A Síndrome de Down, ou trissomia do 21 é o distúrbio cromossômico mais freqüente em recém-nascidos. Esta trissomia é na maioria das vezes (95% dos casos) decorrente da não disjunção meiótica materna durante a meiose I. Com exceção da idade materna avançada, outros fatores de risco para não disjunção meiótica não estão bem estabelecidos. Estudos preliminares recentes sugerem que o metabolismo anormal do folato e polimorfismos no gene MTHFR (C677T e A1298C) podem ser fatores de riscos materno para Síndrome de Down. Com objetivo de verificar a freqüência dos polimorfismos no gene MTHFR (C677T e A1298C) e uma possível associação destes, como fatores de risco materno para Síndrome de Down, realizamos um estudo do tipo caso-controle, em 41 mulheres de filhos com a Síndrome de Down (freqüentadoras da APAE) e 39 controles da cidade de Belém-PA. Nossos resultados demonstraram que a distribuição dos genótipos para os dois polimorfismos entre as mães investigadas e controles se encontravam em equilíbrio de Hardy Weinberg. Não encontramos diferenças entre as freqüências nas mães investigadas e mães controles. Esses resultados sugerem que estas mutações isoladas e/ou em forma de haplótipos, não podem ser consideradas como risco materno para Síndrome de Down, para população analisada e sim polimorfismos comuns dentro desta população.

Comunidades vulneráveis estão sujeitas aos impactos causados por eventos climáticos e aos efeitos nocivos que eles provocam como perturbar o funcionamento da sociedade e a possível ocorrência de perdas materiais, econômicas e sociais. Quanto maior a capacidade de adaptar-se, menores as vulnerabilidades. Em Belém, a presença de cursos hídricos dentro da área urbana e a ocupação irregular ao longo das faixas marginais evidenciam os fatores que levam às frequentes enchentes na região. Provavelmente, as enchentes tenham aumentado a partir da postura das instituições públicas, que favoreceram a ocupação de áreas alagáveis da cidade através dos aterramentos e canalização de igarapés. Dessa forma, o processo de periferização foi intensificado nas áreas de várzea e o espaço, profundamente alterado pela ocupação das áreas não edificáveis, como as bordaduras dos cursos d’água urbanos. Em função disso, a pesquisa objetivou definir a vulnerabilidade e a adaptação autônoma aos impactos das inundações considerando indicadores sociais, habitacionais e de saneamento. Foram analisadas as formas estruturais de adaptação, a partir da observação das edificações, suas tipologias e técnicas construtivas. Para tal, a metodologia para a realização do trabalho foi dividida em três etapas, que compreendem a delimitação da área de estudo; a descrição das medidas autônomas contra inundações e a análise da população como produtora destas medidas. Os resultados retratam a tentativa da comunidade de diminuir as perdas sobre algo que a representa valor, ou seja, as tomadas de decisão de adaptação quanto à capacidade e ao tipo de ação da população, onde as medidas adaptativas refletem não somente as habilidades da comunidade, mas uma série de outros indicadores sociais, econômicos e cognitivos.

Supernumerary teeth are a relatively frequent disorder of odontogenesis that can interfere with the development of the germs teeth. This pathology is most common in the maxillary anterior area, called mesiodens, and their prevalence is generally reported to occur more frequently in male than female, in a ratio of 2:1. The aim of this article was to report the clinical procedures performed in the Clinic of Pediatric Dentistry at the Araraquara School of Dentistry when the presence of two mesiodens, radiographically shown, caused retention of permanent anterior teeth, resulting in esthetic and funcional disturb. All the surgical procedures, etiology and the importancy of the early diagnostic for the appropriate treatment was detailed described.

Soil compaction is one of the limiting factors in areas subjected to direct seeding. The method used to break up the compacted layer should disturb the soil as little as possible, as well as maintain the ground cover. The aim of this study was to evaluate the influence of subsoiling, scarification and use of shaft-type furrowing mechanisms when sowing, on preserving the ground cover, water content and soil density, as well as the effects on maize yield in a dystroferic Red Nitosol, cultivated under a system of direct seeding for ten years. The experimental design was of randomised blocks, with eight soil management treatments: subsoiling to a depth of 0.40 m before sowing the winter crop, subsoiling to 0.40 m before sowing the maize, scarification to 0.30 m before the winter crop, scarification to 0.30 m before the maize, scarification to 0.20 m before the winter crop, scarification to 0.20 m before the maize, direct seeding of the maize with a shaft-type furrowing mechanism and direct seeding of the maize using a double disc furrower. There were four replications. Subsoiling and scarification influenced the preservation of the ground cover, soil density and water content immediately after sowing, but did not interfere in plant development or grain yield in the maize crop. The use of shaft-type furrowing mechanisms in the sowing operation had no effect on any of the parameters under study.

Shiftwork-induced sleep deprivation and circadian disruption probably leads to an increase in the production of cytokines and dysregulation of innate immune system, respectively. This project aims evaluating changes in salivary IL-1 beta, cortisol, and melatonin in night workers. Method. Two day and three night healthy workers participated in this study. Sleep was evaluated by actimetry and activity protocols. Saliva was collected at waking and bedtime the last workday and the following two days-off and was analyzed by ELISA. Results. Neither sleep duration nor efficiency showed any association with salivary IL-1beta. IL-1beta levels were higher at waking than at bedtime during working days for all workers, but only one day and one night-worker maintained this pattern and hormone rhythms during days off. For this night worker, melatonin levels were shifted to daytime. A second one presented clear alterations in IL-1beta and hormone rhythms on days-off. Conclusions. Our preliminary results suggest that night work can disturb the variation pattern of salivary IL-1beta. No association of this variation with sleep was observed. It seems that disruption in hormone rhythms interfere with salivary IL-1beta production. IL-1beta production pattern seems to be maintained when rhythms are present, in spite of a shift in melatonin secretion.

Das Hepatitis C Virus (HCV) ist ein umhülltes RNA Virus aus der Familie der Flaviviridae. Sein Genom kodiert für ein ca. 3000 Aminosäuren langes Polyprotein, welches co- und posttranslational in seine funktionellen Einheiten gespalten wird. Eines dieser viralen Proteine ist NS5A. Es handelt sich hierbei um ein stark phosphoryliertes Protein, das eine amphipatische α-Helix im Amino-Terminus trägt, welche für die Membran-Assoziation von NS5A verantwortlich ist. Welche Rolle die Phosphorylierung für die Funktion des Proteins spielt, bzw. welche Funktion NS5A überhaupt ausübt, ist zur Zeit noch unklar. Beobachtungen lassen Vermutungen über eine Funktion von NS5A bei der Resistenz infizierter Zellen gegenüber Interferon-alpha zu. Weiterhin wird vermutet, das NS5A als Komponente des membranständigen HCV Replikasekomplexes an der RNA Replikation beteiligt ist. Das Ziel dieser Doktorarbeit war es, die Funktion von NS5A für die RNA Replikation zu untersuchen. Zu diesem Zweck wurde eine Serie von Phosphorylierungsstellen-Mutanten generiert, die auf Ihre Replikationsfähigkeit und den Phosphorylierungsstatus hin untersucht wurden. Wir fanden, dass bestimmte Serin-Substitutionen im Zentrum von NS5A zu einer gesteigerten RNA Replikation führten, bei gleichzeitig reduzierter NS5A Hyperphosphorylierung. Weiterhin studierten wir den Einfluß von Mutationen in der Amino-terminalen amphipatischen α-Helix von NS5A auf die RNA-Replikation, sowie Phosphorylierung und subzelluläre Lokalisation des Proteins. Wir fanden, dass geringfügige strukturelle Veränderungen der amphipatischen Helix zu einer veränderten subzellulären Lokalisation von NS5A führten, was mit einer reduzierten oder komplett inhibierten RNA Replikation einherging. Zudem interferierten die strukturellen Veränderungen mit der Hyperphosphorylierung des Proteins, was den Schluß nahe legt, dass die amphipatische Helix eine wichtige strukturelle Komponente des Proteins darstellt, die für die korrekte Faltung und Phosphorylierung des Proteins essentiell ist. Als weitere Aspekte wurden die Trans-Komplementationsfähigkeit der verschiedenen viralen Komponenten des HCV Replikasekomplexes untersucht, sowie zelluläre Interaktionspartner von NS5A identifiziert. Zusammenfassend zeigen die Ergebnisse dieser Doktorarbeit, dass NS5A eine wichtige Rolle bei der RNA-Replikation spielt. Diese Funktion wird wahrscheinlich über den Phosphorylierungszustand des Proteins reguliert.

In der vorliegenden Arbeit wurde die Funktion von Dystroglycan in jungen und späten Stadien des sich entwickelnden ZNS untersucht. Hierzu wurden Antikörper generiert, die fähig waren, in vivo die Interaktion zwischen a-und b-Dystroglycan zu stören. Die Antikörper oder Fab-Fragmente wurden in das Mesencephalon oder Auge lebender Hühnerembryonen injiziert, um aus den beobachteten Veränderungen die Funktion des DAG zu untersuchen. Die Fab-Fragmentinjektionen führten zu Hyperproliferation, verbunden mit morphologischen Veränderungen der Neuroepithelzellen und Zunahme der Anzahl postmitotischer Neuronen. Ebenso wurde die basale und apikale Polarität von Neuroepithelzellen beeinflusst. Auch die Axonorientierung der tectobulbären Axone wurde durch die Injektionen gestört. In älteren embryonalen Stadien kam es, durch Fab-Fragmentinjektionen in die Augen von Embryonen, zu strukturellen Veränderungen der Retina, verbunden mit einer breiteren Verteilung des DAG, wie auch der Synapsen innerhalb der OPL. Die retinalen Zelltypen, wie Müller-Gliazellen und Stäbchen-Bipolarzellen, waren abgerundet und hatten ihre typische Zellform verloren. Die Ergebnisse dieser Arbeit zeigen, dass Dystroglycan einen entscheidenden Einfluss auf die Proliferation, Migration, Polarität und Differenzierung der Neuroepithelzellen ausübt. Außerdem zeigen diese Daten, dass Dystroglycan nicht nur in der frühen embryonalen ZNS-Entwicklung eine maßgebliche Rolle spielt, sondern auch in späten Stadien. Die Ähnlichkeit der beobachteten Veränderungen nach Fab-Fragmentinjektionen legt nahe, dass einige Veränderungen im ZNS bestimmter Muskeldystrophieformen, durch Beeinflussung der Neuroepithelzellen im sich entwickelnden ZNS, verursacht werden.

Indo-Pacific region encompasses about 75% of world's coral reefs, but hard coral cover in this region experienced a 32% region-wide decline since 1970s. This great change is primarily ascribable to natural and anthropogenic pressures, including climate change and human activities effects. Coral reef conservation requires management strategies oriented to maintain their diversity and the capacity to provide ecosystem goods and services. Coral reef resilience, i.e. the capacity to recover after disturbances, is critical to their long-term persistence. The aims of the present study were to design and to test field experiments intended to measure changes in recruitment processes, as a fundamental aspect of the coral reef resilience. Recruitment experiments, using artificial panels suspended in the water column, were carried out in two Indo-Pacific locations affected by different disturbances: a new mine in Bangka Island (Indonesia), and the increased sedimentation due to coastal dynamics in Vavvaru Island (Maldives). One (or more) putatively disturbed site(s) was selected to be tested against 3 randomly selected control sites. Panels’ arrangement simulates 2 proximities to living corals, i.e. the sources of propagules: few centimetres and 2 meters over. Panels were deployed simultaneously at each site and left submerged for about five months. Recruits were identified to the lowest possible taxonomic level and recruited assemblages were analysed in terms of percent cover. In general it was not possible to detect significant differences between the benthic assemblages recruited in disturbed and control sites. The high variability observed in recruits assemblages structure among control sites may be so large to mask the possible disturbance effects. Only few taxa showed possible effects of the disturb they undergo. The field tests have highlighted strengths and weaknesses of the proposed approach and, based on these results, some possible improvements were suggested.

The embryonic head development, including the formation of dental structures, is a complex and delicate process guided by specific genetic programs. Genetic changes and environmental factors can disturb the execution of these programs and result in abnormalities in orofacial and dental structures. Orofacial clefts and hypodontia/ oligodontia are examples of such abnormalities frequently seen in dental clinics. An insight into the mechanisms and genes involved in the formation of orofacial and dental structures has been gradually gained by genetic analysis of families and by the use of experimental vertebrate models such as the mouse and chick models. The development of novel clinical therapies for orofacial and dental pathological conditions depends very much on a detailed knowledge of the molecular and cellular processes that are involved in head formation.

The central nervous system (CNS) has long been regarded as an immune privileged organ implying that the immune system avoids the CNS to not disturb its homeostasis, which is critical for proper function of neurons. Meanwhile, it is accepted that immune cells do in fact gain access to the CNS and that immune responses can be mounted within this tissue. However, the unique CNS microenvironment strictly controls these immune reactions starting with tightly controlling immune cell entry into the tissue. The endothelial blood-brain barrier (BBB) and the epithelial blood-cerebrospinal fluid (CSF) barrier, which protect the CNS from the constantly changing milieu within the bloodstream, also strictly control immune cell entry into the CNS. Under physiological conditions, immune cell migration into the CNS is kept at a very low level. In contrast, during a variety of pathological conditions of the CNS such as viral or bacterial infections, or during inflammatory diseases such as multiple sclerosis, immunocompetent cells readily traverse the BBB and likely also the choroid plexus and subsequently enter the CNS parenchyma or CSF spaces. This chapter summarizes our current knowledge of immune cell entry across the blood CNS barriers. A large body of the currently available information on immune cell entry into the CNS has been derived from studying experimental autoimmune encephalomyelitis (EAE), an animal model for multiple sclerosis. Therefore, most of this chapter discussing immune cell entry during CNS pathogenesis refers to observations in the EAE model, allowing for the possibility that other mechanisms of immune cell entry into the CNS might apply under different pathological conditions such as bacterial meningitis or stroke.
