The efficiency of systems composed of two species of floating aquatic macrophyte (Eichhornia crassipes and Pistia stratiotes) to treat effluents from shrimp culture was evaluated in this research. The effluent originated from a pond populated with Macrobrachium amazonicum. Treatment systems consisted of 12 experimental tanks with aquatic macrophyte and three tanks without plants (control). Water samples were collected from the fresh water supply and before and after passing through the treatment systems. There are no differences in the removal of nutrients between the two species. The higher nutrient removal was observed for total phosphorus (41.9% by control; 71.6% by E. crassipes; 69.9 by P. stratiotes; 72.5% by E. crassipes + P. stratiotes and 72.1 by P. stratiotes + E. crassipes) and turbidity (30.6% by control; 80.2% by E. crassipes; 75.2 by P. stratiotes; 79.8% by E. crassipes + P. stratiotes and 81.5 by P. stratiotes + E. crassipes). The systems containing aquatic macrophytes were efficient in nitrogen and phosphorus removal from M. amazonicum culture. © 2008 Sociedade Brasileira de Zootecnia.

Numerous studies use major element concentrations measured on continental margin sediments to reconstruct terrestrial climate variations. The choice and interpretation of climate proxies however differ from site to site. Here we map the concentrations of major elements (Ca, Fe, Al, Si, Ti, K) in Atlantic surface sediments (36 degrees N-49 degrees S) to assess the factors influencing the geochemistry of Atlantic hemipelagic sediments and the potential of elemental ratios to reconstruct different terrestrial climate regimes. High concentrations of terrigenous elements and low Ca concentrations along the African and South American margins reflect the dominance of terrigenous input in these regions. Single element concentrations and elemental ratios including Ca (e. g., Fe/Ca) are too sensitive to dilution effects (enhanced biological productivity, carbonate dissolution) to allow reliable reconstructions of terrestrial climate. Other elemental ratios reflect the composition of terrigenous material and mirror the climatic conditions within the continental catchment areas. The Atlantic distribution of Ti/Al supports its use as a proxy for eolian versus fluvial input in regions of dust deposition that are not affected by the input of mafic rock material. The spatial distributions of Al/Si and Fe/K reflect the relative input of intensively weathered material from humid regions versus slightly weathered particles from drier areas. High biogenic opal input however influences the Al/Si ratio. Fe/K is sensitive to the input of mafic material and the topography of Andean river drainage basins. Both ratios are suitable to reconstruct African and South American climatic zones characterized by different intensities of chemical weathering in well-understood environmental settings.

Within this doctoral thesis, biogenic emissions of several globally relevant halocarbons (methyl chloride, methyl bromide, methyl iodide, dibromomethane, chloroform and bromoform) have been investigated in different environments. An airborne study was focused on the tropical rainforest ecosystem, while shipborne measurements investigated naturally occurring oceanic plankton blooms. Laboratory experiments using dried plant material were made to elucidate abiotic production mechanisms occurring in organic matter. Airborne measurements over the tropical rainforest of Suriname and French Guyana (3 - 6 °N, 51 - 59 °W) revealed net fluxes of 9.5 (± 3.8 2σ) µg m-2 h-1 methyl chloride and 0.35 (± 0.15 2σ) µg m-2 h-1 chloroform emitted in the long dry season (October) 2005. An extrapolation of these numbers to all tropical forests helped to narrow down the range of the recently discovered and poorly quantified methyl chloride source from tropical ecosystems. The value for methyl chloride obtained (1.5 (± 0.6 2σ) Tg yr-1) affirms that the contribution of the tropical forest ecosystem is the major source in the global budget of methyl chloride. The extrapolated global net chloroform flux from tropical forests (56 (± 23 2σ) Gg yr-1) is of minor importance (5 - 10 %) compared to the global sources. A source of methyl bromide from this region could not be verified. The abiotic formation of methyl chloride and methyl bromide from dead plant material was tested in a laboratory study. The release from plant tissue representative of grassland, deciduous forest, agricultural areas and coastal salt marshes (hay, ash, tomato and saltwort) has been monitored. Incubations at different temperatures (25 - 50 °C) revealed significant emissions even at ambient temperature, and that the emissions increased exponentially with temperature. The strength of the emission was found to be additionally dependent on the availability of halide and the methoxyl group within the plant tissue. However, high water content in the plant material was found to inhibit methyl halide emissions. The abiotic nature of the reaction yielding methyl halides was confirmed by its high activation energy calculated via Arrhenius plots. Shipborne measurements of the atmospheric mixing ratios of methyl chloride, methyl bromide, methyl iodide, dibromomethane and bromoform have been conducted along a South Atlantic transect from the 27.01. - 05.02.2007 to characterize halocarbon emissions from a large-scale natural algae bloom encountered off the coast of Argentina. Mixing ratios of methyl chloride and methyl bromide were not significantly affected by the occurrence of the phytoplankton bloom. Emissions of methyl iodide, dibromomethane and bromoform showed pronounced mixing ratio variations, triggered by phytoplankton abundance. Methyl iodide was strongly correlated with dimethyl sulfide throughout the sampled region. A new technique combining satellite derived biomass marker (chlorophyll a) data with back trajectory analysis was successfully used to attribute variations in mixing ratios to air masses, which recently passed over areas of enhanced biological production.

The effects of abandoned mine drainage (AMD) on streams and responses to remediation efforts were studied using three streams (AMD-impacted, remediated, reference) in both the anthracite and the bituminous coal mining regions of Pennsylvania (USA). Response variables included ecosystem function as well as water chemistry and macroinvertebrate community composition. The bituminous AMD stream was extremely acidic with high dissolved metals concentrations, a prolific mid-summer growth of the filamentous alga, Mougeotia, and .10-fold more chlorophyll than the reference stream. The anthracite AMD stream had a higher pH, substrata coated with iron hydroxide(s), and negligible chlorophyll. Macroinvertebrate communities in the AMD streams were different from the reference streams, the remediated streams, and each other. Relative to the reference stream, the AMD stream(s) had (1) greater gross primary productivity (GPP) in the bituminous region and undetectable GPP in the anthracite region, (2) greater ecosystem respiration in both regions, (3) greatly reduced ammonium uptake and nitrification in both regions, (4) lower nitrate uptake in the bituminous (but not the anthracite) region, (5) more rapid phosphorus removal from the water column in both regions, (6) activities of phosphorus-acquiring, nitrogenacquiring, and hydrolytic-carbon-acquiring enzymes that indicated extreme phosphorus limitation in both regions, and (7) slower oak and maple leaf decomposition in the bituminous region and slower oak decomposition in the anthracite region. Remediation brought chlorophyll concentrations and GPP nearer to values for respective reference streams, depressed ecosystem respiration, restored ammonium uptake, and partially restored nitrification in the bituminous (but not the anthracite) region, reduced nitrate uptake to an undetectable level, restored phosphorus uptake to near normal rates, and brought enzyme activities more in line with the reference stream in the bituminous (but not the anthracite) region. Denitrification was not detected in any stream. Water chemistry and macroinvertebrate community structure analyses capture the impact of AMD at the local reach scale, but functional measures revealed that AMD has ramifications that can cascade to downstream reaches and perhaps to receiving estuaries.

Vertical fluxes of 239+240Pu and 241Am and temporal changes in their inventories in the northwestern Mediterranean Sea have been examined through high-resolution water column sampling coupled with direct measurements of the vertical flux of particle-bound transuranics using time-series sediment traps. Water column profiles of both radionuclides showed well-defined sub-surface maxima (2391240Pu between 100-400 m; 241Am at 100-200 m and 800 m), the depths of which are a result of the different biogeochemical scavenging behavior of the two radionuclides. Comparison of deep water column (0-2,000 m) transuranic inventories with those derived from earlier measurements demonstrate that the total 2391240Pu inventory had not substantially changed between 1976-1990 whereas 241Am had decreased by approximately 24%. Enhanced scavenging of 241Am and a resultant, more rapid removal from the water column relative to 239+240Pu was also supported by the observation of elevated Am/Pu activity ratios in sinking particles collected in sediment traps at depth. Direct measurements of the downward flux of particulate 239+240Pu and 241Am compared with transuranic removal rates derived from observed total water column inventory differences over time, show that particles sinking out of deep waters (1,000-2,000 m) could account for 26-72% of the computed total annual 239+240Pu loss and virtually all of the 241Am removal from the water column. Upper water column (0-200 m) residence times based on direct flux measurements ranged from 20-30 yr for 239+240Pu and 5-10 yr for 241Am. The observation that 241Am/239+240Pu activity ratios in unfiltered Mediterranean seawater are six times lower than those in the north Pacific suggests the existence of a specific mechanism for enhanced scavenging and removal of 241Am from the generally oligotrophic waters of the open Mediterranean. It is proposed that atmospheric inputs of aluminosilicate particles transported by Saharan dust events which frequently occur in the Mediterranean region could enhance the geochemical scavenging and resultant removal of 241Am to the sediments.

Halocarbons from oceanic sources contribute to halogens in the troposphere, and can be transported into the stratosphere where they take part in ozone depletion. This paper presents distribution and sources in the equatorial Atlantic from June and July 2011 of the four compounds bromoform (CHBr3), dibromomethane (CH2Br2), methyl iodide (CH3I) and diiodomethane (CH2I2). Enhanced biological production during the Atlantic Cold Tongue (ACT) season, indicated by phytoplankton pigment concentrations, led to elevated concentrations of CHBr3 of up to 44.7 and up to 9.2 pmol/L for CH2Br2 in surface water, which is comparable to other tropical upwelling systems. While both compounds correlated very well with each other in the surface water, CH2Br2 was often more elevated in greater depth than CHBr3, which showed maxima in the vicinity of the deep chlorophyll maximum. The deeper maximum of CH2Br2 indicates an additional source in comparison to CHBr3 or a slower degradation of CH2Br2. Concentrations of CH3I of up to 12.8 pmol/L in the surface water were measured. In contrary to expectations of a predominantly photochemical source in the tropical ocean, its distribution was mostly in agreement with biological parameters, indicating a biological source. CH2I2 was very low in the near surface water with maximum concentrations of only 3.7 pmol/L. CH2I2 showed distinct maxima in deeper waters similar to CH2Br2. For the first time, diapycnal fluxes of the four halocarbons from the upper thermocline into and out of the mixed layer were determined. These fluxes were low in comparison to the halocarbon sea-to-air fluxes. This indicates that despite the observed maximum concentrations at depth, production in the surface mixed layer is the main oceanic source for all four compounds and one of the main driving factors of their emissions into the atmosphere in the ACT-region. The calculated production rates of the compounds in the mixed layer are 34 ± 65 pmol/m**3/h for CHBr3, 10 ± 12 pmol/m**3/h for CH2Br2, 21 ± 24 pmol/m**3/h for CH3I and 384 ± 318 pmol/m**3/h for CH2I2 determined from 13 depth profiles.

We explore the impact of a latitudinal shift in the westerly wind belt over the Southern Ocean on the Atlantic meridional overturning circulation (AMOC) and on the carbon cycle for Last Glacial Maximum background conditions using a state-of-the-art ocean general circulation model. We find that a southward (northward) shift in the westerly winds leads to an intensification (weakening) of no more than 10% of the AMOC. This response of the ocean physics to shifting winds agrees with other studies starting from preindustrial background climate, but the responsible processes are different. In our setup changes in AMOC seemed to be more pulled by upwelling in the south than pushed by downwelling in the north, opposite to what previous studies with different background climate are suggesting. The net effects of the changes in ocean circulation lead to a rise in atmospheric pCO2 of less than 10 atm for both northward and southward shift in the winds. For northward shifted winds the zone of upwelling of carbon- and nutrient-rich waters in the Southern Ocean is expanded, leading to more CO2 outgassing to the atmosphere but also to an enhanced biological pump in the subpolar region. For southward shifted winds the upwelling region contracts around Antarctica, leading to less nutrient export northward and thus a weakening of the biological pump. These model results do not support the idea that shifts in the westerly wind belt play a dominant role in coupling atmospheric CO2 rise and Antarctic temperature during deglaciation suggested by the ice core data.

A 380 cm long sediment core from Lake Temje (central Yakutia, Eastern Siberia) was studied to infer Holocene palaeoenvironmental change in the extreme periglacial setting of eastern Siberia during the last 10,000 years. Data on sediment composition were used to characterize changes in the depositional environment during the ontogenetic development of the Lake Temje. The analysis of fossil chironomid remains and statistical treatment of chironomid data by the application of a newly developed regional Russian transfer functions provided inferences of mean July air temperatures (T_July) and water depths (WD). Reconstructed WDs show minor changes throughout the core and range between 80 and 120 cm. All the fluctuations in reconstructed water depth lie within the mean error of prediction of the inference model (RMSEP = 0.35) so it is not possible to draw conclusions from the reconstructions. A qualitative and quantitative reconstruction of Holocene climate in central Yakutia recognized three stages of palaeoenvironmental changes. The early Holocene between 10 and 8 ka BP was characterized by colder-than-today and moist summer conditions. Cryotextures in the lake sediments document full freezing of the lake water during the winter time. A general warming trend started around 8.0 ka BP in concert with enhanced biological productivity. Reconstructed mean T_July were equal or up to 1.5 °C higher than today between 6.0 ka and 5.0 ka BP. During the entire late Holocene after 4.8 ka BP, reconstructed mean T_July remained below modern value. Limnological conditions did not change significantly. The inference of a mid-Holocene climate optimum supports scenarios of Holocene climatic changes in the subpolar part of eastern Siberia and indicates climate teleconnections to the North Atlantic realm.

Vertical profiles of dissolved and particulate 230Th and 231Pa were obtained across the Antarctic Circumpolar Current (ACC) in the southern Atlantic. North of the Polar Front, dissolved and total 230Th increase with depth in conformity with published scavenging models. There is no depletion of 230Th or 231Pa in the water column south of the Polar Front, thought to be an area of enhanced biological productivity. 230Th concentrations increase three-fold to the Weddell Sea across the ACC. Dissolved and total 231Pa concentrations are relatively constant below 500 m depth at about 0.3 dpm m**-3, and change little with depth or latitude. The results from the Weddell Gyre are explained by a mixing-scavenging model that takes into account the input of lower Circumpolar Deep Water through upwelling, which is the main source of water in the Weddell Gyre and is enriched in 230Th but not in 231Pa. 230Th accumulates in the Weddell Gyre as a result of a reduction in the scavenging rate and by ingrowth from 234U. Ingrowth is more significant for 230Th than for 231Pa because the residence time of water in the gyre (about 35 years) is similar to the scavenging residence time of Th in the south Atlantic (29 years) but shorter than that of Pa (120 years). It is argued that changes in 230Th accumulation in the past may reflect changes in water residence time and in the formation rate of Weddell Sea Deep Water.

Since the implementation of the activated sludge process for treating wastewater, there has been a reliance on chemical and physical parameters to monitor the system. However, in biological nutrient removal (BNR) processes, the microorganisms responsible for some of the transformations should be used to monitor the processes with the overall goal to achieve better treatment performance. The development of in situ identification and rapid quantification techniques for key microorganisms involved in BNR are required to achieve this goal. This study explored the quantification of Nitrospira, a key organism in the oxidation of nitrite to nitrate in BNR. Two molecular genetic microbial quantification techniques were evaluated: real-time polymerase chain reaction (PCR) and fluorescence in situ hybridisation (FISH) followed by digital image analysis. A correlation between the Nitrospira quantitative data and the nitrate production rate, determined in batch tests, was attempted. The disadvantages and advantages of both methods will be discussed.

A denitrifying microbial consortium was enriched in an anoxically operated, methanol-fed sequencing batch reactor (SBR) fed with a mineral salts medium containing methanol as the sole carbon source and nitrate as the electron acceptor. The SBR was inoculated with sludge from a biological nutrient removal activated sludge plant exhibiting good denitrification. The SBR denitrification rate improved from less than 0.02 mg of NO3-.N mg of mixed-liquor volatile suspended solids (MLVSS)(-1) h(-1) to a steady-state value of 0.06 mg of NO3-.N mg of MLVSS-1 h(-1) over a 7-month operational period. At this time, the enriched microbial community was subjected to stable-isotope probing (SIP) with [C-13] methanol to biomark the DNA of the denitrifiers. The extracted [C-13]DNA and [C-12]DNA from the SIP experiment were separately subjected to full-cycle rRNA analysis. The dominant 16S rRNA gene phylotype (group A clones) in the [C-13]DNA clone library was closely related to those of the obligate methylotrophs Methylobacillus and Methylophilus in the order Methylophilales of the Betaproteobacteria (96 to 97% sequence identities), while the most abundant clone groups in the [C-12]DNA clone library mostly belonged to the family Saprospiraceae in the Bacteroidetes phylum. Oligonucleotide probes for use in fluorescence in situ hybridization (FISH) were designed to specifically target the group A clones and Methylophilales (probes DEN67 and MET1216, respectively) and the Saprospiraceae clones (probe SAP553). Application of these probes to the SBR biomass over the enrichment period demonstrated a strong correlation between the level of SBR denitrification and relative abundance of DEN67-targeted bacteria in the SBR community. By contrast, there was no correlation between the denitrification rate and the relative abundances of the well-known denitrifying genera Hyphomicrobium and Paracoccus or the Saprospiraceae clones visualized by FISH in the SBR biomass. FISH combined with microautoradiography independently confirmed that the DEN67-targeted cells were the dominant bacterial group capable of anoxic [C-14] methanol uptake in the enriched biomass. The well-known denitrification lag period in the methanol-fed SBR was shown to coincide with a lag phase in growth of the DEN67-targeted denitrifying population. We conclude that Methylophilales bacteria are the dominant denitrifiers in our SBR system and likely are important denitrifiers in full-scale methanol-fed denitrifying sludges.

The cause of seasonal failure of a nitrifying municipal landfill leachate treatment plant utilizing a fixed biofilm was investigated by wastewater analyses and batch respirometric tests at every treatment stage. Nitrification of the leachate treatment plant was severely affected by the seasonal temperature variation. High free ammonia (NH3-N) inhibited not only nitrite oxidizing bacteria (NOB) but also ammonia oxidizing bacteria (AOB). In addition, high pH also increased free ammonia concentration to inhibit nitrifying activity especially when the NH4-N level was high. The effects of temperature and free ammonia of landfill leachate on nitrification and nitrite accumulation were investigated with a semi-pilot scale biofilm airlift reactor. Nitrification rate of landfill leachate increased with temperature when free ammonia in the reactor was below the inhibition level for nitrifiers. Leachate was completely nitrified up to a load of 1.5 kg NH4-N m(-3) d(-1) at 28 degrees C. The activity of NOB was inhibited by NH3-N resulting in accumulation of nitrite. NOB activity decreased more than 50% at 0.7 mg NH3-N L-1. Fluorescence in situ hybridization (FISH) was carried out to analyze the population of AOB and NOB in the nitrite accumulating nitrifying biofilm. NOB were located close to AOB by forming small clusters. A significant fraction of AOB identified by probe Nso1225 specifically also hybridized with the Nitrosonlonas specific probe Nsm156. The main NOB were Nitrobacter and Nitrospira which were present in almost equal amounts in the biofilm as identified by simultaneous hybridization with Nitrobacter specific probe Nit3 and Nitrospira specific probe Ntspa662.
