivities of acetylcholinesterases from other species can also be predicted from the sensitivity of rat brain acetylcholinesterase. ^
