BACKGROUND: basic calcium phosphate (BCP) crystals are commonly found in osteoarthritis (OA) and are associated with cartilage destruction. BCP crystals induce in vitro catabolic responses with the production of metalloproteases and inflammatory cytokines such as interleukin-1 (IL-1). In vivo, IL-1 production induced by BCP crystals is both dependant and independent of NLRP3 inflammasome. We aimed to clarify 1/ the role of BCP crystals in cartilage destruction and 2/ the role of IL-1 and NLRP3 inflammasome in cartilage degradation related to BCP crystals. METHODOLOGY PRINCIPAL FINDINGS: synovial membranes isolated from OA knees were analysed by alizarin Red and FTIR. Pyrogen free BCP crystals were injected into right knees of WT, NLRP3 -/-, ASC -/-, IL-1α -/- and IL-1β-/- mice and PBS was injected into left knees. To assess the role of IL-1, WT mice were treated by intra-peritoneal injections of anakinra, the IL-1Ra recombinant protein, or PBS. Articular destruction was studied at d4, d17 and d30 assessing synovial inflammation, proteoglycan loss and chondrocyte apoptosis. BCP crystals were frequently found in OA synovial membranes including low grade OA. BCP crystals injected into murine knee joints provoked synovial inflammation characterized by synovial macrophage infiltration that persisted at day 30, cartilage degradation as evidenced by loss of proteoglycan staining by Safranin-O and concomitant expression of VDIPEN epitopes, and increased chondrocyte apoptosis. BCP crystal-induced synovitis was totally independent of IL-1α and IL-1β signalling and no alterations of inflammation were observed in mice deficient for components of the NLRP3-inflammasome, IL-1α or IL-1β. Similarly, treatment with anakinra did not prevent BCP crystal effects. In vitro, BCP crystals elicited enhanced transcription of matrix degrading and pro-inflammatory genes in macrophages. CONCLUSIONS SIGNIFICANCE: intra-articular BCP crystals can elicit synovial inflammation and cartilage degradation suggesting that BCP crystals have a direct pathogenic role in OA. The effects are independent of IL-1 and NLRP3 inflammasome.

O objetivo deste trabalho foi avaliar o efeito da suplementação alimentar com gordura protegida ruminal sobre as estruturas ovarianas e sobre a concentração sérica de progesterona, em novilhas Nelore mantidas em pasto. Quarenta novilhas foram divididas em dois grupos: um suplementado com gordura protegida Megalac-E (G); e outro sem suplementação de gordura (C). O grupos foram avaliados em delineamento "crossover". Utilizaram-se dietas isoenergéticas e isoproteicas. Após 15 dias de suplementação, os animais foram submetidos a um protocolo hormonal, para avaliação da influência da suplementação com gordura no metabolismo da progesterona. Para isto, em um dia aleatório do ciclo (D0), inseriu-se um implante intravaginal de liberação de progesterona (CIDR), e aplicou-se prostaglandina F2α (PGF2α, i.m.). No D7, o implante foi retirado, e outra aplicaηão de PGF2α foi realizada. No D18, foi feita uma nova aplicaηão de PGF2α e, então, foram observados diariamente os exames ultrassonográficos ovarianos e a ocorrência de estro. Para o ensaio com progesterona, colheu-se sangue 4 dias após a inserção do implante e, novamente, 7 e 14 dias após a ovulação. A concentração de progesterona sérica no D4 foi maior no grupo G. Não houve diferença nas concentrações séricas de progesterona 7 e 14 dias após a ovulação, nem no diâmetro do folículo ovulatório, nem no volume luteal. A suplementação com Megalac-E altera o metabolismo de progesterona, mas não altera a função ovariana em novilhas zebuínas em pasto.

Citalopram, a new bicyclic antidepressant, is the most selective serotonin reuptake inhibitor. In a number of double-blind controlled studies, citalopram was compared to placebo and to known tricyclic antidepressants. These studies have shown their efficacy and good safety. The inefficacy of a psychotropic treatment in at least 20% of depressives has led a number of authors to propose original drug combinations and associations, like antidepressant/lithium (Li), antidepressant/sleep deprivation (agrypnia), antidepressant/ECT, or antidepressant/LT3. The aim of this investigation is to evaluate the clinical effectiveness and safety of a combined citalopram/lithium treatment in therapy-resistant patients, taking account of serotonergic functions, as tested by the fenfluramine/prolactin test, and of drug pharmacokinetics and pharmacogenetics of metabolism. DESIGN OF THE STUDY: A washout period of 3 days before initiating the treatment is included. After an open treatment phase of 28 days (D) with citalopram (20 mg D1-D3; 40 mg D4-D14; 40 or 60 mg D15-D28; concomitant medication allowed: chloral, chlorazepate), the nonresponding patients [less than 50% improvement in the total score on the 21 item-Hamilton Depression Rating Scale (HDRS)] are selected and treated with or without Li (randomized in double-blind conditions: citalopram/Li or citalopram/placebo) during the treatment (D29-D35). Thereafter, all patients included in the double-blind phase subsequently receive an open treatment with citalopram/Li for 7 days (D36-D42). The hypothesis of a relationship between serotoninergic functions in patients using the fenfluramine/prolactin test (D1) and the clinical response to citalopram (and Li) is assessed. Moreover, it is evaluated whether the pharmacogenetic status of the patients, as determined by the mephenytoin/dextromethorphan test (D0-D28), is related to the metabolism of fenfluramine and citalopram, and also to the clinical response. CLINICAL ASSESSMENT: Patients with a diagnosis of major depressive disorders according to DSM III are submitted to a clinical assessment of D1, D7, D14, D28, D35, D42: HDRS, CGI (clinical global impression), VAS (visual analog scales for self-rating of depression), HDRS (Hamilton depression rating scale, 21 items), UKU (side effects scale), and to clinical laboratory examens, as well as ECG, control of weight, pulse, blood pressure at D1, D28, D35. Fenfluramine/prolactin test: A butterfly needle is inserted in a forearm vein at 7 h 45 and is kept patent with liquemine. Samples for plasma prolactin, and d- and l-fenfluramine determinations are drawn at 8 h 15 (base line). Patients are given 60 mg fenfluramine (as a racemate) at 8 h 30. Kinetic points are determined at 9 h 30, 10 h 30, 11 h 30, 12 h 30, 13 h 30. Plasma levels of d- and l-fenfluramine are determined by gas chromatography and prolactin by IRNA. Mephenytoin/dextromethorphan test: Patients empty their bladders before the test; they are then given 25 mg dextropethorphan and 100 mg mephenytoin (as a racemate) at 8 h 00. They collect all urines during the following 8 hours. The metabolic ratio is determined by gas chromatography (metabolic ratio dextromethorphan/dextrorphan greater than 0.3 = PM (poor metabolizer); mephenytoin/4-OH-mephenytoin greater than 5.6, or mephenytoin S/R greater than 0.8 = PM). Citalopram plasma levels: Plasma levels of citalopram, desmethylcitalopram and didesmethylcitalopram are determined by gas chromatography--mass spectrometry. RESULTS OF THE PILOT STUDY. The investigation has been preceded by a pilot study including 14 patients, using the abovementioned protocol, except that all nonresponders were medicated with citalopram/Li on D28 to D42. The mean total score (n = 14) on the 21 item Hamilton scale was significantly reduced after the treatment, ie from 26.93 +/- 5.80 on D1 to 8.57 +/- 6.90 on D35 (p less than 0.001). No Brasil, o uso do trifoliata 'Flying Dragon', como porta-enxerto em citros, tem crescido quando se buscam pomares com altas densidades, devido, principalmente, a induzir nanismo à maioria das variedades de copa compatíveis. No entanto, por possuir desenvolvimento lento, existe a necessidade de se estudar melhor o crescimento desse porta-enxerto. A nutrição é uma importante ferramenta que visa a acelerar o crescimento e a melhorar o vigor e a qualidade das plantas. Diante disso, foi avaliada a aplicação de nutrientes no crescimento do porta-enxerto em três experimentos. No primeiro experimento, utilizaram-se seis doses de N parceladas; no segundo, seis doses de P (aplicadas em dose total, misturadas diretamente no substrato); e no terceiro, seis doses de K parceladas. As seis doses foram constituídas por: D0 = testemunha sem adubação; D1= metade da dose-padrão; D2 = a dose-padrão (920 mg dm-3 de N, 790 mg dm-3 de K e 100 mg dm-3 de P); D3 = uma vez e meia a dose-padrão; D4 = duas vezes a dose-padrão; e D5 = duas vezes e meia a dose-padrão. Assim, coletaram-se dados biométricos da planta de diâmetro e altura, quinzenalmente, e dados de massa seca da parte aérea e da raiz, mensalmente. O delineamento utilizado foi o inteiramente casualizado, com parcela subdividida. O nitrogênio teve papel fundamental no incremento das massas secas de raiz e da parte aérea.

Background Dopamine is believed to be a key neurotransmitter in the development of attention-deficit/ hyperactivity disorder (ADHD). Several recent studies point to an association of the dopamine D4 receptor (DRD4) gene and this condition. More specifically, the 7 repeat variant of a variable number of tandem repeats (VNTR) polymorphism in exon III of this gene is suggested to bear a higher risk for ADHD. In the present study, we investigated the role of this polymorphism in the modulation of neurophysiological correlates of response inhibition (Go/Nogo task) in a healthy, high-functioning sample. Results Homozygous 7 repeat carriers showed a tendency for more accurate behavior in the Go/Nogo task compared to homozygous 4 repeat carriers. Moreover, 7 repeat carriers presented an increased nogo-related theta band response together with a reduced go-related beta decrease. Conclusions These data point to improved cognitive functions and prefrontal control in the 7 repeat carriers, probably due to the D4 receptor's modulatory role in prefrontal areas. The results are discussed with respect to previous behavioral data on this polymorphism and animal studies on the impact of the D4 receptor on cognitive functions.

The immune system is involved in the development of neuropathic pain. In particular, the infiltration of T-lymphocytes into the spinal cord following peripheral nerve injury has been described as a contributor to sensory hypersensitivity. We used the spared nerve injury (SNI) model of neuropathic pain in Sprague Dawley adult male rats to assess proliferation, and/or protein/gene expression levels for microglia (Iba1), T-lymphocytes (CD2) and cytotoxic T-lymphocytes (CD8). In the dorsal horn ipsilateral to SNI, Iba1 and BrdU stainings revealed microglial reactivity and proliferation, respectively, with different durations. Iba1 expression peaked at D4 and D7 at the mRNA and protein level, respectively, and was long-lasting. Proliferation occurred almost exclusively in Iba1 positive cells and peaked at D2. Gene expression observation by RT-qPCR array suggested that T-lymphocytes attracting chemokines were upregulated after SNI in rat spinal cord but only a few CD2/CD8 positive cells were found. A pronounced infiltration of CD2/CD8 positive T-cells was seen in the spinal cord injury (SCI) model used as a positive control for lymphocyte infiltration. Under these experimental conditions, we show early and long-lasting microglia reactivity in the spinal cord after SNI, but no lymphocyte infiltration was found.

Synthetic methods used for the preparation of azaindoles are described in this article. Applications in the preparation of bioactive molecules are given: synthesis of substituted 6-azaindoles as benzodiazepines receptor ligands, substituted 7-azaindoles as dopamine D4 ligands and preparation of an olivacine analogue.

The understanding of the scientific basis of the erectile function expanded rapidly the range of therapies for treating erectile dysfunction in recent years. This article reviews the role of dopamine on the erection mechanisms and its importance for new pro-erectile drug design. The ability of dopaminergic agents to elicit penile erection has been described since 1975 and successively confirmed by numerous studies. The development of apomorphine SL (dopaminergic non selective agonist) to enhance erectile function represents a new pharmacological approach to the management of erectile dysfunction using CNS drugs. The search for selective D4 dopaminergic agents is being explored by some research groups and pharmaceutical companies.
