Despite the resistance developed by the Mycobacterium tuberculosis (MTb) strains, isoniazid (INK) has been recognized as one of the best drug for treatment of Tuberculosis (Tb). The coordination of INH to ruthenium metal centers was investigated as a strategy to enhance the activity of this drug against the sensitive and resistant strains of MTb. The complexes trans-[Ru(NH3)(4)(L)(INH)](2+) (L = SO2 or NH3) were isolated and their chemical and antituberculosis properties studied. The minimal inhibitory concentration (MIC) data show that [Ru(NH3)(5)(INH)](2+) was active in both resistant and sensitive strains, whereas free INK (non-coordinated) showed to be active only against the sensitive strain. The coordination of INH to the metal center in both [Ru(NH3)(5)(INH)](2+) and trans-[Ru(NH3)(4)(SO2)(INH)](2+) complexes led to a shift in the INH oxidation potential to less positive values compared to free INH. Despite, the ease of oxidation of INH did not lead to an increase in the in vitro INH activity against MTb, it might have provided sensitivity toward resistant strains. Furthermore, ruthenium complexes with chemical structures analogous to those described above were synthesized using the oxidation products of INK as ligands (namely, isonicotinic acid and isonicotinamide). These last compounds were not active against any strains of MTb. Moreover, according to DFT calculations the formation of the acyl radical, a proposed intermediate in the INH oxidation, is favored in the [Ru(NH3)(5)(INH)](2+) complex by 50.7 kcal mol(-1) with respect to the free INH. This result suggests that the stabilization of the acyl radical promoted by the metal center would be a more important feature than the oxidation potential of the INH for the antituberculosis activity against resistant strains. A thin-layer electrochemical flow cell coupled to capillary electrophoresis with contactless conductivity detection (EC-CE-(CD)-D-4) was applied for the first time to the derivatization and quantification of neutral species using aliphatic alcohols as model compounds. The simultaneous electrooxidation of four alcohols (ethanol, 1-propanol, 1-butanol, and 1-pentanol) to the corresponding carboxylates was carried out on a platinum working electrode in acid medium. The derivatization step required 1 min at 1.6 V vs. Ag/AgCl under stopped flow conditions, which was preceded by a 10 s activation at 0 V. The solution close to the electrode surface was then hydrodynamically injected into the capillary, and a 2.5 min electrophoretic separation was carried out. The fully automated flow system operated at a frequency of 12 analyses per hour. Simultaneous determination of the four alcohols presented detection limits of about 5 x 10(-5) mol As a practical application with a complex matrix, ethanol concentrations were determined in diluted pale lager beer and in nonalcoholic beer. No statistically significant difference was observed between the EC-CE-(CD)-D-4 and gas chromatography with flame ionization detection (GC-FID) results for these samples. The derivatization efficiency remained constant over several hours of continuous operation with lager beer samples (n = 40).

This work describes the covalent immobilization of an ironporphyrin, 5,10,15,20-tetrakis(pentafluorophenyl)porphyrin iron(III) chloride (FeTFPP), onto maghemite/silica magnetic nanospheres covered with aminofunctionalized silica. The resulting material (gamma-Fe2O3/SiO2-NHFeP) was characterized by diffuse reflectance infrared spectroscopy (DRIFTS) and UV-Vis absorption spectroscopy. The catalytic activity of this magnetic ironporphyrin was investigated in the oxidation of hydrocarbons (styrene, (Z)-cyclooctene and R-(+)-limonene) and an herbicide (simazine) by hydrogen peroxide or 3-chloroperoxybenzoic acid. Hydrocarbon and simazine oxidation reaction products were analyzed by gas chromatography (GC) and high performance liquid chromatography (HPLC), respectively. This catalytic system proved to be efficient and selective for hydrocarbon oxidation, leading to high product yields from styrene (89%), cyclooctene (71%) and R-(+) -limonene (86%). Simazine oxidation was attained with 100% selectivity for a dechlorinated product (OEAT), while several oxidation products were obtained for the same catalyst in homogeneous media. The catalyst can be easily recovered through application of an external magnetic field and washed after reaction. Catalyst reuse experiments for R-(+)-limonene oxidation have shown that the catalytic activity is kept at 90% after 10 consecutive reactions.

A study of the interference of Zn2+ ions on phenol degradation by Fenton reaction (Fe2+/Fe3(+) + H2O2) is reported. One of the first intermediates formed in the reaction, catechol, can reduce Fe3+ to Fe2+ and, in the presence of H2O2 initiates an efficient catalytic redox cycle. In the initial stages of the reaction, this catechol-mediated cycle becomes the principal route of thermal degradation of phenol and its oxidation products. The Zn2+ ion addition enhances the persistence time of catechol, probably by stabilization of the corresponding semiquinone radical via complexation.

Our aim was to investigate and determine the associations between oxidative stress (OS), dyslipidemia and inflammation in patients treated with continuous ambulatory peritoneal dialysis (CAPD) and hemodialysis (HD) using observational cross-sectional study. Twenty patients in CAPD and 48 in HD for at least 8 weeks and aged =18 years were included in the study. Individuals with malignant or acute inflammatory disease were excluded. A control group of 17 healthy individuals was also recruited. The biochemical parameter evaluations were analyzed using colorimetric kits for albumin, serum glucose, total cholesterol (TC) and lipid fractions. To determine the inflammatory status, CRP, IL-6 and TNF-a were analyzed by automated chemiluminescence kits. Plasma advanced oxidation protein products (AOPP) were determined by spectrophotometry. Mean AOPP levels were significantly higher for the HD group compared to the control, and there was no difference in AOPP concentrations between the control and CAPD groups. Dialysis patients had levels of inflammatory parameters higher than controls, and showed a high prevalence of patients with dyslipidemia, especially in CAPD. In the HD group, AOPP was positively correlated with triglycerides (TG) and inversely associated with HDL. Also the HD group was observed to have negative associations between TNF-a and HDL, LDL and TC. In the CAPD group, CRP was inversely correlated with HDL. Hemodialysis patients had increased protein OS and associations of inflammation and dyslipidemia were also observed in these dialysis groups. A more detailed characterization of the relations between oxidative stress and other more traditional risk factors has therapeutic importance, since cardiovascular diseases are the leading cause of death among dialysis patients.

Modifications in low-density lipoprotein (LDL) have emerged as a major pathogenic factor of atherosclerosis, which is the main cause of morbidity and mortality in the western world. Measurements of the heat diffusivity of human LDL solutions in their native and in vitro oxidized states are presented by using the Z-Scan (ZS) technique. Other complementary techniques were used to obtain the physical parameters necessary to interpret the optical results, e. g., pycnometry, refractometry, calorimetry, and spectrophotometry, and to understand the oxidation phase of LDL particles. To determine the sample's thermal diffusivity using the thermal lens model, an iterative one-parameter fitting method is proposed which takes into account several characteristic ZS time-dependent and the position-dependent transmittance measurements. Results show that the thermal diffusivity increases as a function of the LDL oxidation degree, which can be explained by the increase of the hydroperoxides production due to the oxidation process. The oxidation products go from one LDL to another, disseminating the oxidation process and caring the heat across the sample. This phenomenon leads to a quick thermal homogenization of the sample, avoiding the formation of the thermal lens in highly oxidized LDL solutions. Nowadays, it is clear that the target of creating a sustainable future for the next generations requires to re-think the industrial application of chemistry. It is also evident that more sustainable chemical processes may be economically convenient, in comparison with the conventional ones, because fewer by-products means lower costs for raw materials, for separation and for disposal treatments; but also it implies an increase of productivity and, as a consequence, smaller reactors can be used. In addition, an indirect gain could derive from the better public image of the company, marketing sustainable products or processes. In this context, oxidation reactions play a major role, being the tool for the production of huge quantities of chemical intermediates and specialties. Potentially, the impact of these productions on the environment could have been much worse than it is, if a continuous efforts hadn’t been spent to improve the technologies employed. Substantial technological innovations have driven the development of new catalytic systems, the improvement of reactions and process technologies, contributing to move the chemical industry in the direction of a more sustainable and ecological approach. The roadmap for the application of these concepts includes new synthetic strategies, alternative reactants, catalysts heterogenisation and innovative reactor configurations and process design. Actually, in order to implement all these ideas into real projects, the development of more efficient reactions is one primary target. Yield, selectivity and space-time yield are the right metrics for evaluating the reaction efficiency. In the case of catalytic selective oxidation, the control of selectivity has always been the principal issue, because the formation of total oxidation products (carbon oxides) is thermodynamically more favoured than the formation of the desired, partially oxidized compound. As a matter of fact, only in few oxidation reactions a total, or close to total, conversion is achieved, and usually the selectivity is limited by the formation of by-products or co-products, that often implies unfavourable process economics; moreover, sometimes the cost of the oxidant further penalizes the process. During my PhD work, I have investigated four reactions that are emblematic of the new approaches used in the chemical industry. In the Part A of my thesis, a new process aimed at a more sustainable production of menadione (vitamin K3) is described. The “greener” approach includes the use of hydrogen peroxide in place of chromate (from a stoichiometric oxidation to a catalytic oxidation), also avoiding the production of dangerous waste. Moreover, I have studied the possibility of using an heterogeneous catalytic system, able to efficiently activate hydrogen peroxide. Indeed, the overall process would be carried out in two different steps: the first is the methylation of 1-naphthol with methanol to yield 2-methyl-1-naphthol, the second one is the oxidation of the latter compound to menadione. The catalyst for this latter step, the reaction object of my investigation, consists of Nb2O5-SiO2 prepared with the sol-gel technique. The catalytic tests were first carried out under conditions that simulate the in-situ generation of hydrogen peroxide, that means using a low concentration of the oxidant. Then, experiments were carried out using higher hydrogen peroxide concentration. The study of the reaction mechanism was fundamental to get indications about the best operative conditions, and improve the selectivity to menadione. In the Part B, I explored the direct oxidation of benzene to phenol with hydrogen peroxide. The industrial process for phenol is the oxidation of cumene with oxygen, that also co-produces acetone. This can be considered a case of how economics could drive the sustainability issue; in fact, the new process allowing to obtain directly phenol, besides avoiding the co-production of acetone (a burden for phenol, because the market requirements for the two products are quite different), might be economically convenient with respect to the conventional process, if a high selectivity to phenol were obtained. Titanium silicalite-1 (TS-1) is the catalyst chosen for this reaction. Comparing the reactivity results obtained with some TS-1 samples having different chemical-physical properties, and analyzing in detail the effect of the more important reaction parameters, we could formulate some hypothesis concerning the reaction network and mechanism. Part C of my thesis deals with the hydroxylation of phenol to hydroquinone and catechol. This reaction is already industrially applied but, for economical reason, an improvement of the selectivity to the para di-hydroxilated compound and a decrease of the selectivity to the ortho isomer would be desirable. Also in this case, the catalyst used was the TS-1. The aim of my research was to find out a method to control the selectivity ratio between the two isomers, and finally to make the industrial process more flexible, in order to adapt the process performance in function of fluctuations of the market requirements. The reaction was carried out in both a batch stirred reactor and in a re-circulating fixed-bed reactor. In the first system, the effect of various reaction parameters on catalytic behaviour was investigated: type of solvent or co-solvent, and particle size. With the second reactor type, I investigated the possibility to use a continuous system, and the catalyst shaped in extrudates (instead of powder), in order to avoid the catalyst filtration step. Finally, part D deals with the study of a new process for the valorisation of glycerol, by means of transformation into valuable chemicals. This molecule is nowadays produced in big amount, being a co-product in biodiesel synthesis; therefore, it is considered a raw material from renewable resources (a bio-platform molecule). Initially, we tested the oxidation of glycerol in the liquid-phase, with hydrogen peroxide and TS-1. However, results achieved were not satisfactory. Then we investigated the gas-phase transformation of glycerol into acrylic acid, with the intermediate formation of acrolein; the latter can be obtained by dehydration of glycerol, and then can be oxidized into acrylic acid. Actually, the oxidation step from acrolein to acrylic acid is already optimized at an industrial level; therefore, we decided to investigate in depth the first step of the process. I studied the reactivity of heterogeneous acid catalysts based on sulphated zirconia. Tests were carried out both in aerobic and anaerobic conditions, in order to investigate the effect of oxygen on the catalyst deactivation rate (one main problem usually met in glycerol dehydration). Finally, I studied the reactivity of bifunctional systems, made of Keggin-type polyoxometalates, either alone or supported over sulphated zirconia, in this way combining the acid functionality (necessary for the dehydrative step) with the redox one (necessary for the oxidative step). In conclusion, during my PhD work I investigated reactions that apply the “green chemistry” rules and strategies; in particular, I studied new greener approaches for the synthesis of chemicals (Part A and Part B), the optimisation of reaction parameters to make the oxidation process more flexible (Part C), and the use of a bioplatform molecule for the synthesis of a chemical intermediate (Part D).

Lipolysis and oxidation of lipids in foods are the major biochemical and chemical processes that cause food quality deterioration, leading to the characteristic, unpalatable odour and flavour called rancidity. In addition to unpalatability, rancidity may give rise to toxic levels of certain compounds like aldehydes, hydroperoxides, epoxides and cholesterol oxidation products. In this PhD study chromatographic and spectroscopic techniques were employed to determine the degree of lipid oxidation in different animal products and its relationship with technological parameters like feeding fat sources, packaging, processing and storage conditions. To achieve this goal capillary gas chromatography (CGC) was employed not only to determine the fatty acids profile but also, after solid phase extraction, the amount of sterols (cholesterol and phytosterols) and cholesterol oxidation products (COPs). To determine hydroperoxides, primary products of oxidation and quantify secondary products UV/VIS absorbance spectroscopy was applied. Beef and pork meat in this study were analysed. In actual fact, lipid oxidation is a major deterioration reaction in meat, meat products and results in adverse changes in the colour, flavour, texture of meat and develops different compounds which should be a risk to human health as oxysterols. On beef and pork meat, a study of lipid fraction during storage was carried out to evaluate its shelf-life and some nutritional features life saturated/unsaturated fatty acids ratio and sterols content, in according to the interest that has been growing around functional food in the last years. The last part of this research was focused on the study of lipid oxidation in emulsions. In oil-in-water emulsions antioxidant activity of 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) was evaluated. The rates of lipid oxidation of 1.0% stripped soybean oil-in-water emulsions with DOPC were followed by monitoring lipid hydroperoxide and hexanal as indicators of primary and secondary oxidation products and the droplet surface charge or zeta potential (ζ) of the emulsions with varying concentrations of DOPC were tested. This manuscript reports the main results obtained in the three activities briefly summarized as follows: 1. study on effects of feeding composition on the photoxidative stability of lipids from beef meat, evaluated during storage under commercial retail conditions; 2. evaluation of effects of diets and storage conditions on the oxidative stability of pork meat lipids; 3. study on oxidative behavior of DOPC in stripped soybean oil-in-water emulsions stabilized by nonionic surfactant.

Sekundäres organisches Aerosol (SOA) ist ein wichtiger Bestandteil von atmosphärischen Aerosolpartikeln. Atmosphärische Aerosole sind bedeutsam, da sie das Klima über direkte (Streuung und Absorption von Strahlung) und indirekte (Wolken-Kondensationskeime) Effekte beeinflussen. Nach bisherigen Schätzungen ist die SOA-Bildung aus biogenen Kohlenwasserstoffen global weit wichtiger als die SOA-Bildung aus anthropogenen Kohlenwasserstoffen. Reaktive Kohlenwasserstoffe, die in großen Mengen von der Vegetation emittiert werden und als die wichtigsten Vorläufersubstanzen für biogenes SOA gelten, sind die Terpene. In der vorliegenden Arbeit wurde eine Methode entwickelt, welche die Quantifizierung von aciden Produkten der Terpen-Oxidation ermöglicht. Die Abscheidung des größenselektierten Aerosols (PM 2.5) erfolgte auf Quarzfilter, die unter Zuhilfenahme von Ultraschall mittels Methanol extrahiert wurden. Nach Aufkonzentrierung und Lösungsmittelwechsel auf Wasser sowie Standardaddition wurden die Proben mit einer Kapillar-HPLC-ESI-MSn-Methode analysiert. Das verwendete Ionenfallen-Massenspektrometer (LCQ-DECA) bietet die Möglichkeit, Strukturaufklärung durch selektive Fragmentierung der Qasimolekülionen zu betreiben. Die Quantifizierung erfolgte teilweise im MS/MS-Modus, wodurch Selektivität und Nachweisgrenze verbessert werden konnten. Um Produkte der Terpen-Oxidation zu identifizieren, die nicht als Standards erhältlich waren, wurden Ozonolysexperimente durchgeführt. Dadurch gelang die Identifizierung einer Reihe von Oxidationsprodukten in Realproben. Neben schon bekannten Produkten der Terpen-Oxidation konnten einige Produkte erstmals in Realproben eindeutig als Produkte des α Pinens nachgewiesen werden. In den Proben der Ozonolyseexperimente konnten auch Produkte mit hohem Molekulargewicht (>300 u) nachgewiesen werden, die Ähnlichkeit zeigen zu den als Dimeren oder Polymeren in der Literatur bezeichneten Substanzen. Sie konnten jedoch nicht in Feldproben gefunden werden. Im Rahmen von 5 Messkampagnen in Deutschland und Finnland wurden Proben der atmosphärischen Partikelphase genommen. Die Quantifizierung von Produkten der Oxidation von α-Pinen, β-Pinen, 3-Caren, Sabinen und Limonen in diesen Proben ergab eine große zeitliche und örtliche Variationsbreite der Konzentrationen. Die Konzentration von Pinsäure bewegte sich beispielsweise zwischen etwa 0,4 und 21 ng/m³ während aller Messkampagnen. Es konnten stets Produkte verschiedener Terpene nachgewiesen werden. Produkte einiger Terpene eignen sich sogar als Markersubstanzen für verschiedene Pflanzenarten. Sabinen-Produkte wie Sabinsäure können als Marker für die Emissionen von Laubbäumen wie Buchen oder Birken verwendet werden, während Caren-Produkte wie Caronsäure als Marker für Nadelbäume, speziell Kiefern, verwendet werden können. Mit den quantifizierten Substanzen als Marker wurde unter zu Hilfenahme von Messungen des Gehaltes an organischem und elementarem Kohlenstoff im Aerosol der Anteil des sekundären organischen Aerosols (SOA) errechnet, der von der Ozonolyse der Terpene stammt. Erstaunlicherweise konnten nur 1% bis 8% des SOA auf die Ozonolyse der Terpene zurückgeführt werden. Dies steht im Gegensatz zu der bisherigen Meinung, dass die Ozonolyse der Terpene die wichtigste Quelle für biogenes SOA darstellt. Gründe für diese Diskrepanz werden in der Arbeit diskutiert. Um die atmosphärischen Prozesse der Bildung von SOA vollständig zu verstehen, müssen jedoch noch weitere Anstrengungen unternommen werden.

Ein neu entwickeltes globales Atmosphärenchemie- und Zirkulationsmodell (ECHAM5/MESSy1) wurde verwendet um die Chemie und den Transport von Ozonvorläufersubstanzen zu untersuchen, mit dem Schwerpunkt auf Nichtmethankohlenwasserstoffen. Zu diesem Zweck wurde das Modell durch den Vergleich der Ergebnisse mit Messungen verschiedenen Ursprungs umfangreich evaluiert. Die Analyse zeigt, daß das Modell die Verteilung von Ozon realistisch vorhersagt, und zwar sowohl die Menge als auch den Jahresgang. An der Tropopause gibt das Modell den Austausch zwischen Stratosphäre und Troposphäre ohne vorgeschriebene Flüsse oder Konzentrationen richtig wieder. Das Modell simuliert die Ozonvorläufersubstanzen mit verschiedener Qualität im Vergleich zu den Messungen. Obwohl die Alkane vom Modell gut wiedergeben werden, ergibt sich einige Abweichungen für die Alkene. Von den oxidierten Substanzen wird Formaldehyd (HCHO) richtig wiedergegeben, während die Korrelationen zwischen Beobachtungen und Modellergebnissen für Methanol (CH3OH) und Aceton (CH3COCH3) weitaus schlechter ausfallen. Um die Qualität des Modells im Bezug auf oxidierte Substanzen zu verbessern, wurden einige Sensitivitätsstudien durchgeführt. Diese Substanzen werden durch Emissionen/Deposition von/in den Ozean beeinflußt, und die Kenntnis über den Gasaustausch mit dem Ozean ist mit großen Unsicherheiten behaftet. Um die Ergebnisse des Modells ECHAM5/MESSy1 zu verbessern wurde das neue Submodell AIRSEA entwickelt und in die MESSy-Struktur integriert. Dieses Submodell berücksichtigt den Gasaustausch zwischen Ozean und Atmosphäre einschließlich der oxidierten Substanzen. AIRSEA, welches Informationen über die Flüssigphasenkonzentration des Gases im Oberflächenwasser des Ozeans benötigt wurde ausgiebig getestet. Die Anwendung des neuen Submodells verbessert geringfügig die Modellergebnisse für Aceton und Methanol, obwohl die Verwendung einer vorgeschriebenen Flüssigphasenkonzentration stark den Erfolg der Methode einschränkt, da Meßergebnisse nicht in ausreichendem Maße zu Verfügung stehen. Diese Arbeit vermittelt neue Einsichten über organische Substanzen. Sie stellt die Wichtigkeit der Kopplung zwischen Ozean und Atmosphäre für die Budgets vieler Gase heraus.

Questa relazione finale è incentrata sul lavoro sperimentale richiesto dalla collaborazione con l’azienda Natural Salumi ed ha come obiettivo principale quello di verifica dei parametri qualitativi e compositivi di una nuova tipologia di salume in fase di studio. L’azienda ha infatti messo a punto un nuovo “prodotto funzionale” addizionando componenti aventi attività nutraceutica al salame ottenuto da sole parti magre di suino (tipo NaturalFetta) e si propone di arricchire il proprio prodotto con acidi grassi polinsaturi della serie omega-3 (da olio di semi di lino), noti per gli effetti benefici esercitati sulla salute umana. Sul prodotto di nuova formulazione si effettueranno sia la determinazione della percentuale lipidica mediante metodo ufficiale Soxhlet ed automatizzato Soxtec, al fine di individuare il quantitativo totale della parte grassa, che una caratterizzazione del profilo quali-quantitativo in acidi grassi, fondamentale per verificare la conformità con i requisiti indicati dal Reg. 1924/06 (e successivi come il Reg. UE 432/2012) previsti per le indicazioni da inserire in etichetta (claims) per i prodotti arricchiti con acidi grassi polinsaturi (PUFA). La determinazione del profilo in acidi grassi e, nello specifico, del contenuto in acido -linolenico, sarà realizzata mediante gascromatografia previa estrazione della frazione lipidica dei salumi con metodo di Folch modificato, che prevede un'estrazione solido-liquido. La concentrazione di acido alfa-linolenico sarà inoltre monitorata durante tutto il periodo di shelf-life del prodotto (45 gg) al fine di valutare eventuali variazioni durante la conservazione. Per soddisfare quest’ultima finalità, le analisi saranno eseguite sia all’inizio (T0) che alla fine (T1) della vita commerciale del prodotto. Le stesse analisi verranno inoltre condotte sia sul prodotto a formulazione classica che su un prodotto del tutto simile, commercializzato da un’altra azienda e considerato come leader di mercato. In relazione ad un possibile aumento di ossidabilità della frazione lipidica, sarà realizzato un controllo dei parametri chimici, qualitativi e compositivi, con particolare riferimento ai prodotti di ossidazione del colesterolo (date le loro implicazioni in ambito biomedico). La maggiore presenza in formulazione di acidi grassi polinsaturi potrebbe infatti favorire un incremento dei COPs (Cholesterol Oxidation Products), che saranno separati, identificati e quantificati mediante la procedura SPE/GC-MS (estrazione e purificazione in fase solida accoppiata ad analisi gascromatografica con rivelazione mediante spettrometria di massa).
