Recent studies found that isolates of Toxoplasma gondii from Brazil were biologically and genetically different from those in North America and Europe. However, to date only a small number of isolates have been analysed from different animal hosts in Brazil. In the present study DNA samples of 46 T. gondii isolates from cats in 11 counties in Sao Paulo state, Brazil were genetically characterised using 10 PCR restriction fragment length polymorphism markers including SAG1, SAG2, SAG3, STUB, GRA6, c22-8, c29-2, L358, PKI and Apico. An additional marker, CS3, that locates on chromosome VIIa and has previously been shown to be linked to acute virulence of T. gondii was also used to determine its association to virulence in mice. Genotyping of these 46 isolates revealed a high genetic diversity with 20 genotypes but no clonal Type I, II or III lineage was found. Two of the 46 isolates showed mixed infections. Combining genotyping data in this study with recent reported results from chickens, dogs and cats in Brazil (total 125 isolates) identified 48 genotypes and 26 of these genotypes had single isolates. Four of the 48 genotypes with multiple isolates identified from different hosts and locations are considered the common clonal lineages in Brazil. These lineages are designated as Types BrI, BrII, BrIII and BrIV. These results indicate that the T. gondii population in Brazil is highly diverse with a few successful clonal lineages expanded into wide geographical areas. In contrast to North America and Europe, where the Type II clonal lineage is overwhelmingly predominant, no Type II strain was identified from the 125 Brazil isolates. Analysis of mortality rates in infected mice indicates that Type BrI is highly virulent, Type BrIII is non-virulent, whilst Type BrII and BrIV lineages are intermediately virulent. In addition, allele types at the CS3 locus are strongly linked to mouse-virulence of the parasite. Thus, T. gondii has an epidemic population structure in Brazil and the major lineages have different biological traits. (C) 2007 Australian Society for Parasitology Inc. Published by Elsevier Ltd. The aim of this paper was to genetically characterize Toxoplasma gondii isolates from free range chickens in regions of Brazilian territory in the state of Mato Grosso do Sul (MS) where T. gondii strains have never been studied. In total, T. gondii isolates from 22 free range chickens were included in this study. Fifty chickens from Eldorado, thirty from Rio Verde and ten from Aquidauana were sampled between January and April 2007. In relation to the genetic diversity of T. gondii isolates from chickens in MS, the magnitude of the diversity in the isolates sampled in this study was comparable to the overall diversity in a composite data set. These 22 isolates in MS revealed 11 genotypes, whereas the 321 isolates ever genotyped in Brazil have revealed 95 genotypes. The values of Simpson`s Diversity Index for the whole population of T. gondii isolates in Brazil, the whole population of T. gondii isolates from chickens in Brazil and the population surveyed in this study were 0.97, 0.95 and 0.90, respectively. Seven of the 11 genotypes revealed from chicken isolates from MS are newly described genotypes and six of them each have a single isolate. In conclusion, the results obtained from isolates in MS corroborate previous studies on T. gondii isolates in Brazil, thus confirming their diversity and atypicality. Toxoplasma gondii isolates are highly diverse in domestic animals from Brazil. However, little is known about the genetics of this parasite from wild mammals in the same region. Reveal genetic similarity or difference of T. gondii among different animal populations is necessary for us to understand transmission of this parasite. Here we reported isolation and genetic characterisation of three T. gondii isolates from wild animals in Brazil. The parasite was isolated by bioassay in mice from tissues of a young male red handed howler monkey (Alouatta belzebul), an adult male jaguarundi (Puma yagouaroundi), and an adult female black-eared opossum (Didelphis aurita). The monkey and the jaguarundi had inhabited the Zoo of Parque Estadual Dois Irmaos, Pernambuco State, Northeastern Brazil, for 1 year and 8 years, respectively. The wild black-eared opossum was captured in Sao Paulo State, Southeastern Brazil, and euthanised for this study because it was seropositive for T. gondii (titre 1:100 by the modified agglutination test, MAT). Ten PCR-RFLP (Polymerase Chain Reaction-Restriction Fragment Length Polymorphism) markers, SAG1, SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1 and Apico, were used to genotype the isolates. T. gondii was isolated from the brain and heart homogenate of the monkey, the muscle homogenate of the jaguarundi, and the heart homogenate of the black-eared opossum. This was the first isolation of T. gondii from a neotropical fetid from Brazil. The isolate from the monkey (TgRhHmBr1) was not virulent in mice, whereas the isolates from the jaguarundi (TgJagBr1) and the black-eared opossum (TgOpBr1) were virulent in mice. The genotype of the isolate from the monkey has been identified in isolates from a goat and ten chickens in the same region of Brazil, suggesting that it may be a common lineage circulating in this region. The genotypes of the isolates from the jaguarundi and the black-eared opossum have not been previously reported. Although there are already 88 genotypes identified from a variety of animal hosts in Brazil, new genotypes are continuously being identified from different animal species, indicating an extremely high diversity of T. gondii in the population. Recent studies indicated that Toxoplasma gondii isolates of many domestic animal hosts from Brazil are genetically and biologically different from those in USA and Europe. Despite of high pathogenicity of this parasite to small ruminants, the epidemiology and genetic diversity of T. gondii in these animals are not well understood in Brazil. In this study, a total of 28 T. gondii samples (16 isolates from sheep in Sao Paulo state, and 12 isolates from goats in the states of Sao Paulo and Rio Grande do Norte) were genotyped using genetic markers SAG1, SAG2, SAG3, BTUB, CRAG, c22-8, c29-2, L358, PK1, Apico and CS3. Eleven genotypes were identified from these T. gondii isolates. Eight isolates (4 from sheep and 4 from goats) were grouped into the common clonal type Brl lineage. One sheep isolate was grouped to the type BrIII lineage. Five isolates grouped to three previously identified genotypes in Brazil, and 13 isolates grouped to six novel genotypes. Mixed genotype was found in one isolate from goat in Sao Paulo. No classical clonal Type I. II or III isolates were found, confirming previous reports that these clonal lineages are rare in Brazil. The allele types at the CS3 locus are strongly linked to mouse virulence of the parasite. The results of this study indicate that even though a large number of T. gondii genotypes have been identified from a variety of animal hosts in Brazil, high percentage of new genotypes are continuously identified from different animal species, suggesting extremely high diversity of T. gondii in the population. The aim of this study was to determine if Toxoplasma gondii are present in oysters (Crassostrea rhizophorae) and mussels (Mytella guyanensis) under natural conditions using a bioassay in mice and molecular detection methods. We first compared two standard protocols for DNA extraction, phenol-chloroform (PC) and guanidine-thiocyanate (GT), for both molluscs. A total of 300 oysters and 300 mussels were then acquired from the fish market in Santos city, Sao Paulo state, Brazil, between March and August of 2008 and divided into 60 groups of 5 oysters and 20 groups of 15 mussels. To isolate the parasite, five mice were orally inoculated with sieved tissue homogenates from each group of oysters or mussels. For molecular detection of T. gondii, DNA from mussels was extracted using the PC method and DNA from oysters was extracted using the GT method. A nested-PCR (Polymerase Chain Reaction) based on the amplification of a 155 bp fragment from the B1 gene of T. gondii was then performed. Eleven PCR-RFLP (Restriction Fragment Length Polymorphism) markers, SAG1, SAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, CS3 and Apico, were used to genotype positive samples. There was no isolation of the parasite by bioassay in mice. T. gondii was not detected in any of the groups of mussels by nested-PCR. DNA of T. gondii was apparently detected by nested-PCR in 2 groups of oysters (3.3%). Genotyping of these two positive samples was not successful. The results suggest that oysters of the species C. rhizophorae, the most common species from the coast of Sao Paulo, can filter and retain T. gondii oocysts from the marine environment. Ingestion of raw oysters as a potential transmission source of T. gondii to humans and marine mammals should be further investigated. (C) 2010 Elsevier B.V. All rights reserved.

Objective: The aim of this study was to investigate the prevalence of the Eosinophil cationic protein (ECP)-gene polymorphism 434(G > C) in oral squamous cell carcinoma (OSCC) patients and its association with tumor-associated tissue eosinophilia (TATE), demographic, clinical, and microscopic variables. Methods: The ECP genotypes of 165 healthy individuals and 157 OSCC patients were detected by PCR-RFLP analysis after cleavage of the amplified DNA sequence with enzyme PstI. TATE was obtained by morphometric analysis. Chi-square test or Fisher`s exact test was used to analyze the association of ECP-gene polymorphism 434(G > C) with TATE, demographic, clinical, and microscopic variables in OSCC patients. Disease-free survival and overall survival were calculated by the Kaplan-Meier product-limit actuarial method and the comparison of the survival curves were performed using log rank test. Results: Most of healthy individuals (53.33%) and OSCC patients (57.97%) were heterozygous for the ECP 434(G > C) polymorphism. Based on numerical differences, our results showed that OSCC patients with intense TATE and at least one C allele had a higher frequency of bilateral neck dissection, local recurrence, vascular embolization, involved resection margins, and postoperative radiotherapy. No statistically significant differences on survival rates were found in OSCC patients presenting different ECP 434(G > C) genotypes. Conclusions: These results suggest a tendency towards a poor clinical outcome in OSCC patients with intense TATE and 434GC/CC genotypes, probably due to an ECP genetic variant with altered cytotoxic activity.

Mosquito collections were made throughout the mainland of Papua New Guinea to identify the members of the Anopheles punctulatus group present and to determine their distribution. Identification was made using morphology, DNA hybridization, and polymerase chain reaction (PCR)-RFLP analysis. Nine members of the group were identified: An. farauti s.s. Laveran, An.farauti 2, An. koliensis Owen, and An. punctulatus Donitz, were common and widespread; An. farauti 4 was restricted to the north of the central ranges where it was common; An. farauti 6 was found only in the highlands above 1,000 m; and An. farauti 3, An. sp. near punctulatus and An. clowi Rozeboom & Knight were uncommon and had restricted distributions. Identification of An. koliensis and An. punctulatus using proboscis morphology was found to be unreliable wherever An. farauti 4 occurred. The distribution and dispersal of the members of the An. punctulatus group is discussed in regard to climate, larval habitats, distance from the coast, elevation, and proximity to human habitation.

Members of the Culex sitiens subgroup are important vectors of arboviruses, including Japanese encephalitis virus, Murray Valley encephalitis virus and Ross River virus. Of the eight described species, Cx. annulirostris Skuse, Cx. sitiens Wiedemann, and Cx. palpalis Taylor appear to be the most abundant and widespread throughout northern Australia and Papua New Guinea (PNG). Recent investigations using allozymes have shown this subgroup to contain cryptic species that possess overlapping adult morphology. We report the development of a polymerase chain reaction-restriction fragment-length polymorphism (PCR-RFLP) procedure that reliably separates these three species. This procedure utilizes the sequence variation in the ribosomal DNA ITS1 and demonstrates species-specific PCR-RFLP profiles from both colony and field collected material. Assessment of the consistency of this procedure was undertaken on mosquitoes sampled from a wide geographic area including Australia, PNG, and the Solomon Islands. Overlapping adult morphology was observed for Cx. annulirostris and Cx. palpalis in both northern Queensland and PNG and for all three species at one site in northwest Queensland.

1. Sulphotransferases are a superfamily of enzymes involved in both detoxification and bioactivation of endogenous and exogenous compounds. The arylsulphotransferase SULT1A1 has been implicated in a decreased activity and thermostability when the wild-type arginine at position 213 of the coding sequence is substituted by a histidine. SULT1A1 is the isoform primarily associated with the conversion of dietary N -OH arylamines to DNA binding adducts and is therefore of interest to determine whether this polymorphism is linked to colorectal cancer. 2. Genotyping, using polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis, was performed using DNA samples of healthy control subjects (n = 402) and patients with histologically proven colorectal cancer (n = 383). Both control and test populations possessed similar frequencies for the mutant allele (32.1 and 31%, respectively; P = 0.935). Results were not altered when age and gender were considered as potential confounders in a logistic regression analysis. 3. Examination of the sulphonating ability of the two allozymes with respect to the substrates p -nitrophenol and paracetamol showed that the affinity and rate of sulphonation was unaffected by substitution of arginine to histidine at position 213 of the amino acid sequence. 4. From this study, we conclude that the SULT1A1 R213H polymorphism is not linked with colorectal cancer in this elderly Australian population.

Diferenças na susceptibilidade do hospedeiro à infecção, na gravidade e na permanência do quadro clínico da doença podem ser atribuídas, em parte, às variações da resposta imune. Estas variações são associadas a polimorfismos de nucleotídeo único (do inglês: single nucleotide polymorphisms - SNPs). Como estudo prévio, foi realizada a caracterização da população geral do Espírito Santo (ES) - Brasil e de uma subpopulação do estado, de origem Pomerana, quanto aos SNPs -131 H/R, -336 A/G, TaqI, -308 A/G, -590 C/T, -174 G/C e +874 A/T nos genes FcγRIIa, CD209, VDR, TNFα, IL-4, IL-6 e INF-γ, respectivamente. Cem indivíduos da Grande Vitória representaram a população geral do ES e 59 indivíduos de Santa Maria de Jetibá representaram a população de origem Pomerana. Como a fase aguda da dengue é bem caracterizada, este estudo objetivou ampliar o conhecimento da fase de convalescença. Noventa e seis indivíduos diagnosticados com dengue sintomática no final de 2012 e início de 2013, no ES, foram acompanhados por 60 dias a partir do início dos sintomas por meio do preenchimento de um questionário clínico e epidemiológico em quatro entrevistas. A persistência de 37 sintomas clínicos da dengue foi avaliada. Para analisar a influência da genética do sistema imunológico do hospedeiro na persistência de sintomas clínicos da dengue na fase de convalescença, foi determinada a associação entre os sete SNPs, para os quais a população do ES foi caracterizada, e a persistência de sintomas. O DNA genômico dos participantes do estudo foi extraído do sangue periférico e a genotipagem dos SNPs foi realizada por reação em cadeia da polimerase - polimorfismo de comprimento de fragmento de restrição (do inglês: polymerase chain reaction - restriction fragment length polymorphism - PCR-RFLP) As frequências genotípicas de todos os SNPs encontraram-se em equilíbrio de Hardy-Weinberg (do inglês: Hardy-Weinberg equilibrium - HWE), com exceção do SNP no gene IL-6. Não houve diferença estatisticamente significante nas frequências genotípicas dos SNPs nos genes FcγRIIa, CD209, VDR, TNF-α e IL-4 entre as duas populações. Diferença estatisticamente significante foi encontrada entre as duas populações nas distribuições genotípicas dos SNPs nos genes IL-6 (p = 0,03) e INF-γ (p = 0,007). Trinta e sessenta dias após o início dos sintomas, 38,5% e 11,5% dos indivíduos com dengue sintomática reportaram ter pelo menos um sintoma clínico da dengue, respectivamente. Dos sintomas analisados, os mais persistentes foram os relacionados à síndrome da fadiga como mialgia, artralgia, astenia e mal-estar, sendo a mialgia o mais frequente. A persistência de sintomas em 30 dias foi associada ao gênero feminino (p = 0,044) e a persistência de sintomas constitucionais foi associada à dengue secundária (p = 0,041). O SNP no gene FcγRIIa, foi associado à persistência de sintomas em 30 dias, no subgrupo de indivíduos com dengue secundária (p = 0,046), sendo a presença do alelo H associada à não persistência de sintomas (p = 0,014). A presença do alelo A do SNP no gene TNF-α foi associada à não persistência de sintomas no subgrupo de indivíduos com dengue secundária (p = 0,025), sendo o genótipo GG associado à persistência de sintomas neurológicos, psicológicos e comportamentais em 30 dias (p = 0,038). A presença do alelo C do SNP no gene IL-6 foi associado à persistência de sintomas dermatológicos em 30 dias (p = 0,005). O perfil genético desses SNPs pode favorecer o estabelecimento de marcadores imunogenéticos associados à fase convalescente da infecção pelo vírus da dengue (do inglês: dengue virus - DENV).

Acute physical exercise is associated with increased oxygen consumption, which could result in an increased formation of reactive oxygen species (ROS). ROS can react with several organic structures, namely DNA, causing strand breaks and a variety of modified bases in DNA. Physical exercise training seems to decrease the incidence of oxidative stress-associated diseases, and is considered as a key component of a healthy lifestyle. This is a result of exercise-induced adaptation, which has been associated with the possible increase in antioxidant activity and in oxidative damage repair enzymes, leading to an improved physiological function and enhanced resistance to oxidative stress (Radak et al. 2008). Human 8-oxoguanine DNA glycosylase 1 (hOGG1) is involved in the base excision repair (BER) pathway and encodes an enzyme responsible for removing the most common product of oxidative damage in DNA, 8-hydroxyguanine (8-OH-G). The genetic polymorphism of hOGG1 at codon 326 results in a serine (Ser) to cysteine (Cys) amino acid substitution (Ser326Cys). It has been suggested that the carriers of at least one hOGG1Cys variant allele exhibit lower 8-OH-G excision activity than the wild-type (Wilson et al. 2011). The aim of this study was to investigate the possible influence of hOGG1 Ser326Cys polymorphism on DNA damage and repair activity in response to 16 weeks of combined physical exercise training, in thirty healthy Caucasian men. Comet assay was carried out using peripheral blood lymphocytes and enabled the evaluation of DNA damage, both strand breaks and FPG-sensitive sites, and DNA repair activity. Genotypes were determined by PCR-RFLP analysis. The subjects with Ser/Ser genotype were considered as wild-type group (n=20), Ser/Cys and Cys/Cys genotype were analyzed together as mutant group (n=10). Regarding differences between pre and post-training in the wild-type group, the results showed a significant decrease in DNA strand breaks (DNA SBs) (p=0.002) and also in FPG-sensitive sites (p=0.017). No significant differences were observed in weight (p=0.389) and in lipid peroxidation (MDA) (p=0.102). A significant increase in total antioxidant capacity (evaluated by ABTS) was observed (p=0.010). Regarding mutant group, the results showed a significant decrease in DNA SBs (p=0.008) and in weight (p=0.028). No significant differences were observed in FPG-sensitive sites (p=0.916), in ABTS (p=0.074) and in MDA (p=0.086). No significant changes in DNA repair activity were observed in both genotype groups. This preliminary study suggests the possibility of different responses in DNA damage to physical exercise training, considering the hOGG1 Ser326Cys polymorphism.

As leveduras são fungos oportunistas responsáveis pela maior parte das infecções fúngicas nos seres humanos. Este tipo de infecções é mais comum em indivíduos com o sistema imunitário comprometido e têm vindo a aumentar ao longo dos anos. A espécie Candida albicans é a mais frequentemente identificada, como sendo responsável por este tipo de infecções, no entanto, o número de infecções provocadas por outras espécies do género Candida ocorre cada vez com mais frequência. As infecções hospitalares fúngicas constituem uma causa crescente de morbilidade e mortalidade em hospitais, afectando tanto doentes internados, como profissionais de saúde. Do ponto de vista etiológico, a grande maioria das infecções fúngicas hospitalares é causada por espécies do género Candida, principalmente Candida albicans, C. parapsilosis, C. tropicalis e C.glabrata. A sua identificação taxonómica, geralmente exige o seu isolamento inicial em meios de cultura, a realização de provas bioquímicas de assimilação in vitro, com a utilização de “kits” comerciais, ou a sua repicagem para meios cromogénios. O principal objectivo deste trabalho foi a identificação rápida e eficaz de candidoses invasivas através de uma metodologia molecular de diagnóstico que fosse simples e fácil de implementar em laboratórios de diagnóstico microbiológico. Para tal, foram seleccionados 100 isolados clínicos de leveduras obtidas a partir de amostras clínicas enviadas para o Laboratório de Patologia Clínica do Centro Hospitalar Cova da Beira E.P.E. para o diagnóstico laboratorial de infecção fúngica durante um período de 8 meses, desde Novembro de 2008 até Junho de 2009. O trabalho baseou-se na identificação molecular por PCR-RFLP de espécies do género Candida, e os resultados foram comparados com os obtidos pelos métodos de diagnóstico tradicionais (CHROMagar® Candida e VITEK® - bioMérieux) utilizados no laboratório hospitalar. Foi ainda efectuado o estudo da sensibilidade in vitro de espécies do género Candida aos antifúngicos fluconazol, voriconazol, através dos métodos, de difusão em disco e E-Test®, segundo os procedimentos padronizados e publicados pelo CLSI, tendo-se verificado elevada sensibilidade dos isolados para ambos os fármacos. Com este trabalho concluiu-se que a eficiente e rápida identificação dos fungos clinicamente relevantes por parte dos laboratórios de patologia clínica deve ser uma tarefa fundamental para o controle das infecções. A identificação ao nível da espécie é importante para determinar a etiologia da infecção, para detectar novos agentes da doença, para prever resistências intrínsecas a agentes antifúngicos e para detectar causas de infecções nosocomiais. Face ao exposto, os estudos epidemiológicos são de extrema importância, assim como o diagnóstico das infecções fúngicas. O diagnóstico das infecções fúngicas continua a ser efectuado por métodos tradicionais, que avaliam características fisiológicas e bioquímicas dos elementos fúngicos, mas que apesar de serem eficazes são, na sua maioria, demoradas impedindo um início rápido e atempado da terapêutica. Como tal, os métodos moleculares podem constituir uma alternativa mais viável ao diagnóstico micológico, nomeadamente de leveduras do género Candida, como se pode comprovar pelos resultados obtidos neste trabalho.

As micoses estão incluídas entre as doenças infecciosas mais ubíquas em todo o mundo, afectando todos os estratos sociais e todos os grupos etários numa variedade de manifestações superficiais, cutâneas, subcutâneas e sistémicas. Um diagnóstico rápido e eficaz destas doenças, com uma correcta identificação da espécie fúngica responsável pela infecção, é essencial para um planeamento do tratamento mais eficaz para o doente infectado. Tem-se registado um aumento da incidência das infecções fúngicas também em consequência do aumento do número de casos de doentes imunodeprimidos, devido particularmente à crescente utilização de terapêuticas imunossupressivas, procedimentos médicos invasivos, prescrição de tratamentos prolongados, entre outros aspectos. O aparecimento da epidemia da Imunodeficiência Humana no início da década de 80, causada pelo vírus VIH, contribuiu também decisivamente para o aumento das infecções fúngicas oportunistas. A Criptococose é uma infecção fúngica, predominantemente oportunista e com uma distribuição epidemiológica mundial, causada por leveduras encapsuladas do género Cryptococcus. A espécie clinicamente mais relevante é Cryptococcus neoformans, cujas estirpes têm sido tradicionalmente classificadas em cinco serotipos relacionados com os antigénios da respectiva cápsula polissacárida: A (C. neoformans var. grubii); D (C. neoformans var. neoformans); B e C (actualmente reconhecidos como uma espécie distinta mas filogeneticamente próxima, C. gattii); e AD (estirpes híbridas). O presente trabalho teve como principal objectivo determinar retrospectivamente os tipos moleculares de uma colecção alargada de estirpes de C. neoformans, isoladas e mantidas durante os últimos 18 anos no Laboratório de Micologia do IHMT/UNL. A maioria das estirpes foi isolada de doentes imunodeprimidos com criptococose, existindo também algumas estirpes de origem ambiental. Foi utilizada a técnica de PCR-RFLP do gene URA5 para diferenciar as estirpes de Cryptococcus neoformans, tendo sido detectados quatro tipos moleculares: VN1 e VN2 (relacionados com C. neoformans var. grubii, serotipo A); VN3 (relacionado com as estirpes híbridas de serotipo AD); e VN4 (relacionado com C. neoformans var. neoformans, serotipo D). Não foram encontrados entre os isolados de origem clínica perfis de restrição correspondentes aos tipos moleculares VG1, VG2, VG3 e VG4 (relacionados com C. gattii, serotipos B e C). O tipo molecular VN1 foi o mais abundante entre os isolados de origem clínica (45% dos isolados), seguindo-se o grupo de estirpes híbridas do tipo molecular VN3 (31%). Os tipos moleculares menos abundantes entre os isolados foram o VN2 (12%) e VN4 (12%). A estandardização do método de tipagem molecular utilizado neste trabalho permite comparar os resultados obtidos com os de outros estudos epidemiológicos semelhantes, realizados noutras regiões do globo e publicados em anos recentes, contribuindo para um conhecimento melhorado da epidemiologia global deste importante fungo patogénico. Em Portugal obteve-se uma percentagem mais elevada de isolados dos grupos moleculares VN1 e VN3 em relação a outros países da Europa e América Latina, em que os tipos mais abundantes são VN1 e VN2. Nestas mesmas regiões, o tipo molecular VN4, relacionado com as estirpes de C. neoformans var. neoformans do serotipo D, é muito raro. Esta variedade é mais comum em zonas mediterrâneas e está muito associada a casos clínicos de infecções cutâneas associadas a infecções do Sistema Nervoso Central. Este tipo molecular parece ser também significativamente mais abundante no nosso país.

In the present study we report the results of an analysis, based on ribotyping of Corynebacterium diphtheriae intermedius strains isolated from a 9 years old child with clinical diphtheria and his 5 contacts. Quantitative analysis of RFLPs of rRNA was used to determine relatedness of these 7 C.diphtheriae strains providing support data in the diphtheria epidemiology. We have also tested those strains for toxigenicity in vitro by using the Elek's gel diffusion method and in vivo by using cell culture method on cultured monkey kidney cell (VERO cells). The hybridization results revealed that the 5 C.diphtheriae strains isolated from contacts and one isolated from the clinical case (nose case strain) had identical RFLP patterns with all 4 restriction endonucleases used, ribotype B. The genetic distance from this ribotype and ribotype A (throat case strain), that we initially assumed to be responsible for the illness of the patient, was of 0.450 showing poor genetic correlation among these two ribotypes. We found no significant differences concerned to the toxin production by using the cell culture method. In conclusion, the use of RFLPs of rRNA gene was successful in detecting minor differences in closely related toxigenic C.diphtheriae intermedius strains and providing information about genetic relationships among them.

RESUMO O presente estudo teve como principal objectivo avaliar a diversidade genética de uma população parasitária de Leishmania em isolados portugueses de hospedeiros humanos, caninos, vulpinos e do vector, aplicando dois marcadores moleculares: kDNA e microssatélites. No Capítulo 1 fez-se uma revisão bibliográfica sobre as leishmanioses incluindo a epidemiologia da infecção nos países da bacia mediterrânica nomeadamente Portugal. Deu-se especial relevo à epidemiologia molecular que nos últimos anos tem vindo a ser desenvolvida. No Capítulo 2 efectuou-se um inquérito de leishmaniose canina que abrangeu 374 cães provenientes da Região Metropolitana de Lisboa. Foi encontrada uma prevalência total de 19,2%, com a prevalência de 18,4% nos cães com dono e 21,6% nos cães sem dono ou vadios. Os resultados obtidos evidenciaram a importância dos cães vadios na transmissão do parasita e disseminação da doença. A partir dos 72 cães infectados, foram isolados 49 estirpes de Leishmania, tendo estas sido tipadas como L. infantum zimodeme MON-1. Estas estirpes, em conjunto com outras amostras isoladas a partir de humanos, vector e outros canídeos, foram utilizadas para avaliar a diversidade genética. No Capítulo 3 foram desenvolvidas sequências iniciadoras cinetoplastideais, MC1 e MC2, tendo-se estas revelado específicas e sensíveis para a identificação do complexo L. donovani isolados em cultura ou directamente a partir de amostras clínicas. Aplicou-se a metodologia de kDNA-PCR-RFLP na análise de 161 amostras de DNA, das quais 134 eram provenientes de isolados portugueses de L. infantum. Foram identificados 16 genótipos na totalidade das amostras, tendo 13 sido identificados nas amostras portuguesas. Observou-se a predominância do genótipo A, observado exclusivamente na população parasitária portuguesa. Em termos geográficos esta metodologia mostrou estar de acordo com a tipagem isoenzimática, e outros marcadores moleculares, individualizando as amostras provenientes de África num único genótipo. No entanto não se observou individualização ao nível das regiões de Portugal estudadas, sugerindo a existência de fluxo genético entre as diferentes áreas geográficas. No Capítulo 4 aplicou-se a análise de 13 loci de microssatélites, polimórficos para L. infantum, em 154 amostras, das quais 128 eram provenientes de diferentes regiões geográficas de Portugal e de diferentes hospedeiros e vector. Obteve-se um maior grau de polimorfismo com estes marcadores do que com o kDNA, identificando-se 85 genótipos. Observou-se uma maior diversidade molecular nas amostras provenientes do Algarve e Alto Douro e, relativamente ao hospedeiro, estes alvos moleculares mostraram ser muito mais polimórficos no hospedeiro humano que o canino, indo ao encontro dos resultados de tipagem isoenzimática que se conhecem até à actualidade. Foi individualizado um agrupamento de amostras não MON-1 e dentro deste, um sub-agrupamento das amostras de África Oriental (Etiópia e Sudão), como anteriormente sugerido por outros autores. No Capítulo 5 discutiram-se os resultados obtidos permitindo verificar que a variabilidade dos parasitas Leishmania no nosso país é maior do que tem sido considerada até ao presente. Possibilitaram também o conhecimento de que há genótipos predominantes em Portugal e que a variabilidade genética no hospedeiro humano e no vector é superior à do reservatório doméstico e silvático.
