Des preuves archéopalynologiques directes appuient maintenant l’hypothèse d’une lente adaptation horticole durant la néolithisation amérindienne de la région de Montréal. Les sites archéologiques Hector-Trudel (BhFl-1a) de Pointe-du-Buisson et Séminaire de Saint-Sulpice (BjFj-18) dans le Vieux-Montréal ont été retenus pour élaborer une méthodologie archéopalynologique d’étude des sols archéologiques. Cela a permis de caractériser l’impact de la présence humaine sur l'environnement végétal des sites et d’identifier des indices de culture et de gestion de plantes allogènes et indigènes. Un complexe horticole de production à petite échelle de maïs (Zea mays), de tournesol (Helianthus annuus) et de petit tabac (Nicotiana rustica) et une forme de gestion des arbustes à petits fruits sont identifiés au site Hector-Trudel durant le Sylvicole moyen tardif (500 à 1000 A.D.). Ces cultigènes sont aussi identifiés au site du Séminaire pour la fin du Sylvicole supérieur ancien (1200 à 1300 A.D.), dans des proportions toutefois plus importantes, et une activité de gestion forestière au profit des arbres à noix et du tilleul d’Amérique (Tilia americana), reflet des pratiques d’entretien des champs cultivés, témoignent d’une évolution dans les comportements

Many nonlinear optical microscopy techniques based on the high-intensity nonlinear phenomena were developed recent years. A new technique based on the minimal-invasive in-situ analysis of the specific bound elements in biological samples is described in the present work. The imaging-mode Laser-Induced Breakdown Spectroscopy (LIBS) is proposed as a combination of LIBS, femtosecond laser material processing and microscopy. The Calcium distribution in the peripheral cell wall of the sunflower seedling (Helianthus Annuus L.) stem is studied as a first application of the imaging-mode LIBS. At first, several nonlinear optical microscopy techniques are overviewed. The spatial resolution of the imaging-mode LIBS microscope is discussed basing on the Point-Spread Function (PSF) concept. The primary processes of the Laser-Induced Breakdown (LIB) are overviewed. We consider ionization, breakdown, plasma formation and ablation processes. Water with defined Calcium salt concentration is used as a model of the biological object in the preliminary experiments. The transient LIB spectra are measured and analysed for both nanosecond and femtosecond laser excitation. The experiment on the local Calcium concentration measurements in the peripheral cell wall of the sunflower seedling stem employing nanosecond LIBS shows, that nanosecond laser is not a suitable excitation source for the biological applications. In case of the nanosecond laser the ablation craters have random shape and depth over 20 µm. The analysis of the femtosecond laser ablation craters shows the reproducible circle form. At 3.5 µJ laser pulse energy the diameter of the crater is 4 µm and depth 140 nm for single laser pulse, which results in 1 femtoliter analytical volume. The experimental result of the 2 dimensional and surface sectioning of the bound Calcium concentrations is presented in the work.

Im ersten Teil dieser Dissertation stand die Analyse der Motilitätsentwicklung bei Vertretern der Gattung Methylobacterium im Vordergrund. Diese zu den pink pigmentierten fakultativ methylotrophen Mikroorganismen (PPFMs) gehörenden Prokaryoten sind in der Umwelt weit verbreitet. Besonders häufig besiedeln die Mikroben pflanzliche Oberflächen und können als so genannte Phytosymbionten in einer wechselseitigen Beziehung zu pflanzlichen Organismen stehen. In aquatischer Umgebung können Methylobakterien Flagellen aufweisen. Hierbei handelt es sich um spezielle Fortbewegungsorganellen, die den Mikroben eine aktive Beweglichkeit ermöglichen. Die Ausbildung polarer Einzelflagellen bei Methylobacterium-Zellen in planktonischer Lebensweise konnte unter Anwendung verschiedener mikroskopischer Techniken dokumentiert werden. Quantitative Beweglichkeitsstudien zeigten einen charakteristischen Entwicklungsverlauf, korreliert mit den Wachstumsphasen der Bakterienkulturen und machten deutlich, dass die Motilitätsrate durch Umweltfaktoren, wie z. B. die Nährstoffversorgung, beeinflusst werden kann. Es konnte gezeigt werden, dass die Pflanzen-assoziierten PPFMs in der Lage sind, zwischen einer sessilen und planktonischen Lebensweise zu wechseln und dass sowohl die zelluläre Beweglichkeit als auch die Biofilm-Bildung der Prokaryoten ein reversibles, reaktivierbares Verhalten darstellt. Weiterhin konnte belegt werden, dass die Motilität der epiphytischen Mikroben bezüglich der Besiedelung von Pflanzen, z. B. bei der Ausbreitung auf Keimblatt-Oberflächen von Sonnenblumen (Helianthus annuus), keine zentrale Rolle spielt und eine endophytische Lebensweise unwahrscheinlich ist. Ziel der Arbeit war weiterhin die Charakterisierung und Identifizierung eines aus der Phyllosphäre der Echten Feige (Ficus carica, Standort Griechenland) isolierten Bakterien-Stammes (Mtb. sp. Fc1). Die fakultativ methylotrophe Stoffwechseleigenschaft, sowie die auffällige rötliche Pigmentierung belegen, dass es sich um einen Vertreter der PPFMs handelt. Die Analyse morphologischer, physiologischer und biochemischer Eigenschaften bestätigte in Übereinstimmung mit molekularphylogenetischen Untersuchungen zur Klassifizierung und taxonomischen Einordnung, dass es sich um Pflanzen-assoziierte Mikroben der Gattung Methylobacterium handelt. Analysen der 16S rDNA sowie partieller Sequenzen der für Methylobakterien etablierten Marker-Gene mxaF und gyrB verdeutlichten die phylogenetische Stellung und die evolutionären Beziehungen des Ficus-Isolates. Obwohl enge Verwandtschaftsverhältnisse zu anderen Methylobacterium-Arten ermittelt werden konnten, war eine Identifizierung als valide beschriebene Spezies nicht möglich. Die Resultate legen den Schluss nahe, dass es sich um eine neue, unbeschriebene Spezies der epiphytisch lebenden Methylobakterien handelt.

Phytoextraction, the use of plants to extract heavy metals from contaminated soils, could be an interesting alternative to conventional remediation technologies. However, calcareous soils with relatively high total metal contents are difficult to phytoremediate due to low soluble metal concentrations. Soil amendments such as ethylene diaminetetraacetate (EDTA) have been suggested to increase heavy metal bioavailability and uptake in aboveground plant parts. Strong persistence of EDTA and risks of leaching of potentially toxic metals and essential nutrients have led to research on easily biodegradable soilamendments such as citric acid. In our research, EDTA is regarded as a scientific benchmark with which degradable alternatives are compared for enhanced phytoextraction purposes. The effects of increasing doses of EDTA (0.1, 1, 10 mmol kg(-1) dry soil) and citric acid (0.01, 0.05,0.25,0.442, 0.5 mol kg(-1) dry soil) on bioavailable fractions of Cu, Zn, Cd, and Pb were assessed in one part of our study and results are presented in this article. The evolution of labile soil fractions of heavy metals over time was evaluated using water paste saturation extraction (similar to soluble fraction), extraction with 1 M NH4OAc at pH 7 (similar to exchangeable fraction), and extraction with 0.5 M NH4OAc + 0.5 M HOAc + 0.02 M EDTA atpH 4.65 (similar to potentially bioavailable fraction). Both citric acid and EDTA produced a rapid initial increase in labile heavy metal fractions. Metal mobilization remained constant in time for soils treated with EDTA, but metal fractions was noted for soils treated with citric acid. The half life of heavy metal mobilization by citric acid varied between 1.5 and 5.7 d. In the following article, the effect of heavy metal mobilization on uptake by Helianthus annutis will be presented.

Phytoextraction has been proposed as an alternative remediation technology for soils polluted with heavy metals or radionuclides, but is generally conceived as too slow working. Enhancing the accumulation of trace pollutants in harvestable plant tissues is a prerequisite for the technology to be practically applicable. The chelating aminopolycarboxylic acid, ethylene diamine tetraacetate (EDTA), has been found to enhance shoot accumulation of heavy metals. However, the use of EDTA in phytoextraction may not be suitable due to its high environmental persistence, which may lead to groundwater contamination. This paper aims to assess whether ethylene diamine disuccinate (EDDS), a biodegradable chelator, can be used for enhanced phytoextraction purposes. A laboratory experiment was conducted to examine mobilisation of Cd, Cu, Cr, Ni, Pb and Zn into the soil solution upon application of EDTA or EDDS. The longevity of the induced mobilisation was monitored for a period of 40 days after application. Estimated effect half lives ranged between 3.8 and 7.5 days for EDDS, depending on the applied dose. The minimum observed effect half life of EDTA was 36 days, while for the highest applied dose no decrease was observed throughout the 40 day period of the mobilisation experiment. Performance of EDTA and EDDS for phytoextraction was evaluated by application to Helianthus annuus. Two other potential chelators, known for their biodegradability in comparison to EDTA, were tested in the plant experiment: nitrilo acetic acid (NTA) and citric acid. Uptake of heavy metals was higher in EDDS-treated pots than in EDTA-treated pots. The effects were still considered insufficiently high to consider efficient remediation. This may be partly due to the choice of timing for application of the soil amendment. Fixing the time of application at an earlier point before harvest may yield better results. The sexual system of the symbiotic shrimp Thor amboinensis is described, along with observations on sex ratio and host-use pattern of different populations. We used a comprehensive approach to elucidate the previously unknown sexual system of this shrimp. Dissections, scanning electron microscopy, size-frequency distribution analysis, and laboratory observations demonstrated that T amboinensis is a protandric hermaphrodite: shrimp first mature as males and change into females later in life. Thor amboinensis inhabited the large and structurally heterogeneous sea anemone Stichoclactyla helianthus in large groups (up to 11 individuals) more frequently than expected by chance alone. Groups exhibited no particularly complex social structure and showed male-biased sex ratios more frequently than expected by chance alone. The adult sex ratio was male-biased in the four separate populations studied, one of them being thousands of kilometers apart from the others. This study supports predictions central to theories of resource monopolization and sex allocation. Dissections demonstrated that unusually large males were parasitized by an undescribed species of isopod (family Entoniscidae). Infestation rates were similarly low in both sexes (approximate to 11%-12%). The available information suggests that T. amboinensis uses pure search promiscuity as a mating system. This hypothesis needs to be formally tested with mating behavior observations and field measurements on the movement pattern of both sexes of the species. Further detailed studies on the lifestyle and sexual system of all the species within this genus and the development of a molecular phylogeny are necessary to elucidate the evolutionary history of gender expression in the genus Thor.

Experimental evidence shows that the mechanism of pore formation by actinoporins is a multistep process, involving binding of the water-soluble monomer to the membrane and subsequent oligomerization on the membrane surface, leading to the formation of a functional pore. However, as for other eukaryotic pore-forming toxins, the molecular details of the mechanism of membrane insertion and oligomerization are not clear. In order to obtain further insight with regard to the structure-function relationship in sticholysins, we designed and produced three cysteine mutants of recombinant sticholysin I (rStI) in relevant functional regions for membrane interaction: StI E2C and StI F15C (in the N-terminal region) and StI R52C (in the membrane binding site). The conformational characterization derived from fluorescence and CD spectroscopic studies of StI E2C, StI F15C and StI R52C suggests that replacement of these residues by Cys in rStI did not noticeably change the conformation of the protein. The substitution by Cys of Arg(52) in the phosphocholine-binding site, provoked noticeable changes in rStI permeabilizing activity; however, the substitutions in the N-terminal region (Glu(2), Phe(15)) did not modify the toxin`s permeabilizing ability. The presence of a dimerized population stabilized by a disulfide bond in the StI E2C mutant showed higher pore-forming activity than when the protein is in the monomeric state, suggesting that sticholysins pre-ensembled at the N-terminal region could facilitate pore formation. Equacionar aquecimento global, escassez de alimentos e a crescente necessidade energética, tornou-se, atualmente, o grande desafio mundial. Existem diversas culturas agrícolas que podem ser exploradas de maneira estratégica e assim colaborar com a solução deste problema. Dentre elas, pode-se destacar a cultura do girassol (Helianthus Annuus). O girassol é uma das quatro maiores culturas oleaginosas no mundo, cultivado com sucesso nos cinco continentes, ocupando uma área de cultivo superior a 22 milhões de hectares. A participação do Brasil nesse montante é inferior a 1%. Acredita-se que essa pequena participação se deva a fatores sócio-econômicos e tecnológicos. Salienta-se, porém que o Brasil, por suas vantagens comparativas naturais e vantagens competitivas construídas possui condições favoráveis para seu desenvolvimento. Diante dos fatos, o objetivo deste trabalho é aprofundar o conhecimento da cadeia produtiva do girassol e através de sua utilização como estratégia de competitividade, avaliar de maneira sistêmica os impactos na matriz agrícola do País. Dentre as inúmeras vantagens dessa cultura, pode-se destacar: características agronômicas, físicas, químicas, organolépticas e versatilidade, que permitem a utilização e otimização dos fatores de produção já disponíveis; época de plantio (adaptabilidade a diferentes condições edafoclimáticas), podendo ser cultivado desde o Rio Grande do Sul até o Estado de Roraima; sistema radicular (raiz pode chegar a dois metros de profundidade), permitindo o melhor aproveitamento dos nutrientes e da água do solo e promovendo a reciclagem de nutrientes; alto teor de óleo nas sementes (30% a 55%) e; alto valor comercial dos co-produtos. Esse conjunto de características é analisado sob a ótica da teoria das vantagens competitivas e das economias de escala e escopo, demonstrando que com inteligência e pragmatismo, a cultura do girassol pode repetir, com vantagens, o que a soja representou para o agronegócio brasileiro.

The mobilization of food reserves in storage tissues and allocation of their hydrolysis products in the growing axis are critical processes for the establishment of seedlings after germination. Therefore, it is crucial for mobilization of reserves to be synchronized with the growing axis, so that photosynthetic activity can be started before depletion of reserves. For this, integrative approaches involving different reserves, different hydrolysis products and interaction between storage and growing axis tissues, either through hormones or metabolites with signaling role, can contribute greatly to the elucidation of the regulation mechanisms for reserve mobilization. In this study, was hypothesized that hormones and metabolites have different actions on reserve mobilization, and there must be a crossed effect of sugars on the mobilization of proteins and amino acids on lipids and starch mobilization in sunflower seedlings. This study was conducted with seeds of sunflower (Helianthus annuus L.) hybrid Helio 253 using in vitro culture system. Seeds were germinated on Germitest® paper and grown on agar-water 4 g/L without addition of nutrients during 9 days after imbibition (DAI) for growth curve. To verify the effect of metabolites and hormones, seedlings were transferred in the 2nd DAI to agar-water 4 g/L supplemented with increasing concentrations of sucrose or L-glutamine, abscisic acid, gibberellic acid or indolebutyric acid. The results of this study confirm that the mobilization of lipids and storage proteins occurs in a coordinated manner during post-germination growth in sunflower, corroborating the hypothesis that the application of external carbon (sucrose) and nitrogen (L-glutamine) sources can delay the mobilization of these reserves in a crossed way. Moreover, considering the changes in the patterns of reserve mobilization and partition of their products in seedlings treated with different growth regulators, it is evident that the effects of metabolites and hormones must involve, at least in part, distinct mechanisms of action

Six new sesquiterpene lactones, annuolide H ( 3), helivypolides F, H-J ( 4, 11-13), and helieudesmanolide A ( 6), together with known compounds, were isolated from polar bioactive fractions of Helianthus annuus cv. SH-222 and Stella fresh leaf water extracts. Spectroscopic analysis of the new data for 1,2-anhydroniveusin A and 1-methoxy-4,5-dihydroniveusin A corrects some previous assignments. The compounds were tested using the etiolated wheat coleoptile bioassay, and the most active compounds were assayed in standard target species ( STS) ( Lepidium sativum, Allium cepa, Lactuca sativa, Lycopersicon esculentum, and Triticum aestivum) from 5 x 10(-4) to 10(-5) M. The most phytotoxic compounds were helivypolide F and 15-hydroxy-3-dehydrodeoxyfruticin, both of which have a carbonyl group at C-3 conjugated with two double bonds.

Verificou-se o comportamento de cultivares de girassol em relação ao ataque de Chlosyne lacinia saundersii em condições de campo, avaliando-se o número de lagartas por planta, numero de lagartas por metro linear de plantas, porcentagem de plantas atacadas e porcentagem de desfolha. Concluiu-se que: a) os métodos mais adequados para a avaliação da infestação desse inseto foram a porcentagem de plantas atacadas e o número de lagartas por metro linear de plantas; b) as cultivares de girassol menos infestadas pela praga foram 'Cargill-33', 'Contissol', 'Issanka-P, 'PIGB', 'Uruguai' e 'Contissol-621', enquanto 'Contissol-711% 'Contissol-112' e 'Rumano-P4' foram as mais infestadas e, c) a ocorrência de C. lacinia saundersii foi maior na segunda época de cultivo (fevereiro a junho) do que na primeira (outubro a fevereiro).

Sistemas autossustentáveis favorecem as populações microbianas devido à conservação e ao aumento da matéria orgânica no solo. Além disso, as plantas que fazem parte desses sistemas promovem o efeito rizosférico, por meio da zona de influência das raízes, que resulta no aumento da atividade e na modificação da população microbiana. O objetivo deste trabalho foi avaliar o efeito da rotação de culturas de inverno sobre sequências de verão, em sistema de semeadura direta, nos atributos bioquímicos (amilase, urease, celulase e protease) e químicos (carbono orgânico total - COT, carboidratos totais e proteínas totais) em solo rizosférico (SR) e não rizosférico (SNR). Este estudo foi constituído de três culturas de inverno: milho (Zea mays L.), girassol (Helianthus anuus L.) e guandu (Cajanus cajan (L.) Millsp), que estavam em rotação sobre três sequências de verão: soja/soja (Glycine max L.), milho/milho e soja/milho, e duas posições no solo: solo aderido às raízes das plantas (SR) e solo da entrelinha de plantio (SNR). As atividades da amilase, celulase, protease e urease no SR foram 16, 85, 62 e 100 % maiores do que no SNR; para COT e proteínas totais a diferença foi de 21 %. Das culturas de inverno, o milho foi a que mais estimulou as atividades das enzimas amilase, celulase, urease e protease no SR, bem como a atividade das enzimas amilase, urease e protease no SNR. de modo geral, os teores de proteínas totais não foram influenciados pelas culturas de inverno e pelas sequências de verão; os carboidratos totais foram influenciados pelas culturas de inverno milho e girassol. Para o COT houve influência apenas da sequência de verão milho/milho. Os atributos bioquímicos e químicos avaliados neste estudo podem ser utilizados como indicadores das alterações no solo promovidas pelas culturas de inverno e pelas sequências de verão.
