A Presente dissertação apresenta uma aplicação de Inteligência Computacional na área de Geotecnia, com a utilização da Técnica de Neuro-Fuzzy para indicar a suscetibilidade de escorregamento de taludes no município do Rio de Janeiro, a partir de inspeção visual. Neste trabalho, a suscetibilidade corresponde à possibilidade de ocorrência de escorregamento sem considerar os danos relacionados ao evento. Adotou-se como variável de saída a Previsão de Escorregamento (PE) com três adjetivos que correspondem a Suscetibilidades Alta, Média e Baixa. A metodologia utilizada consistiu em, inicialmente, montar um banco de dados com informações preliminares de análise de estabilidade, com a indicação dos condicionantes de escorregamento relacionados à geomorfologia, pluviosidade, capacidade de drenagem, vegetação e ocupação com seus respectivos graus de suscetibilidades de escorregamento obtidos em um conjunto de Laudos de Vistoria da Geo Rio. O banco de dados foi aplicado em um algoritmo de Neuro-Fuzzy. Diversos testes foram realizados com as alterações dos parâmetros do modelo Neuro-Fuzzy para uma combinação de fatores condicionantes de escorregamento e refinamento do banco de dados. Os testes apresentaram diminuição do erro fornecido pelo programa com o aumento de tipos de condicionantes utilizados no treinamento, o que permite inferir que o escorregamento ocorre por uma complexa relação entre diversos fatores condicionantes. O banco de dados utilizado nos testes apresenta descontinuidades nas relações entre os diversos condicionantes, ou seja, para uma mesma faixa de valores de Altura do talude, não é possível obter uma relação para todas as faixas de outro condicionante e, até mesmo, para todas as faixas da Previsão de Escorregamento. As PEs obtidas na validação do modelo tiveram seus valores próximos aos desejados somente nos conjuntos de variáveis utilizadas para o treinamento. O modelo não foi capaz de apresentar valores de suscetibilidades dentro da faixa de valores utilizados no treinamento para combinação de variáveis com pequenos ruídos, o que indica a necessidade de ampliação do banco de dados tanto quantitativamente quanto qualitativamente de modo a cobrir as descontinuidades apresentadas nas relações entre as variáveis.

Background: The impact of nano-scaled materials on photosynthetic organisms needs to be evaluated. Plants represent the largest interface between the environment and biosphere, so understanding how nanoparticles affect them is especially relevant for environmental assessments. Nanotoxicology studies in plants allude to quantum size effects and other properties specific of the nano-stage to explain increased toxicity respect to bulk compounds. However, gene expression profiles after exposure to nanoparticles and other sources of environmental stress have not been compared and the impact on plant defence has not been analysed. Results: Arabidopsis plants were exposed to TiO2-nanoparticles, Ag-nanoparticles, and multi-walled carbon nanotubes as well as different sources of biotic (microbial pathogens) or abiotic (saline, drought, or wounding) stresses. Changes in gene expression profiles and plant phenotypic responses were evaluated. Transcriptome analysis shows similarity of expression patterns for all plants exposed to nanoparticles and a low impact on gene expression compared to other stress inducers. Nanoparticle exposure repressed transcriptional responses to microbial pathogens, resulting in increased bacterial colonization during an experimental infection. Inhibition of root hair development and transcriptional patterns characteristic of phosphate starvation response were also observed. The exogenous addition of salicylic acid prevented some nano-specific transcriptional and phenotypic effects, including the reduction in root hair formation and the colonization of distal leaves by bacteria. Conclusions: This study integrates the effect of nanoparticles on gene expression with plant responses to major sources of environmental stress and paves the way to remediate the impact of these potentially damaging compounds through hormonal priming.

Nos últimos anos, temos nos deparado com a difusão maciça e a popularização crescente de descrições biológicas para aspectos outrora pensados como mentais, sociais, ou relacionais. Visível em diversas arenas leigas e científicas, esta tendência freqüentemente elege o cérebro como o órgão privilegiado da sua atenção. A cada semana é divulgada uma nova localização cerebral correlacionada os mais variados aspectos comportamentais e ou de personalidade. Acompanhando este movimento, é notável o esforço intelectual e financeiro despendido nos últimos anos no campo da saúde mental, no sentido de fazer avançar pesquisas cujo foco central é a descoberta das bases neurobiológicas dos transtornos mentais. Esta tendência apontaria na direção de uma fusão entre a psiquiatria e a neurologia em uma disciplina única, de teor fisicalista, chamada por alguns de cerebrologia. Dentre os acontecimentos que serviram de alicerce para a legitimação e a popularização desta tendência, o desenvolvimento nas últimas décadas de novas técnicas e tecnologias de visualização médica, como a tomografia por emissão de pósitrons (PET scan) e a ressonância magnética funcional (fMRI), foi fundamental. Elas permitiram a construção de imagens das mais diversas categorias nosográficas construídas no campo psiquiátrico, veiculando tacitamente uma série de pressupostos e promessas. Malgrado o imaginário cultural sustentado por estas tecnologias e todo o esforço despendido nas últimas décadas no sentido de se tentar localizar os marcadores biológicos dos transtornos psiquiátricos, não há, até o presente momento, nenhum resultado conclusivo que autorize o diagnóstico por imagem de nosografias como a esquizofrenia, a depressão, e muito menos o jogo patológico. Apesar de todo o alarde midiático e dos montantes milionários direcionados para pesquisas nesta área, os resultados concretos obtidos até agora não estão livres das mais ferozes controvérsias. Entretanto, ainda que estejamos muito longe da construção de mapas precisos para as perturbações mentais é espantoso o poder de convencimento que as neuro-imagens comportam na atualidade. Os scans são exibidos como verdades visuais, ou fatos acerca das pessoas e do mundo, numa proporção muito superior aos dados que apresentam. Alguns críticos chamam este aspecto de neurorealismo, ou de retórica da auto-evidência. A intenção deste trabalho é problematizar o poder persuasivo que as neuro-imagens detém na contemporaneidade, especialmente quando utilizadas com a finalidade diagnóstica no campo da saúde mental. Se estas imagens transmitem uma ideia de neutralidade, transparência imediata e auto-evidência, este trabalho almeja inseri-las num contexto sócio-histórico, a partir do qual puderam adquirir sentido, familiaridade e valor de verdade. O ponto de partida é o de que elas estão localizadas no cruzamento de dois movimentos históricos distintos: o das ilustrações médicas, em sua relação com a produção de conhecimento objetivo; e o das pesquisas acerca da localização no córtex cerebral de comportamentos complexos e traços de personalidade. Além de estabelecer algumas condições históricas de possibilidade para a emergência de um neo-localizacionismo cerebral, mediado pelas novas tecnologias de imageamento, pretende-se enfatizar algumas descontinuidades com projetos anteriores e marcar a influência do contexto cultural da atualidade para o sucesso e poder persuasivo deste tipo de tecnologia.

The evolution of the railway sector depends, to a great extent, on the deployment of advanced railway signalling systems. These signalling systems are based on communication architectures that must cope with complex electromagnetical environments. This paper is outlined in the context of developing the necessary tools to allow the quick deployment of these signalling systems by contributing to an easier analysis of their behaviour under the effect of electromagnetical interferences. Specifically, this paper presents the modelling of the Eurobalise-train communication flow in a general purpose simulation tool. It is critical to guarantee this communication link since any lack of communication may lead to a stop of the train and availability problems. In order to model precisely this communication link we used real measurements done in a laboratory equipped with elements defined in the suitable subsets. Through the simulation study carried out, we obtained performance indicators of the physical layer such as the received power, SNR and BER. The modelling presented in this paper is a required step to be able to provide quality of service indicators related to perturbed scenarios.

Choosing a project manager for a construction project—particularly, large projects—is a critical project decision. The selection process involves different criteria and should be in accordance with company policies and project specifications. Traditionally, potential candidates are interviewed and the most qualified are selected in compliance with company priorities and project conditions. Precise computing models that could take various candidates’ information into consideration and then pinpoint the most qualified person with a high degree of accuracy would be beneficial. On the basis of the opinions of experienced construction company managers, this paper, through presenting a fuzzy system, identifies the important criteria in selecting a project manager. The proposed fuzzy system is based on IF-THEN rules; a genetic algorithm improves the overall accuracy as well as the functions used by the fuzzy system to make initial estimates of the cluster centers for fuzzy c-means clustering. Moreover, a back-propagation neutral network method was used to train the system. The optimal measures of the inference parameters were identified by calculating the system’s output error and propagating this error within the system. After specifying the system parameters, the membership function parameters—which by means of clustering and projection were approximated—were tuned with the genetic algorithm. Results from this system in selecting project managers show its high capability in making high-quality personnel predictions

The nervous system implements a networked control system in which the plants take the form of limbs, the controller is the brain, and neurons form the communication channels. Unlike standard networked control architectures, there is no periodic sampling, and the fundamental units of communication contain little numerical information. This paper describes a novel communication channel, modeled after spiking neurons, in which the transmitter integrates an input signal and sends out a spike when the integral reaches a threshold value. The reciever then filters the sequence of spikes to approximately reconstruct the input signal. It is shown that for appropriate choices of channel parameters, stable feedback control over these spiking channels is possible. Furthermore, good tracking performance can be achieved. The data rate of the channel increases linearly with the size of the inputs. Thus, when placed in a feedback loop, small loop gains imply a low data rate. ©2010 IEEE.

Highly sensitive biosensor for detection of acetylcholine (ACh) and competitive acetylcholinesterase (AChE) inhibitor, eserine, is investigated. Peculiar microelectronic configuration of an ion-sensitive field-effect transistor (ISFET) in addition to a right choice of the pH-transducing nanolayers allows recording a response of the enzyme-modified ISFET (EnFET) to a wide range of ACh concentrations. We demonstrate a remarkable improvement of at least three orders of magnitude in dose response to ACh. Described bioelectronic system reveals clear response, when the catalytic activity of the immobilized AChE is inhibited in a reversible manner by eserine, competitive inhibitor of AChE. ©2007 IEEE.

Interferons (IFNs), consisting of three major subfamilies, type I, type II (gamma) and type III (lambda) IFN, activate vertebrate antiviral defences once bound to their receptors. The three IFN subfamilies bind to different receptors, IFNAR1 and IFNAR2 for type I IFNs, IFN gamma R1 and IFN gamma R2 for type II IFN, and IL-28R1 and IL-10R2 for type III IFNs. In fish, although many types I and II IFN genes have been cloned, little is known about their receptors. In this report, two putative IFN-gamma receptor chains were identified and sequenced in rainbow trout (Oncorhynchus mykiss), and found to have many common characteristics with mammalian type II IFN receptor family members. The presented gene synteny analysis, phylogenetic tree analysis and ligand binding analysis all suggest that these molecules are the authentic IFN gamma Rs in fish. They are widely expressed in tissues, with IFN gamma R1 typically more highly expressed than IFN gamma R2. Using the trout RTG-2 cell line it was possible to show that the individual chains could be differentially modulated, with rIFN-gamma and rIL-1 beta down regulating IFN gamma R1 expression but up regulating IFN gamma R2 expression. Overexpression of the two receptor chains in RTG-2 cells revealed that the level of IFN gamma R2 transcript was crucial for responsiveness to rIFN-gamma, in terms of inducing gamma IP expression. Transfection experiments showed that the two putative receptors specifically bound to rIFN-gamma. These findings are discussed in the context of how the IFN gamma R may bind IFN-gamma in fish and the importance of the individual receptor chains to signal transduction. (c) 2009 Elsevier Ltd. Cell biology is characterised by low molecule numbers and coupled stochastic chemical reactions with intrinsic noise permeating and dominating the interactions between molecules. Recent work [9] has shown that in such environments there are hard limits on the accuracy with which molecular populations can be controlled and estimated. These limits are predicated on a continuous diffusion approximation of the target molecule (although the remainder of the system is non-linear and discrete). The principal result of [9] assumes that the birth rate of the signalling species is linearly dependent on the target molecule population size. In this paper, we investigate the situation when the entire system is kept discrete, and arbitrary non-linear coupling is allowed between the target molecule and downstream signalling molecules. In this case it is possible, by relying solely on the event triggered nature of control and signalling reactions, to define non-linear reaction rate modulation schemes that achieve improved performance in certain parameter regimes. These schemes would not appear to be biologically relevant, raising the question of what are an appropriate set of assumptions for obtaining biologically meaningful results. © 2013 EUCA.

A prototype neuro-stimulus chip for sub-retinal implants in blind patients affected by Age-related Macular Degeneration (AMD) or Retinitis Pigmentosa (RP) is presented in this paper. This retinal prosthetic chip was designed to replace the degenerated photoreceptor cells, and in order to stimulate directly the remaining healthy layers of retinal neurons. The current stimulus circuits are monolithic integrated with photodiodes (PD) array, which can convert the illumination on the eyes into bi-phasic electrical pulses. In addition, a novel charge cancellation circuit is used to discharge the electrodes for medical safty. The prototype chip is designed and fabricated in HJTC 0.18 mu m N-well CMOS 1P6M Mix-signal process, with a +/- 2.5 V dual voltage power supply.

Acute myeloid leukaemia refers to cancer of the blood and bone marrow characterised by the rapid expansion of immature blasts of the myeloid lineage. The aberrant proliferation of these blasts interferes with normal haematopoiesis, resulting in symptoms such as anaemia, poor coagulation and infections. The molecular mechanisms underpinning acute myeloid leukaemia are multi-faceted and complex, with a range of diverse genetic and cytogenetic abnormalities giving rise to the acute myeloid leukaemia phenotype. Amongst the most common causative factors are mutations of the FLT3 gene, which codes for a growth factor receptor tyrosine kinase required by developing haematopoietic cells. Disruptions to this gene can result in constitutively active FLT3, driving the de-regulated proliferation of undifferentiated precursor blasts. FLT3-targeted drugs provide the opportunity to inhibit this oncogenic receptor, but over time can give rise to resistance within the blast population. The identification of targetable components of the FLT3 signalling pathway may allow for combination therapies to be used to impede the emergence of resistance. However, the intracellular signal transduction pathway of FLT3 is relatively obscure. The objective of this study is to further elucidate this pathway, with particular focus on the redox signalling element which is thought to be involved. Signalling via reactive oxygen species is becoming increasingly recognised as a crucial aspect of physiological and pathological processes within the cell. The first part of this study examined the effects of NADPH oxidase-derived reactive oxygen species on the tyrosine phosphorylation levels of acute myeloid leukaemia cell lines. Using two-dimensional phosphotyrosine immunoblotting, a range of proteins were identified as undergoing tyrosine phosphorylation in response to NADPH oxidase activity. Ezrin, a cytoskeletal regulatory protein and substrate of Src kinase, was selected for further study. The next part of this study established that NADPH oxidase is subject to regulation by FLT3. Both wild type and oncogenic FLT3 signalling were shown to affect the expression of a key NADPH oxidase subunit, p22phox, and FLT3 was also demonstrated to drive intracellular reactive oxygen species production. The NADPH oxidase target protein, Ezrin, undergoes phosphorylation on two tyrosine residues downstream of FLT3 signalling, an effect which was shown to be p22phox-dependent and which was attributed to the redox regulation of Src. The cytoskeletal associations of Ezrin and its established role in metastasis prompted the investigation of the effects of FLT3 and NADPH oxidase activity on the migration of acute myeloid leukaemia cell lines. It was found that inhibition of either FLT3 or NADPH oxidase negatively impacted on the motility of acute myeloid leukaemia cells. The final part of this study focused on the relationship between FLT3 signalling and phosphatase activity. It was determined, using phosphatase expression profiling and real-time PCR, that several phosphatases are subject to regulation at the levels of transcription and post-translational modification downstream of oncogenic FLT3 activity. In summary, this study demonstrates that FLT3 signal transduction utilises a NADPH oxidase-dependent redox element, which affects Src kinase, and modulates leukaemic cell migration through Ezrin. Furthermore, the expression and activity of several phosphatases is tightly linked to FLT3 signalling. This work reveals novel components of the FLT3 signalling cascade and indicates a range of potential therapeutic targets.
