The crystal structure of [HgCl2(Pyo)](n) (Pyo = pyridazine, C4H4N2) consists of chloride-bridged strands of octahedrally coordinated mercuric centers, connected by the two neighboring N atoms of pyridazine molecules. All atoms lie in special positions:Hg with site symmetry 2/m and the others on mirror planes.

Supersolitons are a form of soliton characterised, inter alia, by additional local extrema superimposed on the usual bipolar electric field signature. Previous studies of supersolitons supported by three-component plasmas have dealt with ion-acoustic structures. An analogous problem is now considered, namely, dust-acoustic supersolitons in a plasma composed of fluid negative dust grains and two kappa-distributed positive ion species. Calculations illustrating some supersoliton characteristics are presented. © Cambridge University Press 2013.

Interaction of a stream of high-energy electrons with the background plasma plays an important role in the astrophysical phenomena such as interplanetary and stellar bow shock and Earth's foreshock emission. It is not yet fully understood how electrostatic solitary waves are produced at the bow shock. Interestingly, a population of energetic suprathermal electrons were also found to exist in those environments. Previously, we have studied the properties of negative electrostatic potential solitary structures exist in such a plasma with excess suprathermal electrons. In the present study, we investigate the existence conditions and propagation properties of electron-acoustic solitary waves in a plasma consisting of an electron beam fluid, a cold electron fluid, and hot suprathermal electrons modeled by a kappa-distribution function. The Sagdeev pseudopotential method was used to investigate the occurrence of stationary-profile solitary waves. We have determined how the electron-acoustic soliton characteristics depend on the electron beam parameters. It is found that the existence domain for solitons becomes narrower with an increase in the suprathermality of hot electrons, increasing the beam speed, decreasing the beam-to-cold electron population ratio. These results lead to a better understanding of the formation of electron-acoustic solitary waves observed in those space plasma systems characterized by kappa-distributed electrons and inertial drifting (beam) electrons.

We investigate the existence conditions and propagation properties of electron-acoustic solitary waves in a plasma consisting of an electron beam fluid, a cold electron fluid, and a hot suprathermal electron component modeled by a k-distribution function. The Sagdeev pseudopotential method was used to investigate the occurrence of stationary-profile solitary waves. We have determined how the soliton characteristics depend on the electron beam parameters. It is found that the existence domain for solitons becomes narrower with an increase in the suprathermality of hot electrons, increasing the beam speed, and decreasing the beam-to-cold electron population ratio.

Objective: To investigate whether intakes of Ca, vitamin D, casein and whey are associated with periodontitis and to investigate the possibility of interactions between them. Design: Cross-sectional study. An Internet-based, 267-item FFQ was used to assess dietary intake. Intakes of casein (32·0 g/d), whey proteins (9·6 g/d) and vitamin D (5·8 μg/d) were classified as within v. above the 50th percentile. Ca intake was classified as within v. below age-specific recommendations. Severe periodontitis was defined as having ≥2 inter-proximal sites with clinical attachment loss ≥6 mm (not on the same tooth) and ≥1 inter-proximal site with pocket depth ≥5 mm. Since vitamin D influences Ca absorption, models were stratified by lower and higher (<5·8 v. ≥5·8 µg/d) vitamin D intake. Setting Danish Health Examination Survey (DANHES) 2007–2008. Subjects Adult participants (n 3287) in the oral health study of DANHES 2007–2008. Results Intakes of Ca within recommendations (OR=0·76; 95 % CI 0·58, 0·99), whey ≥9·6 g/d (OR=0·75; 95 % CI 0·58, 0·97) and casein ≥32 g/d (OR=0·75 95 % CI 0·58, 0·97) were associated with lower likelihood of severe periodontitis after adjustment for age, gender, education, smoking, sucrose intake, alcohol consumption, number of teeth, daily brushing, regular visits to the dentist and chronic illness, irrespective of vitamin D intake levels. Intake of vitamin D alone was not associated severe with periodontitis. Conclusions Intakes of Ca, casein and whey protein were inversely associated with periodontitis. Consumption of foods rich in Ca, casein and whey (e.g. dairy foods) should be promoted, as they may contribute to the prevention of periodontitis. Further longitudinal studies are required to confirm these associations.

Le cancer de la prostate (CaP) est le plus diagnostiqué chez les hommes au Canada et représente le troisième cancer le plus meurtrier au sein de cette population. Malgré l’efficacité des traitements de première ligne, de nombreux patients finiront par développer une résistance et, le cas échéant, verront leur CaP progresser vers une forme plus agressive. Plusieurs paramètres, essentiellement cliniques, permettent de prédire la progression du CaP mais leur sensibilité, encore limitée, implique la nécessité de nouveaux biomarqueurs afin de combler cette lacune. Dans cette optique nous nous intéressons au facteur de transcription NF-κB. Des études réalisées au laboratoire et ailleurs, associent RelA(p65) à un potentiel clinique dans le CaP, soulignant ainsi l’importance de la voie classique NF-κB. L’implication de la voie alternative NF-κB dans la progression du CaP a aussi été suggérée dans une de nos études illustrant la corrélation entre la distribution nucléaire de RelB et le score de Gleason. Alors que la voie classique est largement documentée et son implication dans la progression du CaP établie, la voie alternative, elle, reste à explorer. La présente thèse vise à clarifier l’implication de la voie alternative NF-κB dans le CaP et répond à deux objectifs fixés dans ce but. Le premier objectif fut d’évaluer l’impact de l'activation de la voie alternative NF-κB sur la biologie des cellules cancéreuses prostatiques. L’étude de la surexpression de RelB a souligné les effets de la voie alternative NF-κB sur la prolifération et l'autophagie. Étant ainsi impliquée tant dans la croissance tumorale que dans un processus de plus en plus associée à la progression tumorale, quoique potentiellement létal pour les cellules cancéreuses, son impact sur la tumorigénèse du CaP reste encore difficile à définir. Il n'existe, à ce jour, aucune étude permettant de comparer le potentiel clinique des voies classique et alternative NF-κB. Le second objectif de ce projet fut donc l'analyse conjointe de RelA(p65) et RelB au sein de mêmes tissus de patients atteints de CaP afin de déterminer l'importance clinique des deux signalisations NF-κB, l'une par rapport à l'autre. Le marquage immunofluorescent de RelA(p65) et RelB en a permis l'analyse quantitative et objective par un logiciel d'imagerie. Nos travaux ont confirmé le potentiel clinique associé à RelA(p65). La variable RelA(p65)/RelB s’est, elle, avérée moins informative que RelA(p65). Par contre, aucune corrélation entre RelB et les paramètres cliniques inclus dans l'étude n’est ressortie. En définitive, mon projet de thèse aura permis de préciser l'implication de la voie alternative NF-κB sur la biologie du CaP. Son impact sur la croissance des cellules cancéreuses prostatiques ainsi que sur l'autophagie, dénote l’ambivalence de la voie alternative NF-κB face à la tumorigénèse du CaP. L’étude exhaustive de la signalisation NF-κB souligne davantage l'importance de la voie classique dont l’intérêt clinique est principalement associé au statut de RelA(p65). Ainsi, bien que RelB n’affiche aucun potentiel en tant que biomarqueur exploitable en clinique, l’analyse de l’intervention de la voie alternative NF-κB sur la biologie des cellules cancéreuses prostatiques reste d’intérêt pour la compréhension de son rôle exact dans la progression du CaP.

Introduction: La circulation extracorporelle (CEC) peut entraîner une dysfonction endothéliale pulmonaire et l’hypertension pulmonaire. Le SN50 agit au niveau de la signalisation cellulaire pour prévenir ces réactions à la CEC et pourrait renverser la dysfonction endothéliale pulmonaire post-CEC sans effets néfastes sur l’hémodynamie. Méthodes: Quatre groups de porcs ont reçu un parmi quatre traîtements avant de subir 90 minutes de CEC et 60 minutes de reperfusion: (1) milrinone nébulisé; (2) sildenafil nébulisé; (3) placebo nébulisé; et (4) SN-50 intraveineux. Un monitoring hémodynamique invasif a été utilisé. La réactivité vasculaire des artères pulmonaires de deuxième ordre a été évaluée face à l’acétylcholine et la bradykinine. Résultats: Le sildénafil produit une augmentation significative de la pression de l’artère pulmonaire (PAP) moyenne à 60 minutes de reperfusion par rapport au début de la chirurgie. Les relaxations dépendantes de l’endothélium face à la bradykinine étaient meilleurs dans les groupes milrinone et SN-50 et surtout dans le groupe sildénafil par rapport au groupe placébo. Le SN-50 produisait de moins bonnes relaxations dépendantes de l’endothélium face à l’acétylcholine que les autres traitements incluant placébo. Conclusion: Le sildénafil prévient mieux la dysfonction endothéliale pulmonaire que les autres traitements. Les bénéfices du SN-50 sont possiblement sous-estimés vu que la dose n’a pas pu être ajustée à la durée de CEC. Le sildenafil inhalé mérite une étude plus importante chez l’humain et le SN-50 dans un model de CEC animal.

The total calcium level of raw skimmed milk was reduced by 10, 19, 29, 40 and 51% using Duolite® ion-exchange resin. The products were examined for concentrations of ionic calcium, sodium and potassium and the pH, ethanol stability, micelle diameter and ζ-potential were also measured. Ionic calcium decreased with removal of calcium and pH increased. Calcium removal resulted in an increase in the ethanol stability from 88% to above 100%. Casein micelle diameter increased as calcium was removed. The ζ-potential of the skimmed bulk milk was -24.4 mV, gradually becoming more negative with calcium removal to -30.6 mV after 51% calcium removal. The milk became more translucent as calcium was removed. To investigate the reversibility of this process, calcium chloride was added back to the depleted samples to restore their original total calcium content. At 51% removal, restoration of the total calcium level resulted in formation of clots. At levels of 10 and 19% calcium removal, the ethanol stability remained above 100%, but at higher levels of calcium removal the alcohol stability was adversely affected when the calcium was added back. Adding back calcium resulted in partial restoration of the original casein micelle diameter.

Different stabilising salts and calcium chloride were added to raw milk to evaluate changes in pH, ionic calcium, ethanol stability, casein micelle size and zeta potential. These milk samples were then sterilised at 121 °C for 15 min and stored for 6 months to determine how these properties changed. Addition of tri-sodium citrate (TSC) and di-sodium hydrogen phosphate (DSHP) to milk reduced ionic calcium, increased pH and increased ethanol stability in a concentration-dependent fashion. There was relatively little change in casein micelle size and a slight decrease in zeta potential. Sodium hexametaphosphate (SHMP) also reduced ionic calcium considerably, but its effect on pH was less noticeable. In contrast, sodium dihydrogen phosphate (SDHP) reduced pH but had little effect on ionic calcium. In-container sterilisation of these samples reduced pH, increased ethanol stability and increased casein micelle size, but had variable effects on ionic calcium; for DSHP and SDHP, ionic calcium decreased after sterilisation but, for SHMP, it remained little changed or increased. Milk containing 3.2 mM SHMP and more than 4.5 mM CaCl2 coagulated upon sterilisation. All other samples were stable but there were differences in browning, which increased in intensity as milk pH increased. Heat-induced sediment was not directly related to ionic calcium concentration, so reducing ionic calcium was not the only consideration in terms of improving heat stability. After 6 months of storage, the most acceptable product, in appearance, was that containing SDHP, as this minimised browning during sterilisation and further development of browning during storage.

The effects of specific nutrients on secretion and plasma concentrations of gut peptides (glucagon-like peptide-1((7-36)) amide (GLP-1), glucose-dependent insulinotropic polypeptide (GIP), and cholecystokinin-8 (CCK)) differ across species, but are not reported for cattle. Our objective was to determine acute (hours) and chronic (1 week) effects of increased abomasal supply of protein, carbohydrate, or fat to the small intestine on dry matter intake (DMI) and plasma concentrations of GLP-1, GIP, CCK, and insulin. Four mid-lactation Holstein cows were used in a 4 x 4 Latin square design experiment. Treatments were 7-day abomasal infusions of water, soybean oil (500 g/d), corn starch (1100 g/d), or casein (800 g/d). Jugular vein plasma was obtained over 7 h at the end of the first and last day of infusions. Oil infusion decreased DMI on day 7, but total metabolizable energy (ME) supply (diet plus infusate) did not differ from water infusion. Casein and starch infusion had no effect on feed DMI; thus, ME supply increased. Decreased DMI on day 7 of oil infusion was accompanied by increased plasma GLP-1 concentration, but decreased plasma CCK concentration. Increased plasma GIP concentration was associated with increased ME supply on day 7 of casein and starch infusion. Casein infusion tended to increase plasma CCK concentration on both days of sampling, and increased plasma GLP-1 and insulin concentration on day 1 of infusion. The present data indicate a sustained elevation of plasma concentration of GLP-1, but not CCK, may contribute to the reduced DMI observed in dairy cows provided supplemental fat. A survey against the draft genome sequence and the cDNA/EST database of Ciona intestinalis identified a number of genes encoding transcription factors regulating a variety of processes including development. In the present study, we describe almost complete sets of genes for Fox, ETS-domain transcription factors, nuclear receptors, and NFkappaB as well as other factors regulating NFkappaB activity, with their phylogenetic nature. Vertebrate Fox transcription factors are currently delineated into 17 subfamilies: FoxA to FoxQ. The present survey yielded 29 genes of this family in the Ciona genome, 24 of which were Ciona orthologues of known Fox genes. In addition, we found 15 ETS aenes, 17 nuclear receptor genes, and several NFkappaB signaling pathway genes in the Ciona genome. The number of Ciona genes in each family is much smaller than that of vertebrates, which represents a simplified feature of the ascidian genome. For example, humans have two NFkappaB genes, three Rel genes, and five NFAT genes, while Ciona has one gene for each family. The Ciona genome also contains smaller numbers of genes for the NFkappaB regulatory system, i.e. after the split of ascidians/vertebrates, vertebrates evolved a more complex NFkappaB system. The present results therefore provide molecular information for the investigation of complex developmental processes, and an insight into chordate evolution.
