Nitric oxide synthase (NOS) inhibitors are largely used to evaluate the NO contribution to pulmonary allergy, but contrasting data have been reported. In this study, pharmacological, biochemical and pharmacokinetic assays were performed to compare the effects of acute and long-term treatment of BALB/C mice with the non-selective NOS inhibitor L-NAME in ovalbumin (OVA)-challenged mice. Acute L-NAME treatment (50 mg/kg, gavage) significantly reduced the eosinophil number in bronchoalveolar lavage fluid (BALF). The inducible NOS (iNOS) inhibitor aminoguanidine (20 mg/kg/day in the drinking water) also significantly reduced the eosinophil number in BALF In contrast, 3-week L-NAME treatment (50 and 150 mg/kg/day in the drinking water) significantly increased the pulmonary eosinophil influx. The constitutive NOS (cNOS) activity in brain and lungs was reduced by both acute and 3-week L-NAME treatments. The pulmonary iNOS activity was reduced by acute L-NAME (or aminoguanidine), but unaffected by 3-week L-NAME treatment. Acute L-NAME (or aminoguanidine) treatment was more efficient to reduce the NO(x) levels compared with 3-week L-NAME treatment. The pharmacokinetic study revealed that L-NAME is not bioavailable when given orally. After acute L-NAME intake, serum concentrations of the metabolite N(omega)-nitro-L-arginine decreased from 30 min to 24 h. In the 3-week L-NAME treatment, the N(omega)-nitro-L-arginine concentration was close to the detection limit. In conclusion, 3-week treatment with L-NAME yields low serum N(omega)-nitro-L-arginine concentrations, causing preferential inhibition of cNOS activity. Therefore, eosinophil influx potentiation by 3-week L-NAME treatment may reflect removal of protective cNOS-derived NO, with no interference on the ongoing inflammation due to iNOS-derived NO. FORMA DE ESTUDO: Experimental. MATERIAL E MÉTODO: Transplante homógeno de traquéia foi realizado em 51 cobaias (Cavia Porcellus). RESULTADO: Observamos reepitelização do enxerto a partir da traquéia receptora da cobaia, evoluindo de epitélio escamoso de 1 a 3 células nos animais com 3, 7 e 14 dias de observação para epitélio ciliado após 60 a 120 dias do transplante. A viabilidade do enxerto traqueal era boa, com reabsorção parcial da cartilagem e formação de estenose traqueal parcial, mas sem sinais clínicos evidentes de obstrução respiratória durante o tempo de observação dos animais.

O estudo teve como objetivo comparar a eficácia do mel e do açúcar mascavo na cicatrização de feridas induzidas cirurgicamente na pele de cobaias, para avaliar o tempo de cicatrização, as variações macroscópicas e microscópicas do tecido cicatricial, a contração das feridas e o grau de contaminação. Quinze cobaias da raça Abissínia, machos inteiros, de seis meses de idade e pesando entre 500 a 850 gramas, foram divididas em três grupos com cinco animais cada, nos quais foram induzidas, cirurgicamente, três feridas em cada animal com dimensão de 1,5cm² e distância entre elas de 1,5cm, na região dorsal. A ferida F1 foi tratada diariamente com mel, a ferida F2 com solução salina 0,9% (controle) e a ferida F3 com o açúcar mascavo. A eutanásia foi realizada no Grupo 1 ao sétimo dia após a cirurgia, no Grupo 2 ao décimo quarto dia e no Grupo 3 no vigésimo primeiro dia após a cirurgia, sendo as biópsias encaminhadas para avaliação histopatológica. Foram também realizados swabs das feridas em todos os grupos no terceiro e sétimo dias após a cirurgia e as amostras enviadas para a cultura e isolamento bacteriano. Concluiu-se que as feridas tratadas com o mel cicatrizaram mais rápido em relação às tratadas com o açúcar mascavo, demonstrando a maior propriedade cicatrizante do mel em relação ao açúcar mascavo.

In this work, we investigated the effects of He-Ne laser irradiation on the inflammatory process induced in the articular cartilage of the right knee of guinea pigs. Through electron microscopy analysis it was possible to identify the induced arthritis in the articular cartilage and its modification after the laser treatment. The laser radiation promoted a reduction in the proliferation of the inflammatory cells in the damaged tissue and also induced the formation of cartilage bridges that tied the destroyed parts favoring the formation of a repaired tissue in the injured cartilage. Comparamos os métodos de Imunofluorescência Indireta (IFI), Imunodifusão Radial Dupla e Aglutinação, para a pesquisa de anticorpos em soros, na tripanossomíase experimental por Trypanosoma evansi, em cobaias. Foram obtidas 20 amostras de soro correspondentes às 4 primeiras semanas de infecção. A IFI foi positiva em apenas 6 animais, com títulos variando de 1:4 a 1:16. Os títulos mais altos foram observados na 3ª semana pós-infecção. Anticorpos aglutinantes foram observados a partir da 1ª semana pós-infecção e, após a 2ª semana, todos os animais apresentaram reação de aglutinação positiva, com títulos variando de 1:8.000 a 1:250.000. O tratamento dos soros com 2-Mercapto-etanol inibiu a reação de aglutinação, sugerindo ser IgM a principal classe dos anticorpos presentes no soro dos animais infectados. Não se constatou a presença de anticorpos precipitantes durante todo o curso da infecção.

Amostra de Paracoccidioides brasiliensis isolada de vísceras (baço e fígado) de um tatu (Dasipus novencinctus) foi estudada do ponto de vista micológico e imunoquímico. O tatu havia sido capturado em área da usina hidroelétrica de Tucuruí (Estado do Pará). Este já havia sido considerado como reservatório enzoótico do Paracoccidioides brasiliensis naquela região. Esta amostra, conservada na Micoteca do Instituto de Medicina Tropical de São Paulo sob o número 135, apresenta todas as características de Paracoccidioides brasiliensis, com elevado poder antigênico e baixa virulência para cobaios e ratos Wistar. A demonstração do exo-antígeno específico do P. brasiliensis, representado pela glicoproteína de peso molecular 43 kDa, foi evidente através das técnicas de Imunodifusão Dupla, Imunoeletroforese, SDS-PAGE e Imunoblotting.

We report biological data of two generations of Amblyomma triste in laboratory and compared the suitability of different host species. Infestations by larval and nymphal stages were performed on guinea pigs (Cavia porcellus), chickens (Gallus gallus), rats (Rattus norvegicus), rabbits (Oryctolagus cuniculus), wild mice (Calomys callosus), dogs (Canis familiaris) and capybaras (Hydrochaeris hydrochaeris). Infestations by adult ticks were performed on dogs, capybaras and rabbits. Tick developmental periods were observed in an incubator at 27degreesC and RH 90%. Guinea pigs were the most suitable hosts for larvae and nymphs, followed by chickens. The remaining host species were less suitable for immature ticks as fewer engorged ticks were recovered from them. Mean larval feeding periods varied from 3.8 to 4.7 d between different host species. Mean larval premolt periods ranged from 8.9 to 10.4 d. Nymphal mean feeding periods varied from 4.2 to 6.2 d for ticks fed on different host species. Premolt period of male nymphs (mean: 15.4 d) was significantly longer than that of female nymphs (14.7 d). Female nymphs were significantly heavier than male nymphs. The overall sex ratio of the adult ticks emerged from nymphs was 0.9:1 (M:F). Capybaras were the most suitable host for the tick adult stage as significantly more engorged females were recovered from them and these females were significantly heavier than those recovered from dogs or rabbits. The life cycle of A. triste in laboratory could be completed in an average period of 155 d. The potential role of guinea pigs, birds and capybaras, as hosts for A. triste in nature, is discussed.

1. The present study was carried out to determine the target cells and tissues for anti-tick immunoglobulins using an indirect immunohistochemical technique.2. Sections in triplicate prepared from unfed ticks Rhipicephalus appendiculatus, R. evertsi and Amblyomma variegatum were used to assess the cross-reactivity of serum from guineapigs naturally infested with these tick species or immunized against them.3. The sections showed slight (+) to strong (++++) labelling of several structures in the tick body, e.g. salivary gland, gut lumen and malpighian tubules, depending on the serum used.4. The immune serum resulting from the immunization of guinea pigs with an extract of unfed nymphs of R. appendiculatus ticks showed the most intense cross-reactivity with the sections examined.

P>The reactivity of sera collected from guinea pigs after three infestations with Amblyomma triste nymphs on histological sections of the same tick species was investigated through immunohistochemistry to identify potential target cells and tissues. Six guinea pigs were infested thrice, at 30 day intervals, with 30 nymphs of A. triste per animal per infestation. Blood samples were collected from the guinea pigs 15 days after each infestation for serum separation; normal serum was obtained before the first infestation as control. Unfed A. triste nymphs' histological sections were submitted to indirect immunohistochemistry technique by using normal or hyperimmune guinea pig serum as primary antibody and a goat IgG-alkaline phosphatase-APase conjugate as secondary antibody. A weak to moderate APase activity was observed in cells of salivary glands, midgut and haemolymph of unfed nymphs incubated with hyperimmune serum.

The disposition of the abdominal aorta branching in Mesocricetus auratus is described, establishing variation groups with relation to the celiac, cranial mesenteric, renal, genital and caudal mesenteric arteries. Sixty animals (30 males and 30 females) of different ages and weights, were anesthetized with chloroform, injected with contrasting substance in the abdominal aorta (50 animals with Neoprene latex and 10 with a radioopaque mass), after which they were dissected with the help of a stereoscopic microscope. The animals with radioopaque masses were radiographed in comparison with the other animals. The results are expressed in relative percentage figures and compared with other mammalian arterial dispositions.

Thrombosis was induced in the jugular vein of guinea pigs by damage of the epithelium with a sclerosing solution. The effect of heparin, Dextran 70, acetylsalicylic acid, dipyridamole and phenylbutazone in preventing the development of the thrombus was investigated. Only heparin in high doses showed this effect.
