El artículo refiere a una investigación doctoral en curso sobre el desarrollo de la comprensión infantil de la muerte, y se centra en los conceptos de lo real, lo posible y lo necesario de las últimas investigaciones piagetianas. Aunque se trata de una discusión teórica, se mencionan algunos estudios precedentes acerca de la comprensión de la muerte, puntuando algunos de sus problemas metodológicos vinculados con la distinción entre creencias infantiles sugeridas y espontáneas. Se presentan algunos datos de nuestras exploraciones piloto con niños de 5 a 10 años. Ellos se utilizan como ilustración en la discusión sobre la posibilidad de analizar el desarrollo del concepto de muerte desde la hipótesis 'tendiente a precisar los mecanismos productores de novedad' de la diferenciación gradual entre lo real, lo posible y lo necesario. Se señala que la aceptación de la universalidad, la inevitabilidad y la irreversibilidad de la muerte es una constante en todos los niños entrevistados. Asimismo, sus ideas acerca de la 'persistencia de la existencia' luego de la muerte, y de la 'relocalización' post mórtem en otro espacio, sugieren la presencia de mecanismos análogos a los hallados en otros estudios. Finalmente, la co-existencia de racionalidades diferentes en un mismo sujeto, parecen manifestar las complejas relaciones entre representaciones sociales y creencias espontáneas

El objetivo de este trabajo es discutir los enfoques principales del debate sobre el tratamiento que debe dar la economía al impacto ambiental. El tema se relaciona con toda la discusión sobre el desarrollo y sobre la relación entre territorio y desarrollo, y atraviesa la problemática de la geografía económica y regional en cuestiones como la planificación regional o el análisis de impacto, sobre todo en territorios con fuerte influencia de la explotación de recursos naturales. Se analiza la manera de incorporar el problema en las bases de la economía neoclásica aceptando sus principios y utilizando sus instrumentos. Luego se aborda la economía ecológica desde el trabajo de Georgescu-Roegen de 1971, el primero que intenta introducir en el razonamiento económico las leyes de la irreversibilidad de los procesos, por fuera de la visión neoclásica y fundando una corriente heterodoxa. Más allá de estas posiciones, están las visiones que analizan la relación entre ambiente y desarrollo manteniendo el concepto de centro-periferia o asumiendo la necesidad de construir discursos contra-hegemónicos, enmarcando la cuestión ambiental en esa lógica: se mencionan la postura del marxismo y el aporte del poscolonialismo -este último, fundamental, por la importancia que asigna a la necesidad de reconstruir la perspectiva ambiental desde las identidades y el territorio-. Las conclusiones plantean el desafío de retomar este tipo de puntos de vista -lo que limita enormemente el uso del herramental de la economía ambiental- y también algunas propuestas neoinstitucionalistas de la economía ecológica

The problem of interdependence between housing and commuting in a city has been analysed within the framework of welfare economics. Uncertain changes overtime in the working population has been considered by means of a dynamic, probabilistic model. The characteristics of irreversibility and durability in city building have been explicitly dealt with. The ultimate objective is that the model after further development will be an auxiliary tool in city planning.

As várias teorias acerca da estrutura de capital despertam interesse motivando diversos estudos sobre o assunto sem, no entanto, ter um consenso. Outro tema aparentemente pouco explorado refere-se ao ciclo de vida das empresas e como ele pode influenciar a estrutura de capital. Este estudo teve como objetivo verificar quais determinantes possuem maior relevância no endividamento das empresas e se estes determinantes alteram-se dependendo do ciclo de vida da empresa apoiada pelas teorias Trade Off, Pecking Order e Teoria da Agência. Para alcançar o objetivo deste trabalho foi utilizado análise em painel de efeito fixo sendo a amostra composta por empresas brasileiras de capital aberto, com dados secundários disponíveis na Economática® no período de 2005 a 2013, utilizando-se os setores da BM&FBOVESPA. Como resultado principal destaca-se o mesmo comportamento entre a amostra geral, alto e baixo crescimento pelo endividamento contábil para o determinante Lucratividade apresentando uma relação negativa, e para os determinantes Oportunidade de Crescimento e Tamanho, estes com uma relação positiva. Para os grupos de alto e baixo crescimento alguns determinantes apresentaram resultados diferentes, como a singularidade que resultou significância nestes dois grupos, sendo positiva no baixo crescimento e negativa no alto crescimento, para o valor colateral dos ativos e benefício fiscal não dívida apresentaram significância apenas no grupo de baixo crescimento. Para o endividamento a valor de mercado foi observado significância para o Benefício fiscal não dívida e Singularidade. Este resultado reforça o argumento de que o ciclo de vida influência a estrutura de capital.

NO2Tyr (3-Nitrotyrosine) is a modified amino acid that is formed by nitric oxide-derived species and has been implicated in the pathology of diverse human diseases. Nitration of active-site tyrosine residues is known to compromise protein structure and function. Although free NO2Tyr is produced in abundant concentrations under pathological conditions, its capacity to alter protein structure and function at the translational or posttranslational level is unknown. Here, we report that free NO2Tyr is transported into mammalian cells and selectively incorporated into the extreme carboxyl terminus of α-tubulin via a posttranslational mechanism catalyzed by the enzyme tubulin–tyrosine ligase. In contrast to the enzymatically regulated carboxyl-terminal tyrosination/detyrosination cycle of α-tubulin, incorporation of NO2Tyr shows apparent irreversibility. Nitrotyrosination of α-tubulin induces alterations in cell morphology, changes in microtubule organization, loss of epithelial-barrier function, and intracellular redistribution of the motor protein cytoplasmic dynein. These observations imply that posttranslational nitrotyrosination of α-tubulin invokes conformational changes, either directly or via allosteric interactions, in the surface-exposed carboxyl terminus of α-tubulin that compromises the function of this critical domain in regulating microtubule organization and binding of motor- and microtubule-associated proteins. Collectively, these observations illustrate a mechanism whereby free NO2Tyr can impact deleteriously on cell function under pathological conditions encompassing reactive nitrogen species production. The data also yield further insight into the role that the α-tubulin tyrosination/detyrosination cycle plays in microtubule function.

The cohesin-dockerin interaction in Clostridium thermocellum cellulosome mediates the tight binding of cellulolytic enzymes to the cellulosome-integrating protein CipA. Here, this interaction was used to study the effect of different cellulose-binding domains (CBDs) on the enzymatic activity of C. thermocellum endoglucanase CelD (1,4-β-d endoglucanase, EC3.2.1.4) toward various cellulosic substrates. The seventh cohesin domain of CipA was fused to CBDs originating from the Trichoderma reesei cellobiohydrolases I and II (CBDCBH1 and CBDCBH2) (1,4-β-d glucan-cellobiohydrolase, EC3.2.1.91), from the Cellulomonas fimi xylanase/exoglucanase Cex (CBDCex) (β-1,4-d glucanase, EC3.2.1.8), and from C. thermocellum CipA (CBDCipA). The CBD-cohesin hybrids interacted with the dockerin domain of CelD, leading to the formation of CelD-CBD complexes. Each of the CBDs increased the fraction of cellulose accessible to hydrolysis by CelD in the order CBDCBH1 < CBDCBH2 ≈ CBDCex < CBDCipA. In all cases, the extent of hydrolysis was limited by the disappearance of sites accessible to CelD. Addition of a batch of fresh cellulose after completion of the reaction resulted in a new burst of activity, proving the reversible binding of the intact complexes despite the apparent binding irreversibility of some CBDs. Furthermore, burst of activity also was observed upon adding new batches of CelD–CBD complexes that contained a CBD differing from the first one. This complementation between different CBDs suggests that the sites made available for hydrolysis by each of the CBDs are at least partially nonoverlapping. The only exception was CBDCipA, whose sites appeared to overlap all of the other sites.

How does a protease act like a hormone to regulate cellular functions? The coagulation protease thrombin (EC 3.4.21.5) activates platelets and regulates the behavior of other cells by means of G protein-coupled protease-activated receptors (PARs). PAR1 is activated when thrombin binds to and cleaves its amino-terminal exodomain to unmask a new receptor amino terminus. This new amino terminus then serves as a tethered peptide ligand, binding intramolecularly to the body of the receptor to effect transmembrane signaling. The irreversibility of PAR1’s proteolytic activation mechanism stands in contrast to the reversible ligand binding that activates classical G protein-coupled receptors and compels special mechanisms for desensitization and resensitization. In endothelial cells and fibroblasts, activated PAR1 rapidly internalizes and then sorts to lysosomes rather than recycling to the plasma membrane as do classical G protein-coupled receptors. This trafficking behavior is critical for termination of thrombin signaling. An intracellular pool of thrombin receptors refreshes the cell surface with naïve receptors, thereby maintaining thrombin responsiveness. Thus cells have evolved a trafficking solution to the signaling problem presented by PARs. Four PARs have now been identified. PAR1, PAR3, and PAR4 can all be activated by thrombin. PAR2 is activated by trypsin and by trypsin-like proteases but not by thrombin. Recent studies with knockout mice, receptor-activating peptides, and blocking antibodies are beginning to define the role of these receptors in vivo.

A 16-amino acid oligopeptide forms a stable β-sheet structure in water. In physiological solutions it is able to self-assemble to form a macroscopic matrix that stains with Congo red. On raising the temperature of the aqueous solution above 70°C, an abrupt structural transition occurs in the CD spectra from a β-sheet to a stable α-helix without a detectable random-coil intermediate. With cooling, it retained the α-helical form and took several weeks at room temperature to partially return to the β-sheet form. Slow formation of the stable β-sheet structure thus shows kinetic irreversibility. Such a formation of very stable β-sheet structures is found in the amyloid of a number of neurological diseases. This oligopeptide could be a model system for studying the protein conformational changes that occurs in scrapie or Alzheimer disease. The abrupt and direct conversion from a β-sheet to an α-helix may also be found in other processes, such as protein folding and protein–protein interaction. Furthermore, such drastic structure changes may also be exploited in biomaterials designed as sensors to detect environmental changes.

While the strong biotin-avidin interaction has been widely used for the detection of biomolecules, its irreversibility complicates their isolation. We report the synthesis of a photocleavable biotin derivative (PCB) which eliminates many limitations of existing methods. This reagent contains a biotin moiety linked through a spacer arm to a photocleavable moiety, which reacts selectively with primary amino groups on any substrate. In experiments using [leucine]-enkephalin as a model substrate, we show that PCB retains its high affinity toward avidin/streptavidin and allows rapid (< 5 min) and efficient (> 99%) photorelease of the substrate in a completely unaltered form. Photocleavable biotins should be useful in numerous applications involving the isolation of proteins, nucleic acids, lipids, and cells.

The Carnot cycle imposes a fundamental upper limit to the efficiency of a macroscopic motor operating between two thermal baths. However, this bound needs to be reinterpreted at microscopic scales, where molecular bio-motors and some artificial micro-engines operate. As described by stochastic thermodynamics, energy transfers in microscopic systems are random and thermal fluctuations induce transient decreases of entropy, allowing for possible violations of the Carnot limit. Here we report an experimental realization of a Carnot engine with a single optically trapped Brownian particle as the working substance. We present an exhaustive study of the energetics of the engine and analyse the fluctuations of the finite-time efficiency, showing that the Carnot bound can be surpassed for a small number of non-equilibrium cycles. As its macroscopic counterpart, the energetics of our Carnot device exhibits basic properties that one would expect to observe in any microscopic energy transducer operating with baths at different temperatures. Our results characterize the sources of irreversibility in the engine and the statistical properties of the efficiency-an insight that could inspire new strategies in the design of efficient nano-motors.

This paper aims to identify drivers of physical capital adjustments in agriculture. It begins with a review of some of the most important theories and modelling approaches regarding firms’ adjustments of physical capital, ranging from output-based models to more recent approaches that consider irreversibility and uncertainty. Thereafter, it is suggested that determinants of physical capital adjustments in agriculture can be divided into three main groups, namely drivers related to: i) expected (risk-adjusted) profit, ii) expected societal benefits and costs and iii) expected private nonpecuniary benefits and costs. The discussion that follows focuses on the determinants belonging to the first group and covers aspects related to product market conditions, technological conditions, financial conditions and the role of firm structure and organization. Furthermore, the role of subjective beliefs is emphasized. The main part of this paper is concerned with the demand side of the physical capital market and one section also briefly discusses some aspects related to supply of farm assets.

We demonstrate that the process of generating smooth transitions Call be viewed as a natural result of the filtering operations implied in the generation of discrete-time series observations from the sampling of data from an underlying continuous time process that has undergone a process of structural change. In order to focus discussion, we utilize the problem of estimating the location of abrupt shifts in some simple time series models. This approach will permit its to address salient issues relating to distortions induced by the inherent aggregation associated with discrete-time sampling of continuous time processes experiencing structural change, We also address the issue of how time irreversible structures may be generated within the smooth transition processes. A közgazdaság-tudomány számos problémája a fizika analóg modelljeinek segítségével nyert megoldást. A közgazdászok körében erőteljesen megoszlanak a vélemények, hogy a közgazdasági modellek mennyire redukálhatók a fizika, vagy más természettudományok eredményeire. Vannak,akik pontosan ezzel magyarázzák,hogy a mai mainstream közgazdasági elmélet átalakult alkalmazott matematikává,ami a gazdasági kérdéseket csak a társadalom-tudományi vonatkozásaitól eltekintve képes vizsgálni. Mások, e tanulmányszerzője is, viszont úgy vélekednek, hogy a közgazdasági problémák egy része, ahol lehetőség van a mérésre, jól modellezhetők a természettudományok technikai arzenáljával. A másik része, amelyekben nem lehet mérni,s tipikusan ilyenek a társadalomtudományi kérdések, ott sokkal komplexebb technikákra lesz szükség. Etanulmány célkitűzése, hogy felvázolja a fizika legújabb, az irreverzibilis dinamika, a relativitáselmélet és a kvantummechanika sztochasztikus matematikai összefüggéseit, amelyekből a közgazdászok választhatnak egy-egy probléma megfogalmazásában és megoldásában. Például az időoperátorok pontos értelmezése jelentős fordulatot hozhat a makroökonómiai elméletekben; vagy az eddigi statikus egyensúlyi referencia pontokat felválthatják a dinamikus,időben változó sztochasztikus egyensúlyi referenciafüggvények, ami forradalmian új megvilágításba helyezhet számos társadalomtudományi, s főleg nemegyensúlyi közgazdasági kérdést.A termodinamika és a biológiai evolúció fogalmait és definícióit Paul A. Samuelson (1947) már adaptálta a közgazdaságtanban, viszont a kvantummechanika legújabb eredményeit, az időoperátorokat stb. nem érintette. E cikk azokat a legújabb fizikai, kémiai és biológiai matematikai összefüggéseket foglalja össze,amelyek hasznosak lehetnek a közgazdasági modellek komplexebb megfogalmazásához. ___________________ The aim of this paper is to out line the newest results of physics,i.e.,the stochastic mathematical relations of relativity theory and quantum mechanics as well as irreversible dynamics which can be applied for some economic problems.For example,the correct interpretation of time operators using for the macroeconomic theories may provide a serious improvement in approach to the reality.The stochastic dynamic equilibrium reference functions will take over the role of recent static equilibrium reference points,which may also reveal some nonequilibrium questions of macroeconomics.The concepts and definitions of thermodynamics and biological evolution have been adopted in economics by Paul A. Samuelson, but he did not concern the newest results of quantum mechanics, e.g., the time operators. Now we do it.In addition, following Samuelson,we show that von Neumann growth model cannot be explained as a peculiar extension of thermodynamic irreversibility.
