O presente estudo teve como objetivos efetuar a caracterização molecular e imunológica da infecção pelo HTLV em 42 portadores assintomáticos da infecção pelo HTLV; e 19 portadores com sintomas neurológicos associados à infecção (16 com PET/MAH e outros três com neuropatia periférica). Outro grupo de 100 indivíduos soronegativos para HTLV procedentes de Belém-PA também foi analisado. As amostras de sangue foram processadas para realização da sorologia para HTLV, para contagem de linfócitos T CD4+ e T CD8+ (incluindo os soronegativos), para as técnicas de quantificação da carga proviral do HTLV e caracterização dos tipos e subtipos de HTLV circulantes nos infectados. Entre os 42 assintomáticos, foi positiva para o HTLV-1 35 amostras (83.3%), e para o HTLV-2 07 amostras (16.7%) (p < 0.0001). Entre os 19 sintomáticos, foi positiva para o HTLV-1 18 amostras (94.7%), e para o HTLV-2 01 amostra (5.3%) (p = 0.0002), onde as 16 amostras que tiveram diagnóstico de PET/MAH foram positivas HTLV-1. As análises filogenéticas das regiões 5âLTR agruparam 34 amostras (60%) de HTLV-1 no Subgrupo Transcontinental do Subtipo Cosmopolita; e 05 amostras (72.2%) de HTLV-2, no subtipo HTLV-2c. As médias de distribuição dos níveis de linfócitos T CD4+ e T CD8+ foi maior entre os sintomáticos, porém não havendo diferenças significantes quando comparados com os assintomáticos e controles soronegativos. Foi observada uma maior média de carga proviral entre os portadores sintomáticos quando comparados aos assintomáticos (p = 0.0123). Os resultados obtidos confirmam a ocorrência de PET/MAH associada à infecção pelo HTLV-1 na região de Belém-PA. A predominância do subtipo A de HTLV-1 corrobora outros resultados que demonstram a presença deste subtipo como o mais prevalente em áreas urbanas do Brasil, assim como a predominância de HTLV-2c entre as infectadas pelo HTLV-2 confirma a maior frequência deste subtipo na Amazônia brasileira, ressaltando que dentre as amostras de HTLV-2 está a de um paciente sintomático (neuropatia periférica). A maior média de carga proviral entre sintomáticos corrobora resultados de achados que associam esta variável ao desenvolvimento de PET/MAH entre os infectados pelo HTLV. Sendo assim, estes resultados indicam ainda a necessidade do monitoramento da descrição de casos de infecção pelo HTLV com diagnóstico clínicolaboratorial adequado.

Fatores de risco e formas de transmissão do Vírus da Imunodeficiência humana (HIV) e o Vírus da hepatite B (VHB) são freqüentemente os mesmos, o qual explica a alta freqüência de co-infecção envolvendo esses dois agentes. O objetivo desta pesquisa foi estimar a prevalência da infecção pelo VHB, analisar possíveis fatores de risco bem como examinar a associação entre a contagem de linfócitos T CD4<sup>+</sup>, CD8<sup>+</sup> e carga viral plasmática em 129 portadores do HIV. Cada participante foi submetido a um questionário específico e tinha uma amostra de sangue testada para os marcadores sorológicos HBsAg, anti-HBc total, anti-HBs, contagem de linfócitos T CD4<sup>+</sup>, CD8<sup>+</sup> e carga viral plasmática. A prevalência total de marcadores para o VHB foi de 46,5%, com 3,1% de soropositividade para o HBsAg, 27,1% para o anti-HBc total e 36,4% para o anti-HBs. Após ajuste por regressão logística, os marcadores sorológicos para hepatite B foram associados com as seguintes variáveis: sexo, preferência sexual e escolaridade. A freqüência de marcadores para hepatite B foi de 39,1% em homens e 16,6% em mulheres. A prevalência de marcadores para hepatite B foi 14,4% em homo/bissexuais e 15,4% em heterossexuais. A menor freqüência de hepatite B (1%) foi encontrada em pessoas com nível superior de escolaridade. Não foram observadas diferenças estatisticamente significante entre contagem de linfócitos CD4<sup>+</sup>, CD8<sup>+</sup> e carga viral plasmática em pacientes co-infectados com VHB comparados com aqueles pacientes infectados somente com HIV.

Considering the importance of umbilical cord blood as a potential source of stem cell and, on the other hand, the use of the domestic swine (Sus scrofa) as a useful model for biomedical research in regenerative medicine and aiming to contribute about the quantification of lymphocyte subsets in umbilical cord blood and peripheral blood of newborn piglets, this study aimed to quantify CD4+, CD5+ and CD8+ cells from umbilical cord blood and peripheral blood from pigs at term blood samples. Were analyzed samples of the umbilical cord blood and peripheral of 48 piglets of Topigs lineage, from healthy mothers, artificially inseminated and natural birth. Blood samples were collected from the umbilical cord at birth, by the umbilical vein, and peripheral blood by venous sinus retro-ophthalmic. The immunological measurements of CD4+, CD5+ and CD8+ were obtained by flow cytometry. The relative average values for the CD4+, CD5+ e CD8+ counts in umbilical cord blood and peripheral blood of newborn piglets were inferior to those reported for peripheral blood in adult pigs, suggesting immunological immaturity. The ratio CD4+:CD8+ in umbilical cord blood (3.2±1.2%) and peripheral blood (3.2±1.7%) showed a predominance of TCD4+ over TCD8+. The percentage of CD4+ and CD8+ cells was 1.37±0.86% and 1.15±0.57%, respectively, in umbilical cord blood and peripheral blood.

Regulatory T (Treg) cells are fundamental in the control of immunity and excessive tissue pathology. In paracoccidioidomycosis, an endemic mycosis of Latin America, the immunoregulatory mechanisms that control the progressive and regressive forms of this infection are poorly known. Due to its modulatory activity on Treg cells, we investigated the effects of anti-CD25 treatment over the course of pulmonary infection in resistant (A/J) and susceptible (B10.A) mice infected with Paracoccidioides brasiliensis. We verified that the resistant A/J mice developed higher numbers and more potent Treg cells than susceptible B10.A mice. Compared to B10.A cells, the CD4(+)CD25(+)Foxp3(+) Treg cells of A/J mice expressed higher levels of CD25, CTLA4, GITR, Foxp3, LAP and intracellular IL-10 and TGF-beta. In both resistant and susceptible mice, anti-CD25 treatment decreased the CD4(+)CD25(+)Foxp3(+) Treg cell number, impaired indoleamine 2,3-dioxygenase expression and resulted in decreased fungal loads in the lungs, liver and spleen. In A/J mice, anti-CD25 treatment led to an early increase in T cell immunity, demonstrated by the augmented influx of activated CD4(+) and CD8(+) T cells, macrophages and dendritic cells to the lungs. At a later phase, the mild infection was associated with decreased inflammatory reactions and increased Th1/Th2/Th17 cytokine production. In B10.A mice, anti-CD25 treatment did not alter the inflammatory reactions but increased the fungicidal mechanisms and late secretion of Th1/Th2/Th17 cytokines. Importantly, in both mouse strains, the early depletion of CD25(+) cells resulted in less severe tissue pathology and abolished the enhanced mortality observed in susceptible mice. In conclusion, this study is the first to demonstrate that anti-CD25 treatment is beneficial to the progressive and regressive forms of paracoccidioidomycosis, potentially due to the anti-CD25-mediated reduction of Treg cells, as these cells have suppressive effects on the early T cell response in resistant mice and the clearance mechanisms of fungal cells in susceptible mice.

Leukotrienes (LTs) are lipid mediators involved in several inflammatory disorders. We investigated the LT pathway in human T-lymphotropic virus type 1 (HTLV-1) infection by evaluating LT levels in HTLV-1-infected patients classified according to the clinical status as asymptomatic carriers (HACs) and HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP) patients. Bioactive LTB4 and CysLTs were both increased in the plasma and in the supernatant of peripheral blood mononuclear cell cultures of HTLV-1-infected when compared to non-infected. Interestingly, CysLT concentrations were increased in HAM/TSP patients. Also, the concentration of plasma LTB4 and LTC4 positively correlated with the HTLV-1 proviral load in HTLV-1-infected individuals. The gene expression levels of LT receptors were differentially modulated in CD4(+) and CD8(+) T cells of HTLV-1-infected patients. Analysis of the overall plasma signature of immune mediators demonstrated that LT and chemokine amounts were elevated during HTLV-1 infection. Importantly, in addition to CysLTs, IP-10 was also identified as a biomarker for HAM/TSP activity. These data suggest that LTs are likely to be associated with HTLV-1 infection and HAM/TSP development, suggesting their putative use for clinical monitoring.

The expression of Langerhans cell (LC) and dermal dendritic cell (dDC) as well as T CD4+ and CD8+ immune responses was evaluated in the skin of BALB/c mice experimentally infected by L. (L.) amazonensis (La) and L. (V.) braziliensis (Lb). At 4th and 8th weeks post infection (PI), skin biopsies were collected to determine the parasite load and CD207+, CD11c+, CD4+, CD8+, iNOS+ cellular densities. Cytokine (IFN-?, IL-4 and IL-10) profiles were also analysed in draining lymph node. At 4th week, the densities of CD207+ and CD11c+ were higher in the La infection, while in the Lb infection, these markers revealed a significant increase at 8th week. At 4th week, CD4+ and CD8+ were higher in the La infection, but at 8th week, there was a substantial increase in both markers in the Lb infection. iNOS+ was higher in the Lb infection at 4th and 8th weeks. In contrast, the parasite load was higher in the La infection at 4th and 8th weeks. The concentration of IFN-? was higher in the Lb infection, but IL-4 and IL-10 were higher in the La infection at 4th and 8th weeks. These results confirm the role of the Leishmania species in the BALB/c mice disease characterized by differences in the expression of dendritic cells and cellular immune response.

One of the greatest challenges in urological oncology is renal cell carcinoma (RCC), which is the third leading cause of death in genitourinary cancers. RCCs are highly vascularized and respond positively to antiangiogenic therapy. Endostatin (ES) is a fragment of collagen XVIII that possesses antiangiogenic activity. In this study, we examined the potential of ES-based antiangiogenic therapy to activate tumor-associated endothelial cells in metastatic RCC (mRCC). Balb/c-bearing Renca cells were treated with NIH/3T3-LendSN or, as a control, with NIH/3T3-LXSN cells. The T-cell subsets and lymphocyte populations of tumors, mediastinal lymph nodes and the spleen were assessed by flow cytometry. The expression of intercellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) was assessed by real-time PCR, flow cytometry and immunohistochemistry analysis. ES gene therapy led to an increase in the percentage of infiltrating CD4-interferon (IFN)-gamma cells (P&lt;0.05), CD8-IFN-gamma cells (P&lt;0.01) and CD49b-tumor necrosis factor-alpha cells (P&lt;0.01). In addition, ES therapy caused an increase at the mRNA level of ICAM-1 (1.4-fold; P&lt;0.01) and VCAM-1 (1.5-fold) (control vs treated group; P&lt;0.001). Through flow cytometry, we found a significant increase in the CD34/ICAM-1 cells (8.1-fold; P&lt;0.001) and CD34/VCAM-1 cells (1.6-fold; P&lt;0.05). ES gene therapy induced a significant increase in both T CD4 and CD8 cells in the lymph nodes and the spleen, suggesting that ES therapy may facilitate cell survival or clonal expansion. CD49b cells were also present in increased quantities in all of these organs. In this study, we demonstrate an antitumor inflammatory effect of ES in an mRCC model, and this effect is mediated by an increase in ICAM-1 and VCAM-1 expression in tumor-associated endothelial cells.

The transcription factor B lymphocyte induced maturation protein-1 (Blimp-1) plays important roles in embryonic development and immunity. Blimp-1 is required for the differentiation of plasma cells, and mice with T cell specific deletion of Blimp-1 (Blimp-1CKO mice) develop a fatal inflammatory response in the colon. Previous work demonstrated that lack of Blimp-1 in CD4(+) and CD8(+) T cells leads to intrinsic functional defects, but little is known about the functional role of Blimp-1 in regulating differentiation of Th cells in vivo and their contribution to the chronic intestinal inflammation observed in the Blimp1CKO mice. In this study, we show that Blimp-1 is required to restrain the production of the inflammatory cytokine IL-17 by Th cells in vivo. Blimp-1CKO mice have greater numbers of IL-17 producing TCR beta(+)CD4(+)cells in lymphoid organs and in the intestinal mucosa. The increase in IL-17 producing cells was not restored to normal levels in wild-type and Blimp-1CKO mixed bone marrow chimeric mice, suggesting an intrinsic role for Blimp-1 in constraining the production of IL-17 in vivo. The observation that Blimp-1 deficient CD4(+) T cells are more prone to differentiate into IL-17(+)/IFN-gamma(+) cells and cause severe colitis when transferred to Rag1-deficient mice provides further evidence that Blimp-1 represses IL-17 production. Analysis of Blimp-1 expression at the single cell level during Th differentiation reveals that Blimp-1 expression is induced in Th1 and Th2 but repressed by TGF-beta in Th17 cells. BACKGROUND: Cellular immunity is the main defense mechanism in paracoccidioidomycosis (PCM), the most important systemic mycosis in Latin America. Th1 immunity and IFN-γ activated macrophages are fundamental to immunoprotection that is antagonized by IL-10, an anti-inflammatory cytokine. Both in human and experimental PCM, several evidences indicate that the suppressive effect of IL-10 causes detrimental effects to infected hosts. Because direct studies have not been performed, this study was aimed to characterize the function of IL-10 in pulmonary PCM. METHODOLOGY/PRINCIPAL FINDINGS: Wild type (WT) and IL-10(-/-) C57BL/6 mice were used to characterize the role of IL-10 in the innate and adaptive immunity against Paracoccidioides brasiliensis (Pb) infection. We verified that Pb-infected peritoneal macrophages from IL-10(-/-) mice presented higher phagocytic and fungicidal activities than WT macrophages, and these activities were associated with elevated production of IFN-γ, TNF-α, nitric oxide (NO) and MCP-1. For in vivo studies, IL-10(-/-) and WT mice were i.t. infected with 1Ã10(6) Pb yeasts and studied at several post-infection periods. Compared to WT mice, IL-10(-/-) mice showed increased resistance to P. brasiliensis infection as determined by the progressive control of pulmonary fungal loads and total clearance of fungal cells from dissemination organs. This behavior was accompanied by enhanced delayed-type hypersensitivity reactions, precocious humoral immunity and controlled tissue pathology resulting in increased survival times. In addition, IL-10(-/-) mice developed precocious T cell immunity mediated by increased numbers of lung infiltrating effector/memory CD4(+) and CD8(+) T cells. The inflammatory reactions and the production of Th1/Th2/Th17 cytokines were reduced at late phases of infection, paralleling the regressive infection of IL-10(-/-) mice. CONCLUSIONS/SIGNIFICANCE: Our work demonstrates for the first time that IL-10 plays a detrimental effect to pulmonary PCM due to its suppressive effect on the innate and adaptive immunity resulting in progressive infection and precocious mortality of infected hosts.

Interferon-gamma is mainly produced by activated T helper cells and cytotoxic T lymphocytes and sustains the immune-defense against viral and bacterial infections. For a better understanding of IFN-gamma promoter regulation in T cells, different DNA-binding motivs were examined. Hereby, a new motiv (-196 to -183) was identified, that binds to the transcription factor AP-1 in T helper cells and Jurkat T cells. This factor acts as an essential activator protein. Further investigation demonstrated that IL-12 and IL-18 induce different regulatory pathways. Both AP-1 and STAT-4 bindings at their cognate DNA elements (-196 to -183 and -224 to -215) are required for the IL-12 dependent activation whereas IL-18 causes direct activation via AP-1.Moreover, the TH2 cytokine IL-4 represses significantly the IFN-gamma promoter activity in CD4+ T cells. IL-4 induces GATA-3, that interacts with two DNA-motivs (-111 to -87) at the IFN-gamma promoter.Furthermore, transgenic mice were generated, yielding a human IFN-gamma promoter construct (410 bp) under the control of a luciferase reporter gene. The data demonstrated a specific IFN-gamma promoter activation by antiCD3 plus antiCD28 in CD4+ and CD8+ T cells. The luciferase activty in CD4+ T cells was reinforced by addition of IL-12 and IL-18 and repressed by IL-4.
