Here we describe the total syntheses and characterization by elemental analyses, infrared and NMR spectroscopy of three new compounds analogous to avenaciolide, a bis-γ-lactone isolated from Aspergillus avenaceus that possesses antifungal activity, where the octyl group of the natural product was replaced by aromatic groups containing chlorine and fluorine atoms. The effects of the avenaciolide, the novel compounds and their synthetic precursors on mycelia development and conidia germination of Colletotrichum gloeosporioides and Fusarium solani were evaluated in vitro. The title compounds were almost as active as avenaciolide. The absolute structures of the chlorinated analogs were determined by X-ray diffraction analysis.

The species Eremanthus mattogrossensis, known as "veludo do cerrado" (cerrado velvet), is native to the Brazilian Cerrado. Because the amount of metabolites present in plants may be influenced by biological and environmental factors, here we conducted an HPLC-DAD-MS/MS investigation of the metabolite concentrations found in the MeOH/H2O extract of the leaves of this species. The main compounds were identified and quantified, and the metabolites were grouped by chemical class (caffeoylquinic acids, flavonoids, and sesquiterpene lactone). Statistical analysis indicated a straight correlation between the quantity of metabolites and seasonality, suggesting that environmental properties elicit important metabolic responses.

The chemical investigation of the MeOH extract from the bryozoan B. dentata MeOH yielded tambjamines A (1), C (3), D (4), K (6), aldehyde 8 and the new tambjamine J1(9), while the extract of its predator, the nudibranch Tambja stegosauriformis, yielded tambjamines C and K, along with aldehyde 8. Furodisinin lactone (11) was isolated from the nudibranch Hypselodoris lajensis, a compound previously isolated from Dysidea sponges. The alkaloid 2,5,6-tribromo-N-methylgramine (12) was isolated from the nudibranch Okenia zoobotryon and from its prey, the bryozoan Zoobotryon verticillatum, the only source of 12 previously known.

The chemical investigation of hexane and ethanol extracts from the aerial parts of Vernonia scorpioides resulted in the isolation and characterization of a new polyacetylene lactone, rel-4-dihydro-4β-hydroxy-5a-octa-2,4,6-triynyl-furan-2-(5H)-one, along with the new ethyl 3,4-dihydroxy-6,8,10-triynyl-dodecanoate, and seven known compounds: taraxasteryl acetate, lupeyl acetate, lupeol, lupenone, β-sitosterol, stigmasterol and luteolin. The structure of all compounds was determined by spectrometric techniques (HR-ESI-MS, ¹H and 13C NMR and IV) and comparison with published spectral data.

Cyclolignan (+)-lyoniresinol (1), veratric acid (2), vanillic acid (3), lupeol, oleanolic acid, 3β-hydroxy-urs-11-en-28,13β-lactone (4), the mixture of α- and β-amyrin, trans-polyisoprene, and β-sitosterol were isolated from the leaves of Maytenus phyllanthoides. The structures of the isolated compounds were established based on spectroscopic data, mainly ¹H and 13C nuclear magnetic resonance (NMR). Compound 1, its acetate analog 1a, and compounds 2, 3, and 4 were tested against Trichomonas vaginalis. (+)-Lyoniresinol showed activity corresponding to IC50 17.57 µM. This is the first report on the occurrence of 3β-hydroxy-urs-11-en-28,13β-lactone (4) in the Celastraceous family and lyoniresinol in the Maytenus genus, and on the antitrichomonal activity of lyoniresinol.

Phytochemical investigation of Eugenia copacabanensis allowed for the isolation and identification of following compounds: β-sitosterol, β-sitosterol-glucoside, eight triterpenes, (mixture of α- and β-amyrins, ursolic acid, 30-hydroxy-ursolic acid, betulin, friedelin, friedelan-3,4-lactone, and taraxerol), a mixture of three sesquiterpenes, (clovandiol, globulol, and viridiflorol), three flavonoids (kaempferol-3-O-β-D-rhamnoside, quercetin-3-O-α-L-arabinoside, and quercetin), and a mixture of four coumaroyl esters (octacosanyl, heptacosanyl, hexacosanyl, and tetracosanyl coumarates). The structures of these compounds were assigned based on comparison with literature data and spectroscopic analysis, including analysis by two-dimensional NMR techniques. Total phenolic content and total flavonoids were evaluated. Antioxidant activities of methanol extracts and fractions were measured by the 1,2-diphenyl-2-picryl-hidrazyl free radical scavenging assay.

Hawthorn (Crataegus sp.) is widely distributed in the northern hemisphere (Asia, Europe and North America). It has been used as a medicinal material and food for hundreds of years both in Europe and in China. Clinical investigations and other research suggest that extracts of hawthorn fruits and leaves have multiple health effects including hypolipidaemic, anti-atherosclerotic, hypotensive, cardioprotective and blood vessel relaxing activities. Hawthorn fruit extracts have also displayed antioxidant and radical scavenging activities. Emblic leafflower fruit (Phyllanthus emblica) is widely used in Chinese and Indian traditional medicine. It has been found to have anti-cancer, hypoglycaemic and hypolipidaemic activities as well as cardioprotective effects and antioxidant activity. The fruit is currently used as a functional food targeted at obese people in China. Phenolic compounds, procyanidins (PCs), flavonols and C-glycosyl flavones in hawthorn and hydrolysable tannins in emblic leafflower fruits are considered among the major bioactive compounds in these berries. Moreover, hawthorn and emblic leafflower fruits are rich in vitamin C, triterpenoids, fruit acids, sugar alcohols and some other components with beneficial effects on the health of human beings. The aim of the thesis work was to characterise the major phenolic compounds in hawthorn fruits and leaves and emblic leafflower fruits as well as other components contributing to the nutritional profile and sensory properties of hawthorn fruits. Differences in the content and compositional profile of the major phenolic compounds, sugars, acids and sugar alcohols within various origins and species of hawthorn were also investigated. Acids, sugars and sugar alcohols in the fruits of different origins/cultivars belonging to three species (C. pinnatifida, C. brettschneideri and C. scabrifolia) of hawthorn were analysed by gas chromatography (GC-FID) and mass spectrometry (Publication I). Citric acid, quinic acid, malic acid, fructose, glucose, sorbitol and myo-inositol were found in all the subspecies. Sucrose was present only in C. scabrifolia and three cultivars of C. pinnatifida var. major. Forty-two phenolic compounds were identified/tentatively identified in fruits of C. pinnatifida var. major by polyamide column chromatography combined with high-performance liquid chromatograph-electrospray ionisation mass spectrometry (HPLC-ESI-MS) (Publication II). Ideain, chlorogenic acid, procyanidin (PC) B2, (-)-epicatechin, hyperoside and isoquercitrin were the major phenolic components identified. In addition, 35 phenolic compounds were tentatively identified based on UV and mass spectra. Eleven major phenolic compounds (hyperoside, isoquercitrin, chlorogenic acid, ideain, (-)-epicatechin, two PC dimers, three PC trimers and a PC dimer-hexoside) were quantified in the fruits of 22 cultivars/origins of three species of Chinese hawthorn by HPLC-ESI-MS with single ion recording function (SIR) (Publication III). The fruits of the hawthorn cultivars/origins investigated fell into two groups, one rich in sugars and flavonols, the other rich in acids and procyanidins. Based on the compositional features, different biological activities and sensory properties may be expected between cultivars/origins of the two groups. The results suggest that the contents of phenolic compounds, acids, sugars and sugar alcohols may be used as chemotaxonomic information distinguishing the hawthorn species from each other. Phenolic compounds in fruits and leaves of C. grayana and their changes during fruit ripening/harvesting were investigated using HPLC-UV-ESI-MS (Publication IV). (-)-Epicatechin, PC B2 and C1, hyperoside and a quercetin-pentoside were the major phenolic compounds in both fruits and leaves. Three C-glycosyl flavones (a luteolin-C-hexoside, a methyl luteolin-C-hexoside and an apigenin-C-hexoside) were present in leaves in abundance, but only at trace levels in fruits. Ideain and 5-O-caffeoylquinic acid were found in fruits only. Additionally, eleven phenolic compounds were identified/tentatively identified in both leaves and fruits (three B-type PC trimers, two B-type PC tetramers, a quercetin-rhamnosylhexoside, a quercetin-pentoside, a methoxykaempferol-methylpentosylhexoside, a quercetin-hexoside acetate, a methoxykaempferol-pentoside, chlorogenic acid and an unknown hydroxycinnamic acid derivative). The total content of phenolic compounds reached the highest level by the end of August in fruits and by the end of September in leaves. The compositional profiles of phenolic compounds in fruits and leaves of C. grayana were different from those of C. pinnatifida, C. brettschneideri, C. scabrifolia, C. pinnatifida. var. major, C. monogyna, C. laevigata and C. pentagyna. Phenolic compounds in emblic leafflower fruits were characterised by Sephadex LH-20 column chromatography combined with HPLC-ESI-MS (Publication V). A mucic acid gallate, three isomers of mucic acid lactone gallate, a galloylglucose, gallic acid, a digalloylglucose, putranjivain A, a galloyl-HHDP-glucose, elaeocarpusin and chebulagic acid represented the major phenolic compounds in fruits of emblic leafflower. In conclusion, results of this study significantly increase the current knowledge on the key bioactive and nutritional components of hawthorn and emblic leafflower fruits. These results provide important information for research on the mechanism responsible for the health benefits of these fruits.

The oil obtained from Brazilian roasted coffee by supercritical CO2 extraction shows considerable aromatic properties, mainly composed by five aromatic compounds, 2-methylpyrazine; 2-furfurylalcohol, 2,5-dimethylpyrazine; γ-butyrolactone and 2-furfurylacetate. Sensory analyses were used to verify the influence of a mixture of these important classes of aromatic coffee compounds (pyrazines, furans and lactones) and of the roasted coffee aromatic oil on the coffee aroma and flavour of black instant freeze and spray-dried coffee beverages. In the acceptance evaluation of the aroma, the samples prepared with freeze-dried instant coffee without the mixture of volatile compounds (sample 4) were not significantly different from the freeze-dried instant coffee in which the aromatic coffee oil was added (sample 5) and from the sample prepared with freeze-dried coffee in which the mixture of the five volatile was added (sample 3), coincidentally from the same drying process. Therefore, sample (3) did not differ from samples prepared with spray dried instant coffee without (sample 1) and to which (sample 2) the mixture of volatile was added. Therefore, with respect to this attribute, the addition of this mixture did not interfere in this drink acceptance. Taking into consideration the flavor, samples prepared with freeze-dried instant coffee in which the aromatic coffee oil was added (5) and the samples with (3) and without (4) the mixture of the five volatile was added did not differ significantly, however sample (4) did not differ from samples (1) and (2). Regarding this attribute, the addition of the aromatic oil of roasted coffee or a mixture of volatile in samples of freeze-dried instant coffee had a better acceptance than those dried by spray dryer (1) and (2). Thus, the enrichment of drinks with the aromatic oil of roasted coffee, or even with the mixture of the five components did not influence the consumer acceptance with respect to the aroma, but exerts influence with respect to flavour.

The deoxy derivative of pancratistatin 1.10 was prepared in good yield through the use of a [4+2] Diels-Alder cycloaddition and Bischler-Napieralski cyclization approach. The Bischler-Napieralski cyclization was shown to yield two additional side products 2.9, 2.10, however, under slightly modified hydrolysis conditions, the tetracyclic product 2.11 was obtained exclusively in greater than 84% yield. Initial screening of the di-hydroxylatgd derivative, and the other complementary pair analogue 1.10' previously prepared in our laboratories gave interesting results. Both of these compounds were shown to exhibit cytostatic activity; the mono-alcohol was marginally active while the di-hydroxylated analogue proved to be more potent although one to two magnitudes less potent than pancratistatin itself Human tumour cell line assay results indicated that the di-hydroxylated derivative exhibited selective cytotoxic inhibition in the following cell lines: non-small cell lung cancer line NCI-H226 (ED50 - 0.65 ^g/mL), leukemia cell lines CCRF-CEM (ED30 = 0.55 Hg/mL) and HL-60(TB) (ED50 = 0.89^ig/mL). Our results demonstrated that the pharmacophore is not a mono-alcohol, and that the minimum pharmacophore contains the hydroxyl group at the C4 position in addition to either, or both, of the hydroxyl groups present at C2 and C3.' The minimum pharmacophore has been narrowed to only three possibilities which are current synthetic targets in several research groups. The controlled Grignard addition to the tartaric acid derived bis-Weinreb amide 1.25 afforded a direct entry to a host of 1,4-diflferentiated tartaric acid derived intermediates (2.12-2.18). This potentially usefiil methodology was demonstrated through the efficient synthesis of the naturally occurring lactone 2.23, which bears the inherent syn-dio\ subunit. Based on this result, a similar approach to the synthesis of syn-dio\ bearing natural products looks very promising? A direct 2,3-diol desymmetrization method using TIPS-triflate was shown to be effective on the selective differentiation of Z,-methyl tartrate (and diisopropyl tartrate). The mono-silyl-protected intermediates 2.31 also proved to be useful when they were selectively differentiated at the 1,4-carboxyl position (2.35, 2.36) through the use of a borohydride reducing agent. Furthermore, the mono-silyl-protected derivative underwent periodate cleavage affording two synthetically useful a,P-unsaturated esters 2.43, 2.44, with one of esters being obtained via a silyl-migration method.''

Plusieurs nouveaux complexes pinceurs de cobalt et de nickel ont été préparés avec le ligand pinceur de type POCOP : 2,6-(i-Pr2PO)2C6H4. Dans le cas du cobalt, une nouvelle voie de synthèse a été développée. Contrairement au cas du nickel, il s’agit ici de cobalt au degré d’oxydation +III. Les composés obtenus sont paramagnétiques. En outre, le dérivé bromé est instable à la lumière et se décompose en perdant un brome pour former le complexe pinceur de Co(II). La réactivité de ces complexes a été étudiée. Pour ce qui est du nickel, la catalyse de l’hydroamination a été élargie aux dérivés de l’acrylonitrile et aux amines aromatiques. En outre, la réaction d’hydroaryloxylation a été étudiée dans les mêmes conditions. Enfin, avec le 4-cyanostyrène et le cinnamonitrile, la formation d’amidines a été observée. Un complexe pinceur portant cette amidine a été isolé. Enfin, le cation réagit avec des anions fortement coordonnants tels le cyanure ou l’isocyanate. En outre, l’anion triflate peut être déplacé par l’eau, l’acrylonitrile et ses dérivés. Enfin, une réactivité particulière a été observée avec la morpholine, l’acétone et un mélange 1:1 aniline/triéthylamine.

La synthèse énantiosélective de la (+)-ambruticine S, un produit naturel antifongique a été effectuée au sein de notre groupe. Trois approches ont été développées pour la synthèse du fragment lactone (cycle A). Ces trois voies d’accès au cycle A ont pour intermédiaire commun le methyl α-D-glycopyranoside déjà porteur du diol requis et disponible commercialement à bon prix. Une désoxygénation de l’hydroxyle en C-4 et l’homologation d’un carbone de la chaine latérale en C-6 ont permis l’obtention du cycle lactonique A. Le deuxième projet est une collaboration entre le groupe Hanessian et ISIS Pharmaceuticals afin de développer de nouveaux oligonucléosides antisens. Les nucléosides antisens [4.3.0]-bicycliques cis et trans ont été synthétisés avec succès à partir d’un monosaccharide naturel commun, L-arabinose, porteur des stéréocentres requis. Un réaction clé d’allylation de Sakurai a permis d’obtenir les diastéréoisomères cis et trans dans des conditions de contrôle de type Felkin-Ahn et de contrôle par chélation respectivement. Les composés bicycliques finaux cibles ont été obtenus par une réaction d’aldol intramoléculaire catalyzéé par la proline, par métathèse de fermeture de cycle et par l’application de la méthode de Vorbrüggen pour la synthèse de nucléosides.

Une nouvelle approche pour la synthèse de l’aliskiren, un puissant inhibiteur de la rénine pour le traitement de l’hypertension chez l’homme, a été développée. De cette approche, des étapes clés tels qu’une allylation avec un des catalyseurs de MacMillan, une cyclisation par métathèse (RCM) pour la préparation d’une lactone à neuf membres et une séquence de réactions d’aziridination diastéréosélective/réarrangement de cycle par une catalyse acide donnant un produit aminohydroxylé ont permis de compléter la synthèse de l’aliskiren en 11 étapes. Durant l’élaboration de la séquence de réactions pour la préparation de l’aliksiren, il a été noté que la lactone à neuf membres avait une facilité à se réarranger via des intermédiaires quinonoïdaux et les produits issus de cette tendance ont été analysés. De plus, une étude sur la réaction de RCM, donnant la lactone à neuf membres, a montré une dépendance envers la nature diastéréomérique du substrat de départ. Une méthodologie d’alpha-allylation asymétrique de cétones catalysée au palladium, tirant avantage d’un ligand chiral PHOX, a été explorée et utilisée en vue de la synthèse de l’aliskiren. De cette méthode, une étude pour la synthèse de produits alpha-allylcétones acycliques a été démontrée et un effet d’additif sur la sélectivité et la vitesse de réaction a été découvert. De plus, la production d’un intermédiaire avancé d’un produit d’intérêts a été accomplie et la nature de la contribution de l’additif a été investiguée. Les produits obtenus depuis la méthodologie d’allylation catalysée au palladium et la formation d’intermédiaire cationique des certains dérivés de la synthèse de l’aliskiren ont inspirés une nouvelle approche faisant appel à des techniques d’alkylation en catalyse acide pour la formation de produits diaryliques. Il a été trouvé que le catalyseur de chlorure d’or(III) et de triflate de bismuth(III) étaient particulièrement efficaces, démontrant une différence de cinétique pour la réactivité d’un mélange diastéréoisomériques d’alcools alpha-substitués de départ.

Ce mémoire présente une poursuite de l’étude vers la synthèse de l’hodgsonox, un sesquiterpénoïde naturel possédant des propriétés insecticides contre la mouche verte d’Australie, Lucilia cuprina. L’hodgsonox comporte six centres stéréogènes et trois cycles : un époxyde fusionné à un cycle à cinq chaînons et une fonction éther cyclique à six chaînons doublement allylique. La stratégie de synthèse de l’hodgsonox proposée comporte dix-neuf étapes linéaires. Elle s’appuie sur les travaux préliminaires de Lise Bréthous, étudiante au doctorat, de Nicolas Lévaray, étudiant à la maîtrise, ainsi que du Dr. Ying Dong Lu et de la Dr. Sonia Diab, qui ont tous travaillé précédemment dans le groupe de la Pr. Lebel. La première étape de cette synthèse consiste en une hydrogénation cinétique dynamique de Noyori permettant d’obtenir un seul diastéréoisomère à partir de l’α-acétylbutyrolactone. Une séquence de six étapes linéaires supplémentaires, comprenant l’ouverture de la lactone ainsi qu’une métathèse d’oléfine, permet d’obtenir le cycle à cinq chaînons avec un rendement global de 37%. L’unité isopropyle est par la suite installée par une addition conjuguée pour former un éther d’énol silylé, qui est directement oxydé en la cétone correspondante avec l’acétate de palladium(II). Une réaction d’hydrosilylation subséquente permet d’obtenir la stéréochimie syn attendue de l’unité isopropyle. Par la suite, la carbonylation d’un intermédiaire triflate permet d’obtenir le squelette de base pour la formation de l’éther cyclique. Enfin, le cycle à six chaînons est formé par insertion O−H intramoléculaire d’un diazo avec un rendement global de 2% sur 17 étapes. Les travaux spécifiques de l’auteure comprennent l’évaluation de conditions catalytiques pour l’oxydation de Saegusa de l’éther d’énol silylé. Les trois dernières étapes ont également été explorées par l’auteure. Il s’agit de l’époxydation de la double liaison endocyclique, de l’insertion dans un lien O−H catalysée par un dimère de rhodium, et de la méthylénation. Enfin, l’exploration d’une voie alternative a été entamée. Cette nouvelle voie consiste à former l’éther cyclique par une substitution nucléophile sur un époxyde. La double liaison exo-cyclique serait installée par une simple réaction de déshydratation.

Cette thèse décrit deux thèmes principaux: 1) la conception, la synthèse, et l'évaluation biophysique des nucléosides tricycliques, et 2) la synthèse de nagilactone B, un produit naturel norditerpenoïde dilactone de la famille de produits naturels “podolactone”. Le premier chapitre décrit la stratégie de design rationnel des nucléosides nommé “restriction conformationnelle double” basée sur les études de modélisation structurales des duplex ADN–ARN modifiés. Cette stratégie implique un blocage du cycle furanose dans une configuration de type N- ou S, et une restriction de la rotation torsionelle autour de l’angle γ. La première contrainte a été incorporée avec un pont méthylène entre l’oxygène en position 2′ et le carbone 4′ du nucléoside. Cette stratégie a été inspirée par les acides nucléiques bloqués (ou “locked nucleic acid”, LNA). La deuxième contrainte a été réalisée en ajoutant un carbocycle supplémentaire dans l'échafaud de l’acide nucléique bloqué. Les défis synthétiques de la formation des nucléotides modifiés à partir des carbohydrates sont décrits ainsi que les améliorations aux stabilités thermiques qu’ils apportent aux duplex oligonucléïques dont ils font partie. Chapitres deux et trois décrivent le développement de deux voies synthétiques complémentaires pour la formation du noyau de nagilactone B. Ce produit naturel a des implications pour le syndrome de Hutchinson–Gilford, à cause de son habilité de jouer le rôle de modulateur de l’épissage d’ARN pré-messager de lamine A. Ce produit naturel contient sept stereocentres différents, dont deux quaternaires et deux comprenant un syn-1,2-diol, ainsi que des lactones à cinq ou six membres, où le cycle à six ressemble à un groupement α-pyrone. La synthèse a débuté avec la cétone de Wieland-Miescher qui a permis d’adresser les défis structurels ainsi qu’explorer les fonctionnalisations des cycles A, B et D du noyau de nagilactone B.

The thesis entitled novel heterocyclic constructions mediated by nucleophilic carbenes and related chemistry, embodies the results of the investigations carried out to explore the reactivity patterns of the 1:1 zwitterions, generated in situ from various nucleophilic carbenes and DiMethyl AcetyleneDicarboxylate(DMAD) towards aldehydes and ketones. The traditional synthesis of complex organic molecules employs stepwise formation of bonds and involves multiple steps. Besides the sequential synthesis, in several instances, the desired product can also be obtained in one pot reactions of three or more starting compounds. Such reactions in which more than two starting materials react to form a product in such a way that the majority of the atoms of the starting materials can be found in the products are called multicomponent reactions(MCRs). The results of our investigations on the application of N-heterocyclic carbenes in multicomponent reaction with DMAD and aromatic aldehydes leading to the one pot synthesis of 2-oxy-maleate and furanone derivatives. It is interesting to note that dihydrofuran and lactone motifs are present in a number of biologically active natural products and other heterocyclic compounds. It is conceivable that the novel multicomponent reactions described herein will find application in the synthesis of a variety of heterocyclic compounds, and in natural product synthesis.
