A divergência genética é um dos mais importantes parâmetros avaliados por melhoristas de plantas na fase inicial de um programa de melhoramento genético. O objetivo deste trabalho foi caracterizar 15 acessos de mamoneira por meio de caracteres morfoagronômicos. O experimento foi conduzido em Lavras, MG, no período de fevereiro a agosto de 2008. O delineamento experimental foi o de blocos ao acaso, com três repetições, e 25 plantas por parcela. Os caracteres avaliados foram: altura da planta, altura do caule, número de internódios, diâmetro do caule e número de racemos. Verificou-se a ocorrência de diferenças significativas pelo teste de F (P < 0,01), para o efeito de acessos para todas as variáveis estudadas. Foram estimadas as distâncias genéticas entre os acessos pelo método euclidiano. de acordo com o agrupamento, utilizando o método de Tocher e o método Hierárquico do Vizinho Mais Próximo, baseado na distância euclidiana houve a formação de quatro grupos distintos. Com base nos resultados obtidos neste trabalho, recomendam-se os cruzamentos entre acessos dos grupos I e IV, II e IV, e III e IV.

A comparative cytogenetic and allozyme analysis of sympatric specimens of Mugil rubrioculus and M. curema from Venezuela is reported. Specimens of M. rubrioculus exhibit a 2n=48 karyotype with exclusively acrocentric (NF=48) chromosomes, one pair of NORs interstitially located on chromosome pair number 8 and constitutive heterochromatin distributed in pericentromeric position of all chromosomes. Specimens of M. curema show cytogenetic features significantly different in comparison to M. rubrioculus in terms of chromosome number and morphology (2n=24 biarmed chromosomes, NF=48) and NORs location (telomeric region of the largest metacentric pair). Starch gel electrophoresis analysis at 20 presumptive loci reveals a reduced genetic difterentiation between the two species. In fact, though a total of ten private alleles are identified; all loci share alleles between the two species and the obtained Nei's genetic distance (D= 0.060) is lower than the values obtained between other congeneric mullet species. Thus, the cytogenetic and allozyme data sets indicate quite different degrees of genetic divergence between M. rubrioculus and M. curema. This could either reflect an underestimate of molecular divergence owing to cryptic variation or different rates of molecular/chromosomal evolution. Whatever the explanation, this study confirms the power of karyological data in discriminating species of Mugilidae.

The analysis of interactions between lineages at varying levels of genetic divergence can provide insights into the process of speciation through the accumulation of incompatible mutations. Ring species, and especially the Ensatina eschscholtzii system exemplify this approach. The plethodontid salamanders E. eschscholtzii xanthoptica and E. eschscholtzii platensis hybridize in the central Sierran foothills of California. We compared the genetic structure across two transects (southern and northern Calaveras Co.), one of which was resampled over 20 years, and examined diagnostic molecular markers (eight allozyme loci and mitochondrial DNA) and a diagnostic quantitative trait (color pattern). Key results across all studies were: (1) cline centers for all markers were coincident and the zones were narrow, with width estimates of 730 m to 2000 m; (2) cline centers at the northern Calaveras transect were coincident between 1981 and 2001, demonstrating repeatability over five generations; (3) there were very few if any putative F1s, but a relatively high number of backcrossed individuals in the central portion of transects: and (4) we found substantial linkage disequilibrium in all three studies and strong heterozygote deficit both in northern Calaveras, in 2001, and southern Calaveras. Both linkage disequilibrium and heterozygote deficit showed maximum values near the center of the zones. Using estimates of cline width and dispersal, we infer strong selection against hybrids. This is sufficient to promote accumulation of differences at loci that are neutral or under divergent selection, but would still allow for introgression of adaptive alleles. The evidence for strong but incomplete isolation across this centrally located contact is consistent with theory suggesting a gradual increase in postzygotic incompatibility between allopatric populations subject to divergent selection and reinforces the value of Ensatina as a system for the study of divergence and speciation at multiple stages. © 2005 The Society for the Study of Evolution. Levels of genetic variability for in situ and ex situ genetic conservation were estimated in a population of Myracrodruon urundeuva using the PCR (polymerase chain reaction) technique with the AFLP (Amplified fragment-length polymorphism) genetic marker. Seeds for progeny tests were collected from 30 open-pollination trees (matrices) at Paulo de Faria Ecological Station - SP. From this genetic material, three progeny tests were installed on the Teaching and Research Farm of Ilha Solteira Faculty of Engineering - University of São Paulo State (UNESP), which is located in Selvlria - MS, Brazil. The analysis by genetic marker was conducted with three combinations of different starters EcoRl-Msel, resulting in a total number of 137 polymorphic bands, thus forming a table of binary data. These data were used for the analysis of genetic divergence and distance between progenies. High levels of genetic divergence were observed among families. Based on the Analysis of Molecular Variance (AMOVA), it was shown that 16.2% of genetic diversity is found among progenies and 83.8% within progenies, which suggests deviances of random matings. The grouping of progenies, based on genetic distances, suggests that progenies deriving from trees which are close to each other tend to be more similar. This, in turn, indicates that the population originating the seeds may be genetically structured.

The objective of this work was to study the effect of selective thinning on the genetic divergence in progenies of Pinus caribaea var. bahamensis, aiming to identify the most productive and divergent progenies for the use of improvement program. The test of progenies containing 119 progenies and two commercial controls were planted in March 1990, using 11 × 11 square lattice design, sextuple, partially balanced, disposed in lineal plots with six trees in the spacing of 3,0 × 3,0m. 13 years after planting thinning was realized (selection for DBH), with 50% selection intensity based on Multi-effect index, leaving three trees per plot in all the experiment. The evaluations were done at four situations: A (before the thinning); B (thinned trees); C (remaining trees after thinning) and D (one year after thinning). The analyzed traits were: height, diameter at breast height (DBH), volume, form of stem and wood density. The genetic divergence among the progenies was studied with aid of the canonical variables and of clustering of Tocher method, using the generalized distance matrix of Mahalanobis (D2) as estimate of the genetic similarity. The progenies were grouped in four groups in situation A, fourteen in the situation B, two in the situation C and three in the situation D. The selective thinning of the trees within of the progenies caused a change in the genetic divergence among the progenies, genetically homogenizing the progenies, as demonstrated by the generalized distances of Mahalanobis, clustering of Tocher' and canonical variables methods. The thinning made possible a high uniformity in respect to the relative contribution of the traits for the total genetic divergence. The techniques of clustering were efficient to identify groups of divergent progenies for the use hybridization and little divergent progenies for the use in backcross program.

The experiment was conducted in the experimental area belonging to the Section of Crop Production and Aromatic Medicinal Plants of the FCAV-UNESP, Jaboticabal Campus - Sao Paulo, Brazil. Tolerance to high temperature was studied in six determinate genotypes of tomato (Lycopersicon esculentum L.): Agrocica 8, Apex 1000, Botu-13, Calmech VFAS, Nemadoro and Jab-2, which were cultivated in a greenhouse at temperatures above 33oC for at least 2 h/day during blooming. The objectives of the study were to identify the genetic diversity of the genotypes studied and to determine their performance associated with tolerance to high temperature. Dissimilarity was determined by the generalized Mahalanobis distance. Delineation groups were optimized with the Tocher technique. A random block design was utilized with six treatments and with three replications. Two similarity groups were identified: 1 - Apex 1000, Botu-13, Calmech VFAS, Jab-2, Nemadoro and 2 - Agrocica 8. Crossing of genotypes within one group has no advantage because little genetic divergence and no heterotic response would be expected. However, the crossing of genotypes between groups is suggested. Knowledge of these groups will be important for efficiency future breeding efforts.

The study was conducted at the Section of Crop Production and Aromatic Medicinal Plants of FCAV-UNESP, Jaboticabal Campus - Sao Paulo, Brazil. Nine indeterminate tomato (Lycopersicon esculentum Mill.) genotypes, Jumbo, Santa Clara, Cláudia VF, Concorde, Débora Plus, FM-9, Carmen, Príncipe Gigante, and CL 5915, were evaluated for high temperature tolerance. Three determinate tomato genotypes, FM-9, Suncoast and TSW-10, were cultivated in a greenhouse at more than 33°C air temperature for at least 2 h/day during bloom. The objective was to identify variable genotypes to determine their tolerance of high temperature. Four replications of fifteen treatments were planted in a randomized block design. Dissimilarity was determined by the generalized Mahalanobis distance. Delineation groups were optimized with the Tocher technique. The tomatoes were classified into six groups of similar temperature responses. CL 5915 was the most tolerant of high temperatures. Crossing of genotypes within one group has no advantage because little genetic divergence and no heterotic response would be expected. However, the crossing of genotypes between groups is suggested. Knowledge of these groups will be important for efficient future breeding efforts.

In addition to the evaluations among genotypes, the use of multivariate techniques enables to restrict errors, mainly concerning genetic diversity, and therefore to predict combinations with greater heterotic effect, and the greater possibility of recovery of superior genotypes. The objective of this study was to evaluate the genetic divergence between 18 soybean cultivars based on six morphological characteristics. Path analysis was performed to verify the contribution of direct and indirect characters on grain yield. The Mahalanobis distance has founded techniques of both Tocher Method and dendrogram by Single Linkage. Five different groups were formed: with nine genotypes considered similar among them; while the cultivars CEP 59, Netuno and Urano formed groups isolated by the two grouping methods. The path analysis showed that the indirect characters had little influence on grain yield, with significant direct relationship with mass of 100 grain, and cultivars Tertulha and CEP 53 standing out with grain yields above 3.7 t.ha-1.

Background: The megadiverse Neotropical freshwater ichthyofauna is the richest in the world with approximately 6,000 recognized species. Interestingly, they are distributed among only 17 orders, and almost 80% of them belong to only three orders: Characiformes, Siluriformes and Perciformes. Moreover, evidence based on molecular data has shown that most of the diversification of the Neotropical ichthyofauna occurred recently. These characteristics make the taxonomy and identification of this fauna a great challenge, even when using molecular approaches. In this context, the present study aimed to test the effectiveness of the barcoding methodology (COI gene) to identify the mega diverse freshwater fish fauna from the Neotropical region. For this purpose, 254 species of fishes were analyzed from the Upper Parana River basin, an area representative of the larger Neotropical region.Results: Of the 254 species analyzed, 252 were correctly identified by their barcode sequences (99.2%). The main K2P intra- and inter-specific genetic divergence values (0.3% and 6.8%, respectively) were relatively low compared with similar values reported in the literature, reflecting the higher number of closely related species belonging to a few higher taxa and their recent radiation. Moreover, for 84 pairs of species that showed low levels of genetic divergence (<2%), application of a complementary character-based nucleotide diagnostic approach proved useful in discriminating them. Additionally, 14 species displayed high intra-specific genetic divergence (>2%), pointing to at least 23 strong candidates for new species.Conclusions: Our study is the first to examine a large number of freshwater fish species from the Neotropical area, including a large number of closely related species. The results confirmed the efficacy of the barcoding methodology to identify a recently radiated, megadiverse fauna, discriminating 99.2% of the analyzed species. The power of the barcode sequences to identify species, even with low interspecific divergence, gives us an idea of the distribution of inter-specific genetic divergence in these megadiverse fauna. The results also revealed hidden genetic divergences suggestive of reproductive isolation and putative cryptic speciation in some species (23 candidates for new species). Finally, our study constituted an important contribution to the international Barcoding of Life (iBOL.org) project, providing barcode sequences for use in identification of these species by experts and non-experts, and allowing them to be available for use in other applications. © 2013 Pereira et al.; licensee BioMed Central Ltd.
