Increased demands on the capacity of the railway network gave rise to new issues related to the dynamic response of railway tracks subjected to moving vehicles. Thus, it becomes important to evaluate the applicability of traditionally used simplified models which have a closed form solution. Regarding simplified models, transversal vibrations of a beam on a visco-elastic foundation subjected to a moving load are considered. Governing equations are obtained by Hamilton’s principle. Shear distortion, rotary inertia and effect of axial force are accounted for. The load is introduced as a time varying force moving at a constant velocity. Transversal vibrations induced by the load are solved by the normal-mode analysis. Reflected waves at the extremities of the full beam are avoided by introduction of semi-infinite elements. Firstly, the critical velocity obtained from this model is compared with results of an undamped Euler- Bernoulli formulation with zero axial force. Contemporaneamente o Homem depara-se com um dos grandes desafios que é o de efetivar a transição para um futuro sustentável. Assim, o setor da energia tem um papel fundamental neste processo de transição, com principal enfoque no setor dos automóveis, sendo este um setor que contribui com elevadas quantidades de gases de efeito estufa libertados para a atmosfera. Também a escassez dos recursos petrolíferos constitui um ponto fundamental no tema apresentado. Com a necessidade de combater esses problemas é que se tem vindo a tentar desenvolver combustíveis renováveis e neutros quanto às emissões. A primeira geração de biocombustíveis obtidos através de culturas agrícolas terrestres preenche em parte esses requisitos, porém, não atinge os valores da procura e ainda competem com a produção de alimentos. Daí o interesse na aposta de uma segunda geração de biocombustíveis produzidos de fontes que não pertencem à cadeia alimentar e são residuais mas, que mesmo assim não permitem satisfazer as necessidades de matériaprima. A terceira geração de biocombustíveis vem justamente responder a estas questões pois assenta em matérias-primas que não competem pela utilização do solo agrícola nem são usadas para fins alimentares, tendo produtividades areais substancialmente superiores às que as culturas convencionais ou biomassas residuais conseguem assegurar. A matéria prima de terceira geração são portanto as microalgas, cujas produtividades em biomassa são extremamente elevadas, para além de produtividades muito superiores em lípidos, hidratos de carbono e/ou outros produtos de valor elevado. No entanto, este tipo de produção de biocombustível ainda enfrenta alguns problemas técnicos que o tornam num processo dispendioso para competir economicamente com outros tipos de produção de biodiesel. Na linha do que foi dito anteriormente, este trabalho apresenta um estudo de viabilidade económica e energética do biodiesel produzido através da Chlorella vulgaris, apresentando as técnicas e resultados de cultivo da Chlorella vulgaris e posteriormente de produção do biodiesel através dos lípidos obtidos através da mesma. Para melhorar a colheita das microalgas, que é uma das fases mais dispendiosas, testou-se o aumento de pH e a adição de um floculante (Pax XL-10), sendo que o primeiro não permitiu obter resultados satisfatórios, enquanto o segundo permitiu obter resultados de rendimento na ordem dos 90%. Mesmo com a melhoria da etapa da colheita, o preço mínimo do biodiesel produzido a partir do óleo de Chlorella vulgaris, com as condições ótimas de cultivo e produtividades máximas encontradas na literatura, foi de 8,76 €/L, pois, na análise económica, o Pax XL-10 revelou-se extremamente caro para utilizar na floculação de microalgas para obtenção de um produto de baixo valor, como é o biodiesel. A não utilização da floculação reduz o preço do biodiesel para 7,85 €/L. O que se pode concluir deste trabalho é que face às técnicas utilizadas, a produção de biodiesel Chlorella vulgaris apenas, não é economicamente viável, pelo que para viabilizar a sustentabilidade do processo seria ainda necessário desenvolver mais esforços no sentido de otimizar a produção de biodiesel, eventualmente associando-a à produção de um outro biocombustível produzido a partir da biomassa extraída residual e/ou da recuperação de outros produtos de maior valor.

RESUMO - Este trabalho consiste numa análise sobre a viabilidade económico-financeira do projecto de construção de uma Unidade de Cuidados Continuados Integrados de Longa Duração e Manutenção pela Fundação COI, que pretende ser uma resposta adequada à diversidade de fenómenos que têm origem no envelhecimento populacional, inserindo-se no modelo de prestação de cuidados de saúde e apoio social preconizado pela Rede Nacional de Cuidados Continuados Integrados. Em termos estruturais, numa primeira fase, o trabalho faz uma análise sobre o estado da arte dos cuidados continuado, nomeadamente através da sua conceptualização e enquadramento, focando os aspectos e características da RNCCI, nomeadamente os aspectos relacionados com a sustentabilidade financeira e os desafios futuros que este sector da saúde se confronta. Numa segunda fase, o trabalho apresenta o projecto de investimento alvo do estudo de caso, mediante o enquadramento histórico e caracterização da Fundação COI, sendo descrita especificamente a tipologia de longa duração e manutenção e o projecto em termos da sua estrutura física e humana. Numa terceira fase, o trabalho estuda a racionalidade do projecto de investimento, através da análise económico-financeira. Tendo-se concluído, com a utilização de indicadores de avaliação e decisão de investimentos, que a ULDM, é viável. Deve-se contudo ter em conta que a sua viabilidade é sensível a hipotéticas alterações de cenários, nomeadamente perante diferentes taxas de ocupação ou alteração do modelo de pagamento. ----------------------- ABSTRACT - This paper consists on analyzing the economical and financial viability of a project of Integrated Continuous Cares on Long-Term and Maintenance Unit (LTMU), managed by COI Foundation, that can create a correct social answer on the various aspects and phenomenon’s that have their origin in an aged population. This kind of project belongs in the role model of social and health cares of the National Network of Integrated Continuous Cares (NNICC). In a first stage the paper is focused on the analysis of the state of the present services of the continuous cares in Portugal, more concretely of his conceptualization and social frame, putting under the lights the aspects and characteristics of the NNICC, more closely the parts related to their financial sustainability and the challenges that this health sector faces nowadays. The next stage is about the investment project applied to this case-study, given his social and historical frame and attending to the specific characteristics of the COI Foundation. It’s also describing its specific LTMU typology under the involved physic and human structure. Finally this paper study the investment project rationality trough scientific methods like its economic–financial analysis. The conclusion, using decision and evaluation procedures that lead to financial indicators, is that the LTMU, is viable. However, the viability of the project is sensible to an hypothetical change of scenario, namely with different rates of change of occupation or type of payment.

De forma a não comprometer o conforto ou a qualidade de vida, nos dias de hoje, é obrigatório que a energia elétrica esteja presente. Sendo indispensável, torna-se necessário assegurar que a sua distribuição seja feita da forma mais qualitativa possível. Uma resposta rápida e eficaz a possíveis falhas que ocorram na rede, irá garantir a tal qualidade de serviço desejada. Para isso, a automatização dos processos é uma grande evolução e objetivo de concretização do setor elétrico. Neste contexto surge o conceito de Smart Grid, que tem como principal objetivo a combinação entre o setor elétrico e a evolução da tecnologia. A par desta característica, estes tipos de redes vêm também trazer evoluções no âmbito ambiental, pois a produção de energia elétrica é feita, maioritariamente, por fontes de energia renovável. Este projeto incide na análise das vantagens técnicas e económicas da inclusão de equipamentos que detêm capacidades de armazenamento de energia, as Baterias de Armazenamento de Energia (BAE), neste tipo de redes. Para tal, procedeu-se à utilização do método do Despacho Económico, que tem como principal objetivo a determinação dos níveis de produção de todas as unidades geradoras do sistema, satisfazendo a carga, ao mais baixo custo de produção. Com este método, foram criados vários cenários de estudo com vista a validar todo o propósito deste projeto. Nesta dissertação, é também realizado um estudo de viabilidade económica destes equipamentos de armazenamento de energia.

We consider an international trade economical model where two firms of different countries compete in quantities and can use three different strategies: (i) repeated collusion, (ii) deviation from the foreigner firm followed by punishment by the home country and then followed by repeated Cournot, or (iii) repeated deviation followed by punishment. In some cases (ii) and (iii) can be interpreted as dumping.We compute the profits of both firms for each strategy and we characterize the economical parameters where each strategy is adopted by the firms.

This work aims to evaluate the feasibility of using image-based cytometry (IBC) in the analysis of algal cell quantification and viability, using Pseudokirchneriella subcapitata as a cell model. Cell concentration was determined by IBC to be in a linear range between 1 × 105 and 8 × 106 cells mL−1. Algal viability was defined on the basis that the intact membrane of viable cells excludes the SYTOX Green (SG) probe. The disruption of membrane integrity represents irreversible damage and consequently results in cell death. Using IBC, we were able to successfully discriminate between live (SG-negative cells) and dead algal cells (heat-treated at 65 °C for 60 min; SG-positive cells). The observed viability of algal populations containing different proportions of killed cells was well correlated (R 2 = 0.994) with the theoretical viability. The validation of the use of this technology was carried out by exposing algal cells of P. subcapitata to a copper stress test for 96 h. IBC allowed us to follow the evolution of cell concentration and the viability of copper-exposed algal populations. This technology overcomes several main drawbacks usually associated with microscopy counting, such as labour-intensive experiments, tedious work and lack of the representativeness of the cell counting. In conclusion, IBC allowed a fast and automated determination of the total number of algal cells and allowed us to analyse viability. This technology can provide a useful tool for a wide variety of fields that utilise microalgae, such as the aquatic toxicology and biotechnology fields.

Coccidioidomycosis is an emerging fungal disease in Brazil; adequate maintenance and authentication of Coccidioides isolates are essential for research into genetic diversity of the environmental organisms, as well as for understanding the human disease. Seventeen Coccidioides isolates maintained under mineral oil since 1975 in the Instituto de Medicina Tropical de São Paulo (IMTSP) culture collection, Brazil, were evaluated with respect to their viability, morphological characteristics and genetic features in order to authenticate these fungal cultures. Only five isolates were viable after almost 30 years, showing typical morphological characteristics, and sequencing analysis using Coi-F and Coi-R primers revealed 99% identity with Coccidioides genera. These five isolates were then preserved in liquid nitrogen and sterile water, and remained viable after two years of storage under these conditions, maintaining the same features.

SUMMARY Inflammation due to Shigella flexneri can cause damage to the colonic mucosa and cell death by necrosis and apoptosis. This bacteria can reach the bloodstream in this way, and the liver through portal veins. Hypoxia is a condition present in many human diseases, and it may induce bacterial translocation from intestinal lumen. We studied the ability of S. flexneri to invade rat hepatocytes and Caco-2 cells both in normoxic and hypoxic microenvironments, as well as morphological and physiological alterations in these cells after infection under hypoxia. We used the primary culture of rat hepatocytes as a model of study. We analyzed the following parameters in normoxic and hypoxic conditions: morphology, cell viability, bacterial recovery and lactate dehydrogenase (LDH) released. The results showed that there were fewer bacteria within the Caco-2 cells than in hepatocytes in normoxic and hypoxic conditions. We observed that the higher the multiplicity of infection (MOI) the greater the bacterial recovery in hepatocytes. The hypoxic condition decreased the bacterial recovery in hepatocytes. The cytotoxicity evaluated by LDH released by cells was significantly higher in cells submitted to hypoxia than normoxia. Caco-2 cells in normoxia released 63% more LDH than hepatocytes. LDH increased 164% when hepatocytes were submitted to hypoxia and just 21% when Caco-2 cells were in the same condition. The apoptosis evaluated by Tunel was significantly higher in cells submitted to hypoxia than normoxia. When comparing hypoxic cells, we obtained more apoptotic hepatocytes than apoptotic Caco-2 cells. Concluding our results contribute to a better knowledge of interactions between studied cells and Shigella flexneri. These data may be useful in the future to define strategies to combat this virulent pathogen.

The in vitro activity of thiabendazole on Ascaris lumbricoides eggs, which were recovered from uteri of worm excreted after chemotherapeutic treatment, was studied. Four concentrations of the drug were used: 1 -- 2.5 -- 5 -- and 10 ppm during 24, 48 and 72 hours of exposure. Subsequently, the eggs were centrifuged, washed three times and H2SO4 0.1N was added. The eggs were maintained in an incubator for 20 days at 28°C. Finally, the percentage of embryonated eggs was determined under a lightmicroscope at a 100X magnification. After 48 and 72 hours of thiabendazole exposure, at a concentration of 10ppm, the drug showed complete inhibition of egg embryonation.

Twenty Coccidioides immitis strains were evaluated. Only 5 of the 20 strains kept under mineral oil maintained their viability while all 5 subcultures preserved in water remained viable and none of the 13 subcultures kept in soil were viable. A 519 bp PCR product from the csa gene confirmed the identity of the strains.

Nowadays, reducing energy consumption is one of the highest priorities and biggest challenges faced worldwide and in particular in the industrial sector. Given the increasing trend of consumption and the current economical crisis, identifying cost reductions on the most energy-intensive sectors has become one of the main concerns among companies and researchers. Particularly in industrial environments, energy consumption is affected by several factors, namely production factors(e.g. equipments), human (e.g. operators experience), environmental (e.g. temperature), among others, which influence the way of how energy is used across the plant. Therefore, several approaches for identifying consumption causes have been suggested and discussed. However, the existing methods only provide guidelines for energy consumption and have shown difficulties in explaining certain energy consumption patterns due to the lack of structure to incorporate context influence, hence are not able to track down the causes of consumption to a process level, where optimization measures can actually take place. This dissertation proposes a new approach to tackle this issue, by on-line estimation of context-based energy consumption models, which are able to map operating context to consumption patterns. Context identification is performed by regression tree algorithms. Energy consumption estimation is achieved by means of a multi-model architecture using multiple RLS algorithms, locally estimated for each operating context. Lastly, the proposed approach is applied to a real cement plant grinding circuit. Experimental results prove the viability of the overall system, regarding both automatic context identification and energy consumption estimation.

The important chemical and food properties conferred to P. angulata make it necessary to conduct studies in seed conservation. This study evaluated the effect of priming of P. angulata seed for varying periods and storage conditions. Lots of seeds were stored in a refrigerator and in ambient conditions for up to 24 months. Some of these seeds were primed before or after storage. The results show that there was variation in moisture content. The germination rate and germination rate index remained high in ambient conditions when primed up to 24 months.

OBJECTIVE: Initial studies with tricyclic antidepressants demonstrated that they jeopardize the immune system activity. Recent studies suggested that selective serotonin reuptake inhibitors would have stimulating immunological effects. Here, we explored the in vitro immunological effects of two antidepressants used in clinical practice, paroxetine (selective serotonin reuptake inhibitor) and bupropion (norepinephrine and dopamine reuptake inhibitor). METHOD: Peripheral blood samples were obtained from 16 healthy volunteers and the peripheral blood mononuclear cells were isolated and cultured in vitro. We evaluated the effects of bupropion and paroxetine on cell viability as well as the ability to suppress phytohemagglutinin-induced lymphocyte proliferation. RESULTS: Both antidepressants produced neither significant effect on cell viability nor on T-cell proliferation. CONCLUSIONS: This could be of valuable information for the clinical practice when these drugs are administered. These results indicate a more favorable effect of such psychopharmacological drugs when compared to reported immunological effects associated with tryciclic antidepressants.
