Este estudo, realizado nas condições do Cerrado do Planalto Central, teve porobjetivo avaliar o efeito da época de suspensão das irrigações por aspersão naprodução, qualidade de frutos e uso de água do tomateiro para processamento. Ostratamentos consistiram de quatorze épocas de suspensão, espaçadas de 7 dias,desde o florescimento até a colheita. A maior produtividade de frutos foi obtidaquando as irrigações foram suspensas 21 dias antes da colheita, com 10% dos frutosmaduros. Houve aumento linear do teor de sólidos solúveis totais a uma taxa de0,34 oBrix para cada 10 dias de antecipação da última irrigação. O máximorendimento de polpa foi obtido suspendendo as irrigações 34 dias antes da colheita,ou seja, quando 20% das plantas apresentaram pelo menos um fruto maduro. A maioreficiência de uso de água, relativa à produtividade de frutos e rendimento de polpa,foi observada quando a última irrigação ocorreu aos 37 e 45 dias após oflorescimento, respectivamente

A irrigação por gotejamento é uma tecnologia poupadora de água e mitigadora de problemas fitossanitários que começa a ser adotada na produção de tomate para processamento em áreas de cerrado de Minas Gerais e Goiás. Visando estabelecer estratégia de manejo de água durante os estádios vegetativo, de frutificação e de maturação do tomateiro, quatro experimentos foram conduzidos na Embrapa Hortaliças, Brasília, DF. Avaliou-se tensões-limite de água no solo entre 5 e 120 kPa. Plantas submetidas a maior déficit hídrico, durante o estádio vegetativo, apresentaram sistema radicular até 10 cm mais profundo que aquelas irrigadas mais freqüentemente. Maior produtividade de frutos foi atingida quando as irrigações foram realizadas de forma a não permitir tensões de água no solo superiores a 35, 12 e 15 kPa durante os estádios vegetativo, de frutificação e de maturação, respectivamente. O teor de sólidos solúveis totais não foi afetado pelos regimes de irrigação avaliados durante os estádios vegetativo e de frutificação, mas aumentou quanto maior o déficit hídrico durante o estádio de maturação.

Objective: To evaluate the histomorphometry and expression of Ki-67 and c-kit in ovarian follicles of pinealectomized or melatonin-treated pinealectomized rats.Study design: Forty adult rats were randomly divided into four groups of 10 animals: Group I - control; Group II - sham-pinealectomized; Group III - pinealectomized (Px), and Group IV - Px treated with melatonin (10 mu g/night, per animal). After two months' treatment, on the night of proestrous, the animals were placed in metabolic cages for night urine collection and subsequent measurement of 6-sulfatoxymelatonin (6-SMT). the rats were anesthetized, blood samples were taken for estrogen and progesterone determinations, and they were then euthanized. the ovaries were dissected out for further histological and immunohistochemical analyses. Data were first submitted to analysis of variance (ANOVA) complemented with the Tukey-Kramer test for multiple comparisons (P < 0.05).Results: the urinary levels of 6-SMT and serum progesterone were lower in the Px group (GIII). Exogenous melatonin treatment restored both blood melatonin and 6-SMT urinary levels. the histomorphometric data in Group III revealed a significant increase of degenerating antral and nonantral follicles with regard to the other groups. in addition no corpora lutea were observed in this group. No significant differences were noticed regarding the number of corpora lutea among the other groups (I, II and IV), but the number of cells and the thickness of the theca interna of Px animals (Group III) were higher than in the other groups. Conversely, the density of progesterone receptors (fmol/g) in the ovaries of Group III was significantly lower than in the other groups.Conclusion: Our data indicate that melatonin exerts a role on the maintenance of a proper follicular function, and is thus important for ovulation and progesterone production. O zooplâncton, particularmente os cladóceros, são organismos de água doce importantes na regulação da produção primária dos ecossistemas de água doce. No entanto, também podem adaptar-se a condições salobras. Tendo em conta as previsões no âmbito das alterações climáticas, a intrusão salina pode ocorrer a par com a subida de temperatura. As populações de água doce podem ficar vulneráveis aos efeitos interativos da salinidade e da temperatura, de acordo com os seus limites de tolerância e capacidade de adaptação ao stress ambiental. Assim, a presente tese analisou as interações resultantes das alterações destes agentes de stress em populações de cladóceros de água doce. Primeiro, comparou-se a halotolerância de diferentes genótipos de Simocephalus vetulus provenientes de populações de água doce e de água salobra de modo a avaliar a existência de uma componente genética de resistência à salinidade. A sensibilidade aguda dos genótipos variou na mesma gama de concentrações; todavia, todos os genótipos da população salobra, exceto um, foram mais tolerantes do que os de água doce, em termos de tempo à imobilização. Contudo, não foi possível estabelecer uma relação entre a performance reprodutiva em condições salobras e o contexto ambiental de origem destes genótipos. Mais, estes ensaios mostraram que as populações de água doce têm potencial para tolerar incrementos de salinidade. Como tal, pode-se concluir que a seleção a que os genótipos estão sujeitos no seu local de origem foi mais fraca do que o esperado. Segundo, investigou-se a capacidade de aclimatação de Daphnia galeata à salinidade e temperatura, de modo a avaliar a halotolerância de Daphnia a duas temperaturas num cenário de aclimatação multigeracional. O objetivo foi compreender se a pré-adaptação ao stress ambiental (20ºC e 25ºC versus 0 g/L e 1 g/L de NaCl) influenciou posteriormente as respostas a estes agentes de stress. Verificou-se uma tendência para um aumento de sensibilidade ao NaCl, a temperaturas mais elevadas. No entanto, este efeito foi anulado após nove gerações, mas apenas quando os organismos foram aclimatados aos dois agentes de stress em simultâneo (salinidade e temperatura elevada). Terceiro, demonstrou-se experimentalmente que a salinidade interferiu com a competição interespecífica, alterando a composição das comunidades zooplanctónicas. Este conjunto de evidências permitiu-nos refletir nos múltiplos impactos de agentes de stress, particularmente os relacionados com as previsões de alterações climáticas. Em paralelo aos estudos de natureza experimental, e numa perspetiva de Educação para o Desenvolvimento Sustentável (EDS), importa também promover o desenvolvimento de competências necessárias à compreensão de mudanças ambientais globais (e.g., o impacto da salinidade e da temperatura) para implementar estratégias de mitigação e adaptação. Neste contexto, foi realizada uma atividade com estudantes do ensino secundário, que se tornou uma boa oportunidade para a sua aprendizagem e aquisição de competências de interpretação de dados experimentais, assim como de sensibilização para as questões ambientais.

Information Visualization is gradually emerging to assist the representation and comprehension of large datasets about Higher Education Institutions, making the data more easily understood. The importance of gaining insights and knowledge regarding higher education institutions is little disputed. Within this knowledge, the emerging and urging area in need of a systematic understanding is the use of communication technologies, area that is having a transformative impact on educational practices worldwide. This study focused on the need to visually represent a dataset about how Portuguese Public Higher Education Institutions are using Communication Technologies as a support to teaching and learning processes. Project TRACER identified this need, regarding the Portuguese public higher education context, and carried out a national data collection. This study was developed within project TRACER, and worked with the dataset collected in order to conceptualize an information visualization tool U-TRACER®. The main goals of this study related to: conceptualization of the information visualization tool U-TRACER®, to represent the data collected by project TRACER; understand higher education decision makers perception of usefulness regarding the tool. The goals allowed us to contextualize the phenomenon of information visualization tools regarding higher education data, realizing the existing trends. The research undertaken was of qualitative nature, and followed the method of case study with four moments of data collection.The first moment regarded the conceptualization of the U-TRACER®, with two focus group sessions with Higher Education professionals, with the aim of defining the interaction features the U-TRACER® should offer. The second data collection moment involved the proposal of the graphical displays that would represent the dataset, which reading effectiveness was tested by end-users. The third moment involved the development of a usability test to the UTRACER ® performed by higher education professionals and which resulted in the proposal of improvements to the final prototype of the tool. The fourth moment of data collection involved conducting exploratory, semi-structured interviews, to the institutional decision makers regarding their perceived usefulness of the U-TRACER®. We consider that the results of this study contribute towards two moments of reflection. The challenges of involving end-users in the conceptualization of an information visualization tool; the relevance of effective visual displays for an effective communication of the data and information. The second relates to the reflection about how the higher education decision makers, stakeholders of the U-TRACER® tool, perceive usefulness of the tool, both for communicating their institutions data and for benchmarking exercises, as well as a support for decision processes. Also to reflect on the main concerns about opening up data about higher education institutions in a global market.

Este trabajo explica la realidad brasileña de las favelas, en particular las que se ubican en la ciudad de Rio de Janeiro. Así mismo, se pretende ubicar al lector en la favela Rocinha, y en uno de los principales problemas que enfrentan sus habitantes: la violencia infantil, o violencia contra los niños. La incapacidad del Estado para dar solución a este problema ha llevado a instituciones como UNICEF a crear alternativas para mitigarlo, por lo que es importante analizar esa nueva agenda, traducida en el primer ciclo del programa "Plataforma de Centros Urbanos". Se ha escogido el trabajo de campo como herramienta metodológica para dar cuenta de tal realidad, y para llegar a la conclusión de que un problema social puede, efectivamente, modificar la determinación de una agenda institucional.

Methods used for lipid analysis in embryos and oocytes usually involve selective lipid extraction from a pool of many samples followed by chemical manipulation, separation and characterization of individual components by chromatographic techniques. Herein we report direct analysis by matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS) of single and intact embryos or oocytes from various species. Biological samples were simply moisturized with the matrix solution and characteristic lipid ( represented by phosphatidylcholines, sphingomyelins and triacylglycerols) profiles were obtained via MALDI-MS. As representative examples, human, bovine, sheep and fish oocytes, as well as bovine and insect embryos were analyzed. MALDI-MS is shown to be capable of providing characteristic lipid profiles of gametes and embryos and also to respond to modifications due to developmental stages and in vitro culture conditions of bovine embryos. Investigation in developmental biology of the biological roles of structural and reserve lipids in embryos and oocytes should therefore benefit from these rapid MALDI-MS profiles from single and intact species.-Ferreira, C. Cutoff lows (COLs) pressure systems climatology for the Southern Hemisphere (SH), between 10 degrees S and 50 degrees S, using the National Center for Environmental Prediction-National Center for Atmospheric Research (NCEP-NCAR) and the ERA-40 European Centre for Medium Range Weather Forecast (ECMWF) reanalyses are analyzed for the period 1979-1999. COLs were identified at three pressure levels (200, 300, and 500 hPa) using an objective method that considers the main physical characteristics of the conceptual model of COLs. Independently of the pressure level analyzed, the climatology from the ERA-40 reanalysis has more COLs systems than the NCEP-NCAR. However, both reanalyses present a large frequency of COLs at 300 hPa, followed by 500 and 200 hPa. The seasonality of COLs differs at each pressure level, but it is similar between the reanalyses. COLs are more frequent during summer, autumn, and winter at 200, 300, and 500 hPa, respectively. At these levels, they tend to occur around the continents, preferentially from southeastern Australia to New Zealand, the south of South America, and the south of Africa. To study the COLs at 200 and 300 hPa from a regional perspective, the SH was divided in three regions: Australia-New Zealand (60 E-130 W), South America (130 degrees W-20 degrees W), and southern Africa (20 degrees W-60 degrees E). The common COLs features in these sectors for both reanalyses are a short lifetime (similar to 80.0% and similar to 70.0% of COLs at 200 and 300 hPa, respectively, persisting for up to 3 days), mobility (similar to 70.0% and similar to 50% of COLs at 200 and 300 hPa, respectively, traveling distances of up to 1200 km), and an eastward propagation.

To analyze the differential recruitment of the raphe nuclei during different phases of feeding behavior, rats were subjected to a food restriction schedule (food for 2 h/day, during 15 days). The animals were submitted to different feeding conditions, constituting the experimental groups: search for food (MFS), food ingestion (MFI), satiety (MFSa) and food restriction control (MFC). A baseline condition (BC) group was included as further control. The MFI and MFC groups, which presented greater autonomic and somatic activation, had more FOS-immunoreactive (FOS-IR) neurons. The MFI group presented more labeled cells in the linear (LRN) and dorsal (DRN) nuclei; the MFC group showed more labeling in the median (MRN), pontine (PRN), magnus (NRM) and obscurus (NRO) nuclei; and the MFSa group had more labeled cells in the pallidus (NRP). The BC exhibited the lowest number of reactive cells. The PRN presented the highest percentage of activation in the raphe while the DRN the lowest. Additional experiments revealed few double-labeled (FOS-IR+ 5-HT-IR) cells within the raphe nuclei in the MFI group, suggesting little serotonergic activation in the raphe during food ingestion. These findings suggest a differential recruitment of raphe nuclei during various phases of feeding behavior. Such findings may reflect changes in behavioral state (e.g., food-induced arousal versus sleep) that lead to greater motor activation, and consequently increased FOS expression. While these data are consistent with the idea that the raphe system acts as gain setter for autonomic and somatic activities, the functional complexity of the raphe is not completely understood. Endurance exercise is known to enhance peripheral insulin sensitivity and reduce insulin secretion. However, it is unknown whether the latter effect is due to the reduction in plasma substrate availability or alterations in beta-cell secretory machinery. Here, we tested the hypothesis that endurance exercise reduces insulin secretion by altering the intracellular energy-sensitive AMP-activated kinase (AMPK) signaling pathway. Male Wistar rats were submitted to endurance protocol training one, three, or five times per week, over 8 weeks. After that, pancreatic islets were isolated, and glucose-induced insulin secretion (GIIS), glucose transporter 2 (GLUT2) protein content, total and phosphorylated calmodulin kinase kinase (CaMKII), and AMPK levels as well as peroxisome proliferator-activated receptor-gamma coactivator-1-alpha (PGC-1 alpha) and uncoupling protein 2 (UCP2) content were measured. After 8 weeks, chronic endurance exercise reduced GIIS in a dose-response manner proportionally to weekly exercise frequency. Contrariwise, increases in GLUT2 protein content, CaMKII and AMPK phosphorylation levels were observed. These alterations were accompanied by an increase in UCP2 content, probably mediated by an enhancement in PGC-1 alpha protein expression. In conclusion, chronic endurance exercise induces adaptations in beta-cells leading to a reduction in GIIS, probably by activating the AMPK signaling pathway.
