La stéatose hépatique et la détérioration du profil lipidique plasmatique sont des pathologies métaboliques favorisées par la carence œstrogénique post-ménopausique. Cependant les mécanismes à la base de ces pathologies n’ont été que très peu étudiés. Le but de cette thèse a été d’investiguer les mécanismes moléculaires possibles à l’origine de l’hypercholestérolémie et de l’accumulation des lipides (triglycérides : TG et cholestérol) dans le foie en utilisant un modèle animal de la ménopause, la rate Sprague Dawley ovariectomisée (Ovx). Nous avons également examiné si le changement des habitudes de vie comme la pratique de l’exercice physique pouvait prévenir ou corriger les modifications induites par l’Ovx. Enfin, rosuvastatine (statine) a été utilisée comme thérapie pharmacologique de l’hypercholestérolémie dans le but de comprendre son effet au niveau moléculaire chez la rate Ovx. L’objectif de la première étude était de déterminer comment l’Ovx peut affecter les niveaux de TG et de cholestérol dans le foie des rates nourries avec une diète riche en lipides (HF : 42% gras). Les rates ont été soumises à la diète HF ou normale pendant 6 semaines avant d’être Ovx ou Sham (ovariectomie simulée), puis maintenues aux mêmes conditions diététiques pour 6 autres semaines. L’Ovx a provoqué une accumulation de TG dans le foie, mais pas la diète HF seule. Cependant, lorsque l’Ovx était combinée à la diète HF, l’accumulation des TG était beaucoup plus importante comparé à ce qui était observé chez les rates Ovx soumises à la diète normale. L’expression génique (ARNm) de CPT1 (Carnitine palmitoyltransferase 1), PGC1α (Peroxisome proliferator-activated receptor gamma, coactivator 1) et PPARα (Peroxysome proliferetor activated receptor alpha) intervenant dans l’oxydation des acides gras dans le foie était augmentée par la diète HF (p ˂ 0.001; p ˂ 0.01; p ˂ 0.05 respectivement) ; mais atténuée (p ˂ 0.05; p ˂ 0.05; p ˂ 0.07 respectivement) lorsque les rates ont été Ovx, favorisant ainsi l’accumulation des TG dans le foie. La combinaison de la diète HF à l’Ovx a également provoqué une hypercholestérolémie et une accumulation de cholestérol dans le foie malgré la diminution de l’expression de la HMGCoA-r (3-hydroxy-3-methylglutaryl-CoA reductase), enzyme clé de la synthèse du cholestérol. Ceci était associé à l’inhibition de l’expression génique de CYP7a1 (Cytochrome P450, family 7, subfamily a, polypeptide 1), suggérant une diminution de la synthèse des acides biliaires. Ayant constaté dans la première étude que l’Ovx élevait les niveaux de cholestérol hépatique et plasmatique, nous nous sommes fixés comme objectif dans la deuxième étude d’évaluer les effets de l’Ovx sur l’expression génique des transporteurs et enzymes responsables du métabolisme du cholestérol et des acides biliaires dans le foie et l’intestin, et de vérifier si l’exercice sur tapis roulant pouvait prévenir ou corriger les changements causés par l’Ovx. L’hypercholestérolémie constatée chez les rates Ovx comparativement aux Sham était accompagnée de la diminution de l’expression génique des récepteurs des LDL (R-LDL), des résidus de lipoprotéines (LRP1), de SREBP-2 (Sterol regulatory element binding protein 2) et de PCSK9 (Proprotein convertase subtilisin/kexin type 9) dans le foie, suggérant une défaillance dans la clairance des lipoprotéines plasmatiques. L’Ovx a aussi inhibé l’expression génique de la MTP (Microsomal triglyceride transfer protein) et stimulé celle de SR-B1 (Scavenger receptor class B, member 1); mais aucun changement n’a été observé avec CYP7a1. Ces changements moléculaires pourraient par conséquent favoriser l’accumulation de cholestérol dans le foie. L’exercice physique n’a pas corrigé les modifications causées par l’Ovx sur l’expression génique de ces molécules au niveau hépatique à l’exception de SREBP-2. Par contre, au niveau intestinal (iléum), l’exercice sur tapis roulant a inhibé l’expression génique des marqueurs moléculaires intervenant dans l’absorption des acides biliaires (OSTα/β, FXR, RXRα, Fgf15) et du cholestérol (LXRα, NCP1L1) au niveau de l’iléum chez les rates Sham entraînées. Ces adaptations pourraient prévenir le développement de l’hypercholestérolémie protégeant en partie contre la survenue de l’athérosclérose. Au vue des effets délétères (hypercholestérolémie et diminution de l’expression du R-LDL, PCSK9, LRP1, SREBP-2 et HMGCOA-r dans le foie) causés par l’Ovx sur le métabolisme du cholestérol constatés dans l’étude 2, la 3ième étude a été conçue pour évaluer l’efficacité de rosuvastatine (Ros) sur l’expression génique de ces marqueurs moléculaires chez les rates Ovx sédentaires ou soumises à l’entraînement volontaire. Ros a été administrée aux rates Ovx pendant 21 jours par voie sous-cutanée à la dose de 5mg/kg/j à partir de la 9ième semaine après l’Ovx. Ros n’a pas diminué la concentration plasmatique de LDL-C et de TC chez les rates Ovx. Par contre, Ros a stimulé (P ˂ 0.05) l’expression génique de PCSK9, SREBP-2, LRP1, HMGCoA-r et ACAT2 (Acyl-CoA cholesterol acyltransferase) mais pas significativement (P = 0.3) celle du R-LDL dans le foie des rates Ovx sédentaires et entraînées. Ros n’a pas réduit la concentration plasmatique de LDL-C probablement à cause de l’induction plus importante de PCSK9 par rapport au R-LDL. Cependant, la stimulation de LRP1 par Ros protège partiellement contre la survenue des maladies cardiovasculaires. En conclusion, les études de cette thèse indiquent que la baisse du niveau des œstrogènes entraîne des changements radicaux du métabolisme hépatique des TG et du cholestérol provoqués par des altérations de l’expression des gènes clés des voies métaboliques associées.

We have compared properties of roots from different lines (genotypes) of tobacco raised either in tissue culture or grown from seed. The different lines included unmodified plants and plants modified to express reduced activity of the enzyme cinnamoyl-CoA reductase, which has a pivotal role in lignin biosynthesis. The size and structure of the rhizosphere microbial community, characterized by adenosine triphosphate and phospholipid fatty acid analyses, were related to root chemistry (specifically the soluble carbohydrate concentration) and decomposition rate of the roots. The root material from unmodified plants decomposed faster following tissue culture compared with seed culture, and the faster decomposing material had significantly higher soluble carbohydrate concentrations. These observations are linked to the larger microbial biomass and greater diversity of the rhizosphere communities of tissue culture propagated plants.

Solvent-free desymmetrisation of meso-dialdehyde 1 with chiral 1-phenylethan-1-ol, led to preparation of 4-silyloxy-6-alkyloxytetrahydro-2H-pyran-2-one (+)-3a with a 96:4 d.r. Deprotected lactone (+)-19a and the related racemic lactones 16a-18a present a lactone moiety resembling the natural substrate of HMG-CoA reductase and their antifungal properties have been evaluated against the phytopathogenic fungi Botrytis cinerea and Colletotrichum gloeosporioides. These compounds were selectively active against B. cinerea, while inactive against C. gloeosporioides.

Background and Objective: Cyclosporine A treatment is important in the therapy of a number of medical conditions; however, alveolar bone loss is an important negative side-effect of this drug. As such, we evaluated whether concomitant administration of simvastatin would minimize cyclosporine A-associated alveolar bone loss in rats subjected, or not, to experimental periodontal disease. Material and Methods: Groups of 10 rats each were treated with cyclosporine A (10 mg/kg/day), simvastatin (20 mg/kg/day), cyclosporine A and simvastatin concurrently (cyclosporine A/simvastatin) or vehicle for 30 days. Four other groups of 10 rats each received a cotton ligature around the lower first molar and were treated similarly with cyclosporine A, simvastatin, cyclosporine A/simvastatin or vehicle. Calcium (Ca(2+)), phosphorus and alkaline phosphatase levels were evaluated in serum. Expression levels of interleukin-1 beta, prostaglandin E(2) and inducible nitric oxide synthase were evaluated in the gingivomucosal tissues. Bone volume and numbers of osteoblasts and osteoclasts were also analyzed. Results: Treatment with cyclosporine A in rats, with or without ligature, was associated with bone loss, represented by a lower bone volume and an increase in the number of osteoclasts. Treatment with cyclosporine A was associated with bone resorption, whereas simvastatin treatment improved cyclosporine A-associated alveolar bone loss in all parameters studied. In addition, simvastatin, in the presence of inflammation, can act as an anti-inflammatory agent. Conclusion: This study shows that simvastatin therapy leads to a reversal of the cyclosporine A-induced bone loss, which may be mediated by downregulation of interleukin-1 beta and prostaglandin E(2) production.

Considering that inflammation contributes to obesity-induced insulin resistance and that statins have been reported to have other effects beyond cholesterol lowering, the present study aimed to it whether atorvastatin treatment has anti-inflammatory action in white adipose tissue of obese mice, consequently improving insulin sensitivity. Insulin sensitivity in vivo (by insulin tolerance test); metabolic-hormonal profile; plasma tumor necrosis factor (TNF)-alpha, interleukin (IL)-6, and adiponectin; adipose tissue immunohistochemistry; glucose transporter (GLUT) 4; adiponectin; INF-alpha; IL-1 beta; and IL-6 gene expression; and I kappa B kinase (IKK)-alpha/beta activity were assessed in 23-week-old monosodium glutamate induced obese mice untreated or treated with atorvastatin for 4 weeks. Insulin-resistant obese mice had increased plasma triglyceride, insulin, TNF-alpha, and IL-6 plasma levels. Adipose tissue of obese animals showed increased macrophage infiltration, IKK-alpha (42%, P < .05) and IKK-beta (73%, P < .05) phosphorylation, and INF-alpha and IL-6 messenger RNA (mRNA) (similar to 15%, P < .05) levels, and decreased GLUT4 mRNA and protein (30%, P < .05) levels. Atorvastatin treatment lowered cholesterol, triglyceride, insulin, INF-alpha, and IL-6 plasma levels, and restored whole-body insulin sensitivity. In adipose tissue, atorvastatin decreased macrophage in and normalized IKK-alpha/beta phosphorylation; INF-alpha, IL-6, and GLUT4 mRNA; and GLUT4 protein to control levels. The present findings demonstrate that atorvastatin has anti-inflammatory effects on adipose tissue of obese mice, which may be important to its local and whole-body insulin-sensitization effects. Layered double hydroxide (LDH) nanocontainers, suitable as carriers for anionic drugs, were intercalated with Pravastatin drug using magnesium-aluminum and zinc-aluminum in a M-II/Al molar ratio equal 2 and different Al3+/Pravastatin molar ratios. Postsynthesis treatments were used in order to increase the materials crystallinity. Hybrid materials were characterized by a set of physical chemical techniques: chemical elemental analysis, X-ray diffraction (XRD), mass coupled thermal analyses, vibrational infrared and Raman spectroscopies, and solid-state C-13 nuclear magnetic resonance (NMR). Results were interpreted in light of computational density functional theory (DFT) calculations performed for Sodium Pravastatin in order to assign the data obtained for the LDH intercalated materials. XRD peaks of LDH-Pravastatin material and the one-dimensional (1D) electron density map pointed out to a bilayer arrangement of Pravastatin in the interlayer region, where its associated carboxylate and vicinal hydroxyl groups are close to the positive LDH. The structural organization observed for the stacked assembly containing the unsymmetrical and bulky monoanion Pravastatin and LDH seems to be promoted by a self-assembling process, in which local interactions are maximized and chloride ion cointercalation is required. It is observed a high similarity among vibrational and C-13 NMR spectra of Na-Pravastatin and LDH-Pravastatin materials. Those features indicate that the intercalation preserves the drug structural integrity. Spectroscopic techniques corroborate the nature of the guest species and their arrangement between the inorganic layers. Changes related to carboxylate, alcohol, and olefinic moieties are observed in both vibrational Raman and C-13 NMR spectra after the drug intercalation. Thus, Pravastatin ions are forced to be arranged as head to tail through intermolecular hydrogen bonding between adjacent organic species. The thermal decomposition profile of the hybrid samples is distinct of that one observed for Na-Pravastatin salt, however, with no visible increase in the thermal behavior when the organic anion is sequestrated within LDH gap.

A focused and commented review on the impact of dermatologic diseases and interventions in the solidary act of donating blood is presented to dermatologists to better advise their patients. This is a review of current Brazilian technical regulations on hemotherapeutic procedures as determined by Ministerial Directive #1353/2011 by the Ministry of Health and current internal regulations of the Hemotherapy Center of Ribeirão Preto, a regional reference center in hemotherapeutic procedures. Criteria for permanent inaptitude: autoimmune diseases (>1 organ involved), personal history of cancer other than basal cell carcinoma, severe atopic dermatitis or psoriasis, pemphigus foliaceus, porphyrias, filariasis, leprosy, extra pulmonary tuberculosis or paracoccidioidomycosis, and previous use of etretinate. Drugs that impose temporary ineligibility: other systemic retinoids, systemic corticosteroids, 5-alpha-reductase inhibitors, vaccines, methotrexate, beta-blockers, minoxidil, anti-epileptic, and anti-psychotic drugs. Other conditions that impose temporary ineligibility: occupational accident with biologic material, piercing, tattoo, sexually transmitted diseases, herpes, and bacterial infections, among others. Discussion: Thalidomide is currently missing in the teratogenic drugs list. Although finasteride was previously considered a drug that imposed permanent inaptitude, according to its short halflife current restriction of 1 month is still too long. Dermatologists should be able to advise their patients about proper timing to donate blood, and discuss the impact of drug withdrawal on treatment outcomes and to respect the designated washout periods.

SAPK/JNK regulieren nach genotoxischem Stress eine Vielzahl von Zielsubstraten, die bedeutsam für Reparatur und Überleben der Zelle sind, somit nehmen sie Einfluss auf das zelluläre Schicksal der Zelle. Ob DNA-Schäden eine Phosphorylierung von Stress-Kinasen nach sich ziehen ist bisher noch wenig untersucht. Mit reparaturdefizienten Zellen wurde der Einfluss von DNA-Schäden, durch Cisplatin/Transplatin/UV-C, auf die SAPK/JNK Aktivierung untersucht. Die Aktivierung der Stress-Kinasen erfolgte agenzspezifisch und abhängig von verschieden Reparaturfaktoren. Die Aktivierung korrelierte in reparaturdefizienten Zellen teilweise mit dem späten Auftreten von DNA-Strangbrüchen, war jedoch unabhängig von erhöhten initialen DNA-Schäden. Diese Befunde zeigten, dass die späte Aktivierung der SAPK/JNK DNA-schadensabhängig verläuft und das Cisplatin und Transplatin bei Verwendung von äquitoxischen Dosen zu einer vergleichbaren Aktivierung von SAPK/JNK führten. Die Hemmung der Rho-GTPasen sowohl durch Statine als auch mittels Clostridium difficile Toxin B zeigte weiterhin, dass Rho-GTPasen möglicherweise die späte DNA-schadensabhängige Aktivierung der Stress-Kinasen vermitteln. Die Hemmung von Rho-GTPasen durch physiologisch relevante Konzentrationen von Statinen führte in primären humanen Endothelzellen (HUVECs) zu einer Protektion vor IR-Strahlung und Doxorubicin. In beiden Fällen konnte eine Hemmung des pro-apoptotischen Transkriptionsfaktors p53 sowie der Chk1, welche einen Zellzyklusarrest reguliert, mit der Statin-Behandlung erreicht werden. Effektor-Caspasen wurden dabei durch den HMG-CoA-Reduktase Hemmer nicht beeinflusst. Ausschließlich bei dem Statin-vermittelten Schutz vor Doxorubicin kam es zu einer Reduktion von initialen DNA-Schäden, in Form von DNA-Strangbrüchen. Die IR-induzierten Strangbrüche in der DNA blieben von der Statin-Inkubation hingegen unbeeinflusst. Aufgrund ihrer protektiven Eigenschaften gegenüber IR- und Doxorubicin-induzierter Zytotoxizität in Endothelzellen und ihrer pro-apoptotischen Wirkung auf Tumorzellen könnten Statine möglicherweise die unerwünschten Nebenwirkungen von Zytostatika und einer Strahlentherapie günstig beeinflussen
