Processing efficiency theory predicts that anxiety reduces the processing capacity of working memory and has detrimental effects on performance. When tasks place little demand on working memory, the negative effects of anxiety can be avoided by increasing effort. Although performance efficiency decreases, there is no change in performance effectiveness. When tasks impose a heavy demand on working memory, however, anxiety leads to decrements in efficiency and effectiveness. These presumptions were tested using a modified table tennis task that placed low (LWM) and high (HWM) demands on working memory. Cognitive anxiety was manipulated through a competitive ranking structure and prize money. Participants' accuracy in hitting concentric circle targets in predetermined sequences was taken as a measure of performance effectiveness, while probe reaction time (PRT), perceived mental effort (RSME), visual search data, and arm kinematics were recorded as measures of efficiency. Anxiety had a negative effect on performance effectiveness in both LWM and HWM tasks. There was an increase in frequency of gaze and in PRT and RSME values in both tasks under high vs. low anxiety conditions, implying decrements in performance efficiency. However, participants spent more time tracking the ball in the HWM task and employed a shorter tau margin when anxious. Although anxiety impaired performance effectiveness and efficiency, decrements in efficiency were more pronounced in the HWM task than in the LWM task, providing support for processing efficiency theory.

Waste frying oil has been used to optimize the production of biodiesel. Biodiesel was prepared through sodium ethoxide catalyzed methanolysis from the transesterification of recycled waste frying oil. Optimization of the transesterification reaction for biodiesel production was carried out by means of statistical analyses using ANOVA. The optimum conditions for reaction were the following: a oil methanol mole ratio of 1:9, temperature of 50 degrees C, catalyst mass fraction of 0.9 %, and reaction time of 40 min, which enabled a yield of 98.7 % determined by gas chromatography/mass spectrometry (GC/MS) analysis. The density and viscosity of biodiesel/diesel blends have been determined as a function of composition at several temperatures.

The silent period is a misunderstood electrophysiological phenomenon leading to several different hypotheses explaining its electrogenesis. It has been studied by different authors and different methodologies giving a wide variability of results, therefore an exact pattern of its normal values does not exist. This work was undertaken to define the normal morphology and duration of the silent period obtained by supramaximal stimulus of the median nerve, during maximum isometric effort of the abductor pollicis brevis muscle against resistance, using 20 adult volunteers without neurological alterations. The normal median duration was 104.6 milliseconds. The same methodology was applied to 20 hands from 20 patients with carpal tunnel syndrome. The silent period showed many types of morphological alterations, but the major alteration observed was a tendency to temporal elongation. No correlation between the severity of the carpal tunnel syndrome and the silent period alterations were observed.

Silent period was evaluated in 20 adult male patients with chronic renal failure undergoing hemodialysis. Readings were obtained by supramaximal stimulus to the median nerve, during maximum isometric effort of the abductor pollicis brevis muscle against resistance. Two types of abnormalities were observed, motor neuron hypoexcitability with elongated silent period, and motor neuron hyperexcitability with reduction or absence of silent period. Some abnormalities are probably linked with dialysis duration, but show no correlation to presence or absence of peripheral neuropathy. The silent period alterations described in this study could possibly correlate with some other clinical feature frequently seen in patients with chronic renal failure such as hypereflexia of the deep tendon reflexes.

The neutral hydrolysis reaction of post-consumer poly(ethylene terephthalate) in solid state was studied through the reaction of the polymer with water at the molar ratio 1:91 with autogenous pressure. Two sizes of post-consumer PET flakes and temperatures of 135 °C, 170°C and 205°C with pressures of 4.0 atm, 7.5 atm and 13.5 atm, respectively, were considered. With reaction time equal to 6h, the method reached 99% depolymerization at 205°C, 8.2% at 170 °C and 1.7% at 135°C. The reaction extension was measured by separating the terephthalic acid formed in the process and calculating by gravimetry how much material could still be reacted. Through the viscosimetry of diluted, solutions and the counting of carboxylic end groups in the remaining material from the gravimetric assay, it was possible to suggest that the reaction occurs randomly and in the whole volume of the polymeric particle and not solely on the surface. The terephthalic acid obtained and then purified was characterized by elemental analysis, magnetic nuclear resonance, size and panicle size distribution and spectrophotometry in the visible spectrum, and it was similar to the petrochemical equivalent, with purity recorded in carbon base equal to 99.9%.

This study aimed to evaluate the motor response time and ability to develop joint torque at the knee and ankle in older women with and without a history of falls, in addition to investigating the effect of aging on these capacities. We assessed 18 young females, 21 older female fallers and 22 older female non-fallers. The peak torque, rate of torque development, rate of electromyography (EMG) rise, reaction time, premotor time and motor time were obtained through a dynamometric assessment and simultaneous electromyography. Surface EMGs of the rectus femoris (RF), vastus lateralis (VL), biceps femoris (BF), gastrocnemius lateralis (GL) and tibialis anterior (TA) muscles were recorded. Knee extension and flexion peak torques were lower in older fallers than in non-fallers. Knee extension and flexion and ankle plantarflexion and dorsiflexion peak torques were lower in both older groups than in the younger group. The rate of EMG rise of the BF and the motor time of the TA were lower and higher, respectively, in older fallers than in the younger adults. The time to reach peak torque in knee extension/flexion and ankle plantarflexion/dorsiflexion and the motor times of the RF, VL, BF and GL were higher in both older groups than in the younger groups. The motor time of the TA during ankle dorsiflexion and the knee extension peak torque were the major predictors of falls in older women, accounting for approximately 28% of the number of falls. Thus, these results further reveal the biomechanical parameters that affect the risk of falls and provide initial findings to support the prescription of exercises in fall prevention programs. © 2013 Elsevier Ltd.

It has been shown that mental rotation of objects and human body parts is processed differently in the human brain. But what about body parts belonging to other primates? Does our brain process this information like any other object or does it instead maximize the structural similarities with our homologous body parts? We tried to answer this question by measuring the manual reaction time (MRT) of human participants discriminating the handedness of drawings representing the hands of four anthropoid primates (orangutan, chimpanzee, gorilla, and human). Twenty-four right-handed volunteers (13 males and 11 females) were instructed to judge the handedness of a hand drawing in palm view by pressing a left/right key. The orientation of hand drawings varied from 0º (fingers upwards) to 90º lateral (fingers pointing away from the midline), 180º (fingers downwards) and 90º medial (finger towards the midline). The results showed an effect of rotation angle (F(3, 69) = 19.57, P < 0.001), but not of hand identity, on MRTs. Moreover, for all hand drawings, a medial rotation elicited shorter MRTs than a lateral rotation (960 and 1169 ms, respectively, P < 0.05). This result has been previously observed for drawings of the human hand and related to biomechanical constraints of movement performance. Our findings indicate that anthropoid hands are essentially equivalent stimuli for handedness recognition. Since the task involves mentally simulating the posture and rotation of the hands, we wondered if "mirror neurons" could be involved in establishing the motor equivalence between the stimuli and the participants' own hands.

The aim of this study is to develop a new enzymeless electroanalytical method for the indirect quantification of creatinine from urine sample. This method is based on the electrochemical monitoring of picrate anion reduction at a glassy carbon electrode in an alkaline medium before and after it has reacted with creatinine (Jaffe's reaction). By using the differential pulse voltammetry technique under the optimum experimental conditions (step potential, amplitude potential, reaction time, and temperature), a linear analytical curve was obtained for concentrations of creatinine ranging from 1 to 80 mu mol L-1, with a detection limit of 380 nmol L-1. This proposed method was used to measure creatinine in human urine without the interference of most common organic species normally present in biological fluids (e.g., uric acid, ascorbic acid, glucose, and phosphocreatinine). The results obtained using urine samples were highly similar to the results obtained using the reference spectrophotometric method (at a 95% confidence level). (C) 2012 Elsevier B.V. The influence of the temperature and reaction time on the sulfation process of a dolomite is investigated in this paper. The sulfation effectiveness was evaluated and correlated with changes in the physical characteristics of a Brazilian dolomite during the reactive process. Calcination and sulfation experiments were performed under isothermal conditions for dolomite samples with average particle sizes of 545 mu m at temperatures of 750 degrees C, 850 degrees C and 950 degrees C at different times of sulfation. Thermogravimetric tests were applied to establish the reactivity variation of the dolomite in function of the time in the sulfation reaction and evaluate the methodology of the samples preparation. Porosimetry tests were performed to study the pore blockage of dolomite during the sulfation reaction. The highest values of BET surface area were 25.55 m(2)/g, 29.55 m(2)/g and 12.62 m(2)/g for calcined samples and after their sulfation processes, conversions of 51.5%, 61.9% and 42.8% were obtained at 750 degrees C, 850 degrees C and 950 degrees C, respectively. Considering the process as a whole, the best fit was provided by a first-order exponential decay equation. Moreover, the results have shown that it is possible to quantify the decreasing in the dolomite reactivity for sulfur dioxide sorption and understand the changes in the behavior of the sulfation process of limestones when applied to technologies, as fluidized bed combustor, in which sulfur dioxide is present. This thesis was aimed at verifying the role of the superior colliculus (SC) in human spatial orienting. To do so, subjects performed two experimental tasks that have been shown to involve SC’s activation in animals, that is a multisensory integration task (Experiment 1 and 2) and a visual target selection task (Experiment 3). To investigate this topic in humans, we took advantage of neurophysiological finding revealing that retinal S-cones do not send projections to the collicular and magnocellular pathway. In the Experiment 1, subjects performed a simple reaction-time task in which they were required to respond as quickly as possible to any sensory stimulus (visual, auditory or bimodal audio-visual). The visual stimulus could be an S-cone stimulus (invisible to the collicular and magnocellular pathway) or a long wavelength stimulus (visible to the SC). Results showed that when using S-cone stimuli, RTs distribution was simply explained by probability summation, indicating that the redundant auditory and visual channels are independent. Conversely, with red long-wavelength stimuli, visible to the SC, the RTs distribution was related to nonlinear neural summation, which constitutes evidence of integration of different sensory information. We also demonstrate that when AV stimuli were presented at fixation, so that the spatial orienting component of the task was reduced, neural summation was possible regardless of stimulus color. Together, these findings provide support for a pivotal role of the SC in mediating multisensory spatial integration in humans, when behavior involves spatial orienting responses. Since previous studies have shown an anatomical asymmetry of fibres projecting to the SC from the hemiretinas, the Experiment 2 was aimed at investigating temporo-nasal asymmetry in multisensory integration. To do so, subjects performed monocularly the same task shown in the Experiment 1. When spatially coincident audio-visual stimuli were visible to the SC (i.e. red stimuli), the RTE depended on a neural coactivation mechanism, suggesting an integration of multisensory information. When using stimuli invisible to the SC (i.e. purple stimuli), the RTE depended only on a simple statistical facilitation effect, in which the two sensory stimuli were processed by independent channels. Finally, we demonstrate that the multisensory integration effect was stronger for stimuli presented to the temporal hemifield than to the nasal hemifield. Taken together, these findings suggested that multisensory stimulation can be differentially effective depending on specific stimulus parameters. The Experiment 3 was aimed at verifying the role of the SC in target selection by using a color-oddity search task, comprising stimuli either visible or invisible to the collicular and magnocellular pathways. Subjects were required to make a saccade toward a target that could be presented alone or with three distractors of another color (either S-cone or long-wavelength). When using S-cone distractors, invisible to the SC, localization errors were similar to those observed in the distractor-free condition. Conversely, with long-wavelength distractors, visible to the SC, saccadic localization error and variability were significantly greater than in either the distractor-free condition or the S-cone distractors condition. Our results clearly indicate that the SC plays a direct role in visual target selection in humans. Overall, our results indicate that the SC plays an important role in mediating spatial orienting responses both when required covert (Experiments 1 and 2) and overt orienting (Experiment 3).

Alcune patologie dell’occhio come la retinopatia diabetica, il pucker maculare, il distacco della retina possono essere curate con un intervento di vitrectomia. I rischi associati all’intervento potrebbero essere superati ricorrendo alla vitrectomia enzimatica con plasmina in associazione o in sostituzione della vitrectomia convenzionale. Inoltre, l’uso di plasmina autologa eviterebbe problemi di rigetto. La plasmina si ottiene attivando il plasminogeno con enzimi quali l’attivatore tissutale (tPA) e l’urochinasi ( uPA ) . La purificazione del plasminogeno dal sangue avviene normalmente attraverso cromatografia di affinità con resina. Tuttavia, le membrane di affinità costituiscono un supporto ideale per questa applicazione poiché possono essere facilmente impaccate prima dell’intervento, permettendo la realizzazione di un dispositivo monouso che fornisce un processo rapido ed economico. Obiettivo di questo lavoro è la preparazione di membrane di affinità per la purificazione del plasminogeno utilizzando L-lisina come ligando di affinità. Per questo scopo sono state usate membrane in cellulosa rigenerata ad attivazione epossidica, modificate con due diversi protocolli per l’immobilizzazione di L-lisina. La densità ligando è stata misurata mediante un saggio colorimetrico che usa l’acido arancio 7 come indicatore. La resa di immobilizzazione è stata studiata in funzione del tempo di reazione e della concentrazione di L-lisina. Le membrane ottimizzate sono state caratterizzate con esperimenti dinamici usando siero bovino e umano, i risultati sono stati confrontati con quelli ottenuti in esperimenti paralleli condotti con una resina commerciale di affinità con L-lisina. Durante gli esperimenti con siero, le frazioni provenienti da ogni fase cromatografica sono state raccolte e analizzate con HPLC ed elettroforesi SDS-PAGE. In particolare, l’elettroforesi dei campioni eluiti presenta una banda del plasminogeno ben definita indicando che le membrane di affinità con L-lisina sono adatte alla purificazione del plasminogeno. Inoltre, è emerso che le membrane hanno maggiore produttività della resina commerciale di riferimento.

Herz-Kreislauf-Erkrankungen zählen weltweit zu den Hauptursachen, die zu frühzeitigem Tod führen. Pathophysiologisch liegt eine Gefäßwandverdickung durch Ablagerung arteriosklerotischer Plaques (Arteriosklerose) vor. Die molekulare Bildgebung mit den nuklearmedizinischen Verfahren SPECT und PET zielt darauf ab, minderperfundierte Myokardareale zu visualisieren, um den Krankheitsverlauf durch frühzeitige Therapie abschwächen zu können. Routinemäßig eingesetzt werden die SPECT-Perfusionstracer [99mTc]Sestamibi und [99mTc]Tetrofosmin. Zum Goldstandard für die Quantifizierung der Myokardperfusion werden allerdings die PET-Tracer [13N]NH3 und [15O]H2O, da eine absolute Bestimmung des Blutflusses in mL/min/g sowohl in der Ruhe als auch bei Belastung möglich ist. 2007 wurde [18F]Flurpiridaz als neuer Myokardtracer vorgestellt, dessen Bindung an den MC I sowohl in Ratten, Hasen, Primaten als auch in ersten klinischen Humanstudien eine selektive Myokardaufnahme zeigte. Um eine Verfügbarkeit des Radionuklids über einen Radionuklidgenerator gewährleisten zu können, sollten makrozyklische 68Ga-Myokard-Perfusionstracer auf Pyridaben-Basis synthetisiert und evaluiert werden. Die neue Tracer-Klasse setzte sich aus dem makrozyklischen Chelator, einem Linker und dem Insektizid Pyridaben als Targeting-Vektor zusammen. Struktur-Affinitätsbeziehungen konnten auf Grund von Variation des Linkers (Länge und Polarität), der Komplexladung (neutral und einfach positiv geladen), des Chelators (DOTA, NODAGA, DO2A) sowie durch einen Multivalenzansatz (Monomer und Dimer) aufgestellt werden. Insgesamt wurden 16 neue Verbindungen synthetisiert. Ihre 68Ga-Markierung wurde hinsichtlich pH-Wert, Temperatur, Vorläufermenge und Reaktionszeit optimiert. Die DOTA/NODAGA-Pyridaben-Derivate ließen sich mit niedrigen Substanzmengen (6 - 25 nmol) in 0,1 M HEPES-Puffer (pH 3,4) bei 95°C innerhalb 15 min mit Ausbeuten > 95 % markieren. Für die DO2A-basierenden Verbindungen bedurfte es einer mikrowellengestützen Markierung (300 W, 1 min, 150°C), um vergleichbare Ausbeuten zu erzielen. Die in vitro-Stabilitätstests aller Verbindungen erfolgten in EtOH, NaCl und humanem Serum. Es konnten keine Instabilitäten innerhalb 80 min bei 37°C festgestellt werden. Unter Verwendung der „shake flask“-Methode wurden die Lipophilien (log D = -1,90 – 1,91) anhand des Verteilungs-quotienten in Octanol/PBS-Puffer ermittelt. Die kalten Referenzsubstanzen wurden mit GaCl3 hergestellt und zur Bestimmung der IC50-Werte (34,1 µM – 1 µM) in vitro auf ihre Affinität zum MC I getestet. In vivo-Evaluierungen erfolgten mit den zwei potentesten Verbindungen [68Ga]VN160.MZ und [68Ga]VN167.MZ durch µ-PET-Aufnahmen (n=3) in gesunden Ratten über 60 min. Um die Organverteilung ermitteln zu können, wurden ex vivo-Biodistributionsstudien (n=3) vorgenommen. Sowohl die µ-PET-Untersuchungen als auch die Biodistributionsstudien zeigten, dass es bei [68Ga]VN167.MZ zwar zu einer Herzaufnahme kam, die jedoch eher perfusionsabhängig ist. Eine Retention des Tracers im Myokard konnte in geringem Umfang festgestellt werden.

Der Free Fatty Acid Receptor 1 (FFAR1) ist ein G-Protein gekoppelter Rezeptor, welcher neben einer hohen Expression im Gehirn auch eine verstärkte Expressionsrate auf den β-Zellen des Pankreas aufweist. Diese Expressionsmuster machen ihn zu einem idealen Target für die Visualisierung der sogenannten β-Zell-Masse mittels molekularer bildgebender Verfahren wie der PET. Eine Entwicklung geeigneter Radiotracer für die β-Zell-Bildgebung würde sowohl für die Diagnostik als auch für die Therapie von Typ-1- und Typ-2-Diabetes ein wertvolles Hilfsmittel darstellen.rnAufbauend auf einem von Sasaki et al. publiziertem Agonisten mit einem vielversprechendem EC50-Wert von 5,7 nM wurden dieser Agonist und zwei weitere darauf basierende 19F-substituierte Moleküle als Referenzverbindungen synthetisiert (DZ 1-3). Für die 18F-Markierung der Moleküle DZ 2 und DZ 3 wurden die entsprechenden Markierungsvorläufer (MV 1-3) synthetisiert und anschließend die Reaktionsparameter hinsichtlich Temperatur, Lösungsmittel, Basensystem und Reaktionszeit für die nukleophile n.c.a. 18F-Fluorierung optimiert. Die abschließende Entschützung zum fertigen Radiotracer wurde mit NaOH-Lösung durchgeführt und die Tracer injektionsfertig in isotonischer NaCl-Lösung mit radiochemischen Ausbeuten von 26,9 % ([18F]DZ 2) und 39 % ([18F]DZ 3) erhalten.rnZusätzlich wurde ein Chelator zur 68Ga-Markierung an den Liganden gekoppelt (Verb. 46) und die Markierungsparameter optimiert. Nach erfolgter Markierung mit 95 % radiochemischer Ausbeute, wurde der Tracer abgetrennt und in vitro Stabilitätsstudien durchgeführt. Diese zeigten eine Stabilität von mehr als 90 % über 120 min in sowohl humanem Serum (37 °C) als auch isotonischer NaCl-Lösung.rnMit einem ebenfalls synthetisierten fluoreszenzmarkierten Derivat des Liganden (Verb. 43) wurden erste LSM-Bilder an sowohl Langerhansschen Inseln als auch FFAR1-tragenden RIN-M Zellen durchgeführt, welche einen vielversprechenden Uptake des neuen Liganden in die Zellen zeigen. Weitere Untersuchungen und biologische Evaluierungen stehen noch aus. Mit den Referenzsubstanzen wurden zusätzlich Vitalitätsstudien an Langerhansschen Inseln durchgeführt, um einen negativen toxischen Einfluss auszuschließen.rn
