The aim of this work was to determine PAs levels in pith tissues and callus cultures from haploid and diploid tobacco plants, explanted from the apical and basal regions of the stem. These explants were cultured in an RM-64 medium supplied with IAA and kinetin, under light or in the dark, during successive subcultures. PAs levels followed a basipetal decrease in diploid and an increase in haploid, pith tissues. A similar pattern of total PAs (free + conjugated) was observed for the callus of diploid and haploid plants maintained in the light, and for the haploid callus in the dark, whereas the diploid callus in the dark showed a constant increase in total PAs levels until the end of culture. The PA increase in the diploid callus in the dark was related to free Put levels increase. The ploidy status of the plants could express different PA gradients together with the plant pith and in vitro callus cultures.

Strategies to promote bone repair have included exposure of cells to growth factor (GF) preparations from blood that generally include proteins as part of a complex mixture. This study aimed to evaluate the effects of such a mixture on different parameters of the development of the osteogenic phenotype in vitro. Osteoblastic cells were obtained by enzymatic digestion of human alveolar bone and cultured under standard osteogenic conditions until subconfluence. They were subcultured on Thermanox coverslips up to 14 days. Treated cultures were exposed during the first 7 days to osteogenic medium supplemented with a GFs + proteins mixture containing the major components found in platelet extracts [plate I et-derived growth factor-BB, transforming growth factor (TGF)-beta 1, TGF-beta 2, albumin, fibronectin, and thrombospondin] and to osteogenic medium alone thereafter. Control cultures were exposed only to the osteogenic medium. Treated cultures exhibited a significantly higher number of adherent cells from day 4 onward and of cycling cells at days 1 and 4, weak alkaline phosphatase (ALP) labeling, and significantly decreased levels of ALP activity and mRNA expression. At day 14, no Alizarin red-stained nodular areas were detected in cultures treated with GFs + proteins. Results were confirmed in the rat calvaria-derived osteogenic cell culture model. The addition of bone morphogenetic protein 7 or growth and differentiation factor 5 to treated cultures upregulated Runx2 and ALP mRNA expression, but surprisingly, ALP activity was not restored. These results showed that a mixture of GFs + proteins affects the development of the osteogenic phenotype both in human and rat cultures, leading to an increase in the number of cells, but expressed a less differentiated state.

A wide range of animals suffer from periodontal disease. However, there is very little reported on disease and oral micro-biota of Australian animals. Therefore, the oral cavity of 90 marsupials was examined for oral health status. Plaque samples were collected from the subgingival margins using curettes; or swabs. Plaque samples were plated onto. non-selective trypticase soy agar plates, selective trypticase soy agar, non-selective and selective Wilkens Chalgrens, Agar. Plates were incubated in an anaerobic atmosphere and examined after 7-14 days for the presence of black-brown-pigmented colonies. A combination of morphological and biochemical tests were used (colonial morphology, pigmentation, aerobic growth, Gram reaction, fluorescence under long-wave UV light (360 nm), production of catalase, enzymatic activity with fluorogenic substrates and haemagglutination of sheep red cells) to identify these organisms. Black-pigmented bacteria were cultivated from the plaque of 32 animals including six eastern grey kangaroos, a musky rat kangaroo, a whiptail and a red-necked wallaby, 18 koalas, a bandicoot and five brushtail possums. No black-pigmented colonies were cultivated from squirrel or sugar gliders or quokkas or from marsupial mice. The majority of isolates were identified as Porphyromonas gingivalis-like species with the higher prevalence of isolation from the oral cavity of macropods (the kangaroos and wallabies). Oral diseases, such as gingivitis can be found in native Australian animals with older koalas having an increase in disease indicators and black-pigmented bacteria. Non-selective Wilkens Chalgren Agar was the medium of choice for the isolation of black-pigmented bacteria. (C) 2002 Elsevier Science Ltd. All rights reserved.

Remyelination can be studied in aggregating rat brain cell cultures after limited demyelination. Demyelination was induced using a monoclonal antibody against myelin/oligodendrocyte glycoprotein (MOG mAb), in the presence of complement. De- and remyelination were assessed by measuring myelin basic protein (MBP). Two days after removing the MOG mAb, MBP levels reached 50% of controls and after 7 days 93%. During this period, cell proliferation determined by [14C]thymidine incorporation was similar in remyelinating and control cultures. Hormones and growth factors were tested for possible stimulatory effect on remyelinating cultures. Bovine growth hormone (bGH), triiodothyronine (T3), basic fibroblast growth factor (bFGF) and platelet-derived growth factor (PDGF) did not improve remyelination. Only epidermal growth factor (EGF) increased the level of remyelination. PDGF increased the rate of cell proliferation in both control and remyelinating cultures. A significant proportion of oligodendrocytes entered the cell division cycle and were not available for remyelination. The results obtained with PDGF and FGF (inhibition) support the idea that a pool of progenitor cells was still present and able to proliferate and differentiate into myelinating oligodendrocytes. The levels of myelin protein mRNAs were investigated during de- and remyelination. During demyelination, myelin protein mRNA levels decreased to approximately 50% of control cultures and returned to normal during remyelination. These preliminary results indicate that normal levels of gene transcription are sufficient to meet the increased need for newly synthesized myelin proteins during remyelination.

Serum-free aggregating cell cultures of fetal rat telencephalon treated with low doses (0.5 nM) of epidermal growth factor (EGF) showed a small, transient increase in DNA synthesis but no significant changes in total DNA and protein content. By contrast, treatment with high doses (13 nM) of EGF caused a marked stimulation of DNA synthesis as well as a net increase in DNA and protein content. The expression of the astrocyte-specific enzyme, glutamine synthetase, was greatly enhanced both at low and at high EGF concentrations. These results suggest that at low concentration EGF stimulates exclusively the differentiation of astrocytes, whereas at high concentration, EGF has also a mitogenic effect. Nonproliferating astrocytes in cultures treated with 0.4 microM 1-beta-D-arabinofuranosyl-cytosine were refractory to EGF treatment, indicating that their responsiveness to EGF is cell cycle-dependent. Binding studies using a crude membrane fraction of 5-day cultures showed a homogeneous population of EGF binding sites (Kd approximately equal to 2.6 nM). Specific EGF binding sites were found also in non-proliferating (and nonresponsive) cultures, although they showed slightly reduced affinity and binding capacity. This finding suggests that the cell cycle-dependent control of astroglial responsiveness to EGF does not occur at the receptor level. However, it was found that the specific EGF binding sites disappear with progressive cellular differentiation.

Sekä organisaatiokulttuuria, luottamusta että innovatiivisuutta on tutkittu paljon, mutta toistaiseksi nämä käsitteet yhdistävää kokonaisvaltaista tutkimusta ei juuri ole tehty tai ainakaan raportoitu tieteellisissä julkaisuissa. Tätä tutkimus käsitteli organisaatiokulttuurin, luottamuksen ja innovatiivisuuden suhteita erilaisissa organisaatiokulttuureissa. Tutkimuksen tavoitteena oli tutkia organisaatiokulttuurin vaikutusta luottamukseen (sekä kompetenssiin, hyväntahtoisuuteen että rehellisyyteen perustuvaan lateraaliin, vertikaaliin ja institutionaaliseen luottamukseen), innovaatioilmastoon ja innovaatiotoiminnan tuloksellisuuteen. Organisaatiokulttuurin, luottamuksen ja innovatiivisuuden yhteyttä tarkasteltiin neljässä erityyppisessä organisaatiokulttuurissa (klaani-, adhokratia-, hierarkia- ja markkinakulttuurit), jotka pohjautuvat kilpailevien arvojen malliin. Tutkimuksen empiirinen osa toteutettiin posti ja Internet -pohjaisena kyselytutkimuksena 40 organisaatioyksikössä tilastollisen analyysin menetelmin. Yleisellä tasolla työssä saatiin selville, että luottamuksen ja innovatiivisuuden tasot vaihtelevat erityyppisissä organisaatio-kulttuureissa. Tarkemmin sanottuna klaani- ja adhokratiakulttuureissa luottamus ja innovatiivisuus olivat korkeita, ja näillä kulttuureilla oli myös positiivinen vaikutus innovaatiotoiminnan tuloksellisuuteen. Erityisesti institutionaalisen luottamuksen ja innovaatiotuen merkitykset olivat tärkeitä, sillä ne toimivat mediaattoreina organisaatiokulttuurin ja innovatiivisuuden välisessä suhteessa. Luottamuksella ja innovatiivisuudelle ei sitä vastoin ollut vaikutusta hierarkia- ja markkinakulttuureissa, tai vaikutus oli negatiivinen. Tässä työssä osoitettiin myös aiemmin hyvin vähän tutkitun institutionaalisen organisatorisen luottamuksen merkitys organisaatioiden innovatiivisuudessa.

This research focuses on the career experiences of women managers in the IT industry in China and Finland, two countries with different cultures, policies, size of population, and social and economic structures regarding work-life support and equal opportunities. The object of this research is to present a cross-cultural comparison of women’s career experiences and how women themselves understand and account for their careers. The study explores how the macro and the micro levels of cultural and social processes become manifested in the lives of individual women. The main argument in this thesis is that culture plays a crucial role in making sense of women’s career experiences, although its role should be understood through its interrelationship with other social processes, e.g., institutional relations, social policies, industrial structures and organizations, as well as globalization. The interrelationship of a series of cultural and social processes affects individuals’ attitudes to, and arrangement and organization of, their work and family lives. This thesis consists of two parts. The first part introduces the research topic and discusses the overall results. The second part comprises five research papers. The main research question of the study is: How do cultural and social processes affect the experiences of women managers? Quantitative and qualitative research methods, which include in-depth interviews, Q-methodology, interpretive analysis, and questionnaires, are used in the study. The main theoretical background is culturally sensitive career theory and the theory of individual differences. The results of this study are viewed through a feminist lens. The research methodology applied allows new explorations on how demographic factors, work experiences, lifestyle issues, and organizational cultures can jointly affect women’s managerial careers. The sample group used in the research is 42 women managers working in IT companies in China (21) and Finland (21). The results of the study illustrate the impact of history, tradition, culture, institutional relations, social politics, industry and organizations, and globalization on the careers of women managers. It is claimed that the role of culture – cultural norms within nations and organizations – is of great importance in the relationship of gender and work. Women’s managerial careers are affected by multiple factors (personal, social and cultural) reflecting national and inter-individual differences. The results of the study contribute to research on careers, adding particularly to the literature on gender, work and culture, and offering a complex and holistic perspective for a richer understanding of pluralism and global diversity. The results of the study indicate how old and new career perspectives are evidenced in women managers in the IT industry. The research further contributes to an understanding of women’s managerial careers from a cross-culture perspective. In addition, the study contributes to the literature on culture and extends understanding of Hofstede’s work. Further, most traditional career theories do not perceive the importance of culture in determining an individual’s career experience and this study richens understanding of women managers’ careers and has considerable implications for international human resource management. The results of this study emphasize the need, when discussing women managers’ careers, to understand the ways by which gendering is produced rather than merely examining gender differences. It is argued that the meaning of self-knowledge is critical. Further, the environment where the careers under study develop differs greatly; China and Finland are very different – culturally, historically and socially. The findings of this study should, therefore, be understood as a holistic, specific, and contextually-bound.

Characterizing Propionibacterium freudenreichii ssp. shermanii JS and Lactobacillus rhamnosus LC705 as a new probiotic combination: basic properties of JS and pilot in vivo assessment of the combination Each candidate probiotic strain has to have the documentation for the proper identification with current molecular tools, for the biological properties, for the safety aspects and for the health benefits in human trials if the intention is to apply the strain as health promoting culture in the commercial applications. No generalization based on species properties of an existing probiotic are valid for any novel strain, as strain specific differences appear e.g. in the resistance to GI tract conditions and in health promoting benefits (Madsen, 2006). The strain evaluation based on individual strain specific probiotic characteristics is therefore the first key action for the selection of the new probiotic candidate. The ultimate goal in the selection of the probiotic strain is to provide adequate amounts of active, living cells for the application and to guarantee that the cells are physiologically strong enough to survive and be biologically active in the adverse environmental conditions in the product and in GI tract of the host. The in vivo intervention studies are expensive and time consuming; therefore it is not rational to test all the possible candidates in vivo. Thus, the proper in vitro studies are helping to eliminate strains which are unlikely to perform well in vivo. The aims of this study were to characterize the strains of Propionibacterium freudenreichii ssp. shermanii JS and Lactobacillus rhamnosus LC705, both used for decades as cheese starter cultures, for their technological and possible probiotic functionality applied in a combined culture. The in vitro studies of Propionibacterium freudenreichii ssp. shermanii JS focused on the monitoring of the viability rates during the acid and bile treatments and on the safety aspects such as antibiotic susceptibility and adhesion. The studies with the combination of the strains JS and LC705 administered in fruit juices monitored the survival of the strains JS and LC705 during the GI transit and their effect on gut wellbeing properties measured as relief of constipation. In addition, safety parameters such as side effects and some peripheral immune parameters were assessed. Separately, the combination of P. freudenreichii ssp. shermanii JS and Lactobacillus rhamnosus LC705 was evaluated from the technological point of view as a bioprotective culture in fermented foods and wheat bread applications. In this study, the role ofP. freudenreichii ssp. shermanii JS as a candidate probiotic culture alone and in a combination with L. rhamnosus LC705 was demonstrated. Both strains were transiently recovered in high numbers in fecal samples of healthy adults during the consumption period. The good survival through the GI transit was proven for both strains with a recovery rate from 70 to 80% for the JS strain and from 40 to 60% for the LC705 strain from the daily dose of 10 log10 CFU. The good survival was shown from the consumption of fruit juices which do not provide similar matrix protection for the cells as milk based products. The strain JS did not pose

The thermotolerant capacity of several lactic acid bacteria strains isolated from cooked commercial sausages was determined. Four strains were positively identified as Lactobacillus plantarum, Lactobacillus curvatus, Pediococcus pentosaceus and Pediococcus acidilacti, after surviving thermal treatment (70 °C during 60 minutes). Thermotolerant strains were inoculated in sausage batters before cooking in order to determine their effect on color, texture, acceptance and inhibition of Enterobacteria during 12 days at 8 °C. No significant effect of the inoculated strains was detected on color parameters. Textural profile parameters, cohesiveness and resilience, were not affected by the inoculation of thermotolerant lactic acid bacteria, but L. curvatus sausages resulted softer than the rest of the treatments. Samples inoculated with L. curvatus also obtained the lowest scores for the sensory attributes evaluated, with the remaining treatments causing no unfavorable effects on sausage acceptance. There was a reduction in enterobacterial counts after 12 days of cold storage in inoculated samples. The performance of inoculated lactic acid bacteria strains can be explained in a similar way as that of starter cultures in dry-fermented sausages, where the growth in nests impairs other pathogenic microorganisms present in the rest of the sausage, since environmental conditions and the early inoculation of these thermotolerant strains favor them to become the dominant flora.

The effect of inulin addition and starters (Kefir grains or commercial starter culture) on the microbial viability, texture, and chemical characteristics of Kefir beverages prepared with whole or skim milk was evaluated during refrigerated storage. The type of starter did not influence microbial viability during the storage of the beverages, but the chemical and textural changes (decreases in pH, lactose concentration, and inulin and increased acidity, firmness, and syneresis) were more pronounced in the formulations fermented with grains than those fermented with the starter culture. The addition of inulin did not influence acidity or viability of lactic acid bacteria, but in general, its effect on the survival of acetic acid bacteria, Lactococcus and yeasts, firmness, and syneresis depended on the type of milk and starter culture used. Generally, the yeast, acetic acid bacteria, and Leuconostoc counts increased or remained unchanged, while the total population of lactic acid bacteria and Lactococcus were either reduced by 1 to 2 logs or remained unchanged during storage.

The aim of this study was to compare the rheology of spreadable cheeses elaborated with autochthonous lactic starter cultures without the addition of exopolysaccharide-producing strain in the same starter with exopolysaccharide-producing strain. From a rheological standpoint, both samples were characterized as weak viscoelastic gels and pseudoplastic products. It was concluded that cheese made with exopolysaccharide-producing strain showed smaller G', G", and η* values over the range of frequencies studied and smaller critic stress values than the cheese without exopolysaccharide-producing strain. The results obtained indicate that cheeses without exopolysaccharide-producing strain need to be added with any texture enhancer product.

The topic of this thesis is marginaVminority popular music and the question of identity; the term "marginaVminority" specifically refers to members of racial and cultural minorities who are socially and politically marginalized. The thesis argument is that popular music produced by members of cultural and racial minorities establishes cultural identity and resists racist discourse. Three marginaVminority popular music artists and their songs have been chosen for analysis in support of the argument: Gil Scott-Heron's "Gun," Tracy Chapman's "Fast Car" and Robbie Robertson's "Sacrifice." The thesis will draw from two fields of study; popular music and postcolonialism. Within the area of popular music, Theodor Adorno's "Standardization" theory is the focus. Within the area of postcolonialism, this thesis concentrates on two specific topics; 1) Stuart Hall's and Homi Bhabha's overlapping perspectives that identity is a process of cultural signification, and 2) Homi Bhabha's concept of the "Third Space." For Bhabha (1995a), the Third Space defines cultures in the moment of their use, at the moment of their exchange. The idea of identities arising out of cultural struggle suggests that identity is a process as opposed to a fixed center, an enclosed totality. Cultures arise from historical memory and memory has no center. Historical memory is de-centered and thus cultures are also de-centered, they are not enclosed totalities. This is what Bhabha means by "hybridity" of culture - that cultures are not unitary totalities, they are ways of knowing and speaking about a reality that is in constant flux. In this regard, the language of "Otherness" depends on suppressing or marginalizing the productive capacity of culture in the act of enunciation. The Third Space represents a strategy of enunciation that disrupts, interrupts and dislocates the dominant discursive construction of US and THEM, (a construction explained by Hall's concept of binary oppositions, detailed in Chapter 2). Bhabha uses the term "enunciation" as a linguistic metaphor for how cultural differences are articulated through discourse and thus how differences are discursively produced. Like Hall, Bhabha views culture as a process of understanding and of signification because Bhabha sees traditional cultures' struggle against colonizing cultures as transforming them. Adorno's theory of Standardization will be understood as a theoretical position of Western authority. The thesis will argue that Adorno's theory rests on the assumption that there is an "essence" to music, an essence that Adorno rationalizes as structure/form. The thesis will demonstrate that constructing music as possessing an essence is connected to ideology and power and in this regard, Adorno's Standardization theory is a discourse of White Western power. It will be argued that "essentialism" is at the root of Western "rationalization" of music, and that the definition of what constitutes music is an extension of Western racist "discourses" of the Other. The methodological framework of the thesis entails a) applying semiotics to each of the three songs examined and b) also applying Bhabha's model of the Third Space to each of the songs. In this thesis, semiotics specifically refers to Stuart Hall's retheorized semiotics, which recognizes the dual function of semiotics in the analysis of marginal racial/cultural identities, i.e., simultaneously represent embedded racial/cultural stereotypes, and the marginal raciaVcultural first person voice that disavows and thus reinscribes stereotyped identities. (Here, and throughout this thesis, "first person voice" is used not to denote the voice of the songwriter, but rather the collective voice of a marginal racial/cultural group). This dual function fits with Hall's and Bhabha's idea that cultural identity emerges out of cultural antagonism, cultural struggle. Bhabha's Third Space is also applied to each of the songs to show that cultural "struggle" between colonizers and colonized produces cultural hybridities, musically expressed as fusions of styles/sounds. The purpose of combining semiotics and postcolonialism in the three songs to be analyzed is to show that marginal popular music, produced by members of cultural and racial minorities, establishes cultural identity and resists racist discourse by overwriting identities of racial/cultural stereotypes with identities shaped by the first person voice enunciated in the Third Space, to produce identities of cultural hybridities. Semiotic codes of embedded "Black" and "Indian" stereotypes in each song's musical and lyrical text will be read and shown to be overwritten by the semiotic codes of the first person voice, which are decoded with the aid of postcolonial concepts such as "ambivalence," "hybridity" and "enunciation."

Problème: Ma thèse porte sur l’identité individuelle comme interrogation sur les enjeux personnels et sur ce qui constitue l’identification hybride à l’intérieur des notions concurrentielles en ce qui a trait à l’authenticité. Plus précisément, j’aborde le concept des identifications hybrides en tant que zones intermédiaires pour ce qui est de l’alternance de codes linguistiques et comme négociation des espaces continuels dans leur mouvement entre les cultures et les langues. Une telle négociation engendre des tensions et/ou apporte le lien créatif. Les tensions sont inhérentes à n’importe quelle construction d’identité où les lignes qui définissent des personnes ne sont pas spécifiques à une culture ou à une langue, où des notions de l’identité pure sont contestées et des codes communs de l’appartenance sont compromis. Le lien créatif se produit dans les exemples où l’alternance de code linguistique ou la négociation des espaces produit le mouvement ouvert et fluide entre les codes de concurrence des références et les différences à travers les discriminations raciales, la sexualité, la culture et la langue. Les travaux que j’ai sélectionnés représentent une section transversale de quelques auteurs migrants provenant de la minorité en Amérique du Nord qui alternent les codes linguistiques de cette manière. Les travaux détaillent le temps et l’espace dans leur traitement de l’identité et dans la façon dont ils cernent l’hybridité dans les textes suivants : The Woman Warrior de Maxine Hong Kingston (1975-76), Hunger of Memory de Richard Rodriguez (1982), Comment faire l’amour avec un nègre sans se fatiguer de Dany Laferrière (1985), Borderlands/La Frontera de Gloria Anzalduá (1987), Lost in Translation de Eva Hoffman (1989), Avril ou l’anti-passion de Antonio D’Alfonso (1990) et Chorus of Mushrooms de Hiromi Goto (1994). Enjeux/Questions La notion de l’identification hybride est provocante comme sujet. Elle met en question l’identité pure. C’est un sujet qui a suscité beaucoup de discussions tant en ce qui a trait à la littérature, à la politique, à la société, à la linguistique, aux communications, qu’au sein même des cercles philosophiques. Ce sujet est compliqué parce qu’il secoue la base des espaces fixes et structurés de l’identité dans sa signification culturelle et linguistique. Par exemple, la notion de patrie n’a pas les représentations exclusives du pays d’origine ou du pays d’accueil. De même, les notions de race, d’appartenance ethnique, et d’espaces sexuels sont parfois négativement acceptées si elles proviennent des codes socialement admis et normalisés de l’extérieur. De tels codes de la signification sont souvent définis par l’étiquette d’identification hétérosexuelle et blanche. Dans l’environnement généralisé d’aujourd’hui, plus que jamais, une personne doit négocier qui elle est, au sens de son appartenance à soi, en tant qu’individu et ce, face aux modèles locaux, régionaux, nationaux, voire même globaux de la subjectivité. Nous pouvons interpréter ce mouvement comme une série de couches superposées de la signification. Quand nous rencontrons une personne pour la première fois, nous ne voyons que la couche supérieure. D’ailleurs, son soi intérieur est caché par de nombreuses couches superposées (voir Joseph D. Straubhaar). Toutefois, sous cette couche supérieure, on retrouve beaucoup d’autres couches et tout comme pour un oignon, on doit les enlever une par une pour que l’individualité complète d’une personne soit révélée et comprise. Le noyau d’une personne représente un point de départ crucial pour opposer qui elle était à la façon dont elle se transforme sans cesse. Sa base, ou son noyau, dépend du moment, et comprend, mais ne s’y limite pas, ses origines, son environnement et ses expériences d’enfance, son éducation, sa notion de famille, et ses amitiés. De plus, les notions d’amour-propre et d’amour pour les autres, d’altruisme, sont aussi des points importants. Il y a une relation réciproque entre le soi et l’autre qui établit notre degré d’estime de soi. En raison de la mondialisation, notre façon de comprendre la culture, en fait, comment on consomme et définit la culture, devient rapidement un phénomène de déplacement. À l’intérieur de cette arène de culture généralisée, la façon dont les personnes sont à l’origine chinoises, mexicaines, italiennes, ou autres, et poursuivent leur évolution culturelle, se définit plus aussi facilement qu’avant. Approche Ainsi, ma thèse explore la subjectivité hybride comme position des tensions et/ou des relations créatrices entre les cultures et les langues. Quoique je ne souhaite aucunement simplifier ni le processus, ni les questions de l’auto-identification, il m’apparaît que la subjectivité hybride est aujourd’hui une réalité croissante dans l’arène généralisée de la culture. Ce processus d’échange est particulièrement complexe chez les populations migrantes en conflit avec leur désir de s’intégrer dans les nouveaux espaces adoptés, c’est-à-dire leur pays d’accueil. Ce réel désir d’appartenance peut entrer en conflit avec celui de garder les espaces originels de la culture définie par son pays d’origine. Ainsi, les références antérieures de l’identification d’une personne, les fondements de son individualité, son noyau, peuvent toujours ne pas correspondre à, ou bien fonctionner harmonieusement avec, les références extérieures et les couches d’identification changeantes, celles qu’elle s’approprie du pays d’accueil. Puisque nos politiques, nos religions et nos établissements d’enseignement proviennent des représentations nationales de la culture et de la communauté, le processus d’identification et la création de son individualité extérieure sont formées par le contact avec ces établissements. La façon dont une personne va chercher l’identification entre les espaces personnels et les espaces publics détermine ainsi le degré de conflit et/ou de lien créatif éprouvé entre les modes et les codes des espaces culturels et linguistiques. Par conséquent, l’identification des populations migrantes suggère que la « community and culture will represent both a hybridization of home and host cultures » (Straubhaar 27). Il y a beaucoup d’écrits au sujet de l’hybridité et des questions de l’identité et de la patrie, toutefois cette thèse aborde la valeur créative de l’alternance de codes culturels et linguistiques. Ce que la littérature indiquera Par conséquent, la plate-forme à partir de laquelle j’explore mon sujet de l’hybridité flotte entre l’interprétation postcoloniale de Homi Bhabha concernant le troisième espace hybride; le modèle d’hétéroglossie de Mikhail Bakhtine qui englobent plusieurs de mes exemples; la représentation de Roland Barthes sur l’identité comme espace transgressif qui est un modèle de référence et la contribution de Chantal Zabus sur le palimpseste et l’alternance de codes africains. J’utilise aussi le modèle de Sherry Simon portant sur l’espace urbain hybride de Montréal qui établit un lien important avec la valeur des échanges culturels et linguistiques, et les analyses de Janet Paterson. En effet, la façon dont elle traite la figure de l’Autre dans les modèles littéraires au Québec fournisse un aperçu régional et national de l’identification hybride. Enfin, l’exploration du bilinguisme de Doris Sommer comme espace esthétique et même humoristique d’identification situe l’hybridité dans une espace de rencontre créative. Conséquence Mon approche dans cette thèse ne prétend pas résoudre les problèmes qui peuvent résulter des plates-formes de la subjectivité hybride. Pour cette raison, j’évite d’aborder toute approche politique ou nationaliste de l’identité qui réfute l’identification hybride. De la même façon, je n’amène pas de discussion approfondie sur les questions postcoloniales. Le but de cette thèse est de démontrer à quel point la subjectivité hybride peut être une zone de relation créatrice lorsque l’alternance de codes permet des échanges de communication plus intimes entre les cultures et les langues. C’est un espace qui devient créateur parce qu’il favorise une attitude plus ouverte vis-à-vis les différents champs qui passent par la culture, aussi bien la langue, que la sexualité, la politique ou la religion. Les zones hybrides de l’identification nous permettent de contester les traditions dépassées, les coutumes, les modes de communication et la non-acceptation, toutes choses dépassées qui emprisonnent le désir et empêchent d’explorer et d’adopter des codes en dehors des normes et des modèles de la culture contenus dans le discours blanc, dominant, de l’appartenance culturelle et linguistique mondialisée. Ainsi, il appert que ces zones des relations multi-ethniques exigent plus d’attention des cercles scolaires puisque la population des centres urbains à travers l’Amérique du Nord devient de plus en plus nourrie par d’autres types de populations. Donc, il existe un besoin réel d’établir une communication sincère qui permettrait à la population de bien comprendre les populations adoptées. C’est une invitation à stimuler une relation plus intime de l’un avec l’autre. Toutefois, il est évident qu’une communication efficace à travers les frontières des codes linguistiques, culturels, sexuels, religieux et politiques exige une négociation continuelle. Mais une telle négociation peut stimuler la compréhension plus juste des différences (culturelle ou linguistique) si des institutions académiques offrent des programmes d’études intégrant davantage les littératures migrantes. Ma thèse vise à illustrer (par son choix littéraire) l’identification hybride comme une réalité importante dans les cultures généralisées qui croissent toujours aujourd’hui. Les espaces géographiques nous gardent éloignés les uns des autres, mais notre consommation de produits exotiques, qu’ils soient culturels ou non, et même notre consommation de l’autre, s’est rétrécie sensiblement depuis les deux dernières décennies et les indicateurs suggèrent que ce processus n’est pas une tendance, mais plutôt une nouvelle manière d’éprouver la vie et de connaître les autres. Ainsi les marqueurs qui forment nos frontières externes, aussi bien que ces marqueurs qui nous définissent de l’intérieur, exigent un examen minutieux de ces enjeux inter(trans)culturels, surtout si nous souhaitons nous en tenir avec succès à des langues et des codes culturels présents, tout en favorisant la diversité culturelle et linguistique. In this study, an attempt has been made to gather enough information regarding lactic acid bacteria from fish and shellfish of tropical regions. The occurrence and distribution of lactic acid bacteria in fresh and frozen marine fish and shellfish, farmed fish and shellfish, cured and pickled fish and shellfish have been investigated. Lactic Acid Bacteria (LAB) have for centuries been responsible for the fermentative preservation of many foods. They are used to retard spoilage and preserve foods through natural fermentations. They have found commercial applications as starter cultures in the dairy, baking, meat, fish, and vegetable and alcoholic beverage industries. They are industrially important organisms recognized for their fermentative ability as well as their nutritional benefits. These organisms produce various compounds such as organic acids, diacetyl, hydrogen peroxide and bacteriocins or bactericidal proteins during lactic fermentations.Biopreservation of foods using bacteriocin producing LAB cultures is becoming widely used. The antimicrobial effect of bacteriocins and other compounds produced during fermentation of carbohydrates are well known to inhibit the growth of certain food spoiling bacteria as well as a limited group of food poisoning and pathogenic bacteria LAB like Lactobacillus plantarum are widely used as starter cultures for the Production of fish ensilage. The present study is the first quantitative and qualitative study on the occurrence and distribution of lactic acid bacteria in fresh and frozen fish and prawn. It is concluded that Lactobacillus plantaruni was the predominant lactobacillus species in fresh and frozen fish and shellfish. The ability of selected Lactobacillus cultures to grow at low temperatures, high salt content, produce bacteriocins, rapidly ferment sugars and decrease the pH make them potential candidates for biopreservation of fish and shellfish.
