Neuronal communication relies on synaptic vesicles undergoing regulated exocytosis and recycling for multiple rounds of fusion. Whether all synaptic vesicles have identical protein content has been challenged, suggesting that their recycling ability may differ greatly. Botulinum neurotoxin type-A (BoNT/A) is a highly potent neurotoxin that is internalized in synaptic vesicles at motor nerve terminals and induces flaccid paralysis. Recently, BoNT/A was also shown to undergo retrograde transport, suggesting it might enter a specific pool of synaptic vesicles with a retrograde trafficking fate. Using high-resolution microscopy techniques including electron microscopy and single molecule imaging, we found that the BoNT/A binding domain is internalized within a subset of vesicles that only partially co-localize with cholera toxin B-subunit and have markedly reduced VAMP2 immunoreactivity. Synaptic vesicles loaded with pHrodo-BoNT/A-Hc exhibited a significantly reduced ability to fuse with the plasma membrane in mouse hippocampal nerve terminals when compared with pHrodo-dextran-containing synaptic vesicles and pHrodo-labeled anti-GFP nanobodies bound to VAMP2-pHluorin or vGlut-pHluorin. Similar results were also obtained at the amphibian neuromuscular junction. These results reveal that BoNT/A is internalized in a subpopulation of synaptic vesicles that are not destined to recycle, highlighting the existence of significant molecular and functional heterogeneity between synaptic vesicles.

During a 1995 aerial video survey of the coastline of Johnstone Strait, an unusual shoreline feature was noted and termed “clam terraces” (inset) because of the terrace-type morphology and the apparent association with high clam productivity on the sandflats. Typical alongshore lengths of the terrace ridges are 20-50m, and across-shore widths are typically 20-40m. An area with an especially high density of clam terraces was noted in the Broughton Archipelago, between Broughton and Gilford Islands of southeastern Queen Charlotte Strait. Clam terraces in this area were inventoried from the aerial video imagery to quantify their distribution. The terraces accounted for over 14 km of shoreline and 365 clam terraces were documented. A three-day field survey by a coastal geomorphologist, archeologist and marine biologist was conducted to document the features and determine their origin. Nine clam terraces were surveyed. The field observations confirmed that: the ridges are comprised of boulder/cobblesized material, ridge crests are typically in the range of 1-1.5m above chart datum, sandflats are comprised almost entirely of shell fragments (barnacles and clams) and sandflats have very high shellfish production. There are an abundance of shell middens in the area (over 175) suggesting that the shellfish associated with the terraces were an important food source of aboriginal peoples. The origin of the ridges is unknown; they appear to be a relict feature in that they are not actively being modified by present-day processes. The ridges may be a relict sea-ice feature, although the mechanics of ridge formation is uncertain. Sand accumulates behind the ridge because the supply rate of the shell fragments exceeds the dispersal rate in these low energy environments. The high density areas of clam terraces correspond to high density areas of shell middens, and it is probable that the clam terraces were subjected to some degree of modification by aboriginal shellfish gatherers over the thousands of years of occupation in the region. Sand seatrout (Cynoscion arenarius) and silver seatrout (C. nothus) are both found within the immediate offshore areas of the Gulf of Mexico, especially around Texas; however information is limited on how much distributional overlap really occurs between these species. In order to investigate spatial and seasonal differences between species, we analyzed twenty years of bay and offshore trawl data collected by biologists of the Coastal Fisheries Division, Texas Parks and Wildlife Department. Sand seatrout and silver seatrout were distributed differently among offshore sampling areas, and salinity and water depth appeared to correlate with their distribution. Additionally, within the northernmost sampling area of the gulf waters, water depth correlated significantly with the presence of silver seatrout, which were found at deeper depths than sand seatrout. There was also an overall significant decrease in silver seatrout abundance during the summer season, when temperatures were at their highest, and this decrease may have indicated a migration farther offshore. Sand seatrout abundance had an inverse relationship with salinity and water depth offshore. In addition, sand seatrout abundance was highest in bays with direct passes to the gulf and correlated with corresponding abundance in offshore areas. These data highlight the seasonal and spatial differences in abundance between sand and silver seatrout and relate these differences to the hydrological and geological features found along the Texas coastline.

Recreational creel survey data from 28,923 intercepts collected from Biscayne National Park, Florida and surrounding waters were analyzed for January 1976 through July 1991, prior to disruptions caused by Hurricane Andrew in 1992. A total of 261,268 fish and shellfish representing 170 species or higher taxa were recorded. The average trip landed 9.03 fish and/or shellfish. Mean annual landings per angler were 4.77 fish/angler/trip (from 3.8 in 1991 to 5.83 in 1981) and dropped significantly for each of the 2 years following Florida's adoption of mutiple new minimum size limits in 1985 and 1990. The relative contribution to total numerical landings by recreational party type were: skilled anglers (34.0%), food (19.8%), family (14.5%), novice (11.5%), spearfishing (10.3%), lobstering (9.6%), and other (0.3%). FIve species or higher taxa accounted for more than 50% of total landings by number: white grunt, Haemulon plumieri, 15.8%; spiny lobster, Panulirus argus, (10.6%; gray snapper, Lutjanus griseus, 10.6%; unidentified grunts, Haemulon spp., 7.3%; and dolphin, Coryphaena hippurus, 6.6%. An average of 4.39 fish or shellfish were reported released per trip. Five taxa accounted for 67% of all releases. Lobster divers reported the highest average release rate (5.73 per trip) and spearfishing the lowest (0.70 per trip). The ratio of releases to landings was 0.49:1 for all taxa, but ranged from 0.03:1 for dolphin to 1.19:1 for unidentified grunts. Spearfishing accounted for 12.0% of the total fishing trips sampled but only 10.3% of the total number organisms landed and 7.6% of all organisms caught. Hogfish, Lachnolaimus maximus, accounted for 49% if total spearfishing landings (13,286 of 27,015) and 84.3% of total 15,762 hogfish landed.

Catch and mesh selectivity of wire-meshed fish traps were tested for eleven different mesh sizes ranging from 13 X 13 mm (0.5 x 0.5") to 76 x 152 mm (3 X 6"). A total of 1,810 fish (757 kg) representing 85 species and 28 families were captured during 330 trap hauls off southeastern Florida from December 1986 to July 1988. Mesh size significantly affected catches. The 1.5" hexagonal mesh caught the most fish by number, weight, and value. Catches tended to decline as meshes got smaller or larger. Individual fish size increased with larger meshes. Laboratory mesh retention experiments showed relationships between mesh shape and size and individual retention for snapper (Lutjanidae), grouper (Serranidae), jack (Carangidae), porgy (Sparidae), and surgeonfish (Acanthuridae). These relationships may be used to predict the effect of mesh sizes on catch rates. Because mesh size and shape greatly influenced catchability, regulating mesh size may provide a useful basis for managing the commercial trap fishery.

This is the episodic variations in stream water chemistry associated with acid rainfall and run-off and the effect on aquatic ecosystems, with particular reference to fish populations in North West England produced by the North West Water Authority in 1985. This report looks at the biological, physical and chemical information collected over a five year period from over 100 sites on upland streams in the North West Region of which drained rocks of low buffering capacity. In both Lake District and South Pennine sites striking differences were found between the composition of invertebrate communities inhabiting acid-stressed and less acid-stressed streams. Electric fishing surveys showed that acidic streams (geometric mean pH <5.5) generally had abnormally low densities of salmonids ( < 0 .2m2) and that 0+ fish were very few or absent. The latter indicates recruitment failure. Salmon were more sensitive than trout to low pH.

植物克隆生长型主要由遗传结构决定,不同的克隆植物具有不同的克隆生长型,同时植物的克隆生长型又依具体生境和个体发育阶段不同而变化。Harper认为植物构件的结构由分枝角度、节间长度和芽的成活率决定。大量的研究表明克隆植物生长型主要由3个形态参数决定:节间长度、分枝角度和分枝强度。 植物的生境条件无论在时间还是空间上都是异质性的,即使在很小的尺度上这种异质性也是存在的。在这种具有异质性的生境条件下几乎所有植物都有表型可塑性。植物的表型可塑性是指植物在不同的环境因子条件下,在形态、生物量、生理等方面产生的一系列不同。表现型可塑性是植物种群克服环境异质性的重要途径,也是克隆植物实现觅养行为的途径。克隆植物的觅养行为是通过根茎或匍匐茎的长度和分枝强度的变化以及生物量的分配来实现其迁移和对生境的选择,从而将分株安置在各种微环境中。 克隆植物构型的可塑性可使它在土壤水分斑块性分布的环境中通过克隆生长,调整对不同斑块的土壤水分获取对策。在一田间实验中,匍匐茎草本蛇莓(Duchesnea indica Focke)经历了不同土壤水分水平(土壤最大含水量的40%、60%、80%、100%等)处理,以研究土壤水分对蛇莓克隆构型的影响。结果表明:间隔子长度、分株密度、分枝角度和分枝强度呈二次曲线变化,土壤含水为最大含水量的80%为最适生境。 克隆植物构型的可塑性可使它在养分斑块性分布的环境中通过克隆生长调整相应于对不同斑块的养分获取对策。在一田间实验中,蛇莓经历了不同土壤养分水平(高、中、低和对照)处理,以研究土壤养分对蛇莓克隆构型的影响。结果表明:随着土壤养分水平的增加,间隔子的长度、分枝角度均逐渐降低,分枝强度和分株密度增加。 克隆植物构型的可塑性可使它在不同光强的环境中通过克隆生长,调整其对不同光强的资源获取对策。在控制性光资源异质性生境(模拟浓密林荫、稀疏林荫、农田间套作、裸地等)下,研究和模拟了光资源的时空异质性和蛇莓的克隆可塑性变化。结果表明:随着光照强度的增加,间隔子长度的长度逐渐降低,分枝角度、分株强度呈二次曲线变化。 克隆植物构型的可塑性可使它在不同海拔的环境中通过克隆生长,调整其对不同海拔的资源获取对策。在一海拔高度实验中,匍匐茎草本蛇莓经历了不同海拔(400 m、800 m、1200 m和1600 m)处理,以研究不同海拔对蛇莓克隆构型的影响。结果表明:随着海拔的增加,间隔子长度、分株密度、分枝角度和分枝强度呈二次曲线变化。 从克隆植物生长环境(小气候)定量分析了克隆植物生长必需资源如水分、养分、光强、海拔等的变化特性。具体描述和分析了异质环境尺度的大小和等级数。在前人研究成果的基础上,验证了克隆植物生长必需资源分布异质性的数学模型并建立相应的运算模块。在不同水分、养分、光强和海拔等异质性生境中,蛇莓克隆构型相关特征的可塑性变化可用动态Logistic模型进行模拟和预测,拟合效果较好。结合植物对环境异质性的利用对策,对所揭示的蛇莓克隆构型可塑性进行了讨论。 用分形技术描述了蛇莓在资源斑块性分布的生境中,通过克隆生长调整相应于对不同斑块的资源获取对策。植物克隆构型的形态特征在一定尺度范围内具有自相似特征。蛇莓克隆构型的分形维数直接反映了在异质性生境中蛇莓克隆生长的差异。蛇莓克隆生长越发达分形维数越高。相对小的分形维数,反映出蛇莓克隆生长相对较弱。基于计算机图像技术和分形理论,建立了植物克隆生长分形度量的计算机模型,实现了对植物克隆生长过程的计算机模拟。模拟的蛇莓克隆生长形态与实际生长不仅具有相近的分形维数,而且形态也非常相似。利用克隆生长模型的预测能力克服实验生态学难以逾越的某些研究盲点,其研究成果将对克隆植物利用资源异质性的生态对策研究具有重要的指导意义。
